Psathyrostachys huashanica Keng ex Kuo (2n = 2x = 14, NsNs), a source of wheat stripe rust, take-all fungus, and powdery mildew resistance with tolerance to salinity and drought, has been successfully hybridized as the pollen parent to bread wheat without using immature embryo rescuing culture for the first time. All of the CSph2b × P. huashanica hybrid seeds germinate well. Backcross derivatives were successfully obtained. F1 hybrids were verified as intergeneric hybrids on the basis of morphological observation, cytological and molecular analyses. The results obviously showed the phenotypes of the hybrid plants were intermediate between bread wheat and P. huashanica. Chromosome pairing at MI of PMCs in the F1 hybrid plants was low, and the meiotic configuration was 26.80 I + 0.60 II (rod). Cytological analysis of the hybrid plants revealed the ineffectiveness of the ph2b gene on chromosome association between the parents. Eight RAPD-specific markers for Ns genome were selected for RAPD analysis, and the results indicated that F1 hybrids contained the Ns genome of P. huashanica. Furthermore, the significance of the finding for bread wheat improvement was discussed.  相似文献

Dasypyrum villosum (L.) P. Candargy is discussed as a species commonly used in wheat improvement. Chromosomal localization of the potentially useful traits and chromosomal position of some morphological and isozyme markes are shown. The investigations using molecular RAPD, AFLP, SSR, RFLP markers and in situ (GISH, FISH) hybridization experiments on D. villosum itself and in wide hybrids with Triticum are summarized. The article also presents the information about designation of D. villosum chromosomes and the current knowledge on the phenomenon of two-coloured D. villosum caryopses.  相似文献

Summary Crossing experiments between Dasypyrum villosum (2 x) and 14 taxa of Triticum sensu lato (seven diploids, five tetraploids and two hexoploids) were performed. Adult hybrids were obtained in all but three of the combinations with diploid species. A haploid plant was obtained from the combination T. aestivum x D. villosum. Cytogenetic data on the meiotic pairing in the intergeneric hybrids revealed that, in general, very little pairing occurred between the V genome of Dasypyrum and the different genomes in Triticum. There may be comparatively large differences in pairing behaviour in hybrids including different parental accessions, which shows that the Triticum as well as the Dasypyrum genotypes may influence the pairing. The combination T. aestivum x D. hordeaceum (4 x) was also produced and from the meiotic pairing in the hybrid it is evident that D. hordeaceum is an autoploid.  相似文献

Chromosome pairing was analysed in F1 hybrids of the wheat cultivar ‘Chinese Spring’ (CS) and its ph1b mutant (CSphlb) with Dasypyrum villosum. On average, 1.61 chromosomes per cell paired in the hybrid CS ×D. villosum, but 14.43 in the hybrid CS ph1b×D. villosum. Genomic fluorescence in situ hybridization (GISH) revealed three types of homoeologous association between wheat (W) and D. villosum (D) chromosomes (W‐D, D‐W‐W and D‐W‐D) in pollen mother cells of the CS ph1b×D. villosum hybrid, and only one type (W‐W), in the CS ×D. villosum hybrid. Both F1 hybrids were self‐sterile. The seed set of the backcross of CS ×D. villosum with CS was 6.67% and that of CS ph1b×D. villosum with CS or CS ph1b was only 0.45%. The chromosome number of BC1 plants varied from 48 to 72. Translocations of chromosome segments or entire arms between wheat and D. villosum chromosomes were detected by GISH in the BC1 plants from the backcross of CS ph1b×D. villosum to CS ph1b.  相似文献

Summary One hexaploid wheat cultivar (Triticum aestivem) and two tetraploid wheat lines (T. durum) were crossed with seventeen inbred lines of rye (Secale cereale). Seed set, degree of hybrid embryo differentiation at the time of excision for in vitro culture and recovery of amphihaploid plantlets from various embryo categories were studied. Degree of embryo differentiation was predominantly determined by maternal wheats, paternal rye genotypes appearing to be of minor importance. T. aestivum x rye hybrid embryos were superior to those produced from T. durum for degree of differentiation. The proportion of plantlets developing from differentiated embryos was high for all wheat parents, whereas undifferentiated embryos were mostly unsuitable for plantlet production. The results revealed that cross-incompatibility in hexaploid wheat x rye crosses was due to failure of fertilization, while in tetraploid wheat x rye crosses it was caused by lack of embryo differentiation. Correlation analyses showed that seed set provided a criterion to predict the amphihaploid plantlets to be expected from a particular wheat x rye combination.  相似文献

Summary Fifty four hybrid plants between Elymus canadensis and Psathyrostachys juncea were obtained by handpollination and embryo culture. The average cross compatibility between both species was 31.2 percent. One amphiploid plant was induced by colchicine treatment. The hybrid and amphiploid plants resembled P. juncea in appearance but showed a higher plant height and dry matter yield than the parents. The hybrids showed extremely low pollen stainability and were completely sterile. With the exception of one plant (2n=3x+1=22), all hybrid plants were allotriploids (SHN, 2n=3x=21). The amphiploid plant (SSHHNN, 2n=6x=42) showed 58.9% pollen stainability and 11.6% seed fertility.Mean chromosome associations of the hybrids and amphiploid at metaphase I were 0.02IV+0.06III+2.03II+16.91I and 0.07III+18.00II+5.85I, respectively. Lagging chromosomes, chromosome bridges, abnormal cytokinesis, and micronuclei were occasionally observed at the anaphase, telophase, or tetrad stage.  相似文献

Dasypyrum villosum (L.) Candargy (DV) is adiploid (2n = 14, VV genomes), allogamous grass of theMediterranean region. It may be hybridized with wheatand is thus a gene resource for wheat improvement. Westudied grain protein concentration andSDS-sedimentation (SED) as indicators of end-usequality. The latter is a good predictor of glutenstrength. A-PAGE and SDS-PAGE were used to identifymonomeric and polymeric seed storage proteins,respectively, to relate proteins of DV to those foundin Chinese Spring (CS), Triticum aestivum L.,wheat. Two full-sib lines of DV had high grain protein(19.3 and 20.3%), but one had very low mean SED (69mm) and one had very high (118 mm) based on onegreenhouse and one field test. CS had very low grainprotein (12.0%) and weak gluten (33 mm). Single-DVchromosome addition and substitution lines and twoDV-wheat recombinant lines all had higher grainprotein than CS (range 13.9 to 16.7%). SED valuesshowed a different pattern. CS=4V and CS=6V hadlow SED, 63 and 44 mm, similar to CS, whereas CS=1Vand full sib DV 200 had very strong gluten, 118 mm, asdid substitution lines CS1V (1A) and CS1V (1B), 125and 131 mm, respectively. One hybrid-derived line withDV-wheat 1V recombinant chromosome had SED of 99 mmand one line with a 6V added chromosome had SED of 64mm. The large positive effects of quality in the wheathaving DV chromosome 1V are believed to be due to DValleles at the Glu-V1 and Gli-V1/Glu-V3loci. DV chromosomes 4V and 6V did not contribute toimproved quality probably due to Gli-V2 and Gli-V3 which, as the orthologous loci in wheat, donot enhance wheat quality. Based on the positiveeffects of alleles on DV chromosome 1V in a breadwheat background, we conclude that D. villosumis a source of allelic diversity that can beconsidered for improving end-use quality in breadwheat.  相似文献

Summary The meiotic pairing behaviour at metaphase I of a Triticum aestivum×Triticum monococcum hybrid has been studied by means of the C-banding technique to ascertain the homology between the chromosomes in the A genome of the two species. The technique allowed the A and B genome chromosomes and the 2D, 3D and 5D chromosomes to be identified. Differences in the level of chromosome pairing in the A genome were noted. The T. monococcum 4A chromosome did not pair with any of the T. aestivum chromosomes in any of the metaphase I cells analysed. Two reciprocal translocations between the 2B and 2D chromosomes on one side and the 2A and 3D on the other side have been identified. The usefulness of the C-banding technique in the study of chromosome homology among species related to wheat is discussed.  相似文献

Summary Triticum aestivum L. em Thell. (2n=42; AABBDD), and T. durum Desf. (2n=28; AABB) genomes were substituted into the cytoplasms of Aegilops mutica Boiss. (2n=14; MtMt), Ae. heldreichii Holzm. (2n=14; MM), Ae. uniaristata Vis. (2n=14; MuMu), and Ae. ovata L. (2n=28; CuCuMoMo), to identify the M-genome diploid cytoplasm donor of Ae. ovata. Substitution of the T. durum genome into Ae. uniaristata cytoplasm resulted in a large proportion of shriveled inviable seeds. A few plump viable seeds were obtained all of which produced male-sterile plants having one univalent or telocentric chromosome from Ae. uniaristata. The T. aestivum plants having Ae. uniaristata or Ae. mutica cytoplasms were fertile. However, Ae. mutica was similar to Ae. ovata in the induction of delayed maturity and tall robust growth habit to the T. durum and T. aestivum plants. Cytoplasms of the other C- and M-genome diploids Ae. umbellulata Zhuk. (2n=14; CuCu) and, Ae. heldreichii (2n=14; MM) earlier had been shown to differ from that of Ae. ovata. Therefore, Ae. mutica is the most likely cytoplasm and M-genome donor to Ae. ovata.  相似文献

Summary The meiotic behaviour of a hybrid between Triticum aestivum and the amphiploid Hordeum chilense x T. turgidum conv. durum, was studied using a C-banding staining method. This hybrid has the genome formula of AA BB D Hch with 2n=6x=42 chromosomes. The durum wheat chromosomes (genomes A and B) were easily recognized, whereas the D and Hch chromosomes were recognized as a whole. Meiotic pairing was homologous, as expected (14 bivalents from A and B genomes +14 univalents from D and Hch genomes). However, some pollen mother cells at metaphase-I presented pseudobivalents that could have been caused by either homoeologous or autosyndetic pairing amongst D and Hch chromosomes.  相似文献

Summary A self-fertile trigeneric hybrid in the Triticeae involving species from the Hordeum, Triticum and Secale genera has been produced. The trigeneric hybrid was obtained by crossing octoploid triticale (x Triticosecale Wittmack) with octoploid tritordeum (H. chilense × T. aestivum amphiploid). The trigeneric hybrid presented a genome constitution AABBDDRHch and 2n=8X=56 chromosomes. The cytogenetical analyses showed no chromosome instability nor homeologous pairing between Hordeum and Secale chromosomes. In the F2 generation the chromosome number ranged from 42 to 52. Within this range, the plants with smaller numbers of chromosomes were more frequent. A preferential transmission of rye chromosomes could be inferred.  相似文献

Summary Wheat has traditionally been grown by the Beduin population in the semi-arid (150 to 200 mm, mean total annual rainfall) northern Negev region of Israel.A collection was made in this area (the size of which is 150 km2) from small (0.1 to 0.5ha) fields of mixed wheat, resulting in 1553 collected spikes. Each spike was planted in a 1 m row at Bet Dagan, and grown under favorable conditions. Qualitative and quantitative data were collected from each row. Qualitative data were submitted to hierarchial clustering and the results were compared with published information on the identification, classification and distribution of the land-races of wheat in the Middle East. Triticum durum was represented in 84% of the collection. It was clustered into 22 populations, identified as 11 known varietas of T. durum. They were aggregated into five groups, similar to groups of old varieties recognized by Jacubziner (1932). While 38.5% of the collection consisted of T. durum groups villosa and sinaica, aboriginal to the northern Negev, it included also forms similar to several land-races found in the past in other parts of the Middle East. Each of the populations, and the durum collection as a whole, was very diverse for the quantitatively measured plant attributes. Triticum aestivum was represented in 15.6% of the collection, clustered into six populations. Most of the common wheat accessions were analogous to the old locally grown variety Hirbawi. This paper reports the waxy proteins and apparent amylose contents of the tetraploid species Triticum dicoccum, Triticum polonicum and Triticum durum. Three waxy proteins were found in the three species; two showed the same electrophoretic mobility as the alleles Wx-A1a and Wx-B1a of the hexaploid variety ‘Chinese Spring’, while the third showed the same mobility as the allele Wx-B1e belonging to the hexaploid wheat variety ‘Bai Huo’. In apparent amylose content no significant differences between the alleles Wx-B1a and the Wx-B1e were found for each species. However, the mean amylose contents of T. durum and T. polonicum were significantly greater than that of T. dicoccum, regardless of which allele was present. Among the progenies of a hybrid between common wheat Triticum aestivum L. cv. Yannong 15 and Thinopyron intermedium, plant E99018 was identified with the chromosome number 2n = 42 and stable agronomic traits. An analysis of the metaphase chromosome pairing indicated that it formed 21 bivalents but that 2 univalents were present in the F1 hybrid of this plant with common wheat. Resistance verification by race 15 and with mixed races of Blumeria graminis f. sp. tritici at the seedling and adult stages showed that at both stages, the plant was immune to powdery mildew. In situ hybridization with the genomic Th. intermedium and the St genome DNAs as probes and wheat DNA as a block has shown that it contained a pair of Th. intermedium chromosomes. On the basis of the hybridization pattern of the St genome probe to the critical chromosome, a conclusion was reached that this pair of chromosomes belonged to the E genome. Therefore, plant E99018 was a spontaneously formed substitution line. An analysis by 116 SSR markers indicated that the substituted wheat chromosome was 2D and the most likely substitution in E99018 is 2E(2D).  相似文献

Summary Durum or macaroni wheat (Triticum turgidum L., 2n = 4x = 28; AABB genomes) arose as a natural hybrid between two wild species, Aegilops speltoides Tausch (2n = 2x = 14; BB genome) and Triticum urartu Tumanian (2n = 2x = 14; AA genome). The two progenitors hybridized in nature about half a million years ago and gave rise to tetraploid wheat, presumably in one step as a result of functioning of unreduced gametes in their hybrid BA (amphihaploid). It is easily possible to go back on the evolutionary scale and obtain durum haploids BA, and then regenerate tetraploid durum plants from them. Interestingly, such a reversion to haploidy does occur in nature as well, although at a very low frequency. This article reports on the occurrence of two spontaneous durum haploids and describes their chromosomal characteristics. The haploids (euhaploids, to be precise) had 14 somatic chromosomes, which, on fluorescent genomic in situ hybridization (fl-GISH), could be distinguished as 7 A-genome and 7 B-genome chromosomes. At meiosis, only 2.3 and 2.7% of the chromosomes paired in the two haploids, because of the presence of the homoeologous pairing-suppressor gene, Ph1. Elongated glumes are present in thetetraploid wheat species T.polonicum, T. turanicum, T.durum convar. falcatum and in thehexaploid species T. petropavlovskyi.Inheritance of glume length was studiedwith the aim to map the respective lociusing wheat microsatellite markers. In T. polonicum and T. petropavlovskyiloci conferring long glume were mapped nearthe centromere on chromosome 7A. These twoloci are designated P-A pol 1 andP-A pet 1, respectively. It isshown that both are probably homoeoallelicto each other and to the P gene ofT. ispahanicum on chromosome 7B. The loci determining elongated glumes in T. turanicum and T. durum conv. falcatum are not homoeologous to the P loci in the centromeric region of thegroup 7 chromosomes.  相似文献

Isolation distance is the main barrier to crop-to-crop gene-flow. A 3-year study assessed the maximum potential outcrossing under field conditions between two wheat cultivars (Triticum aestivum L.) and between wheat and durum wheat (Triticum turgidum L. var. durum). Outcrossing was measured by seed set on emasculated recipient plants placed at four sides with different distances from a 3 m × 3 m T. aestivum (cultivar Chinese Spring) pollen source. Frequencies of seed set at 0 m distance were 45% (37–56%) for T. aestivum cultivars and 18% (5–30%) with T. turgidum. These values agree with hybridization in non-limiting pollen conditions measured by manual crosses in greenhouse. The number of pollen grains and the outcrossing frequencies decreased at increasing distances influenced by the prevailing wind direction. Under semiarid conditions of this assay, viable pollen was found 14 m from the pollen source, with a maximum distance of 8 m at which cross-pollination decreases below 1%. Ambient conditions affect pollen viability, hybridization and pollen dispersal. Data presented in this paper emphasize the major role played by environmental conditions in outcrossing. Data obtained in one area may therefore not coincide with the prevailing situation in different locations and climates.  相似文献

Summary The intergeneric hybrid Hordeum vulgare x Psathyrostachys fragilis was fairly easily obtained. During each growing season the intermediate, perennial hybrid yielded haploid tillers of H. vulgare. Late in one season few, hybrid tillers headed. Summary Resistance to septoria nodorum blotch in Triticum monococcum, T. tauschii, T. timopheevii, T. dicoccum and T. durum was evaluated on plants at the three-leaf stage in greenhouse tests. A high frequency of resistant genotypes was found in T. monococcum, T. tauschii and T. timopheevii, but not in T. dicoccum and T. durum. The resistance of F1 plants of crosses of resistant T. monococcum (PI 289599) and T. timopheevii (PI 290518) accessions with susceptible common wheat cv. Park and durum wheat cv. Wakooma, respectively, was evaluated on the basis of percentage leaf necrosis, lesion number, lesion size and incubation period. No dominance was found for long incubation period, but various dominance relationships occurred for low percentage leaf necrosis, low lesion number and small lesion size, depending on the cross. Multiple regression analysis showed that lesion number contributed more to percentage leaf necrosis than lesion size or incubation period. Resistance to septoria nodorum blotch was transferred successfully from T. timopheevii to cultivated durum wheat. Resistant BC1F7 lines, recovered from the T. timopheevii (PI 290518) × Wakooma cross, showed normal chromosome behaviour at meiosis (14 bivalents) and were self-fertile. However, an effective level of resistance was not recovered in lines derived from the other interspecific crosses.  相似文献
