Though artificial propagation of Asian seabass Lates calcarifer (Bloch) in captivity through induced breeding techniques is standardized under Indian conditions, larval and nursery rearing techniques including suitable nursery feeds have to be standardized to obtain better survival and growth. Feeding experiments in triplicate were conducted to evaluate the suitability of the marine copepod Acartia clausi as live prey for fourteen day-old seabass larvae (6.53 ± 0.06 mm; 8.58 ± 0.33 mg) and compared with the traditional live prey, rotifers and Artemia nauplii. While A. clausi and rotifers were mass produced using algae Isochrysis galbana, Chaetoceros affinis and Chlorella marina, Artemia nauplii were produced using cysts. Nutritional quality of cultured copepods was evaluated based on the proximate composition, amino acid and fatty acid composition, and compared with that of rotifers and Artemia nauplii. Proximate composition varied significantly (P < 0.05) among the different live feeds. A. clausi showed higher protein (63.12%) and lipid (16.65%) content than Artemia nauplii and rotifers. Total essential amino acids content was 2% lower in A. clausi compared to that in Artemia nauplii. Fatty acid profiles of the live feed organisms showed that A. clausi is a rich source of n − 3 fatty acids. The total n − 3 fatty acid content of A. clausi was 33.94%. Length, weight overall weight gain and survivorship were significantly (P < 0.05) different among the dietary treatments, and weight gain was comparatively higher in A. clausi fed larvae. Survival of seabass larvae fed A. clausi was obtained highest as 58.13% against the lower values of 39.93% and 41.62% in larvae fed rotifer and Artemia nauplii respectively. Final carcass composition of the larvae of L. calcarifer fed different live-food organisms showed significant differences (P < 0.05) among the dietary treatments. The fatty acid composition of the dietary treatments was reflected to a certain extent in the fatty acid composition of the seabass larvae. The present investigation revealed the nutritional value of calanoid copepod and thus underlining its usefulness as a suitable live-food organism for rearing larvae of the commercially valuable Asian seabass.  相似文献

White spot syndrome virus (WSSV) is the most lethal pathogen of cultured shrimp. Previous studies done with undefined WSSV titers showed that high water temperature (32-33 °C) reduced/delayed mortality of WSSV-infected shrimp. This study evaluated the effect of high water temperature on the clinical and virological outcome of a WSSV infection under standardized conditions. Groups of specific pathogen-free Litopenaeus vannamei were challenged either by intramuscular or oral routes with a low (30 SID50) or a high (10,000 SID50) virus titer. Shrimp were kept (i) continuously at 27 °C, (ii) 30 °C or (iii) 33 °C; (iv) maintained at 33 °C before challenge and 27 °C afterwards, or (v) kept at 27 °C before challenge and 33 °C afterwards. Shrimp were maintained at the respective temperatures for 120 h before challenge and 120-144 h post challenge (hpc). Gross signs and mortality were monitored every 12 h until the end of the experiment. Dead and surviving shrimp were screened for WSSV infection (VP28-positive cells) by indirect immunofluorescence (IIF). Shrimp kept continuously at 27 °C or 30 °C, or switched to 27 °C post challenge developed gross signs within 24 hpc, first mortalities at 36-60 hpc and 100% cumulative mortality between 60 and 144 hpc depending on the virus titer. All dead shrimp were WSSV-positive. In contrast, shrimp kept at 33 °C continuously or after WSSV challenge showed no signs of disease and low mortalities (0-30%) regardless of the virus titer. Dead and surviving shrimp were WSSV-negative. Further, early virus replication was studied in two groups of shrimp: one maintained at 27 °C before and after challenge and one switched from 27 °C to 33 °C after challenge with 10,000 SID50. Immunohistochemistry (IHC) analysis showed that WSSV-positive cells were first displayed at 12 hpc in shrimp kept at 27 °C and by 24 hpc the infection became systemic. In contrast, shrimp kept at 33 °C did not display WSSV-positive cells at 12 or 24 hpc. This work confirms previous reports that high water temperature prevents the onset of disease and significantly reduces mortality of WSSV-inoculated shrimp regardless of the route of inoculation or virus titer used. This strategy may have practical applications to control WSSV in tropical shrimp farming countries.  相似文献

This is the first report on the successful larval rearing of captive bred population of Chitala chitala (Hamilton). C. chitala is one of the endangered fresh water fish species in India for which the development of controlled larval rearing procedures are needed for stock enhancement. Fifteen days old post-hatchlings were stocked for 28 d in a 30 L recirculatory tanks using eight different diets i.e. live feed (tubifex worms, chironomous larvae, zooplanktons,), dry feed (dry tubifex, spirulina, daphnia) and other non-conventional feed (fish eggs and boiled egg-yolk). Fishes accepted all types of diets. The study revealed that specific growth rate (SGR) was higher in post-hatchlings fed on live tubifex worms (2.40 ± 0.72) followed by fish eggs (2.15 ± 0.71), dry tubifex (2.12 ± 0.40), chironomous larvae (1.91 ± 0.44), spirulina (1.79 ± 0.38), daphnia (1.42 ± 0.79) and planktons (1.37 ± 0.77) whereas minimum SGR was recorded with boiled egg-yolk (0.63 ± 0.5). A highly significant difference (p < 0.01) in SGR was observed in fish fed on live feed (tubifex worms, chironomous larvae, planktons, spirulina), dry tubifex and fish eggs whereas for daphnia and boiled egg-yolk it was only significant (p < 0.05). The final mean weight and weight gain showed highly significant difference (p < 0.01) in live tubifex, zooplanktons, spirulina, chironomous larvae, dry tubifex and fish eggs, whereas daphnia and boiled egg-yolk fed larvae showed significant difference (p < 0.05). Highest mean survival rate on day 28 was observed in live tubifex worms (94%) and chironomous larvae (92%). The post-hatchlings reared with spirulina and daphnia showed same survival rate of 88% whereas the lowest mean survival of 66% was recorded in boiled egg-yolk. The experiments showed that captive bred post-hatchlings of C.chitala could be reared in experimental recirculatory system for attaining higher growth and survival during early life stages. However, methods to improve the larval rearing have to be improved further for commercial farming of the species.  相似文献

Cell membranes from the rotifer, Brachionus urceus, were obtained by centrifugation and found to specifically bind white spot syndrome virus (WSSV) in vitro. This finding suggests that there is likely a WSSV receptor on the rotifer cell membrane and provides evidence that rotifers may be a host for WSSV.  相似文献

Chilled storage of spermatophores from white shrimp (Litopenaeus vannamei) is needed to generate a consistent and reliable supply of spermatozoa for domestication purposes. The objective of this study was to develop a protocol for the chilled storage of white shrimp spermatophores and to evaluate bacterial propagation during such storage. In the first experiment, spermatophores were immersed in four extenders, mineral oil, Ringer's solution, phosphate buffer and 0.85% NaCl, and stored at low temperature (2-4 °C) for 35 days. Characteristics of preserved spermatophores changed the least and viable sperm was highest when spermatophores were stored in mineral oil. Spermatophores preserved with mineral oil appeared morphologically normal. Bacillus circulans, Staphylococcus hominis and S. lugdunensis, S. sciuri, S. xylosus and Micrococcus spp. were identified as the predominant bacteria during chilled storage, and total bacterial counts gradually increased during the experiment. A second experiment investigated the effect of antibiotic on chilled storage. Spermatophores were preserved in only mineral oil or mineral oil with 0.1% penicillin-streptomycin. These were evaluated for changes in external morphology of spermatophores, sperm viability and total bacteria count every week during a 35-day experimental period. Percentages of viable sperm (69.5 ± 3.9%) were significantly higher (P < 0.05) among spermatophores preserved in mineral oil with 0.1% antibiotic compared with those preserved only in mineral oil (57.7 ± 3.4%) over 35 days. The number of total bacteria in the treatment with mineral oil ranged between 28.3 ± 4.8 and 2416.7 ± 299.4 CFU/g, but in mineral oil containing antibiotic bacteria were undetectable. This study suggests that chilled storage of spermatophores is a feasible approach for the management and spawning of white shrimp broodstock.  相似文献

The spider crab Maja brachydactyla is overexploited on the NW coast of Spain. Aquaculture of this species can be the solution to the problem, and consequently, several attempts of intensive larval rearing have been conducted. However, most of the studies already published do not provide enough zoo technical data, especially in terms of larval and prey densities or the nutritional quality of diets used for rearing.Three experiments were carried out to evaluate the conditions for intensive larval rearing of M. brachydactyla. Larval stocking density (10, 50 and 100 larvae L− 1), prey:larva ratio (15, 30 and 60 enriched Artemia larva− 1) and diet (enriched Artemia, non-enriched Artemia and polychaete supplement) effects on growth and survival of this species were studied. For larval culture nine, 35 L, 150 μm mesh-bottomed PVC cylinders (triplicates for each treatment and larval stage) connected to a recirculation unit, were used. Temperature and salinity were kept constant at 18 °C and 36‰ respectively. A 12 to 18 day trial was conducted for each experiment and samples of larvae were collected at each larval stage (zoea I, zoea II, megalopa) in the inter-molt phase and at first juvenile. Survival, carapace length and width, dry weight (DW), and proximate biochemical content (protein, carbohydrates and total lipid) as well as lipid class composition were determined.Stocking densities of 100 larvae L− 1 resulted in higher growth in DW and higher content in lipids and protein for zoea I (ZI) and zoea II (ZII) than 10 larvae L− 1. However, survival decreased with increasing stocking density.The use of 60 preys larva− 1 produced larvae with significantly higher DW and protein content, especially at ZII stage, than lower prey densities. Survival rate obtained feeding 60 preys larva− 1 up to the megalopa stage was almost two-fold (42.2%) the rate obtained using 15 preys larva− 1 (24.8%).Larvae fed on enriched Artemia (EA) showed an increase in weight up to megalopa (518.9 ± 26.5 μg) in contrast to larvae fed on non-enriched prey (A) (467.9 ± 6.9 μg). Variation in DW correlated with the total lipid content (L) of the larvae (LEA = 70.1 ± 37.5 μg ind− 1; L= 28.9 ± 3.2 μg ind− 1) especially in terms of neutral lipids. The use of an initial density of at least 50 larvae L− 1 and 60 enriched Artemia larva− 1 can be considered the most adequate rearing parameters in order to obtain good results in growth and survival of M. brachydactyla.  相似文献

As has been demonstrated in previous studies, Octopus maya can be fed on artificial diets. In the present study six different diets were assayed. Five diets were designed to test the effect of percentage of inclusion of fish protein concentrate (CPSP: 0, 5, 10, 15, and 20%) and were offered to octopuses as a specifically designed artificial diet. The sixth diet consisted of frozen crab (Callinectes spp) and was used as control diet. Blood metabolites and energy budget of octopuses were evaluated to determine how CPSP levels modulate the digestive capacity and allow retaining energy for growth. Wild animals (316.4 ± 9.8 g) were used in the study. Results showed that CPSP produced a positive specific growth rate (SGR, % day− 1) with high value in octopuses fed 15% CPSP level. A maximum growth rate of 0.86% day− 1 was recorded in these animals, a value that is extremely low when compared with the SGR obtained when animals were fed fresh crab (3.7% day− 1). In general, blood metabolites were affected by diet composition, indicating that some metabolites could reflect the nutritional and/or physiological status of octopus. Preliminary reference values for O. maya fed crab were found for glucose (0.09 ±0.02 mg/ml), lactate (0.004 ± 0.002 mg/ml), cholesterol (0.16 ± 0.02 mg/ml), acylglycerol (0.14 ± 0.01 mg/ml), protein (0.37 ± 0.04 mg/ml), hemocyanin (1.85 ± 0.04 mmol/l), and digestive gland glycogen (1.86 ± 0.3 mg/g). Total energy content can be used as an indicator of tissue metabolic reserves. In the present study, higher energy content in the digestive gland and muscle was observed in octopuses fed crab, followed by animals fed 15% CPSP. Results from the digestive gland indicated that the retained energy derived from glycogen, suggesting that lipids and protein were the main sources of variation linked with energy content. In general, digestive gland proteases activity and trypsin were induced in octopuses fed 15% CPSP. The capacity of O. maya juveniles to adjust their digestive enzymes to different types of food was evidenced. Essential amino acid content (EAA) of the diet was not a limiting factor. When dietary EAA profiles were compared with O. maya EAA profiles, all dietary EAA resulted in a higher concentration than whole body octopus composition. In the present study, all experimental groups ingested between 3300 and 4106 kJ wk− 1 kg− 1 without statistical differences among treatments, indicating that experimental diets were as attractive as crab. Differences were recorded in the proportion of absorbed energy (Ab, %) between CPSP-based and crab meat diets, suggesting digestion limitations associated with artificial diets. The present results indicate that the 15% CPSP diet had characteristics that stimulate digestive enzymes and reduce energetic costs associated with its digestion (HiE or SDA), channeling more biomass production than the other experimental diets.  相似文献

Stimulation of the non-specific defence enhances the disease resistance and growth, and has good potentials as a measure for increased microbial control in juvenile production of marine fish and shellfish. So far, the most commonly used immunostimulants are β-(1 → 3, 1 → 6)-glucans, and in this study the stimulatory potential of a β-(1 → 3, 1 → 6)-glucan of marine origin, the storage polysaccharide from the marine diatom Chaetoceros mülleri, was examined. The glucan (chrysolaminaran) was extracted from cultures of C. mülleri, and used as a dietary supplement in two first feeding experiments with larvae of Atlantic cod Gadus morhua L. In one experiment the microalgal glucan was compared to the commercial yeast-glucan product MacroGard®, and in the other to an alginate with a high content of mannuronic acid (High-M alginate) isolated from Durvillaea antarctica. The stimulants were given via rotifers, and weaning to formulated feed was initiated at day 17 or 18 after hatching. The survival ± SEM at day 27 after hatching was 24.5 ± 2.0%, 14.8 ± 4.5% and 13.1 ± 1.4% for the groups fed C. mülleri-glucan, yeast glucan and for the control, respectively, in the first experiment. The group fed C. mülleri-glucan group had higher survival compared to the control (< 0.05) group, whereas the yeast glucan had no positive effect on the survival (p > 0.05). The dry weights of the groups at day 27 were low, with 203.2 ± 52.2, 165.2 ± 43.4 and 198.5 ± 58.1 μg per larva for the C. mülleri-glucan, yeast glucan and control groups, respectively. In the second experiment the survival in the period of feeding formulated feed (days 18-30) were 44.6 ± 4.3%, 44.7 ± 1.3%, and 33.8 ± 4.1% survival for the C. mülleri-glucan, High-M alginate and control group, respectively. The cod larvae fed C. mülleri-glucan reached an average weight of 531.6 ± 17.2 μg at day 30, which was significantly higher (p < 0.05) than the control group that had an average of 473.6 ± 3.5 μg. The larvae fed High-M alginate had an average weight of 470.3 ± 31.6 μg per larva at day 30, and not significantly different from the control (p > 0.05). The early weaning to formulated diet had detrimental effect on the growth of the larvae. In both experiments the C. mülleri-glucan group was the only group showing a positive growth rate in the period of weaning to dry feed. The microbial conditions in larval gut and water were monitored with respect to total colony forming units on Marine agar, and Vibrio- and Pseudomonas-like species on selective agars (TCBS and marine Pseudomonas Agar with CFC-supplement). The larvae were rapidly colonised after hatching, but no or weak effects of the stimulants were observed on the colonisation rates or the composition. The total CFU varied from 101 to 102 CFU per μg larva after initiation of the first feeding. The percentages of Pseudomonas-like bacteria increased throughout the period, whereas the levels of Vibrio-like bacteria were low and stable. The chrysolaminaran from the diatom C. mülleri was shown to be a promising candidate for use as an immunostimulatory feed additive, and which should be further explored.  相似文献

As natural diets of fish larvae, a number of calanoid copepod species are being investigated for use as live prey in aquaculture hatcheries. One of these, the tropical calanoid copepod, Acartia sinjiensis, has good potential as a live feed for tropical reef fish larvae. However, the rearing techniques for A. sinjiensis require further development to improve productivity. This study was carried out to investigate the population growth and egg hatching success of A. sinjiensis when fed a range of mono-and binary algal diets, including algae in the form of frozen paste.For the population growth experiment, the final A. sinjiensis population, including eggs, nauplii, copepodites and adults, was determined after feeding eight algal diets (two frozen algae, four live monoalgal and two live binary algal diets) for 8 days at temperature 28 ± 1 °C; salinity 34 ± 1 psu and photoperiod 12 L:12 D. Five replicates, with an initial 12 adult A. sinjiensis per replicate, were set up for each treatment. In a separate experiment, effects of diets on egg hatching success were examined after 48 h incubation of eggs produced by A. sinjiensis fed the same eight diets.The results showed that diet significantly affected both population growth and hatching success of A. sinjiensis. Of the diets tested, the binary algal diets were more successful than monoalgal diets, while the frozen algae had little dietary value. The highest population growth was recorded on A. sinjiensis fed a binary diet of Tetraselmis chuii and the Tahitian strain of Isochrysis sp. (T-ISO) (final population: 1091 ± 80), which was significantly higher (P < 0.001) than all other diets tested except for the other binary diet of Nannochloropsis sp. and T-ISO (final population: 897 ± 123). Diet also had a significant effect (P < 0.001) on egg hatching rate, though the highest hatch rate was recorded with eggs produced by A. sinjiensis fed binary diet Nannochloropsis sp. and T-ISO (88.1 ± 2.1%), this was not significantly different from that of eggs produced by A. sinjiensis fed either T-ISO alone (88.0 ± 1.7%) or the binary algal diet of T. chuii and T-ISO (76.4 ± 7.1%). The results of this study suggest that among the diets tested, a combination of live T. chuii and T-ISO was the best for the culture of A. sinjiensis.  相似文献

The effect of the inclusion of probiotics (Vibrio alginolyticus) and β-1,3/1,6-glucans in Penaeus vannamei larviculture was evaluated by measuring the immune response and survival of shrimp juveniles subjected to white spot syndrome virus (WSSV) challenge tests (per os) and pond culture. Treatments were designed to contrast the probiotic factor (inclusion vs non-inclusion) and β-1,3/1,6-glucans supplementation in various larvae stages; starting from early stage (Zoea II), to middle stage (PL 12), late stage (15 days pre-challenge) or non-supplemented. In larviculture, the highest survival was obtained in animals treated with β-1,3/1,6-glucans from Zoea II. The use of probiotics enhanced survival during the first 0-52 h post-WSSV challenge period. During 56-156 h post-WSSV challenge period, interactions were observed between β-1,3/1,6-glucans × time and β-1,3/1,6-glucans × time × probiotics. Significant differences in final survival rates between treatments were not observed. In the second WSSV challenge, immune parameters were analysed. Significant interaction between probiotics and β-1,3/1,6-glucans was observed for plasmatic protein (PP) concentration, super oxide anion (O2) generation, antibacterial activity (AA), and total haemocyte count (THC). The use of probiotics in larviculture had a negative effect on the PP, but increased the AA and THC, while β-1,3/1,6-glucans had a negative effect on the O2 generation. The most relevant results were obtained from treatments T2 (probiotics in larviculture, β-1,3/1,6-glucans from Z II) and T4 (probiotics in larviculture, β-1,3/1,6-glucans 15 days before challenge). Treatment T2 presented the highest survival rate in larviculture. After WSSV infection, the animals of this treatment displayed resistance to the virus, a strong AA and increase of THC. Treatment T4 increased the amount of PP, increased the O2 generation and THC. Histological analysis showed that the animals of treatment T2 and T4 were able to limit the spread of the virus during the first hour after challenge with WSSV. The survivors from treatments T2 and T4 had a high THC, accompanied by a lack of white spot disease (WSD) injuries. A bioassay was carried out under farm conditions during the warm-rainy season using larvae from treatments T2 and T4. The animals were stocked at 18 animals/m2 in earth ponds of 0.20 ha (three ponds/larvae kind). WSD outbreak was not detected, and the survival was significantly higher in ponds stocked with larvae from treatment T4 (70 ± 3%) than in ponds stocked with larvae from treatment T2 (49 ± 9%).  相似文献

Two infection protocols, individual oral (IO) and waterborne infection (WB), were evaluated to challenge Penaeus (Litopenaeus) vannamei with White Spot Syndrome Virus (WSSV). Five different batches of full and half-sib families were infected experimentally and tested for growth performance under commercial growth conditions. The genetic variance for WSSV resistance was estimated using a linear sire-dam repeatability model that considers test-day survival as the dependent variable. The heritability estimates using the IO protocol ranged from 0.01 ± 0.00 to 0.02 ± 0.01, whereas the heritability estimates using the WB infection protocol were not significantly different from zero. The genetic correlations measured as the product moment correlation between full-sib family breeding values for resistance to WSSV and harvest body weight in ponds were unfavourable in three of the five batches and favourable in one of them. The dosage of WSSV was better controlled with IO oral infection than with other methods, with all animals being exposed to approximately the same risk of infection at the same time. This should improve the accuracy of the genetic parameters and hence improve the accuracy of the breeding values. It should, however, be noted that once the outbreak was established and the mortalities began, the shape and the magnitude of the slope of the mortality curves showed little difference in the infection pattern between batches irrespective of the dosage and infection protocol, and in most of the cases the cumulative mortality was greater than 80%. The main reason for this is probably the high densities of animals in the tanks needed for the genetic evaluations.  相似文献

The white shrimp, Litopenaeus vannamei, has become a very important species for the development of shrimp aquaculture in Northwest Mexico. However, viral and bacterial diseases are considered a major threat to the development of this industry. In the present study a trial was conducted to evaluate the tissue distribution, maximum concentration, and elimination of the widely used antibiotic oxytetracycline (OTC) in L. vannamei using indoor tanks under laboratory-controlled conditions. OTC was given to shrimp simulating a therapeutic treatment through medicated feed for 14 days followed by a period of feeding without antibiotic for another 14 days to evaluate the elimination pattern. Samples of hemolymph, muscle, and hepatopancreas were taken from medicated animals every two days for 28 days. All tissues were removed and frozen immediately in liquid nitrogen. OTC levels were analyzed by High Performance Liquid Chromatography (HPLC). Results showed an important OTC increase during consumption of medicated feed in all examined tissues. OTC maximum concentrations were 33.54 ± 11.19, 194.37 ± 16.11, and 18.79 ± 5.87 µg g− 1 for muscle, hepatopancreas and hemolymph, respectively. Although the highest OTC level was found in the hepatopancreas, it required only two days after the start of dosing to reach this value, whereas the maximum OTC for muscle and hemolymph was detected after eight days of dosing. Ten days after the cessation of medicated feeding, the drug content in the shrimp tail muscle was under the detectable limit for the method (0.01 µg g− 1 of OTC).  相似文献

In three separate experiments, harpaticoid copepods Tisbe monozota (alive and dead) and a microparticulate microbound diet were evaluated as alternatives to live Artemia nauplii as food, beginning at either stage PZ2 or M1, in the larval culture of Litopenaeus vannamei. Larvae were cultured in 2 L round bottom flasks at a density of 150 L− 1 (Experiment 1) and 100 L− 1 ( 3.2 and 3.3) at 28 °C, 35‰ salinity and 12:12 LD photoperiod, and fed 4×/day- 1. Larvae were initially fed a mixture of phytoplankton to stages PZ2 or M1 and then fed either live Artemia, live or dead copepods, or a microparticulate microbound diet. The experiments were terminated and all larvae were harvested when more than 80% of larvae had molted to postlarvae 1 (PL1) within any flask representing any of the treatments. The comparative value of the different diets and feeding regimes was determined by mean survival, mean dry weight and total length of individual larva, and percentage of surviving larvae that were PL1. Trypsin activity of samples of larvae from each treatment was also determined. The microparticulate microbound diet effectively served as a complete substitute for Artemia nauplii when fed beginning at stage M1. When fed at the beginning of the PZ2 stage, survival was comparable to that of larvae fed Artemia, but mean dry weight, mean total length, and percent of surviving larvae that were PL1 generally were significantly less. Responses to the feeding of copepods, whether fed dead or live, as a substitute were generally significantly less than those of larvae fed either the Artemia nauplii or the microparticulate diet. Values of trypsin activity (10− 5 IU/μg- 1 dry weight) corresponded to the relative proportions of the different larval stages within a treatment, with higher activity being characteristic of early stages. Previously demonstrated successful results with another species of crustacean suggest that the microparticulate microbound diet has characteristics that should be effective in the culture of the carnivorous stages of other crustacean and fish larvae that are currently fed live Artemia nauplii.  相似文献

White spot syndrome virus has been a threat to the global shrimp industry since it was discovered in Taiwan in 1992. Thus, shrimp-producing countries have launched regulations to prevent import of WSSV-infected commodity shrimp from endemic areas. Recently, cooked shrimp that is infected with WSSV tested positive by PCR. However, there is no study to determine the infectivity of WSSV in cooked shrimp that tested positive by PCR. In the present study, WSSV-infected shrimp were cooked at boiling temperature for different times including 0, 1, 3, 5, 10 and 30 min. Upon exposure to boiling temperature, WSSV-infected shrimp were fed to SPF shrimp (Litopenaeus vannamei). The result showed experimentally challenged shrimp from 0-min treatment (positive control) indeed got infected with WSSV. However, experimentally challenged shrimp that were fed tissues boiled at 1, 3, 5, 10 and 30 min were not infected with WSSV. Mortality data showed that only the positive control (0-min) treatment displayed high mortality, whereas no mortality was observed in any other treatment category. These findings suggest that cooking shrimp at boiling temperature for at least 1 min might prevent any potential spread of WSSV from endemic countries to other geographical areas where WSSV has not yet been reported.  相似文献

In mass culture of Pacific bluefin tuna Thunnus orientalis, a marked growth variation is observed after they start feeding at 6–7 mm in body length (BL) on yolk‐sac larvae of other species, and the growth variation in tuna larvae is a factor leading to the prevalence of cannibalism. To examine the relationship between prey utilization and growth variation, nitrogen stable isotope ratios (δ15N) of individual larvae were analysed. A prey switch experiment was conducted under two different feeding regimes: a group fed rotifers (rotifer fed group), and a group fed yolk‐sac larvae of spangled emperor, Lethrinus nebulosus (fish fed group) from 15 days after hatching (6.87 mm BL). The fish fed group showed significantly higher growth than the rotifer fed group. Changes in the δ15N of the fish fed group were expressed as an exponential model and showed different patterns from those of the rotifer fed group. The δ15N of fast‐growing tuna larvae collected in an actual mass culture tank after the feeding of yolk‐sac larvae was significantly higher than those of the slow‐growing larvae, indicating that slow glowing larvae depended largely on rotifers rather than the yolk‐sac larvae.  相似文献

The effects of the dietary medicinal herbs Massa medicata, Crataegi fructus (Cf), Artemisia capillaries, Cnidium officinale and their mixture (HM), on growth and resistance against Vibrio anguillarum were examined using larval red sea bream, Pagrus major. The methanol extracts of Cf and HM inhibited the proliferation of seven infectious bacterial strains including the genera Aeromonas, Edwasiella, Photobavterium, Pseudomonas and Vibrio. Rotifers enriched with emulsified bonito oil containing extracts of Cf and HM lowered the bacterial counts in their body on TCBS medium for the genus Vibrio in comparison with rotifers enriched by emulsified bonito oil alone. Although statistically not significant, larvae fed rotifer fortified with Cf from 3 to 20 days after hatching showed the best growth and final body weight followed by those given rotifer with HM and control enrichment. However, final body length of larvae fed rotifer with Cf was significantly larger than larvae fed on rotifers enriched with other herb extracts. After a challenge test with V. anguillarum, survival of larvae fed rotifer with Cf and HM was significantly higher than for the control treatment. These results revealed that Cf and HM in rotifer is useful to promote growth and resistance against V. anguillarum in red sea bream larvae, providing a new technology for mass production of disease‐resistant fry and fingerlings.  相似文献

Systemic granulomatosis is the most frequent disease in juvenile and adult meagre, but studies regarding the first appearance of granulomas in larvae do not exist. In order to evaluate this, meagre larvae were fed four different feeding regimes as follows: RS and RO (rotifer enriched with Easy DHA Selco or Ori‐Green from 3 to 30 dph respectively), RAS and RAO (rotifer enriched with Easy DHA Selco or Ori‐Green from 3 to 21 dph and Artemia enriched with Easy DHA Selco or Ori‐Green from 12 to 30 dph respectively). All treatments were also fed with commercial microdiet from 20 to 30 dph. At 30 dph weight, length, specific growth rate and survival were significantly higher in Artemia‐fed larvae, regardless of the enrichment. Microscopic first appearance of granulomas was observed in 20 dph larvae fed RS and RO. At 30 dph granulomas and thiobarbituric acid reactive substances (TBARS), values were significantly higher in RS and RO‐fed larvae than in RAS and RAO‐fed larvae. The results showed that granulomas first appeared in meagre larvae at 20 dph when fed rotifers only. Conversely, a reduced appearance of granulomas and lipid peroxidation occurs when Artemia is included in the feeding sequence reinforcing the hypothesis of a nutritional origin of the systemic granulomatosis.  相似文献

Taurine is an essential or conditionally essential nutrient for many species of marine fish, especially during early development. There is growing evidence that marine fish larvae benefit from taurine‐enriched rotifers; however, it is unknown if larvae benefit from taurine‐enriched Artemia. We investigated the effects of taurine‐enriched rotifers (Brachionus plicatilis) and Artemia franciscana on the growth and whole‐body taurine concentrations of California yellowtail (Seriola lalandi; CYT) larvae. The approach used in this study was to encapsulate taurine within microparticles (liposomes), which were then fed to rotifers and Artemia. We found that feeding taurine liposomes to rotifers and Artemia resulted in taurine concentrations in these prey species that were similar to or above those previously reported in copepods. At the end of the rotifer phase, CYT larvae fed taurine‐enriched rotifers showed increased growth (final dry weights; DW) and had higher whole body taurine concentrations when compared to larvae fed unenriched rotifers. At the end of the Artemia phase, CYT whole body taurine concentrations varied among dietary treatments. Larval lengths and DWs were not significantly different among treatments at the end of the Artemia phase, suggesting that the taurine concentrations of unenriched Artemia were sufficient to support the growth of CYT larvae.  相似文献

The effect of algae with different DHA contents used for the enrichment of rotifers on the growth performance, survival, and swim bladder inflation of larval amberjack Seriola dumerili was investigated. Rotifers were enriched with freshwater Chlorella vulgaris containing three levels of DHA (rotifer containing DHA 0.04, 0.60, 1.32?g DHA 100?g?1 DM) and Nannochloropsis (0.04?g DHA 100?g?1 DM; 2.54?g EPA 100?g?1DM). The larvae were fed the enriched rotifers in triplicate from 3?days post-hatch for 7?days in static condition. The same algae used for rotifer enrichment were supplied to the larval tanks. Growth and survival rate of fish fed the rotifers enriched with Nannochloropsis were higher than those of fish fed the rotifers enriched with all three Chlorella treatments. Swim bladder inflation was lowest in fish fed the rotifers enriched with Nannochloropsis. The proportion of EPA was higher in fish fed the rotifers enriched with Nannochloropsis, while that of DHA increased proportionally with the DHA levels in the rotifers enriched with Chlorella. These results suggest that rotifers enriched with Nannochloropsis (EPA-rich rotifers) are effective to enhance growth and survival, but DHA instead of EPA is essential to improve the swim bladder inflation in amberjack larvae.  相似文献

At present, clownfishes are the best example of successfully captive bred ornamental specimens but little is known about the relationship between food enrichment and both larval growth and development. In fact, it is well known that a certain percentage of these fishes cultured in captive conditions show a miss-band pigmentation. In the present study, the effects of live prey enrichment on growth and pigmentation in false percula clownfish (Amphiprion ocellaris) larvae were tested.Newly hatched A. ocellaris larvae were divided in three different groups and fed as follows: group A fed on enriched (Algamac 2000) B. plicatilis (10 ind./mL) from day 1 to day 5 post-hatch (ph); group B fed on enriched (Algamac 2000) B. plicatilis (10 ind./mL) followed by Artemia nauplii (5 ind./mL;) and group C fed on Algamac 2000 enriched B. plicatilis and Algamac 2000 enriched Artemia nauplii.Samples of the larvae were collected on day 5 from group A and on day 11 ph in group B and C for morphometric and molecular analysis. On day 11 ph food enrichment resulted in a better growth of group C larvae respect to those of group B fed on not enriched Artemia nauplii (15.8 ± 0.2 mg and 8.78 ± 0.02 mm vs. 6.8 ± 0.2 mg and 6.93 ± 0.01 mm). Moreover, 36 ± 2% of the juveniles obtained from group B showed a miss-band pigmentation as compared to 29 ± 1% of the juveniles obtained from group C. At molecular level, the results obtained by Real-Time PCR are in agreement with the morphometric ones: a positive induction of the Insulin-like Growth Factor II (IGFII) and Peroxisome Proliferator Activated Receptor α and β (PPARα and PPARβ) gene expression and a reduction of Myostatin (MSTN) was observed in group C larvae fed on enriched live prey. IGFI gene expression was higher in group B.The present study provides clear evidences of the positive role of Algamac 2000 on growth and pigmentation of captive cultured false percula clownfish.  相似文献
