Summary Simple sequence repeat (SSR) markers linked to quantitative trait loci (QTL) associated with resistance to sorghum shoot fly, Atherigona soccata resistance were used to characterize the genetic and phenotypic diversity of 12 cytoplasmic male-sterile (CMS) and maintainers, 12 restorer lines, and 144 F1 hybrids. The genetic diversity was quite high among the shoot fly-susceptible parents and the hybrids based on them, as indicated by high polymorphic information content (PIC) values, while limited genetic diversity was observed among shoot fly-resistant lines. The phenotypic and genotypic dissimilarity analysis indicated that the shoot fly-resistant and -susceptible parents were 73.2 and 38.5% distinct from each other, and the morphological and genetic distances of certain resistant and susceptible cross combinations was more than their resistant or susceptible parents. Genetic variability among the groups was low (10.8%), but high within groups (89.2%). The genetic and morphological distances suggested that the F1 hybrids were closer to CMS (5 to 12% dissimilar) than the restorer (11 to 87% dissimilar), suggesting that CMS influences the expression of resistance to sorghum shoot fly. The SSR markers can be used to characterize the homologous traits in sorghum germplasm.  相似文献

The spotted stem borer, Chilo partellus, is one of the most important pests of sorghum, and host plant resistance is an important component for the management of this pest. Most of the sorghum hybrids currently under cultivation are based on cytoplasmic male-sterility (CMS). In order to develop a strategy for resistance to stem borer, we studied the traits associated with resistance, and their nature of gene action in F1 hybrids derived from resistant, moderately resistant, and susceptible CMS and restorer lines. The hybrids based on stem borer-resistant, moderately resistant, or susceptible CMS and restorer lines were equally resistant or susceptible as the parents for leaf feeding [Damage rating (DR) 5.8 to 6.6 vs. 5.9 to 6.6], and had significant and decreasing trend in deadheart formation (resistant CMS × resistant restorer lines < moderately resistant CMS × moderately resistant restorer lines < susceptible CMS × susceptible restorer lines), respectively. Proportional contributions of restorer lines were greater than those of the CMS lines for leaf feeding, deadhearts, recovery and overall resistance, stalk length, nodes per plant, stem borer holes per plant, and peduncle tunneling. The general (GCA) and specific combining ability (SCA) estimates suggested that leaf feeding score, number of nodes, overall resistance score, panicle initiation, recovery score, and stalk length (dominance type of gene action) have been found to be associated with resistance to spotted stem borer, governed by additive type of gene action, their correlation and direct effects in the same direction, and explained 65.3% of the variation in deadhearts, and thus could be used as marker traits to select and breed for resistance to C. partellus in sorghum. The parents having significant SCA effects for two or more resistance traits for either or more parents have also been discussed for their use in the stem borer resistance breeding.  相似文献

Summary The shoot fly, Atherigona soccata is an important pest of sorghum, and host plant resistance is one of the most effective components for managing this pest. Most of the hybrids grown in India based on milo cytoplasm (A1 cytoplasm) are highly susceptible to shoot fly. Therefore, the present studies were undertaken to evaluate different male-sterile cytoplasms (CMS) for their relative susceptibility to sorghum shoot fly. Oviposition and deadheart formation were significantly lower on the maintainer lines as compared to the corresponding male-sterile lines. Among the cytoplasms tested, A4M cytoplasm showed antixenosis for oviposition and suffered lower deadheart formation than the other cytoplasms tested. The A4G1 and A4M cytoplasms suffered lower deadhearts in tillers than the other cytoplasms. Recovery following shoot fly damage in A4M, A3, and A2 cytoplasms was better than in the other cytoplasms tested. The larval and pupal periods were longer and male and female pupal weights lower in A4M and A4VzM CMS backgrounds compared to the other CMS systems. Fecundity and antibiosis indices on CMS lines were lower than on the B-lines. The A4M cytoplasm was found to be relatively resistant to sorghum shoot fly, and can be exploited for developing shoot fly-resistant hybrids for sustainable crop production in future.  相似文献

Sorghum midge, Stenodiplosis sorghicola (Coquillett) is one of the most important pests of grain sorghum worldwide. We studied the inheritance of resistance to sorghum midge and compensation in grain weight and volume in panicles of sorghum hybrids and their parents under uniform infestation (40 midges per panicle for two consecutive days). Sorghum midge damage ranged from 8.2 to 82.4% in the maintainer lines (B-lines) of the females parents (A-lines), and 9.0 to 67% in the male parents (restorer lines). Hybrids involving resistant × resistant parents were highly resistant, while those involving resistant ×susceptible and susceptible × resistant parents showed moderate susceptibility. Susceptible × susceptible hybrids were susceptible. Compensation in (percentage increase) grain weight and volume in midge-infested panicles of midge-resistant parents and their F1 hybrids was greater than in midge-susceptible parents and hybrids. General combining ability effects for midge damage, and grain weight and volume were significant and negative for the midge-resistant females (ICSA 88019 and ICSA 88020), whereas those for the midge-susceptible females (ICSA 42 and 296A) were significant and positive. However, the reverse was true in case of compensation in grain weight and volume. Inheritance of compensation in grain weight and volume and resistance to sorghum midge is controlled by quantitative gene action with some cytoplasmic effects. One of the reasons of poor nutritive value of sorghum grain is resistance of its seed storage proteins (kafirins) to protease digestion. To reveal sorghum entries with increased kafirin digestibility, the sodium dodecyl sulfate – polyacrylamide gel electrophoresis (SDS‐PAGE) of endosperm proteins of 10 lines [cytoplasmic male sterility (CMS)‐lines and fertility restorers] and five F1 hybrids before and after pepsin digestion was carried out. For quantitative estimation of kafirin digestibility the SDS‐PAGE banding patterns were scanned by laser densitometer. Significant variability for both individual fractions and total kafirin digestibility was found. The line KVV‐45, fertility restorer for the Indian ‘M35‐1A’ type of CMS, had the highest level of kafirin digestibility (30% and 25% of undigested γ‐ and α1‐kafirins, respectively), while in some entries 80–90% of kafirins remained undigested. Increased α1‐kafirin digestibility coincided with relatively high γ‐kafirin digestibility. High‐molecular weight kafirins (HMWK) (45 kDa and 66 kDa) resistant to pepsin digestion were found in some lines, the F1 hybrids had the same HMWK as parental lines. These data demonstrate possibility for isolation of sorghum genotypes with increased nutritive value by screening their flour for in vitro protein digestibility.  相似文献

Sorghum shoot fly, Atherigona soccata is an important pest of sorghum during the seedling stage, which influences both fodder and grain yield. To understand the nature of inheritance of shoot fly resistance in sorghum, we performed generation mean analysis using two crosses IS 18551 × Swarna and M 35-1 × ICSV 700 during the 2013–2014 cropping seasons. The F1, F2, BC1 and BC2 progenies, along with the parental lines were evaluated for agronomic and morphological traits associated with resistance/susceptibility to sorghum shoot fly, A. soccata. The cross IS 18551 × Swarna exhibited significant differences between the parents for shoot fly deadhearts (%) in the postrainy season. The progenies of this cross exhibited lower shoot fly damage, suggesting that at least one of the parents should have genes for resistance to develop shoot fly-resistant hybrids. Leaf glossiness, leafsheath pigmentation and plant vigor score during the seedling stage exhibited non-allelic gene interactions with dominant gene action, whereas 100 seed weight showed both additive and dominant gene interactions. Presence of awns showed recessive nature of the awned gene. Generation mean analysis suggested that both additive and dominance gene effects were important for most of the traits evaluated in this study, but dominance had a more pronounced effect.  相似文献

Wide crosses were made to identify new cytoplasmic male sterility (CMS) systems in faba beans, based on the interaction of cytoplasm with restorer and maintainer alleles. A total of 330 F1 hybrids were produced in both reciprocal forms. Male sterile segregates were observed in one reciprocal version in the F2 generation of six crosses. Two of these crosses with female parents originating from Afghanistan and Egypt expressed stable male sterility in subsequent backcross generations. Based on the female parents of the two crosses, these two CMS systems were designated CMS 199 and CMS 297. CMS 199 was more stable than CMS 297 during backcross generations and across different environments. Maintainer and restorer lines for both CMS systems were identified. Lower expression of male sterility occurred in CMS 297 in the greenhouse during the winter generations than in isolation cages during the summer generations, which may be utilized to maintain male sterile lines by selfing. Regarding practical applications, the CMS 199 shows great promise for hybrid breeding in faba beans.  相似文献

The stability of biochemical constituents and their association with resistance to shoot fly (Atherigona soccata Rondani) was evaluated for reducing sugars, total sugars, nitrogen, phosphorus, potassium, chlorophyll and moisture contents at weekly intervals of seedling growth (7, 14, 21 and 28 days after emergence) in 14 selected grain sorghum genotypes [five resistant accessions (IS nos. 1054, 2146, 2312, 3962 and 4664); three susceptible checks (CK 60B, CSV 1 and CSH 1); one national variety (CSV 8R); and five post-rainy advanced generation (F6) breeding lines (148 × CS 3541, SPV 103 × IS 4664, CSV 8R × SPV 104, SPV 104 × M 35-1, and PD 3-1-11 derivative)]. The genotypes IS 2312 and IS 4664 showed stability of antixenosis for oviposition during post-rainy season advanced generation lines compared to the susceptible checks. Deadheart formation was low and the expression of resistance was stable across different seedling growth stages in IS 1054 and IS 2146. Depletion in levels of reducing sugars and phosphorus in resistant genotypes played a significant role in deadheart formation in the test genotypes. Positive association of nitrogen and potassium with oviposition at early seedling stages indicated their role in releasing chemical cues for oviposition. Low levels of reducing sugars and total sugars seemed to enhance the degree of resistance to sorghum shoot fly. The total chlorophyll content had no relationship with antixenosis for oviposition. No relationship was observed between moisture content of sorghum seedlings and shoot fly resistance. Low concentrations of reducing sugars, total sugars, nitrogen, phosphorus and potassium in sorghum seedlings greatly enhanced the degree of antixenosis for oviposition/feeding and deadheart formation, and can be used as selection criteria for resistance to shoot fly.  相似文献

Summary Ten cytoplasmic male sterile (CMS) sunflower (Helianthus annuus L.) lines were crossed with nine maintainer or male fertility restorer lines in a diallel crossing scheme. Based on fertility restoration of the F1 generation, CMS lines were divided into four groups. At least two new sources of CMS, CMS PET2 and CMS GIG1, were found to be potentially useful for commercial production of hybrids. Environment had an influence on fertility restoration of one CMS line, CMS MAX1. Effective restoration of male fertility for CMS RIG1, CMS ANN2, and CMS ANN3 was not found.  相似文献

选用农家种改良过的6个食葵细胞质雄性不育系和4个优良的分枝型恢复系为亲本,按NCⅡ遗传交配设计,配成24个F1杂种,研究了9个主要经济性状,即株高、茎粗、生育期、盘径、单盘总粒数、单盘实粒数、结实率、百粒重、单株产量的配合力及杂种优势表现。结果显示:(1)不育系中有8个性状,恢复系中有7个性状的一般配合力差异达显著或极显著水平;有4个性状的特殊配合力差异达到显著或极显著水平;(2)株高、茎粗、生育期、盘径、百粒重等5个性状主要受基因的加性效应控制;单盘总粒数、单盘实粒数、结实率、及单株产量等4个性状则受基因的加性效应和非加性效应共同影响;(3)从GCA分析看,参试的不育系以A6最为理想,A2次之;恢复系则以C3最为理想。从SCA分析看,组合A2C1单株产量效应值最高;(4)杂种优势分析表明,大部分经济性状存在杂种优势,甚至相当一部分存在超亲优势,并且这些优势都是可利用的,F1多倾向于早熟、粗壮、高杆、大盘、多粒亲本。  相似文献

Sorghum shoot fly, Atherigona soccata, is one of the important pests of postrainy season sorghums. Of the 90 sorghum genotypes evaluated for resistance to this pest, RHRB 12, ICSV 713, 25026, 93046 and 25027, IS 33844‐5, Giddi Maldandi and RVRT 3 exhibited resistance in postrainy season, while ICSB 463, Phule Anuradha, RHRB 19, Parbhani Moti, ICSV 705, PS 35805, IS 5480, 5622, 17726, 18368 and 34722, RVRT 1, ICSR 93031 and Dagidi Solapur showed resistance in rainy season, suggesting season‐specific expression of resistance to A. soccata. ICSB 461, ICSB 463, Phule Yasodha, M 35‐1, ICSV 700, 711, 25010, 25019 and 93089, IS 18662, Phule Vasudha, IS 18551 and 33844‐5 and Barsizoot had fewer deadhearts than plants with eggs across seasons, suggesting antibiosis as one of the resistance mechanism. Five genotypes exhibited resistance with high grain yield across seasons. Correlation, path and stepwise regression analyses indicated that leaf glossiness, seedling vigour, trichome density, oviposition and leaf sheath pigmentation were associated with the expression of resistance/susceptibility to shoot fly, and these can be used as marker traits to select and develop shoot fly‐resistant sorghums.  相似文献

Summary Possible unfavourable influence of sterility inducing cytoplasm on physico-chemical grain quality traits in rice hybrids is one of the important concerns hindering the large-scale adoption of hybrid rice technology. Produce from 23 pairs of CMS line × restorer (AF1) and maintainer line × restorer (BF1) cross combinations carrying different cyto-sterile sources (WA, ARC, Mutagenized IR 62829B and Kalinga I) was compared for various grain and cooking quality traits. The milling recovery in rice hybrids was not influenced by the sterile cytoplasm. For kernel dimensions before and after cooking there were both favourable and unfavourable cytoplasmic effects, which varied in magnitude depending upon the sterile cytoplasm and parental combinations. Similar results were obtained for kernel elongation and gelatinization temperature. The most widely used WA cytoplasm had minimum instances of unfavourable influence. In general, the cytoplasmic influence was found to be highly cross-specific and depended on the nuclear background of CMS line and fertility restorer. Availability of alloplasmic CMS lines carrying different cyto-sterile sources in the same nuclear background would help in ascertaining the cytoplasmic influence in a more comprehensive manner.  相似文献

Four bacterial blight (BB) resistance genes, Xa7, Xa21, Xa22 and Xa23, were introgressed into an elite hybrid rice restorer line Huahui 1035, by marker-assisted selection (MAS). Ten promising BB resistant lines identified by MAS approach in Huahui 1035 restorer background and their respective F1 hybrids with a cytoplasmic male sterile line i.e. Jinke 1A, three BB resistant gene donors and their recurrent parent were evaluated for BB resistance by inoculating them with eleven representative races of Xanthomonas oryzae pv. oryzae (Xoo) in China. Further, the ten marker assisted selection of BB (MAS-BB) resistant restorer lines and their F1 hybrids were also characterized for agro-morphological traits and grain yield. Results revealed that restorer lines with Xa23 introgression i.e. HBQ809 and HBQ810 were found to be resistant to all eleven Chinese representative Xoo races, showing broad spectrum resistance to BB. However, the lines possessing Xa7 or Xa7+Xa21 were resistant to ten out of eleven Xoo races. While restorer lines with Xa21 were resistant to nine out of eleven Xoo races. Interestingly, the F1 hybrids with Xa23, Xa7 or Xa7+Xa21 were resistant to two severe epidemic Xoo races of China. Restorer lines i.e. HBQ807 and HBQ808 possessing Xa22 were only resistant to six or five out of inoculated eleven Xoo races. Lesion length comparisons between restorer lines and their F1 hybrids for the above four BB resistance genes showed them to be dominant in heterozygote genotype. Three promising high yielding F1 hybrids with BB resistance were identified for immediate exploitation for hybrid rice production in China.  相似文献

Non‐pungent bell pepper (Capsicum annuum L.) lacks the cytoplasmic male sterility (CMS) nuclear restorer allele, Rf, and CMS cannot be employed in its F1 hybrid seed production. To demonstrate that the genic male sterility (GMS) system in non‐pungent bell pepper can be converted to the CMS male sterility system, the conversion of GMS to CMS for non‐pungent bell pepper line GC3 was conducted by introgression of S‐type cytoplasm and the Rf allele from tropical pungent donors. After morphological traits were evaluated, two lines from BC1F1 containing S‐type cytoplasm and four lines from BC2F2 containing Rf allele, phenotypically similar to GC3, were obtained and could be employed as CMS male sterile lines and restorer lines for non‐pungent bell pepper. Four molecular markers potentially linked to traits of interest were also evaluated in BC1F1 and BC1F2 populations. This is the first time that GMS has been successfully converted to CMS in bell pepper, a significant contribution for bell pepper hybrid seed production.  相似文献

Sorghum midge, Stenodiplosis (Contarinia) sorghicola (Coquillett), is an important pest of grain sorghum, and host plant resistance is an important aspect of control of this pest. This research investigated how cytoplasmic male-sterility and source of pollen influence the expression of resistance to sorghum midge. Sorghum midge emergence was significantly lower in panicles of midge-resistant and midge-susceptible cytoplasmic male-sterile lines when pollinated with AF 28 - a midge-resistant restorer line, than those pollinated with Swarna - a midge susceptible restorer line, indicating the presence of xenia effects. Maintainer lines (B-lines) of midge-resistant parents had significantly lower numbers of eggs and larvae than the B-lines of midge-susceptible parents. Male-sterile lines of the both midge-resistant and midge-susceptible lines were equally susceptible, indicating that resistance to sorghum midge is influenced by factors in the cytoplasm of the B-line. These findings will have an important bearing on the production of hybrids with resistance to insects. Significant heterosis for seed yield in oilseed rape has created interest in the development of hybrid cultivars. The DNA‐based marker protocol, sequence‐related amplified polymorphism (SRAP) was used to determine genetic diversity among oilseed rape maintainer and restorer lines. This measure was used in an attempt to establish an association between genetic distance and heterosis in hybrids for various agronomic traits. A total of 118 polymorphic loci were generated by 18 SRAP primer combinations. Based on the polymorphism generated by the markers, calculated similarity index values ranged from 0.46 to 0.97. Cluster analysis grouped 10 maintainer and 12 restorer lines into three groups, with the exception of two maintainer lines, PM5 and PM9, which fell outside these groups. The grouping of the lines was largely in agreement with the available pedigree data on their origin and agronomic performance. Analysis of variance among inbred lines and their resulting F1 hybrids over two locations revealed significant differences for plant height, days to maturity and seed yield, but not for oil content. Substantial mid‐parent heterosis was observed only for seed yield, and ranged from 26% to 169%. All hybrids surpassed their respective inbred lines for this trait, except for a single cross combination of related lines. In general, crosses of lines located in different clusters yielded more than those from the same clusters. Regression analysis revealed a statistically significant relationship between the genetic distance of the parents and seed yield in their hybrid, and their derived mid‐parent and high‐parent heterosis. The correlation coefficient between genetic distance and yield (0.64) indicated a moderately strong relationship, so it is possible that some of the SRAP markers might be linked to quantitative trait loci for seed yield.  相似文献

Significant differences of callus induction, green plant regeneration and culture efficiency were observed among restorer lines and their hybrids in rice. Average callus induction percentage of F1, hybrids was about twice that of their parents. Twenty doubled haploid (DH) lines that showed normal fertility with both indica and japonica CMS lines, were selected as widely compatible restorers (WCRs). TG8 derived from CPSLO17/Minghui 63 not only showed normal fertility to both indica and japonica test varieties and CMS lines, but also exhibited superior agronomic traits, in particular short plant, desired plant type, high tillerability, large panicles, good grain quality and resistance to rice blast disease. Strong heterosis for yield was observed in crosses between indica or japonica CMS lines and TG8. TG8 possessed a dominant, widely compatible gene (WCG) that was inherited from the variety CPSLO17. Results confirmed that the anther culture technique is a quick and effective way of developing widely compatible restorers in rice and that it is applicable to the direct use of intersubspecific heterosis with the three-line method.  相似文献

利用大豆质核互作雄性不育系NJMCS3A的质、核供体亲本N21566和N21249构建F2和BC1F1育性分离群体进行雄性育性的遗传分析与基因定位。结果表明, F1正反交可育,F2和BC1F1的可育株与不育株分离比例经χ2测验分别符合3∶1和1∶1,表明NJCMS3A供体亲本雄性育性由一对基因控制,可育等位基因为显性。该基因可能是NJCMS3A的一个恢复基因。选用793对SSR引物对F2和BC1F1群体分别进行育性基因定位,发现该育性基因位于O连锁群上,在Satt331和Satt477标记之间,与Satt331、CSSR133和Satt477标记距离的次序一致,分别为8.1~10.4 cM、11.4~16.4 cM、13.3~19.2 cM。  相似文献

Cytoplasmic male sterility (CMS) is considered an efficient genetic tool in pearl millet hybrid breeding. Of the several CMS sources available in pearl millet, A1 is the only CMS widely exploited to produce commercial hybrids in India. To explore the possibility of using alternate CMS sources, we studied the cytoplasmic effects of different CMS sources on agronomic characters in pearl millet. Five CMS (A) lines representing A1, A2, A3, A4 and A5 cytoplasms, their respective maintainer (B) lines and eight restorer (R) lines were used to generate 40 A × R and B × R experimental crosses. The experimental material was evaluated at two different locations in India. Analysis of combining ability and heterosis revealed that A4 and A5 cytoplasms had desirable effects for earliness. The A5 CMS was found to be particularly promising, as compared to other CMS sources for improving grain yield. The study also indicated that the cytoplasmic effects on general combining ability (GCA) for various agronomic characters were largely non-significant. However, cytoplasmic effects on specific combining ability and heterosis were found to be modulated by cytoplasmic-nuclear interactions and influenced by the environmental conditions. The study also demonstrated the advantage of utilizing diverse male-sterile and restorer combinations in maximizing the productivity as well as for genetic and cytoplasmic diversification of hybrids in pearl millet.  相似文献

以84份不同类型高粱品种(系)为试验材料,研究高粱中胚轴伸长特性,并筛选不同中胚轴伸长特性种质,为进一步研究高粱出苗能力奠定基础。结果表明,所有参试材料中胚轴平均长度为6.5cm,不育系、保持系、杂交种、恢复系和地方品种中胚轴平均长度分别为4.7、5.1、5.8、7.0和8.4cm。不同类型高粱中胚轴伸长能力不同,其伸长能力表现为地方品种>恢复系>杂交种>不育系、保持系。以中胚轴长度为指标的UPGMA(类平均法)聚类分析结果显示,参试的84份种质可以分为5种类型,即极长中胚轴类型、长中胚轴类型、中胚轴长度中间类型、短中胚轴类型和极短中胚轴类型。杂交种中胚轴伸长的杂种优势属于负向中亲优势。  相似文献
