黑穗醋栗花粉生活力及萌发力研究   总被引:2,自引:2,他引:2  
TTC法对黑穗醋栗初花期花粉生活力鉴定表明,6种供试材料花粉生活力表现出明显差异;离体培养试验表明,各供试材料在蔗糖的不同浓度培养基中萌发是不一致的,差异极显著.同时发现硼对花粉萌发影响很大;利用荧光显微镜技术,测定了不同传粉时间花粉在柱头上的萌发力.结果表明,体内萌发与离体萌发有相同规律,各试验材料萌发率之间的差异与以上试验结果一致.  相似文献

增施锌硅肥对小麦有显著的减少小穗退化、降低空秕率作用,达到增加每穗粒数、提高产量的目的。过量增施锌硅肥对强筋小麦蛋白质、湿面筋、赖氨酸等营养品质有一定负效应,不利于强筋小麦营养品质的提高。生产上以施用腐殖酸锌硅肥37.5kg/hm2为宜。  相似文献

Nisin is a posttranslationally modified antimicrobial peptide that is widely used as a food preservative. It contains five cyclic thioethers of varying sizes that are installed by a single enzyme, NisC. Reported here are the in vitro reconstitution of the cyclization process and the x-ray crystal structure of the NisC enzyme. The structure reveals similarities in fold and substrate activation with mammalian farnesyl transferases, suggesting that human homologs of NisC posttranslationally modify a cysteine of a protein substrate.  相似文献

为了进一步了解强筋小麦的后熟过程、探明强筋小麦在收获后不同储藏期的品质变化特点,以津强小麦及其衍生系中筛选出的8个强筋小麦品种(系)为试材,在室温条件下分别储藏10,90 d后进行品质测定。结果表明,储藏90 d时,8个品种的稳定时间均超过7 min;储藏10,90 d,小麦的蛋白质含量变异幅度均较小,湿面筋含量均有所增加,平均增幅3.69%,面团吸水率均有所下降,形成时间和稳定时间的平均增幅分别为13.41%和56.34%。t测验结果表明,储藏10,90 d的湿面筋含量和面团稳定时间之间的差异均达显著水平,而蛋白质含量、吸水率、形成时间间的差异均不显著。  相似文献

【目的】探索不同萌发状态小麦麸质蛋白、醇溶蛋白和谷蛋白等主要致敏蛋白含量的动态变化规律,以及醇溶蛋白、谷蛋白亚基的含量变化,为无麸质食品的研发和发芽小麦的利用提供科学依据。【方法】控制发芽条件获得7种不同萌发状态的小麦,采用SDS-PAGE分析不同萌发状态小麦醇溶蛋白(Gliadins)、谷蛋白(Glutenin)的亚基组成变化,通过R5 ELISA和RP-HPLC进一步测定小麦发芽过程中麸质蛋白、醇溶蛋白和谷蛋白亚基的含量变化。【结果】以R5 ELISA法和RP-HPLC法两种方法可有效测定小麦中麸质蛋白、醇溶蛋白和谷蛋白亚基的含量,发芽处理对上述过敏蛋白及亚基有不同程度的影响;麸质蛋白含量在发芽初期变化不大,后期显著减少;ω-醇溶蛋白相对含量变化不显著;α-/β-醇溶蛋白的相对含量在发芽过程中大幅度降低(从未处理小麦籽粒的41.85%降低到发芽处理后的31.51%—35.35%,P<0.01),γ-醇溶蛋白相对含量从31.37%显著增加到发芽处理后的36.69%—39.02%(P<0.05);高分子量麦谷蛋白亚基(high molecular weight glutenin subunit,HMW-GS)和低分子量麦谷蛋白亚基(low molecular weight glutenin subunit,LMW-GS)的相对含量变化不显著,HMW-GS略有减少,从8.66%(未处理组)降低到5.94%(芽长1/4),再回升到7.28%(芽长=籽粒长);LMW-GS略有增加,从8.30%(未处理组)增加到10.45%(芽长=籽粒长)。【结论】R5 ELISA法和RP-HPLC法两种方法都可以有效进行小麦致敏蛋白定量分析;发芽过程中小麦的致敏蛋白含量总体呈下降趋势,特别是在发芽芽长达籽粒长1/2时,含有最多致敏肽的α-/β-醇溶蛋白下降尤为显著,提示小麦进行适度发芽处理可以降低致敏性。  相似文献

在前文研究的基础上,进一步对小麦单细胞培养和未熟胚培养再生植株无性系第7代和第8代子粒蛋白质和干、湿面筋含量进行测定与分析。结果表明,体细胞无性系子粒蛋白含量为14%~17%,干面筋含量为11%~14%,湿面筋含量为32%~42%。面筋与蛋白质含量之间虽然存在正相关关系,但未达显著水平。湿面筋与干面筋含量之间则高度显著正相关。  相似文献

采用离体蛙心灌流和离体回肠灌流的方法将小麦面筋蛋白胃蛋白酶酶解液分别加入蛙心体外循环系统和豚鼠回肠体外循环系统中,通过BL-420生物机能实验系统测定蛙心心肌和肠道平滑肌收缩力变化,并随即加入特异性阿片肽阻断剂——纳洛酮,以检测小麦面筋蛋白胃蛋白酶酶解液的生物学活性,并将两种方法所得的实验结果经处理后进行相关性比较研究。结果表明,小麦面筋蛋白胃蛋白酶酶解液能显著引起蛙心心肌和肠道平滑肌收缩力加强,并可被纳洛酮逆转此现象,说明小麦面筋蛋白胃蛋白酶酶解液含有外啡肽,对两种方法具有显著的相关性,表明用蛙心灌流法测定外啡肽活性与经典的豚鼠回肠法测定结果是一致的,但蛙心灌流法更易推广。  相似文献

本试验分析了浙江省主要小麦品种浙麦1号、浙麦2号、浙麦3号和品系“119”籽粒的物理性状及其面粉品质。结果表明,“119”和浙麦3号的千粒重,容重、角质率都相对较高,而浙麦1号和浙麦2号相对较低。面粉品质属弱面粉或中等面粉,其中“119”小麦的面筋含量较高,面团形成时间、稳定时间和断裂时间都相对较长,面团曲线较平稳,谱带较宽,软化度也较小,总评价值较高,加工品质相对较好。 年度间和不同收获期之间,麦粒的物理性状和面粉品质均有差异。延迟收获的小麦籽粒容重、千粒重、角质率等物理性状和面团的弹性、延伸性变劣,软化度增加,而出粉率和面筋含量稍有提高,可见,小麦产量、籽粒的物理性状和加工品质等以适时收获为好。  相似文献

This study evaluated the quality potential of seven synthetic hexaploid wheats (2n=6x=42, AABBDD) expressing only allelic variation at Glu-D1 of Aegilops tauschii (SHWSD). Major quality parameters related to dough strength, gluten proteins (including high-molecular-weight glutenin subunits (HMW-GS) and low-molecular-weight glutenin subunits (LMW-GS), gliadins), and their ratios between SHWSD and the weak gluten wheat control Chuannong 16 (CN16) were measured in at least three environments (except STD7). The zeleny sedimentation value (ZSV), dough development time (DDT), dough stability time (DST), and farinograph quality number (FQN) of SHWSD were considered stable under different environments, with their respective ranges being 8.00–17.67 mL, 0.57–1.50 min, 0.73–1.80 min, and 9.50–27.00. The ZSV, DDT, DST, and FQN of SHWSD were smaller than those of CN16, suggesting that SHWSD had a weaker dough strength than CN16. Although SHWSD had a lower gluten index than CN16, its wet and dry gluten contents were similar to or even higher than those of CN16 in all environments tested. The protein content of grains (12.81–18.21%) and flours (14.20–20.31%) in SHWSD was higher than that in CN16. The amount of HMW-GS in SHWSD sharply decreased under the expression of fewer HMW-GS genes, and the LMW-GS, gliadins, and total glutenins were simultaneously increased in SHWSD in comparison with CN16. Moreover, SHWSD had higher ratios of LMW-GS/glutenin and gliadin/glutenin but a lower ratio of HMW-GS/glutenin than CN16. These results provide necessary information for the utilization of SHWSD in weak-gluten wheat breeding.  相似文献

 【目的】探讨氮素形态对强筋小麦豫麦34和弱筋小麦豫麦50籽粒内源激素含量的影响。【方法】在盆栽条件下, 设酰胺态氮、铵态氮和硝态氮3种氮素形态处理并加入硝化抑制剂双氰铵(DCD),在小麦开花后定期取样,用酶联免疫法测定籽粒中IAA、GA3、ABA和ZR含量。【结果】酰胺态氮处理下,豫麦34籽粒形成初期,ZR含量较高;籽粒灌浆盛期前,IAA,ABA,GA3含量较高,灌浆速率较早达到高峰,与铵态氮和硝态氮处理相比,粒重分别增加9.16%和5.74%。铵态氮处理下,豫麦50籽粒形成初期,ZR含量较高;籽粒灌浆盛期前,IAA,GA3含量高;籽粒灌浆后期,IAA含量仍较高,籽粒灌浆速率较快且下降较为平缓,与酰胺态氮和硝态氮相比,粒重分别增加3.88%和11.52%。【结论】氮素形态对两个品种的影响不同,酰胺态氮有利于调节豫麦34内源激素水平,促进籽粒灌浆,提高粒重;而铵态氮对豫麦50较为有利。  相似文献

本文汇总了环境与大米汞污染的来源、汞与甲基汞对人体的健康危害、水稻对甲基汞的吸收与累积、包括大米在内的食品中汞的含量与形态分析方法、全球大米汞污染的现状,以及人群摄食大米的甲基汞暴露方面的研究进展。我国贵州和东南亚汞矿开采地区产出的大米存在严重的汞污染,且大米为当地居民的主要粮食,摄食大米给当地居民带来了严重的甲基汞暴露健康风险。未来有必要在全球更广泛地开展大米汞污染与相应的人群摄食暴露健康风险评估工作,并加强水稻对环境中汞的吸收、转运和累积机制以及这一系列过程中汞形态转化方面的研究。  相似文献

以强筋型小麦郑麦9023、中筋型小麦温麦4号和弱筋型小麦豫麦50为试验材料,在大田条件下研究了不同类型小麦籽粒灌浆期碳水化合物、淀粉及其组分含量以及相关酶活性的动态变化。结果表明,不同类型小麦籽粒灌浆期的蔗糖含量均随灌浆进行呈降低趋势,葡萄糖含量在灌浆前期亦呈逐渐降低趋势。郑麦9023以花后6 d籽粒果糖含量最高,而后逐渐降低;豫麦50和温麦4号籽粒的果糖含量亦在花后6 d达到峰值,之后迅速降低,至花后24 d时略有升高,而后又迅速降低。郑麦9023籽粒可溶性总糖含量的变化呈双峰曲线,峰值分别出现在花后6和18 d,且第1个峰值显著高于第2个;而豫麦50和温麦4号均呈单峰曲线,峰值均出现在花后12 d。随着灌浆的进行,不同类型小麦籽粒总淀粉、直链淀粉、支链淀粉含量均呈增加趋势,且豫麦50明显高于郑麦9023和温麦4号。不同类型小麦籽粒腺苷二磷酸葡萄糖焦磷酸化酶(AGPP)、尿苷二磷酸葡萄糖焦磷酸化酶(UGPP)和可溶性淀粉合成酶(SSS)活性的变化均呈单峰曲线,3类小麦AGPP活性的峰值均出现在花后18 d,UGPP和SSS活性的峰值,温麦4号分别出现在花后12和18 d,豫麦50和郑麦9023则分别出现在花后18和12 d。  相似文献

对强筋小麦和弱筋小麦施用不同肥料组合来验证对不同类型小麦的影响。结果表明:强筋小麦的产量、千粒重、穗数和穗粒数随有机肥施用量的增加呈抛物线增加趋势。在N、P、K总量相同的条件下,当化肥与精制有机肥比例为6∶4时弱筋小麦产量最高。在一定程度上减少化肥用量而增施精制有机肥,小麦产量可明显提高。  相似文献

以5个不同品质类型的小麦品种(强筋品种烟农15、郑麦9023、济麦20、中筋品种鲁麦21、弱筋品种扬麦9号)为材料,在大田条件下,研究了3个氮肥水平(0kg/hm2、120kg/hm2、240kg/hm2)对籽粒蛋白质及其组分的影响,结果表明:增加施氮量可显著提高各品质类型小麦籽粒蛋白质及各组分含量。增加施氮量提高强筋小麦籽粒总蛋白质、清蛋白和球蛋白、醇溶蛋白和麦谷蛋白含量较中、弱筋小麦幅度大。各品质类型小麦的谷/醇比是随着施氮量的增加而增加的。  相似文献

Skin color and nutrient photolysis: an evolutionary hypothesis   总被引:7,自引:0,他引:7  
Human populations native to areas of intense sunlight tend to be heavily melanized. Previous explanations for this relationship have invoked only weak selective pressures. To test the hypothesis that dark pigmentation may protect against photolysis of crucial light-sensitive vitamins and metabolites by ultraviolet light, folate was used as a model. It was found that exposure of human plasma in vitro to simulated strong sunlight causes 30 to 50 percent loss of folate within 60 minutes. Furthermore, light-skinned patients exposed to ultraviolet light for dermatologic disorders have abnormally low serum folate concentrations, suggesting that photolysis may also occur in vivo. Deficiency of folate, which occurs in many marginally nourished populations, causes severe anemia, fetal wastage, frank infertility, and maternal mortality. Prevention of ultraviolet photolysis of folate and other light sensitive nutrients by dark skin may be sufficient explanation for the maintenance of this characteristic in human groups indigenous to regions of intense solar radiation.  相似文献

Antibodies were raised against a synthetic peptide corresponding to 14 amino acid residues at the COOH-terminus of a protein deduced from the human c-erbB-2 nucleotide sequence. These antibodies immunoprecipitated a 185-kilodalton glycoprotein from MKN-7 adenocarcinoma cells. Incubation of the immunoprecipitates with (gamma-32P)ATP resulted in the phosphorylation of this protein on tyrosine residues. These results indicate that the human c-erbB-2 gene product is the 185-kilodalton glycoprotein that is associated with tyrosine kinase activity. Although the c-erbB-2 protein was predicted to encode a protein very similar to epidermal growth factor (EGF) receptor, EGF did not stimulate this kinase activity either in vivo or in vitro.  相似文献

功能性水稻的开发与推广应用,对改善我国以稻米为主食人群的营养与健康状况、促进我国农业 产业结构转变、提高农民种粮效益及助推乡村振兴具有重要意义。功能性水稻主要包括有色稻、低谷蛋白水稻、 高抗性淀粉水稻、富微营养水稻和药物制造生物反应器水稻等 5 大类。有色稻由于种子果皮积累花色素苷而呈 紫色或红褐色,有红米和黑米两大类,分别受 2 个和 3 个基因控制,是体弱者药食同源的重要滋补食品,国内 已育成一大批有色稻品种推广应用。低谷蛋白水稻由于稻米谷蛋白含量低,适于肾病患者作主粮使用,目前中 国和日本已育成多个品种用于商业化开发应用。高抗性淀粉水稻由于有部分淀粉无法被人体淀粉酶分解而不能 被肠道吸收,可降低糖尿病患者餐后血糖峰值,已成为糖尿病患者重要的主食,目前国内已育成多个品种推广 应用。富微营养水稻由于稻米特种维生素、有益微量元素等含量高而有益于不同缺素症患者治疗与保健,国内 外已育成以“黄金大米”为代表的多个富微营养水稻品种。药物制造生物反应器水稻利用水稻为药用重组蛋白、 多肽或其他生物活性物质生产平台,合成与提取目标功能成分物质,目前国内外都有成功报道,并已尝试进行 商业化开发应用。全球对功能性水稻的需求日益增大,功能性水稻研究具有广阔前景。功能性水稻育种未来将 从功能的单一型向复合型发展,由保健型向保健与辅助疗效相结合转变。多组学技术和常规育种相结合的方法, 不断挖掘与创制新种质以培育功能性水稻新品种,是功能性水稻研究的发展方向。  相似文献

Effect of Gluten on Pasting Properties of Wheat Starch   总被引:2,自引:0,他引:2  
The effect of gluten on pasting properties of wheat starch was studied to provide a scientific basis for the application of gluten in food production and quality improvement in wheat breeding. The pasting properties of blends were analyzed using PH 1391 wheat starch mixed with five different additions of three kinds of gluten (strong-, medium-, and weak-gluten) and the structures of network were observed with microscope. The significant downtrends of peak viscosity, trough viscosity, final viscosity, area of viscosity, setback, and peak time were observed with the increase in the addition of gluten. In general, the average value of them decreased respectively by 3.6, 4.8, 3.4, 3.8, 4.0, and 1.18% of those corresponding indexes of pure starch for every 2% increase in gluten. The decreasing rate of the indexes mentioned above exceeded more than 2% except peak time, but there were no significant influence of gluten addition on breakdown, pasting temperature and pasting time. The inter layer composed of gluten was not observed when the addition of gluten was 10%, as the compound formed of gluten inlaid in the paste of starch, but obvious inter layer was detected when the addition of gluten was 18%. There was significant or remarkable difference among the effects of three different kinds of gluten on the peak viscosity, trough viscosity, area of viscosity, setback, and peak time, but it had no significant difference among the effects of different glutens on pasting temperature and pasting time. The descending order of the effect of different glutens on peak viscosity, trough viscosity, and area of viscosity was strong-, medium-, and weak-gluten, but the order of them for setback was opposite. Both addition and types of gluten significantly affected peak viscosity, trough viscosity, area of viscosity, setback, and peak time, but there were no significant effects of it on peak time and peak temperature.  相似文献
