Embryo of lilacs (Syringa L) culture in vitro and the rapid propagation were studied. The orthogonal experiments, including the selection of basal medium, embryo age and other factors such as sugar, benzyladenine (BA), naphthalene acetic acid (NAA) and glutamine (Gln), were carried out. The results indicated that the optimal medium for embryo culture was Monnier medium supplemented with NAA (0.001 mg.L^-1), BA (0.1 mg.L^-1), sugar (50 g.L^-1), and Gin (400 mg.L^-1), with a germination rate of 91.7% at least; the optimal embryo age was 50 d; and Gln had significant effects on the germination rate of embryo.Moreover, the optimal medium for subculture was MS BA (2 mg.L^-1) NAA (0.001 mg.L^-1) Gln (0.5 mg.L^-1), with the propagation coefficient of 3.6 at least.  相似文献

For improving seed germination of Prosopis koelziana and Prosopis juliflora, different treatments of seeds were conducted, including scarification with sulfuric acid 98% for 10 and 15 min, sandy paper, hot water for 5 and 10 min, potasium nitrate 0.1%, gibberellic acid at 250 mg·L−1 and 500 mg·L−1 and combinational treatment of scarification with gibberellic acid of 250 mg·L−1 and 500 mg·L−1. The results show that scarifications with sandy paper and sulfuric acids 98% were the most effective treatments on breaking seed dormancy and seed germination induction. Scarification with sulfuric acid 98% for 15 min was the best treatment. According to the positive effect of scarification and lack of reaction of seeds against KNO3 and gibberellic acid, the kind of seed dormancy was determined as exogenous.  相似文献

Leaves of fine Populus tomentosa genotype TC152 were used as explants to establish cell suspension lines. The effects of plant growth regulators on callus induction and establishment of cell suspension lines were studied. The callus induction rate was the highest on a MS solid medium supplemented with 1.0 mg·L^-1 2,4-D. A cell suspension line could be obtained by inoculating calli which were not subcultured into a MS liquid medium supplemented with 1.5 mg·L^-1 2,4-D. The best subculture medium was MS + 0.8 mg'L-1 2,4-D + 30 g·L^-1 sucrose with a subculture cycle of seven days.  相似文献

Twigs of 2–3-year-old Platanus occidentalis L. were used as experimental material to find the causes for the contamination and browning in the initial stages of tissue cultures. To compare the degree of browning of explants picked off from different growing seasons, the experimental material was excised from trees on each of the first ten days in January, March, May and July, 2006. The results indicated that the contamination and browning rates of the material cut off in January (14.2% and 30.6%, respectively) and March were somewhat lower than those in July. The pretreatment of soaking the explants in different anti-oxidants and absorbents at the same time could diminish some side effects. The pretreatment of using 10 g·L–1 vitamin C reduced the contamination and brown-ing rate effectively. An orthogonal experiment showed that the optimal factor and level arrangement is 0.5 mg·L–1 BA, 2.0 g·L–1 ac-tive carbon and 1.5 g·L–1 PVP which resulted in a browning rate of only 16.5%. In general, sampling period, physical properties and pretreatment of explants are the main factors responsible for the contamination and browning of material in the initial stages of P. occidentalis tissue cultures.  相似文献

Cottonhead windhairdaisy (Saussurea laniceps Hand.-Mazz.) is one of the most famous and important medicinal herbs in China. Illegal collection from wild populations is increasingly threatening the present environment of S. laniceps. Establishment of an efficient method for micropropagation is the best way to change its endangered situation. When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supplemented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg•L–1 BA plus 0.2 mg•L-1 NAA. In the presence of 0.2 mg•L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization.  相似文献

Optimizing SSR-PCR system of Panax ginseng by orthogonal design   总被引:1,自引:0,他引:1  
An orthogonal design was used to optimize SSR-PCR amplification system using Panax ginseng genomic DNA as template. Four levels of five factors (DNA template, Taq DNA polymerase, Mg^2+, primer, and dNTP) and annealing temperature have been tested separately in this system. The results demonstrated the reaction efficiency was affected by these factors. Based on the results, a stable, productive and reproducible PCR system and cycling program for amplifying a ginseng SSR locus were obtained: 20 μL system containing 1.0 U Taq DNA polymerase, 2.0 mmol·L^-1 Mg^2+, 0.2 mmol·L^-1 dNTPs, 0.3 μmol·L^-1 SSR primer, 60 ng· μla^-1 DNA template, performed with a program of 94℃ for 5 min, 94℃ for 30 s, annealing at 56.3℃ for 30 s, 72℃ for 1 min, 37 cycles, finishing at 72℃ for 7 min, and storing at 4℃.  相似文献

The cutting seedlings ofLiriodendron chinense xtulipifera were treated with the different concentrations of auxin (treatment1: IBA of 50 g·kg−1+NAA of 300 g·kg−1; treatment2; IBA of 100 g·kg−1+NAA of 300 g·kg−1). The biomass nutrient element contents for different organs (root, stem, leaf) of cutting seedling ofLiriodendron chinense xtulipifera were measured by the dry method, Kjeldahl method and Atomic Absorption Spectroscopy method. The result showed that the biomass of root, stem, and leaf of the cutting seedling treated with auxin was all remarkably increased. The contents of element C in root, stem and leaf had no significant difference between the control and auxin treatments, while the contents of N, P, K and Ca in stem were much lower than that in leaf and root. Variance analysis showed that for the same organ with different concentration treatment of auxin, the four nutrient elements (N, P, K, and Ca) had no significant difference in contents, while there existed significant or very significant difference in contents of the four nutrient elements in different organs with the same concentration auxin treatment. The N, P, K and Ca contents were very low in cutting seedlings; as a result, additional fertilizer should be applied to the seedlings when they were planted in the field. In order to investigate the enzymatic properties of the 4CL1 of Populus tomentosa, the recombinant expression vector pQE31-4CL1 was constructed. The recombinant was identified by three restriction endonucleases, then the vector pQE31-4CL1 was transformed into expression host M15 (pREP4) and induced by isopropyl-α-D-thiogalactoside (IPTG) to express 60 kD fused protein Pt4CL1. The biologically active Pt4CL1, expressed as soluble protein, was achieved with 0.6 mmol·L-1 IPTG induction as the ex-pression temperature declined from 37 to 28°C. The 6×His tag facilitates affinity binding to Ni2 -nitrolotriacetic acid (NTA) and enables one-step purification to acquire the molecular SDS-PAGE electrophoresis purity of the active 4CL1 protein by agarose cou-pled with Ni2 -NTA affinity chromatography. The time processes of photosynthetic induction responses to various irradiances in Korean pine (Pinus koraiensis) seedlings grown in open-light environments and in understory of forest were studied in an area near the Research Station of Changbai Mountain Forest Ecosystems, Jilin Province, China from July 15 to August 5, 1997. The results showed that at 200 μmol·m−2·s−1 photosynthetic photon flux density (PPFD) and 500 μmol·m−2·s−1 PPFD, the induction time for the photosynthetic rates of understory-grown seedlings to reach 50% and 90% steady-state net photosynthetic rates was longer than that of the open-grown seedlings. The induction responses of open-growth seedlings at 500 μmol·m−2·s−1 PPFD were slower than those at 200 μmol·m−2·s−1 PPFD, but it was the very reverse for understory-growth seedlings, which indicates that the photosynthetic induction times of Korean pine seedlings grown in the understory depended on the sunfleck intensity. 研究赤霉素(GA3),冷湿和温度 对五个种源的印度冷杉(Abies pindrow)和长叶云杉(Picea smithiana)种子萌发的影响.种子被浸泡在GA3 (10 mg(L-1)中24小时,然后在3(5(C温度的条件下冷藏15天.设计4个温度(10(C, 15(C, 20(C 和 25(C)条件来促进种子的萌发.结果表明,浸泡和冷湿处理明显增加种子的萌发率.在10 (C时种子的萌发率最高.总体结果表明,种子被浸泡在GA3 (10 mg(L-1)中24小时,冷湿藏15天,可以有效地促进印度冷杉和长叶云杉的种子萌发.统计数据表明,浸泡处理、温度和种源以及与温度的相互作用都对种子的萌发有明显的影响.  相似文献

Experiments were conducted to study plant regeneration through direct somatic embryogenesis using mature zygotic embryo and cotyledonary explants from seeds of Melia volkensii stored for <3 and >12 months. Explants were cultured on Murashige and Skoog (MS) medium supplemented with BAP, NAA and 2,4-D (0.5, 1.0 and 2.0 mg l−1) alone, and BAP (0.5, 1.0, 2.0 and 4.0 mg l−1) in combination with 2,4-D or NAA (0.2 and 0.5 mg l−1). After 4 weeks in culture, up to 60% of cotyledonary explants from the seeds stored for <3 months produced direct somatic embryos on BAP (0.5–4.0 mg l−1) in combination with 2,4-D (0.2 mg l−1). The number of somatic embryos ranged from 5 to 14 per explant in BAP (0.5 mg l−1) and 2,4-D (0.2 mg l−1) combination. Only 20% of cotyledonary explants from seeds stored for >12 months produced somatic embryos. Mature zygotic embryos failed to produce any somatic embryos. Subcultures of somatic embryos from cotyledonary explants of seeds stored for <3 months formed clusters of shootlets on semi solid MS and 1/2 MS media. After 6 weeks of subculture on multiplication MS media augmented with BAP (0.5 mg l−1) and IAA (0.2 mg l−1), 70% of the shoot tips formed 4–7 shoots per explant. Up to 33% of the multiplied shoots were rooted in MS medium supplemented with 2.0 mg l−1 IBA. Plantlets developed normally into seedlings in the greenhouse.  相似文献

3年生白桦同时接受3种外源糖溶液(蔗糖、果糖、葡萄糖)和3种高浓度CO2(700、1400、2100μL·μL-1CO2)处理.处理1个月后,测定了叶片的总糖、蔗糖、果糖和蛋白质含量.结果表明:在700μL·L-1和1400μL·L-1 CO2下,外源糖溶液增加了叶片的可溶性糖和蛋白质含量,其中外源蔗糖的效果最好:外源糖溶液与2100μL·L-1CO2结合,会抑制叶片积累总糖和蛋白质:在700μL·L-1和1400μL·L-1CO2下,喷施葡萄糖、果糖的叶片在蛋白质含量上没有明显差别:同700、1400μL·L-1CO2相比,除喷施果糖植株外,2100μL·L-1 CO2明显增加了叶片的总糖、蔗糖、果糖和蛋白质含量:在喷施同种外源糖溶液的情况下,叶片的糖含量与CO2浓度呈正相关性.图6参7.  相似文献

A study was conducted to determine the fitting soil moisture for the normal growth of two-year-old W. sinensis (Sims) Sweets by using gas exchange technique. Remarkable threshold values of net photosynthetic rate (Pn), transpiration rate (Tr) and water use efficiency (WUE) were observed in the W. sinensis leaves treated by various soil moisture and photosynthetic available radiation (PAR). The fitting soil moisture for maintaining a high level of Pn and WUE was in range of 15.3%-26.5% of volumetric water content (VWC), of which the optimal VWC was 23.3%. Under the condition of fitting soil moisture, the light saturation point of leaves occurred at above 800μmol.m^2.s^-1, whereas under the condition of water deficiency (VWC, 11.9% and 8.2%) or oversaturation (VWC, 26.5%), the light saturation point was below 400μmol.m^-1.s^-1. Moreover, the light response curves suggested that a special point of PAR occurred with the increase in PAR. This special point was considered as the turning point that indicated the functional transition from stomatal limitation to non-stomatal limitation. The turning point was about 600, 1000, 1000 and 400 μmol.m^-2.s^-1, respectively, at VWC of 28.4%, 15.3%, 11.9% and 8.2%. In conclusion, W. sinensis had higher adaptive ability to water stress by regulating itself physiological function.  相似文献

Dalbergia sissoo Roxb. is one of the promising multipurpose tree species of South Asia. Most of the plantations of D. sissoo from seeds are facing severe threats due to the die-back disease, which ultimately causes death of this potential tree-species within a few months. Vegetative propagation could avoid the die-back disease. Thirty mother trees of different age-groups of D. sissoo were selected for evaluating the rooting behaviour of branch cuttings from D. sissoo as influenced by auxins (IAA or IBA at 100, 200, 500 mg·L−1), ages of mother trees (10, 4 and 2 years old) and different environment conditions, i.e., different mediums (soil and sand) or light conditions (in shade and open condition). The results show that application of IAA and IBA induced more numbers of cuttings (collected from 10-year-old mother trees) to root compared to control. Branch cuttings of D. sissoo collected from 10-year-old mother trees and planted in soil bed in open conditions had 100.0% of cuttings to root in IAA (100 mg·L−1) and IBA (200 mg·L−1) treatments. Both rooting medium (Soil and sand) influenced significantly (p<0.05) on rooting response of branch cuttings. Soil medium was found to achieve maximum no. of branch cuttings to root, compared to sand medium.  相似文献

孟加拉东南部土壤中的砷含量很高,不仅威胁人的健康,而且对土壤也有破坏作用。云南石梓(Gmelina arborea)在孟加拉是个快速生长的树种,也是含砷土壤中的潜力树种。研究评价了含砷试验土中丛枝菌根真菌对云南石梓(Gmelina arborea)生长的影响。播种前,四种不同浓度的砷(10mg·kg-1、25mg·kg-1、50mg·kg-1和100mg·kg-1)被加入到试验土中。记录生长参数,如,植物的根、苗鲜重、干重、冠幅径、根长和苗高、根瘤菌和孢子菌群落。菌根植株较非菌根植株生长好。与其它含砷量高的土壤中植株的生长情况相比,在含砷量为10-mg·kg-1的土壤中,菌根植株和菌根生长效果最佳,菌根植株生物量最高。随着砷浓度的增加,种苗生长,根瘤菌和孢子菌群落均明显降低p0.05)。与非菌根植株比较,菌根植株高生长增加了40%,生物量增加了2.4倍。研究表明,根瘤菌接种可以减少有害土壤中的云南石梓(Gmelina arborea)的初生长的影响。  相似文献

The effect of Thidiazuron (TDZ), basal media and light quality on adventitious shoot regeneration from in vitro cultured stem of Populus albaxP berolinensis were determined to establish a high efficiency shoot regeneration system from stem explants of P. alba~P berolinensis. Stems ofPopulus alba~P berolinensis were collected from cultured shoots in vitro derived from dormancy buds of 3-year-old seedlings. The stem explants were cultured on MS medium containing 0.02-mg·L-1NAA (naphthaleneacetic acid), and 0.1, 0.3, 0.5 and 1.0 mg·L-1 concentrations of TDZ to determine the effect of TDZ on shoot regeneration. Three basal media, i.e. MS, woody plant medium (WPM) and B5, were used to test their influences of different media on adventitious shoot regeneration. Green, red, blue and yellow plastic films in comparison with florescent light as control were used to observe their effects on shoot regeneration. The results showed that differ- ent concentrations of TDZ had an evident influence on shoot regeneration. Lower concentration of TDZ (0.1 mg·L-1) resulted in more ad- ventitious shoot regeneration and higher concentration of TDZ (〉0.1 mg·L-1) inhibited shoot regeneration. Among different media, MS medium exhibited a high efficiency for shoot regeneration, followed by WPM medium, while B5 medium inhibited shoot regeneration. Normal light and yellow light exhibited better effects on shoot regeneration, compared with other light.  相似文献

We developed a shoot multiplication protocol for Syringa reticulata Blume var. mandshurica Hara from in vitro cultured seedlings that derived from in vitro germinated seeds. The shoots could be induced on Murashige and Skoog (MS) medium with proper plant growth regulator combinations of 6-benzylaminopurine (BA) and indole-3-butyric acid (IBA). The better medium for shoot multiplication and growth was MS + 5 mg L?1 BA + 0.5 mg L?1 IBA + 20 g L?1 sucrose + 7 g L?1 agar, and the corresponding shoot induction rate was 75 %. The plantlets grew well after rooting on 1/2MS medium (macro-elements of MS medium are at half-strength) supplemented with 1 mg L?1 IBA, and the survival percentage was >80 % at 16 weeks after transplanting.  相似文献

Nursery practice using microbial inoculants was performed to find out the efficacy of the inoculants on seed germination and seedling growth of sissoo (Dalbergia sissoo Roxb.). Microbial inoculants or effective microorganisms (EM) are a mixture of many different beneficial microorganisms in a solution. The seedlings were grown in a mixture of sandy soil and cowdung (3:1) kept in polybags with pouring EM solution at different concentrations (0.1%, 0.5%, 1%, 2%, 5% and 10%) before and after a week of sowing the seeds. Seed germination rate and growth parameters of seedlings were measured, such as, shoot and root length, vigor index, fresh and dry weight of shoot and root and total biomass increment. The nodulation status influenced by EM was also observed along with the measurement of pigment contents in leaves. The highest germination rate (69%) was observed in 2% EM treatment, followed by 67% and 65% in 1% and 5% EM. The highest shoot length (33.2 cm) was in 2% EM, whereas highest root length (26.3 cm) was in 1% EM. Both fresh and dry weights from shoot and root, were maximum (4.16 g and 1.57 g; 2.12 g and 0.83 g respectively) in 2% EM and were significantly (p ≤ 0.05) different from control. Vigor index was highest (4071) in 2% EM, which was significantly (p ≤ 0.05) different from control. Total dry weight increment was highest in 2% EM treatment, followed by 1% and 5% concentrations of EM. Nodulation number was higher at very low (0.1%) concentration of EM solution but it normally decreased with the increase of EM concentrations. The contents of chlorophyll a, chlorophyll b and carotenoid were highest (60.11, 17.05 and 42.48 mg·L−1 respectively) in 2% EM treatment and lowest (39.35, 13.55 and 27.29 mg·L−1 respectively) in control treatment. Therefore, low concentration of EM (up to 2%) can be used for getting maximum seed germination rate and seedling development of Dalbergia sissoo Roxb.  相似文献

The AhDREB1 gene, cloned from Atriplex hortensis L., was transferred into black locust (Robiniapseudoacacia L.) by an Agrobacterium-mediated transformation. The results suggest that stems of black locust sub-cultured in vitro for 20 d are suitable for genetic transformation. The optimum concentrations of kanamycin and cefotaxime were 30 and 150 mg.L-1, respectively. Important factors affecting the transformation efficiency were studied by means of a L9(3^4) orthogonal design. An effective system for genetic transformation in black locust was developed as follows: the stems were pre-cultured for 2 d, immersed in the Agrobacterium solution (OD600 = 0.7) with 10 mg·L^-1 acetosyringone for 21 min and then co-cultured for 2 d. The selection pressures, changing from low to high, could improve transformation efficiency. The transgenic plants were identified by a PCR method. The PCR results indicated that the AhDREB1 gene had been integrated into the genome of black locust and two lines of the transgenic plants were obtained.  相似文献
