Isolation and characterization of cellulase‐producing aeorobic bacterial flora in the intestine of omnivorous tilapia (Oreochromis mossambica) and phytophagous Chinese grass carp (Ctenopharyngodon idella) have been carried out using selective carboxymethylcellulose‐agar (CMC‐agar) medium. The cellulolytic activity was measured both qualitatively and quantitatively. It was found that the ability of different strains in degrading cellulose varies within a wide range. Among the strains isolated from the gut of each test fish, TM1 and CI3 isolated from O. mossambica and C. idella, respectively exhibited maximum cellulolytic activity (67.02 and 35.8 U mL?1 respectively). Pure cultures of these strains were selected for morphological, physiological and biochemical characterization. On the basis of these tests, the isolated strains were identified as Bacillus circulans (TM1) and Bacillus megaterium (CI3). Both the strains are rod‐shaped, motile and show better temperature (15–42°C) and pH (5–11) tolerance. The selected strains were further quantitatively assayed for amylase and protease activities. Maximum amylase and protease activities were exhibited by TM1 and CI3 respectively. Information generated from the present study might contribute towards better‐feed formulation incorporating plant ingredients.  相似文献

实验对野生和养殖银鲳胃、幽门盲囊、前肠、中肠、后肠菌群结构进行了定性对比分析,并对产蛋白酶、淀粉酶、脂肪酶、纤维素酶的菌株进行了鉴定。结果发现,野生和养殖种群的消化道菌群结构存在较大差异,且同一种群消化道各部分之间菌群结构也存在较大差异。尽管养殖和野生银鲳均在幽门盲囊中具有最多的可培养细菌菌株,但野生银鲳消化道内主要菌群为嗜冷菌属(Psychrobacter)和Pseudochrobactrum,养殖银鲳消化道主要菌群为不动杆菌属(Acinetobacter)和假单孢菌属(Pseudomonas),两种群共有细菌仅一株,即Psychrobacter piscatorii strain VSD503,但其分别存在于野生与养殖种群银鲳消化道的不同部位。在产酶菌株筛选中发现,野生银鲳消化道内分离到16株产酶菌,其中44%可培养菌能产蛋白酶,56%能产淀粉酶,11%能产脂肪酶,56%能产纤维素酶,部分菌株可产2株以上的消化酶,其中产3种以上酶的菌株有5株,且产酶量丰富。相对于野生银鲳,养殖银鲳消化道内分离到22株产酶菌,主要以产蛋白酶和淀粉酶为主,70%可培养菌可产蛋白酶,21%可产淀粉酶,仅Bacillus thuringiensis strain VITGS可产纤维素酶,无一株菌产脂肪酶,其中只有Bacillus thuringiensis strain VITGS产3种酶但产酶量相对较少。研究可为银鲳人工养殖中潜在益生菌的筛选提供理论依据。  相似文献

为了解近江牡蛎养殖水体中细菌的产酶特性 ,筛选出有应用潜力的有益菌 ,本实验从近江牡蛎养殖水体中分离到 32株菌。革兰氏染色表明 ,10株为革兰氏阳性菌 ,2 2株革兰氏阴性菌。在此基础上研究了它们蛋白酶、脂肪酶、淀粉酶和纤维素酶的产酶能力。结果表明 ,6 1.9% ( 17株 )的菌株能分泌蛋白酶 ,71.9% ( 2 3株 )菌株能分泌脂肪酶或淀粉酶 ,34.4 % ( 11株 )的菌株产纤维素酶。这些产酶菌株均以革兰氏阴性菌为主。在这 32株菌中 ,产四种酶的有 5株菌 ,产三种酶的有 9株菌 ,产两种酶的有 9株菌 ,产一种酶的有 9株菌 ,也就是说 ,所有的菌株都能产酶。由此表明近江牡蛎养殖水体中细菌在该环境物质循环中的重要地位。  相似文献

Enzymes synthesized by the gut microbiome are recognized as vital for assisting host metabolism. This study was conducted to screen and identify bacterial isolates with enzyme-producing abilities from the intestine of milkfish Chanos chanos. Overall, 114 isolates were collected, of which 58% showed clear zones indicating various enzyme activities via plate-based assays. Specifically, 65 isolates were amylolytic, 1 isolate was cellulolytic, and 18 isolates exhibited both activities. The two most promising isolates—CC8 and CC27—were further subjected to spectrophotometric enzyme assays to determine their amylase, cellulase and protease activities. Results showed that amylase, cellulase and protease activities (6.00 ± 0.01, 0.52 ± 0.02 and 18.62 ± 0.16 U mg?1 protein, respectively) were higher in CC8 than in CC27. Using 16S rRNA gene sequencing, the isolates were putatively identified as Vibrio sp. and Bacillus cereus respectively. At a suspension (mL) to feed (g) ratio of 3:25, dietary supplementation of putative probiotics was carried out to evaluate the growth performance, survival and enzymatic activities of juvenile Nile tilapia Oreochromis niloticus. Results revealed that probiotic-supplemented fish exhibited better growth performance—final average body weight, weight gain, specific growth rate and feed conversion ratio—along with increased intestinal enzymatic activities compared with the control (p < 0.05). Survival rates ranged from 80% to 90% and were statistically similar in all treatments (p > 0.05). This study concludes that the application of Vibrio sp. CC8 and Bacillus cereus CC27, supplemented either in monostrain or multistrain preparation, can promote growth and enzymatic activities in juvenile Nile tilapia.  相似文献

Herbivorous grass carp (Ctenopharyngodon idellus) has a powerful capability to digest cellulose from aquatic plants, depending on the cellulase complex produced by the cellulolytic bacterial community in the gastrointestinal (GI) tract. However, it remains uncertain which bacteria taxa may actively participate in the digestion of food fibre. In this study, a total of 499 cellulolytic bacteria from the gut content of grass carp fed on Sudan grass (242 strains) and artificial feedstuffs (257 strains) were randomly isolated and characterized using carboxymethyl‐cellulose, microcrystalline cellulose and cellobiose agar media. The results showed that more than half of the isolates were capable of degrading carboxymethyl‐cellulose and cellobiose, while the remaining isolates were restricted to microcrystalline cellulose decomposition, exclusively. The cellulolytic bacterial community was dominated by Aeromonas, followed by Enterobacter, Enterococcus, Citrobacter, Bacillus, Raoultella, Klebsiella, Hydrotalea, Pseudomonas, Brevibacillus and some unclassified bacteria, as revealed by 16S rDNA sequence analysis. Notably, grass carp fed on grass with high‐fibre content harboured a higher diversity of cellulolytic bacteria than the ones fed on low‐fibre feedstuffs. Our results provided evidence for a positive correlation between the content of food fibre and the diversity of cellulolytic bacteria in grass carp intestines. Thus, improving growth conditions and cellulase activities for GI cellulolytic microorganisms in grass carp intestines are critical for effective utilization of feedstuffs containing high fibre levels.  相似文献

Isolation and enumeration of phytase‐producing bacterial flora in the foregut and hindgut regions of the gastrointestinal tracts of 10 culturable freshwater teleosts of different feeding habits, namely rohu (Labeo rohita), catla (Catla catla), mrigal (Cirrhinus mrigala), bata (Labeo bata), kalbasu (Labeo calbasu), Nile tilapia (Oreochromis niloticus), climbing perch (Anabas testudineus), common carp (Cyprinus carpio), silver carp (Hypophthalmichthys molitrix) and grass carp (Ctenopharyngodon idella), have been carried out. Microbial culture of the gut mucosa on selected nutrient media following the enrichment culture technique was performed for bacterial isolation. The bacterial isolates were screened on the basis of their enzyme‐producing ability. The bacterial population on the tryptone soya agar (TSA) plate was maximum in the hindgut region of bata, followed by mrigal and minimum in the foregut region of Nile tilapia. In modified phytase screening medium (MPSM), phytase‐producing strains were recorded at higher densities in the foregut region of mrigal and grass carp and minimum in the foregut region of bata. In case of the hindgut, maximum phytase‐producing strains were present in grass carp and mrigal and minimum in rohu. In general, in MPSM, the bacterial population was lower in the hindgut region of all the 10 species of fish examined. The phytase‐producing ability of the selected 31 strains (16 from the foregut and 15 from the hindgut region) was determined by clearing zones on phytate‐containing plates. Among these isolates, 22 strains (12 from the foregut and 10 from the hindgut region) were selected as potent phytase producers according to a quantitative enzyme assay. The highest phytase activity was observed in the bacterial strains LF1 and LH1 isolated from the fore and the hindgut regions of rohu respectively. Both the strains were identified as Bacillus licheniformis on the basis of phenotypic characteristics as well as 16S rDNA sequence analysis.  相似文献

从汕尾健生鲍鱼场养殖水体和九孔鲍肠道中共分离筛选到25株弧菌,其中14株来自成鲍肠道,11株来自养殖水体,并对它们产多种胞外酶的能力进行了比较分析。试验结果表明:78.6%的肠道弧菌能分泌蛋白酶,21.4%能分泌脂肪酶,64.3%能分泌卵磷脂酶或淀粉酶,92.9%能分泌明胶酶;水体中的弧菌63.6%能分泌蛋白酶,54.5%能分泌脂肪酶或明胶酶,72.7%能分泌卵磷脂酶,100%能分泌淀粉酶。同时筛选出5株胞外酶分泌能力超强的菌株,除分泌蛋白酶能力最强的弧菌(Bh14)来自鲍鱼肠道外,其余分泌其它4种胞外酶能力最强者(Sh05、Sh03、Sh11、Sh08)均来自养殖水体。研究还发现来自水体的弧菌分泌多种胞外酶能力强于来自肠道的弧菌。由此提示,应用有益微生物以改善鲍鱼肠道中的菌群结构,可能会提高鲍鱼饲料利用率。  相似文献

为了解近江牡蛎肠道厌氧菌在食饵消化过程中的作用,为微生态饲用添加剂的开发研究打下基础,从近江牡蛎肠道中分离出8株厌氧菌,其中有5株为革兰氏阳性菌。并研究了它们蛋白酶、脂肪酶、淀粉酶、纤维素酶的产酶能力,结果表明仅有3株菌具有产酶能力:2株菌能分泌脂肪酶,1株菌能分泌蛋白酶。由此可见.厌氧菌在分泌消化酶、消化食饵中所起的作用不大。  相似文献

对主养草鱼(Ctenopharyngodon idellus)池塘中4种鱼类消化器官的胃蛋白酶、胰蛋白酶、脂肪酶和淀粉酶的活性与鱼体长、体重及主要水质指标的相关性进行了研究。结果显示,草鱼肠道和肝胰脏的胃蛋白酶与水温具有显著正相关(P<0.05),而草鱼肠道胰蛋白酶、肠道和肝胰脏的脂肪酶以及鲢(Hypophthalmichthys molitrix)肝胰脏中淀粉酶与水温呈负相关(P>0.05)。4种鱼的消化酶活性与总氮和总磷呈正相关。匙吻鲟(Polyodon spathula)的脂肪酶和淀粉酶活性与化学耗氧量呈负相关;除鲢外,其他3种鱼的胃蛋白酶活性均与水体硬度和碱度呈正相关,而胰蛋白酶、脂肪酶和淀粉酶均与水体硬度和碱度呈负相关。草鱼和鳙(Aristichthys nobilis)肠道胰蛋白酶、脂肪酶和淀粉酶以及匙吻鲟消化道和肝脏胰蛋白酶与鱼的体长、体重均具有显著的正相关。  相似文献

运用酶动力学方法研究了鲢、鳙的蛋白酶和淀粉酶的活性分布。鲢、鳙的蛋白酶活性在肝胰脏中最强,脾脏中也较强,而在胆汁中未测出。该酶活性在肠粘膜的第Ⅱ段最大,第Ⅰ段向第Ⅱ段递增,第Ⅱ段向肠后端递减,但第Ⅴ段的该酶活性比前段要高些。鲢的淀粉酶活性分布规律与上述情况相似,但鳙的淀粉酶活性在肠粘膜各段中由前向后呈递减趋势,并以第Ⅰ段为最大。在相同的器官组织中,鳙蛋白酶活性高于鲢,但差异不显著(P>0.05),而淀粉酶活性则相反。这两种鱼的蛋白酶活性适宜pH为8.0~8.5,淀粉酶活性在pH7.5时最适。在鲢的肠内容物中发现有纤维素酶活性,但在肠粘膜中未检出该酶。  相似文献

草鱼和银鲫肠道产消化酶细菌的研究   总被引:4,自引:1,他引:3  
检测了分别从草鱼(Ctenopharyngodon idellus)和银鲫(Carassius auratus gibelio)肠道中分离的180株细菌的蛋白酶、脂肪酶、淀粉酶和纤维素酶的产酶能力。结果显示,两种鱼肠道内可分泌胞外消化酶的细菌包括Aero-monas(气单胞菌属,Aer.)、Vibrio(弧菌属,Vib.)、Bacillus(芽孢杆菌属,Bac.)、Pseudomonas(假单胞菌属,Pse.)四个种属的细菌,Aer.在其中占主要优势,45.71%的Aer.可分泌胞外消化酶。草鱼可分泌上述四种胞外消化酶的菌株共有33株,占肠道菌总数的36.67%;银鲫43株,占47.78%。产酶菌的分布上,草鱼中肠内产消化酶细菌数量显著多于前肠和后肠(P<0.05),前、中、后肠分别是6株、20株和7株;银鲫中肠和后肠数量差异不显著,前肠分布最少。草鱼分泌蛋白酶、脂肪酶、淀粉酶和纤维素酶的菌株分别有21株(23.33%)、10株(11.11%)、30株(33.33%)和16株(17.78%)。银鲫肠道内未检测到可分泌纤维素酶的细菌,蛋白酶、脂肪酶和淀粉酶菌株的数量分别是21株、37株和17株。可见鱼类肠道细菌对食饵消化有重要作用。  相似文献

从100 g左右的健康草鱼(Ctenopharyngodon idellus)肠道中分离出3株疑似芽孢杆菌(Bacillus)菌株(G1、G2、G3),并通过对其进行生理生化特征、16S rDNA全序列和产酶能力的分析,筛选出1株高产酶能力的益生菌株。结果显示:G1、G2和G3菌株与枯草芽孢杆菌在16S rDNA序列相似性≥99%的水平上聚为同一分支,结合形态观察和生理生化特征,最终鉴定G1、G2和G3菌株均为枯草芽孢杆菌(Bacillus subtilis);以水解圈直径/菌落直径比值评定了G1、G2和G3菌株的产纤维素酶、淀粉酶和蛋白酶的能力,其中G1菌株产纤维素酶、淀粉酶和蛋白酶能力(比值分别为2.9、2.2和3.3)均高于G2、G3菌株,具有作为益生菌的潜力。  相似文献

Isolation and enumeration of amylase, cellulase and protease‐producing autochthonous bacteria in the proximal intestine (PI) and distal intestine (DI) of three species of Indian major carps, catla (Catla catla), mrigal (Cirrhinus mrigala) and rohu (Labeo rohita), were investigated using the conventional culture‐based technique. Population levels of amylolytic strains were the highest in the PI of catla and the lowest in the DI of rohu. The highest viable count of cellulase and protease‐producing bacteria was recorded in the DI and PI of mrigal respectively. Among the bacteria isolated, 10 strains (five from PI and five from DI) were selected as potent enzyme producers according to a quantitative enzyme assay. The chosen strains were further identified by 16S rRNA gene sequence analysis. The five strains isolated from catla showed high similarity to Citrobacter sp. clone W2, Enterobacter sp. JA24, Bacillus coagulans strain TR, uncultured bacterial clone Hel3bc04 and Bacillus cereus strain UST2006‐BC004. The four strains isolated from mrigal were most closely related to Bacillus sp. KCd2, uncultured bacterial clone Hel3bd09, B. cereus strain BU040901‐020 and Citrobacter freundii strain YRL11, while the strain isolated from rohu probably belonged to Bacillus sp. GV.  相似文献

鳜鱼、青鱼、草鱼、鲤、鲫、鲢肝脏蛋白酶活性低于肠蛋白酶。肠蛋白酶活性,鳜鱼最高,其余依次为青鱼、鲤、鲢、草鱼、鲫。鳜鱼、鲢的肠蛋白酶活性由前肠向后肠递减;而青鱼、鲤、草鱼的则由前肠向后肠递增;鲫则中肠活性最低。六种鱼不同组织的脂肪酶活性因鱼而异。青鱼、鳜鱼、鲤、鲢肝脏中脂肪酶活性低于肠脂肪酶活性(P<0.01);而草鱼和鲫肝脏中脂肪酶活性与肠脂肪酶活性差异不明显(P>0.05)。六种鱼肝脏中脂肪酶活性由高到低依次为鳜鱼>鲢>鲫>草鱼>青鱼>鲤;肠脂肪酶活性顺序为鳜鱼>鲢>青鱼>鲤>草鱼>鲫。脂肪酶活性与鱼类食性无明显相关性。不同组织间淀粉酶活性存在差异。鳜鱼的肝脏和肠均有较高淀粉酶活性,青鱼和鲫肝脏中淀粉酶活性低于肠中的;草鱼、鲤和鲢肝脏中淀粉酶活性高于肠中的,但差异不显著。六种鱼中鳜鱼的肝脏淀粉酶活性明显高于其它五种无胃鱼,它们的淀粉酶活性顺序依次为鳜鱼>鲢>鲤>草鱼>青鱼>鲫;肠淀粉酶活性顺序为鳜鱼>鲫>鲤>青鱼>鲢>草鱼。  相似文献

Integration of tilapia to shrimp culture is currently being practiced to minimize the growth of pathogenic luminous bacteria. The microorganisms that are associated in tilapia may contribute to the inhibition of the growth of Vibrio harveyi through the production of secondary metabolites. In this study, two Bacillus strains (MJA1.1, MJA2.1) isolated from mucus of tilapia were evaluated for their possible application in shrimp culture. The inhibitory property of these isolates against V. harveyi was determined in vitro using co-culture assay in a liquid medium. Also qualitative extracellular enzyme assay was conducted to assess whether the bacterial isolates produce extracellular enzymes. Furthermore, the potential use of these isolates as shrimp feed additive was tested. Thereafter, shrimps were exposed to lethal dose of ammonia (140 mg l?1) to test the effects of the isolates in vivo. The results showed that in vitro co-culture assay after 72 h caused a significant decline in the population of V. harveyi in treatments with potential probiotic isolates. Both isolates showed protease, amylase, and cellulase activities. Although no significant difference was observed in growth, survival was significantly higher in shrimp fed with diets added with either of the isolates. The shrimp exposed to lethal dose of ammonia demonstrated better survival when supplemented with the probionts compared to the control group. Thus, the efficiency of the isolates in inhibiting V. harveyi population and the improvement of survival and resistance of cultured shrimp to ammonia stress indicate their potential as probionts for shrimp culture.  相似文献

本文就鲤鱼肠道细菌对其宿主的影响和细菌淀粉酶对宿主消化的影响作了探讨。从鲤鱼肠道筛选细菌菌株63株,这上结细菌中淀粉酶产生菌分布较广,溶血毒素产生菌较少,且产毒素的6株菌无致病性。淀粉酶活力较高的CCI139菌株的淀粉酶和鲤鱼肝胰淀粉酶的最适pH和反应产物都不相同。原位酶反应法显示鲤鱼肠道食物混合液及肠壁样品中均有细菌淀粉酶。本文结果证明了鲤鱼肠道不稳定的细菌群落,细菌淀粉酶对鲤鱼消化食物的淀粉起  相似文献

进行了黄鳝喜食的天然动物性饵料蚯蚓、鲢肌肉和配合饲料对黄鳝消化酶活性影响的比较研究。结果表明:(1)黄鳝各消化组织中蛋白酶活性均以摄食蚯蚓组增加幅度最大,其次为摄食鲢肌肉组,而以摄食配合饲料的蛋白酶活性增加最小;(2)胰蛋白酶的活性变化则以摄食鲢肌肉组的增加幅度最大,以摄食蚯蚓组次之,摄取配合饲料组的增加幅度最小;(3)淀粉酶的活性以摄取配合饲料的黄鳝的增加幅度最大,其次为摄食鲢肌肉组,增加幅度最小的为摄取蚯蚓组个体;(4)脂肪酶的活性变化以摄取鲢肌肉的增加幅度最大,其次为摄食蚯蚓的,摄食配合饲料组增加幅度最小。这表明不同饲料对黄鳝消化器官消化酶的活性影响不同,这可能与不同饲料的营养组成、性质有关,食物中的不同营养物质会刺激黄鳝消化器官分泌相应的消化酶,以便更好地消化所摄取的食物。  相似文献

两株芽孢杆菌的生长特性及其对仿刺参的益生作用   总被引:1,自引:0,他引:1  
本实验室前期从大连湾地区健康的仿刺参养殖池塘沉积物中分离出两株芽孢杆菌即B6和B7。首先,通过分析二者的生长特性发现,B6和B7的最适生长温度为15~35℃;最适pH范围为6~8;最适盐度为3%~5%。菌株产酶特性检测结果表明,B6和B7均可产生淀粉酶和蛋白酶,B7产脂肪酶。将两种菌悬液投喂仿刺参后发现,B6和B7可有效提高仿刺参肠道内的消化酶活性以及体腔液中多种非特异性免疫酶活性。本研究结果表明:B6和B7可作为益生菌剂的基础菌株添加至仿刺参生长环境中,并且对仿刺参的消化及免疫功能具有正向刺激作用。  相似文献

测定了中华鲟四种消化器官消化酶的活性,其淀粉酶及蛋白酶的活性高高到低的顺序均为:幽门盲囊>肠道>胃>肝脏,其脂肪酶活性由高到低的顺序为:肠道>胃>肝脏>幽门盲囊。中华鲟幽门盲囊中淀粉酶和蛋白酶的活性较高,而脂肪酶几乎没有活性,肝脏中淀粉酶几乎没有活性,蛋白酶和脂肪酶的活性也比较小;肠道中各种酶的活性都较高,而胃中各种酶的活性则居中。  相似文献
