The purpose of this study was to determine whether gilthead sea bream and sea bass treated with combination of trimethoprim and sulfamethoxazole (TMP-SMX) differed in terms of physiological and innate immune biomarkers. Fish were exposed to TMP-SMX at 40 ppm concentration for 1 h as a prophylactic usage. Plasma cortisol, glucose, electrolytes (Ca, P, Na, K, Cl, and Mg) as well as plasma lysozyme activity, C-reactive protein (CRP), and ceruloplasmin (Cp) were measured soon after treatment and following 24 and 48 h in normal sea water for recovery. Treatment with TMP-SMX in both gilthead sea bream and sea bass led to an increase in plasma cortisol and glucose. Fluctuations in some electrolytes were found after treatment and during recovery period, however, the ratios of monovalent ions in treated sea bream were similar to control. Hematocrit values as well as plasma lysozyme activity in gilthead sea bream and sea bass were not affected by the treatment. CRP in gilthead sea bream and Cp in sea bass responded to the treatment with decreased levels. Both gilthead sea bream and sea bass displayed a rapid physiological stress response and sensitivity to TMP-SMX exposure, which requires more than 48-h period for regaining homeostasis.  相似文献

The present study examined the pharmacokinetics and bioavailability of oxytetracycline (OTC) in vannamei shrimp (Penaeus vannamei) after intra-sinus (10 mg/kg) and oral (10 and 50 mg/kg) administration and also investigated the net changes of OTC residues in the shrimp after the thermal, acid and alkaline processing methods. The hemolymph concentrations of OTC after intra-sinus dosing were best described by a two-compartment open model. The oral bioavailability was found to be 48.2 and 43.6% at doses of 10 and 50 mg OTC/kg, respectively. The peak hemolymph concentrations after 10 and 50 mg OTC/kg doses were 3.37 and 17.4 μg/ml; the times to peak hemolymph concentrations were 7 and 10 h. The elimination half-lives were found to be 15.0 and 11.5 h for the low and high dose, respectively. The residual OTC was rapidly eliminated from muscle with the elimination half-life value of 19.4 and 15.4 h, respectively, for the groups treated with doses of 10 and 50 mg/kg. The residual OTC levels in the muscle fell below the MRL (0.2 μg/g) at 72 and 96-h post-dosing at dose levels of 10 or 50 mg/kg, respectively. Residual OTC levels in muscle and shell were approximately 20–50% lower in the thermal treatment such as boiling, baking and frying. By the acid treatment, OTC residues were reduced to >80%, while those were reduced to around 30% by alkaline treatment.  相似文献

The white shrimp, Litopenaeus vannamei, has become a very important species for the development of shrimp aquaculture in Northwest Mexico. However, viral and bacterial diseases are considered a major threat to the development of this industry. In the present study a trial was conducted to evaluate the tissue distribution, maximum concentration, and elimination of the widely used antibiotic oxytetracycline (OTC) in L. vannamei using indoor tanks under laboratory-controlled conditions. OTC was given to shrimp simulating a therapeutic treatment through medicated feed for 14 days followed by a period of feeding without antibiotic for another 14 days to evaluate the elimination pattern. Samples of hemolymph, muscle, and hepatopancreas were taken from medicated animals every two days for 28 days. All tissues were removed and frozen immediately in liquid nitrogen. OTC levels were analyzed by High Performance Liquid Chromatography (HPLC). Results showed an important OTC increase during consumption of medicated feed in all examined tissues. OTC maximum concentrations were 33.54 ± 11.19, 194.37 ± 16.11, and 18.79 ± 5.87 µg g− 1 for muscle, hepatopancreas and hemolymph, respectively. Although the highest OTC level was found in the hepatopancreas, it required only two days after the start of dosing to reach this value, whereas the maximum OTC for muscle and hemolymph was detected after eight days of dosing. Ten days after the cessation of medicated feeding, the drug content in the shrimp tail muscle was under the detectable limit for the method (0.01 µg g− 1 of OTC).  相似文献

Olive flounder is the most important species for the Northeast Asian fish farming industry. However, this species is substantially affected by multiple infectious agents, including Streptococcus parauberis. Evaluation of antibiotics before their application is critical to treat infections and prevent drug resistance. Therefore, in this study, the pharmacodynamics of amoxicillin (AMX) and other antimicrobials against the planktonic‐ and biofilm‐forming bacteria were assessed. The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and Time–kill curve assay were analysed using micro‐dilution method. The minimum biofilm eradicating concentration (MBEC) was determined using the Calgary Biofilm device. The effects of temperature, pH, hardness and salinity were detected for both planktonic‐ and biofilm‐forming bacteria. The MIC of AMX ranged from 0.015 to 2 μg/ml, whereas that of cephalexin (CEP), enrofloxacin (ENR) and oxytetracycline (OTC) ranged from 0.125 to 256, 0.125 to >512 and 0.25 to >512 μg/ml respectively. No bacteria were resistant against AMX, while the percentage of resistance to CEP, OTC and ENR were 68.7%, 52.6% and 11.1% respectively. The IC50 of AMX, CEP, ENR and OTC was 0.03, 0.091, 0.015 and 0.213 μg/ml respectively. The MBEC of amoxicillin against S. parauberis ranged from 0.5 to 16 μg/ml. Higher rates of bacterial growth were obtained at 30°C, pH = 8 and salinity of 7.5–10 ppt. The hardness of the media suppressed the bacterial growth. In conclusion, AMX was found to be effective against both the planktonic and the biofilm forms of the prominent fish pathogen, S. parauberis.  相似文献

The present study was designed to explore pharmacokinetics of enrofloxacin and its metabolite ciprofloxacin in healthy and Vibrio alginolyticus-infected large yellow croaker (Pseudosciaena crocea) after a single 10 mg/kg oral dose. Concentrations of enrofloxacin and ciprofloxacin in serum, liver, kidney, muscle and skin of fish were determined using high-performance liquid chromatography. Pharmacokinetic parameters were analysed based on classical compartmental model analysis. The overall changes in enrofloxacin concentration–time curves in serum and tissues of diseased fish were similar to those of healthy fish. However, the peak concentration and peak time of enrofloxacin in serum and tissues were different in healthy and diseased fish. A delay of enrofloxacin peak time in serum and all tissues appeared in the diseased fish. The peak concentrations in serum and tissues of the diseased fish were lower than those of healthy fish. In healthy fish, the area under the concentration–time curve (AUC) was in the order serum >liver > kidney >muscle > skin, while AUC was serum >live > muscle >kidney > skin in the diseased fish. The peak concentrations of ciprofloxacin in the liver, serum, kidney, muscle and skin of healthy fish were 0.93 μg/g, 0.55 μg/ml, 0.36 μg/g, 0.37 μg/g and 0.12 μg/g respectively. Tmax of ciprofloxacin in the corresponding tissues was 8, 24, 12, 12 and 16 h respectively. In the diseased fish, the peak concentrations of ciprofloxacin in the corresponding tissues were 0.52 μg/g, 0.52 μg/ml, 0.41 μg/g, 0.27 μg/g and 0.13 μg/g respectively. Tmax in the corresponding tissues were 0.5, 8, 12, 16 and 48 h respectively. These data indicate that the health status of fish affects drug absorption and metabolism.  相似文献

Gilthead sea bream cultured along the northern Mediterranean coast are affected by the winter season when low temperatures reduce fish feed intake and growth. The coldest episodes can provoke a fish pathology known as ‘winter disease’. The effects of low temperatures, as well as concurrent fasting, have been studied by transferring three groups of gilthead sea bream from 16 °C to 14 °C, 12 °C and 8 °C. Fish at 12 °C and 8 °C refused food, whereas those at 14 °C were not fed following the temperature drop. Changes in body indices, organ composition, liver metabolism, and in particular, lipid fractions and their fatty acids were analysed on days 7 and 20 after the temperature shift. Only the rapid reduction of non-polar lipids in muscle was common for the three conditions. Fasting effects were linked to the maintenance temperature, being maximal after 20 days at 14 °C where fish body weight, hepatosomatic index, and perivisceral fat were reduced by 18%, 40%, and 60%, respectively. In this group, liver lipids did not change, as was the case for the enzymatic activities of liver glucose-6-phosphate and phosphogluconic acid dehydrogenases (G6PDH and PGADH) and lipoprotein lipase (LPL). In contrast, the liver of sea bream submitted to 8 °C accumulated large amounts of non-polar lipids (from 80 mg to 125 mg in 20 days), changing in size and aspect (bigger, pale, and friable). Simultaneously, liver LPL and hepatic lipase (HL) activities decreased. After 20 days at 8 °C, sea bream exhibited incipient acclimation responses to low temperatures: rising levels of unsaturation ratio in gill and liver polar lipids, of docosahexaenoic acid (DHA) in muscle polar lipids, and of G6PDH and PGADH hepatic activities.Fish at 12 °C presented some changes similar to those of the group at 14 °C (e.g., in morphological indices, and LPL and HL activities) and others like the group at 8 °C (increases in G6PDH and PGADH), suggesting a temperature threshold for gilthead sea bream (below 13 °C).  相似文献

To control black disease infecting fairy shrimp Branchinella thailandensis, the effects of concentrations and exposure time to three effective antimicrobials, which inhibited the pathogens in vitro, were evaluated. Exposure to sodium hypochlorite (NaOCl) caused a great toxicological response in the shrimp, 100% mortality was observed within 30 min–2 h at 5–20 μg mL?1. For oxytetracycline dihydrate (OTC) and chloramphenicol (CP), short‐term exposure to four high concentrations up to 5 h and long‐term exposure (12 days) to four low concentrations were used to determine an appropriate method for bath efficacy. Long‐term exposure to low concentrations was more toxic than the short‐term. Short‐term exposure to OTC showed the highest survival rate and CP was considered more toxic. The minimum survival rate of the shrimp exposed to both antibiotics at 250 μg mL?1 for 3 h was 83.3%. For determination of the bath efficacy, a short–term exposure (3 h) to OTC and CP was conducted using artificially infected shrimp. Administration of OTC and CP at 250 and 500 μg mL?1 resulted in the highest survival rates of 56.7% and 46.7% respectively. This study demonstrated that bath administration with OTC could be an alternative method for the treatment of black disease in fairy shrimp cultivation.  相似文献

采用高效液相色谱法检测土霉素,研究土霉素口灌给药途径下在锯缘青蟹体内的药代动力学。锯缘青蟹口灌给药土霉素50 mg/kg后,其血浆、肌肉和肝胰脏中的药峰浓度分别为16.78±1.98 mg/L、9.39±2.12μg/g和32.12±6.12μg/g,达峰时间分别为4 h、8 h和4 h。血浆中土霉素浓度-时间关系曲线符合一级吸收的二室开放动力学模型。土霉素在锯缘青蟹体内分布广泛,其表观分布容积(Vd)为2.129 L/kg;分布半衰期(t1/2α)和消除半衰期(t1/2β)分别为3.200 h和47.856 h,总体清除率(CLs)为0.063 mL/(kg.h)。肌肉和肝胰脏中土霉素浓度与时间关系的药动学参数采用统计矩原理分析,其消除半衰期(t1/2 z)分别为60.145 h和71.009 h,总体清除率(CLz)分别为0.054 g/(kg.h)和0.037 g/(kg.h)。土霉素在精巢和卵巢中达峰时间分别为8 h和12 h,峰浓度分别为9.83μg/g和10.26μg/g。给药后24 d时,血浆、肌肉、肝胰脏、精巢和卵巢中土霉素含量都已低于0.10μg/g。土霉素在锯缘青蟹体内消除比较缓慢。  相似文献

The efficacy of four chemical reagents, iodophor, formalin, hydrogen peroxide and bronopol as fish egg surface disinfectants were evaluated in bluefin sea bream (Sparidentex hasta). Fertilized eggs were counted and subjected to a static bath dip treatment in different concentrations of the above chemicals for 4 min before being incubated at 20 ± 0.5°C for 40 h. Treatment efficacy of the different disinfectants was evaluated by assessing the bactericidal activity, egg hatch percentage and survival of larvae up to 3 days post hatch. Results showed that iodophor at medium concentrations (75 and 100 ppm) was the best of all tested disinfectants in bacterial killing ability (12% reduction in the bacterial counts), egg hatching per cent (99.8% and 99.6% respectively) and larval survival up to 3 days post hatch (50.8% and 54.8% respectively). Formalin was the second best disinfectant at levels of 100 and 150 ppm. Hydrogen peroxide gave good results compared with the control while, bronopol showed discouraging results. In conclusion, iodophor appeared to be suitable for bluefin sea bream eggs disinfection with a 4 min exposure to 75–100 ppm when applied 14–16 h after egg fertilization.  相似文献

在14±2℃水温条件下,连续5d对红鳍东方鲍口灌剂量为100mg/kg的土霉素,采用高效液相色谱法测定了停药后血清、肌肉、肝脏组织中的药物含量、消除速率常数和消除半衰期,提出了该温度下的休药期。研究表明,在停药2d后红鳍东方鲍血清和肌肉中的药物浓度达到峰值,分别为1.092μg/ml和0.806μg/g;停药3d后肝脏内土霉素浓度达到峰值1.229μg/g,土霉素在血清、肌肉和肝脏中的消除半衰期分别为23.8、22.4和26.8d;土霉素在红鳍东方纯肌肉组织中降到0.1μg/g残留限量的时间为58d,降到0.05μg/g残留限量的时间为81d。  相似文献

This research was designed to study Sparus aurata (sea bream) biotransformation and detoxification responses to acute exposure to cadmium (Cd). Sexually immature gilthead sea bream were treated by intraperitoneal injection of Cd chloride (200 μg kg−1) for 6, 12, 24 and 48 h. Cd accumulation was quantified in sea bream liver by graphite furnace atomic absorption spectroscopy after the various exposure periods. The following biological responses were measured: (1) ethoxyresorufin-O-deethylase (EROD) activity as phase I biotransformation parameter, (2) liver glutathione-S-transferase (GST) activity as a phase II conjugation enzyme and metallothionein (MT) content as specific response to Cd contamination. Cd bioaccumulation in the liver resulted in an increasing uptake up to 10.3 μg g−1 wet weight after 48 h of exposure. EROD showed a significant activation only after 6 h exposure and a return to control levels after 12 h. GST revealed significant activation starting from 12 h exposure. MT accumulation in liver showed the same behavior as GST activation.  相似文献

Two experiments were conducted to investigate the effect of dietary taurine and cystine on growth and body composition of juvenile red sea bream Pagrus major. In Experiment I, a casein-based semi-purified diet included a small amount of fish meal were supplemented with taurine at the levels of 0 (control) and 1.0%. The experimental diets in Experiment II were without fishmeal and supplemented with taurine at 0 (control), 0.5, 1.0 and 2.0% or cystine at 1.0 and 2.0%. These diets were fed three times a day for 6 weeks to fish (average body weight: 2.3 g in Experiment I and 2.5 g in Experiment II). In Experiment I, fish fed the taurine-supplemented diet showed significantly (P < 0.05) improved growth, feed efficiency and feed consumption relative to fish fed the unsupplemental diet. The whole body taurine content increased, whereas the non-essential amino acid contents decreased, in fish fed the taurine-supplemental diet compared to fish fed the unsupplemented diet. In Experiment II, the growth, feed efficiency and feed consumption of fish fed the taurine-supplemented diets, irrespective of the dietary taurine levels, were significantly higher than those of fish fed the control diet and the cystine-supplemented diets. Taurine content in the whole body increased with the dietary taurine level, while the taurine contents did not increase by the supplemental cystine. Other free amino acid contents in the taurine-supplemented diet groups followed similar trends to those in Experiment I. These results indicate that supplemental taurine to a casein-based semi-purified diet at more than 0.5% improved the growth and feed performance of juvenile red sea bream. It is also suggested that juvenile red sea bream cannot metabolize cystine into taurine.  相似文献

In the present study biotransformation and detoxification responses to acute exposure to the polycyclic aromatic hydrocarbons benzo[a]pyrene (B[a]P) were investigated in the liver of Sparus aurata (sea bream). Sexually immature gilthead sea bream were treated by intraperitoneal injection of B[a]P (20 mg kg−1) for 6, 12, 24, and 48 h. B[a]P accumulation was quantified in sea bream liver by mean of gas phase chromatography (GPC-MS) after the various exposure periods. The following biological responses were measured: (1) ethoxyresorufin-O-deethylase (EROD) activity, as a phase I biotransformation parameter; (2) liver glutathione S-transferase (GST) activity as a phase II conjugation enzyme. DNA damage was assessed over time using the single-cell gel electrophoresis comet assay. B[a]P bioaccumulation in the liver resulted in a biphasic curve with an increasing uptake up to 5.55 ± 0.67 μg g−1 dry weight after only 6 h exposure and 4.67 ± 0.68 μg g−1 dry weight after 48 h exposure. EROD activity showed a nonsymmetrical bell-shaped kinetic with a maximum at 24 h and lower but significant activities at 12 and 48 h with respect to control animals. Hepatic GST activities were only significant after 48 h exposure. Comet assay showed an increase in liver cells DNA damage with a maximum after 48 h exposure reaching up to 12.17 %DNA in the tail.  相似文献

首次报道了黑鲷口服土霉素的药物代谢动力学特征,用高效液相色谱法测定组织中的药物含量,药物在肌肉,血液,肝脏中的平均回归率分别为85.61%,85.38%,82.005,该方法的检测限可达0.01μg/g,黑鲷1次口服剂量为75mg/kg的土壤素后,其血液药物浓度-时间数据符合一室开放动力学模型,吸收速率常数(ka)为0.296/h,达峰时间(Tamx)为10.635h,峰浓度(Cmax)为1.398μg/ml, 分布半衰期(T1/2a)为2.339h ,消除半衰期(T1/20β)为46.663h,药时曲线下面积(AUC)为110.25mg/L.h,黑绸口服药物0.5h后在血液,肥肉,肝脏,肾脏4种组织中就可以检测到药物的存在,药物在16h的采样点浓度达最高,分别为1.68μg/ml,1.68,2.52,6.77μg/g。  相似文献

为探究日粮中不同蛋白质与碳水化合物水平对黑鲷肌肉营养组成和质地的影响,实验配置了41.0%和37.5%2种蛋白质水平下3种碳水化合物水平(以小麦面粉为碳水化合物源,使用量分别为19%,24%,29%)的实验饲料,分别记为L1(19/41),L2(24/41),L3(29/41),L4(19/37.5),L5(24/37.5),L6(29/37.5)。实验以黑鲷幼鱼[均重(9.95±0.22)g)]为研究对象,投喂上述实验饲料养殖8周后,分析肌肉常规营养与氨基酸组成,测定肌纤维密度,并采用TA.XT plus质构分析仪进行质地多面剖析(TPA)模拟测试。结果显示,不同饲料蛋白质与碳水化合物比例对黑鲷背肌粗蛋白、粗脂肪无显著性影响(P0.05);对背肌谷氨酸、丙氨酸、亮氨酸和蛋氨酸含量均具有显著影响(P0.05),而对其余氨基酸含量与总量均无显著性影响(P0.05);此外,对背肌硬度产生显著性影响(P0.05)。在同一碳水化合物水平下,蛋白质水平降低,背肌硬度、胶性显著减小(P0.05),恢复性显著增大(P0.05);而在同一蛋白水质平下,随着碳水化合物水平升高,背肌硬度、恢复性出现显著性差异(P0.05);蛋白水平降低,肌纤维密度下降(P0.05)。研究表明,2种蛋白质水平下不同碳水化合物水平对黑鲷背肌的常规营养组成无显著性影响,但对氨基酸组成和质地有一定影响。  相似文献

The aim of this study was to determine the gill histopathological characteristics of Caspian roach (Rutilus rutilus caspicus) fingerlings exposed to different concentrations of potassium permanganate (PM) and formalin (FO). The fish were exposed to different PM and FO concentrations over different periods: 1 ppm PM for 24 h (1‐P24), 2 ppm PM for 24 h (2‐P24), 5 ppm PM for 0.5 h (5‐P0.5), 10 ppm PM for 0.5 h (10‐P0.5), 10 ppm FO for 24 h (10‐F24), 20 ppm FO for 24 h (20‐F24), 40 ppm FO for 0.5 h (40‐F0.5) and 80 ppm FO for 0.5 h (80‐F0.5). After exposure, gill samples were collected for histological examination. The most frequent gill histopathological symptoms were hyperplasia and epithelial lifting. The order of the gill injury index (Ig.i) among the treatments was as follow: 1‐P24, 2‐P24 < 5‐P0.5, 40‐F0.5 < 10‐F24 < 10‐P0.5, 20‐F24, 80‐F0.5. Comparing the two agents, it is concluded that there is no significant difference in the gill damages between the agents during the short‐bath treatments. However, PM causes less pathological symptoms than FO during the long‐bath treatments. Comparing the bath durations, the agents showed different behaviours. PM‐treated fish showed significantly lower gill damage during the long‐bath compared to the short‐bath treatment; whereas, pathological symptoms of FO‐treated fish was dose‐dependent for each exposure period, separately. Overall, exposure of Caspian roach to PM, at the concentrations recommended for long bath, resulted in milder gill damages compared to FO.  相似文献

The effects of dietary amino acid profile (based on muscle (M) or whole body composition (WB) and the balance between indispensable (IAA) and dispensable amino acids (DAA) in the diet, on plasma levels of insulin and glucagon, were analysed in rainbow trout and gilthead sea bream. Plasma insulin values (baseline and 6 h postfeeding) were higher in trout than in sea bream, but the relative postfeeding increase was more pronounced in sea bream. Within the same dietary amino acid profile, diets with lower IAA/DAA, had a lower effect on the postfeeding secretion of insulin in both species. Circulating levels of glucagon (baseline and postfeeding relative increases) were higher in sea bream. In trout, diets with WB amino acid profile had a greater secretory effect on postfeeding glucagon than did diets with M profile, while gilthead sea bream showed an inverse response to circulating glucagon with respect to diet. Muscle insulin and insulin growth factor-I binding parameters were not affected by the dietary regimen. The postfeeding glucagon response depends on both the dietary AA profile and the fish species, while that of insulin seems to be more uniform, and is affected in a similar way regardless of the species.  相似文献

The common dentex (Dentex dentex) is a sparid fish which is considered a suitable candidate for Mediterranean aqua-culture. A seven-weeks feeding trial was conducted over the summer period with common dentex, sea bream (Sparus aurata) and sea bass (Dicentrarchus labrax). All three species were fed to visual satiety with two practical diets with varying crude protein and crude fat levels (55 % protein, 9 % lipids; 46 % protein, 17 % lipids). The initial body weights were 8.2, 11.6 and 17.3 g for common dentex, sea bass and sea bream, respectively. In all cases, specific growth rates were not significantly affected by dietary treatment, but they varied among species (sea bass 1.7–1.8 %; sea bream 2.1–2.3 %; common dentex 3.1–3.2 %). When comparisons between fish species were made, we observed that the increase in growth rates was linked to a decrease of the whole body fat content, as a consequence of a greater utilisation of dietary lipids as energy substrates. Furthermore, in common dentex, the 17 % lipid diet was able to improve protein retention, but this diet effect was less significant in sea bream and sea bass. In contrast to sea bream, the 17 % lipid diet did not up-regulate plasma GH levels in common dentex, which provides additional evidence for a more efficient utilisation of dietary lipids.  相似文献

Substitution of marine ingredients (FM‐FO) by plant protein and oil sources can modify selenium (Se) levels in feeds. Se plays an important role in the antioxidative defence by forming part of selenoproteins. Se requirements of gilthead sea bream are not accurately determined; therefore, this study was conducted to define Se supplementation levels in low FM‐FO practical diets for sea bream fingerlings. A plant‐based diet containing 0.45 mg Se/kg diet was used as the basal diet. Four other diets were supplemented to contain 0.68, 0.86, 1.00 or 1.70 mg Se/kg diet, supplied as sodium selenite. Sea bream, weighing 12.6 ± 1.4 g, were distributed in triplicate groups per diet and fed for 42 days. Se supplementation up to 1.00 mg Se/kg significantly improved the growth of sea bream, whereas further increase up to 1.70 mg Se/kg diet reduced growth. The results of this study suggest that the optimum dietary levels of sodium selenite in diets with low FM‐FO with basal levels of 0.45 mg Se/kg are around 0.94 mg Se/kg to promote growth of gilthead sea bream juveniles. On the contrary, dietary levels of 1.70 mg Se/kg were found to be excessive and caused growth reduction, increased catalase expression and hydropic degeneration in the liver.  相似文献

Asian seabass (Lates calcarifer) is an economically important fish in Asian and Australian markets, but few pharmacokinetic (PK) data of antimicrobial drugs in this species is available. The present study investigated the PK behaviour of florfenicol (FF) through medicated feed in Asian seabass cultured at 25°C. The serum and muscle/skin concentrations of FF and its metabolite florfenicol amine (FFA) were determined by the HPLC-FLD method and analysed by one-compartmental model. The optimal dosages were determined by pharmacokinetic-pharmacodynamic (PK-PD) approach and the linear regression analysis was used to determine the withdrawal time (WDT). The PK study following a single oral administration of 15 mg/kg FF via medicated feed revealed that the absorption half-life (t1/2Ka), elimination half-life (t1/2K), peak concentration (Cmax), area under the concentration-time curve (AUC), volume of distribution (Vd/F) and clearance (CL/F) were 1.47 h, 8.07 h, 8.61 μg/ml, 146.41 h·μg/ml, 1.19 L/kg and 0.102 L/kg/h, respectively. The muscle/skin concentration-time profile was similar to that of the serum, suggesting well distribution but only a small fraction of FF was metabolized to FFA. The optimal dosage for a minimum inhibitory concentration of 2 μg/ml was calculated as 13.38 mg/kg/day. The appropriate WDT after multiple oral medications with 15 mg/kg FF once daily for 7 days was determined as 8 days. Information obtained from the current study can potentially be applied for the treatment of bacterial diseases in farming Asian seabass.  相似文献
