Follicle selection is associated with an increase in the expression of vascular endothelial growth factor (VEGF) and its receptors in granulosa cells, however, the roles of VEGF in regulating the function of these or other non-endothelial cells in the ovary have not been explored in detail. The current study used bovine cell cultures to investigate potential roles of VEGF in the regulation of granulosa cell function during follicle development. Granulosa cells were obtained from morphologically healthy follicles 4 to 8 mm or 9 to 14 mm in diameter (corresponding to diameters before and after the establishment of dominance, respectively, during a bovine follicular wave) and exposed to a range of VEGF concentrations (1 to 100 ng/mL) encompassing concentrations found naturally in bovine dominant follicles. A concentration of VEGF of 1 ng/mL induced significant proliferation of granulosa cells from 4- to 8-mm follicles (P = 0.024) and increased the proliferative response of these cells to follicle-stimulating hormone (FSH; P = 0.045); whereas higher doses of VEGF had no effect on proliferation (P = 0.9). Treatment with VEGF induced an overall increase in mean extracellular signal-regulated kinase 1/2 (ERK1/2) phosphorylation (P = 0.02). In contrast, VEGF, alone or in combination with FSH, had no effect on expression of the steroidogenic enzyme, CYP11A1, by cells from 4- to 8-mm follicles (P = 0.9). Granulosa cells from 9- to 14-mm follicles responded to 1 ng/mL VEGF with an increase in expression of the ovulation-associated gene, PTGS2 (P = 0.003) but higher VEGF doses had no effect (P = 0.9). The PTGS2 response to 1 ng/mL VEGF was similar to that induced by treatment with luteinizing hormone (LH). Interestingly, the stimulatory effects of LH on ERK1/2 phosphorylation (P = 0.003) and PTGS2 expression (P < 0.01) in granulosa cells from 9- to 14-mm follicles were abolished (P = 0.2) by specific chemical inhibition of VEGF receptor 2 (VEGFR2). These results suggest novel and important roles of VEGF and its receptor, VEGFR2, in mediating and/or enhancing the effects of gonadotropins in granulosa cells.  相似文献

Previous studies in cattle have shown influences of dietary unsaturated fatty acid (UFA) supplementation on ovarian function. However, it is unclear whether these UFA exert direct or indirect effects on ovarian steroid production or their mechanisms of action. We have recently shown that 5′AMP-activated protein kinase (AMPK) regulates progesterone secretion through mitogen-activated protein kinase/extracellular signal-regulated kinase 1/2 (MAPK ERK1/2) in rodent granulosa cells. Here, we investigated the effects of 3 UFAs, oleic acid (OA), linoleic acid (LA), and α-linolenic acid (ALA) on progesterone secretion in goat granulosa cells. Finally, we examined the effects of UFAs on MAPK ERK1/2 and AMPK phosphorylation in these granulosa cells. Oleic acid and LA (10 μM each), but not ALA (100 μM), increased progesterone secretion (P < 0.05) in the presence or absence of insulin-like growth factor (IGF)-1 (10-8 M) or FSH (5 × 10−8 M). The different AMPK subunits, except for γ3, are present in the goat ovary. Treatment with metformin (10 mM), an activator of AMPK, increased AMPK phosphorylation (P < 0.05) and reduced progesterone secretion by 50% (P < 0.05) in the basal state and in response to IGF-1 or FSH in goat granulosa cells. Oleic acid and LA had no effect on AMPK phosphorylation, whereas they rapidly increased MAPK ERK1/2 phosphorylation (P < 0.05). Finally, U0126, a MAPK ERK1/2 inhibitor, decreased OA- and LA-induced progesterone secretion (P < 0.05), suggesting that these UFAs could stimulate progesterone secretion partly through MAPK ERK1/2 in the absence of IGF-1 and FSH in goat granulosa cells. The involvement of AMPK in this process remains to be demonstrated. Taken together, some fatty acids could improve ovarian steroidogenesis through the MAPK ERK1/2 signaling pathway and, consequently, have beneficial effects on goat fertility.  相似文献

Oxidative stress parameters and erythrocyte characteristics were studied in 15 normal healthy dogs and 33 renal azotaemic dogs from Small Animal Hospital, Faculty of Veterinary Science, Chulalongkorn University. Dogs with renal azotaemia had reduced mean corpuscular volume (MCV) (P < 0.01), packed cell volume (PCV) (P < 0.001) and increased mean corpuscular hemoglobin concentration (MCHC) (P < 0.001). The relationship was found between degree of azotaemia and MCV, PCV and MCHC. Dogs with severe renal azotaemia had higher intraerythrocytic sodium contents (RBC-Na) (P < 0.05). The red blood cell catalase activity and glutathione and plasma malondialdehyde were unaltered while urinary malondialdehyde-creatinine ratio (U-MDA/Cr) increased significantly (P < 0.001). The U-MDA/Cr was correlated significantly with plasma creatinine concentration (P < 0.05), urinary protein-creatinine ratio (P < 0.05) and fractional excretion of sodium (P < 0.001). The results suggest some changes in RBC characteristics and urine oxidative stress marker in renal azotaemic dogs. Moreover, the U-MDA/Cr is a sensitive biochemical parameter which increased along with degree of renal dysfunction.  相似文献

This investigation studied the effects of intravaginal administration of a mixture of lactic acid bacteria (LAB) on the incidence of purulent vaginal discharges (PVD), plasma haptoglobin concentrations, and milk production in dairy cows. A total of 82 pregnant primiparous and multiparous Holstein dairy cows were used in this study. Half of the cows received intravaginally 1 mL of LAB at 1010–1012 cfu/mL and the other half 1 mL of reconstituted skim milk (i.e., carrier) (controls). Administration of LAB was conducted once per wk during 2 and 1 wk before the expected day of calving and at 1, 2, 3, and 4 wk postpartum. Data demonstrated that intravaginal administration of LAB decreased the occurrence of PVD at 3 wk postpartum (P < 0.05). Concentrations of plasma haptoglobin, an acute phase protein often associated with uterine infections, was lower in cows treated with the LAB mixture at 2 wk (P < 0.001) and 3 wk (P < 0.05) postpartum. Treatment with LAB did not improve overall pregnancy rate, but the treated multiparous cows produced more milk than their control counterparts (P < 0.05), whereas no difference was observed in primiparous cows regarding milk yield (P > 0.05). Overall, this is the first study demonstrating that intravaginal LAB administration lowers the incidence of PVD and enhances milk production in dairy cows. Further research is warranted to evaluate the effects of LAB on reproductive performance in a larger cohort of cows.  相似文献

The use of herbs as additives in livestock nutrition as an alternative to antibiotics is becoming a new goal in animal production. It is known that green tea exerts antimicrobial activity owing to specific flavonoid compounds named catechins, primarily represented by epigallocatechin-3-gallate (EGCG). Remarkably, despite many potential benefits of green tea and EGCG consumption, it is also important to get an insight on the possible reproductive-related consequences of feeding supplementation. To this purpose, granulosa cells were harvested from follicles > 5mm and treated with 5 and 50 microg/ml of EGCG in order to evaluate the effects on the main parameters of granulosa cell function: steroidogenesis, by measuring progesterone and estradiol-17beta production, and proliferation, one of the major feature of ovarian follicular growth. Moreover, as the genesis of new vessels has been demonstrated to be fundamental for follicle development, we evaluated the effect of EGCG on the production of the main angiogenetic factor, VEGF, by swine granulosa cells. Finally, since reactive oxygen species (ROS) might be involved in the control of female reproductive activity, we studied the effect of EGCG on superoxide anion (O2-) and hydrogen peroxide (H2O2) production by swine granulosa cells and on the activity of the scavenging enzyme superoxide dismutase (SOD). EGCG significantly (p < 0.05) inhibited proliferation, steroidogenesis, VEGF and O2- production by swine granulosa cells; on the contrary, H2O2 levels and SOD activity were stimulated (p < 0.05) by the catechin. Therefore, since our data demonstrate that EGCG has a negative effect on reproductive performances in swine, feeding supplementation should be carefully considered.  相似文献

This study aimed to determine the erythrocytic lipid peroxidation and haemoglobin oxidation as contributory factors causing anaemia in cattle (Friesian × Egyptian native breed) infected with Babesia bigemina. Blood was collected from 32 cows infected with B. bigemina along with 18 healthy cows as controls for determination of erythrocytic malondialdehyde (MDA), blood methaemoglobin (MetHb), plasma free haemoglobin (PHb), corpuscular osmotic fragility (COF), red blood cell count (RBC), total haemoglobin (Hb) and packed cell volume (PCV). Percentage of parasitaemia varied from 14% to 36%. MDA, MetHb, COF and PHb were significantly increased (P < 0.001) in infected cows versus controls. Parasitaemia was positively correlated (P < 0.001) with MDA, MetHb, COF and PHb. MDA was positively correlated (P < 0.001) with COF and PHb and negatively correlated (P < 0.001) with RBC, Hb and PCV. MetHb was negatively correlated (P < 0.001) with RBC, Hb and PCV and positively correlated (P < 0.001) with COF. In conclusion, B. bigemina infection in cattle is associated with a parasitic burden-dependent corpuscular oxidative damage as indicated by membrane lipid peroxidation and methaemoglobin formation, which are contributed to COF and intravascular haemolysis.  相似文献

Two bovine mammary cell types (BME-UV1 and MeBo cells) were used to evaluate the effect of natural retinoids, retinoid analogs, and bovine lactoferrin (bLf) on cell viability in vitro. Experiments with Alamar Blue showed a linear relationship between fluorescence and cell viability index. The BME-UV1 cells exhibited twice the metabolic activity but required half the doubling time of the MeBo cells. The BME-UV1 cells were very sensitive to all-trans retinoic acid (atRA) inhibition of cell viability (P < 0.05) and exhibited a dose-dependent inhibition with 9-cisRA (9cRA; P < 0.05). The MeBo cells exhibited some inhibition with these natural ligands (P < 0.05), but they were not as sensitive. The addition of bLf had similar inhibitory effects (P < 0.05) on cell viability of the 2 mammary cell types. Applications of RA receptor (RAR) agonist indicated that the stimulation of the RAR in both mammary cell types was highly effective in inhibition of cell viability (P < 0.05), whereas the application of an RAR antagonist stimulated MeBo cell viability (P < 0.05) and inhibited BME-UV1 cell viability (P < 0.05). Finally, the use of the RAR antagonist in conjunction with bLf indicated a rescue of the bLf effect in the MeBo cells, suggesting that bLf is acting through the RAR receptor. Conversely, bLf reverted inhibition of cell viability by 9cRA in the BME-UV1 cell type (P < 0.05). We conclude that RAR interaction in bovine mammary cell types regulates cell viability in vitro; we hypothesize that the natural ligands mediate regulation of bovine mammary cell viability in vivo and that bLf can either enhance or reverse the retinoid-induced inhibition of cell viability, depending on the type of bovine mammary cell studied.  相似文献

Despite a large amount of work on compensatory growth in pigs it continues to be poorly understood with many conflicting reports. The aim of this work was to conduct four similar trials using the same genotype of pig, facilities, feed and growth restriction period to determine whether it was possible to obtain consistent results using a constant trial set-up. A total of 576 pigs (Hampshire sire×(Large White×Landrace) dam) were used. Pigs were weaned onto trial at 26.8±0.11 (mean±SE) days of age at a mean weight of 8.1±0.07 kg and remained on trial until slaughter, approximately 150.9±0.66 days of age at a mean weight of 98.4±0.65 kg. In each of the four trials the restriction period was for 3 weeks immediately following weaning. Pigs received either a high (Control; 17.5 g/kg) or a low (weaner restrict (WR); 8.0 g/kg) lysine diet during these 3 weeks, all pigs then received a high lysine diet up until slaughter, 15.5 and 12.0 g/kg of lysine for the grower and finisher diets respectively. Pigs from trial 1 ate (P<0.001) and gained (P<0.001) more throughout the weaner stage than all other trials. Growth performance was successfully reduced during the weaner phase. WR pigs grew more slowly (P<0.001) and less efficiently (P<0.001) than Control pigs. WR pigs from trial 1 demonstrated compensatory gains throughout the grower stage, growing 6% faster than Control pigs from trial 1 due to an improvement in feed efficiency (P<0.001). WR pigs from trial 2 demonstrated compensatory gains during the finisher stage, increasing their rate of gain by 7.0% compared to Control pigs from trial 2 again due to an improvement in feed efficiency (P<0.1). However previous lysine restriction did not result in compensatory growth in trials 3 and 4. WR pigs from trials 3 (P<0.05) and 4 (P<0.1) had a lower feed intake compared to Control pigs during the grower stage and an overall lower lysine intake throughout the trial (P<0.05; P<0.05). Although pigs from trials 1 and 2 demonstrated compensatory growth following a reduction in performance when dietary lysine levels as low as 8.0 g/kg lysine were fed for a period of 3 weeks immediately post-weaning, pigs from experiments 3 and 4 did not, thus compensatory growth was not consistently observed.  相似文献

Effects of grain source and dietary oil supplement on dry matter intake (DMI), feeding, chewing behavior, and production performance of lactating dairy cows were evaluated using eight multiparous Holstein cows (77±22.1 days in milk; mean±SD) in a duplicated 4×4 Latin square design with a 2×2 factorial arrangement of treatments. Experimental diets contained either ground barley or ground corn supplemented with either fish oil or soybean oil at 2% of dietary dry matter (DM). Geometric mean particle size of dietary treatments was 4.1 mm. Dry matter intake tended (P=0.09) to be greater for barley- vs. corn-based diets (23.2 vs. 22.3 kg/d), but was reduced for the fish oil compared to soybean oil supplemented diets (21.1 vs. 24.3 kg/d; P<0.001). This reduction in DMI was attributed to smaller meal size (1.24 vs. 1.55 kg of DM; P=0.004) and slower eating rate (0.082 vs. 0.098 kg of DM/min; P<0.001) for fish oil compared to soybean oil supplemented diets. Main treatment effects interacted (P=0.005) for DMI of particles retained on 19 mm sieve but not for sorting index. Eating rate (0.090 kg of DM/min) was similar between barley- and corn-based diets, however, rumination time was greater for barley- compared to corn-based diets as result of longer rumination bout duration (32.5 vs. 28.5 min/bout; P=0.01). Barley- compared to corn-based diets increased total chewing time by 71 min (709 vs. 638 min) for cows fed fish oil, but not for cows fed soybean oil. Grain source did not affect milk yield or milk composition. Compared to soybean oil, fish oil negatively affected milk yield (40.4 vs. 43.4 kg/d; P=0.01), and thereby, both milk fat (0.91 vs. 1.26 kg/d; P<0.001) and protein (1.23 vs. 1.33 kg/d; P=0.007) production. However, feed efficiency (milk yield/DMI) was greater in fish oil compared to soybean oil supplemented diets (1.94 vs. 1.80; P=0.003). Results indicated that grain source and oil supplement can interact to affect feeding and chewing behavior, but not lactational performance of lactating cows.  相似文献

It is widely accepted that transport is stressful for horses, but only a few studies are available involving horses that are transported regularly and are accustomed to transport. We determined salivary cortisol immunoreactivity (IR), fecal cortisol metabolites, beat-to-beat (RR) interval, and heart rate variability (HRV) in transport-experienced horses (N = 7) in response to a 2-d outbound road transport over 1370 km and 2-d return transport 8 d later. Salivary cortisol IR was low until 60 min before transport but had increased (P < 0.05) 30 min before loading. Transport caused a further marked increase (P < 0.001), but the response tended to decrease with each day of transport. Concentrations of fecal cortisol metabolites increased on the second day of both outbound and return transports and reached a maximum the following day (P < 0.001). During the first 90 min on Day 1 of outbound transport, mean RR interval decreased (P < 0.001). Standard deviations of RR interval (SDRR) decreased transiently (P < 0.01). The root mean square of successive RR differences (RMSSD) decreased at the beginning of the outbound and return transports (P < 0.01), reflecting reduced parasympathetic tone. On the first day of both outbound and return transports, a transient rise in geometric HRV variable standard deviation 2 (SD2) occurred (P < 0.01), indicating increased sympathetic activity. In conclusion, transport of experienced horses leads to increased cortisol release and changes in heart rate and HRV, which is indicative of stress. The degree of these changes tended to be most pronounced on the first day of both outbound and return transport.  相似文献

The severity of host response to some disease agents differs between sexes and this dimorphism has been attributed to the immunomodulating effects of steroid hormones. Our objective was to determine in heifers whether the phase of estrous cycle affected immune response mediators after endotoxin challenge (LPS, 2.5 μg/kg BW, i.v.). Sixteen beef heifers (426 ± 9 kg) were reproductively synchronized with the two-injection protocol of dinoprost tromethamine (Lutalyse®, Pfizer) to establish diestrus and estrus stages of the estrous cycle. Heifers were challenged with LPS on day 3 (E, estrus; n = 8) or day 10 (D, diestrus, n = 8) after the last i.m. injection of Lutalyse®. In all heifers, plasma concentrations of tumor necrosis factor-α (TNF-α) peaked 2 h after LPS treatment (P < 0.01) and returned to basal level by 7 h. However, the integrated TNF-α response (area under the time × concentration curve, AUC) was greater in E than in D (P < 0.05). Plasma concentrations of nitrate + nitrite (NOx, an estimate of NO production) increased (P < 0.01) in all heifers at 7 and 24 h after LPS; plasma NOx AUC after LPS was greater in E than D (P < 0.01). Plasma xanthine oxidase activity (XO, a mediator of superoxide production) responses were also greater in E than D (P < 0.05). A companion LPS challenge study in steers validated that the protocol for and use of Lutalyse® did not affect any of the immune parameters studied in heifers in response to LPS. Results indicate that the underlying physiological attributes of the estrus and diestrus phases of the estrous cycle constitute a major source of variability in the magnitude of proinflammatory response to bacterial toxins like LPS.  相似文献

The aim of this study was to compare the growth, carcass, histochemical, and meat quality characteristics in Large White pig groups that were categorized by live weight (Heavy and Light) and type I fiber percentage (High and Low), a procedure which resulted in four groups (Heavy-High, Heavy-Low, Light-High, and Light-Low). As expected, the Heavy group showed heavier live weight (114 vs. 94.7 kg, P<0.001) and larger loin eye area (53.3 vs. 47.8 cm2, P<0.001), as well as, higher total number (1,223,000 vs. 1,140,000, P<0.05) and greater mean value cross-sectional area (CSA; 4031 vs. 3798 μm2P<0.05) of muscle fibers than the Light group. However, there were no significant differences in start and finish days among the groups (P>0.05). Heavier pigs harboring a higher percentage of type I fibers (HH) exhibited a similar mean CSA (3894 vs. 4101 μm2) and total number (1,249,000 vs. 1,198,000) of muscle fibers, even though these pigs had a greater CSA of type I fibers (3181 vs. 2719 μm2, P<0.05) and a smaller CSA of type IIB fibers (4048 vs. 4457 μm2, P<0.05) compared to heavier pigs harboring a lower percentage of fiber type I (HL). Both the HL and Light-Low groups exhibited a rapid decline of muscle pH at the early postmortem period (5.90 and 5.85 vs. 6.08, P<0.05), paler surfaces (43.07 and 43.55 vs. 40.73,P<0.05), and higher degrees of fluid loss by exudation (6.26 and 6.39 vs. 4.22%, P<0.05) compared to the HH group due to their muscle fiber type composition. Thus, the HH pigs showed better meat quality characteristics without significant differences in growth and carcass performance compared to the HL pigs. Therefore, selection for increased live weight at the same age and muscle fiber characteristics, especially the increased type I fiber CSA and proportion, is one of the relevant indicators to improve and control meat quality without reducing the growth and carcass performance.  相似文献

The effects of fibre in diets for periparturient sows are poorly documented. Three weeks before farrowing, 41 sows (LACT) were fed a diet containing 3.8% crude fibre. Other 40 sows (FIBRE) received a diet containing 7% crude fibre. We estimated the intestinal activity of the sows with a daily qualitative evaluation of their faeces. The FIBRE group had a qualitative faeces score value of 2.1 ± 1.3 and the LACT group had a value of 1.2 ± 1.1 (P < 0.001). Individual daily water consumption was higher in the FIBRE group than in the LACT group (P < 0.001). Piglet weight gain at day 5 was higher in the FIBRE group (P < 0.05). The energy balance related parameters did not differ between the treatments.Concluding, diets containing more fibre can be successfully used around farrowing reducing prolonged constipation of sows with no negative effect on their energy balance related parameters.  相似文献

Different vaccination strategies against Mycoplasma hyopneumoniae have been adopted worldwide. Reports from the field indicate varying levels of protection among currently available vaccines. The goal of the present study was to compare the efficacies of three widespread commercial vaccination strategies against M. hyopneumoniae under field conditions. 20 farms were included. 14 farms used different single dose vaccines (vaccine 1 [V1], 8 herds; vaccine 2 [V2], 6 herds); another 6 farms (V3) used a two dose vaccination strategy. Gross lesions of 854 lungs and histopathology from 140 lungs were quantified, and a quantitative PCR was applied to detect M. hyopneumoniae and porcine circovirus 2 (PCV2) DNA in lung tissue (n = 140). In addition, porcine reproductive and respiratory disease virus (PRRSV), swine influenza virus (SIV), Actinobacillus pleuropneumoniae, Haemophilus parasuis and Pasteurella multocida were tested by qualitative PCR. 53% of lungs were positive for M. hyopneumoniae. 55.9% of lungs showed macroscopic enzootic pneumonia (EP)-like lesions. Lung lesion scores (P < 0.001) and M. hyopneumoniae-loads (P < 0.008) differed significantly among the vaccination groups, with the most severe cases and highest amounts occurring in V1. Histological alterations differed (P < 0.001) between V1 and V3. Lung lesion scores and histopathological changes were significantly correlated, with prevalence and load of M. hyopneumoniae indicating that the applied diagnostic tools are valuable in confirming the prevalence and severity of M. hyopneumoniae infections. Comparing different vaccination strategies against M. hyopneumoniae indicates varying levels of protection. M. hyopneumoniae is still a major problem despite the widely applied vaccination.  相似文献

The temporal pattern and sex effect of immune and stress hormone responses to a lipopolysaccharide (LPS) challenge were assessed using a pig model. Secretion of the pro-inflammatory cytokines tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 increased in a time-dependent manner following LPS infusion. There was also a time-dependent increase in secretion of the stress-related hormones cortisol, epinephrine (E), and norepinephrine (NE) following LPS, with peak concentrations attained within 30 min. The magnitude of the TNF-α and IL-1β responses were both positively associated (P < 0.05) with the magnitude of cortisol response following LPS, whereas serum IL-1β and IL-6 were positively correlated with the magnitude of E and NE responses following LPS. Acute-phase protein production was also time-dependently increased following LPS. The concentration of immune cells in circulation was decreased (P < 0.05) at 5.5 h post-LPS and negatively correlated with pro-inflammatory cytokine production. By 24 h post-LPS, immune cell counts increased (P < 0.05) and were positively associated with both pro-inflammatory cytokine and stress hormone production. The amplitude of pro-inflammatory cytokine response following LPS was affected (P < 0.05) by sex classification; however, the magnitude of elevated cytokine concentrations was not. The magnitude of the NE response, but not of the E and cortisol responses, to LPS was influenced by sex (P < 0.05). Similar to the pro-inflammatory cytokines, the magnitude of exposure to the stress hormones following LPS was not influenced by sex. The production of serum amyloid A (SAA) was influenced by sex, with barrows producing more SAA than gilts at 24 h post-LPS (P < 0.05). Collectively, these results demonstrate sex-specific, concomitant temporal changes in innate immune- and stress-related hormones.  相似文献

The aim of this study was to investigate total antioxidant (TAC), and oxidant capacity (TOC) and nitric oxide (NO) levels in milk of cows with subclinical mastitis. Brown Swiss and Holstein breed cows were screened with California Mastitis Test (CMT) to determine mammary glands with subclinical mastitis. Moreover, somatic cell counts (SCC) were determined electronically in all milk samples. Mammary quarters were classified as healthy (n = 25) or subclinical mastitis (n = 35) based on CMT scores and somatic cell count (SCC: ?200,000/ml or >200,000/ml) in milk. Nitric oxide, TOC and SCC levels were significantly higher (< 0.001, < 0.005 and < 0.001, respectively) in milk from mammary quarters with subclinical mastitis compared to those from healthy mammary quarters. In conclusion, subclinical mastitis results in higher NO concentrations, TOC and SCC, and NO and TOC were positively correlated with SCC. The performance of spring calving Holstein-Friesian (HF) and Jersey×Holstein-Friesian (J×HF) dairy cows was examined during three successive years (mean of 35 HF cows and 31 J×HF cows per year). Throughout the experiment cows were managed on one of three grassland-based systems of milk production, namely low concentrate (LC), medium concentrate (MC) or high concentrate (HC). Post-calving, cows were housed and offered grass silage, supplemented with 6.0, 8.0 and 10.0 kg concentrate/cow/d in systems LC, MC and HC, respectively (mean period from calving to start of full time grazing, 69 days). During the grazing period target concentrate feed levels were 0, 2.5 and 5.0 kg/cow/d for systems LC, MC and HC, respectively (mean period from start of full time grazing to start full time re-housing, 206 days). Full lactation concentrate inputs were 530, 1092 and 1667 kg/cow, in systems LC, MC and HC, respectively. There were no significant genotype×system interactions for any of the milk production parameters examined. Food intake during the confinement and grazing periods was unaffected by genotype. Milk yield was highest with HF cows while milk fat and milk protein content were highest with the J×HF cows (P<0.001). Genotype had no effect on fat plus protein yield. Milk yield and fat plus protein yield were higher with systems MC and HC than with LC (P<0.001). HF cows were on average 44 kg heavier than J×HF cows, while the mean condition score of the J×HF cows was approximately 0.2 units higher than that of the HF cows (P<0.001). Live weight and condition score changes during the lactation followed similar trends with both genotypes. The J×HF cows had fewer days to first observed heat (P<0.05), a higher conception rate to first service (P<0.01), first plus second service (P<0.001), and a higher pregnancy rate at the end of the breeding season (P<0.05). Although mean somatic cell score was unaffected by genotype, the proportion of cows with one or more cases of mastitis was lower with the J×HF cows (P<0.05). In summary, while the J×HF cows had improved fertility performance compared to the HF cows, both genotypes exhibited similar levels of tissue mobilisation and deposition throughout the lactation, while there was no evidence of a genotype×grassland system interaction for any of the milk production parameters examined.  相似文献

Two basal diets containing an adequate and a low supply of available P (4.7 vs 2.5 g/kg diet) and three others containing a low available P but supplemented with 250, 500 and 12,500 units of phytase (FTU) per kg diet, respectively, were prepared. Each diet was fed ad libitum to birds in six metabolism cages (five birds in each cage) in a randomised block design. It was found that birds fed a high (12,500 FTU) dosage of phytase grew faster (P < 0.001) and retained proportionally more carotenoids in the liver (P < 0.05) compared to birds fed unsupplemented diet. The results suggest that dietary phytase may improve the anti-oxidative status of birds fed low P diets through enhancing dietary carotenoid availability.  相似文献

The effects of fasting on insulin-like growth factor (IGF)-I, IGF-II, and IGF-binding protein (IGFBPs) mRNA in channel catfish were examined. Fed control fish (Fed) were compared to fish that had been fasted for 30 d followed by 15 d of additional feeding (Restricted). Sequence alignment and similarity to orthologous proteins in other vertebrates provided structural evidence that the 3 catfish sequences identified in the present research were IGFBP-1, -2, and -3. Prolonged fasting (30 d) reduced body weight approximately 60% (P < 0.001) and decreased IGF-I mRNA in the liver and muscle (P < 0.01). Fifteen days of re-feeding restored concentrations of hepatic and muscle IGF-I mRNA. Liver IGF-II mRNA was not affected by fasting but was increased 2.2-fold after 15 d of re-feeding (P < 0.05). Abundance of muscle IGF-II mRNA was similar between the fed control group and the restricted group throughout the experimental period. Fasting also increased liver IGFBP-1 mRNA (P < 0.05) and decreased IGFBP-3 mRNA (P < 0.01), whereas abundance of IGFBP-2 mRNA was not significantly affected. Interestingly, re-feeding for 15 d did not restore concentrations of IGFBP-1 and IGFBP-3 mRNA relative to fed control concentrations. The IGF results suggest that IGF-I and IGF-II are differently regulated by nutritional status and probably have a differential effect in promoting muscle growth during recovery from fasting. Similar to mammals, IGFBP-1 mRNA in catfish is increased during catabolism, whereas IGFBP-3 mRNA is decreased during inhibited somatic growth. The IGFBP results provide additional evidence of the conserved nature of the IGF-IGFBP-growth axis in catfish.  相似文献
