Two carbohydrases (cellulase, lysozyme), three proteases (trypsin, aminopeptidase and non-specific protease), a non-specific lipase, and semiquantitative tests of 19 digestive enzymes were assayed in different gut sections of juvenile red abalone, Haliotis rufescens, in order to identify the regions where digestion takes place and investigate the extent to which diet composition can modify the digestive capacity of abalone. The abalone were fed either fresh kelp (K) or balanced diets containing 25 or 38% crude protein for 6 months. Enzyme assays were carried out on different sections of the abalone's gut at the end of this period. On a weight-specific basis, the digestive gland was the site containing most of the enzymes. On a protein-specific basis, two main digestion regions were identified: the digestive gland-stomach region that is characterized by high activities of cellulase and lysozyme, chymotrypsin and protease, and the mouth-intestine region with a typically high activity of lipase and amino peptidase. Significant dietary effects were observed on the activity of enzymes, especially in the digestive gland. Abalone fed with 25 and 38% crude protein diets exhibited higher cellulase (39.8 ± 4.6 and 14.2 ± 0.8 mU mg− 1 protein, respectively) and lysozyme activities (88.0 ± 20.4 and 56.6 ± 15.7 U, respectively) than those fed with fresh kelp (5.5 ± 0.7 mU mg− 1 protein and 17.1 ± 1.8 U). In contrast, higher protease activity was found in kelp-fed organisms (234.1 ± 20.4 μg product/mg protein) than those fed the 25 and 38% crude protein diets (109.5 ± 20.7 and 119.5 ± 20.5 μg product/mg protein, respectively). Semiquantitative API ZYM assays resulted in no clear food-specific effects on the activity of carbohydrases, proteases, ester hydrolases or phosphohydrolases, yet organ-specific differences were conspicuous in various cases, and generally agreed with quantitative results. It is suggested that the increased carbohydrase activity exhibited by organisms fed the balanced diets resulted from a combination of an increased number of resident bacteria in the abalone's gut and facilitated contact between dietary substrates and digestive cells. The present results indicate that H. rufescens can adjust their enzyme levels in order to maximize the acquisition of dietary protein and carbohydrates. This characteristic can be advantageously used to search for suitable diets in abalone aquaculture.  相似文献

As has been demonstrated in previous studies, Octopus maya can be fed on artificial diets. In the present study six different diets were assayed. Five diets were designed to test the effect of percentage of inclusion of fish protein concentrate (CPSP: 0, 5, 10, 15, and 20%) and were offered to octopuses as a specifically designed artificial diet. The sixth diet consisted of frozen crab (Callinectes spp) and was used as control diet. Blood metabolites and energy budget of octopuses were evaluated to determine how CPSP levels modulate the digestive capacity and allow retaining energy for growth. Wild animals (316.4 ± 9.8 g) were used in the study. Results showed that CPSP produced a positive specific growth rate (SGR, % day− 1) with high value in octopuses fed 15% CPSP level. A maximum growth rate of 0.86% day− 1 was recorded in these animals, a value that is extremely low when compared with the SGR obtained when animals were fed fresh crab (3.7% day− 1). In general, blood metabolites were affected by diet composition, indicating that some metabolites could reflect the nutritional and/or physiological status of octopus. Preliminary reference values for O. maya fed crab were found for glucose (0.09 ±0.02 mg/ml), lactate (0.004 ± 0.002 mg/ml), cholesterol (0.16 ± 0.02 mg/ml), acylglycerol (0.14 ± 0.01 mg/ml), protein (0.37 ± 0.04 mg/ml), hemocyanin (1.85 ± 0.04 mmol/l), and digestive gland glycogen (1.86 ± 0.3 mg/g). Total energy content can be used as an indicator of tissue metabolic reserves. In the present study, higher energy content in the digestive gland and muscle was observed in octopuses fed crab, followed by animals fed 15% CPSP. Results from the digestive gland indicated that the retained energy derived from glycogen, suggesting that lipids and protein were the main sources of variation linked with energy content. In general, digestive gland proteases activity and trypsin were induced in octopuses fed 15% CPSP. The capacity of O. maya juveniles to adjust their digestive enzymes to different types of food was evidenced. Essential amino acid content (EAA) of the diet was not a limiting factor. When dietary EAA profiles were compared with O. maya EAA profiles, all dietary EAA resulted in a higher concentration than whole body octopus composition. In the present study, all experimental groups ingested between 3300 and 4106 kJ wk− 1 kg− 1 without statistical differences among treatments, indicating that experimental diets were as attractive as crab. Differences were recorded in the proportion of absorbed energy (Ab, %) between CPSP-based and crab meat diets, suggesting digestion limitations associated with artificial diets. The present results indicate that the 15% CPSP diet had characteristics that stimulate digestive enzymes and reduce energetic costs associated with its digestion (HiE or SDA), channeling more biomass production than the other experimental diets.  相似文献

Duplicate groups of Atlantic salmon (Salmo salar), kept in saltwater, were fed fish meal based cold-pelleted diets where graded levels of native or extruded non-starch polysaccharides (NSP) from soybeans replaced cellulose, at a total NSP level of 100 g kg− 1 for 28 days. The study also included a diet where defatted soybean meal (SBM) constituted the NSP at a level of 100 g kg− 1 and a reference diet without NSP supplementation.The SBM diet resulted in a significant reduction in faecal dry matter content, apparent digestibilities of starch and organic material, and growth, and an increased faecal output of several elements (K, Na, Zn), compared to all the other diets. Morphological changes were only seen in the intestine of the fish fed the SBM, i.e. the diets holding purified soy-NSP did not induce enteritis. When compared to the diet without NSP, cellulose addition increased faecal dry matter, while inclusion of native soy-NSP reduced it. Dry matter in faeces and apparent digestibility of crude protein decreased in a linear manner, while the digestibility of starch and faecal output of K and Na increased linearly when native soy-NSP replaced graded levels of cellulose from 0 to 100 g kg− 1 feed. When diets with 75 or 100 g kg− 1 of native NSP and extruded soy-NSP were compared, fish fed native soy-NSP had reduced faecal dry matter, higher digestibility of starch, and increased faecal output of Cu, Fe, and K. Dry matter in faeces and faecal output of Cu was lower for the highest inclusion level, while digestibility of starch and faecal output of Mn and K were higher.In conclusion, soy-NSP was inert compared to the fish meal reference, with respect to nutrient digestibilities and intestinal pathologies, but affected faecal mineral excretion in Atlantic salmon.  相似文献

The spider crab Maja brachydactyla is overexploited on the NW coast of Spain. Aquaculture of this species can be the solution to the problem, and consequently, several attempts of intensive larval rearing have been conducted. However, most of the studies already published do not provide enough zoo technical data, especially in terms of larval and prey densities or the nutritional quality of diets used for rearing.Three experiments were carried out to evaluate the conditions for intensive larval rearing of M. brachydactyla. Larval stocking density (10, 50 and 100 larvae L− 1), prey:larva ratio (15, 30 and 60 enriched Artemia larva− 1) and diet (enriched Artemia, non-enriched Artemia and polychaete supplement) effects on growth and survival of this species were studied. For larval culture nine, 35 L, 150 μm mesh-bottomed PVC cylinders (triplicates for each treatment and larval stage) connected to a recirculation unit, were used. Temperature and salinity were kept constant at 18 °C and 36‰ respectively. A 12 to 18 day trial was conducted for each experiment and samples of larvae were collected at each larval stage (zoea I, zoea II, megalopa) in the inter-molt phase and at first juvenile. Survival, carapace length and width, dry weight (DW), and proximate biochemical content (protein, carbohydrates and total lipid) as well as lipid class composition were determined.Stocking densities of 100 larvae L− 1 resulted in higher growth in DW and higher content in lipids and protein for zoea I (ZI) and zoea II (ZII) than 10 larvae L− 1. However, survival decreased with increasing stocking density.The use of 60 preys larva− 1 produced larvae with significantly higher DW and protein content, especially at ZII stage, than lower prey densities. Survival rate obtained feeding 60 preys larva− 1 up to the megalopa stage was almost two-fold (42.2%) the rate obtained using 15 preys larva− 1 (24.8%).Larvae fed on enriched Artemia (EA) showed an increase in weight up to megalopa (518.9 ± 26.5 μg) in contrast to larvae fed on non-enriched prey (A) (467.9 ± 6.9 μg). Variation in DW correlated with the total lipid content (L) of the larvae (LEA = 70.1 ± 37.5 μg ind− 1; L= 28.9 ± 3.2 μg ind− 1) especially in terms of neutral lipids. The use of an initial density of at least 50 larvae L− 1 and 60 enriched Artemia larva− 1 can be considered the most adequate rearing parameters in order to obtain good results in growth and survival of M. brachydactyla.  相似文献

Different assays related to the first feeding of Octopus vulgaris Cuvier, 1797 are compiled in this paper. They include: age at initial feeding age, prey size selection and optimal density, attack timing after feeding, and effect of dose number on the number of captures. Prey capture and ingestion processes were also analysed. Food supplied was cultured Artemia sp. Each assay lasted 15 min.Although paralarvae already start to feed on the hatching day (day 0), it is during day 2 when a greater number of attacks is recorded (81.7 ± 14.7% paralarvae attack). They mainly prefer (significance level α = 0.05) large Artemia, 1.4 ± 0.4 mm (77.0 ± 5.6% of the total attacks) than small Artemia, 0.8 ± 0.1 mm (23.0 ± 5.6%). There is also a slight predilection for the lowest Artemia concentration (33.3 ±12.6% paralarvae attack in a 0.1 Artemia ml− 1 density, opposite 16.7 ± 7.6 and 18.3 ± 7.6% in densities of 0.5 and 1 Artemia ml− 1 respectively). The greatest predatory activity is recorded during the first 5 min after food is supplied (72.2 ± 25.5%). An increase in the predatory activity was also observed when food was distributed in several doses instead of a single dose (75.0 ± 10.0% and 46.7 ± 17.6% respectively). It was proved for the first time that paralarvae completely ingest their preys (including their exoskeletons), in this case Artemia. Time needed for their total ingestion ranges between 4 and 10 min.  相似文献

As natural diets of fish larvae, a number of calanoid copepod species are being investigated for use as live prey in aquaculture hatcheries. One of these, the tropical calanoid copepod, Acartia sinjiensis, has good potential as a live feed for tropical reef fish larvae. However, the rearing techniques for A. sinjiensis require further development to improve productivity. This study was carried out to investigate the population growth and egg hatching success of A. sinjiensis when fed a range of mono-and binary algal diets, including algae in the form of frozen paste.For the population growth experiment, the final A. sinjiensis population, including eggs, nauplii, copepodites and adults, was determined after feeding eight algal diets (two frozen algae, four live monoalgal and two live binary algal diets) for 8 days at temperature 28 ± 1 °C; salinity 34 ± 1 psu and photoperiod 12 L:12 D. Five replicates, with an initial 12 adult A. sinjiensis per replicate, were set up for each treatment. In a separate experiment, effects of diets on egg hatching success were examined after 48 h incubation of eggs produced by A. sinjiensis fed the same eight diets.The results showed that diet significantly affected both population growth and hatching success of A. sinjiensis. Of the diets tested, the binary algal diets were more successful than monoalgal diets, while the frozen algae had little dietary value. The highest population growth was recorded on A. sinjiensis fed a binary diet of Tetraselmis chuii and the Tahitian strain of Isochrysis sp. (T-ISO) (final population: 1091 ± 80), which was significantly higher (P < 0.001) than all other diets tested except for the other binary diet of Nannochloropsis sp. and T-ISO (final population: 897 ± 123). Diet also had a significant effect (P < 0.001) on egg hatching rate, though the highest hatch rate was recorded with eggs produced by A. sinjiensis fed binary diet Nannochloropsis sp. and T-ISO (88.1 ± 2.1%), this was not significantly different from that of eggs produced by A. sinjiensis fed either T-ISO alone (88.0 ± 1.7%) or the binary algal diet of T. chuii and T-ISO (76.4 ± 7.1%). The results of this study suggest that among the diets tested, a combination of live T. chuii and T-ISO was the best for the culture of A. sinjiensis.  相似文献

The effect of replacing fish meal with soybean meal (SBM) in practical feeds for cuneate drum was evaluated in an 8-week net pen trial. The cuneate drum fingerlings (initial body weight 29.8 ± 1.3 g fish− 1) were fed six isonitrogenous and isocaloric feeds containing 39% digestible protein and 16 MJ kg− 1 digestible energy. The control feed was formulated to contain 40% herring meal, whereas in the other five feeds SBM was included at 11.3, 22.5, 33.8, 45.0 and 56.3% to replace 20, 40, 60, 80 or 100% of the fish meal. There were no significant differences in feed intake between fish fed the control feed and feeds in which SBM replaced 20 to 80% of the fish meal, but fish fed the fish meal free feed had higher feed intake than the other treatments. Weight gain linearly declined with the decrease of fish meal level. Final body weight (FBW) of fish fed the feeds in which SBM replaced 20% of the fish meal did not significantly differ from fish fed the control feed. Replacing 40 to 100% of the fish meal resulted in lower FBW and nitrogen retention efficiency (NRE), and higher feed conversion ratio (FCR) than those of fish fed the control feed. Fish fed the feeds in which SBM replaced 60 to 100% of the fish meal had lower condition factor and hepatosomatic index than those of fish fed the control feed. No significant differences in carcass protein content was found among the treatments, but fish fed the feeds in which SBM replaced 60 to 100% of the fish meal had higher moisture and lower lipid content in carcass than those of fish fed the control feed. Results of the present study appear to indicate that cuneate drum has a limited ability to utilize SBM as a protein source in practical feeds.  相似文献

Hatchery broodstock conditioning and nursery culture of the Chilean flat oyster Ostrea chilensis have been hampered by the poor performance of oysters fed typical microalgal hatchery diets. To determine the feeding capabilities of this species the selective removal and consumption of natural planktonic assemblages and artificial inert particles (polystyrene beads) by juvenile and adult oysters were examined experimentally. The arrangement of the eulaterofrontal cirri of the ctenidia was also examined to infer their potential efficiency of particle selection for feeding. Polystyrene beads of 45 and 15 μm in diameter had high rates of removal from suspensions by both juvenile (45 μm = 70%, 15 μm = 73%) and adult (45 μm = 88%, 15 μm = 87%) oysters. In contrast, beads of 6 μm diameter had lower rates of removal (adults = 68%, juveniles = 53%), while 1 μm beads were not removed at all. Both adult and juvenile oysters feeding upon natural plankton assemblages removed only microphytoplankton (20-75 μm in size) despite the presence of nanophytoplankton (2-20 μm), picophytoplankton (< 2 μm), cyanobacterium Synechoccocus spp. (< 2 μm), and bacterial cells (< 75 μm) in the experimental suspensions. Eulaterofrontal cirri of both juvenile (15.2 μm ± 0.9 SE) and adult oysters (18.9 μm ± 0.3 SE) are the shortest that have been reported for any ostreid species which helps to explain the inability of this species to retain small food particles. The clearance rates for oysters feeding on microphytoplankton only were 1.49 (± 0.05 SE) and 7.1 (± 1.2 SE) l h− 1g− 1 for juveniles and adults respectively. These values are much higher than previously reported for this species being fed smaller sized cultured microalgae. Our results strongly suggest that the difficulties in the nursery and broodstock hatchery culture of this oyster may be due to inappropriate phytoplankton diets. We recommend the provision of cultured microalgae of 20-75 μm in diameter for improving the performance of hatchery maintained juvenile and adult O. chilensis.  相似文献

Three potentially valuable red seaweeds, Chondrus crispus Stackhouse, Gracilaria bursa pastoris (S.G. Gmelin) P.C. Silva and Palmaria palmata (L.) O. Kuntze, collected in northern Portugal, were cultivated using the nutrient-rich effluents from a local turbot (Scophthalmus maximus Linnaeus) and sea bass (Dicentrarchus labrax Linnaeus) farm. The algae were cultivated in a two level cascade system. Several arrangements of the cascade system, stocking densities (3, 5, 7 and 8 kg m− 2) and water fluxes (140 and 325 l h− 1) were tested to optimize biomass yield and nitrogen uptake rate and efficiency. The yield and the total ammonium nitrogen (TAN) uptake of the three species were highly seasonal. Palmaria could not survive culture conditions during the summer when water temperature was above 21 °C. In the spring, Palmaria had an average yield of 40.2 (± 12.80) g DW m− 2 day− 1 and a nitrogen uptake efficiency (NUE) of 41.0% (± 17.26%). NUE expresses, in percentage, the average reduction in TAN concentration between the inflows and the outflows of the tanks. Chondrus performed better in summer with an average yield of 37.0 (± 11.10) g DW m− 2 day− 1 and removing 41.3% (± 17.32%) of nitrogen. Gracilaria grew year round, but also performed better during spring/summer, producing an average of 29.1 (± 2.90) g DW m− 2 day− 1, and only 7.3 (± 5.08) g DW m− 2 day− 1 during autumn. Yield of C. crispus did not differ significantly when grown at two different stocking densities (5 kg m− 2 and 8 kg m− 2). On the other hand, Gracilaria had significantly higher yields at 5 than at 7 kg m− 2. Better NUE, on average 76.7% (± 22.13%), was also obtained with 5 kg m− 2 stocking density and only 63.8% (± 24.62%) with 7 kg m− 2. The yield of Gracilaria increased significantly with the increase of water flux from 140 to 325 l h− 1 and more nitrogen was removed from the water. However, NUE decreased from 48.4% to 33.4% at 140 and 325 l h− 1, respectively. Biofiltration was highly improved by a cascade system with a NUE as high as 83.5%.  相似文献

The current study investigated acute toxicity to ammonia of the South African abalone, Haliotis midae, from three size classes relevant to mariculture operations, and the chronic impact of sub-lethal ammonia levels on growth of juvenile abalone.Results showed that tolerance to ammonia (at pH 7.8 and Ta = 15 °C) increases with body size (i.e. age) as indicated by 36 h LC50 values: juvenile abalone (1-2.5 cm shell length) had the lowest LC50 of 9.8 μg l− 1 FAN, whereas LC50 was 12.9 μg l− 1 FAN in “cocktail”-size abalone (5-8 cm shell length). The highest LC50 of 16.4 μg l− 1 FAN was observed in “brood stock”-size animals (10-15 cm). When “cocktail”-size abalone were allowed to acclimatize to sub-lethal ammonia levels for 48 h, their ammonia tolerance increased compared with non-acclimatized abalone of the same size: LC50 was 2.0 μg l− 1 FAN higher at 14.8 μg l− 1 FAN.Growth of juvenile abalone (1-2.5 cm shell length) during chronic exposure to sub-lethal FAN levels is inhibited: specific growth rate (SGR) is significantly reduced by 58.7% to 0.10 ± 0.03% d− 1 (weight) compared with 0.24 ± 0.06% d− 1 of abalone of a control group (no ammonia).The results demonstrate the negative effects of ammonia not only on survival but also on growth of farmed abalone, both impair profitability of the farming operation. The information from the present study will be helpful in determining water quality requirements in South African abalone farms.  相似文献

An 8 weeks growth experiment was conducted to determine the effects of dietary vitamin C on the survival, growth, tissue ascorbic acid contents and immunity of large yellow croaker (Pseudosciaena crocea) with initial weight of 17.82 ± 0.68 g. Seven practical diets were formulated to contain 0.1, 12.2, 23.8, 47.6, 89.7, 188.5 and 489.0 mg ascorbic acid equivalent kg− 1 diet, supplied as l-ascorbyl-2-polyphosphate (LAPP). Each diet was fed to triplicate groups of fish in seawater floating cages (1.5 × 1.5 × 2.0 m), and each cage was stocked with 120 fish. Fish were fed twice daily (05:00 and 17:00) to apparent satiation for 8 weeks. The water temperature fluctuated from 19.5 to 25.5 °C, the salinity from 25 to 28‰ and dissolved oxygen content was approximately 7 mg l− 1 during the experimental period. Results showed that the specific growth rate (SGR) (from 1.80 to 1.96% d− 1) had an increasing trend with the increase of dietary vitamin C, but no significant difference was observed among dietary treatments. No gross deficiency signs were observed in any of the experimental fish. Survival rate, however, significantly increased with increasing dietary vitamin C (P < 0.05). The vitamin C contents in liver and muscle correlated positively with the vitamin C in diets. The vitamin C requirement was estimated to be 28.2 mg kg− 1 based on survival, and 87.0 mg kg− 1 on liver content of vitamin C. The activities of serum lysozyme and alternative complement pathway (ACP), phagocytosis percentage (PP) and respiratory burst activity of head kidney significantly increased with increasing dietary vitamin C. The challenge experiment with Vibrio harveyi showed that fish fed the diets with supplementation of vitamin C had significantly lower cumulative mortality compared to the control group (66.7%), and the cumulative mortality (16.7%) in fish with 489.0 mg kg− 1 ascorbic acid was significantly lower than that (41.7%) in fish with 23.8 mg kg− 1 ascorbic acid. These results suggested that vitamin C significantly influenced the immune response and disease resistance of large yellow croaker.  相似文献

Stimulation of the non-specific defence enhances the disease resistance and growth, and has good potentials as a measure for increased microbial control in juvenile production of marine fish and shellfish. So far, the most commonly used immunostimulants are β-(1 → 3, 1 → 6)-glucans, and in this study the stimulatory potential of a β-(1 → 3, 1 → 6)-glucan of marine origin, the storage polysaccharide from the marine diatom Chaetoceros mülleri, was examined. The glucan (chrysolaminaran) was extracted from cultures of C. mülleri, and used as a dietary supplement in two first feeding experiments with larvae of Atlantic cod Gadus morhua L. In one experiment the microalgal glucan was compared to the commercial yeast-glucan product MacroGard®, and in the other to an alginate with a high content of mannuronic acid (High-M alginate) isolated from Durvillaea antarctica. The stimulants were given via rotifers, and weaning to formulated feed was initiated at day 17 or 18 after hatching. The survival ± SEM at day 27 after hatching was 24.5 ± 2.0%, 14.8 ± 4.5% and 13.1 ± 1.4% for the groups fed C. mülleri-glucan, yeast glucan and for the control, respectively, in the first experiment. The group fed C. mülleri-glucan group had higher survival compared to the control (< 0.05) group, whereas the yeast glucan had no positive effect on the survival (p > 0.05). The dry weights of the groups at day 27 were low, with 203.2 ± 52.2, 165.2 ± 43.4 and 198.5 ± 58.1 μg per larva for the C. mülleri-glucan, yeast glucan and control groups, respectively. In the second experiment the survival in the period of feeding formulated feed (days 18-30) were 44.6 ± 4.3%, 44.7 ± 1.3%, and 33.8 ± 4.1% survival for the C. mülleri-glucan, High-M alginate and control group, respectively. The cod larvae fed C. mülleri-glucan reached an average weight of 531.6 ± 17.2 μg at day 30, which was significantly higher (p < 0.05) than the control group that had an average of 473.6 ± 3.5 μg. The larvae fed High-M alginate had an average weight of 470.3 ± 31.6 μg per larva at day 30, and not significantly different from the control (p > 0.05). The early weaning to formulated diet had detrimental effect on the growth of the larvae. In both experiments the C. mülleri-glucan group was the only group showing a positive growth rate in the period of weaning to dry feed. The microbial conditions in larval gut and water were monitored with respect to total colony forming units on Marine agar, and Vibrio- and Pseudomonas-like species on selective agars (TCBS and marine Pseudomonas Agar with CFC-supplement). The larvae were rapidly colonised after hatching, but no or weak effects of the stimulants were observed on the colonisation rates or the composition. The total CFU varied from 101 to 102 CFU per μg larva after initiation of the first feeding. The percentages of Pseudomonas-like bacteria increased throughout the period, whereas the levels of Vibrio-like bacteria were low and stable. The chrysolaminaran from the diatom C. mülleri was shown to be a promising candidate for use as an immunostimulatory feed additive, and which should be further explored.  相似文献

Sea urchin fisheries are valuable commercial resources in the United States with processed gonads sold in Japanese and American markets and maximum US sales of $150M US dollars in 1996. Wild populations of sea urchins on all coasts of the US have been dramatically over-fished. Aquaculture of sea urchins in land-based facilities can help restore commercial populations and preserve this ecologically important herbivore. In this study, we used invariant summer photoperiod to prevent gametogenesis in the North American green sea urchin (Strongylocentrotus droebachiensis) maintained in a land-based aquaculture system and provided a commercially available formulated feed that promotes maximum growth of intra-gonadal somatic nutrient storage cells called nutritive phagocytes. Results were compared with individuals fed the same formulated feed under ambient photoperiod in cages in the ocean. Monthly samples of gonads from both treatments were evaluated for gonad index, volume fractions of cellular constituents of the germinal epithelium, oocyte diameters and taste. Over the 5 months of this study, gonad indices increased significantly (p < 0.001) in both treatments from 4.8% ± 0.9 (all values ± SE) initially to 20.5% ± 2.1 under invariant and 23.2% ± 1.4 under ambient photoperiod with no significant difference between treatments (p = 0.55). Volume fractions of nutritive phagocytes increased to 80.3% ± 5.9 (initial 37.9% ± 7.1) in males and 71.0% ± 6.7 (initial 10.3% ± 4.0) in females (p < 0.001) only under invariant photoperiod. Nutritive phagocyte lengths increased under both photoperiod treatments, but the volume fraction containing nutrients was higher under invariant photoperiod. Volume fractions of gonial/gametogenic cells increased significantly (p < 0.001) only under ambient photoperiod from 20.4% ± 5.5 to 37.8% ± 1.8 in males and 0% to 22.6% ± 3.6 in females. The volume fraction of residual oocytes from last year's oogenesis increased under invariant photoperiod while that of both residual and new oocytes increased under ambient photoperiod. Residual oocyte diameters increased from 56.2 μm ± 2.2 initially to 93.5 μm ± 3.7 under invariant and those of residual and new oocytes to 126.0 μm ± 7.3 under ambient photoperiod. Invariant photoperiod yields gonads in both sexes of S. droebachiensis that do not initiate fall gametogenesis but attain large size as their nutritive phagocytes grow substantially in size. A Canadian study of wild-collected S. droebachiensis indicated that gonads taste best when they contain pre-dominantly nutritive phagocytes and not copious gametes, however gonad taste in our study was unsatisfactory suggesting that the only commercially available sea urchin diet requires modification to support commercial development of land-based aquaculture.  相似文献

The white shrimp, Litopenaeus vannamei, has become a very important species for the development of shrimp aquaculture in Northwest Mexico. However, viral and bacterial diseases are considered a major threat to the development of this industry. In the present study a trial was conducted to evaluate the tissue distribution, maximum concentration, and elimination of the widely used antibiotic oxytetracycline (OTC) in L. vannamei using indoor tanks under laboratory-controlled conditions. OTC was given to shrimp simulating a therapeutic treatment through medicated feed for 14 days followed by a period of feeding without antibiotic for another 14 days to evaluate the elimination pattern. Samples of hemolymph, muscle, and hepatopancreas were taken from medicated animals every two days for 28 days. All tissues were removed and frozen immediately in liquid nitrogen. OTC levels were analyzed by High Performance Liquid Chromatography (HPLC). Results showed an important OTC increase during consumption of medicated feed in all examined tissues. OTC maximum concentrations were 33.54 ± 11.19, 194.37 ± 16.11, and 18.79 ± 5.87 µg g− 1 for muscle, hepatopancreas and hemolymph, respectively. Although the highest OTC level was found in the hepatopancreas, it required only two days after the start of dosing to reach this value, whereas the maximum OTC for muscle and hemolymph was detected after eight days of dosing. Ten days after the cessation of medicated feeding, the drug content in the shrimp tail muscle was under the detectable limit for the method (0.01 µg g− 1 of OTC).  相似文献

A major problem in the development of Senegalese sole Solea senegalensis intensive culture is the poor control on reproduction, in part due to the lack of knowledge on the precise role of photoperiod and temperature. Thus, gonadal maturation was evaluated by assessing the sequential changes in plasma levels of 17β-estradiol (E2), 11-ketotestosterone (11-KT), and testosterone (T) in both female and male cultured Senegalese sole (F1 generation) exposed to various combinations of constant or naturally-fluctuating daylength and water temperature. Under natural photoperiod (NP; 36° N), exposure to constant temperature (t0; 18-20 °C) disrupted gonadal development, as indicated by a lower incidence (in comparison with naturally-fluctuating water temperature; 14-24 °C) of females at advanced maturation (from February to April: 12 vs. 33%) and running males (from February to May: 46% vs. 57%), and the reduced mean (± S.E.M.) sex steroid plasma levels (female peak E2 levels: 2.9 ± 0.28 vs. 1.8 ± 0.3 ng ml− 1; male peak T levels: 1.5 ± 0.14 vs. 0.9 ± 0.06 ng ml− 1). Therefore, the onset and progression of gonadal development in this species seem to be strongly (“proximally”) influenced by fluctuating water temperature. When compared to NP and t0, exposure to continuous light (LL) under t0 significantly reduced steroid production (female peak E2 levels: 1.8 ± 0.28 vs. 0.5 ± 0.05 ng ml− 1; male peak 11-KT levels: 9.4 ± 1.06 vs. 5.4 ± 1.33 ng ml− 1) and subsequently gonadal development (lower proportions of females at intermediate [46 vs. 6%] and advanced maturation [12 vs. 0%] from February to April and of RM [46 vs. 33%] from February to May). Thus, the seasonal changes of daylength would be crucial for normal gonadal development, being its cueing effects of higher magnitude than those of water temperature. The present report constitutes the first systematic study focused on the environmental control of reproductive events in Senegalese sole.  相似文献

Periphyton grown on substrates is known to improve water quality in aquaculture ponds. Five different substrates, (i) bamboo pipe (ii) plastic sheet (iii) polyvinylchloride (PVC) pipe (iv) fibrous scrubber, and, (v) ceramic tile were evaluated for the formation of biofilm in this experiment. The substrates were suspended 25 cm below the water surface. Each type of substrate was collected fortnightly to analyze the abundance and biomass of different periphytic algae and of the biofouling organism. The study was terminated after 60 days due to severe fouling by polychaete. Results showed that pond water nutrients were high on day 60 with mean total ammonia-N, nitrite-N and soluble reactive phosphorus concentrations of 309.6 ± 8.6 μg L− 1, 26.0 ± 2.7 μg L− 1 and 87.2 ± 7.1 μg L− 1 respectively. During the first two weeks the substrates were colonized by 19 periphytic algae. The most abundant family was Bacillariophyta (8 genera) followed by Chlorophyta (7 genera) and Cyanophyta (4 genera). Periphyton colonization on bamboo pipe showed the highest (p < 0.05) biomass in terms of chlorophyll a amongst all the substrates used. The biomass varied from 179 to 1137 μg m− 2 with mean values of 1137.2 ± 0.6, 929.6 ± 0.6, 684.2 ± 1.2, 179.1 ± 0.6 and 657.0 ± 0.6 μg m− 2 on bamboo pipe, PVC pipe, plastic sheet, fibrous scrubber and ceramic tile respectively for the first 15 days. From 3rd week, polychaetes began to form tubes on the substrate. By day 60, the whole surface of all substrates was covered with tightly packed polychaete tubes with mean densities of 168.0 ± 15.4, 121.0 ± 13.5, 72.8 ± 9.8, 72.4 ± 7.4 and 56.0 ± 6.8 polychaete tubes cm− 2 for bamboo, PVC, plastic, fibrous scrubber and ceramic tile respectively. This study illustrated the invasive nature of attached polychaete thus hampering the formation of periphyton biofilm on substrates which could have been used for improving water quality in enriched brackishwater shrimp ponds.  相似文献

African catfish (Clarias gariepinus) (initial body weight: 34.8 ± 4.8 g) and vundu catfish (Heterobranchus longifilis) (initial body weight: 39.1 ± 8.2 g) fingerlings were stocked at densities of 4, 6 or 8 fish m− 3 in traditional fish ponds (whedos) constructed in the floodplain of the Oueme River (South Benin, West Africa), for 70 days from March to June 2005. Fish were fed twice a day with 34% crude protein feed formulated with locally available ingredients. The effects of stocking density were evaluated in growth responses, gross production and body composition. Water quality variables were similar (p > 0.05) in all compartments. Temperature and pH were at the optimum level for fish. Dissolved oxygen ranged from 0.9 to 1.2 mg l− 1 during the experiment and secchi disc transparency was low (< 14 cm). In both species, growth responses increased with the increasing density, significantly in African catfish stocked at density of 8 fish m− 3 compared to the other densities (4 and 6 fish m− 3) but not significantly in vundu catfish. Production data ranged from 3.1 ± 0.5 to 22.8 ± 4.5 t ha− 1 year− 1 in African catfish and from 6.1 ± 1.2 to 15.1 ± 3.1 t ha− 1 year− 1 in vundu catfish. Production increased with increasing stocking densities but only significantly (p < 0.05) between the density of 8 fish m− 3 and the other densities. In both species, carcass fat increased with increasing density (p < 0.05) while carcass protein and moisture decreased (p > 0.05). These results are important because they indicate that, as far as growth rate and production are concerned, African catfish is more profitable than vundu catfish for culture at high density in whedo.  相似文献

Giant freshwater prawns, Macrobrachium rosenbergii (17.9 ± 2.7 g), exposed to different concentrations of saponin at 0, 0.3, 0.6, 0.9 and 1.2 mg l− 1 for 168 h were examined for osmolality, electrolyte levels, oxyhemocyanin, protein levels, acid-base balance status, total hemocyte count (THC), phenoloxidase activity, and respiratory bursts. Hemolymph oxyhemocyanin, protein, and pO2 were inversely related to the saponin concentration. Hemolymph oxyhemocyanin, protein, pO2, pCO2, and pH of prawns exposed to 1.2 mg l− 1 saponin were significantly lower than those of prawns exposed to 0.3 mg l− 1 and control solutions. However, no significant difference was observed in osmolality or electrolyte levels of prawns exposed to different concentrations of saponin for 168 h. The THC of prawns following 168 h of exposure to 0.9 and 1.2 mg l− 1 saponin increased, but the phenoloxidase activity decreased suggesting that the decrease in phenoloxidase activity under saponin stress was not a consequence of the increase in THC. We concluded that saponin at as low as 0.9 mg l− 1 decreases the respiratory protein level and acid-base balance, and modulates the immune system of M. rosenbergii.  相似文献

This experiment was done to study the effects of dietary soybean meal (SBM) and inulin (a prebiotic) on the capacity for digestive hydrolysis and amino acid absorption by Atlantic salmon, and how a dietary supplement of the broad-spectrum antibiotic oxytetracycline (OTC) modulated these responses. A control diet (FM) was made from fish meal, fish oil and extruded wheat. Two similar diets were made with 250 g soybean meal (SBM) or 75 g inulin kg− 1. Each diet was made with or without a supplement of 3 g OTC kg− 1. All six diets contained yttrium oxide for estimation of apparent nutrient absorption. Each diet was fed to two groups of 172 g salmon kept in 1 m2 tanks with 9 °C saltwater for 3 weeks. Intestinal organs were then sampled and weighed. Gastrointestinal tracts (GIT) were sectioned for analyses of brush border alkaline phosphatase (ALP) and leucine aminopeptidase (LAP) activities. Tissue from the distal intestine (DI) was also fixed for histological examination. Digesta from the different sections were freeze dried for estimation of trypsin and amylase activities, and of apparent absorption of amino acids (AA), nitrogen (N), and sulphur (S). About 85% of the trypsin activity, 70% of the amylase activity, 85% of the ALP activity, and 82% of the LAP activity were found in the proximal (PI) and mid (MI) intestine of fish with functional DI, and the absorption of AA, N, and S was quantitatively completed in the MI. Dietary OTC resulted in lower relative liver weight, but apart from increased ALP and LAP activities in DI when feeding OTC in combination with inulin, OTC did not modify the responses to dietary SBM or inulin. Dietary SBM resulted in lower relative liver weight, and induced pathomorphological changes in the DI mucosa, thus lower the ALP and LAP activities in the DI. SBM also stimulated absorption of AA, N, and S in the PI, but at the same time increased the activities of trypsin and amylase in the DI, indicating reduced re-absorption and increased faecal losses of these endogenous enzymes. Dietary inulin did not damage the DI, and stimulated intestinal growth and higher relative mass of the GIT. Inulin without OTC did not affect the hydrolytic and absorptive capacity of the salmon GIT.  相似文献

The phytoplanktivorous silver carp is an important biomanipulation fish to control cyanobacterial blooms and is also a food fish with the greatest production in China. The accumulation of the hepatotoxic microcystins (MCs) determined by LC-MS in various organs of silver carp was studied monthly in Lake Taihu dominated by toxic Microcystis aeruginosa. Average recoveries of spiked fish samples were 78% for MC-RR and 81% for MC-LR. The highest content of MCs was found in the intestine (97.48 μg g− 1 DW), followed by liver (6.84 μg g− 1 DW), kidney (4.88 μg g− 1 DW) and blood (1.54 μg g− 1 DW), and the annual mean MC content was in the order of intestine > liver > kidney > blood > muscle > spleen > gallbladder > gill. Silver carp could effectively ingest toxic Microcystis cells (up to 84.4% of total phytoplankton in gut contents), but showed fast growth (from 141 g to 1759 g in 1 year in mean weight). Silver carp accumulated less microcystins in liver than other animals in the same site or other fish from different water bodies at similar level of toxin ingestion. There was possible inhibition of the transportation of the most toxic MC-LR across the gutwall. Muscle of silver carp in Lake Taihu should not be consumed during period of dense Microcystis blooms while viscera were risky for consumption in more months.  相似文献
