The metabolism of fats,proteins and carbohydrates and change of enzyme ofseeds of pinus bungeana during the germination were conducted by the methods of gas-—liquid chromatography,flow injection,colorimetric analysis of spectrophotometer,toprovide theoretical basis for seeds dormancy,germination and storage of seeds of foresttree.The results indicate that(1)carbohydrates were first utilized during germination ofseed of pinus bungeana;(2)stored substances in seeds began to decompose quickly afterradicale broke through seed coats;(3)the activity of enzymes in the seeds does not alwayscoincides with the increase or decrease in quantity of its responsible substance duringgermination of the seed.Changes in stored substance relate to metabalism of othermetabolism of other materials and the use of hydrolysates.  相似文献

Juniperus procera Endl. is economically important timber species, but its populations are extremely small and fragmented in its natural habitat, thus, calling for immediate ex situ conservation. Here we examined the effects of seed sources and storage temperature on the longevity of Juniperus procera seed lots through collection and preservation of seeds in seed banks. Seeds were collected from nine sites across the species natural distribution in Ethiopia and stored in four warehouses: modern cold room (5℃), mud house (15℃), concrete block house (17℃ or corrugated iron house (20℃) for 42 months. Every three months, a random sample of stored seeds were drawn and tested for germination. A highly significant variation (p < 0.01) in germination of stored seeds was observed among different storage environments, seed lots, and duration of storage. Over the storage period, seeds stored in the cold room had the highest mean percentage germination, followed by the mud house, corrugated house and blocket house. The cold room (41%) and the mud house (38%) maintained the same level of germination as the intitial germination of the seedlotds (42%). The variation in longevity of stored seeds was significnatly correlated with the initial germination of seed lots (r > 0.80; p < 0.01). Cold storage also resulted in enhancement of germination through its stratification effect that terminated the non-deepphysiological dormancy of juniper seeds. In conclusion, seed lots with good initial germination can be effectively stored in cold room (5℃) up to four years. In the absence of modern cold stores, mud houses can be used as a good alternative to store seeds at local level.  相似文献

We performed desiccation and storage trials to better under- stand storage behavior of Knema attenuata seeds. Mature seeds with moisture content (MC) of 31% exhibited 73% germination. During the period of desiccation (open lab condition) seeds with MC 23% showed 40% germination. After further drying to MC 21% germination was reduced to 16%. Complete loss in viability resulted when seed moisture was reduced to 18%. The seeds stored at -10°C, 0°C, 10°C and 28±2°C (open lab condition) lost their viability within 10 days. Seeds stored in sealed polythene bags and moist sand retained viability for more days than did seeds stored under all other storage conditions. Sensitivity of seeds to lower temperature and desiccation suggest that the storage be- havior of K. attenuata seeds is recalcitrant. Seeds stored in moist condi- tions can, at best, be stored for a period of two months.  相似文献

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献

Seed germination process has closely relation with material transformation and energy exchange within the seed. Study on its thermal effect is important for understanding the mechanism and the influencing factors of the seed germination. The thermogenetic curves of seed germination ofRobinia pseudoacacia was measured by a new-type conductive microcalorimeter made in Wuhan University. The relationship was analyzed between the germination thermogenetic regulation and seed germination physiology. The thermogentic curves were further analyzed by thermokinetic theory to obtain the dynamic parameters and the thermokinetic model on seed germination ofRobinia pseudoacacia. Seeds of Pinus bungeana are very dormant due to the impermeability of their seed coats. We have found that treatment with microorganisms (Trichoderma-4030) can effectively soften the seed coats, and thus promote germination. The first record of a “field” infestation of stored lettuce seeds (Lactuca sativa) byTineola bisselliella (TB) is reported. The pest is able to consume almost the whole kernel ofL. sativa including the shell. The ability od TB to damage selected stored cereal, pulse and oil seeds under laboratory condition was evaluated. In maize and wheat, germ area was damaged preferably and the germination decreased considerably. TB was unable to infest and damage sound seeds of rape and beans ofPhaseolus vulgaris and soya. These findings enabled the development of a vegetarian diet for laboratory rearing of TB. The pest status and hazards resulting from cross-infestation is discassed.  相似文献

Dimorphandra jorgei MF Silva (Caesalpinaceae) and Swartzia macrostachya Benth. var. riedelii Cowan (Fabaceae) are tree species from the southern Bahian Atlantic Forest, identified for the restoration of degraded areas. The objective of this research is to study their seed germination and seedling growth to develop simple methods for seedling production in small scale nurseries. Because the species have distinct dormancy and germination characteristics, the seeds were submitted to different treatments. D. jorgei seeds were immersed in hot water, scarified and stored for 12 weeks in the laboratory environment (25°C). S. macrostachya seeds were submitted to different combinations of drying and storage in the laboratory environment or refrigerator (4°C). Seedlings were submitted to two light treatments for 3 months: partial shade (52% of full sun) and full shade (12% of full sun). In D. jorgei, the highest germination occurred for scarified seeds (87%) and for seeds stored for 12 weeks and then scarified (69%). In S. macrostachya the undried control seeds had the highest germination, followed by that of the refrigeration storage treatment, without predrying (99% and 97%, respectively). The average values for the growth parameters were greater in full shade for D. jorgei, while S. macrostachya seedlings showed greater growth rates in partial shade. Two months growth is recommended before D. jorgei seedlings are of the sufficient size and leaf number for field planting. On the other hand, a period of 4 months is recommended for S. macrostachya, due to its relatively slow growth.  相似文献

Germination tests in the three sympatricCarpinus species,C. laxiflora, C. tshonoskii, andC. cordata, were carried out to clarify their germination traits in relation to the regeneration strategies. Seeds did not germinate just after ripening under any conditions in the three species. After one or two rounds of cold stratification, however,C. laxiflora andC. tshonoskii showed high germination percentages (>80%) at alternating temperature in light whileC. cordata did not. After a long-term (10 months) cold stratification, about 100% ofC. cordata seeds germinated, although about 70% of these seeds were in “conditional dormancy” and cannot germinate in the dark. Seed germination in the threeCarpinus species was enhanced by alternating temperature, but not by light exposure. These results suggest that the seeds of the three species are dormant just after ripening and the dormancy is broken by cold stratification. with intra-generic variations in the breakage. Germination traits in each species may be closely related to the regeneration strategy;C. cordata maintains a persistent seed bank with seed dormancy and regenerates after disturbances as a seed bank strategist, whereas the other two species germinate in the next spring after seedfall and regenerate depending either on existing disturbed-sites or sites of recent disturbances after seedling establishment as a seed rain strategist (C. tschonoskii) or seedling bank strategist (C. laxiflora). Throughout the southeastern United States, Rhus and Rubus species are common associates of the southern pines on a wide array of upland site and stand conditions. Because of their ability to overrun disturbed sites, these species are categorized as competitors to pine during stand regeneration. Since prescribed burning is often used for site preparation in advance of pine regeneration, this study investigated the effect of fire on the germination of seeds from three pine competitors (Rubus argutus Link, Rhus copallina L. and Rhus glabra L.). During dormant-season burns, sumac seeds were located 45 cm above litter, within the F layer of a reconstructed forest floor, and at the interface of the forest floor and mineral soil. During growing-season burns, fresh blackberry fruits were placed at heights of 0, 15, 30, and 45 cm above the surface litter of a reconstructed forest floor. In subsequent germination tests, sumac seeds from within the F layer of burned litter had significantly higher germination rates for smooth sumac (31%) and shining sumac (42%) as compared to unburned control seeds (1–5%). In general, germination rates for sumac seeds placed in the air or on mineral soil during burning were no better than control seeds. Seeds from blackberry fruits that were located at heights of 15, 30, and 45 cm had germination rates that were comparable to unburned control seeds (18%), but seeds from fruits placed on the litter during burning had <1% germination. Results suggest that sumac seed germination may be enhanced by the heat from prescribed burning, whereas blackberry seeds showed more germination response to multiple germination cycles which indicated a potential for long-term storage in the soil seed bank.  相似文献

Abstract

The effect of soaking on germination and occurrence of fungal infections on Norway spruce (Picea abies (L.) Karst.) seeds and germinants was studied in three commercial seed lots. Treatments in which seeds were soaked in water for 24 h or in which the water was changed during the soak did not have an effect on the species diversity or on the abundance of the fungi isolated from the seeds. Different fungi were found in different seed lots. Most of those isolated are saprophytic or weakly pathogenic, but pathogens such as Sirococcus conigenus (D.C.) P.F. Cannon &; Minter and Trichothecium roseum (Pers.) Link were also isolated. Soaking increased germination energy but had no effect on final germination percentage. The number of mouldy seeds and germinants with disease symptoms was different among seed lots when seeds were germinated on water agar. When germinated in low humified Sphagnum peat, no differences in the emergence of disease symptoms were observed among seed lots. The frequency of disease was lower in peat than in water agar. Soaking had no effect on disease emergence in germination trials on either water agar or in Sphagnum peat.  相似文献

Albizia saman (Jacq.) F. Muell. commonly known as rain tree seeds were treated with five pre-sowing treatments to study the effect of pre-sowing treatments on germination and initial seedling development in the nursery.The experiment was established in the nursery of the Institute of Forestry and Environmental Sciences.University of Chittagong, Chittagong, Bangladesh. Results revealed that Nail clipping in one side of the seed (at the distal end of the seed) (T4) provides the highest (50%) seed germination. The second highest germination (42%) was obtained for the seeds treated with immersion in cold water for 24 h (T1). Germination was completely inhibited when the seeds immersed in boiled water for 30 s followed in cold water soaking for 24 h. Other germination parameter and initial morphological growth and biomass production of the seedlings was also higher for the treatments T4and Ti in comparison to the control (Ta)treatment. Pre-sowing treatments of T4 e.g. Nail clipping in one side of the seed (at the distal end of the seed) and T1 (Seeds immersed in cold water for 24 h) may be recommended for maximum germination and initial vigorous seedlings growth of Albizia sarnan in the nursery.  相似文献

We measured physiological parameters including water uptake, in-vitro embryo germination ratio, and seed coat structure observed by scanning electron microscopy (SEM) to explore the influence of seed coat on the germination of seeds of tallow tree (Sapium sebiferum (Linn) Roxb.). Tallow tree seeds had good water permeability. We found that germination of cabbage seeds was inhibited when cabbage seeds were soaked in extracted solutions from tallow tree seed coat. Seed coat structure at the side of the radicle appeared to be a barrier to seed germination. We tested methods to break tallow tree seed dormancy. Dormancy of tallow tree seeds was overcome by soaking the seeds in 500 mg·L-1 or 1000 mg·L-1 GA3, followed by 100 days of cold stratification.  相似文献

The effect of seed water content (WC) (2–3, 5–6 and 22–25%, on a fresh weight basis), storage temperature (+4, −20, −80 and −196°C) and storage duration (6, 12, 24, 48 and 60 months) on the germination of white spruce (Picea glauca (Moench) Voss), black spruce (Picea mariana (Mill.) B.S.P.) and lodgepole pine (Pinus contorta Dougl. ex Loud. var. latifolia Engelm.) seed was investigated. Germination of white spruce control (untreated) seeds and seeds adjusted to 2–3% and 5–6% WC declined after 48 months of storage at −80 and −196°C, with a further decline at 60 months at −20, −80, −196°C. Germination remained high when control white spruce seeds and seeds with 2–3, 5–6% WC were stored at +4°C, over all storage durations. Generally, black spruce and lodgepole pine exhibited high germination at all storage temperatures at 2–3% and 5–6% WC as well as the control (untreated) seed, for up to 60 months in storage. Germination declined for all three species when seed was conditioned to 22–25% WC. This loss in germination was partially recovered in white spruce seed stored at +4, −20 and −80°C after storage durations of 24, 12 and 48 months, respectively, and in black spruce seeds stored at −20 and −196°C after storage durations of 24 months. Mean germination time (MGT) was relatively constant for all species, under all conditions, except for seed conditioned to 22–25% WC, where MGT increased for white spruce seed stored 48 months at −80 and −196°C, and for black spruce seed stored 24 months at +4 and −80°C and 60 months at −196°C. These results show that the optimal storage temperatures are 4°C for white spruce, and 4, −20, −80, and −196°C for black spruce and lodgepole pine, and 2–6% water content is optimal for all 3 species at these temperatures.  相似文献

Allanblackia parviflora A. Chev, is an indigenous fruit tree species that could be used in agroforestry systems with both environmental and economic benefits. The seed oil is of prime importance as a foreign exchange earner and is being developed as a rural based enterprise in many African countries notably Ghana, Nigeria, Cameroon and Tanzania. Methods for propagation and conservation of the species are therefore of prime importance. The seeds of the species are however difficult to germinate hindering its domestication process. Allanblackia parviflora seeds were collected from seven different populations in Ghana and were subjected to four different seed germination trials at the nursery of CSIR-Forestry Research Institute of Ghana at Fumesua, Kumasi. This was followed by studying the effect of stage of seed germination on establishment after potting. Intact seeds began germination at 7 months after sowing and at 24 months, significant (P < 0.001) variations were observed in the seed germination ability of 74 accessions. Mean seed germination ranged from 0 to 35%. Significant differences in seed germination percentages among populations (P < 0.001) and provenances (P < 0.05) were also observed. Removal of seed coat significantly (P < 0.001) enhanced seed germination. Germination percentages of seeds with seed coat removed ranged from 6 to 43% while seed germination percentages for intact seeds were less or equal to 3%. Incubation of seeds with seed coat removed in polythene bags gave an added advantage. Seeds with seed coat removed and kept in polythene bags started germinating from 2 weeks and within 10 months, 75 and 68% germination were obtained for plane and black polythene bags, respectively. Removal of germinants and potting when shoot length was at least 1 cm gave significantly higher growth (P < 0.001 for shoot height, P < 0.05 for stem diameter, P < 0.001 for number of leaves produced) compared to when only radicle had emerged. It became evident in this study that seeds should be classified as fully germinated only when shoots begin to grow.  相似文献

用剪刀剪、锉子锉、与瓜子石混搓等机械处理法以及用5%氢氧化钾、98%浓硫酸等化学处理法对神黄豆种子进行预处理,3倍种子体积沸水浸泡并自然冷却浸种48 h,观察并统计种子吸胀粒数、种皮破裂数后置床进行发芽测定。结果表明,除与瓜子石混搓处理外,其余处理均能促进种子吸水膨胀,显著提高神黄豆种子发芽率。其中以98%浓硫酸浸泡2 h处理效果最好,其吸胀率达98.0%、发芽率最高,达88.3%,吸胀破皮率、不正常发芽率均为0%;其次是锉子锉处理效果好,其吸胀率达99.0%,发芽率可达84.7%,吸胀破皮率、不正常发芽率均为0%;用5%氢氧化钾进行浸泡处理,对种子发芽有一定的促进作用,但发芽不整齐,发芽速度慢,处理14 d吸胀率为59.7%、发芽率最高为48.0%;与瓜子石混搓处理效果相对较差,对神黄豆种子发芽的促进作用不明显,吸胀率为5.0%、发芽率为5.0%。  相似文献

Leucaena leucocephala and Acacia farnesiana are tree species used for several agricultural purposes in the Mediterranean region. The seeds of these species exhibit dormancy, causing delayed germination. Two experiments were conducted to investigate the effect of pre-sowing treatments (scarification, hot water, or soaking) on seed germination of L. leucocephala and A. farnesiana. In one experiment, seeds were exposed to three pre-sowing treatments: control, sandpaper scarification, or soaking in 70°C water for 4, 8, 12, 16, 20, or 24 min. In another experiment, seeds were soaked in 70°C water for 20 min, and then soaked in water at room temperature for an additional 24, 48, or 72 h or blade scarified. In general, soaking the seeds of the two species in hot water was more effective in breaking seed dormancy than scarification. Sandpaper scarification was not effective for either species. Blade scarification increased A. farnesiana seed germination to 56%, indicating that seed dormancy was mainly a consequence of hardseededness. L. leucocephala seeds collected from Jordan University of Science and Technology (JUST) site and soaked in 70°C water for 20 min and then soaked for 24, 48, or 72 h had germination rates above 97%. Our results suggest that blade scarification of A. farnesiana seeds and soaking of L. leucocephala seeds in 70°C water for 20 min are effective treatments to break seed dormancy and enhance seed germination of these vital species.  相似文献

Abstract

The response of common alder [Alnus glutinosa (L.) Gaertn.] and downy birch (Betula pubescens Ehrh.) seeds to germination temperature was examined following chilling and priming. Seeds of two seed lots of each species were subjected to combinations of chilling (4±1°C) and priming (20°C) treatments in fully imbibed (FI) state or a lower target seed moisture content (TMC) level (30% and 35% in alder and birch, respectively). After treatment, the seeds were allowed to germinate for 56 days at constant temperatures of 7.5, 10, 15, 20, 25 or 30°C. The response to temperature and pretreatment differed between species, but the effect was consistent in each seed lot within each species. In alder, the TMC seeds germinated well across the full range of temperatures, whereas there was an optimum temperature (22–23°C) for seeds given the FI pretreatment. Priming had no significant effect on the germination response of the TMC seeds in alder, but priming greatly improved germination in the FI seeds, especially at the lower germination temperatures (optimum 18–19°C). In contrast, in birch, the TMC seeds germinated better across the full temperature range, but the optimal germination temperature (15°C) was the same for all seed pretreatments. Priming improved germination in both the FI and TMC seeds in birch.  相似文献

The germination of Casuarina equisetifolia Forst seed lots is low (40–50%) even when freshly collected. This study aimed at upgrading the germination performance by removing non-germinable seeds using petroleum flotation technique. Seeds collected from 19 individual trees in seedling seed orchards in India were assessed using X-ray to determine the proportion of different seed lot fractions. The feasibility of petroleum ether as a separation medium was tested. Morphological characters of floating and sunken seeds were further studied using image analysis technique to examine their effects on the separation efficiency. The X-ray analysis revealed a large quantity of shrivelled, empty and insect-damaged seeds, altogether accounting 50%, which were the causes of low percentage germination of un-graded seed lots (48%). Petroleum flotation resulted in 90% germination in the sunken fraction and 4% in the floating fraction. The separation was distinct for most seed lots, except few seed lots that had relatively low germination in the sunken fractions and more than 10% germination in the floating fractions. As a whole, petroleum flotation appears to be a feasible technique to upgrade the germination of C. equisetifolia seed lots, and its efficacy is influenced by wing surface area, seed density and wing quotient of filled and empty seeds.  相似文献
