In autumn 2000, a study was carried out on 25 dairy farms in the vicinity of Utrecht with the aim to estimate infectivity levels for nematode parasites in cows. On each farm, faecal samples were collected from 15 cows, blood samples from 5 of these and herbage samples from 2 cow pastures. Faecal examination demonstrated a variation between farms and within farms in faecal egg output with a mean number of 4 eggs/g faeces (EPG) and Ostertagia spp. and Cooperia oncophora being the dominant species. In 6 out of 21 farms examined, lungworm larvae were detected in at least 1 cow. Serum pepsinogen values and serology using ELISA's with crude adult Ostertagia, crude adult C. oncophora and a specific recombinant C. oncophora protein as antigens indicated low to moderate infection levels. Pasture infectivity levels varied between farms with again Ostertagia spp. and C. oncophora as the dominant larval types and correlated with the crude worm Ostertagia ELISA, the crude worm Cooperia ELISA and the pepsinogen values. Exposure levels were high enough to enable the possible occurrence of production losses on the majority of farms.  相似文献

Interactions between gastrointestinal worms (Ostertagia ostertagi, Cooperia oncophora) and lungworms (Dictyocaulus viviparus) in calves were studied by assessing the effect of primary infections with either group of worms on the development of homologous or heterologous challenge infections. Primary infections with lungworms resulted in some degree of resistance to challenge with gastrointestinal worms, but this resistance was lower than that found after homologous infection. Primary infections with gastrointestinal worms did not confer any resistance to challenge with lungworms. On the contrary, an indication was found of some enhancing effect of previous gastrointestinal worm infection on the establishment of lungworms. The highest degree of resistance against lungworm challenge was found where calves have been primarily infected with lungworms. Lungworm infections produced some elevation of serum pepsinogen levels. Gastrointestinal worms evoked a rise in circulating eosinophils, although this rise was smaller and occurred later than in lungworm-infected calves. Under the conditions of the experiment, the effect of 6000 infective lungworm larvae on weight gain was larger than the effect of 100,000 L3 of Ostertagia ostertagi and 100,000 L3 of Cooperia oncophora.  相似文献

Control of ostertagiasis is a major economic problem in the temperate areas of the world. An immunological control scheme for cattle is an important aspect of integrated control and its achievement is dependent on our understanding of how immunity is acquired. Also, host mechanisms regulated by Ostertagia need to be understood in order to address immunological questions. Little is known about acquired immunity to Ostertagia infection in cattle. The degree of immunity is incomplete and its development slow. Evidence is accumulating for impairment of antibody and cellular immune responses by Ostertagia which is responsible for the survival of the parasite in the young host. Other experimental work suggests that local factors, such as eosinophil accumulation, mucosal mast cell kinetics and other cellular and humoral immune responses, may play a role in pathogenesis and immunity. At the present time these areas are also totally unexplored in the study of Ostertagia infection.  相似文献

Bovine viral diarrhoea virus (BVDV) is the most prevalent infectious disease of cattle. It causes financial losses from a variety of clinical manifestations and is the subject of a number of mitigation and eradication schemes around the world. The pathogenesis of BVDV infection is complex, with infection pre- and post-gestation leading to different outcomes. Infection of the dam during gestation results in fetal infection, which may lead to embryonic death, teratogenic effects or the birth of persistently infected (PI) calves. PI animals shed BVDV in their excretions and secretions throughout life and are the primary route of transmission of the virus. These animals can usually be readily detected by virus or viral antigen detection assays (RT-PCR, ELISA), except in the immediate post-natal period where colostral antibodies may mask virus presence. PI calves in utero (the ‘Trojan cow’ scenario) currently defy detection with available diagnostic tests, although dams carrying PI calves have been shown to have higher antibody levels than seropositive cows carrying non-PI calves.Acute infection with BVDV results in transient viraemia prior to seroconversion and can lead to reproductive dysfunction and immunosuppression leading to an increased incidence of secondary disease. Antibody assays readily detect virus exposure at the individual level and can also be used in pooled samples (serum and milk) to determine herd exposure or immunity. Diagnostic tests can be used to diagnose clinical cases, establish disease prevalence in groups and detect apparently normal but persistently infected animals. This review outlines the pathogenesis and pathology of BVD viral infection and uses this knowledge to select the best diagnostic tests for clinical diagnosis, monitoring, control and eradication efforts. Test methods, types of samples and problems areas of BVDV diagnosis are discussed.  相似文献

Serum fructosamine concentration falls with an increase in plasma protein turnover. Since this increase is a feature of gastrointestinal parasite infection at all sites, a falling serum fructosamine level may be of general application in the interpretation of parasite-related events. The circulating fructosamine level was investigated in the lamb as an indicator of protein-losing gastroenteropathy associated with two parasites at different sites, Ostertagia circumcincta (abomasum) and Nematodirus spathiger (small intestine). Infection with 10,000 N spathiger infective larvae daily for seven weeks produced only slight clinical signs, and only a small change in serum fructosamine levels. 2500 O circumcincta L3 daily gave no clinical signs in most animals, but serum fructosamine was more strongly affected in this group. Concurrent infection with both organisms caused some degree of diarrhoea in all lambs, and a sustained fall in serum fructosamine, more accentuated than that observed in either of the single infection groups, supporting the value of serum fructosamine determination in monitoring gastrointestinal parasitism.  相似文献

Equine coronavirus (ECoV) is an emerging virus associated clinically and epidemiologically with fever, depression, anorexia and less frequently colic and diarrhoea in adult horses. Sporadic cases and outbreaks have been reported with increased frequency since 2010 from Japan, the USA and more recently from Europe. A faeco‐oral transmission route is suspected and clinical or asymptomatic infected horses appear to be responsible for direct and indirect transmission of ECoV. A presumptive clinical diagnosis of ECoV infection may be suggested by clinical presentation, haematological abnormalities such as leucopenia due to lymphopenia and/or neutropenia. Confirmation of ECoV infection is provided by specific ECoV nucleic acid detection in faeces by quantitative PCR (qPCR) or demonstration of coronavirus antigen by immunohistochemistry or electron microscopy in intestinal biopsy material obtained ante or post mortem. The disease is generally self‐limiting and horses typically recover with symptomatic supportive care. Complications associated with disruption of the gastrointestinal barrier have been reported in some infected horses and include endotoxaemia, septicaemia and hyperammonaemia‐associated encephalopathy. Although specific immunoprophylactic measures have been shown to be effective in disease prevention for closely‐related coronaviruses such as bovine coronavirus (BCoV), such strategies have yet not been investigated for horses and disease prevention is limited to basic biosecurity protocols. This article reviews current knowledge concerning the aetiology, epidemiology, clinical signs, diagnosis, pathology, treatment and prevention of ECoV infection in adult horses.  相似文献

An experiment was conducted to determine the efficiency of a method of experimental infection of weaner beef calves with Bunostomum phlebotomum and to compare such infection with that established by natural infection. Six calves, maintained on a concrete-floored pen, were inoculated with B. phlebotomum L3 by placing the larval inocuulum, in small volume, in the outer chamber of the ear while the animal was restrained for 18 min. Inoculation doses of 20, 30, 40, 50, 60, and 80 thousand L3 were used. Six other calves were grazed on pasture known to be heavily contaminated with hookworm. All animals were killed 72 days after experimental infection and 93 days after initial exposure to pasture infection. The experimental and naturally-infected calves became patent at 55 and 66 days, respectively, after exposure to L3. Red blood cell counts and hemoglobin values were markedly depressed in both groups and lowest values coincided with onset of patency. There was difference in liveweight changes, but both groups lost weight during the prepatent period of infection and gained weight with the onset of patency. The largest number of hookworms was established at the 30000 L3 inoculation level; little or no establishment was observed at the 2 highest levels. Sizeable adult hookworm burdens were established in 4 out of 6 pastured calves. Intestinal pathology was generally more severe in experimentally-infected calves, consisting of a thickened mucosa and masses of punctate, hemorrhagic foci. Pastured calves also acquired large burdens of Ostertagia ostertagi, particularly inhibited early fourth-stage larvae. Moderate to severe abomasal pathology and elevated plasma pepsinogen were associated with ostertagiasis in the pastured calves. The experimental infection method is efficient in establishing high levels of B. phlebotomum infection in calves currently or previously infected with other gastrointestinal nematodes.  相似文献

Groups containing six lambs that previously had been infected five times each week for four, eight or 12 weeks, either with 1000 Trichostrongylus vitrinus third stage larvae (L3) or 1000 T vitrinus L3 and 2500 L3 Teladorsagia (previously Ostertagia) circumcincta were challenged, along with six worm-free control lambs, with 3000 75Se-labelled T vitrinus L3. The lambs were killed 10 days after challenge. Those given a dual infection for four and eight weeks had significantly lower total T vitrinus numbers than monospecifically infected lambs. Significantly fewer radiolabelled larvae were recovered from the dual infected group killed after four weeks exposure to infection than from lambs infected monospecifically. The negative interaction between these two nematode species appears to be physiologically mediated and may be an important factor in accounting for differences in their seasonal prevalence.  相似文献

Laboratory diagnosis of feline leukemia virus (FeLV) and feline immunodeficiency virus (FIV) usually involves both viruses, as the clinical signs are similar and coinfection may occur. Serological methods may not represent an accurate diagnosis: maternal antibodies or cross-reactions may give false positive results to FIV, and false negative results may occur in latent FeLV status, or in certain FIV infection stages. A nested polymerase chain reaction (PCR) technique was designed to detect FeLV, FIV and feline endogenous retrovirus simultaneously. The detection of endogenous sequences was considered indicative of successful DNA extraction. The technique was used to diagnose FIV and FeLV in the blood cells of 179 cats. The kappa value with the serological data was 0.69 for FeLV and 0.87 for FIV. The joint detection of FeLV and FIV by this novel nested PCR is sensitive, specific, fast and convenient, and its applicability for clinical diagnosis is promising, as the direct evidence of the presence of the virus is more realistic than the indirect data provided by the serological detection.  相似文献

Breed susceptibility to nematode infection was evaluated in Angus (Bos taurus) and Brangus (B. indicus crossbreed) cattle. A cow-calf herd and a yearling replacement heifer herd were monitored during one grazing season. Calves were born in March and were weaned in October. Individual rectal fecal samples were collected monthly from the two herds and processed for fecal egg counts (FEC) and coprocultures. Cow and calf FEC increased from April, reaching maximum values during the summer. Angus cows and calves had significantly (p<0.05) greater FEC than Brangus cows and calves, and Haemonchus and Cooperia were the predominant genera. Replacement heifer FEC showed a similar pattern with maximum levels during late summer/fall, and Haemonchus was the predominant genus. No significant differences were seen between breeds, however, infection levels were consistently lower in Brangus heifers. Ostertagia was present in cows and heifers only in fall/winter, which is consistent with summer inhibition. The data suggested that cows were an important source of pasture contamination for their susceptible calves and that the Brangus breed was relatively more resistant to infection. The use of B. indicus crossbreeds may help in alleviating reliance on chemical control by reducing the rate of pasture contamination and subsequent infection losses.  相似文献

This paper describes the signs, clinical pathology, and postmortem findings in 14 young African grey parrots (Psittacus erithacus erithacus) that were naturally infected with psittacine beak and feather disease (PBFD) virus (psittacine circovirus). All but two of the parrots had severe leukopenia at clinical presentation. Two other parrots also had severe anemia. All birds died within 3 wk after presentation. Postmortem examination documented liver necrosis in 11 of 14 birds and secondary bacterial or fungal infections in 9 of 14 birds. Tests for Chlamydia psittaci, polyomavirus, and Salmonella sp. were negative. PBFD viral infection could be demonstrated in all birds by polymerase chain reaction. Supporting evidence of PBFD viral infection was gathered by histologic examination of the bursa of Fabricius, electron microscopy, and DNA in situ hybridization. Electron microscopic examination of both the bursa of Fabricius and liver revealed virus particles resembling circovirus. DNA in situ hybridization of six liver tissue samples confirmed the presence of PBFD virus and excluded the presence of avian polyomavirus. Our findings suggest that a specific presentation of peracute PBFD viral infection, characterized by severe leukopenia, anemia, or pancytopenia and liver necrosis in the absence of feather and beak abnormalities, may occur in young African grey parrots.  相似文献

This is the first report of an intestinal Eimeria infection in Asian seabass (Lates calcarifer) at the histopathological and ultrastructural levels. The Eimeria infection was often associated with severe pathology and significant mortality in the absence of other pathogens. This showed that it is an important disease of juvenile L. calcarifer in small scale nurseries in Vietnam. Heavy infection and high prevalence levels of the Eimeria infection are suspected to be linked to the low daily water exchange rates practised in these nurseries. Although systemic iridovirus infection was concurrently observed in some of the fish examined, it was not as consistently present in diseased fish as the Eimeria infection.  相似文献

SUMMARY: Resistance to a range of benzimidazole anthelmintics was investigated in 2 strains of Ostertagia spp. One strain (SRBO) had been exposed to fenbendazole, oxfendazole and thiabendazole, the other strain (KR79) only to thiabendazole. Both strains showed a high degree of resistance to albendazole, fenbendazole, oxfendazole and thiabendazole, which had efficiencies of 11–38% and 0–60% against all developmental stages of SR80 and KR79, respectively. There was no significant reduction in the KR79 worm count by thiabendazole at 132 mg kg-1 given either as a single dose or divided into 12 equal portions, one administered every 6h. Nematodirus spp were also found to be resistant to all benzimidazoles tested. Levamisole at 7 mg kg-1 and naphthalophos at 30 mg kg-1 had efficiences of 89 and 66%, respectively, against SR80 Ostertagia and 99 and 19% against Nematodirus, all but the last of these being significant reductions in worm burdens. The resistant SR80 Ostertagia occurred on a research station, but possibly originated from a property which 3 years earlier had supplied ewes to the station. The use of benzimidazole anthelmintics and subsequent grazing on worm-free pasture may have enhanced the level of resistance. A relationship was established between egg counts and adult worm counts 10 days post-treatment, which suggested that for Ostertagia the worm count could be predicted from the geometric mean egg count from about 10 animals. Thus, where an Ostertagia population is suspected of being resistant, an anthelmintic efficiency assay using pre-and post-treatment faecal egg counts should provide a satisfactory diagnostic procedure.  相似文献

An unusual epidemiological pattern in bovine ostertagiasis is described in which clinical ostertagiasis occurred in calves grazed on a silage aftermath from late July. The calves were treated with the anthelmintic levamisole prior to grazing the aftermath and the latter had not been grazed since the previous autumn. Analysis of the herbage larval counts suggests that delayed emergence of overwintered Ostertagia ostertagi L3 from the soil was the most likely source of the infection.  相似文献

The sequential development of Type I and Type II ostertagiasis over a 2-year period in the same naturally infected cattle is described for the first time. Particular reference is made to biochemical and serological changes. Positive relationships were demonstrated between the clinical signs of both Type I and Type II disease, and marked increases in the levels of plasma pepsinogen, plasma gastrin and antibody titres to adult Ostertagia antigen. At necropsy, there were significant relationships between the combined total of adult and developing 5th stage larvae of Ostertagia spp. and the levels of both plasma pepsinogen and gastrin. By the end of the second grazing season the cattle had acquired an immunity to infection with Ostertagia spp. and had very low burdens of this parasite at necropsy. However some of these cattle maintained elevated plasma pepsinogen levels when under natural challenge by Ostertagia spp. larvae and the aetiology of these changes and the problems of diagnosis using this parameter are discussed. Similar trends of infection were observed for Cooperia oncophora, although resistance to the parasite developed more rapidly.  相似文献

The significance of the worm burden in the diagnosis of parasitic gastroenteritis is examined. It is emphasised that the worm population is in a dynamic state with the calf constantly losing part of its worm burden while picking up new infective larvae. The numbers and proportion of late fourth stage Ostertagia ostertagi larvae gives an indication of the success of the animal in controlling its worm burden. During an examination of the worm population it is also necessary to look at the condition and degree of development of adult female worms to assess the efficiency of the calf's acquired resistance to infection.  相似文献

Infections with bovine virus diarrhoea virus (BVDV) are widespread throughout the world. Although the prevalence of infection varies among surveys, the infection tends to be endemic in many populations, reaching a maximum level of 1-2% of the cattle being persistently infected (PI) and 60-85% of the cattle being antibody positive. Persistently infected cattle are the main source for transmission of the virus. However, acutely infected cattle as well as other ruminants, either acutely or persistently infected, may transmit the virus. Transmission is most efficient by direct contact. However, as infections have been observed in closed, non-pasturing herds, other transmission routes seem likely to have some practical importance. Differences in BVDV prevalence among regions or introduction of virus in herds previously free of BVDV are often associated with particular epidemiological determinants such as cattle population density, animal trade and pasturing practices. However, on a few occasions there have been no obvious explanations for infection of individual herds. Estimates of economic losses due to BVDV infection vary depending on the immune status of the population and the pathogenicity of the infecting virus strains. Introduction of the infection into a totally susceptible population invariably causes extensive losses until a state of equilibrium is reached. Infection with highly virulent BVDV strains causing severe clinical signs and death after acute infection gives rise to substantial economical losses. At an estimated annual incidence of acute infections of 34%, the total annual losses were estimated as US$ 20 million per million calvings when modeling the losses due to a low-virulent BVDV strain. At the same incidence of infection, the losses due to a high-virulent BVDV strain were estimated as US$ 57 million per million calvings. Low-virulent BVDV infections caused maximum losses at an incidence of 45%, whereas high-virulent BVDV infections caused maximum losses at an incidence of 65%. Thus, cost-benefit analyses of control programs are highly dependent on the risks of new infections under different circumstances and on the strains of the virus involved.  相似文献

The antimuscarinic drug atropine caused a marked fall in plasma pepsinogen values of sheep with burdens of Ostertagia circumcincta and this response was greater in animals which had higher plasma pepsinogen values before administration of the drug. The response was shown to occur whether the elevated plasma pepsinogen values were a consequence of a larval infection in previously naive or exposed animals or of adult parasites directly transplanted into the abomasum of naive lambs.  相似文献

Worm-free Holstein tracer calves were used to assess the seasonal variation in number and course of development of nematode parasites of cattle on an irrigated pasture located in the California Sierra foothills. The most common genera of nematodes found in the abomasum and small intestine were Ostertagia and Cooperia, respectively. The infective Ostertagia larvae on herbage were most numerous in early spring and lowest in summer months. Arrested development of Ostertagia occurred in mid-to-late spring. The percentage of the population of Ostertagia found to be arrested larvae was constant (73% to 95%) over 4 years, even though the total number of nematodes in the genus was markedly different. Levels of infection with Ostertagia and Cooperia were found to correlate with pasture contamination occurring in the preceding fall and concurrent winter months. In the cecum and large intestine, Oesophagostomum venulosum was found to be the most common nematode. It is suggested that this species may replace Os radiatum as the cattle nodular worm in regions where climate is similar to that of the southwestern coast of North America. Evidence of seasonal hypobiotic development of Oe venulosum was found. Other genera of nematodes observed in tracer calves were Trichostrongylus, Haemonchus, Bunostomum, Trichuris, and Dictyocaulus.  相似文献
