The vulvar labia of ewes naturally infected with ureaplasma were significantly more swollen and red than those of uninfected ewes. Similar changes were observed following experimental infection of previously uninfected ewes. These differences, although statistically significant, were not sufficiently marked to be useful in diagnosing infection by clinical examination.  相似文献

OBJECTIVE: To investigate intrauterine infection of foetuses with Mycobacterium avium subsp paratuberculosis and the presence of infection in mammary secretions of sheep. DESIGN: A study of 142 late-pregnant ewes and their foetuses from two heavily infected flocks. PROCEDURE: Infection of ewes was determined at necropsy by histopathology and culture of tissues and mammary secretions. Antemortem tests (clinical assessment, faecal culture and serology) were also applied. Foetuses from 59 infected ewes and 47 apparently uninfected ewes were examined by culture and histopathology. RESULTS: Five of five ewes with clinical ovine Johne's disease had infected foetuses. Only one of 54 subclinically affected ewes, and none of 47 uninfected ewes had an infected foetus. M a paratuberculosis was cultured from mammary secretions or mammary glands of only two of 76 ewes, both of which were clinical cases and had infected foetuses. CONCLUSION: Although intrauterine or transmammary transmission of Mycobacterium avium subsp paratuberculosis may occur frequently in clinically affected sheep, these are less common in subclinically infected ewes. Therefore these modes of transmission are unlikely to compromise existing control programs for ovine Johne's disease on most farms, especially if programs include the immediate culling of clinically affected sheep.  相似文献

An alternative control option for heartwater (Cowdria ruminantium infection) is the establishment and maintenance of endemic stability which would lessen the existing dependence on acaricides. In an endemically stable state, animals become infected by vaccination or natural challenge at an early age, following which the immunity so created is boosted by continuing tick challenge. In this study, growth rates, health and hematological parameters were monitored at regular intervals for lambs born to two matched groups of ewes until weaning at 4 mo of age. One group of ewes was infected multiple times with Cowdria ruminantium; the other group remained uninfected. The overall mean leucocyte count of lambs born to infected ewes was significantly lower than that of lambs born to uninfected ewes (P=0.04). However, there were few other significant differences in the other hematological data between the two groups. The mean birth weight of single lambs born to uninfected ewes (4.6 kg) was significantly higher than the mean birth weight of single lambs born to infected ewes (4.4 kg) (P=0.02). Trends in milk consumption and growth rates were similar for the two groups, with few significant differences detected. Likewise, there were no significant differences in the incidences of health problems or pre-weaning mortalities between the two groups of lambs. The results of this study indicate that there is no detectable effect on productivity of pre-weaning lambs when their dams are carriers of C. ruminantium--a situation likely to occur in an endemically stable state. Hence, maintenance of endemic stability would be a suitable control option for heartwater.  相似文献

A chronic infection was established in a group of twenty 18-month-old non-reproductive Merino ewes by oral administration of 3000 infective H. contortus larvae twice weekly for 12 weeks. Their live-weights and wool production were compared with those of 20 uninfected ewes grazing the same pasture. In the infected sheep, faecal egg counts increased over a period of 9 weeks to reach a mean of 5000 eggs per gram, accompanied by small, but significant effects on packed-cell volume and live-weight. The effects of infection on wool growth were also small, and not statistically significant, although there was evidence of a faster seasonal decline in wool growth in infected sheep. It was concluded that mortality induced by acute infection is the most important economic effect of this parasite.  相似文献

Eight 18-month-old ewes were infected orally with Listeria monocytogenes between 77 and 91 days of pregnancy. Only one ewe aborted, 10 days after the first infecting dose, at 94 days of gestation; L monocytogenes was isolated from several sites in both its aborted fetuses. Two days after the first infecting dose all the ewes exhibited mild illness and pyrexia lasting for two to three days but the ewe which aborted was seriously ill until nine to 10 days after aborting. Agglutination tests carried out on 2-mercaptoethanol reduced sera revealed a strong immunological response in all the infected ewes but in the ewe which aborted this response was delayed. Four uninfected ewes which were kept as controls remained healthy throughout the experiment and showed no evidence of 2-mercaptoethanol resistant antibodies to L monocytogenes. Growth retardation lines, occurring at the time of and after experimental infection, were found in the bones of 14 of 17 newborn lambs in both the infected and control groups; in the aborted lambs these lines occurred before the infection.  相似文献

Two groups of pregnant ewes were experimentally infected oronasally in midpregnancy. A faecal and an abortion-source isolate of Chlamydophila abortus were used. They were derived from a healthy ewe from a flock with no history of abortion, and from an aborted foetus in a farm with enzootic abortion. As assessed by modified Ziehl-Neelsen (MZN) staining, egg culture, antigen ELISA, the Clearview test and immunohistochemistry, inoculation resulted in placental and/or foetal infection in all ewes. Histopathology revealed placentitis in two and four ewes inoculated with the enteric or abortion-source isolate, respectively, in addition, these samples were immunohistochemically positive for chlamydial antigen. All six ewes infected with the enteric isolate and five of seven ewes infected with the abortion-source isolate showed evidence for a serological response by an indirect ELISA or CFT. Neither chlamydiae nor lesions were detected in the placentae and lambs of the uninfected control ewes, which remained seronegative. Our results suggest that enteric C. abortus can be associated with placental and foetal lesions in sheep.  相似文献

Milk and blood samples from 704 lactating ewes were examined for the diagnosis of Brucella melitensis infection by milk-ELISA, serum-ELISA, RBPT, SAT and culture of milk. Of these ewes, 209 were from brucellosis free sheep flock, 443 from brucellosis infected sheep flock and 52 were from private sheep flocks of which status for brucellosis was not known. All the 209 ewes belonging to uninfected sheep flock were found negative in all the tests and of the remaining 495 ewes 105 were positive in serum-ELISA, 103 in milk-ELISA, 92 in RBPT, 85 in SAT, and B. melitensis biovar-1 was isolated from the milk of 29 ewes. Of the 105 serum-ELISA positive ewes, 99 were positive and 6 were negative in milk-ELISA, whereas of the 103 milk-ELISA positive ewes, 4 were negative in serum-ELISA. All together, 99 ewes were positive and 386 were negative in both the assays while 10 ewes yielded variable results. The specificity of milk-ELISA in brucellosis free flock was 100% and sensitivity and positive predictive value were 96.11% and 94.28%, respectively, in infected flocks. The Brucella antibody levels in milk and serum samples as determined by milk-ELISA and serum-ELISA were correlated significantly. The milk-ELISA for brucellosis appears to be an attractive alternative of serum-ELISA particularly in the lactating ewes.  相似文献

Effectiveness of vaccination in controlling ovine footrot   总被引:2,自引:0,他引:2  
Efficacy of reducing footrot by use of a commercial vaccine was evaluated over 2 yr with 223 ewes from five crossbred genotypes. Ewes were assigned randomly within genotype, age and initial footrot status to a 2 x 3 factorial array of vaccination in the 1st yr and fall vaccination, winter vaccination or control in the 2nd yr. The flock contained a single Bacteroides nodosus serotype (XV) known to be highly cross-reactive with one of the vaccine serotypes (British serotype H). Vaccination reduced footrot incidence by 61% (P less than .01) in the 1st yr and by 45% (P less than .01) in the 2nd yr. Vaccination of previously uninfected ewes did not significantly reduce the rate of new infection, but vaccination of infected ewes reduced re-infection by 92% (P less than .01). Vaccination in the 1st yr failed to produce any carryover or additive protection in the 2nd yr. Genotypes differed in both footrot incidence (P less than .05) and footrot reduction following vaccination (P less than .05), but the effect was inconsistent between years. Vaccination increased serum antibody titers in both years (P less than .01). Among vaccinates in the 1st yr, infected ewes had lower titer levels than uninfected ewes (P less than .01), but no correlation between titer level and incidence was observed in the 2nd yr.  相似文献

Lundén, A., A. Näsholm and A. Uggla: Long term study of Toxoplasma gondii infection in a Swedish sheep flock. Acta vet. scand. 1994,35,273-281.– The infection rate of Toxoplasma gondii was studied during 6 years in a sheep flock in central Sweden. The flock consisted of 165-249 breeding ewes of which 20-35% were lambs less than 1 year old. Most ewes were slaughtered when 5 years old. The sheep were kept indoors from end of September to early May. Lambing took place in March and April. Individual serum samples were collected twice a year, once just before turning the sheep out to pasture in the spring, and again after housing in the autumn. Sera were analysed by ELISA for antibodies to T. gondii. The seroprevalence varied between 10% and 45% during the 6 years of observation. Seroconversion was detected predominantly at the autumn sampling, indicating that in most cases infection was acquired at pasture. Subclinical effects of T. gondii infection on lamb weight, litter size, total litter weight and ewe weight were also studied. Lambs born to chronically infected ewes were lighter at birth than those of uninfected ewes, but this disparity was no longer evident at weaning.  相似文献

The peripheral immune response, and its relationship with the outcome of the infection according to the age of the animal, has been investigated in young lambs and adult ewes experimentally infected with two different doses of Mycobacterium avium subspecies paratuberculosis (Map). Sixteen 1.5-month-old lambs out of 24 and 23 adult ewes out of 30 were orally challenged with an ovine Map field isolate. Animals were divided into two groups: HD, infected with a higher dose of Map and LD, with a lower dose. The remaining animals were used as uninfected control groups. Animals were euthanized at 110-120 and 210-220 days post-infection (dpi). Along the experiment, the humoral response and the specific and non-specific IFN-γ production were assessed. An intradermal skin test (IDT), using avian PPD, was also performed at 90 and 195 dpi. Samples of intestine and related lymphoid tissue were taken for histological, bacteriological and PCR studies. The Ab and IFN-γ production as well as the IDT response appeared earlier and with more intensity in the adult ewes compared to the lambs. The basal non-specific IFN-γ levels increased only in the adult ewes from the HD group. Animals from the LD and HD groups were positive to PCR; however, lesions consistent with paratuberculosis were exclusively observed in the HD group, both in lambs and in adult sheep, but they only progressed to more advanced stages in the former. These results suggest that the peripheral immune response induced by Map infection in the adult ewes is more efficient to control the progression of the infection than in lambs. This could likely be due to the existence of previous contacts with Map or other mycobacteria in the adult sheep compared to the young lambs.  相似文献

OBJECTIVE: To determine whether tracer sheep could be used to detect S strain Mycobacterium avium subsp paratuberculosis on pasture, and to provide further insight into the early stages of infection. DESIGN: A field study on two farms in an endemic area for ovine Johne's disease in New South Wales. Procedure Lambs, weaners and adult ewes were introduced to pasture with varying amounts of M. a. paratuberculosis contamination and monitored using skin tests, gamma interferon assay, faecal culture and serial necropsy of small groups for up to 15 months after first exposure. RESULTS: Culture from tissues was the most sensitive method for detecting early infection in sheep after natural exposure to S strain M. a. paratuberculosis. The organism was detected in at least one introduced sheep from every exposed group, 6 to 12 months after first exposure. Histopathological lesions were detected in only 17% of culture-positive sheep, and only after at least 8 months of exposure. Similarly, antemortem diagnostic tests had low sensitivity during the early stages of naturally acquired infection. There was no evidence of any differences in infection rate between sheep first exposed as neonates, as weaners or as adults. A higher proportion of lambs born to ewes from an infected flock were infected than lambs suckling uninfected ewes introduced to the same infected environment, and infection was detected earlier in these 'resident' lambs. CONCLUSION: These findings indicate that groups of unexposed 'tracer' sheep, tested by culture of tissues at slaughter 6 to 12 months after first exposure, might be a useful way to assess pasture infectivity in control programs for ovine Johne's disease.  相似文献

During the epidemic of foot-and-mouth disease (FMD) in The Netherlands in 2001, a sheep farm was identified that had been subclinically infected with the disease. The FMD virus genome was detected in 12 of 16 probang samples collected from the sheep and the virus was isolated from four of these samples. Linear defects were observed, 1 to 3 cm from the coronary band, in the hooves of several of the sheep. The defects were thought to have been caused by the FMD infection. It was thought that the distance of the defects from the coronary band might be an indication of the time since the animals had been infected. To determine the growth rate of the claws of sheep, the growth of the hoof horn of uninfected lambs and ewes was measured; in the lambs the growth rate was 0.44 mm per day and in the ewes it was 0.29 mm per day.  相似文献

Eleven maiden Merino ewes, free of antibody to bluetongue virus serotype 20 (BTV-20) in agar gel immunodiffusion and serum neutralisation tests, were mated once with a ram. Ten ewes were inoculated with BTV-20 35 to 42 days after service, and one ewe was left as an uninoculated control. One of the inoculated ewes and the control ewe remained uninfected throughout the experiment. Eight of the remaining 9 ewes showed clinical signs ranging from mild to moderate, and the other showed no clinical signs of infection. BTV-20 viraemia was detected in ewes between days 3 and 11 post inoculation, and the serum antibody response was followed. The control ewe and 5 of the 9 infected ewes were pregnant when examined 90 to 97 days after service. Each of these animals produced a normal lamb. There was no evidence of abortion in the remaining 5 ewes, and no transplacental transfer of virus was detected in the lambs of the 5 infected ewes. At necropsy, 46 days after the birth of the last lamb, no gross or microscopic lesions were observed in either the ewes or lambs.  相似文献

Maedi visna virus (MVV) vertical transmission in sheep via infected colostrums is a very important route of infection in lambs. To verify colostral transmission and to study early viral entry in lambs, colostrum samples, and small intestine and mesenteric lymph nodes of lambs born from experimentally infected ewes were examined by histopathology, immunohistochemistry (IHC) and in situ hybridisation (ISH) studies. In particular, newborn lambs were naturally fed maternal colostrum and humanely killed at 10, 24, 48, 72, 96 h and 7 and 10 days after birth; two caesarian-derived lambs served as uninfected controls. No lesions suggestive of MVV infection were found, but marked immunoreactions for MVV capsid antigen (CA, p28) were detected in lambs fed maternal colostrum and in macrophages cultured from colostrum. IHC results in lambs suggest an initial viral absorption by intestinal epithelial cells at the tip of the villi, passage to mononuclear cells in the lamina propria and involvement of ileum Peyers' patches and mesenteric lymph nodes, with different staining patterns depending on infection times. ISH on intestinal sections of the 72 h lamb revealed the presence of proviral DNA in epithelial cells at the tip of the villi, suggesting a role for these cells in early MVV replication. The results contribute to knowledge about the pathogenesis of ovine lentivirus infection suggesting that the small intestine and mesenteric nodes are the sites of entry and propagation of MVV in lambs fed colostrums from infected ewes.  相似文献

The technique of embryo transfer was used to investigate the maternal transmission of scrapie in sheep. Embryo donor ewes were experimentally infected with scrapie (all eventually developing the disease) and artificially inseminated six months later with semen from an uninfected scrapie-susceptible ram. Embryos were harvested five and six days after insemination and transferred by laparoscopy, unwashed, into recipient ewes which had been genetically selected for very low susceptibility to scrapie. Six of the 26 lambs born to these recipients developed scrapie.  相似文献

This study describes responses to artificial infection with Haemonchus contortus in ewes and lambs of 50% Dorset, 25% Rambouillet, and 25% Finn-sheep ancestry and provides estimates of genetic parameters for measures of parasite resistance. One hundred ninety-eight ewes out of 64 sires, and 386 lambs out of 25 sires were evaluated in autumn and spring of 2 yr. Ewes were dewormed shortly after weaning their lambs and lambs were dewormed at about 120 d of age. One week after deworming, ewes and lambs were dosed with approximately 10,000 infective larvae of H. contortus. After infection, BW, fecal egg counts (FEC), and packed cell volume (PCV) were measured weekly for 7 wk in lambs kept in drylot and fortnightly for 11 wk in ewes on pasture. Summary traits were defined as initial PCV, mean BW across all times, and means for PCV (MPCV) and log-transformed FEC (MLFEC) at wk 3 to 7 after infection for lambs and wk 3 to 11 after infection for ewes. Ewes and lambs did not lose weight overall in any year or season, but there was no consistent effect of year or lambing season on mean LFEC or mean PCV during infection in either ewes or lambs. Yearling ewes were less resistant to infection than older ewes, with lower PCV (P < 0.05) and higher LFEC (P < 0.05). During infection, PCV was positively correlated with BW and negatively correlated with LFEC in both ewes and lambs. In lambs, heritabilities were 0.39 (P < 0.01) for PCV, 0.10 (P < 0.05) for LFEC across all measurement times, and 0.19 (P < 0.01) for three measures of LFEC taken at the peak of infection. Heritability estimates for ewes were 0.15 (P < 0.05) for PCV and 0.31 (P < 0.01) for LFEC. Repeatabilities for LFEC and PCV across measurement times were moderate in ewes and lambs. Correlations between dam and lamb records for MLFEC were generally low, suggesting different mechanisms of resistance in lambs and nonlactating ewes. Ewes with higher genetic merit for growth as lambs were less resistant to infection as adults, but genetic merit for fertility and prolificacy were not related to parasite resistance. Lambs with higher genetic merit for body weight were more resistant to infection. Selection for resistance to H. contortus is therefore possible and should not adversely affect growth of lambs and fertility of ewes in this production environment.  相似文献

Attempted venereal transmission of Chlamydia psittaci in sheep   总被引:2,自引:0,他引:2  
Attempts were made to transmit Chlamydia psittaci, the causal agent of enzootic abortion of ewes, in three different ways. Ten ewes were inseminated artificially with freshly collected semen containing 10(5) CELD50 chlamydia. Serological evidence of infection was found two weeks before parturition in nine ewes and the organism was recovered from three of them. By six weeks post partum antibody titres had fallen and were negative in six ewes. Twenty-four hours after service two groups of 10 ewes were infected intravaginally with 10(8) CELD50 and 10(3) CELD50 chlamydia respectively. Positive complement fixation titres were present in the first half of pregnancy in all the ewes in the high dose group but not the low dose group. None of the ewes showed evidence of infection at parturition. Fourteen ewes were served by four rams which had been intravenously infected with 10(8) CELD50 chlamydia four to six days earlier. Following service seroconversion occurred but titres became negative again by late pregnancy. No microbiological evidence of infection was detected in any of the ewes at parturition but complement fixation titres were positive in 12 of 14 ewes sampled six weeks post partum. The 14 ewes were sampled during their pregnancies the next year and none showed any evidence of chlamydial infection. It is concluded that venereal transmission of C psittaci is biologically feasible in sheep, but that under normal systems of flock management in Britain it is unlikely to contribute greatly to the epidemiology of enzootic abortion of ewes.  相似文献

The onset of growth retardation was investigated in fetal lambs following experimental infection of pregnant ewes with Border Disease virus ( ) on day 53 of pregnancy. Fetuses from control and infected ewes were harvested at weekly intervals between day 60 and day 95 of gestation and morphometric studies were completed on tibial radiographs and tibial growth cartilage metaphyseal junctions. Mean tibial areas were significantly reduced (P<0·01) in fetuses from infected ewes at 35 and 42 days after infection and growth cartilage metaphyseal junctions were less mature in fetuses from infected ewes at 42 days after infection. Positive immunostaining for BDV antigen was demonstrated in the brains of all fetuses from infected ewes between 14 and 42 days after infection. Attempts to demonstrate BDV antigen in bone proved unsuccessful. It is concluded that intrauterine growth retardation is an early manifestation of BDV infection in lambs and that the process is initiated shortly following infection of the fetus.  相似文献

A cohort study was conducted in order to measure the effect of the chronic indurative lymphocytic mastitis caused by the South African strain of maedi visna virus (MVV) on the pre-weaning growth of lambs born either of naturally infected or uninfected ewes kept under similar conditions. Fifty naturally infected ewes as well as another 40 from a maedi-visna-free source to be used as control animals, were purchased and kept in separate flocks which were managed in a similar way. All the ewes were of the same breed and 3-4 years old. During the adaptation period, and through the mating, pregnancy and lactation periods they were periodically monitored for the presence of MVV serum antibodies. The lambs were weighed at birth and thereafter every 2 weeks until the age of 90 days, when they were weaned. The ewes were then slaughtered, and their udders examined histologically and the number of lymphocytic follicles were counted and assessed. Although the calculated values indicated a correlation between the number of follicles in the udder and the reduction in the growth rate of the lambs, this was not statistically significant. Similarly, despite higher counts of lymphoid follicles in the udders of sero-positive ewes as compared to those that were sero-negative and the lower ewe productivity indexes in infected ewes, no statistically significant differences were found in the indexes of ewes in different follicle categories.  相似文献

The effects of Trypanosoma brucei brucei infection during the first, second or third trimesters of pregnancy in 13 ewes were studied. All infected ewes were anaemic with the anaemia being most severe, moderate and least in ewes infected in the second, third and first trimesters, respectively. Weight loss occurred in all infected ewes but was most severe in ewes infected in the third trimester. Three of the four ewes infected in the first trimester died without aborting while one aborted and later died. Of the four ewes infected in the second trimester, three died without aborting while one lambed and later died. In the third trimester ewes, one aborted, two lambed and all three later died while one died without aborting. None of the lambs was viable. The control ewe lambed normally. The infection resulted in 16.7% abortion, 100% death and 33.3% neonatal deaths. This study demonstrates that T. brucei brucei has a devastating effect on pregnancy irrespective of the trimester of infection.  相似文献
