An experiment was conducted with 64 first-litter sows to evaluate the efficacy of a D2O dilution procedure for measuring in vivo body composition during the reproduction cycle. Eight gilts were each infused at breeding, 57 and 105 d postcoitum and at 5 and 25 d postpartum, with equivalent numbers of nongravid controls infused at corresponding periods except at 5 d postpartum. Results from D2O dilution were compared with body water estimates obtained from chemical analysis. An early-equilibrating D2O pool (before 15 min) was similar quantitatively to empty body (ingesta free) water in nongravid and lactating animals, but not in pregnant sows. Because of inconsistent D2O equilibration patterns in gravid sows, the early pool was considered to have equilibrated with part but not all of the water in the conceptus products. Total body D2O space measurement obtained from data following equilibration of D2O in the entire body (1 to 2 h) overestimated total body water (including gastrointestinal water) by approximately 19%. Coefficients of determination for equations relating total body D2O space to empty body and maternal body water were .96 and .88, respectively, in gestating sows and .67 and .74, respectively, for lactating sows, while coefficients of variation were below 6% in all cases. Prediction equations were developed to estimate empty and maternal body components (protein, fat and ash) from body weight and D2O space. Accuracy of protein and ash weight prediction is lowest with this procedure because it involves the composite error of estimation of the other body components.  相似文献

Current methods of estimating milk production in beef cows can be time-consuming, labor-intensive, and subject to high variability. The weigh-suckle-weigh (WSW) method requires repeated separation of offspring from their dams. Machine milking requires that animals be acclimated to the equipment prior to the estimation. The objective of Exp. 1 was to validate a deuterium oxide (D2O) dilution method of estimating milk production in cattle. In Exp. 1, Holstein calves (n = 5) averaging 29+/-2 d of age and 52.6+/-2.5 kg (+/- SE) were used as the model. Blood was collected for baseline D2O measurements followed by an injection of 300 mg D2O/kg BW. Syringes were weighed before and after the injection to gravimetrically determine the dose. Another blood sample was collected after D2O was allowed to equilibrate with body water for 2 h, and on each of the next five consecutive days, prior to feeding. Actual milk intake was measured by disappearance (i.e., amount of milk replacer offered to the calf minus the amount refused). Deuterium oxide in plasma was measured by mass spectrometry and milk intake was computed from the disappearance curve of D2O in blood plasma for each calf. Accumulated milk intake estimated by D2O dilution was highly correlated (y = 0.9x + 0.6; R2 = 0.99; P < 0.001) with actual milk intake. The objectives of Exp. 2 were to determine whether 1) D2O dilution was comparable to a standard measure of milk production in beef heifers and 2) growth hormone (GH) response to GH-releasing hormone (GHRH) in heifers at weaning is predictive of subsequent milk production. Deuterium oxide dilution and WSW were compared using 14 first-calf Angus heifers and their calves. Deuterium oxide dilution was used to estimate milk production of 40 first-calf Angus heifers that had been challenged with GHRH at weaning. Results indicate that the D2O dilution method is correlated (R2 = 0.89; P = 0.04) to the WSW estimation of milk production. Growth hormone response to GHRH in weanling heifers is positively related (R2 = 0.22; P = 0.03) to their subsequent milk production. Deuterium oxide dilution in calves offers an additional approach to the estimation of milk production of the dam in typical beef cattle production settings.  相似文献

The efficacy of the urea dilution technique in estimating the empty body composition of pigs weighing 50 kg was evaluated in three trials using 17 contemporary (Large White X Landrace X Hampshire X Duroc) and 8 Nebraska Gene Pool X contemporary pigs. Blood samples were collected via ear catheter before infusion (-60, -30 and 0 min) and at various times (3 to 90 min) after urea infusion (2.16 mmol/kg live BW), and analyzed for plasma urea. Backfat thickness of live pigs from the contemporary line was measured ultrasonically. Pigs then were killed by euthanasic injection, and total bodies (with gastrointestinal contents removed) were analyzed for water, protein and fat. In Trials 1 and 2, there were linear relationships (P less than .001) between chemically determined body water and fat and between body water and protein. Urea space was related (P less than .05) to empty body components with few exceptions, but regression coefficients for urea space in Trial 3 were different from those of Trials 1 and 2. Inclusion of additional independent variables with urea space improved estimation of empty body components. Although backfat alone did not estimate empty body components (except fat) as well as urea space alone, the addition of other common independent variables resulted in better estimates using backfat than urea space. The results of this experiment indicate that the urea dilution technique can be used to estimate the body composition of growing pigs. However, the accuracy obtained depended on the population of pigs being investigated and was no greater than the accuracy with appropriate equations based on backfat.  相似文献

The milk intake in piglets from four lactating sows was measured in three 48 h periods, starting on days 3, 10 and 17 of lactation, using the deuterium oxide (D 2 O) dilution technique. The milk intake was estimated from the water turnover in the piglets corrected for the production of metabolic water. The water turnover was estimated from the water dilution space and the fractional turnover of body water, determined by the rate of D 2 O dilution. To determine the magnitude of isotopic recycling, a randomly selected piglet in each litter was not enriched with D 2 O. Milk production of the four lactating sows was also measured by the weigh-suckle-weigh (WSW) method on days 4, 11 and 18 of lactation. The average daily milk production determined by the D 2 O dilution technique (9.91 - 0.69 kg) was higher ( P <0.05) than that determined by the uncorrected WSW method (8.65 - 0.81 kg). Different procedures for selecting piglets were evaluated to obtain the best possible estimation of total milk production from three piglets in each litter. The total milk production of a sow based on three piglets in the litter was obtained from a quadratic model of the relationship between milk intake and daily gain. When the daily gain of all piglets were known the Pearson correlation between milk production determined for the whole litter ( n -1) and only three median piglets was 0.98.  相似文献

OBJECTIVE: To evaluate energy expenditure (EE) in dogs by estimating rate of CO2 production (rCO2). ANIMALS: 15 Beagles. PROCEDURE: Food was withheld for 24 hours, and all dogs received an IV infusion of 13C sodium bicarbonate for 8 hours. Breath samples were collected before infusion and at 30-minute intervals from 4 to 8 hours, and 13C enrichment in breath CO2 was measured, using gas chromatography-isotopic ratio mass spectrometry. Food was withheld from 6 dogs, and rCO2 and O2 consumption were measured, using a conventional indirect calorimeter. The CO2 production and O2 consumption were measured by use of indirect calorimetry in 6 other fed dogs that were injected with 2H2O and H2(18)O. Blood samples were collected before tracer injection, 4 hours later, and on days 4, 7, and 11. Deuterium and 18O enrichments in plasma water were determined. RESULTS: Mean rCO2 measured by indirect calorimetry was 516 +/- 34 and 410 +/- 16 micromol/kg(0.75)/min in 6 fed and 6 food-deprived dogs, respectively. The rCO2 calculated from 13C-bicarbonate dilution was 482 +/- 30 micromol/kg(0.75)/min. Mean rCO2 determined by use of the double-labeled water method was 1,036 +/- 46 mmol/kg(0.75)/d. Mean energy expenditure calculated from rCO2 determined by infusion of 13C bicarbonate, indirect calorimetry in fed and food-deprived dogs, and infusion of double-labeled water was 386 +/- 39, 379 +/- 25, 338 +/- 14, and 552 +/- 25 kJ/kg(0.75)/d, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Energy expenditure calculated by indirect calorimetry in unfed dogs can be considered representative of basal metabolic rate.  相似文献

The pharmacokinetics of ascorbic acid were studied in 29 horses after intravenous (iv), subcutaneous, intramuscular (im) and oral administration. Following iv injection of 5 and 10 g ascorbic acid, respectively, a biphasic decline of ascorbic acid serum levels was found, indicating that the vitamin distributes in the body according to a two-compartment open model. The apparent volume of distribution (average value for Vd(ss)= 0.6 litre/kg) was approximately equivalent to the volume of total body water. The terminal half-life of the biexponential serum level-time curve (t1/2β) varied between 5 and 17 h. Both distribution and elimination were found to be positively correlated with the iv dose administered. Following subcutaneous and im injection, the average bioavailability of ascorbic acid amounted to 82 and 61 per cent, respectively. However, both routes of administration gave rise to marked local irritation. Following oral administration, the systemic availability of ascorbic acid was very poor. Serum levels in most experiments were not increased above the endogenous pre-administration values of the vitamin. Thus, in horses iv injection appears to be the only satisfactory route of administration of ascorbic acid if supplementation is required.  相似文献

The current study was undertaken to determine the effects of human growth hormone-releasing factor [hpGRF-(1-44)-NH2] on growth performance in pigs and whether this response was comparable to exogenous porcine growth hormone (pGH) treatment. Preliminary studies were conducted to determine if GRF increased plasma GH concentration after iv and im injection and the nature of the dose response. Growth hormone-releasing factor stimulated the release of pGH in a dose-dependent fashion, although the individual responses varied widely among pigs. The results from the im study were used to determine the dose of GRF to use for a 30-d growth trial. Thirty-six Yorkshire-Duroc barrows (initial wt 50 kg) were randomly allotted to one of three experimental groups (C = control, GRF and pGH). Pigs were treated daily with 30 micrograms of GRF/kg body weight by im injection in the neck. Pigs treated with pGH were also given 30 micrograms/kg body weight by im injection. Growth rate was increased 10% by pGH vs C pigs (P less than .05). Growth rate was not affected by GRF; however, hot and chilled carcass weights were increased 5% vs C pigs (P less than .05). On an absolute basis, adipose tissue mass was unaffected by pGH or GRF. Carcass lipid (percent of soft-tissue mass) was decreased 13% by GRF (P less than .05) and 18% by pGH (P less than .05). Muscle mass was significantly increased by pGH but not by GRF. There was a trend for feed efficiency to be improved by GRF; however, this was not different from control pigs. In contrast, pGH increased feed efficiency 19% vs control pigs (P less than .05). Chronic administration of GRF increased anterior pituitary weight but did not affect pituitary GH content or concentration. When blood was taken 3 h post-injection, both GRF- and pGH-treated pigs had lower blood-urea nitrogen concentrations. Serum glucose was significantly elevated by both GRF and pGH treatment. This was associated with an elevation in serum insulin. These results indicate that increasing the GH concentration in blood by either exogenous GH or GRF enhances growth performance. The effects of pGH were more marked than for GRF. Further studies are needed to determine the optimal dose of GRF to administer in growth trials and the appropriate pattern of GRF administration in order to determine whether GRF will enhance pig growth performance to the extent that exogenous pGH does.  相似文献

The present study was conducted to determine the effects of different dietary protein levels and recombinant porcine somatotropin (rpST) administration on deposition rates of protein, fat, water, ash, and collagen in pigs. Ten groups of six barrows (30 kg BW) were restrictively fed (80% of ad libitum) one of five diets containing 11, 15, 19, 23, or 27% CP. Diets were isoenergetic and all contained equivalent amounts of lysine. Thirty barrows were treated daily with rpST (100 micrograms/kg) by i.m. injection; remaining pigs were treated with diluent for 42 d. At all levels of dietary protein intake, carcass and empty body accretion rates of protein, water, and ash were greater in rpST-treated pigs than in respective controls. The magnitude of change elicited by rpST was lowest in pigs consuming 11% CP. Administration of rpST resulted in a 34% decrease in the accretion rate of fat; increasing protein intake resulted in a linear decrease in fat accretion in control and rpST-treated pigs. Accretion rates of protein, water, ash, and fat were increased in viscera of rpST-treated pigs compared with respective controls; rates of visceral protein and water accretion were increased as dietary protein was increased, whereas deposition of fat was decreased in control and rpST-treated pigs. Administration of rpST resulted in an overall 66% increase in the utilization efficiency of dietary protein for empty body protein deposition. Protein intake had minimal effect on the concentration of collagen in the carcass; however, rpST treatment increased concentrations of total and soluble collagen by 30 and 33%, respectively. Recombinant pST had little influence on collagen crosslinking or maturation. Deposition rate of carcass collagen was increased 63% in rpST-treated pigs compared with respective controls.  相似文献

An experiment was conducted to test the hypothesis that inclusion of Cu oxide (Cu2O) in diets for growing–finishing pigs improves body weight (BW) and bone mineralization, and reduces accumulation of Cu in the liver compared with pigs fed diets containing Cu sulfate (CuSO4). Two hundred growing pigs (initial BW: 11.5 ± 0.98 kg) were allotted to a randomized complete block design with 2 blocks of 100 pigs, 5 dietary treatments, 5 pigs per pen, and a total of 8 pens per treatment. Treatments included the negative control (NC) diet that contained 20 mg Cu/kg, and 4 diets in which 125 or 250 mg Cu/kg from CuSO4 or Cu2O were added to the NC diet. The experiment was divided into 4 phases and concluded when pigs reached market weight. Pig weights were recorded on day 1 and at the end of each phase and feed provisions were recorded throughout the experiment. On the last day of phases 1 and 4, 1 pig per pen was sacrificed to obtain samples of liver and spleen tissue, and the right metacarpal was collected. Results indicated that pigs fed diets containing 250 mg Cu/kg from CuSO4 had greater BW at the end of phases 1 and 2 than pigs fed NC diets. Pigs fed diets containing 250 mg Cu/kg from Cu2O had greater (P < 0.05) BW at the end of phases 1, 2, 3, and 4 compared with pigs fed NC diets, and these pigs also had greater BW at the end of phases 3 and 4 than pigs fed all other diets. Pigs fed the diets with 250 mg Cu/kg tended to have greater (P < 0.10) feed intake than pigs fed the NC diet at the end of phase 2, and for the overall experimental period, pigs fed diets containing 250 mg Cu/kg from Cu2O had greater (P < 0.05) feed intake than pigs on all other treatments. However, no differences in gain:feed ratio were observed among treatments. Copper accumulation in liver and spleen increased with Cu dose, but at the end of phase 1, pigs fed 250 mg Cu/kg from CuSO4 had greater (P < 0.05) Cu concentration in liver and spleen than pigs fed 250 mg Cu/kg from Cu2O. Pigs fed diets containing 250 mg Cu/kg from Cu2O had greater (P < 0.05) quantities of bone ash and greater (P < 0.05) concentrations of Ca, P, and Cu in bone ash than pigs fed NC diets or the 2 diets containing CuSO4, but Zn concentration in bone ash was less (P < 0.05) in pigs fed diets containing 250 mg Cu/kg from Cu2O. To conclude, supplementing diets for growing pigs with Cu2O improves growth performance and bone mineralization with less Cu accumulation in liver compared with pigs fed diets containing CuSO4.  相似文献

The pharmacokinetic behaviour and bioavailability of enrofloxacin (ENR) were determined after single intravenous (iv) and intramuscular (im) administrations of 5mg/kg bw to six healthy adult Angora rabbits. Plasma ENR concentrations were measured by high performance liquid chromatography. The pharmacokinetic data were best described by a two-compartment open model. ENR pharmacokinetic parameters were similar (p>0.05) for iv and im administrations in terms of AUC0-infinity, t1/2beta and MRT. ENR was rapidly (t1/2a, 0.05 h) and almost completely (F, 87%) absorbed after im injection. In conclusion, the pharmacokinetic properties of ENR following iv and im administration in Angora rabbits are similar to other rabbit breeds, and once or twice daily iv and im administrations of ENR at the dose of 5mg/kg bw, depending upon the causative pathogen and/or severity of disorders, may be useful in treatment of infectious diseases caused by sensitive pathogens in Angora rabbits.  相似文献

The efficacy of an Escherichia coli-derived phytase preparation   总被引:1,自引:0,他引:1  
Five experiments were conducted to evaluate the effect of an Escherichia coli-derived phytase on phytate-P use and growth performance by young pigs. The first experiment involved time course, pH dependence, and phytase activity studies to investigate the in vitro release of P from corn, soybean meal, and an inorganic P-unsupplemented corn-soybean meal negative control diet. In Exp. 2, which was designed to determine the efficacy of the E. coli-derived vs. fungal phytase-added diets at 0, 250, 500, 750, 1,000, or 1,250 FTU/kg (as-fed basis; one phytase unit or FTU is defined as the quantity of enzyme required to liberate 1 micromol of inorganic P/min, at pH 5.5, from an excess of 15 microM sodium phytate at 37 approximately C) and a positive control diet, eight individually penned 10-kg pigs per diet (12 diets, 96 pigs) were used in a 28-d growth study. The third experiment was a 10-d nutrient balance study involving six 13-kg pigs per diet (four diets, 24 pigs) in individual metabolism crates. In Exp. 4, eight pens (four pigs per pen) of 19-kg pigs per treatment were used in a 42-d growth performance study to examine the effect of adding the E. coli-derived phytase to corn-soybean diets at 0, 500, or 1,000 FTU/kg (as-fed basis) and a positive control (four diets, 128 pigs). In Exp. 5, six 19-kg pigs per treatment were used in a 10-d nutrient balance study to investigate the effects of the E. coli-derived phytase added to diets at 0, 250, 500, 750, or 1,000 FTU/kg (as-fed basis) and a positive control diet (six diets, 36 pigs). The in vitro study showed that the E. coli-derived phytase has an optimal activity and pH range of 2 to 4.5. Inorganic phosphate release was greatest for soybean meal, least for corn, and intermediate for the negative control diet. Dietary supplementation with graded amounts of E. coli-derived phytase resulted in linear increases (P < 0.05) in weight gain, feed efficiency, and plasma Ca and P concentrations in 10-kg pigs in Exp. 2. Phytase also increased P digestibility and retention in the 13-kg pigs in Exp. 3. In Exp. 4, dietary supplementation with E. coli-derived phytase resulted in linear increases (P < 0.05) in weight gain and feed efficiency of 19-kg pigs. Supplementation of the diets of 19-kg pigs with the E. coli-derived phytase also improved Ca and P digestibility and retention in Exp. 5. In the current study, the new E. coli-derived phytase was efficacious in hydrolyzing phytate-P, both in vitro and in vivo, in young pigs.  相似文献

Systolic time intervals were measured in 15 stress-susceptible (SS) pigs to derive regression equations to determine to what extent their ventricular functions differed from those of stress-resistant (SR) pigs. Regression analysis revealed that the RR interval was the variable that was significantly (P less than 0.01) related to electromechanical systole and left ventricular ejection time. The preejection period (PEP) was independent of the RR interval. A normal increase of all systolic time intervals with age, independent of the RR interval, was also observed in SR and SS pigs. The 55-kg SS pigs had a higher noradrenergic tone than did 15-kg and 90-kg SS pigs, because their mean arterial blood pressure was higher and because their index values for electromechanical systole and their PEP were shorter than those of SR pigs of the same body weights. The cardiovascular responses to biogenic amines were also different according to the degree of development and stress susceptibility. Changes in mean arterial pressure and PEP were not as pronounced after injections of epinephrine and norepinephrine (10(-8) mol/kg of body weight) were given. Subsequently, a decrease in cardiovascular responses to epinephrine and norepinephrine injections in 90-kg SS pigs were recorded. These results indicate that 55-kg SS pigs have a higher level of circulating catecholamines and that the myocardium becomes less sensitive to epinephrine and norepinephrine after it is chronically exposed to these amines.  相似文献

The effects of feeding various dietary Ca:P level sequences on gain and feed efficiency, leg structural soundness and bone indices of growing-finishing swine were evaluated as an incomplete 3 X 3 factorial arrangement of treatments in a split-plot design. A total of 664 pigs were fed one of three total dietary Ca:P levels (.52:.40, .65:.50, .80:.60%) from 19-kg to 56-kg body weights followed by one of three Ca:P levels (.45:.32, .52:.40, .65:.50%) to market weight. The .80:.60% and .65:.50% Ca:P mineral sequence was not evaluated. Diets were formulated to 14.5% crude protein using a corn-soybean meal mixture with proportions of dicalcium phosphate and limestone altered to attain the desired dietary Ca:P levels. Maximum gains occurred at the .65:.50% and .52:.40% Ca:P level during the grower (P less than .01) and finisher (P less than .01) periods, respectively. No grower X finisher phase pig gain or feed intake interaction resulted, providing evidence of no carry-over response on these measurements from the grower to the finisher period. Serum P concentration increased and plateaued at the same dietary Ca:P level, as did rate of gain at both 56-kg and 95-kg body weights. Leg soundness subjectively evaluated at 56-kg and 95-kg body weights revealed no effect of dietary Ca:P level on soundness scores at 56 kg, but at 95-kg body weight, the interaction between grower and finisher diets was significant. Percentage bone ash of the humerus, shaft thickness and bending moment of the femur increased as dietary Ca:P level increased at both 56-kg and 95-kg body weights.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Little information is available on the effects of growth hormone (GH) and growth hormone-releasing factor (GRF and GHRH) treatment on bone metabolism in pigs. Thus, tibial bending moments and ash contents were studied in 12, 6-wk-old pigs weighing 13 +/- .2 kg. Six pigs (GRF group) were injected s.c. twice daily with 75 micrograms GRF (hGRF [1-29] NH2)/kg BW for 52 d and six remained untreated (control group, C). Average daily gain was slightly (5%; P less than .10) increased in treated pigs. At slaughter, plasma measurements related to calcium homeostasis, such as concentrations of Ca, inorganic P, and vitamin D metabolites (25-OH and 1,25-(OH)2 vitamin D3), were not changed by GRF injection. At slaughter, plasma GH levels were 3.3 times greater in treated (11.3 +/- 3 ng/ml) than in untreated pigs (3.4 +/- .5 ng/ml, P less than .02), whereas those of insulin-like growth factor I were increased by approximately 38%. No difference was observed between the two groups at slaughter in tibial weight, density, bending moment, ash relative to bone volume (29 +/- 1 vs 30 +/- 2 g/100 cm3, GRF vs C), total ash content, or ash relative to dry matter in cortical or medullary bone. Six experiments were conducted to determine the true digestible valine requirement of 5- to 20-kg pigs. In Exp. 1, a valine-deficient diet for 5- to 10-kg pigs was developed and validated in terms of growth performance in response to supplemental L-valine. A different basal diet was validated for 10- to 20-kg pigs in Exp. 2. Both diets were demonstrated to be deficient in valine and to support performance equivalent to typical nursery diets when fortified with L-valine. In Exp. 3, true ileal digestibility of valine in the two basal diets was determined in eight pigs fitted with a simple T-cannula at the terminal ileum. Another four pigs received an enzymatically hydrolyzed casein-based diet to determine endogenous contributions to collected ileal digesta. The two diets were found to have true valine digestibilities of 82% (5- to 10-kg pigs) and 86% (10- to 20-kg pigs). In Exp. 4, 80 weaned pigs (5.8 kg) were offered the basal diet fortified with five incremental doses (0.08%) of L-valine. Weight gain increased quadratically (P < 0.05) with increasing levels of valine. Broken-line analysis revealed a true digestible valine requirement of 0.86 +/- 0.03%. In Exp. 5, the true digestible valine requirement of 10- to 20-kg pigs was estimated with 120 pigs (10.9 kg) using the second basal diet fortified with six incremental doses (0.05%) of L-valine. The data suggested a digestible valine requirement level of about 0.775%, which was reevaluated in Exp. 6, wherein pigs did not respond to levels of digestible valine higher than 0.775%. In conclusion, requirement estimates were 2.50 and 2.22 g of true digestible valine per megacalorie of ME for 5- to 10- and 10- to 20-kg pigs, respectively. These empirical estimates are in close agreement with recent estimates of the National Research Council Subcommittee on Swine Nutrition of 2.48 and 2.11 g of true digestible valine per megacalorie of ME, respectively.  相似文献

Twenty-eight 4-d-old nursing pigs were used in two trials to evaluate the effects of im injection of iron (Fe) from Fe-dextran upon subsequent serum mineral concentrations. An im injection of 150 mg of Fe from Fe-dextran was given to all pigs (n = 17) in trial 1. Serum Fe concentration increased rapidly from an initial mean of 65 to 3,466 micrograms/dl at 6 h postinjection (PI), 5,712 micrograms/dl at 24 h PI, then fell to 317 micrograms/dl at 4 d PI, 143 micrograms/dl at 7 d PI and remained about 100 micrograms/dl at 14, 21 and 28 d PI. Concentrations of other serum minerals did not appear to be influenced by the Fe-dextran injection. Treatments of Pigs (n = 11) in trial 2 were no injection, injection of dextran only, injection of 100 mg Fe from Fe-dextran and injection of 200 mg Fe from Fe-dextran. Serum Fe concentration increased rapidly after Fe-dextran injection, rising to a peak of 11,028 micrograms/dl at 6 h PI from the 100-mg Fe injection and 17,012 micrograms/dl at 6 h PI from the 200-mg Fe injection. Serum Fe concentration dropped steadily in pigs that were given no injection or injected with dextran only. A two-component, nontoxic, quantifiable animal/carcass tracing system was developed using riboflavin as an on-premises, initial carcass identifier visible under longwave ultraviolet (UV) light and deuterium oxide (D2O) as a tracer analytically quantified via fixed wavelength infrared spectrophotometry. Twenty-four cull cows and heifers were allocated into eight antemortem treatment groups (1, 12, 24, 48, 72, 96, 120, 144 h) for evaluation of the efficacy of riboflavin and D2O as tissue tracers in postmortem meat tissues. All cattle were slaughtered using conventional procedures and inspection. To study postmortem riboflavin marker changes due to constant light exposure over time, fluorescence and emission intensity scores were obtained by a trained panel 24, 48, and 168 h postslaughter. The riboflavin marker intensity rating means for UV fluorescence were classified as identifiable on all carcasses when evaluated under UV light, but were classified as not identifiable when evaluated under ambient light. Deuterium oxide levels in all tissue water samples, regardless of antemortem infusion group, contained D2O concentrations at least 2.5 times greater than those found in background water. Deuterium oxide was shown to disperse rapidly throughout living tissues. Correlations within animals for D2O levels from blood and muscle were all highly significant (r = .99).  相似文献

Chlortetracycline hydrochloride was administered intra-arterially (11 mg/kg) and as an oral drench (33 mg/kg) to ten 21.0-31.5-kg pigs. Five of the pigs were fasted 18 h prior to dosing and five of the pigs were fed ad libitum prior to dosing. The mean volume of distribution determined by area-under-the-curve calculations for the fasted pigs (0.967 +/- 0.210 l/kg) was significantly less (P less than 0.05) than the mean volume of distribution for the fed pigs (1.39 +/- 0.31 l/kg). Mean total body clearance of the drug was also significantly less (P less than 0.05) in the fasted pigs (0.165 +/- 0.055 l/kg/h) as compared to the fed pigs (0.307 +/- 0.053 l/kg/h). The elimination constants (beta) were not found to be statistically different (P less than 0.05): 0.1811 +/- 0.0057 for the fasted pigs; 0.2260 +/- 0.0461 for the fed pigs. The bioavailability for both groups was similar; 19.12 +/- 8.3% for the fasted pigs and 17.88 +/- 5.3% for the fed pigs. In a second experiment three groups of six pigs which weighed 34.5-44.1 kg were fed a corn-soy diet ad libitum. The rations were fortified with chlortetracycline at 100, 400 or 1000 mg chlortetracycline hydrochloride/kg feed. Chlortetracycline concentrations were determined in plasma samples collected over a 6-day period. Plasma chlortetracycline concentrations reach a plateau within 24 h after initial access to the trial diets and were highly correlated with the dose of the drug consumed (r2 = 0.97).  相似文献

Nineteen dogs were assigned randomly to one of three groups. Animals in Group 1 were pre-medicated with acepromazine, 50 μg/kg bodyweight (bwt) intramuscularly (im) and received 10 ml of 0.9 per cent saline intravenously (iv) at the time of skin incision. Dogs in Group 2 were pre-medicated with acepromazine, 50 μg/kg bwt im, and received fentanyl 2 μg/kg bwt iv at skin incision. Dogs in Group 3 were pre-medicated with acepromazine, 50 μg/kg bwt and atropine, 30 to 40 μg/kg bwt, im and received fentanyl, 2 μg/kg bwt iv at skin incision. Pulse rate, mean arterial blood pressure, respiratory rate and end tidal carbon dioxide were measured before and after fentanyl or saline injection. Fentanyl caused a short-lived fall in arterial blood pressure that was significant in dogs premedicated with acepromazine, but not in dogs pre-medicated with acepromazine and atropine. A significant bradycardia was evident for 5 mins in both fentanyl treated groups. The effect on respiratory rate was most pronounced in Group 3, in which four of seven dogs required intermittent positive pressure ventilation (IPPV) for up to 14 mins. Two of six dogs in Group 2 required IPPV, whereas respiratory rate remained unaltered in the saline controls. The quality of anaesthesia was excellent in the fentanyl treated groups; however, caution is urged with the use of even low doses of fentanyl in spontaneously breathing dogs under halothane-nitrous oxide anaesthesia.  相似文献

Six gilts, with an average BW of 70 kg, were fitted with a simple T-cannula at the distal ileum to study the effect of continuous starch infusion on urea kinetics by means of a radioisotope dilution technique. The pigs were fed twice daily 600 g of a cornstarch-based diet formulated to contain 16% CP by supplementation with isolated soy protein. Infusion of starch, compared with water, decreased (P < .05) plasma urea concentration, urea pool size, and entry, excretion, and degradation rates; urea turnover rate and urea space were not affected (P > .05). Expressed as a percentage of total entry rate, approximately 40% of urea was recycled into the digestive tract in both infusion treatments. The stimulation of microbial fermentation in the large intestine resulted in an increase (P < .05) in fecal N excretion, which was mainly due to an increased excretion of bacterial N. This increase could not be attributed to a greater secretion of urea into the large intestine and its subsequent utilization by the intestinal microflora. The increased bacterial N assimilation after starch infusion led to a reduction in ammonia absorption from the large intestine, which in turn was reflected by a reduced urinary N excretion. As a result, the overall N balance was not affected. In a second experiment, two barrows, with an average BW of 80 kg, were fed twice daily 1.4 kg of a cereal-based diet.
