In the high altitude regions of Africa, coffee berry disease (CBD), caused by Colletotrichum kahawae, is the main constraint for arabica coffee (Coffea arabica) production. However, certain agricultural practices can reduce losses caused by the disease and thereby promote optimum production. On small family farms in Cameroon, mixed cropping with fruit trees, intercropping with food crops and maintenance pruning of coffee trees are very widespread agricultural practices that can affect CBD epidemics. Consequently, an epidemiological study was conducted to assess how cultural practices affected the disease in an arabica coffee smallholding in Cameroon. The disease was monitored on a weekly basis over four successive years (2002–2005) on coffee trees in diverse cultural situations. Cultural practices likely to reduce losses due to CBD were identified. The infection rate was significantly lower on coffee trees grown intensively than on coffee trees grown in the traditional manner. Coffee trees located under the shade of fruit trees were significantly less attacked than those located in full sunlight. In addition, berries on the leafless parts of branches, near the main trunk of the coffee tree, were less infected than those on leafy sections. These results show that maintenance pruning, removal of mummified berries, and mixed cropping with shade plants are cultural practices which create environmental conditions that limit CBD development.  相似文献

The development of coffee berry disease (CBD) epidemics (caused by Colletotrichum kahawae) in Cameroon was monitored over two successive years (2004 and 2005) on coffee trees protected from rainfall by transparent plastic sheets and on unprotected control trees. This work was done to assess how rain affected disease development when it did not fall directly onto the coffee trees and to determine the influence of primary inoculum on the severity of CBD. Weekly observations over the 2 years showed that there were 1·1% diseased berries on coffee trees completely protected from rainfall, compared with 45% diseased berries on unprotected coffee trees. Disease severity on unprotected trees during the 2 years of the experiment was estimated at 53% diseased berries, compared with 27% on trees only protected in the first year. These results confirmed rainfall as one of the key physical factors in the development of Arabica CBD. They also provided evidence of a subsequent effect of protecting coffee trees from rainfall in 2004 on the severity of CBD in 2005. This suggested some practices that might lead to very effective cultural control of CBD in regions where severe epidemics of the disease occur.  相似文献

ABSTRACT Grape berries are highly susceptible to powdery mildew 1 week after bloom but acquire ontogenic resistance 2 to 3 weeks later. We recently demonstrated that germinating conidia of the grape powdery mildew pathogen (Uncinula necator) cease development before penetration of the cuticle on older resistant berries. The mechanism that halts U. necator at that particular stage was not known. Several previous studies investigated potential host barriers or cell responses to powdery mildew in berries and leaves, but none included observation of the direct effect of these factors on pathogen development. We found that cuticle thickness increased with berry age, but that ingress by the pathogen halted before formation of a visible penetration pore. Cell wall thickness remained unchanged over the first 4 weeks after bloom, the time during which berries progressed from highly susceptible to nearly immune. Autofluorescent polyphenolic compounds accumulated at a higher frequency beneath appressoria on highly susceptible berries than on highly resistant berries; and oxidation of the above phenolics, indicated by cell discoloration, developed at a significantly higher frequency on susceptible berries. Beneath the first-formed appressoria of all germinated conidia, papillae occurred at a significantly higher frequency on 2- to 5-day-old berries than on 30- to 31-day-old fruit. The relatively few papillae observed on older berries were, in most cases (82.8 to 97.3%), found beneath appressoria of conidia that had failed to produce secondary hyphae. This contrasted with the more abundantly produced papillae on younger berries, where only 35.4 to 41.0% were located beneath appressoria of conidia that had failed to produce secondary hyphae. A pathogenesis-related gene (VvPR-1) was much more highly induced in susceptible berries than in resistant berries after inoculation with U. necator. In contrast, a germin-like protein (VvGLP3) was expressed within 16 h of inoculation in resistant, but not in susceptible berries. Our results suggest that several putative barriers to infection, i.e., cuticle and cell wall thickness, antimicrobial phenolics, and two previously described pathogenesis-related proteins, are not principal causes in halting pathogen ingress on ontogenically resistant berries, but rather that infection is halted by one or more of the following: (i) a preformed physical or biochemical barrier near the cuticle surface, or (ii) the rapid synthesis of an antifungal compound in older berries during the first few hours of the infection process.  相似文献

Isolates of Colletotrichum gloeosporioides associated with anthracnose disease on coffee berries in Vietnam were characterized by morphological and molecular methods. Random amplified polymorphic DNA (RAPD) and microsatellite-primered PCR (MP-PCR) analyses were employed to investigate the genetic variation among 38 and 51 isolates of C. gloeosporioides , respectively. According to both methods, the isolates mainly grouped in accordance with geographical origins. Higher genetic variation ( H  = 0·312 and 0·335) in the northern population of C. gloeosporioides than in the southern population ( H  = 0·261 and 0·186), according to the RAPD and MP-PCR markers, respectively, was indicative of a difference between the northern and southern populations. Moderate gene differentiation ( G st = 0·1) between populations from the north and the south was found. However, there was no differentiation between locations within the northern or southern populations, indicating significant gene flow. A four-gamete test indicated a high level of recombination, particularly in the south. The geographic differences may be explained by different histories of coffee cultivation in different parts of Vietnam. The symptoms caused by the Vietnamese isolates on both hypocotyls and green berries were less severe than symptoms caused by the reference CBD (coffee berry disease; Colletotrichum kahawae ) isolates originating from Africa.  相似文献

Coffee berry disease (CBD) caused by Colletotrichum kahawae is a major constraint to Arabica coffee (Coffea arabica) production in Africa. One source of resistance to the disease is a natural interspecific hybrid between C. arabica and C. canephora and its derivatives. This study is aimed at deciphering the genetic basis of the host resistance and identification of molecular markers associated with it. CBD is a mature stage disease and in the absence of a mature mapping population, early detection of disease reaction phenotypes of mapping individuals is required. Two F2 populations from crosses of cv. Catimor (resistant) and cv. SL28 (susceptible) were screened for resistance by a two step procedure. First, half of each population was screened 6 weeks after germination by inoculating hypocotyls with the pathogen. The surviving seedlings (G1) were considered to be resistant and were raised in a nursery together with the other unscreened halves (G2). Secondly, after one year, all the seedlings (G1 + G2) were screened by inoculation. Analysis of 57 microsatellites and 31 AFLP markers in 56 and 95 seedlings from G1 and G2, respectively, were performed. Eight AFLP and two microsatellites markers linked tightly to the resistant phenotype were identified and mapped to one unique chromosomal fragment introgressed from C. canephora. The gene conferring the resistance was localized within an 11 cM segment. It is concluded that the locus carries a major resistance gene designated Ck‐1, which is likely to be synonymous to the T gene described in previous studies.  相似文献

On the maturing bark of cut branches ofCoffea arabica previously sprayed with copper fungicides perithecia ofGlomerella cingulata were easily found after two to ten days of incubation. Without fungicides the number of perithecia was decidedly lower. On prunings left on the ground under the coffee trees for 6 to 24 weeks the perithecia could also be found, but the numbers declined rapidly with time. Perithecia ofG. cingulata could forcibly discharge ascospores under laboratory conditions. Monospore cultures obtained by catching ascospores on agar, invariably belonged to threeColletotrichum types. It was rarely possible to isolate aColletotrichum able to infect green coffee berries. Growthrate, colour of the mycelium, number of conidia producedin vitro and infectivity on green coffee berries, however, differed substantially fromC. coffeanum, the cause of coffee berry disease. In Kenya no evidence has been obtained that ascospores from perithecia on bark could infect wounded or unwounded green coffee berries. Neither has any infection been obtained with ascospores from perithecia grownin vitro. Possible explanations for the difference with previous findings are offered. Based on the data presented in this paper, it is concluded thatG. cingulata is not likely to play a role in the epidemiology of the coffee berry disease in Kenya.  相似文献

On the basis of pathogenicity tests on green berries or hypocotyls of coffee and by morphological and biochemical characteristics in culture, 31 isolates of Colletotrichum were classified into C. kahawe (24 isolates), C. gloeosporioides (six isolates) or C. acutatum (one isolate). Within these groups of isolates, vegetative compatibility groups (VCGs) were determined by complementation tests with mutants in the nitrate assimilation pathway. There were distinct incompatibility barriers between the three species. Among the C. gloeosporioides group, the three isolates tested were self-compatible but incompatible with each other. Within C. kahawe , 18 isolates were self-compatible and only one main VCG was detected. However, partial compatibility in C. kahawe was also indicated by variation in the intensity of heterokaryon formation between different pairs of isolates and between different types of mutant. The existence of only one VCG in C. kahawe is consistent with the low level of variation found in previous work on DNA polymorphism.  相似文献

ABSTRACT A fundamental principle of integrated pest management is that actions taken to manage disease should be commensurate with the risk of infection and loss. One of the less-studied factors that determines this risk is ontogenic, or age-related resistance of the host. Ontogenic resistance may operate at the whole plant level or in specific organs or tissues. Until recently, grape berries were thought to remain susceptible to powdery mildew (Uncinula necator) until late in their development. However, the development of ontogenic resistance is actually quite rapid in berries, and fruit become nearly immune to infection within 4 weeks after fruit set. Our objective was to determine how and at what stage the pathogen was halted in the infection process on ontogenically resistant berries. Adhesion of conidia, germination, and appressorium formation were not impeded on older berries. However, once berries were approximately 3 weeks old and older, few germlings were able to form secondary hyphae. Ontogenically resistant berries responded rapidly to infection by synthesis of a germin-like protein that had been previously shown to play a role in host defense against barley powdery mildew. On susceptible berries, cell discoloration around penetration sites indicated the oxidation of phenolic compounds; a process that was followed by localized cell death. However, the pathogen was still able to infect such cells prior to their death, continue secondary growth, and thereby colonize young berries. Formation of papillae was not involved in the differential resistance mechanism of older berries. In susceptible berries, papillae formed frequently at infection sites but did not always contain the pathogen, whereas in resistant berries, the pathogen was always halted prior to the formation of papillae. The host defense, which conditions ontogenic resistance, operates in the earliest stages of the infection process, in the absence of gross anatomical barriers, prior to the formation of a functional haustorium and prior to the development of a conspicuous penetration pore. We also found that diffuse powdery mildew colonies that were not visible in the field predisposed berries to bunch rot by Botrytis cinerea, increased the levels of infestation by spoilage microorganisms, and substantially degraded wine quality. Our improved understanding of the nature, causes, and stability of ontogenic resistance in the grapevine/ powdery mildew system has supported substantial changes in how fungicides are used to control the disease. Present applications are more focused on the period of maximum fruit susceptibility instead of following a calendar-based schedule. This has improved control, reduced losses, and in many cases reduced the number of fungicide applications required to suppress the disease. Particularly where fungicides are deployed in a programmatic fashion and ontogenic resistance is dynamic, there may be equivalent improvements to be made in other hostpathogen systems through studies of how host susceptibility changes through time.  相似文献

The volume of the cells ofErwinia amylovora (Burrill) Winslow et al. hardly changes with changing water potential, but its extracellular slime swells strongly with increasing water potential. When bacterial slime has accumulated in intercellular spaces of the host plant and the water potential rises, the slime will swell. If this slime cannot escape, it will exert a pressure on the surrounding plant cells. This ‘swelling pressure’ can rise to Δψ, being the change in water potential (up to 3 MPa). The pressure may lead to compression of soft host cells and to formation of large slime-filled holes in the plant tissue. Moreover, the swelling slime may force its way to the outside of the plant (exudation) or to healthy parts. Cork barriers, being formed by the plant after infection, may be breached if the mechanical pressure is high and the cork barrier is incomplete or not yet fully developed. Then, the resistance reaction of the plant, the attempt to seal off, is not effective. The swelling pressure would explain how the extracellular slime may function as a virulence factor.  相似文献

Colletotrichum kahawae is a specialized plant pathogen of arabica coffee in Africa, able to infect green berries. The economic impact of this pathogen means there is an urgent need to better understand its pathogenic lifestyle, in particular its aggressiveness. In this study, several quantitative traits including disease severity, latent period and incubation period were measured to concomitantly assess the aggressiveness of 26 C. kahawae isolates. The results show that the area under disease progression curve is the most informative variable, particularly when joined together with the index disease intensity 10 days after inoculation and latency period, while the incubation period is not a reliable trait to distinguish aggressiveness levels in C. kahawae. This study also confirms the suitability of hypocotyls and detached green berries to perform C. kahawae aggressiveness assays, revealing that hypocotyls are a more reproducible testing material. Based on isolate profiles, three aggressiveness classes were established (high, moderate and low). A cytological analysis of representative isolates from each class showed that aggressiveness can be related to the development of post-penetration stages, rather than conidia germination and appressoria differentiation. This study provides, for the first time, the best metrics to evaluate C. kahawae aggressiveness, characterizing the profile of a broad range of isolates, and defining a set of parameters that can be used to classify new isolates. Furthermore, the collected information will contribute to the improvement of coffee breeding programmes, through the selection of tester isolates for prescreening of resistant coffee materials, and offers the opportunity to engage on future genotype–phenotype studies.  相似文献

Loose smut fungi of barley and wheat (Ustilago nuda and U. tritici, respectively) colonize the plant without causing obvious disease symptoms before heading. The availability of diagnostic methods to detect and follow the growth of these pathogens in the plant would therefore be highly advantageous for both resistance breeding and the development of effective seed treatments. Using seed lots of barley and wheat highly infected with loose smut, we studied the early establishment of the loose smut pathogens in the plant by fluorescence microscopy. In hand-cut sections stained with the fluorochrome Blankophor?, fungal hyphae were observed to invade the shoot apical meristem and leaf primordia during the first days after the onset of germination. At the first node stage the ear and leaf primordia were generally extensively colonized. Hyphae of U. nuda were also regularly observed in high density in the nodes. A protocol was developed for the specific amplification of U. nuda and U. tritici DNA extracted from infected plant tissue. PCR screening of U nuda in seedlings from infected and healthy seed lots was compared to ELISA, microscopy and ultimately head infection of mature plants derived from tillers of the tested seedlings. The results indicated that a prediction of loose smut infection by real-time PCR is possible at the second leaf stage, and that the assay is equally suited for use with spring and winter varieties of barley and wheat.  相似文献

Twenty-three isolates of Colletotrichum gloeosporioides, five isolates of C. acutatum, two isolates of C. capsici and six isolates of C. boninense associated with anthracnose disease on coffee (Coffea spp.) in Vietnam were identified based on morphology and DNA analysis. Phylogenetic analysis of DNA sequences from the internal transcribed spacer region of nuclear rDNA and a portion of mitochondrial small subunit rRNA were concordant and allowed good separation of the taxa. We found several Colletotrichum isolates of unknown species and their taxonomic position remains unresolved. The majority of Vietnamese isolates belonged to C. gloeosporioides and they grouped together with the coffee berry disease (CBD) fungus, C. kahawae. However, C. kahawae could be distinguished from the Vietnamese C. gloeosporioides isolates based on ammonium tartrate utilization, growth rate and pathogenicity. C. gloeosporioides isolates were more pathogenic on detached green berries than isolates of the other species, i.e. C. acutatum, C capsici and C. boninense. Some of the C. gloeosporioides isolates produced slightly sunken lesions on green berries resembling CBD symptoms but it did not destroy the bean. We did not find any evidence of the presence of C. kahawae in Vietnam.  相似文献

Coffee berry disease (CBD) is caused by the fungus Colletotrichum kahawae and is restricted to the African continent, where it generates losses of up to 80 % of coffee production. Weather conditions in certain growing areas at high altitudes in Colombia appear to be very favourable for the development of this disease. Certain genotypes of Coffee arabica are resistant to this pathogen, such as the Timor Hybrid and some Ethiopian accessions. It is important to identify the proteins in these coffee genotypes that are associated with resistance to this fungus. Therefore, we compared the proteomes of two genotypes that are resistant to different isolates of C. kahawae with the proteome of the susceptible coffee genotype Caturra. We optimized the methodology applied for the extraction, cleaning and purification of proteins from the green fruit pericarp at 150 to 170 days after flowering. Through two-dimensional differential gel electrophoresis, proteomic map images were obtained for the resistant and susceptible genotypes. Fifty-two protein spots that were significantly different between the resistant and susceptible genotypes were detected. These protein spots were isolated and sequenced via mass spectrometry. The sequence analysis identified 14 proteins in the Timor Hybrid and 14 in CCC1147 that were associated with resistance and pathogen defence.  相似文献

The decline of cork oak (Quercus suber) stands in Iberian Peninsula is associated with infection by Phytophthora cinnamomi. Most Phytophthora species secrete elicitins, which can enhance defence reactions against some pathogens. Here cytological and physiological effects of the elicitins cryptogein and capsicein on cork oak root infection by P. cinnamomi were evaluated. The progression of the pathogen in root tissue and its effects on total fatty acid (TFA) composition of roots and leaves were analysed in seedlings. Net photosynthesis (P n ), stomatal conductance (g s ), chlorophyll a fluorescence measurements (quantum yield of linear electron transport ? e , photochemical quenching q P, non-photochemical quenching NPQ) and carotenoid determinations were carried out in well established (4?months) plants. In elicitin-treated roots, 2?days after inoculation, the pathogen which presented loss of viability and membrane degradation was mainly restricted to the intercellular spaces of the cortical parenchyma, and did not reach the vascular cylinder. Electron dense materials accumulated in the intercellular spaces of the cortex next to disorganized hyphae, suggested to be related with defence reactions. Cryptogein (or its interaction with P. cinnamomi) induced enhanced lipid synthesis in leaves, which may contribute to preserve membrane stability. P. cinnamomi decreased P n , g s , ? e , and q P, whereas elicitin-treated plants displayed values similar to controls. Overall, the results indicated a resistance response of cork oak against this oomycete, induced by the elicitins.  相似文献

Between 1977 and 1981, seven qualitatively distinct new races of coffee leaf rust (Hemileia vastatrix) were detected in breeding plots in the State of São Paulo, Brazil. Four races carry complex virulence against the resistance genes SH1, SH2, SH4 and SH5 ofCoffea arabica. Three races match unidentified resistance genes ofC. canephora. Two of these were isolated from cv. Kouillou and one from the hybrid population Icatu. Pending further identification, these races were indicated by the number of their type cultures (Is. 2, 10 and 11). Is. 2 and Is. 10 showed extra virulence to some coffee genotypes and decreased virulence to other coffee genotypes, suggesting stabilizing selection. Three rust isolates were detected which differed quantitatively from the common rust race II. Is. 1 was moderately virulent to the coffee differential for SH3 in the laboratory but avirulent in the greenhouse, indicating a host x pathogen x environment interaction. Is. 3 and 12 showed levels of virulence intermediate between race II and Is. 2 and 10, respectively. The results show that incomplete resistance of coffee toH. vastatrix, at various levels, can be race-specific. The nature of race formation of coffee leaf rust in Brazil and breeding strategies for obtaining durable resistance are discussed.  相似文献

ABSTRACT Grape berries become resistant to powdery mildew early in development and are nearly immune to infection within 4 weeks after bloom. In this study, ontogenic resistance did not reduce attachment, germination, or appressorium formation of Uncinula necator on 3- to 4-week-old berries of Vitis vinifera 'Chardonnay' or 3-week-old berries of V. labruscana 'Concord'. Pathogen ingress halted at the cuticle before formation of a penetration pore. As berries aged, hyphal elongation and colony growth slowed until finally no secondary hyphae formed on fully resistant berries. More appressoria formed per unit of hyphal length as berries aged, indicating that failure to penetrate older berries led to increased attempts to penetrate resistant fruit. Additionally, hyphae within the colonies began to die as berries aged. Finally, the number of degree-hours between germination and sporulation of the colony (latent period) increased and sporophore density decreased with berry age at time of inoculation. Thus, ontogenic resistance both slows, and eventually halts disease development on grape berries, and limits the likelihood of spread by reducing absolute supply of conidia and delaying their formation. It furthermore has a consistent, stable, and predictable impact on grape powdery mildew and operates in a similar fashion and to a similar degree in both V. labruscana and V. vinifera, although at a slightly earlier phenological stage in V. labruscana.  相似文献

为评价东北地区玉米主推品种对禾谷镰孢根腐病的抗性水平,探究根腐病发生与苗势、产量损失间的关系,采用人工接种方法鉴定东北地区117个玉米主推品种对禾谷镰孢根腐病的抗性水平,应用室内盆栽试验分析自交系LN810在施用氮、磷、钾及复合肥后对禾谷镰孢根腐病发生和苗势的影响,在田间对先玉335接种禾谷镰孢菌Fusarium graminearum后探究根腐病发生程度对其苗势及产量的影响。结果显示,117个玉米品种对禾谷镰孢根腐病的抗性差异明显,鉴定出高抗品种14个,抗病品种44个,中抗品种47个,感病品种12个,其中表现中抗以上的品种占89.74%,且中早熟材料均表现为抗性。与未施肥相比,施用磷钾肥后玉米禾谷镰孢根腐病发生率最低,为22.03%。按照禾谷镰孢根腐病发生程度从轻到重将先玉335群体划分为一、二、三类苗,级别越高苗势越弱,产量测定发现二、三类苗的平均产量较一类苗的平均产量分别下降了39.97%和76.39%。表明东北地区主推玉米品种大部分对禾谷镰孢根腐病表现出抗性,但仍有部分品种存在较大感病风险,且该病害的发生程度与幼苗长势和产量呈负相关,施用磷钾肥可降低该病害的发生率。  相似文献

Magnaporthe oryzae chrysovirus 1 strain A (MoCV1-A) is associated with an impaired growth phenotype of its host fungus, Magnaporthe oryzae. In this report, we assayed the virulence and pathogenicity of MoCV1-A-infected and MoCV1-A-free M. oryzae on rice plants. MoCV1-A infection did not affect virulence-associated fungal traits, such as conidial germination and appressorium formation. However, after punch inoculation of leaves on rice plants, MoCV1-A-infected strain formed smaller lesions than the MoCV1-A-free strain did on all rice varieties tested, showing that MoCV1-A infection resulted in reduced virulence of host fungi in rice plants. In contrast, after spray inoculation of rice seedlings, in some cases, MoCV1-A-infected and MoCV1-A-free strains caused different lesion types (resistance to susceptible, or vice versa) on individual international differential rice varieties. However, we did not find any gain/loss of the fungal avirulence genes by PCR, suggesting that MoCV1-A infection can convert the pathogenicity of the host M. oryzae from avirulence to virulence, or from virulence to avirulence, depending on the rice variety. We also confirmed the correlation of these race conversion events and invasive hyphae growth of the fungi in a leaf sheath inoculation assay. These data suggested that MoCV1-A infection generally confers hypovirulence to the fungal host and could be a driving force to generate physiological diversity, including pathogenic races.  相似文献

Ultrastructural aspects of host–parasite interactions were investigated in fruits and leaves of citrus (satsuma mandarin) infected with Elsinoe fawcettii. Fungal infection induced host tissues to form cork layers bordering the necrotic areas below the infected sites. The cork layers were composed of compact host cells with convoluted cell walls and alternating lamellations, indicating ligno–suberized tissues in the wound periderm. No host tissues below the cork layers were invaded by hyphae. Hyphae grew intercellularly and intracellularly, often causing hypertrophy and compartmentalization of infected host cells. Also, host cells adjacent to invading hyphae showed accumulation of electron-dense materials and the formation of host cell wall protuberances in intercellular spaces. Hyphae had concentric bodies that showed an electron-transparent core surrounded by an electron-dense layer with radiating filamentous structures on their surface. One or more intrahyphal hyphae were found in the cytoplasm of intercellular or intracellular hyphae. These results suggest that the ligno–suberized cork layers in the wound periderm of citrus act as a protective barrier, which leads to restricted growth of E. fawcettii in bordered scab lesions. The fungus is thought to form concentric bodies and intrahyphal hyphae as a survival mechanism against the water- and nutrient-deficient environments that occur in the cork layers of necrotic host parts.  相似文献

Of 14 varieties and lines of cucumber and seven of cantaloup tested for resistance againstPseudoperonospora cubensis, only the cucumbers Poinsett and Chipper were found to be highly resistant. Growth chamber studies showed that resistance of young as well as of old (> 8-leaf-stage) plants was stable under considerably high (300 sporangia/cm2) inoculum loads. Resistance was conferred by both restriction of fungal growth and prevention of sporangial formation.  相似文献
