Oocyte maturation and gonadosomatic index (GSI) of eyestalk ablated Penaeus monodon females collected from the wild and from two first‐generation domesticated lines were assessed. Frequency and diameter of the different oocytes, and the intensity of oocyte atresia, were compared among groups through histological assessments of the sections of the middle ovarian lobe. Digitized images from ovary sections were used to record the frequency and diameter of different oocyte types. Spawning performance of the three groups were expressed in terms of the percentage of females that spawned at least once (productive females), time from eyestalk ablation to first spawning (latency period) and the number of spawnings per female stocked. Final ovarian maturation was attained in all groups, as indicated by the presence of mature oocytes with cortical rods (cortical oocytes), dark‐green ovarian colour and high GSI values (5.83–6.86%). However, domesticated groups presented significant larger immature oocyte types (previtellogenic and yolky oocytes) and smaller cortical oocytes compared with wild females, indicating a reduced vitellogenic activity during final oocyte maturation. Additionally, the frequency of atresia was comparatively higher for both domesticated groups, which could be related to their inferior spawning performance. The implications of these results on the reproductive potential and development of domesticated P. monodon are discussed.  相似文献

Prostaglandin reductase 1 (PTGR1; also called NADP+-dependent leukotriene B4 12-hydroxydehydrogenase, LTB4DH) is the key enzyme responsible for biological inactivation of prostaglandins and related eicosanoids. In this study, the full-length cDNA of PTGR1 in the giant tiger shrimp (Penaeus monodon) was characterized. PmPTGR1 was 2405 bp in length with an ORF of 1035 bp encoding a polypeptide of 344 amino acids. Interestingly, its 3′ UTR contained the nucleotide sequence (825 bp) that significantly matched positions 3–277 (with 4 amino acid variants) of the deduced P. monodon peritrophin2 protein. PmPTGR1 was more preferentially expressed in ovaries than testes of P. monodon broodstock. In intact broodstock, PmPTGR1 was up-regulated in early cortical rod (stage III) ovaries (P < 0.05) and comparably expressed afterwards (P > 0.05). In eyestalk-ablated broodstock, PmPTGR1 was temporally lower in early cortical rod compared to previtellogenic (I) and vitellogenic (II) ovaries (P < 0.05) and returned to the previous level in mature (IV) ovaries (P < 0.05). More importantly, the relative expression level of PmPTGR1 in each ovarian stage in eyestalk-ablated females was lower than that in intact P. monodon broodstock (P < 0.05). This strongly suggested that eyestalk ablation potentially affects the expression of PmPTGR1 allowing the stimulating effects of prostaglandins and related eicosanoids on vitellogenesis and ovarian maturation of P. monodon. The level of ovarian PmPTGR1 protein seemed to increase during ovarian development in intact broodstock but slightly reduced in mature ovaries in eyestalk-ablated broodstock. Results suggested the possible contribution of PmPTGR1 in ovarian development of P. monodon.  相似文献

Immunocytochemical localization using antibodies against five isoforms of gonadotropin-releasing hormone (GnRH), namely, luteinizing hormone-releasing hormone (LHRH), salmon (s)GnRH, octopus (oct)GnRH, lamprey (l)GnRH-I, and lGnRH-III, showed that only lGnRH-I immunoreactivity (ir-lGnRH-I) was localized in follicular cells of proliferative, vitellogenic, and mature ovaries. The effects of exogenous GnRHs on the ovarian maturation cycle of Penaeus monodon were compared by treating female broodstocks with LHRH, sGnRH, and lGnRH-I. The cycle of ovarian maturation in both eyestalk-ablated and eyestalk-intact shrimp administered with the three isoforms of GnRH was significantly shorter than that of the control animals. Moreover, administrations of all GnRH isoforms showed similar numbers of spawned eggs and the percentage of successful fertilization as in the control animals. These findings suggest that GnRHs may be highly conserved peptides that play an important role in inducing the ovarian maturation in the shrimp.  相似文献

The present study investigated the external and histological changes in the ovary and measured the gonadosomatic index (GSI) and hepatosomatic index (HSI) during the re‐maturation of the Chinese mitten crab Eriocheir sinensis. The results show that the ovarian re‐maturation cycle of this crab species can be divided into four stages, i.e. Stage I: the ovary had an X shape and appeared milky white or with a mottled buff colour, and the dominant gametocyte at the stage were oogonia (OG, 28.3%) and previtellogenic oocytes (PR, 31.6%); Stage II: the ovary appeared light yellow or tan in colour, dominated by endogenous vitellogeic oocytes (EN, 69.3%). The yolk globules of unreleased mature oocytes (MO) from the previous spawn were absorbed, leaving many vacuoles in the retrogressing MO; Stage III: the ovary appeared crimson red or deep purple, and although the main type of gametocyte was exogenous vitellogenic oocytes (EX), near‐mature oocytes (NO) could also be found in the late phase of the ovarian development stage; Stage IV: the ovary was ripe and filled with MO that contained large yolk globules. During the period of the second ovarian maturation cycle, the GSI increased significantly from the time just after the first spawning (P<0.01). However, although HSI appeared to decrease within days after spawning (DAS), no significant correlation was found between GSI and HSI or between HSI and DAS (P>0.05). Furthermore, the final GSI and the volume of MO of E. sinensis during the second maturation cycle were drastically lower than those of the first maturation cycle.  相似文献

A possible factor behind the successful use of polychaetes for prawn broodstock diet was investigated. Some steroid hormones in polychaetes have been suspected as being a potential component for prawn reproductive system development. Progesterone (P4) and 17-alpha hydroxyprogesterone (17α-OHP4) were extracted from polychaetes (Perinereis sp.) and female Penaeus monodon broodstock at various maturation stages. Concentration of both hormones was determined using RIA or HPLC. Immature natural polychaetes (2 months old) displayed the highest level of P4 and 17α-OHP4 compared to more mature ones (4, 6 and 8 months old polychaetes) (47.48 ± 6.70 ng P4/mg Protein and 143.74 ± 28.70 ng 17α-OHP4/mg Protein) whereas opposite results were received with cultured polychaetes in which a reverse relationship between P4 and its derivative with polychaete maturation was obtained. Diverse concentrations of both hormones may be linked with other factors such as types of polychaetes, habitats, seasons, and feed intakes. Twenty-four hours in vitro incubation of prawn previtellogenic oocytes with P4 or 17α-OHP4 extracted from polychaetes or with synthetic hormones, significantly increased percentages of vitellogenic oocytes and oocytes with cortical rod compared to the control with no hormones. P4 was more effective in enhancing the final maturation of oocytes while 17α-OHP4 had more effects on vitellogenic oocytes. Synthetic steroid hormones at equal hormone concentrations produced similar results to steroid hormones extracted from natural polychaetes.  相似文献

Retinoblastoma (Rb) is a multifunctional regulator involved in several key cellular processes, such as cell cycle control, cell differentiation, tumorigenesis and senescence. In this study, an Rb‐like gene, PmRBL, was cloned from black tiger shrimp (Penaeus monodon). The full‐length cDNA sequence of PmRBL is 4,069 bp with an open reading frame of 3,243 bp, which encodes 1,080 amino acids. Quantitative real‐time PCR (qRT‐PCR) analysis indicated that PmRBL was highly expressed in the gills, hepatopancreas and ovaries of P. monodon. The highest PmRBL expression levels were observed in stage III of the ovarian development in P. monodon. RNA interference experiments were conducted to examine the expression profiles of PmRBL, PmCDK2 and PmCyclin E. The knockdown of PmRBL in the ovary and hepatopancreas by dsRNA‐RBL was successful. After dsRNA‐RBL was injected into the shrimp, the relative expression levels of PmCDK2 and PmCyclin E were upregulated at 12–72 hr in the ovaries and hepatopancreas. The localization and level of PmRBL expression in the ovary and hepatopancreas were investigated through in situ hybridization, which revealed consistent results with those of qRT‐PCR. Therefore, PmRBL, PmCDK2 and PmCyclin E may be involved in vitellogenin synthesis and ovarian maturation in P. monodon.  相似文献

A histological study was made of the reproductive cycle of captive sharpsnout seabream Diplodus puntazzo during the 3rd year of life. All specimens contained both ovarian and testicular tissue. Ovarian tissue prevailed in 64 out of 120 fish sampled throughout one year. The ovary underwent a long resting phase from October to August, during which time it contained oogonia, primary growth phase (PGP) and occasional yolk vesicle stage oocytes. Intense ovarian development took place in September leading to the formation of advanced vitellogenic oocytes. Final maturation and spawning were not achieved in any of the fish. The testicular area in the predominantly female gonads contained only spermatogonia and was found to be histologically normal in 76% of individuals, but severely regressed in the remaining fish. The testicular portion was found to be dominant in 56 out of 120 fish sampled throughout one year. These underwent a resting phase from November to July, a maturing/pre-spawning phase in August, and a spawning phase with spontaneous sperm release in September to October, as in the natural environment. The ovarian portion in the functional male gonads contained only oogonia and oocytes which did not develop beyond the perinucleolus stage at any point during the reproductive cycle. Apart from the pre-spawning/spawning phase, during which the gonadosomatic index (GSI) was comparable in the two groups, the GSI values were approximately ten-fold higher in those fish in which the ovary predominated than in those in which the testis predominated.  相似文献

Tiger shrimp Penaeus monodon were intensively grown from PL15 for 56 d in tank systems at stocking densities of 1000 and 2000 shrimp m− 3, with and without the addition of artificial substrates (AquaMat® (buoyant and non-buoyant) and polyethylene mesh) at each density. Shrimp growth was significantly greater at the lower density and when substrates were added. Mean shrimp weight at harvest ranged from 0.64 ± 0.06 g (2000 shrimp m− 3, no added substrate) to 1.17 ± 0.01 g (1000 shrimp m− 3, added substrate). Survival was high and averaged 79.5 ± 2.7% across all treatments. The addition of substrates significantly increased survival at both stocking densities; however, survival was not significantly affected by stocking density. A maximum harvest density of 1645 shrimp m− 3 and biomass of 1.27 kg m− 3 were produced at a stocking density of 2000 m− 3 with added substrates. Both harvest density and biomass significantly increased with stocking density and addition of substrates. The feed conversion ratio (FCR) of formulated feed was significantly lower when substrates were added. The results show that growth of P. monodon juveniles was inversely related to stocking density during intensive production. However, production output was significantly increased by addition of artificial substrates, which enhanced both growth and survival.  相似文献

The optimization of reproductive parameters in shrimp farming continues to be a challenge for most producing countries. Although the crustacean neuropeptides have been studied extensively in the last two decades, the functions of most of these neuropeptides remained putative. Among them, molt‐inhibiting hormone isoform II (MIH II) has shown an important role in vitellogenesis. In this study, the cDNA encoding mature MIH II peptide was isolated by RT‐PCR from the L. vannamei eyestalk. The cDNA was cloned into pET28a bacterial expression vector. Recombinant MIH II was obtained in the form of insoluble inclusion bodies and purified to ~88% purity. Two doses of rMIH II and a negative control group were assayed in vivo. The stages of ovarian maturation and spawning were recorded during 72 hr post‐injection. The results showed that ovarian maturation occurred approximately in 9% and 33% of females injected with rMIH II at the doses of 300 and 600 ng/gbw respectively. Neither maturation nor spawning was detected in the negative control group. Females injected with 600 ng/gbw, which showed vitellogenic stages III and IV, spawned. These preliminary results argue that the hormone rMIH II could be a promising candidate to induce spawning in L. vannamei shrimp.  相似文献

High resolution ultrasound scanning was tested as a non-invasive technique for monitoring oocyte maturation and ovulalion in the Atlantic halibut Hippoglossus hippoglossus (L.). Female broodstock halibut were examined using a 7.5-Mhz linear ultrasound transducer, prior to and during spawning. Representative images of halibut ovaries are presented and discussed. Individual oocytes of spawning females were discernible during oocyte final maturation, due to the large increase in volume caused by water uptake. The yolky (vitellogenic) cocytes of pre-spawning fish were more reflective to ultrasound than hydrating oocytes. The resulting differences in depth of ultrasound penetration permitted easy distinction of pre-spawning from spawning females. In addition, short-term changes in the depth of ultrasound penetration were observed during repeated scanning of a spawning female, owing to progressive hydration of the oocyte batch destined for ovulation. Although of similar diameter, hydrating oocytes in the ovarian tissue could be discerned from eggs in the ovarian lumen because of the different acoustic properties of the surrounding media. The findings of the present study are considered promising for the future routine use of ultrasonography in halibut broodstock management.  相似文献

The greasyback shrimp Metapenaeus ensis is widely distributed along the coast of India and the West Pacific where it is an important fisheries species. We have examined seasonal changes in ovarian development, spermatogenesis, and mating of Me. ensis in histological studies and by external observations on specimens collected in Ise Bay, its northernmost habitat. Ovaries were found to be previtellogenic from February to May, with the first signs of development being the accumulation of yolk in oocytes in late May. Ovarian shadow ratios were high during the period late July to mid-September. The formation of cortical rods in the peripheries of oocytes and germinal vesicle breakdown were observed in ovaries from late June to September. Male shrimps had sperm in the testes during the period early June to early October, and female shrimps had spermatophores in spermatheca after early July. In late July, some post-spawn female shrimps had exogenous vitellogenic oocytes in their ovaries, indicating that ovarian development had been repeated in preparation for the next spawning. Ovarian shadow ratios, which were positively correlated with gonadosomatic indices and ovarian development, seem to be a useful marker to determine ovarian development. Our results indicate that mating in Me. ensis started in early July and that the spawning season ranged from July to September with more than two cycles of spawning in Ise Bay.

Seasonal changes In oocyte growth and plasma estradiol, testosterone, thyroid hormones and vitellogenin levels were monitored in three captive adult female orangemouth corvina Cynoscion xanthulus subjected to a condensed (8 mo) seasonal cycle of photoperiod and water temperature. Plasma concentrations of estradiol and testosterone began to rise when temperature increased to 28 C and photoperiod to 15 h light (midsummer conditions). This was accompanied by elevated circulating levels of vitellogenin and the appearance of vitellogenic oocytes (diameter > 100 μm). Estradiol concentrations and mean oocyte size increased concurrently during late summer conditions and were maximum during fall conditions, approximately 8 wk after the beginning of ovarian recrudescence. In contrast, plasma levels of thyroid hormones did not show any distinct seasonal changes. Gonadally recrudesced females contained several size classes of vitellogenic oocytes. Injection of these fish with a luteinizing hormone-releasing hormone analog (LHRHa) caused maturation and spawning of the largest oocytes (mean diameter of follicles × 2500 μm). Another group of vitellogenic oocytes had been recruited into this size class by 2 wk after the end of spawning, which suggests that this species is capable of repeated spawning during the reproductive season. Injection of LHRHa resulted in increased plasma levels of gonadotropin and repeated spawning over several days. LHRHa-induced final oocyte maturation, ovulation and spawning were preceded by increases in plasma levels of two teleostean maturation-inducing steroids, 17α,20β, 21-trihydroxy-4-pregnen-3-one (20β-S) and 17α,20β-dihydroxy-4-pregnen-3-one (17α,20β-P4). The results provide preliminary evidence that oocyte growth is stimulated in orangemouth corvina subjected to an abbreviated seasonal cycle in captivity under midsummer photoperiod and temperature conditions and is associated with seasonal increases in plasma estradiol concentrations and vitellogenin production. However, the relative importance of 20β-S versus 17α, 20β-P4 in the control of final oocyte maturation in this species could not be determined from the results of the present study.  相似文献

We evaluated the annual process of oocyte and ovarian development of the planktivorous Amazonian catfish, the mapará (Hypophthalmus marginatus), aiming to support captive reproduction for domestication. A total of 149 females were captured from the Tocantins River at the Tucuruí Dam (3°49'55"S, 49°39'9"W) in 13 sampling events. For each individual, we evaluated ovary histology and plasma steroid concentrations. Two annual reproductive cycles, with similar characteristics, including a long period of rest, a short vitellogenic stage and a well‐defined spawning season from November to March, were described. A 17β‐estradiol (E2) rise and a 17α,20β‐dihydroxy‐4‐pregnen‐3‐one (DHP) peak were associated with vitellogenesis and spawning season, respectively, in the first cycle but not in the second reproductive cycle. In conclusion, the studied species presents a reproductive cycle similar to that of other migratory total spawners; however, unusually for this group of fish, there were three (but not two) batches of vitellogenic oocytes in “in maturation” and “mature” stage ovaries, pointing to the possibility of more than one spawn during the spawning season.  相似文献

Chilled storage of spermatophores from white shrimp (Litopenaeus vannamei) is needed to generate a consistent and reliable supply of spermatozoa for domestication purposes. The objective of this study was to develop a protocol for the chilled storage of white shrimp spermatophores and to evaluate bacterial propagation during such storage. In the first experiment, spermatophores were immersed in four extenders, mineral oil, Ringer's solution, phosphate buffer and 0.85% NaCl, and stored at low temperature (2-4 °C) for 35 days. Characteristics of preserved spermatophores changed the least and viable sperm was highest when spermatophores were stored in mineral oil. Spermatophores preserved with mineral oil appeared morphologically normal. Bacillus circulans, Staphylococcus hominis and S. lugdunensis, S. sciuri, S. xylosus and Micrococcus spp. were identified as the predominant bacteria during chilled storage, and total bacterial counts gradually increased during the experiment. A second experiment investigated the effect of antibiotic on chilled storage. Spermatophores were preserved in only mineral oil or mineral oil with 0.1% penicillin-streptomycin. These were evaluated for changes in external morphology of spermatophores, sperm viability and total bacteria count every week during a 35-day experimental period. Percentages of viable sperm (69.5 ± 3.9%) were significantly higher (P < 0.05) among spermatophores preserved in mineral oil with 0.1% antibiotic compared with those preserved only in mineral oil (57.7 ± 3.4%) over 35 days. The number of total bacteria in the treatment with mineral oil ranged between 28.3 ± 4.8 and 2416.7 ± 299.4 CFU/g, but in mineral oil containing antibiotic bacteria were undetectable. This study suggests that chilled storage of spermatophores is a feasible approach for the management and spawning of white shrimp broodstock.  相似文献

Based on the light microscopic observations of cells' sizes, chromatin patterns, amount of lipid droplets and yolk granules, the female germ cells could be classified into four different phases, which include 1) oogonia (Oog), 2) primary oocytes (pOc), 3) secondary oocytes (sOc), and 4) mature oocyte (mOc). Oog are small oval-shaped cells with irregular-shaped nuclei sizing 4–6 μm in diameter. They rest on the connective tissue germinal cord at the tip of each ovarian pouch (lobule). Oogonia increase their number through mitotic division, and the daughter cells move into ovarian pouch where they undergo first meiotic division to become primary oocytes, which have various steps of 1st meiotic prophase accumulating at the innermost zone of the ovarian pouch. The primary oocytes are small oval-shaped cells (8.5–10 μm in diameter) with large nuclei containing chromatin in various states of condensation that finally transform into chromatids. Their nuclei are surrounded by thin rim of faint blue-stained cytoplasm. The secondary oocytes derived from 2nd meiosis and comprise five steps: Oc1 and Oc2, classified as previtellogenic oocytes, Oc3 and Oc4, classified as vitellogenic oocytes, and mature oocyte (mOc) The zones of ovarian pouch are defined based on the accumulation of various steps of developing oocytes, namely, oogenic, previtellogenic, vitellogenic and mature zones, respectively. The ovarian cycle is divided into five stages based on the number and types of oocytes present in each stage. Stage 0 and I are spawn and spent stages. Stage II and III are proliferative and premature stages, while stage IV is mature stage. During ovarian stage I, each ovarian pouch contains primarily oogonia, primary oocytes, Oc1 and a few Oc2. In stage II, the pouch contains mainly Oc2 and Oc3, while in stage III the predominant cells are Oc4. Mature oocytes appear synchronously, in stage IV. The ovulating mature oocytes pass through the thin disrupted wall of ovarian pouch into subcapsular space, that leads into the oviduct situated on the ventro-lateral side of the ovarian lobe. At spawning, the ovarian pouches break down and only connective sheaths and hemolymph sinuses remain. The germinal cords and islets of oogonia remain in the central area of stage 0 ovary. The ovarian capsule, including the muscular layer, becomes attenuated as the ovary progresses from stage 0 to IV. The hemolymph vessels become highly convoluted in the central area of the ovary, and they branch radially into smaller hemolymph sinuses around each oogenic pouch.  相似文献

This study investigated the effects of serotonin (5-hydroxytryptamine or 5HT) on ovarian development in Macrobrachium rosenbergii de Man. Adult female prawns at the ovarian stage I (spent) were injected with 5HT at 1, 5, 10, 20 and 50 μg g− 1 body weight (BW) intramuscularly on days 0, 5 and 10, and sacrificed on day 15. The doses as related to the effect could be categorized into three levels: low (1 and 5 μg g− 1 BW of 5HT), medium (10 and 20 μg g− 1 BW of 5HT) and high (50 μg g− 1 BW of 5HT). The low-dose, especially at 1 μg g− 1 BW, caused prawns to exhibit a significant increase in ovarian index (ovarian weight/body weight × 100) (5.79 ± 0.09%) as compared to the control (1.49%). The ovaries of most of these prawns could develop to stage IV (mature) and contained synchronously mature oocytes while most of the control ovaries remained at stage I and II (proliferative), and contained only oogonia to previtellogenic (Oc1, Oc2) and early vitellogenic oocytes (Oc3). The medium- and high-dose treated prawns exhibited ovaries that could reach stages III and IV and contained various types of oocytes of different maturity. Pretreatment with 5HT receptor antagonist, cyproheptadine (CYP), at 10 μg g− 1 BW before 5HT injection significantly suppressed the effect of 5HT. Intramuscular injection of the 5HT-primed thoracic ganglion culture medium into CYP-pretreated prawns resulted in the increase of ovarian index about 5–6 times more than in the control, and in the groups injected with 5HT-primed media from muscle strip, eyestalk and brain. The ovaries of most prawn could develop up to stage IV and contained synchronously developed vitellogenic (Oc4) and mature oocytes (Oc5). These findings suggest that 5HT indirectly induces ovarian development and oocytes maturation in M. rosenbergii, probably via a putative ovarian stimulating factor released from the thoracic ganglia.  相似文献

A variety of natural foods have been reported to induce ovarian maturation of penaeid shrimp. In this study, conducted from 19 June to 11 September 1980, four single-food diets (clams, shrimp, squid, and worms) and one composite diet consisting of all four foods were fed to Penaeus vannamei in 3.7-m diameter indoor circular tanks. The shrimp, averaging 28 g, were stocked at a rate of 6.9 m-2 with a 1:1 male to female ratio. Unilateral eyestalk ablation was performed on half of the females in each tank. Tanks received one turnover of water volume daily and the water temperature and salinity averaged 27.0°C and 28.6 ppt, respectively. Daily photoperiod was maintained at 13 hours of light. Ovarian maturation, spawning, molting rate, and survival were monitored daily and growth and gonad size were measured at termination. The composite diet was the best overall diet, while squid was the best single-food diet, followed by shrimp, worms, and clams. No maturation of unablated females was observed with the exception of one female that developed to stage 3. Although the size of the shrimp was considered marginal for spawning, 5 mated females were observed, but only one (squid treatment) produced viable nauplii. After termination of this study, all shrimp were fed the composite diet for 41 days. During this period, ovarian maturation and spawning occurred in the previously unproductive treatment receiving only clams, and growth rate of shrimp in all previously single-food treatments increased 4 to 7 fold. The composite diet was also fed to two tanks of P. stylirostris (mean initial weight = 45.4 g) for a period of 118 days. During that time, 50 spawns were obtained with up to 3 spawns per female. A total of 4.3 × 105 P. stylirostris nauplii were produced.  相似文献

Outbreaks of multidrug-resistant pathogens have been constraint on aquaculture production in Thailand, thereby controlling shrimp pathogens by preventive probiotics being importance to sustain the aquaculture system. In this study, the effect of potential probiotics, Bacillus subtilis and Enterococcus sp. was related to growth, digestive enzyme activities (trypsin and chymotrypsin) and pathogenic resistance by postlarval black tiger shrimp (Penaeus monodon). The experiment was divided into two treatments, in triplicate, with and without supplementation of probiotics as a food additive. Shrimp fed with probiotics for a culture period of 84 days showed a significant increase (P < 0.05) in weight (2.03 ± 0.29 g) and survival (71.91 ± 3.15 %) in comparison with non-treated shrimp (1.53 ± 0.28 g and 65.20 ± 5.68 %, respectively). Trypsin activity of treated shrimp was significantly higher (P < 0.05) than that of control shrimp whereas chymotrypsin activities of the two treatments were not significantly different (P > 0.05). After challenging P. monodon postlarvae with a shrimp pathogen, Vibrio harveyi for 10 days, percentage of mortality of P. monodon postlarvae fed with probiotics was 46.67 ± 1.44 %, significantly lower (P < 0.05) than that of non-treated shrimp (61.67 ± 6.29 %). This study showed that potential probiotics was appropriate for application in P. monodon postlarvae cultivation under laboratory condition due to improvement of shrimp weight and survival, enhancement of trypsin activity and reduced mortality causing by pathogenic V. harveyi. This is the first publication reported the effect of probiotics on trypsin and chymotrypsin activities of P. monodon postlarvae.  相似文献

Spontaneous ovarian tunica albuginea contractility was evaluated in gilthead seabream (Sparus aurata L.) at different phases of the reproductive cycle. Fourteen adult females were sampled from February to November 2012 in a commercial fish farm, and ovaries were removed and processed for histological and contractility analyses. Fish reproductive stages were evaluated on haematoxylin–eosin-stained ovary sections or by simple macroscopic observation of hydrated oocytes in spawning individuals. Tunica albuginea spontaneous contractility was recorded by using ovary wall strips mounted in an organ bath containing modified Ringer’s solution. Ovary macro- and microscopic analyses allowed the identification of three different reproductive conditions: vitellogenesis, spawning and regressing. The gilthead seabream tunica albuginea was capable to contract spontaneously, and significant differences were found in mean contraction amplitude among the three reproductive states, with the highest value recorded in individuals in regressing condition and the lowest in individuals at spawning stage. No differences in mean contractility frequency among the three different groups were found. Possible involvement of spontaneous contractility in facilitating developing follicle advancement towards the ovarian lumen within the ovary and in supporting recovery of regressing ovaries may be hypothesized. The low contractility observed during the final oocyte maturation and spawning phases does not seem to support a role of tunica albuginea during ovulation, which could conversely involve theca cell contraction. Alternatively, possible single instantaneous contractions of tunica albuginea muscle fibres, not detected in the present study, could occur during ovulation in response to neuro-hormonal stimulations; a role of abdominal wall musculature in ovary “squeezing” and consequent release of ovulated eggs cannot be excluded.  相似文献

In this study, four species of penaeid shrimp from Pakistan's inshore waters were studied. These were Penaeus penicillatus (Alcock), Penaeus merguiensis (de Man), Metapenaeus affinis (Milne Edwards) and Parapenaeopsis stylifera (Milne Edwards). During maturation, shrimp ovaries pass through a series of colour changes. The various colours observed in the ovaries of the four species of penaeid shrimps were translucent, white, cream, yellow, green‐yellow, green‐white, light green and dark green. The different type of oocytes observed in histological sections of the penaeid ovaries are as follows: chromatin nucleolar oocytes, perinucleolar oocytes, yolkless oocytes, yolky oocytes and oocytes with cortical bodies. The ovarian developmental stages recognized in these shrimps are: undeveloped, developing, nearly ripe, fully ripe, resorbing and resorbing/ developing. The ovarian developmental stages were similar in all the four species with the exception of the fully ripe stage. In the fully ripe stage of P. penicillatus and P. merguiensis, the cortical bodies were present at the periphery in two shapes; spherical as well as rod‐like. On the other hand, in the fully ripe stage of M. affinis and P. stylifera, the cortical bodies were present only at the periphery and were only spherical. A relationship between colour of ovaries and histological stages of ovarian development was established. A female shrimp with a dark‐green ovary is an indication of the ready‐ to‐spawn condition. In P. merguiensis and Penaeus penicillatus, respectively, 100% and 88.1% females with dark‐green ovaries had oocytes with cortical bodies, whereas in M. affinis and P. stylifera, respectively, 50% and 28.6% females with dark‐green ovaries had oocytes with cortical bodies.  相似文献
