The expression pattern of TIR8 is conserved among vertebrates   总被引:1,自引:0,他引:1  
IL-1R/TLRs play an important role in the inflammatory responses. Their signaling pathway is tightly regulated to keep inflammation under control. The regulation involves both extracellular and intracellular mechanisms. TIR8, also known as SIGIRR (Single Immunoglobulin IL-1R-Related molecule), is an orphan receptor belonging to the IL-1R/TLRs super-family. Recent studies suggest its role in modulating the inflammatory response in the gastrointestinal tract in mice, acting as an IL-1 receptor family antagonist. We identified the sequence and analyzed the expression of TIR8 in a wide panel of organs and tissues in different animals. There was high homology of the cDNA sequence among human, mouse and the domestic species (74–85%). The pattern of expression of this receptor was similar in all the species examined (high levels in kidney and gastrointestinal tract) and similar to the human. These results demonstrate that TIR8 is conserved, in evolutionary terms, both with regard to sequence and pattern of expression, a finding consistent with a key function of this molecule in modulating inflammation in the gastrointestinal tract.  相似文献

Nonsteroidal anti-inflammatory drugs (NSAIDs) have long been used for the treatment of pain and inflammation because of their inhibitory effects on cyclooxygenase (COX). For almost as long as NSAIDs have been in use, multiple adverse effects have been noted. Assessment of many of these adverse effects have been complicated because of the discovery of multiple splice variants of the cox gene, and a greater array of COX inhibitors, especially the COX-2 selective inhibitors have become available. Some of these adverse effects cannot be readily explained by the effect of these drugs on COX. This has sparked a new field of investigation into the COX-independent effects of the COX inhibitors. The major noncyclooxygenase targets of the COX inhibitors of particular relevance to inflammation and the gastrointestinal tract are phosphatidylinositol 3'-kinase Akt signaling, uncoupling of oxidative phosphorylation, PPARgamma, nuclear factor KB, mitogen activated protein kinases, and heat shock proteins.  相似文献

OBJECTIVE: To determine an infusion rate of butorphanol tartrate in horses that would maintain therapeutic plasma drug concentrations while minimizing development of adverse behavioral and gastrointestinal tract effects. ANIMALS: 10 healthy adult horses. PROCEDURE: Plasma butorphanol concentrations were determined by use of high-performance liquid chromatography following administration of butorphanol by single IV injection (0.1 to 0.13 mg/kg of body weight) or continuous IV infusion (loading dose, 17.8 microg/kg; infusion dosage, 23.7 microg/kg/h for 24 hours). Pharmacokinetic variables were calculated, and changes in physical examination data, gastrointestinal tract transit time, and behavior were determined over time. RESULTS: A single IV injection of butorphanol was associated with adverse behavioral and gastrointestinal tract effects including ataxia, decreased borborygmi, and decreased defecation. Elimination half-life of butorphanol was brief (44.37 minutes). Adverse gastrointestinal tract effects were less apparent during continuous 24-hour infusion of butorphanol at a dosage that resulted in a mean plasma concentration of 29 ng/ml, compared with effects after a single IV injection. No adverse behavioral effects were observed during or after continuous infusion. CONCLUSIONS AND CLINICAL RELEVANCE: Continuous IV infusion of butorphanol for 24 hours maintained plasma butorphanol concentrations within a range associated with analgesia. Adverse behavioral and gastrointestinal tract effects were minimized during infusion, compared with a single injection of butorphanol. Continuous infusion of butorphanol may be a useful treatment to induce analgesia in horses.  相似文献

动物胃肠道被视为抵御外界病原体的一道重要屏障,胃肠道健康与动物机体健康密不可分。动物胃肠道中栖息着大量多样性的微生物种群,微生物之间的相互作用复杂且多样,微生物的功能和种类组成影响着胃肠道的稳态平衡。微生物群通过定植在胃肠道黏膜,在黏膜免疫系统发育中起着至关重要的作用,同时也对胃肠道健康与功能起着重要作用。调控胃肠道健康的方式有很多种,目前主要通过添加益生元、益生素等非营养物质和营养调控剂来调控胃肠道功能和维护胃肠道健康。但相较于其他营养调控剂来说,免疫球蛋白调控胃肠道的研究报道较少,属于新的免疫调控技术,其具有特异专一性、易生产且成本低、效果显著等优点。免疫调节是一种高效且相对安全的调节方式。作者介绍了4种免疫球蛋白(IgG、IgA、IgY、纳米抗体)的主要理化性质,总结了现阶段免疫球蛋白对胃肠道功能调节的进展及展望,从而为应用免疫球蛋白调控动物胃肠道健康和促进营养代谢提供指导。  相似文献

反刍动物胃肠道中存在着复杂、多样且动态的微生物菌群,它们对维持免疫功能、营养物质消化和代谢动态平衡具有重要作用。氧化应激是自由基产生过多、抗氧化能力不足或两者同步的结果,严重情况下可危害动物健康,并造成经济损失。越来越多的研究表明,胃肠道菌群与氧化应激之间存在密切的联系,它们通过相关代谢物诱导局部和全身炎症,并调节氧化应激。本文综述了部分反刍动物胃肠道菌群与氧化应激的相关研究,对胃肠道菌群代谢物和微生物-肠道-器官轴对氧化应激的调节进行了讨论,旨在为未来进一步探讨反刍动物胃肠道菌群调控氧化应激的机制提供理论依据。  相似文献

丁酸钠是一种绿色新型饲料添加剂,目前在动物饲料中应用广泛,且效果良好。作者主要介绍了丁酸钠的生物学功能、对犊牛胃肠道发育的影响及其在犊牛日粮中的应用效果。重点总结了丁酸钠促进犊牛胃肠道发育的影响机制,包括作为胃肠道组织的能量来源调控激素和生长因子的分泌,提高消化和吸收相关酶的分泌及活性,提高肠道免疫细胞保护性蛋白的分泌等。现有研究表明,日粮干物质中添加0.3%丁酸钠就能达到促进犊牛生长和胃肠道发育的效果,但具体添加水平、添加阶段和方式等尚不明确。此外,日粮粗饲料水平、精料发酵所产生的内源丁酸对外源丁酸钠的作用效果是否有影响也鲜有报道,有待进一步开展研究。  相似文献

Normal individuals maintain tolerance to the endogenous bacterial flora residing within their alimentary tract, a phenomenon mediated by the gastrointestinal lymphoid tissue. Loss of this tolerance is a key factor in the development of chronic intestinal inflammation. Manifestations of such uncontrolled inflammation in humans include inflammatory bowel disease and celiac disease. Dogs may similarly be affected, and although the etiopathogenesis is likely similar, the lesions differ. This review includes discussion of the factors involved in breakdown of mucosal tolerance, the immunologic basis of canine enteropathies, and the use of novel immunotherapies for these diseases.  相似文献

Idiopathic inflammatory bowel disease was the diagnosis for 58 dogs and 26 cats, with signs of persistent gastroenteritis, failed responses to dietary trials, and histologic evidence of cellular infiltrates unrelated to other causes of gastrointestinal tract inflammation. Clinical signs of large intestinal dysfunction, watery diarrhea, vomiting, and anorexia with weight loss were common. Nonspecific hematologic, biochemical, and radiographic abnormalities frequently were observed. Mucosal biopsy specimens, obtained endoscopically, were histologically evaluated for severity of mucosal epithelial damage. Mucosal erythema, friability, enhanced granularity, and ulceration or erosion were the predominant endoscopic lesions. Inflammatory bowel disease lesions of moderate severity predominated in the stomach, duodenum, and colon. Lymphocytic/plasmacytic infiltrates were limited to the lamina propria in biopsy specimens from all regions of the gastrointestinal tract. Inflammatory bowel disease commonly is associated with chronic gastroenteritis in dogs and cats.  相似文献

Choice of an analgesic for gastrointestinal pain requires consideration of the cause of the pain, desired duration of pain relief, need for sedation, and potential side effects and toxicity, particularly in light of other drugs being used and effects on the gastrointestinal tract. It is imperative that close monitoring be continued to ensure that surgical lesions or worsening conditions are detected. Recent research in the field may lead to new drugs, drug combinations, and avenues of treatment that minimize the side effects of these drugs while maximizing their efficacy.  相似文献

OBJECTIVE: To determine concentrations of 2 acute-phase proteins (serum amyloid A [SAA] and lipopolysaccharide-binding protein [LBP]) in serum samples obtained from horses with colic and identify relationships among these acute-phase proteins and clinical data. ANIMALS: 765 horses with naturally developing gastrointestinal tract diseases characterized by colic (ie, clinical signs indicative of abdominal pain) and 79 healthy control horses; all horses were examined at 2 university teaching hospitals. PROCEDURE: Serum concentrations of SAA and LBP were determined by immunoturbidometric and dot-blot assays, respectively. RESULTS: SAA and LBP concentrations were determined for 718 and 765 horses with colic, respectively. Concentrations of SAA were significantly higher in nonsurvivors than in survivors, and horses with enteritis or colitis and conditions characterized by chronic inflammation (eg, abdominal abscesses, peritonitis, or rectal tears) had SAA concentrations significantly greater than those for horses with other conditions. Serum concentrations of LBP did not correlate with outcome, disease process, or portion of the gastrointestinal tract affected. CONCLUSIONS AND CLINICAL RELEVANCE: Circulating concentrations of SAA were significantly higher at admission in horses with colic attributable to conditions having a primary inflammatory cause (eg, enteritis, colitis, peritonitis, or abdominal abscesses) and were higher in horses that failed to survive the episode of colic, compared with concentrations in horses that survived. Serum concentrations of LBP did not correlate with survival. Analysis of these findings suggests that evaluation of SAA concentrations may be of use in identifying horses with colic attributable to diseases that have inflammation as a primary component of pathogenesis.  相似文献

OBJECTIVE: To determine the pharmacokinetics and effects of the morphine antagonist N-methylnaltrexone (MNTX) on gastrointestinal tract function in horses when administered alone and in combination with morphine. ANIMALS: 5 healthy adult horses. PROCEDURES: Horses were treated with MNTX (1 mg/kg, IV), and serial blood samples were collected for determination of drug pharmacokinetics. For evaluation of effects on the gastrointestinal tract when administered alone, MNTX was administered at a dosage of 0.75 mg/kg, IV, twice daily for 4 days. For evaluation of effects when administered concurrently with morphine, MNTX (0.75 mg/kg, IV, q 12 hours) and morphine (0.5 mg/kg, IV, q 12 hours) were administered for 6 days. Gastrointestinal variables evaluated were defecation frequency, weight of feces produced, fecal moisture content, intestinal transit time, and borborygmus scores. RESULTS: The time-concentration data for MNTX disposition best fit a 2-compartment model with a steady-state volume of distribution of 244.6 +/- 21.8 mL/kg, t1/2 of 47.04 +/- 11.65 minutes, and clearance of 11.43 +/- 1.06 mL/min/kg. 复杂和多样化的肠道微生物群落对宿主健康发挥着重要作用。产丁酸菌代表了宿主肠道微生态一个重要的功能性群体,多数为革兰阳性厌氧菌。人们发现,该类菌和微生物失调引发的消化道炎症等现象息息相关,因此产丁酸菌成为行业研究热点。产丁酸菌能够利用食物(饲料)中不易消化的碳水化合物合成丁酸,通过调控肠道菌群结构、为肠道上皮细胞供能、增强肠黏膜屏障功能等维护肠道健康,发挥对宿主健康的有益作用。本文从产丁酸菌的生物学分类、消化道产酸机制、肠道炎症调控作用及应用进展等方面做了系统综述,旨在阐明产丁酸菌与宿主健康的互作机制,并提出产丁酸菌的未来发展方向。  相似文献

欧洲猪生产中抗菌饲料添加剂的禁用及对猪胃肠道微生物发酵的有益调控的研究 ,促进了非消化性寡糖的研究。非消化性寡糖 ,作为化学益生素最显著的作用就是对有益菌 ,如双歧杆菌的特殊的刺激作用 ,并通过竞争性排斥病源微生物如大肠杆菌的生长 ,抑制有害菌的作用 ,从而有助于保持胃肠道合适的菌丛。然而 ,猪日粮中包含的特异性非消化性寡糖对微生物生态系统、消化过程及动物的生产性能的作用存在着争议。由于大多数非消化性寡糖在胃和小肠中快速的发酵速率 ,因此上段消化道的糖分解活力不能在整个消化道中维持。若使用在整个胃肠道部分包括大肠能够发酵的非消化性寡糖的复合物可延长化学益生的作用 ,并且证明其作为饲料添加剂在猪饲料中的使用是有效的  相似文献

OBJECTIVE: To estimate prevalence of and identify risk factors for fecal Salmonella shedding among hospitalized horses with signs of gastrointestinal tract disease. DESIGN: Cross-sectional study. ANIMALS: 465 hospitalized horses with gastrointestinal tract disease. PROCEDURE: Horses were classified as positive or negative for fecal Salmonella shedding during hospitalization by means of standard aerobic bacteriologic methods. The relationship between investigated exposure factors and fecal Salmonella shedding was examined by means of logistic regression. RESULTS: The overall prevalence of fecal Salmonella shedding was 13%. Salmonella serotype Newport was the most commonly isolated serotype (12/60 [20%]), followed by Anatum (8/60 [13%]), Java (13%), and Saint-paul (13%). Foals with gastrointestinal tract disease were 3.27 times as likely to be shedding Salmonella organisms as were adult horses with gastrointestinal tract disease. Adult horses that had been treated with antimicrobial drugs prior to hospitalization were 3.09 times as likely to be shedding Salmonella organisms as were adult horses that had not been treated with antimicrobial drugs prior to hospitalization. Adult horses that underwent abdominal surgery were 2.09 times as likely to be shedding Salmonella organisms as were adult horses that did not undergo abdominal surgery. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that a history of exposure to antimicrobial drugs prior to hospitalization and abdominal surgery during hospitalization were associated with Salmonella shedding in adult horses with gastrointestinal tract disease. Foals with gastrointestinal tract disease were more likely to shed Salmonella organisms than were adult horses with gastrointestinal tract disease.  相似文献

鸽毛滴虫病是由鸽毛滴虫引起的一种侵害消化道上段的原虫病,其特征是在口腔、食道、嗉囊、前胃出现坏死性溃疡,常常造成上消化道和呼吸道阻塞,妨碍进食及正常呼吸进而引起饥饿造成宿主死亡。世界各地均有鸽的病例报道,毛滴虫首先感染上消化道,引起其溃疡,然后从溃疡处进入循环系统,引起肝脏等器官的病变,可以通过直接涂片镜检、人工培养检查等病原的诊断方法对该病进行确认。鸽毛滴虫对硝唑类药物敏感,但极易产生抗药性。目前对鸽毛滴虫分子生物学方面的研究不多,充分利用现代分子生物学技术开展鸽毛滴虫的生理、生化,特别是安全有效的分子疫苗是今后的一个重要研究方向。  相似文献

Transfer of functional blood IgG1 to the gastrointestinal tract was measured in neonatal calves. Radiolabelled immunoglobulin G1 (IgG1) anti-DNP antibody was administered to 2 day old calves by intravenous injection. The serum clearance rate was measured and was compared to the rate of protein-bound 125I excretion in the feces over a 10 day period to determine the importance of transfer to the gastrointestinal tract as a mechanism of serum IgG1 clearance. The amount of protein-bound and DNP-binding 125I present in the gastrointestinal tract of 10 day old calves at necropsy was also measured. Fecal excretion of protein-bound 125I accounted for 32% of the serum 125I-IgG1 clearance. Protein-bound 125I was present in the gastrointestinal tract at necropsy in amounts estimated to account for 68% of the total 125I-IgG1 clearance, and retained 65% of the DNP-binding ability of the original antibody. The discrepancy between the fecal excretion (32% of total IgG1 clearance) and the GI clearance estimated from protein-bound 125I in the gut (68% of total IgG1 clearance) is explained in part by IgG1 proteolysis occurring after transfer to the gastrointestinal tract but before fecal excretion. These results indicate that transfer to the calf gastrointestinal tract accounts for most IgG1 clearance in young calves, and that the intestinal antibody retains antigen binding function and may contribute to intestinal immunity.  相似文献
