Intravenous infusion of Pasteurella hemolytica endotoxin caused marked increases in the plasma levels of thromboxane B2 (TxB2), prostaglandins (PG) and serotonin in sheep. The control values for TxB2, 6-keto-PGF1 alpha, PGF2 alpha, and serotonin before endotoxin infusion averaged 283 +/- 53 (standard error of mean), 281 +/- 14 and 199 +/- 27 pg/ml and 57 +/- 3 ng/ml, respectively. At 50 min during endotoxin infusion, these values were increased to their maximum of 376, 339, 325 and 202% of control, respectively. Body temperature increased from the control value of 39.5 +/- 0.1 degrees C to a maximum of 41.5 +/- 0.1 degrees C at 200-300 min of infusion. In the second part of this study, we have examined the effects of ibuprofen on endotoxin-induced increases in plasma PG, TxB2, and serotonin levels and body temperature. The control values for TxB2, 6-keto-PGF1 alpha, PGF2 alpha, and temperature prior to ibuprofen and endotoxin infusion averaged 238 +/- 35, 335 +/- 44 and 248 +/- 28 pg/ml, 65 +/- 3 ng/ml and 40.1 +/- 0.2 degrees C, respectively. A loading dose (15 mg/kg) of ibuprofen was followed by infusion of endotoxin (12 micrograms/kg) and ibuprofen (43.3 mg/kg) over 500 min. Plasma levels of 6-keto-PGF1 alpha and serotonin increased only to 131 and 149% of control at 50 min of infusion, and levels of PGF2 alpha and TxB2 decreased to 50 and 80% of control at 100 and 150 min of infusion, respectively. Temperature remained unchanged. Ibuprofen effectively suppressed endotoxin-induced increases in the plasma levels of TxB2, 6-keto-PGF1 alpha, PGF2 alpha, and serotonin and body temperature. It was concluded from the present study that nonsteroidal anti-inflammatory drugs as an adjunct to antibiotic therapy might have a rational basis in treatment of endotoxin toxicity.  相似文献

Calves given 2 subcutaneous inoculations (4 ml, 4.5 weeks apart) of an inactivated bluetongue virus serotype 17 (BTV-17), aluminum hydroxide adjuvant, and cimetidine (600 mg) or levamisole (819 mg, 6 ml) combination were challenge exposed with virulent BTV-17 (2.5 x 10(5) embryo lethal dose) 9 weeks after the 1st inoculation and were monitored for 35 days. Plasma prostaglandins (PG) and thromboxane (Tx) B2 were measured by radioimmunoassay. Histamine was assayed spectrofluorometrically. During the inoculation period (9 weeks from the 1st inoculation to challenge exposure) PGE and histamine increased from base-line concentrations of 34 +/- 3 pg/ml and 1.2 +/- 0.1 ng/ml to 83 +/- 8 pg/ml and 2.0 +/- 0.1 ng/ml, respectively, whereas PGF2 alpha decreased from base-line values of 356 +/- 41 pg/ml to 226 +/- 16 pg/ml. Significant (P less than or equal to 0.05) changes from base-line TxB2 values (110 +/- 7 pg/ml) were not observed during the inoculation period. After challenge exposure, maximum increases were observed in TxB2 (157 +/- 10 pg/ml), PGF2 alpha (713 +/- 93 pg/ml), PGE (140 +/- 30 pg/ml), and histamine (3.6 +/- 0.2 ng/ml) concentrations at 4, 7, 7, and 14 days after challenge exposure, respectively. Concentrations of PGF2 alpha and TxB2 decreased from base-line values to 211 +/- 42 pg/ml and 75 +/- 11 pg/ml, respectively, 21 days after challenge exposure and then returned to base-line values. Significant changes were not observed in plasma concentrations of 6-keto-PGF1 alpha. Results indicate that PG, TxA2, and histamine may be involved in the hypersensitivity reaction to BTV in cattle.  相似文献

Arachidonic acid metabolites (AAM) were measured in milk and plasma during the course of acute endotoxin-induced mastitis in 12 lactating cows. Mastitis was induced by intramammary challenge exposure with 10 micrograms of Escherichia coli (026:B6) endotoxin. Endotoxin was injected into the teat cistern via the teat canal of a single randomly selected rear quarter of each cow. Concentrations of prostaglandin (PG) F2 alpha and thromboxane (Tx) B2 in fat-free unextracted milk and of 15-keto-13,14-dihydro-PGF2 alpha in plasma were measured by radioimmunoassay. Total production of AAM in milk was determined by measuring quarter milk production. The AAM were compared in 6 cows administered flunixin meglumine (1.1 mg/kg of body weight) and in 6 cows administered saline solution. Concentrations of TxB2 in milk were significantly (P less than 0.001) increased during the early course of acute mastitis in endotoxin-treated quarters of cows not administered flunixin meglumine. Peak concentrations of TxB2 in milk occurred at 8 hours after endotoxin inoculation. Flunixin meglumine treatment produced significant (P less than 0.05) reductions in milk TxB2 and plasma 15-keto-13,14-dihydro-PGF2 alpha concentrations. Concentrations of PGF2 alpha in milk and total PGF2 alpha and TxB2 production per quarter per milking were not significantly influenced by endotoxin challenge or by flunixin meglumine treatment.  相似文献

Four doses (5 to 100 micrograms, 1 dose/quarter) of Escherichia coli endotoxin were introduced into lactating mammary glands of 2 cows. There was no effect on milk prostaglandin (PG) E2 concentration, except that the concentration was increased from 200 pg/ml of milk to 1,060 pg/ml at post-treatment hour (PTH) 8 in cow 1 and from 75 to 420 pg/ml at PTH 4 in cow 2 after the highest dose 100 micrograms. Endotoxin caused a dose-dependent increase in milk PGF2 alpha concentrations in both cows. After the highest dose, PGF2 alpha was maximally increased from 200 to 3,500 pg/ml at PTH 4 in cow 1 and from 250 to 2,000 pg/ml in cow 2 at PTH 8. The instillation of 50 micrograms of endotoxin in all 8 quarters of 2 more lactating cows caused no significant (P greater than 0.05) changes in milk PGE2 and thromboxane B2 concentrations, whereas milk PGF2 alpha was significantly increased from the base-line value of 642 to 2,683, 1,189, and 2,281 pg/ml at PTH 4, 8, and 12, respectively. The 6-keto-PGF1 alpha was also significantly increased from the base-line value of 305 to 871, 631, and 600 pg/ml at the corresponding times, respectively. A marked increase in vascular permeability, as judged by high concentrations of serum albumin in the whey, was observed as early as PTH 4 and peaked at PTH 12 followed by a gradual decline, although it remained significantly increased over the control for 48 hours after treatment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Mastitis was induced in 4 lactating cows by inoculation of Klebsiella pneumoniae (10(7) organisms/ml) via the teat canal. Sterile isotonic saline solution (1 ml) was instilled into designated control quarters via the teat canal. Changes in milk leukotriene B4 and C4 (LTB4, LTC4) concentrations, milk somatic cell counts, and milk bovine serum albumin concentration were monitored over a 24-hour postinoculation period. Milk LTB4 concentration before inoculation in control quarters and quarters later to be infected was 376 +/- 45 and 326 +/- 56 pg/ml of milk, respectively. A significant (P less than 0.05) increase in milk LTB4 concentration in the infected quarters was first observed at postinoculation hour 6, and milk LTB4 concentration in infected quarters generally remained significantly high through postinoculation hour 14. Thereafter, milk LTB4 concentration in infected quarters was not significantly different from the concentration in control quarters. Measurable amounts of LTC4 were not detected in the milk of either control or infected quarters. Milk bovine serum albumin concentration in the infected quarters generally was high throughout the study, as were milk somatic cell counts. The results of this study suggested that LTB4 contributes to the pathogenesis of bovine mastitis.  相似文献

Subclinical mastitis caused by streptococcal infections affected 27 of 83 cows in a commercial dairy herd. Between three and six weeks after intramammary treatment of these cows with cloxacillin, 16 (59 per cent) of the treated cows developed acute clinical mastitis associated with Mycobacterium smegmatis. None of the untreated cows was affected. Infected quarters were moderately hypertrophied and fine clots were present in the milk for three to four weeks. No cows showed systemic signs of illness. Studies carried out over 12 months showed that infected cows shed M smegmatis for three to four months and affected quarters remained hypertrophied in all but one cow after 12 months. The mean milk cell count of affected quarters fell slowly from 4,850,000/ml in the acute stage to 810,000/ml five months later and 620,000/ml 12 months later, suggesting that the organism persisted in the udder. The estimated mean loss in lactation yield for cows with M smegmatis mastitis was 10.8 per cent. Losses were greatest when the hind quarters were involved (mean 28 per cent for cows with both hind quarters affected). Ten of the 16 affected cows were ultimately culled owing to serious reductions in yield.  相似文献

Ten Holstein cows were fed a selenium-deficient (SeD) diet containing 0.04 mg of Se/kg of dry matter for 3 months before and throughout their first lactation. A selenium-supplemented (SeS) group of 10 cows was fed an additional 2 mg of Se/head/d to increase dietary Se concentration of the dry matter to approximately 0.14 mg/kg of body weight. An intracisternal challenge exposure of 40 to 60 colony-forming units (CFU) of Staphylococcus aureus was administered into 1 or 2 quarters of the udder of each trial cow at about the twenty-second week of lactation. Blood Se concentration (micrograms/ml +/- SEM) at the time of challenge exposure was 0.035 +/- 0.002 in SeD and 0.139 +/- 0.006 in SeS cows. Infections were established in 14/16 of the challenge-exposed quarters in SeD and 16/19 of the challenge-exposed quarters in SeS cows. The infection in 1 quarter of each Se group cleared without treatment by the end of the 8-week trial period. Log10 peak bacterial concentrations in milk from infected SeD quarters (5.04 +/- 0.25 CFU/ml) were higher (P less than 0.05) than those of infected SeS quarters (4.40 +/- 0.12 CFU/ml). Log10 peak somatic cell count (SCC) in milk from infected SeD quarters (7.18 +/- 0.08 cells/ml) did not differ from that of SeS quarters (7.17 +/- 0.05 cells/ml). Peak bacterial concentrations were attained sooner (P less than 0.05) in SeD quarters (9.5 +/- 4.0 days) than in SeS quarters (20.7 +/- 3.1 days). Four experiments assessed changes in serum IGF-I under various physiologic conditions in postpartum cows. In Exp. 1, anestrous suckled cows (n = 25) were infused for 6 d with either saline or glucose at two different infusion rates. In Exp. 2, anestrous cows (n = 29) received either a saline (weaned and suckled controls) or 3 g/d phlorizin (weaned phlorizin) infusion for 3 d. Calves from the weaned groups were removed from 15 h before and throughout infusions. In Exp. 3, cycling suckled cows (n = 20) received prostaglandin F2 alpha (PGF2 alpha) when the 5-d saline or phlorizin infusion began. In Exp. 4, suckled cows (n = 20) had ad libitum access to feed or received 50% of control feed consumption from 30 to 40 d postpartum. Increasing glucose availability (Exp. 1) increased (P less than .05) serum IGF-I by 30 to 35%. IGF-I remained stable after weaning (Exp. 2) in phlorizin-infused cows (128.8 +/- 12.7 ng/ml), but increased (P less than .05) by 3 d after calf removal in weaned control cows (152.2 +/- 7.5 ng/ml). IGF-I also remained stable in phlorizin-infused cows following PGF2 alpha injection (Exp. 3), but increased in control cows by 2 d after PGF2 alpha (156.8 +/- 18.3 on d 2 vs. 133.7 +/- 9.8 ng/ml pre-injection; P less than .05) and remained elevated (P less than .05) during the periovulatory period. Reproductive management programmes based on strategic use of prostaglandin F2 alpha (PGF2 alpha) to induce and synchronize oestrus in post-partum dairy cows are widespread. Repeated shortening of the oestrous cycle during early lactation in high-yielding dairy cows, however, could impair corpus luteum function and thus decrease fertility. The objective of this study was to analyse the effect of repeated treatments with the prostaglandin F2 alpha analogue D (+) cloprostenol sodium on progesterone concentrations indicative of a functional corpus luteum in post-partum dairy cows. Furthermore, the influence of milk production, parity and endometritis on progesterone concentrations under these circumstances were studied. Eighty-four cows of a commercial dairy operation were treated three to four times with D (+) cloprostenol sodium (Preloban; Hoechst Roussel Vet, Wiesbaden, Germany) at 14-day intervals, starting 22-28 days post-partum. Blood samples were collected prior to treatment 1 (sample 1) and 14 days after treatments 1, 2 and 3 (samples 2-4) and serum progesterone (P4) levels were determined. The percentage of cows with P4 levels < 1 ng/ml decreased from 51% in sample 1 to 23% in samples 3 and 4. More primiparous cows had low P4 levels 14 days after the second treatment than older cows (P < 0.05). Cows with low progesterone levels in sample 3 or 4 had lower protein contents in milk on the second milk test day post-partum and in their cumulative milk yield of the first 100 days of lactation. Clinical endometritis at post-partum examination did not influence progesterone levels after treatment with PGF2 alpha. Repeated application of PGF2 alpha (more than twice) in the post-partum period does not influence serum progesterone levels 14 days after treatment. Failure to develop luteal tissue after treatment contributed to the conception failures after first service.  相似文献

Twelve dairy herds that had participated in the Pennsylvania Dairy Herd Improvement Association (DHIA) program for at least 12 months, that had a 12-month mean DHIA somatic cell count greater than 700,000 cells/ml, and that had greater than 25% of lactating cows infected with Streptococcus agalactiae participated in a herd blitz treatment program. Initially, quarter milk samples for bacteriologic culturing were collected from all lactating cows. Subsequently, all cows identified as infected with Str agalactiae were treated, using a commercial penicillin-novobiocin intramammary infusion product. In addition, a herd mastitis management program of postmilking teat dipping and treatment of all cows at the start of the nonlactating period was instituted. Thirty days after the initial herd visit, samples from all lactating cows were again cultured, and cows infected at that time were treated. Twelve months after the initial herd visit, samples from all lactating cows were again cultured. Mean prevalence of infection with Str agalactiae decreased (P less than 0.05) from 23.0% of quarters and 41.6% of cows initially to 3.4% of quarters and 9.3% of cows at 30 days and 1.6% of quarters and 4.2% of cows at 1 year. It was the objective of this study to analyse shedding patterns and somatic cell counts in cows and quarters infected with Prototheca spp. and to evaluate two approaches to identify infected animals by somatic cell count (SCC) or by bacteriological analysis of pooled milk samples. Five lactating dairy cows, chronically infected with Prototheca spp. in at least one quarter were studied over 11 weeks to 13 months. Quarter milk samples and a pooled milk sample from 4 quarters were collected aseptically from all quarters of the cows on a weekly basis. Culture results of quarter milk and pooled samples were compared using cross tabulation. SCC of quarter milk samples and of pooled samples were related to the probability of detection in the infected quarters and cows, respectively. Shedding of Prototheca spp. was continuous in 2 of 8 quarters. In the other quarters negative samples were obtained sporadically or over a longer period (1 quarter). Overall, Prototheca spp. were isolated from 83.6% of quarter milk samples and 77.0% of pooled milk samples of infected quarters and cows. Somatic cell counts were higher in those samples from infected quarters that contained the algae than in negative samples (p < 0.0001). The same applied for composite samples from infected cows. Positive samples had higher SCC than negative samples. However, Prototheca spp. were also isolated from quarter milk and pooled samples with physiological SCC (i.e. < 10(5)/ml). Infected quarters that were dried off did not develop acute mastitis. However, drying off had no effect on the infection, i.e. samples collected at calving or 8 weeks after dry off still contained Prototheca spp. Results indicate that pre-selection of cows to be sampled for Prototheca spp. by SCC and the use of composite samples are probably inadequate in attempts to eradicate the disease. However, due to intermittent shedding of the algae in some cows, single herd sampling using quarter milk samples probably also fails to detect all infected cases. Therefore, continuous monitoring of problem cows with clinical mastitis or increased SCC in herds during eradication programs is recommended.  相似文献

Pregnant cows at 4- to 5-months' of gestation were exposed to Sarcocystis cruzi or Campylobacter fetus. Plasma prostaglandin F (PGF) and 13,14-dihydro-15-ketoprostaglandin metabolite (PGM) concentrations were determined at intervals from before exposure until abortion or parturition. The plasma PGF concentration of pregnant infected cattle remained at 0.02 +/- 0.04 ng/ml until 24 to 48 hours before abortion or parturition when it increased 5-fold to 0.11 +/- 0.12 ng/ml. The plasma PGM concentration of these cattle remained at 0.10 +/- 0.07 ng/ml until 24 to 48 hours before abortion or parturition when it increased over 10-fold to 1.36 +/- 0.60 ng/ml. This change in PGF and PGM was similar to that of cattle exposed to each of the infective agents and to that of normal cows at parturition. Thus, changes in PGF and PGM concentrations in bovine plasma cannot be used as a diagnostic tool to determine fetal distress or fetal death for these infections.  相似文献

Single-dose pharmacokinetics of norfloxacin after intravenous administration of norfloxacin nicotinate at 10 mg norfloxacin/kg body weight was investigated in cows with healthy udders and in cows with chronic subclinical and postacute clinical mastitis. An HPLC method was used to determine the norfloxacin concentrations in serum and milk. Significant differences were observed in norfloxacin pharmacokinetics when administered to cows with infected udder quarters. The clearance (Cl) values were 10.4+/-2.5, 13.2+/-1.9 and 14.2+/-2.1 mL/min/kg (mean +/-SD) in the control (healthy udder) cows and in cows with subclinical and postacute clinical mastitis, respectively. There appeared to be a trend of increasing clearance according to severity of the disease. The volume of distribution at steady state (Vss) in the respective groups was 3.1+/-0.7, 2.2+/-0.6 and 1.3+/-0.2 L/kg. The volume of distribution was significantly decreased in the cows with postacute clinical mastitis. The half-lives (t1/2) and mean residence times (MRT) of norfloxacin were 353, 206 and 115 min (harmonic means) and 306+/-76, 168+/-39 and 95+/-9 min in control cows or in cows with subclinical and postacute clinical mastitis, respectively. The half-lives in the clinical mastitis group were significantly shorter than in the control group and the mean residence times were significantly shorter in the two mastitis groups when compared to the control group. Norfloxacin concentrations in milk were extremely high when compared to the respective serum concentrations. The area under the concentration vs. time curve (AUC) of norfloxacin in milk was 23899+/-6206 mg/L x min in the control cow group. The AUC in milk was significantly lower in the infected udder quarters of the mastitis groups (5075+/-1887 mg/L x min and 7484+/-4645 mg/L x min in the subclinical and the clinical group). The AUC values were significantly lower in milk from the infected udder quarters of the cows with chronic subclinical and postacute clinical mastitis when compared to the values in milk from the healthy quarters of the same udder. Norfloxacin was marginally bound to serum protein. The binding was concentration dependent and was 19, 13 and 6% at 0.2, 1.0 and 8.4 mg/L, respectively. Binding to milk protein was 46-51% and concentration independent. An in vitro dialysis model was used to simulate drug transport between serum and milk as a function of protein binding. The results showed that the rate of norfloxacin disposition from milk to serum was slower than from serum to milk, which was in agreement with the findings obtained in the pharmacokinetic study. Norfloxacin was poorly soluble in organic solvents and our results suggest that changes in the degree of ionization of the drug in different body fluids considerably affect its disposition.  相似文献

To evaluate clinical effects of autogenous toxoid-bacterin treatment for Staphylococcus aureus subclinical mastitis in lactating cows, 22 cows which had at least one S. aureus infected quarter were selected from among cows at a S. aureus prevalent dairy farm. Eleven cows were injected with their own autogenous toxoid-bacterin and the others were maintained as non-injected control. In the toxoid-bacterin injected group, 27% of infected quarters were cured during the 12-week trial, compared to 5% in the control group. New intramammary infections with S. aureus were only detected in 3 quarters of the control group. Mean IgG antibody titer against S. aureus somatic antigens and alpha-toxin in serum and milk were significantly increased in the toxoid-bacterin injected group (p<0.05) and remained higher than those of the control group which showed no significant changes (p<0.05). In contrast to the control group, from 3 weeks after the second injection of the toxoid-bacterin injected group, mean S. aureus cfu/ml in milk samples from injected quarters with S. aureus was significantly decreased until the end of the study (p<0.05). In the toxoid-bacterin injected group, significant decreases of mean SCC were detected from milk samples from infected quarters with S. aureus from week 7 to week 10 (p<0.05). These data show that autogenous toxoid-bacterin treatment against S. aureus subclinical mastitis in lactating cows may increase the cure rate of the infections, reduce the severity of the infections and also prevent occurrence of the new infections.  相似文献

This study was conducted to examine the effects of metestrus administration of SyncroMate-B (SMB) on PGF2alpha secretion and corpus luteum (CL) development. In a study replicated over 2 yr, cows were observed for spontaneous estrus in yr 1, and cows received an injection of 25 mg of PGF2alpha and were observed for subsequent estrus in yr 2. At standing estrus (estrus = d 1), cows were randomly allotted to receive either the standard SMB regimen (n = 40) on d 3 of the estrous cycle or no treatment (n = 8). Fifty percent (n = 20) of SMB-treated cows were administered PGF2alpha on d 10 of the estrous cycle 48 h prior to implant removal. Twice-daily blood samples were collected in the morning (AM) and evening (PM) from d 2 AM through d 14 AM of the treated estrous cycle and subsequently analyzed for progesterone (P4) and PGF2alpha metabolite (PGFM). Prior to statistical analysis, SMB- and SMB/PGF2alpha-treated cows were sorted according to P4 concentration at d 10 of the treated estrous cycle to either a CL functional group (P4 > or = 1 ng/mL; n = 20) or a CL nonfunctional group (P4 < 1 ng/mL; n = 17). Following d 10 AM administration of PGF2alpha, functional and nonfunctional groups were further subdivided based on treatment. The groups were as follows: untreated control cows (n = 8); SMB-treated cows retaining a functional CL (SMB-F; n = 8); SMB-treated cows with a nonfunctional CL (SMB-N; n = 11); SMB/PGF2alpha-treated cows retaining a functional CL (SMB/PG-F; n = 12); and SMB/PGF2alpha-treated cows with a nonfunctional CL (SMB/PG-N; n = 6). Of all SMB-treated cows, 54% retained a functional CL through d 10 AM of the treated estrous cycle. Mean serum P4 concentrations increased for cows in all groups until d 7, after which P4 concentrations increased for cows in SMB/PG-F, SMB-F, and control groups and decreased for cows in SMB/PG-N and SMB-N groups. Following PGF2alpha administration on d 10, mean serum P4 concentrations remained < 1 ng/mL for cows in SMB/PG-N and SMB-N groups, decreased to < 1 ng/mL for cows in the SMB/ PG-F group, and remained > 1 ng/mL for cows in SMB-F and control groups. Mean serum PGFM concentrations tended (P = .06) to increase in cows with nonfunctional CL compared with control cows on d 8 AM and were greater (P < .05) in cows with functional CL on d 8 PM through d 9 PM. These results indicate that retention of a functional rather than a nonfunctional CL following metestrus administration of SMB is dependent on a premature release of uterine PGF2alpha.  相似文献

Prevalence and effects of intramammary infection in 322 beef cows was determined during three calving intervals. Intramammary infection was confirmed in 37% of cows and 18.1% of quarters. Coagulase-positive staphylococci accounted for 17.9% of infections with Staphylococcus aureus isolated from 7.1% of cows. Coagulase-negative staphylococci and micrococci accounted for the remainder of infectious organisms. Butterfat and total protein levels were reduced 27.3 (P less than .05) and 25.5% (P less than .01), respectively, in milk from quarters infected with S. aureus. Somatic cell counts were elevated (P less than .001) with 3,827 X 10(3) cells/ml for S. aureus-infected quarters as compared with 555 X 10(3) cells/ml for uninfected quarters. Somatic cell counts were negatively correlated with 210-d calf weaning weights. Staphylococcus aureus-infected cows weaned calves weighing 19.1 kg less (P less than .01) than uninfected cows. At a present market value of $1.65/kg, economic losses were placed at $31.43/calf from cows infected with S. aureus in one or more quarters.  相似文献

Prostaglandin F(2alpha) (PGF(2alpha)) is the primary luteolysin in the cow, and luteal endothelin-1 (ET-1) interacts with PGF(2alpha) during the process of luteolysis. In contrast, a developing corpus luteum (CL) is refractory to exogenous administration of PGF(2alpha). Thus, the present study was aimed to investigate the functional relationship between ET-1 and PGF(2alpha) in the mid-CL (PGF(2alpha)-sensitive) and early-CL (PGF(2alpha)-refractory). In the mid-CL model, cows (n = 6/treatment) were assigned to receive one of five types of treatments on day 10 of the estrous cycle: (1) an injection of saline; control, (2) a 500 microg of PGF(2alpha) analogue (sufficient dose to induce luteolytis); full-PG, (3) an intraluteal injection of 0.25 mg ET-1; ET-1, (4) a 125 micro g of PGF(2alpha) (insufficient dose to induce luteolytis); 1/4PG or (5) an intraluteal injection of 0.25 mg ET-1 after administration of a insufficient dose of PGF(2alpha) analogue; 1/4PG/ET. In the early-CL model, cows were assigned to receive one of two types of treatments on day 5 of the estrous cycle: (1) a sufficient dose of PGF(2alpha) analogue; PG (n = 5) or (2) an intraluteal injection ET-1 after a sufficient dose of PGF(2alpha); PG/ET (n = 7). In the mid-CL model, 1/4PG/ET resulted in a rapid reduction of progesterone (P) concentrations similar to that in full-PG from the next day. However, the levels of P in 1/4PG/ET (1.5-2.5 ng/ml) kept significantly higher than that in full-PG (< 0.5 ng/ml). ET-1 or 1/4PG did not decrease plasma P concentrations (4-6 ng/ml). The plasma ET-1 levels increased with the full-PG administration. In the early-CL model, both treatments had no effect on plasma P increase and ET-1 levels. The overall results indicate that the intraluteal ET-1 injection after administration of insufficient dose of PGF(2alpha) induces the depression of P secretion in vivo during the mid luteal phase in the cow, supporting the concept that ET-1 is one of a local mediator of functional luteolysis in the cow. The result further indicates that the early-CL is not only PG-refractory but also ET-1-refractory.  相似文献

Prevalence of intramammary infection was studied in primiparous beef cows preceding and following parturition. Aseptic mammary quarter samples were collected from Brahman-sired cows (Group 1; n = 19) on d −143, −30, and 110 ± 4 from parturition, and from Angus-sired cows (Group 2; n = 54) on d -36 and 37 ± 2 from parturition. In Group 1, 67.6, 55.6, and 9.3% of quarters sampled were infected with mastitis pathogens on d −143, −30, and 110 ± 4 from parturition, respectively. In Group 2, 35.3 and 10.4% of the quarters were infected on d −36 and 37 ± 2 from parturition, respectively. In Group 2, over 22% of the quarters that had previously been infected with Staphylococcus aureus, and over 26% of the quarters that had previously been infected with Staphylococcus species were still infected with the same organism on d 37 ± 2 postpartum. In Group 2, representative cows were selected for blood chemistry analyses (n = 30) and machine milking (n = 46). Serum potassium was higher (4.64 vs 4.36 ± 0.09 mmol/L; P<0.03) and serum chloride was lower (106.19 vs 107.24 ± 0.31 mmol/L; P<0.02) in infected than in non-infected cows, respectively. Based on the d 37 ± 2 postpartum milk yield, cows were retrospectively divided into a low-milk (below the median milk production) or high-milk (above the median milk production) group. Serum cholesterol was higher in high-milk than in low-milk cows (145 vs 124 ± 5.3 mg/dL; P<0.01). In summary, mastitis-causing pathogens were present in quarters of primiparous beef cows prior to parturition; however, prevalence was considerably lower during lactation.  相似文献

An enzyme-linked immunosorbent assay (ELISA) procedure was used to quantitate milk and serum antibodies (IgG) to Staphylococcus aureus alpha and beta toxins, and S. aureus 2-8 and Smith diffuse strain capsular antigens. Milk samples were collected on two occasions. A comparison was made between levels of milk antibodies specific for the two toxins and capsular antigens for 41 cows that were infected with S. aureus on both sampling dates, and 18 cows not S. aureus-infected on either date. Staphylococcus aureus-infected cows were grouped according to somatic cell counts. All groups of infected cows, regardless of somatic cell counts, had significantly higher milk antibody levels to alpha and beta toxins than did the non-infected cows (P less than .002). Serum samples taken for 13 infected and 4 non-infected cows also indicated that significant elevations in anti-alpha toxin and anti-beta toxin IgG were present in S. aureus-infected cows, compared to non-infected cows. A similar immune response was not seen to capsular antigens, however. No significant differences were present between the two groups of cows for either milk or serum antibodies to Smith diffuse strain capsular antigens. Milk antibodies to 2-8 capsule were significantly elevated only in infected cows with somatic cell counts greater than 10(6)/ml, compared to non-infected cows; no differences were present for serum antibodies to 2-8 capsule between infected and non-infected cows. These results indicate that significant increases in milk (and possibly serum) antibodies to alpha and beta toxins are present in cows with chronic staphylococcal mastitis, apparently resulting from a systemic immune response to these toxins. There does not appear to be a similar immune response to capsular antigens.  相似文献
