Experiments were performed with young two-humped camels exposed to 36-hour starvation with free access to water. The renal functions were measured by the standard clearance method. In spite of the administration of 20 micrograms DDAVP, a higher urine flow rate was recorded in the camels subjected to control measurements (feed intake) than in the fasting period (1.45 +/- 0.06 vs. 0.96 +/- 0.06 ml . min-1, P less than less than 0.001). On the second day of fasting the camels had a significantly reduced glomerular filtration rate (GFR 317.5 +/- 23.2 vs. 170.2 +/- 17.4 ml . min-1, P less than 0.001), urea output (700.5 +/- 62.9 vs. 352.2 +/- 64.7 mumol . min-1, P less than 0.005), and fractional excretion of urea (26.9 +/- 2.8 vs. 17.9 +/- 1.7%, P less than 0.01), whereas their tubular resorption. of urea (Reab urea/GFR) increased (6.28 +/- 0.61 vs. 9.12 +/- 0.82 mumol . ml-1, P less than 0.02). No significant difference was found in the concentration of urea in plasma in the fed camels and in fasting camels (8.55 +/- 0.64 vs. 11.18 +/- 1.09 mmol . l-1, N. S.). The creatinine inulin clearance ratio (C creat/Cin) was 0.92 +/- 0.07 when the animals were fed and 1.17 +/- 0.05 when the animals starved (P less than 0.001); this suggests that the clearance of endogenous creatinine is not suitable for GFR measurement in camels under different conditions of nutrition. The kidneys of camels regulate the excretion of urea during short-time fasting mainly through the reduction of glomerular filtration rate and just partly through an increased tubular resorption.  相似文献

The renal excretion of selenium was investigated in ewes with an excretion of hypotonic urine (control group) and in ewes with a formation of highly concentrated urine. Chronic stimulation of the urinary concentrating activity of sheep kidneys was induced by a long-term treatment with 1-desamino-8-D-arginine vasopressin (dDAVP), a synthetic analogue of vasopressin with prolonged effects. Young animals with 22 to 25 kg b.w. were fed a normal protein diet providing a daily intake of 129.25 g of crude protein, 12.03 MJ of digestible energy and 0.18 mg of selenium for 3 weeks. The vasopressin treated sheep (n = 11) were given subcutaneous injections of 12.5 micrograms of dDAVP in glycerol twice daily for one week before the clearance measurement of renal functions. The control group (n = 11) was treated with glycerol only. The administration of dDAVP resulted in a highly significant decrease of the urinary flow rate (from 3.19 +/- 0.50 in control group to 0.33 +/- 0.03 mL.min-1 in dDAVP animals, P < 0.001) without changes in the glomerular filtration rate (80.18 +/- 6.36 in controls vs. 77.86 +/- 6.26 mL.min-1, NS). No effects on plasma selenium level were observed (0.17 +/- 0.03 in controls vs. 0.20 +/- 0.03 mumol.L-1, NS) but the amounts of selenium excreted were found to be highly significantly reduced (from 0.29 +/- 0.05 in controls to 0.03 +/- 0.01 nmol.min-1, P < 0.001) in dDAVP treated sheep. Despite a large reduction in urinary flow rate, the selenium concentration in urine was actually the same in both groups (0.09 +/- 0.01 mumol.L-1) resulting in a sharp fall in the renal clearance of selenium (2.20 +/- 0.54 in controls vs. 0.18 +/- 0.03 mL.min-1, P < 0.01) due to dDAVP. This seems to be a consequence of the large increase in the selenium solvent drag induced by a vasopressin treatment. The results presented suggest that vasopressin may contribute to maintenance of the selenium balance in sheep via its effects on renal function.  相似文献

Benzylpenicillin kinetics were investigated in a breed of sheep adapted to life in barren areas. Important pharmacokinetic differences, dependent on reproductive status (control, pregnancy, lactation), were demonstrated. In control ewes, the steady state volume of distribution was 0.23 +/- 0.15 litre kg-1 and the plasma clearance was 12.4 +/- 3.19 ml kg-1 min-1. During pregnancy, these two parameters were not significantly modified. During lactation, the plasma clearance and volume of distribution were significantly higher than during the control period and pregnancy (27.4 +/- 3.22 ml kg-1 min-1 and 0.696 +/- 0.20 litre kg-1, respectively (P less than 0.01). In contrast the mean residence time (25.0 +/- 4.6 minutes) was similar to those of the control period (17.54 +/- 6.25 minutes) and pregnancy (23.18 +/- 5.94 minutes). These modifications in benzylpenicillin kinetics were explained in terms of fluid balance adjustments and renal function adaptation observed during pregnancy and lactation, and it is suggested that an appropriate dosage regimen for benzylpenicillin should take into account the physiological status of the animal.  相似文献

Urea excretion was studied in an experiment with two sheep breeds (steppe fat-tailed and merino) on the second day of fasting when the urea concentration in blood increases in fasting animals. The control group in the two breeds was given free-choice feed and water while fasting sheep were given ad libitum only water. Diuresis in both breeds was steady during the experiment. Glomerular filtration rate was not found to vary, in comparison with the control, although the plasma urea concentration rose in fat-tailed sheep (P less than 0.01) as well as in the sheep of merino breed (P less than 0.001). Fractional excretion of urea decreased in fat-tailed sheep (P less than 0.05) and also in the sheep of merino breed (P less than 0.02) while total output of urea remained steady in fat-tailed sheep but it increased in merino sheep (P less than 0.02). Tubular reabsorption of urea on the second day of fasting was observed to be higher by 65% in merino sheep (P less than 0.001), but in steppe fat-tailed sheep the increase was much higher--by up to 180% (P less than 0.001), in comparison with the control. It was demonstrated by the results that the increased tubular reabsorption of urea contributes to the rise of plasma urea concentration in sheep on the second day of fasting.  相似文献

Respones to bumetanide were studied in five ewes. During ADH infusion urine flow increased from less than 1-0 to approximately 11-5 cm3 min-1 within 30 min of intravenous injection of 0-02 mg kg-1 bumetanide and returned to approximately 2-0 cm3 min-1 within 3 h of dosing. The diuresis was accompanied by large increases in sodium and chloride excretion and smaller increases in potassium and free hydrogen ion excretion. Bicarbonate excretion and TCH20 were reduced. Plasma potassium and chloride concentrations decreased slightly while arterial bicarbonate pH and pCO2 slightly increased. A transient increase in GFR and RPF was followed by a small reduction in GFR. No change in CH20 was observed after bumetanide injection during water diuresis. Increasing the dose of bumetanide over the range 0-002 to 0-20 mg kg-1 resulted in more pronounced and prolonged responses. The results show that bumetanide is a potent diuretic in sheep with its main site of action on the ascending limb of Henle's loop.  相似文献

Fasting is associated with unconjugated hyperbilirubinemia in several species, including the horse. Studies in ponies showed that a 3-day fast decreased plasma clearance of bilirubin, cholic acid, and sulfobromophthalein (BSP). Since these organic anions are conjugated with different substrates, it is possible that observed differences in plasma clearance result from a general decrease in hepatic conjugating capacity during the animals' fasting. To test this hypothesis, the effects of a 3-day fast on plasma clearance of IV injected BSP (4.4 to 5.1 mg/kg), which is conjugated to glutathione, and indocyanine green (ICG; 0.8 to 1.1 mg/kg), which is not conjugated, were studied in 10 healthy horses and 2 ponies with diverted enterohepatic circulations (indwelling T tubes). Blood samples were obtained for 30 minutes after injection, and bile samples from ponies were obtained for 3 hours. Fasting increased plasma bilirubin concentration in all animals studied (from 1.03 +/- 0.337 mg/dl in control animals to 3.49 +/- 1.01 mg/dl in fasted animals). Kinetic values of ICG disappearance were determined from single exponential functions, and those for BSP were determined from both single and curvilinear (2-exponential) functions. Plasma clearance of BSP in fed horses (8.65 +/- 1.02 ml X min-1 X kg-1) was greater than clearance of ICG (3.54 +/- 0.67 ml X min-1 X kg-1), results similar to those reported in dogs, cats, rats, and persons. Simultaneous measurement of the renal clearance of endogenous creatinine and exogenous inulin in eight sheep showed similar mean +/- SD (n = 32) values of 13.8 +/- 1.3 and 13.2 +/- 2.0 ml min-1 (10 kg)-1 bodyweight respectively. These results demonstrate that the renal clearance of endogenous creatinine is a satisfactory measure of glomerular filtration rate (GFR) in sheep. The plasma concentrations of endogenous creatinine and urea were significantly higher because of haemoconcentration during summer, resulting in lower GFR than in winter. Besides glomerular filtration and back diffusion, the renal handling of urea in sheep seems to involve mechanisms analogous to active tubular secretion.  相似文献

Recent evidence has linked alpha-receptor and beta-receptor activations with ventricular arrhythmia genesis. In order to assess the relative contribution of specific adrenoceptors (alpha 1, alpha 2, beta 1) on ventricular arrhythmogenic activity during xylazine (1.1 mg X kg-1 X hr-1)-halothane (1.35%) anesthesia, the arrhythmogenic dose of epinephrine (ADE) was repeatedly determined before and after prazosin (alpha 1 antagonist; 0.1 mg X kg-1), metoprolol (beta 1 antagonist; 0.5 mg X kg-1), and yohimbine (alpha 2 antagonist; 0.125 mg X kg-1) administration in 6 dogs. The ADE was expressed as infusion rate and total dose. The ADE was defined as the dose which produced 4 or more intermittent premature ventricular contractions within 15 s during a 3-minute infusion period or within 1 minute from end of infusion. Control ADE was 2.69 +/- 0.372 (micrograms X kg-1 X min-1) and 4.17 +/- 0.544 (micrograms X kg X -1) for infusion rate and total dose, respectively. The ADE significantly increased after prazosin (P less than 0.005), metoprolol (P less than 0.005), and yohimbine (P less than 0.05) administration. The ADE values increased to 5.42 +/- 1.22 (rate) and 8.10 +/- 1.95 (dose) after alpha 2 blockade, but were significantly less than the alpha 1 and beta 1 blockade ADE values. In conclusion, although both alpha- and beta-adrenoceptor blockade depressed ventricular arrhythmia genesis in xylazine-halothane-anesthetized dogs, alpha 2 blockade, which was achieved with the recommended dose of yohimbine for reversal of anesthetic-induced CNS depression, was not as protective as alpha 1 (prazosin) or beta 1 (metoprolol) blockade.  相似文献

The oral and intravenous disposition of the anti-thyroid drug propylthiouracil (PTU) was determined in six clinically healthy cats and four cats with naturally occurring hyperthyroidism. Compared with the normal cats, the mean plasma elimination half-life of PTU was significantly (P less than 0.001) shorter in the hyperthyroid cats (77.5 +/- 5.8 minutes compared with 125.5 +/- 3.7 minutes) and the total body clearance of PTU was significantly (P less than 0.05) more rapid in the cats with hyperthyroidism (5.1 +/- 0.8 ml kg-1 min-1 compared with 2.7 +/- 0.2 ml kg-1 min-1). Following oral administration, both the bioavailability (59.7 +/- 4.9 per cent compared with 73.3 +/- 3.7 per cent) and peak plasma concentrations (14.5 +/- 1.6 micrograms ml-1 compared with 18.9 +/- 0.9 micrograms ml-1) of PTU were significantly (P less than 0.05) lower in the hyperthyroid cats than in the control cats. No difference was noted, however, between the apparent volume of distribution for PTU in the two groups of cats. Overall, results of this study indicate that the oral bioavailability of PTU is decreased and PTU disposition is accelerated in cats with hyperthyroidism.  相似文献

Pharmacokinetics and renal clearance of ampicillin were investigated in 13 sheep, following one single oral dose of 750 mg. A peak concentration in plasma 0.38 +/- 0.04 microgram/ml (mean +/- SEM) was achieved 95.3 +/- 5.95 min after drug administration. Absorption half-life was 44.4 +/- 4.4 min. The area under the plasma concentration curve was 94.6 +/- 4.5 micrograms.hour.ml-1, while in the case of urine it was 370.5 +/- 28.3 micrograms.hour.ml-1. Biological half-life of ampicillin was 110 +/- 3 min, with an elimination rate constant of 0.0064 +/- 0.0002 min-1. The values for volume of distribution and total body clearance were 8.2 +/- 0.71/kg or 52.0 +/- 4.2 ml/kg/min, respectively. The priming and maintenance doses, using MIC as 0.05 microgram/ml, were suggested to be 8.8 or 8.4 mg/kg, respectively, at an 8-h interval. For MIC of 0.5 microgram/ml, this dose should be 10 times higher. Renal clearance of ampicillin seemed to involve active tubular secretion. Renal excretion indicated either extensive metabolism or excretion through routes other than kidneys.  相似文献

Clinical pathologic alterations in horses during a water deprivation test   总被引:1,自引:0,他引:1  
A 72-hour water deprivation test was performed in 12 horses to determine clinical pathologic changes. Reference values for electrolyte (X) clearance, expressed as a percentage of creatinine clearance (CLCR; %CLCRX), were also determined. A comparison was made between urine concentration measurement techniques. Results of %CLCRX determination in 12 horses before water deprivation were 0.034 +/- 0.095 %CLCRNa, 42.4 +/- 9.8 %CLCRK, 0.352 +/- 0.190 %CLCRCl, and 0.710 +/- 0.250 %CLCRP. During water deprivation, there was individual variation for electrolyte clearances, but Na excretion increased significantly (P less than 0.01) at 24 and 48 hours. After 48 hours' water deprivation, %CLCRNa decreased significantly, but was still greater than the initial clearance. Plasma protein was a better indicator of water deprivation (dehydration) in the horse than was PCV. Electrolyte concentrations in serum and urine were determined. Little significant (P less than 0.01) change in acid-base values was noticed after 72 hours' water deprivation. Urine osmolality (as determined by osmometry) was compared with sp gr (determined by refractometry) in determining urine concentration. Initially, sp gr correlated well with urine osmolality determinations, but this correlation decreased after 48 hours.  相似文献

Plasma disappearance of creatinine as a renal function test in the dog   总被引:2,自引:0,他引:2  
The serum concentration of creatinine at 120 minutes (SC120) after intravenous injection of 88 mg kg-1 of creatinine, the plasma half-life (t1/2) and the plasma clearance of creatinine (PCC) were evaluated as renal function tests in 30 healthy adult dogs and six adult dogs with known or suspected renal disease. The mean SC120 in the normal dog was 0.31 +/- 0.08 mmol litre-1 and in the clinical cases 0.71 +/- 0.19 mmol litre-1. The correlation coefficients between SC120 and renal creatinine clearance (RCC) for the normal dogs and the clinical cases were -0.76 and -0.69, respectively. At 120 minutes after injection, 95 per cent of normal dogs would be predicted to have a serum creatinine concentration below 0.46 mmol litre-1. The mean plasma t1/2 of creatinine for the normal dogs was 107.7 +/- 17.96 minutes, while the clinical cases had a wide range of values (148.8 to 620.1 minutes). Plasma t1/2 of creatinine was correlated with RCC for both the normal dogs and the clinical cases (r = -0.55, r = -0.91, respectively). The mean PCC for the normal dogs was 7.42 +/- 2.22 ml min-1 kg-1 (range 4.95 to 13.28 ml min-1 kg-1). There was a good correlation between RCC and PCC (r = 0.7). The PCC for the clinical cases ranged from 0.76 to 3.37 ml min-1 kg-1. The correlation between RCC and PCC was significant (r = 0.91). Thus SC120, t1/2 and PCC may be useful methods of assessing renal function in dogs with renal impairment insufficient to cause azotaemia.  相似文献

The pharmacokinetics of amikacin sulphate were investigated in calves and sheep. Five animals of each species were given 7.5 mg kg-1 intravenously and intramuscularly. After intravenous administration the pharmacokinetic parameters significantly different (P less than 0.01) between calves (first value) and sheep (second value), were: the initial concentration (87.05, 146.6 micrograms ml-1), the apparent distribution volume (350, 200 ml kg-1), the area under curve (5512, 11,018 min micrograms ml-1) and the clearance (1.5, 0.7 ml min-1 kg-1). After dosing intramuscularly the peak concentration (23.5, 34.36 micrograms ml-1), the peak time (45, 75 min) and the area under curve (5458, 9191 min micrograms ml-1) were significantly different (P less than 0.01). No significant differences were observed in the terminal halflife values, suggesting that elimination rate was independent of both route of administration and animal species. The drug in aqueous solution showed a good bioavailability in both animal species (about 0.87 in sheep and greater than 0.99 in calves) despite the greater serum concentrations always attained in sheep.  相似文献

The disappearance of IV injected [24-14C]cholic acid from plasma was studied in normal and mutant Corriedale and Southdown sheep exhibiting hereditary defects in hepatic organic anion transport. Hepatic cholic acid clearance was determined from the integral of the 40-minute disappearance curves fit to the sums of two exponential functions. Cholic acid clearance among Corriedale sheep was significantly less (P less than 0.05) for mutant sheep (8.44 +/- 0.86 SEM ml/minute/kg of body weight) than for normal sheep (12.7 +/- 0.58 ml/minute/kg). Cholic acid clearance in the Southdown mutant (1.97 +/- 0.59 ml/minute/kg) was less than 15% of normal clearance rate (13.3 +/- 2.2 ml/minute/kg). Clearance of [14C]taurocholic acid (curves fit to three exponential function) in the Southdown mutant (10.8 +/- 0.4 ml/minute/kg) was significantly greater (P less than 0.01) than cholic acid clearance, yet was not significantly different (P greater than 0.05) from normal taurocholate clearance (17.8 +/- 2.5 ml/minute/kg). Hepatic regurgitation of conjugated bile acid was not detected after [14C]cholic acid injection. Both the mutant Corriedale and Southdown sheep, which exhibited inherited defects in hepatic bilirubin transport similar to Dubin-Johnson syndrome and Gilbert's disease in man, exhibited defects in hepatic bile acid clearance.  相似文献

Nine N metabolism trials were conducted in a N-depletion-repletion regimen using five breed crosses of steers with 0, 25 or 50% Brahman. Three steers from each breed cross (avg initial wt 239 kg) were standardized for 3 wk on an 18% protein diet and depleted for 5 wk on a 7.5% protein diet and then repleted for 4 wk on the 18% protein diet. Nitrogen balance and blood and urinary N metabolites were monitored to determine changes in metabolic status due to changes in dietary N intake. Reduced N intake during the depletion period almost completely inhibited fiber digestion (P less than .01), while ruminal ammonia concentration fell from an average concentration of 156 during standardization to 22 mg/liter (P less than .01) during depletion. Organic matter digestion was depressed (P less than .05) due to the reduced fiber digestion. Absolute amounts of intake, fecal, urinary, absorbed and retained N were severely depressed during depletion. Apparent digestibility of N was reduced from an average of 68% during standardization to 42% (P less than .05) during depletion, but truly digested N was not affected. Nitrogen retained, as a percentage of intake, or absorbed was reduced only during the first week of depletion. Urea N comprised the major identified urinary fraction during standardization and repletion, whereas creatinine was the major fraction during depletion. Brahman-cross steers excreted more urinary N (P less than .1) and retained less N (P less than .13) during repletion than Angus X Hereford steers. Creatinine excretion was also higher (P less than .01) during depletion for Brahman-cross steers. Blood urea and albumin N concentrations were higher (P less than .05) during both depletion and repletion among Brahman crosses. These experiments suggest that blood measurements were more sensitive than N balance to changes in dietary N state and that faster recharge of blood N measurements in Brahman cattle may help mitigate adverse effects of N depletion stress.  相似文献

Four consecutive 6-hour urine sample collections were performed on 7 healthy adult Holstein cows fed a diet of coastal Bermuda hay with ad libitum water consumption. Urine (via indwelling urinary catheter) and venous blood samples were collected at 6, 12, 18, and 24 hours. Total 24-hour urine production for the 7 cows ranged from 4,515 to 7,130 ml/d (mean +/- SD, 5,633 +/- 946 ml/d) or 0.02 to 0.04 ml/kg of body weight/d (mean +/- SD, 0.03 +/- 0.007 ml/kg/d). Renal clearance (C) of creatinine (Cr), sodium (Na), calcium (Ca), and magnesium (Mg) varied significantly (P less than 0.05) among individuals, but did not vary significantly among the four 6-hour collection periods. Clearance of chloride (Cl) and phosphorous (P) did not vary significantly either among individuals or among the four 6-hour periods. Clearance of potassium (K) varied significantly (P less than 0.05) among individuals and among the four-6-hour periods. Creatinine clearance was significantly (P less than 0.01) correlated with C(Cl), C(Ca), C(P), and C(Mg) when all data were considered. Significant (P less than 0.05) correlations were also found between C(Cl) and C(K), C(Ca), C(P), and C(Mg); between C(Ca) and C(P) and C(Mg); and between C(P) and C(Mg). Fractional excretion (FE) of Na, K, Cl, Ca, P, and Mg did not vary significantly among the four 6-hour periods. Kochia hay that had elicited mild toxicosis in sheep and cattle when fed for 21 d was fed to young, growing steers to evaluate the efficacy of metoclopramide for preventing kochia toxicosis. Twelve steers (271 +/- 39 kg) were given ad libitum access to Kochia hay for 38 d. Six steers were dosed orally with 15 mg of metoclopramide/kg of BW three times each week in gelatin capsules, and six steers received empty gelatin capsules. Steers were housed in individual outside pens for the first 28 d, then inside in metabolism stalls for the last 10 d. Kochia intake averaged 1.2 +/- .04 and 1.1 +/- .05% of BW throughout 38 d for kochia-fed control and kochia plus metoclopramide-dosed steers, respectively. During the last 10 d, metoclopramide had no effect (P greater than .15) on digestibility of DM, OM, NDF, ADF, or CP. However, metoclopramide reduced N retention (P less than .01; 20 vs 8 g/d). Kochia hay decreased serum prolactin and insulin concentrations (P less than .01) from 12.4 to 1.5 ng/ml and from .53 to .23 ng/ml, respectively. Metoclopramide had no effect (P greater than .50) on prolactin or insulin in steers fed kochia hay. Serum growth hormone was not affected by kochia but was suppressed by metoclopramide in steers fed kochia hay (P less than .07). Serum bilirubin (total and unconjugated) was elevated (P less than .05), indicating early, mild hepatotoxicosis characteristic of kochia toxicosis. The arrhythmogenic dose of epinephrine (ADE) was determined in 6 pigs during steady-state anesthesia (1.5% halothane in O2) and steady-state anesthesia plus xylazine (1.1 mg X kg-1 X hr-1; IV infusion) and after either prazosin (alpha 1) or metoprolol (beta 1) adrenergic blockade during halothane-xylazine (H-X) anesthesia. A constant infusion (1, 2, 3, 5, and 10 micrograms X kg-1 X min-1) of freshly mixed epinephrine (100 micrograms X ml-1 in saline solution) was used to determine ADE. The ADE was defined as the total dose of epinephrine which produced 4 or more continuous or intermittent, premature, ventricular contractions within a 15-s period. The mean epinephrine total dose values during 1.5% halothane anesthesia, H-X anesthesia alone, or H-X anesthesia after either prazosin (0.1 mg X kg-1) or metoprolol (0.5 mg X kg-1) adrenergic blockade were 3.60 +/- 0.844, 2.68 +/- 0.402, 11.85 +/- 3.804, and 5.17 +/- 0.587 micrograms X kg-1, respectively. Xylazine administration did not significantly decrease ADE, although mean arterial pressure significantly increased. Prazosin administration significantly increased ADE and was associated with an increased heart rate and a decreased mean arterial pressure. We conclude that alpha 1-blockade with prazosin is more protective to epinephrine-induced arrhythmias in H-X-anesthetized pigs than is beta 1-blockade with metoprolol.  相似文献

Urea recycling in ruminants has been studied extensively in the past, but the mechanisms regulating the amount of urea recycled or excreted remain obscure. To elucidate the role of urea transporters (UT) in N recycling, nine Dorset-Finn ewe lambs (20.8 +/- 0.8 kg) were fed diets containing 15.5, 28.4, and 41.3 g of N/kg of DM for 25 d. Nitrogen balance and urea N kinetics were measured during the last 3 d of the period. Animals were then slaughtered and mucosa samples from the rumen, duodenum, ileum, and cecum, as well as kidney medulla and liver, were collected. Increasing N intake tended to increase N balance quadratically (1.5, 5.1, and 4.4 +/- 0.86 g of N/d, P < 0.09), and linearly increased urinary N excretion (2.4, 10, and 16.5 +/- 0.86 g N/d, P < 0.001) and plasma urea N concentration (4.3, 20.3, and 28.4 +/- 2.62 mg of urea N/dL, P < 0.001), but did not affect fecal N excretion (5.0 +/- 0.5 g of N/d; P < 0.94). Urea N production (2.4, 11.8, and 19.2 +/- 0.83 g of N/d; P < 0.001) and urinary urea N excretion (0.7, 7.0, and 13.4 +/- 0.73 g N/d; P < 0.001) increased linearly with N intake, as well as with the urea N recycled to the gastrointestinal tract (1.8, 4.8, and 5.8 +/- 0.40 g of N/d, P < 0.001). No changes due to N intake were observed for creatinine excretion (518 +/- 82.4 mg/d; P < 0.69) and clearance (46 +/- 10.7 mL/min; P < 0.56), but urea N clearance increased linearly with N intake (14.9, 24.4, and 34.9 +/- 5.9 mL/min; P < 0.04). Urea N reabsorption by the kidney tended to decrease (66.3, 38.5, 29.1 +/- 12.6%; P < 0.06) with increasing N content of the diet. Increasing the level of N intake increased linearly the weight of the liver as a proportion of BW (1.73, 1.88, and 2.22 +/- 0.15%, P < 0.03) but only tended to increase the weight of the kidneys (0.36, 0.37, and 0.50 +/- 0.05%, P < 0.08). Urea transporter B was present in all the tissues analyzed, but UT-A was detected only in kidney medulla, liver, and duodenum. Among animals on the three diets, no differences (P > 0.10) in UT abundance, quantified by densitometry, were found. Ruminal-wall urease activity decreased linearly (P < 0.02) with increasing level of N intake. Urease activity in duodenal, ileal, and cecal mucosa did not differ from zero (P > 0.10) in lambs on the high-protein diet. In the present experiment, urea transporter abundance in the kidney medulla and the gastrointestinal tract did not reflect the increase in urea-N reabsorption by the kidney and transferred into the gut.  相似文献

Sheep of the Merino breed, given 10.11 g nitrogen daily (after Czechoslovak Standard CSN 46 7007) were studied for the effect of the expansion of the extracellular fluid (ECF) by an infusion of an isotonic solution of NaCl (at a rate of 5% of live weight), as exerted on the renal excretion of urea and on the sodium and potassium electrolytes. The high natriuresis, induced by the expansion of ECF, resulted in a statistically significant increase in the clearance of urea, free water, and solutions, and in higher diuresis. An increase was observed in the excretion of urea with urine (UureaV; P < 0.005) and in the excretion of the studied electrolytes of sodium (UNaV; P < 0.001) and potassium (UKV; P < 0.005) by the same route. No variation was recorded in the clearance of inulin as a measure of the glomerular filtration rate (GFR). The fractional excretion of urea, sodium, and potassium also showed a significant increase. The results imply that the fractional excretion of urea increases under the conditions of artificially induced natriuresis.  相似文献
