Three hundred and three chicks of both sexes, from a synthetic dam line (SDL) of broiler chickens, were studied for economic traits (body weights at 4, 5 and 6 weeks of age) and immunological traits (humoral and cell mediated immune responses, and serum lysozyme concentration). The objective was to evaluate these traits and to estimate their genetic and non-genetic parameters. The humoral immune response was assessed by estimating the antibody response to sheep red blood cells using the haemagglutination (HA) test and serum IgG concentration using single radial immunodiffusion (SRID). The cell mediated immune (CMI) response was estimated as in vivo response to a mitogen (PHA-P). Serum lysozyme was measured by lysoplate assay. Least squares means for body weight at 4, 5 and 6 weeks were 684 +/- 20, 920 +/- 19 and 1205 +/- 28 g, HA titre was 6.289 +/- 0.246, CMI was 0.438 +/- 0.015 mm, lysozyme was 1.860 +/- 0.047 microg/ml and IgG was 6.287 +/- 0.194 mg/ml. There was an effect of sire on HA titre and on body weight at 4, 5 and 6 weeks of age; males were heavier than females. Heritability estimates were high for body weights but low for immunological traits. Phenotypic correlations (rp) among body weights were high and positive but were very low between body weights and most immunological traits. Among the immunological traits all rp were very low. Genetic correlations (rg) of body weights were positive and medium to high with CMI and HA and negative with serum IgG.  相似文献

Influenza and tetanus-specific antibodies of the IgG sub-isotypes are posively transferred to foals via colostrum and inhibit their response to inactivated influenza vaccines and tetanus toxoid. High titres of influenza antibodies of IgGa and IgGb subisotypes and tetanus antibodies of the IgGa, IgGb and IgG(T) subisotypes were detected in postsucking serum samples collected from foals born to mares that had received booster doses of multicomponent vaccines during the last 2 months of gestation. Thereafter, titres declined in an exponential manner but were still detectable in all foals at age 26 weeks, regardless of whether they had been vaccinated prior to age 26 weeks. Mean +/- s.e. half-life of decline of influenza IgGa antibodies (27.0 +/- 2.3 days) was significantly shorter than that of influenza IgGb antibodies (39.1 +/- 2.7 days; P<0.005). Tetanus IgGa and IgGb antibodies declined with half-lives of 28.8 +/- 3.0 and 34.8 +/- 5.1 days, respectively. Titres of tetanus IgG(T) antibodies were substantially higher than those of influenza IgG(T) antibodies in postsucking samples and remained so through age 26 weeks, declining with a half-life of approximately 35 days. Postsucking titres of tetanus and influenza antibodies of the IgA isotype were low and declined rapidly to undetectable levels. Yearlings showed significant increases in titre of influenza IgGa, IgGb and IgG(T) subisotype antibodies but no increase in influenza IgA antibodies in response to 2 doses of multicomponent vaccines containing tetanus toxoid and inactivated influenza A-1 and A-2 antigens. Yearlings also showed strong tetanus IgGa, IgGb and IgG(T) subisotype responses to one dose of vaccine and a substantial further rise in titre in response to administration of a second dose 3 weeks later, but failed to show an increase in titre of tetanus IgA antibodies. The influenza and tetanus IgGa, IgGb and IgG(T) subisotype responses of 6-month-old foals to vaccination followed the same pattern as those shown by yearlings but titres were generally lower. In contrast, 3-month-old foals failed to show increases in titre of either influenza or tetanus IgG subisotypes in response to 2 doses of vaccine and generally needed 1-3 additional booster doses of vaccine to achieve titres similar to those achieved by yearlings after 2 doses. Based on the finding that maternal antibodies exert a significant inhibitory effect on the response of foals to tetanus toxoid and inactivated influenza antigens, it is recommended that primary immunisation of foals born to vaccinated mares should not commence before age 6 months.  相似文献

Serum samples from 20 out of 180 (11.1%) apparently healthy Nigerian indigenous chickens were negative for antibodies against chicken anaemia virus using the enzyme-linked immunosorbent assay (ELISA). Of the 160 positive sera (88.9%), 12 (7.5%) had titres ranging from 1500-3000, 46 (28.8%) had titres from 3000-5000 while 102 (63.8%) had titres between 5000-11000. The overall mean titre value was 5845 +/- 2402. This appears to be evidence of a natural outbreak of the infection since the chickens had no history of vaccination against any poultry disease.  相似文献

1. A total of 433 birds (7 weeks old) of both sexes belonging to Indian native breeds, including, Aseel, Kadakanath, Naked Neck and Frizzle fowl along with the imported breeds Dahlem Red, White Leghorn, Synthetic dam line broiler (SDL) and Naked Neck broiler were utilised to test the primary antibody response to sheep erythrocytes by haemagglutination test. The effect of genotype (breed), sex and their interactions on antibody response were also studied. 2. The results revealed the presence of natural antibodies in all groups under study. 3. All groups except broilers showed the highest HA titre on day 5 post immunisation, which gradually declined until the end of the experiment (19th day post immunisation). In broilers, the peak HA titre was observed on day 12. 4. Dahlem Red showed the highest response throughout. The lowest antibody response was recorded for broilers except on day 19 post immunisation when it exceeded the White Leghorn value. 5. Amongst the native Indian breeds, the Naked neck had the highest titre on day 5 post immunisation but the Aseel titre was highest on days 12 and 19. 6. Males tended to have higher titres than females in Aseel, Kadakanath, Naked Neck, White Leghorn and Naked Neck broilers whereas Frizzle, Dahlem Red and SDL broilers showed the converse. 7. Statistical analysis revealed significant variation in HA response among the various genetic groups on different days post immunisation. The apparent differences between sexes were not significant. However, interactions between breed and sex were significant on day 5 and 19 post immunisation.  相似文献

Levels of passively transferred maternal antibodies to three canine viruses, rabies virus (RV), canine distemper virus (CDV) and infectious canine hepatitis (ICH) virus, in serum specimens from 14 fetal pups and in serial serum specimens collected up to 45 days after whelping from 14 neonate pups were compared with levels of antibodies to these viruses in milk and sera collected concurrently from their respective dams. Radioimmunoassays using RV-, CDV- and ICH virus-specific antigens showed that sera from all fetal pups had detectable levels of antibodies to all three canine viruses and ICH neutralising antibodies were detected in sera from 10 of the 14 fetal pups. As the time after whelping increased, titres of RV-, CDV- and ICH virus antibodies measured by radioimmunoassay and ICH virus neutralising antibody tests in serially collected specimens of milk from dams rapidly decreased, while titres of the antibodies in serum specimens from newborn pups in their litters steadily decreased. Individual fetal and newborn pups with a high titre of antibody to one virus also had high titres to the other two viruses, although a wide range of titres was observed among pups in each of the litters studied. Markedly higher titres of antibody to all three viruses were observed in serially collected specimens of sera from dams than in sera from fetal and newborn pups in their litters. Results show that maternal RV, CDV and ICH virus antibodies are transferred from dams to pups in utero and by nursing. Levels of these maternal virus-specific antibodies in newborn pup sera decreased at similar rates as time after whelping increased.  相似文献

An indirect ELISA has been developed to detect Salmonella typhimurium antibodies in chicken sera, using whole bacterial cell protein, flagellar protein or lipopolysaccharide as antigens. In experimental infections high concentrations of S typhimurium-specific IgG persisted after the faecal excretion of S typhimurium had ceased, whereas the specific IgM response was transitory. Some uninfected chickens placed in contact with experimentally infected birds developed high IgG titres in the absence of detectable faecal excretion. Other S typhimurium strains, which varied in their invasive abilities, also induced high titres of IgG. The ELISA allowed chickens infected experimentally with S typhimurium to be differentiated from chickens infected with 10 other serotypes, including S enteritidis. The use of whole blood in place of serum in the ELISA reduced the titres slightly. The storage of serum dried on to filter paper strips for four weeks produced little change in ELISA antibody titre, and the treatment of such strips with phenol or chloroform vapour had little or no effect on the antibody titre.  相似文献

We studied the problems of sarcocystosis of lambs in regard to the transfer of antibodies over ewes to lambs. Antibodies in the blood serum of 26 ewes found out by help of indirect immunofluorescent reaction (NFR) varied within the range from 640 to 5120 titres, while in six cases their level was higher than in blood. In none of new-born lambs there were proved antibodies to the sarcocystosis before the drinking of colostrum but three days later we already detected antibodies in the dilution 1:4 to 1:128. Then we studied the titre dynamics of antibodies in weaned lambs. In the age of 10-18 weeks we proved only low titres of antibodies, from the 20th to the 24th weeks the titres gradually increased and from the 26th to the 30th weeks the level of antibodies increased significantly (5120 titres) with its maximum on about the 28th week. Animals had no clinic symptoms of disease. In the other part of the work we examined the muscular substance of 342 lambs, killed in the slaughter house, by a digestive method and we carried out the blood serologic examination (NFR). Microcysts were found out in 35.67% of lambs while macrocysts were so in none of them. Specific antibodies of sarcocystosis were detected in 94.44% of young animals. A high discrepancy of the both methods (61.69%) in young animals is a result of a significant antibody reaction at still undeveloped muscular cysts.  相似文献

The aim of this study was to evaluate the effects of domperidone, a dopamine D2 receptor antagonist, in dogs naturally infected by Leishmania infantum. Ninety-eight dogs were treated with single-agent domperidone at 1mg/kg twice a day orally for 1 month. Clinical, serological, biochemical and immunological examinations were conducted for the following 12 months. Domperidone was effective in controlling and reducing clinical signs and antibody titre. Significant decreases in reciprocal serum antibodies were seen in 74.3% of the dogs with mild clinical signs and 40% of the dogs became seronegative. In dogs with several clinical signs and high antibody titres, clinical improvement occurred in 86% of animals and the reciprocal serum antibody titres decreased in 38% of these dogs. A significant increase was noted in the immune cellular status, as measured by the leishmanin skin test and a lymphocyte proliferation assay.  相似文献

Rabies is a highly fatal viral infection of the central nervous system affecting all warm-blooded animals including humans. To implement the preventive and control measures, it is important to decide the status of anti-rabies antibodies in dogs. Out of 120 serum samples, 47 (39.2 %) serum samples, showed an antibody titre equal to or above the cut off value of 0.5 IU/ml. The maximum number of dogs showed anti-rabies antibody titres equal to or above the cut-off value of 0.5 IU/ml after <1 month pre-exposure to the rabies vaccine. In 15 serum samples of pet dogs, we observed 13 (86.66 %) dogs with protective anti-rabies antibody titre. Statistical analysis suggests that the age of the animal had no significant effect on anti-rabies antibody titre in vaccinated pet dogs. The overall low seroprevalence of anti-rabies antibody in stray dogs indicates their susceptibility to rabies infection and thus posing a risk of rabies to other animals and humans.  相似文献

Young Thoroughbred racehorses (222 yearlings entering training and 246 2-year-old horses already in training) from eight flat-training yards in Newmarket, UK were used to monitor serological responses to vaccination with an inactivated influenza virus vaccine. Blood samples taken prior to and after vaccination were tested by single radial haemolysis (SRH) to determine antibody titres (expressed as area of haemolysis in mm(2)). Prior to vaccination, yearlings had mean antibody titres (64+/-4 mm(2)) that were approximately half of those of 2-year-olds (115+/-3 mm(2)) and 89% of yearlings and 73% of 2-year-olds had SRH titres <140 mm(2). Extrapolation from experimental and field studies suggests that these levels would not protect against homologous influenza virus infection. Both age-groups showed anamnestic responses to vaccination resulting in similar peak mean titres ( approximately 160+/-2mm(2)) with 67% of yearlings and 73% of 2-year-olds achieving levels > or =140 mm(2). A second dose of vaccine administered a month after the first in yearlings did not increase the mean titre but 75% of horses had levels of antibody > or =140 mm(2). The vaccination history in the official passport of yearlings showed that 23% had no record of previous vaccination and were probably fully susceptible to infection. For yearlings entering training, the important predictors from multiple-regression analyses of SRH titres prior to vaccination were "Time since last vaccination," "Total number of previous vaccines" and "Age at first vaccination." In 2-year-olds and following two doses of vaccine in yearlings, there was no significant relationship between these factors and SRH titre.  相似文献

The risk of infection with avian influenza viruses for poultry workers is relatively unknown in China, and study results are often biased by the notification of only the severe human cases. Protein microarray was used to detect binding antibodies to 13 different haemagglutinin (HA1‐part) antigens of avian influenza A(H5N1), A(H7N7), A(H7N9) and A(H9N2) viruses, in serum samples from poultry workers and healthy blood donors collected in the course of 3 years in Guangdong Province, China. Significantly higher antibody titre levels were detected in poultry workers when compared to blood donors for the most recent H5 and H9 strains tested. These differences were most pronounced in younger age groups for antigens from older strains, but were observed in all age groups for the recent H5 and H9 antigens. For the H7 strains tested, only poultry workers from two retail live poultry markets had significantly higher antibody titres compared to blood donors.  相似文献

Virulent Newcastle disease virus (NDV) was isolated from susceptible and immune chickens following intra-ocular challenge with the Essex '70 strain. Challenge virus was isolated from the trachea and cloaca of susceptible birds until they died 7 to 9 days after challenge. This virus was isolated from immunised chickens for up to 14 days after challenge. The duration of excretion was influenced by the prechallenge serum antibody titre to NDV. It persisted longest in chickens with titres of 2(3) to 2(7) and decreased in length and frequency from chickens with titres in the range 2(8) to 2(12). Chickens with pre-challenge titres of 2(3) to 2(5) developed 2- to 3- fold increases in post-challenge titres, whereas those with higher pre-challenge titres had smaller proportional increases in titre. Excretion of virulent virus from immunised birds should be considered in the development of Newcastle disease control programs.  相似文献

Measurement of anti-A and anti-B haemagglutinating alloantibody titres was performed on serum samples from both pedigree (n = 61) and non-pedigree (n = 43) cats previously typed using a commercial blood typing kit. All 40 type B cats had anti-A antibody with titres ranging from 4 to 1600. Among the 61 type A sera tested, an anti-B agglutination titre of greater than 2 was recorded in 16.4 per cent. The adjuvant vaccine against infectious bovine rhinotracheitis (IBR) was tested as to its innocuousness and immunogenicity. The immunity response induced by a single or double application of different vaccine doses was evaluated according to the content of neutralization antibodies (NP) in the blood serum. A direct dependence was revealed between the size of the inoculum and NP content in the blood serum, with NP titres of 1 : 9.3; 1 : 26.6, 1 : 80 and 1 : 149 after doubled application of 1, 2, 5, and 10 ml of the vaccine. The calves inoculated at an age of one week produced antibodies in the same titres as one- to five-month-old calves. Singly inoculated animals mostly showed zero-level or low antibody titres, but revaccination induced general serum-positivity with NP titres 1 : 4 to 1 : 128. The animals which had been in contact with the IBR virus and were serologically negative during inoculation or had an NP content in the blood serum at a titre of 1 : 4 or less, gave an anamnestic response to inoculation, but revaccination did not lead to a significant rise in antibody content. Double administration of 2 ml of vaccine in four production charges induced the production of antibodies with average titres of 1 : 36, 1 : 25; 1 : 31 and 1 : 24. Inoculation of susceptible animals in non-infected herds and of clinically healthy animals in infected herds did not cause any health disorders. Fifteen minipigs were infected intratracheally with three different doses (10(8), 10(5) or 5 x 10(3) colony forming units) of the reference strains of serotypes 2 or 4 of Actinobacillus pleuropneumoniae, and three remained as controls. The titre of specific IgG, IgM and IgA in the serum was measured weekly for 15 weeks with an indirect ELISA using monoclonal antibodies specific for each isotype. IgG attained the highest titres, IgM lower and IgA the lowest, being only detected in four animals. Serotype 4 evoked significantly higher titres than serotype 2 (P < or = 0.01). In general the highest IgG titres were attained at four to six weeks after infection. Some of the minipigs were reinfected after seven weeks but this evoked an increased titre in only two instances.  相似文献

The associations between the levels of canine distemper virus neutralising antibodies and vaccination history, age and gender were investigated in a cross-sectional study of a sample of 4627 dogs from the Finnish urban dog population. Dogs vaccinated with either Canlan (Langford Laboratories) or Dohyvac (Solvay Animal Health) or with both, had significantly lower titres than those vaccinated with Candur (Behringwerke), Duramune (Fort Dodge Laboratories) or Nobivac (Intervet), or with combinations including at least one of these. The vaccines were classified as having low and high immunogenicity on the basis of the geometric mean titre achieved by the vaccine when compared with the geometric mean titre of the entire dataset. The low geometric mean titre of Canlan, Dohyvac and their combination groups resulted from the large proportion of dogs without detectable titres, especially dogs under one year of age, rather than from uniformly low titres. An age-stratified comparison of vaccine usage and titres showed that the division of the vaccines into low and high immunogenicity vaccines was apparent in the dogs less than two years of age but not in the older dogs. The first vaccination with the high immunogenicity vaccines resulted in a higher proportion of dogs with detectable antibodies than even repeated vaccination with the low immunogenicity vaccines. Neither the time elapsed since the most recent vaccination nor the gender of the dogs was associated with the titre of antibody.  相似文献

Preruminant calves fed heated soyabean flour produced high titres of serum antibodies with specificity mainly for soyabean glycinin and beta-conglycinin. Various soyabean products that had been processed by different methods were analysed by immunological techniques to determine their content of glycinin and beta-conglycinin in soluble and undenatured form. The presence of such proteins in soyabean flour fed to preruminant calves was associated with the development of digestive disturbances and the production of serum antibodies. It is suggested that the suitability of soyabean products for calf nutrition could be assessed by simple immunological methods to detect these constituents.  相似文献

As immunosuppression in pigeons is common and results in reduced post-vaccination immunity and lower health status of the birds, studies have been taken up aimed at evaluation of the effect of three doses of methisoprinol on the percentage of CD4+ and CD8+ T lymphocyte subpopulation in peripheral blood and in the spleen and the titre of anti-NDV antibodies in the serum of pigeons in four groups (A, B, C, D), with 20 birds each. Pigeons in each group were immunised against paramyxovirosis at week 6 and 9 of life. Water for injection (group A - control) or methisoprinol at 100 mg/kg of body weight (group B), 200 mg/kg of body weight (group C) and 600 mg/kg of body weight (group D) was administered intramuscularly for 3 days before each vaccination. The immunological analyses were carried out by flow cytometry and the ELISA test. The findings indicate that methisoprinol administered intramuscularly at 100 and 200 mg/kg of body weight for 3 successive days before vaccination against paramyxovirosis mainly stimulates the mechanisms of non-specific humoral and cellular immunity, which is indicated by a higher percentage of the subpopulation of CD4+ T lymphocytes in peripheral blood and in the spleen and a higher titre of anti-NDV antibodies.  相似文献

In the present study the occurrence of morbillivirus infection in harbour porpoises (Phocoena phocoena) from the Black Sea was investigated. Blood and tissue specimens of lung, brain and spleen from 73 stranded or by-caught harbour porpoises derived from the three Black Sea subregions such as Bulgaria, Georgia and Ukraine were collected between 1997 and 1999 and processed for histology, immunohistochemistry and serology. Age determination was performed according to dental growth layers and body length. The age of the investigated population ranged from neonates to a 12-year-old animal. Morbillivirus-specific neutralizing antibodies were detected in 53% of harbour porpoises. Generally, titres were very low and ranged from 20 to 270. There was no correlation between age, geographical origin and titre levels. The most common histological finding (97%) consisted of a mild to severe granulomatous bronchopneumonia due to lung worm infection. There were no changes indicative of a morbillivirus infection. Using immunohistology none of the animals were positive for morbillivirus antigen. However, the serological data are suggestive of a continuously circulating morbillivirus among harbour porpoises from the Black Sea indicating that harbour porpoises may serve as carriers for fatal diseases in susceptible cetacean species.  相似文献
