Eleven pathotype groups (A-K), including five not previously reported, ofDidymella rabiei (anamorphAscochyta rabiei), representing isolates of the pathogen from Ascochyta blight-affected chickpeas mainly from India, Pakistan, Spain and the USA, were characterized using 44 single-spore isolates tested against seven differential chickpea lines. Of 48 isolates tested for mating type, 58% belonged to MAT 1-1 and 42% to MAT 1-2. Thirty-nineD. rabiei isolates, as well as two isolates ofAscochyta pisi and six isolates of unrelated fungi, were analyzed using Randomly Amplified Polymorphic DNAs (RAPDs) employing five primers (P2 at 40°C, and OPA3, OPC1, OPC11 and OPC20 at 35°C). Computer cluster analysis (UPGMA / NTSYS-PC) detected a relatively low level of polymorphism among all theD. rabiei isolates, although atca 7% dissimilarity,ca 10 RAPD groups [I-X] were demarcated, as well as subclustering within the larger groups. By the same criteria, the maximum dissimilarity for the whole population ofD. rabiei isolates wasca 13%. No correlation was found between different RAPD groups, pathotype, or mating type ofD. rabiei, although some evidence of clustering based on geographic origin was detected. The use of RAPDs enabled us to identify specific DNA fragments that may have a potential use as genetic markers in sexual crosses, but none which could be used as virulence markers.  相似文献

Sharka disease has a limited distribution in Turkey and does not present a problem for stone fruit production. However, sharka is the most common virus disease of apricots, plums and peaches in Ankara, although it is not a common disease in other cities in Turkey. In different parts of Ankara, 212 private gardens in 21 locations were surveyed forPlum pox virus (PPV) incidence. All together 935 trees of apricot, plum, peach, sweet cherry and sour cherry were sampled and tested for the presence ofPPV by double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA).PPV was identified in numerous trees,viz. 286 apricots, 172 plums and 65 peaches. Strain differentiation ofPPV was accomplished using double-antibody sandwich indirect enzyme-linked immunosorbent assay (DASI-ELISA). These assays confirmed the presence of isolates belonging toPPV-M, PPV-D and mixed infections ofPPV-M+D. This work was undertaken to determine the effects ofTrichoderma spp. combined with label and sublabel rates of metam sodium on survival ofRhizoctonia solani in soil. Soils were infested with wheat bran preparations ofTrichoderma hamatum Tri-4,T. harzianum Th-58,T. virens Gl-3, andT. viride Ts-1-R3. Soil was also infested with sterile beet seeds that were colonized withR. solani. Beet seeds were later recovered, plated onto water agar plus antibiotics, and the growth ofR. solani was recorded. Preliminary experiments showed thatT. hamatum andT. virens reduced survival and saprophytic activity ofR. solani when the biocontrol fungi were incorporated into soil at 1.5% (w:w) or greater. Based on these data, biocontrol fungi in subsequent experiments were incorporated into soil at 2%. Metam sodium at label rate killed all biocontrol fungi andR. solani. At 1:2 and 1:5 dilutions, metam sodium reduced survival ofR. solani and allTrichoderma spp. When biocontrol fungi plus the label rate of metam sodium and 1:5, 1:10, 1:50 or 1:100 dilutions of the label rate were tested together, there were no interactions between any biocontrol agent and the fumigant with respect to colony diameter, reflecting that allTrichoderma isolates tested reacted similarly to increasing concentrations of metam sodium. At the label rate of metam sodium, allTrichoderma spp. significantly reduced colony diameter, but not growth rate, ofR. solani from beet seed. For the levels of metam sodium tested in combination withTrichoderma, it does not appear feasible to use a reduced rate of metam sodium to controlR. solani. Typical symptoms of corky root were observed on iceberg lettuce (Lactuca sativa L.) in the Netherlands and England, on prickly lettuce (Lactuca serriola L.) in Spain, and on sowthistle (Sonchus oleraceus L.) in Greece. Slow-growing bacteria with similar colonies as strains ofRhizomonas suberifaciens or otherRhizomonas species were isolated from soil surrounding plants with corky root symptoms using lettuce seedlings as bait. Crude lysate from all strains was tested for DNA homology with DNA fromR. suberifaciens strain CA1 (R. sub. CA1) andRhizomonas sp. strain WI4 (R. sp. WI4). Strains that had homology values higher than that ofR. sp. WI4 orR. sub. CA1 were tested for pathogenicity on 1-wk-old lettuce seedlings, cv. Salinas, Two strains from the Netherlands induced typical symptoms of corky root on lettuce. These strains tested negative with monoclonal antibody MAb-Rs1 specific forRhizomonas suberifaciens in an enzyme-linked immunosorbent assay (ELISA). They had low DNA homology withR. sub. CA1 (4–9%) and low to moderate DNA homology withR. sp. WI4 (4–17%). Some nonpathogenic strains had moderate to high levels of DNA homology withR. sub. CA1 orR. sp. WI4 (19–84%). All strains had fatty acid profiles similar to those ofRhizomonas species. This is the first report ofRhizomonas sp. causing corky root of lettuce in Europe.  相似文献

The host range specificity of Greek isolates ofCucumber green mottle mosaic virus (CGMMV) was determined and the ability of the virus to retain its infectivity in naturally contaminated soil, after storage at 4°C for at least 10 months, was established. An immunocapture RT-PCR protocol was developed for the detection of the virus and proved to be 105 times more sensitive than DAS-ELISA and 102 times more sensitive than F(ab’)2-ELISA using F(ab’)2 fragments that were prepared from an antiserum, which was raised against a local virus isolate. In order to clarify dissimilarities that were revelaed between the coat protein (CP) genes of the Greek isolates after restriction mapping of their respective PCR products, the CP genes of three isolates were sequenced and found to be very similar but not identical to the CP gene of CGMMV-SH. http://www.phytoparasitica.org posting Aug. 27, 2002.  相似文献

Four-hundred-sixty-eightFusarium andFusarium-like isolates were obtained from crowns and subcrown internodes of winter wheat grown in Erzurum, Turkey. Of these isolates, 34.8% wereFusarium acuminatum, 32.3% wereF. equiseti, 16.9% wereF. oxysporum, 15.0% wereMicrodochium nivale (formerlyFusarium nivale), 0.6% wereF. tabacinum and 0.4% wereF. solani. In pathogenicity tests on wheat, the highest disease severity was caused by isolates ofM. nivale, whereas isolates ofF. acuminatum, F. equiseti, F. oxysporum andF. solani were slightly virulent; isolates ofF. tabacinum were nonpathogenic. Abstrast  Three-hundred-twenty-five isolates ofRhizoctonia (215R. solani and 110 binucleateRhizoctonia) were obtained from roots and crowns of alfalfa, sainfoin and common vetch grown in Erzurum, Turkey. The isolates were assigned to five anastomosis groups (AG) ofR. solani (AG-2-1, AG-3, AG-4, AG-5, and AG-10) and two anastomosis groups of binucleateRhizoctonia (AG-I and AG-K). In pathogenicity tests on alfalfa, sainfoin and common vetch, the highest disease severities were caused by isolates of AG-4 and AG-5. Isolates of AG-10 and AG-I were not pathogenic on the three tested forage legumes, whereas isolates of AG-K on alfalfa and sainfoin, and of AG-2-1 on sainfoin, were moderately virulent. Alfalfa isolate AG-3 was moderately virulent on sainfoin. This is the first report ofR. solani AG-3, AG-5, AG-10 and binucleateRhizoctonia AG-I on alfalfa. Thirty-four isolates ofFusarium oxysporum f.sp.melonis (F.o.m.) obtained from 205 fields in melon-producing areas in the southeastern Anatolia Region of Turkey were identified on the basis of colony morphology and pathogenicity by the root dip method. In this region the mean prevalence of wilt disease was 88.1% and the mean incidence of disease was 47.5%. Physiologic races 0, 1, 2, and 1,2 of the pathogen were determined by their reactions on differential melon cultivars ‘Charentais T,’ ‘Isoblon’, ‘Isovac’ and ‘Margot’ in the greenhouse. Race 1,2, representating 58.8% (20/34) of all isolates, was widely distributed. Of the other pathogenic isolates, eight were identified as race 0, five as race 1, and one as race 2. This is the first report of physiologic races ofF.o.m. in Turkey. Of 44 melon cultivars tested in the greenhouse for resistance toF.o.m. races, 36 were found to be moderately resistant to race 0, 17 were susceptible to race 1,2, 34.1% were highly resistant to race 1, and 52.2% had moderate resistance to race 2. http://www.phytoparasitica.org posting July 16, 2002.  相似文献

Potential antagonists ofFusarium solani f. sp.pisi (Fsp) were selected from soil samples with varying degrees of receptivity to this pathogen. They were tested against Fsp isolate 48 (Fs48), in increasingly complex systems. Most species testedin vitro were able to antagonize Fs48. No relation could be establishedin vitro between the receptivity of the soil from which an isolate originated and its antagonism to Fs48. In soils naturally infested with pea root rot pathogens, which were stored humid at 4°C for a period longer than a year, various isolates ofFusarium, Gliocladium andPenicillium spp. were able to reduce root rot. After sterilization of these soils, onlyGliocladium roseum isolates, added at 105 conidia g–1 dry soil, significantly reduced disease severity and prevented root weight losses caused by Fs48 at 104 conidia g–1 dry soil. In soils in which the biota were activated by growing peas before the assays, doses of 106 and 107 ofG. roseum were required to reduce root rot. In these soils, the antagonistic effects of fluorescent pseudomonad strains from soil of low receptivity to Fsp were variable. Some strains of fluorescent pseudomonads, from soil moderately receptive to Fsp and from highly infested soils, were also able to reduce root rot. Disease suppression by pseudomonad strains was more evident in the absence than in the presence ofAphanomyces euteiches in the root rot pathogen complex. The role of receptiveness of the soil with regard to potential antagonists is discussed.  相似文献

Sclerotia ofRhizoctonia solani collected from potato tubers from different countries were assayed for the presence of mycoparasites. Among the mycoparasites observedVerticillium biguttatum predominated. Its geographical distribution was not restricted to certain latitudes or soil types;V. biguttatum occurred worldwide in potato fields.The minimum growth temperature of 57V. biguttatum isolates was found to be in the narrow range from 10 to 13°C, irrespective of their geographical origin. A non-linear logistic growth model was used to describe the radial growth onRhizoctonia mycelium and nutrient agar plates. At near-minimum temperature the maximum colony radii varied considerably; they were up to 3.8 times that of the reference isolate M73. Based on parameter values for logistic growth, fast-and slow-growing isolates could be distinguished. Although the growth properties ofV. biguttatum isolates from different locations varied, the presence of fast- and slow-growing isolates was not restricted to particular areas and both types could be found in the same field. However, bioassays with selected fast- and slow-growing isolates do not support the assumption that growth at near-minimum temperatures is a relevant criterion for screening isolates ofV. biguttatum in terms of effectiveness for biological control ofR. solani.  相似文献

Complementary auxotrophic nitrate-nonutilizing (nit) mutants were used to investigate vegetative compatibility within 27 strains ofVerticillium dahliae isolated from several hosts originating from Africa, Asia, Europe and the United States. Using about 500nit mutants generated from these strains, three vegetative compatibility groups, 1, 2 and 4, were identified. Simultaneously, virulence of each strain was assessed on cultivars ofGossypium hirsutum, G. barbadense andG. arboreum, based upon Foliar Alteration Index (FAI) and Browning Index (BI) estimation. The strains in VCG1 were of both the cotton-defoliating pathotype and race 3 (on cotton) but were non pathogenic on tomato; those in VCG2 and VCG4 were of the nondefoliating pathotype and belonged to different races on cotton and on tomato. Hyaline mutants deriving from parental wild-type strain showed differences in pathogenicity but were always assigned to the parental VCG. A relationship was established between VCGs and the taxonomic position of host plants. Data fromnit pairings indicated that the sub-populations ofV. dahliae (VCGs) may not be completely isolated genetically.  相似文献

Anthracnose, caused byColletotrichum gloeosporioides, is the major disease of almond in Israel. Pathogen attack of young fruit results in fruit rot and leaf wilting. Seventy isolates ofC. gloeosporioides were obtained from affected almond fruits collected at 11 sites during 1991–2 and 1994. Chlorate-resistant nitrate-nonutilizing (nit) mutants were generated from each isolate and used in complementation (heterokaryon) tests. The formation of complementary stable heterokaryons between mutants from different isolates showed that all the isolates belonged to a single vegetative compatibility group. Representative isolates ofC. gloeosporioides from almond did not form heterokaryons with local isolates ofColletotrichum from anemone and avocado, indicating that the almond isolates constitute a distinct subspecific group withinC. gloeosporioides.  相似文献

Root and crown rot of raspberry (Rubus idaeus L.) was observed in a plantation at the experimental station of small fruits in Kostinbrod, Bulgaria. Isolates ofPhytophthora spp. were obtained from diseased plants. Colony morphology, growth rates, features of asexual and sexual structures were studied and as a result twoPhytophthora species were identified:Phytophthora citricola Saw. andPhytophora citrophthora (R.E. Sm. & E.H. Sm.) Leonian. Their pathogenicity was confirmed in artificial inoculation experiments. The isozyme (-esterase) patterns ofP. citrophthora andP. citricola isolates from raspberry and from the collection of the CBS, Baarn the Netherlands were compared, using micro-gel electrophoresis. Both species are reported for the first time as pathogens of raspberry in Bulgaria. This is only the second report in phytopathological literature ofP. citrophthora on raspberry, the first being from Chile [Latorre and Munoz, 1993].  相似文献

Vegetative compatibility among 17 isolates ofVerticillium dahliae obtained from watermelon, originating from eight regions of Greece, was investigated using complementation tests between nitrate-nonutilizing(nit) mutants. Among 529 chlorate-resistant sectors obtained, only 107 werenit mutants. These mutants were paired with tester strains (from Greece and other countries) of previously described vegetative compatibility groups (VCGs), and also were paired in many combinations among themselves. All isolates were self-compatible. Sixteen isolates were found to belong to VCG2. Only isolate V75 could not be assigned to a VCG, because the threenit mutants obtained from it showed negative reactions with the tester strains of four VCGs and with complementary mutants from other isolates. Based on this sample, we conclude that the population ofV. dahliae from watermelon in Greece is homogeneous in respect to VCG.  相似文献

The distribution of N-acetylglucosamine residues in the cell wall of the white-rot pathogenic fungus, Rigidoporus lignosus, was studied by using gold labelled wheatgerm agglutinin bound to ovomucoid-colloidal gold. Ultrastructural investigation of R. lignosus-infected root tissues of Hevea brasiliensis showed a modification of the fungal cell wall throughout the infection process. Gold particles were found to occur on both thick- and thin-walled hyphae of R. lignosus rhizomorphs at the root surface. Walls of hyphae that had penetrated the roots were only labelled when they were out of the host cell, suggesting that modification of chitin molecules may be related to the excretion of host cell wall degrading enzymes. Variation in the distribution of gold particles was observed over hyphal walls of both colonized phellem and xylem cells. The observation that N-acetylglucosamine residues were released in the host cell cytoplasm suggests that lytic enzymes alter the fungal cell walls. Released chitin oligosaccharides may play a role in the induction of the root's defence system against fungal attack.  相似文献

Pepper cultivars from diverse geographic origins were evaluated for resistance to different isolates ofPhytophthora capsici under controlled environmental conditions. All accessions tested were susceptible at the four-leaf stage to the six isolates ofP. capsici. Inoculation at the eight-leaf stage resulted in significantly different interactions among the accessions andP. capsici isolates. The Korean and U.S. cultivars tested were highly susceptible to the isolates ofP. capsici at this stage. In contrast, PI 201234 and PI 201238 had a differential interaction with someP. capsici isolates. At the twelve-leaf stage, Phytophthora blight developed slowly in the Korean and U.S. cultivars that were highly susceptible at the eight-leaf stage. Furthermore, the accessions from the Asian Vegetable Research and Development Center (AVRDC) became highly resistant toP. capsici at this stage.  相似文献

Three strains ofPseudomonas fluorescens (63-49, 63-28, and 15), one strain ofPseudomonas corrugata (13) and one strain ofSerratia plymuthica (R1GC4) were tested on rockwool-grown cucumbers for their ability to reduce Pythium root-rot caused byPythium aphanidermatum. These strains were previously selected for biocontrol ability from collections of >4000 bacteria. Strains 63-49 and 63-28 were tested on cucumber plants grown in rockwool in two replicatedPythium-inoculated trials conducted in British Columbia (B.C). Another inoculated, replicated trial was conducted in Quebec with all five strains. Cucumber yields (fruit number and weight) were measured over a ten-week harvest period. Strain 63-49 caused an early promotion of plant growth and increased cucumber yields at early harvests. No measurable effect ofPythium inoculation on disease development was observed in the Quebec trial, due to unfavourable cool weather. However, 63-49 significantly increased the total number of cucumbers (12%) and cucumber weight (18%), compared to the non-treated control. Strains 13, 15 and R1GC4 slightly increased the cumulative cucumber yields, but strain 63-28 had no effect. In the B.C. trial, inoculation withP. aphanidermatum reduced the number and weight of cucumbers by 27%. Treatments ofPythium-inoculated cucumbers with 63-49 significantly increased fruit number and weight by 18%, compared to thePythium-inoculated control. Strain 63-28 increased the cumulative number of cucumbers over time, compared to thePythium-inoculated control, but the increase was less than with 63-49. The use ofPseudomonas spp. in rockwool-grown cucumbers can increase yields, both in the presence and absence of Pythium root rot, and with variable seasonal conditions and disease pressures.  相似文献

Thirteen strains ofPhytophthora porri from five different hosts were compared with respect to their morphology, cardinal temperatures for growth, pathogenicity to leek and cabbage and restriction fragment patterns of mitochondrial DNA. Morphology of vegetative growth was rather similar in most isolates. Those characters which differed among isolates showed overlapping variability and could not be used to distinguish groups, with the exception of production of oogonia and sporangia and the antheridium type. Considerable differences were found in restriction patterns of mitochondrial DNA, isolates from the same host mostly showing identical patterns. Isolates from differentAllium species showed relatively similar restriction patterns if compared to the other isolates. Isolates fromBrassica oleracea proved to be a homogeneous group, quite different from the others with respect to restriction patterns, production of sporangia, production of oogonia, antheridium type and pathogenicity. One isolate, CBS 366.59, isolated from and pathogenic toA. porrum, deviated in many characters from the other isolates. It showed the restriction patterns ofPhytophthora nicotianae and also the high cardinal temperatures for growth typical for this species. The sporangia, however, were distinctly non-papillate and the majority of antheridia was of the paragynous type.  相似文献

Roots of seedlings of wheat and barley affected by bare patch disease at a field site in Western Australia were assessed for root damage and plated to isolate fungi. The patches were variable in shape and size and had the most severely affected plants in the centre. Of the 165 isolates ofRhizoctonia spp. obtained, 90% were multinucleate and 10% binucleate, the former being predominant in the plants at the centre of the patch. The relative frequency of binucleate isolates increased with proximity to the periphery. The increase in activity of avirulent binucleate isolates towards the periphery of the patch may be related to the sharp and abrupt edging of the patch. A variety of other species of fungi such asFusarium spp.,Mortierella spp.,Bipolaris sorokiniana, Pythium sp. andTrichoderma sp. were encountered within the patches. The multinucleate isolates belonging to anastomosis groups (Ag) 2–1, 2–2 and 8 (Thanatephorus cucumeris) were most pathogenic to wheat. The binucleate isolates of Ag C, D, E, and K (Ceratobasidium sp.) were less pathogenic. De tweekernige isolaten van de anastomosegroepen Ag C, D, E en K (Ceratobasidium sp.) waren minder pathogeen. Isolates ofAzotobacter chroococcum were found to be promising for the control of infestation of potato plants withRhizoctonia solani. Inoculation with an isolate ofVerticillium biguttatum in combination with isolates ofA. chroococcum effectively protected sprouts, stems and stolons against infestation withR. solani.
