对进口的加拿大牛冻胚胎进行解冻后移植效果观察。结果共解冻24枚冻胚,解冻后直接移植给24头受体牛,妊娠19头,移植妊娠率79.2%,其中荷斯坦奶牛冻胚移植21头,妊娠17头,移植妊娠率达81.0%,创造我国牛冷冻胚胎移植成功率的最高记录。  相似文献

甘肃省进口肉牛冷冻胚胎移植的研究   总被引:1,自引:1,他引:0  
1999年5月至2000年12月在甘肃省武威市、平凉市、信县、灵台县开展了进口肉牛冷冻胚胎移植工作。经过对386头当地黄牛母牛及杂交母牛进行了诱导同期发情,结果获得84.9%的同期发情率和77.1%的黄体合格率。对于黄体合格的253头母牛进行了进口肉牛冷冻胚胎移植,获移植妊娠率50.9%,其中45头受体牛由甘肃省家畜繁育中心技术人员移植,获得51.1%的移植妊娠率。对同一批次经产牛与青年牛移植妊娠结果、常规法与一步法解冻胚胎移植妊娠结果进行了对比分析,结果显示,经产牛移植妊娠率(37.5%)显著低于青年牛(56.6%)常规胚胎解冻法与一步细管解冻法解冻胚胎,移植妊娠率差异显著(41.6%,69.5%,P<0.05),后者较高。  相似文献

胚胎移植技术是MOET奶牛育种的重要手段.但由于胚胎移植受胎率偏低,尚不能在生产中广泛应用,如何尽快将这一技术在生产中应用和推广,是摆在广大畜牧科技人员面前亟待解决的问题。从2009年1月至2010年6月,经过技术人员的共同努力.初步确立了MOET核心群方案,同时胚胎移植取得了较好的效果:供体牛同期发情率达95.6%,受体牛同期发情率达89%,供体牛冲胚平均每头(6.2±0.8)枚,移植受体牛128头(其中移植鉴别母胚84枚、混合胚44枚),妊娠78头,妊娠率达60.9%(母胚妊娠率63.1%),鲜胚性别鉴定率为100%.均达到了国内先进水平。  相似文献

2000年4月至2002年12月,在塔城市、额敏县、塔城种牛场开展了进口美国褐牛冷冻胚胎移植工作。经过对126头新疆褐牛及杂交母牛进行了诱导同期发情,结果获得78.6%的同期发情率和72.7%的黄体合格率。对于黄体合格的72头母牛进行了进口美国褐牛冷冻胚胎移植,获移植妊娠率51.4%。其中24头受体牛由塔城地区育种站和塔城种牛场的技术人员移植,获得45.8%的移植妊娠率。对同一批次牛的常规法与一步法解冻胚胎移植妊娠结果(分别为43.5%和64.7%)进行对比分析,结果显示,移植妊娠差异显著(P<0.05)。  相似文献

2002-2004年,从饲养的812头奶牛群中,选择69头高产奶牛作供体进行超数排卵处理,63头供体牛共获胚胎495枚,超排成功率达91.3%(63/69),平均获胚胎7.17枚/头,可用胚胎402枚,平均可用胚胎5.82枚。其中采用加拿大生产的促卵泡素超排处理奶牛供体10头,获卵106枚,可用胚胎87枚,平均获可用胚胎8.7枚/头;采用鲜胚移植奶牛受体8l头,妊娠44头,妊娠率为54.3%,产犊28头,待产16头;采用冷冻胚胎移植黄牛受体278头,妊娠114头,妊娠率为41.0%。产犊22头,待产92头。  相似文献

用简易分割方法,分割奶牛冷冻胚胎16枚,获得半胚32枚,分割成功率为100%(16/16),可移植半胚29枚,移植于16头受体牛,到第3个情期未返情者经直肠检查有6头妊娠,妊娠率为37.5%(6/16)。其中,在分割前或者分割后经恢复培养0.5~2h再移植的10头受体牛,5头妊娠;而未经恢复培养,分割后直接移植的6头受体牛中,只有1头妊娠。移植后3个月,直肠检查确定2头流产。已有1头受体黄牛生出1头奶牛牛犊(母)和1头受体奶牛产1头奶牛牛犊。其余的2头妊娠受体牛将于9月份产犊。此外,用简易分割法分割奶牛胚胎5枚,得到半胚10枚,裸半胚直接冷冻,解冻后回收可移植半胚5枚,移植于4头受体牛,无一头妊娠。结果表明,冷冻胚胎的分割半胚优于分割后冷冻半胚移植效果;冷冻胚胎分割前或者分割后恢复培养移植优于未经恢复培养而直接移植;简易分割法可应用于冷冻胚胎的分割。  相似文献

1991-1996年,对6个品种138头供体母牛超排处理199次,共获得A、B级胚胎1148枚,平均每次每头获可用胚5.77枚;共移植受体1299头(包括鲜胚和冻胚),妊娠产犊593头,总移植妊娠率为45.7%。对牛的超数排卵技术进行了深入研究,对不同超排方法的超排效果、不同品种肉牛的超排反应及重复超排的效果进行了分析,获得了适于重复应用、效果稳定的超排方案,邓用FSH-PG法,FSH总剂量在4-6mg,5d 10次递减注射。同时,对受体牛饲养管理条件与受体诱导同期发情效果和移植妊娠率之间的关系进行了探讨,对经产牛和育成牛受本移植妊娠效果进行了比较,并对移植人员技术熟练程度对妊娠率的影响进行了分析,从而为大规模实施牛ET计划提供了受体牛选择依据。  相似文献

Transfer of porcine embryos after 3 days of in vitro culture   总被引:1,自引:0,他引:1  
Two experiments were conducted to determine the viability of porcine embryos transferred after long-term in vitro culture. In Exp. 1, four-cell embryos were kept in culture for 120 h. Embryos that were exposed to fresh culture medium every 12 h survived better than embryos kept in the same medium throughout the culture period. In Exp. 2, four- and eight-cell embryos were cultured in vitro for 72 h before transfer to estrus-induced recipient gilts. Each gilt received, on average, 19 embryos. If recipients were synchronous with donors 3/32 (9%) recipients remained pregnant with an average of 4.0 +/- .6 viable young. If the sexual cycle of the recipients was 24 h behind that of the donors the pregnancy rate was 18/34 (53%) with 4.4 +/- .5 viable young. Average embryo survival rate for the two groups was 1.8 and 12.5%, respectively. A 24-hourly medium replacement during the in vitro culture period had no significant effect on transfer results. When transferring freshly collected blastocysts, pregnancy rate, number of viable young and survival rate of embryos were 6/10 (60%), 7.8 +/- 1.4, and 23.9% for synchronous recipients and 7/10 (70%), 9.3 +/- 1.8, and 32.9% for asynchronous recipients, respectively. Recipients with very high plasma progesterone levels or numerous follicular cysts at the time of transfer were less likely to remain pregnant than others.  相似文献

试验就胚胎的发育阶段、胚胎保存时间及移植胚胎数等因素对胚胎移植效果的影响情况进行了研究。结果表明,胚胎发育日龄对受体妊娠率没有显著影响(P>0.05),但移植8~16细胞的胚胎的妊娠率略高于2~4细胞(58.49%vs 61.47%)。人员不是影响受体妊娠率的主要因素。胚胎在体外保存0~120min对妊娠率没有显著影响,但以在体外保存0~30 min内移植的妊娠率最高(65.61%)。黄体与移植胚胎的相对位置对受体的妊娠率没有显著影响(P>0.05),但移植双胚的受体的妊娠率较移植单胚高(61.76%vs 45.17%)。同时,试验统计了经胚胎移植产生的羔羊和自然繁殖羔羊之间的差异,发现经胚胎移植产生的波尔山羊母羔的初生重极显著地高于自然繁殖的母羔(P<0.01),但公羔间的初生重以及公、母羔间的体高、体长、胸围、死亡率和性别比例与自然繁殖羔羊差异不显著(P>0.05)。  相似文献

本研究对胚胎质量、胚胎移植数量、胚胎发育阶段、冷冻方法、移植方法及饲养管理水平等因素对移植产羔率的影响进行研究,以提高绵羊胚胎移植效率。结果表明:采用1.5mol/L乙二醇做冷冻保护剂对胚胎进行常规冷冻,冻前A级胚胎冷冻/解冻后可用胚率、存活率、降级率均高于B级,差异极显著(P<0.01)。移植1枚冷冻解冻后A级、B级或2枚(A+B)胚胎的受体移植产羔率分别为44.48%、46.84%和44.81%。经χ2检验分析,三者之间无显著差异(P>0.05)。移植双胚的受体双羔率为22(%53/241),以胚胎为单位计算,产羔率仅为33.40%。A级囊胚和桑椹胚的产羔率之间无显著差异(P>0.05)。1.5 mol/L乙二醇常规冷冻保存的体内胚胎移植产羔率高于EFS40玻璃化冷冻保存,但经统计学分析,二者无显著差异(P>0.05)。子宫手术移植和腹腔内窥镜法子宫移植之间产羔率不具统计学差异(P>0.05)。在牧场条件下大规模移植的平均产羔率为43.95%,范围在20% ̄70%之间,受体饲养管理水平对绵羊移植产羔率有较大影响。  相似文献

In this study, we attempted to produce piglets by non-surgically transferring blastocysts produced in vitro, using a flexible catheter as the transfer instrument. Cumulus-oocyte complexes (COCs) were aspirated from the follicles of ovaries obtained at a local slaughterhouse. They were then matured in modified North Carolina State University (NCSU)-37 medium for 44-46 h and fertilized in porcine gamete medium (PGM). Ten hours after in vitro fertilization (IVF), presumptive zygotes were removed from the cumulus cells and cultured in porcine zygote medium (PZM)-5. Blastocysts were cultured for five days after IVF and, using a catheter for deep intrauterine insemination without sedation, they were transcervically transferred into the uterine horn of six recipients (45-50 blastocysts/recipients) whose estrous cycles were synchronized, at 5 days after human chorionic gonadotropin (hCG) injection. Of the six recipients, one sow became pregnant and farrowed seven piglets (four live piglets) 119 days after hCG injection. The body weight at birth of the newborns ranged from 0.8 to 1.4 kg. These results indicate that it is possible to obtain piglets by transcervically transferring blastocysts produced by IVF and in vitro cultures in chemically defined media.  相似文献

为了有效提高绵羊体细胞克隆和转基因等研究工作的效率,缩短胚胎移植试验羊群中空怀个体的利用周期间隔,试验对采用不同移植方法的胚胎移植受体及供体绵羊进行了移植前后的发情规律研究。结果表明:采用自制孕激素海绵栓和进口阴道孕激素释放装置(CIDR)对绵羊进行同期发情处理时发情率均接近90%,且92%以上的个体发情起始时间均集中在撤栓后24~48 h。在胚胎移植试验结束后,未参加胚胎移植个体和子宫角法未移植受体第2情期发情起始时间主要集中在上次发情后的14~20 d,而输卵管法未移植受体和输卵管法冲卵供体的发情时间则比较分散,呈现明显延后状态。未参加胚胎移植个体和子宫角法未移植受体人工授精后的受胎率接近往年羊场的平均水平,输卵管法未移植受体的受胎率略有下降,且输卵管法冲卵供体的受胎率明显降低。  相似文献

The purpose of this study was to produce quail–duck chimeras by transferring stage X blastoderm cells and to detect the distribution of donor cells in heterogeneous embryos using PCR. Four experimental groups were made by transferring different amounts of quail blastoderm cells into duck recipients. In early embryonic stages, donor cells labeled with PKH26 fluorescent dye were observed in the head, neural tube and gonads by fluorescent microscopy. A total of 194 duck recipient embryos were injected and 93 survived to hatch. The average hatching rate was 48% (93/194); the hatching rate showed a significant difference among all the groups (P < 0.05). Sixteen somatic chimeras were obtained, 10 of which had black feathers derived from the donor quail. The PCR results showed that donor cells were distributed in various tissues and organs of the phenotypic chimeras. This is the first report on producing Japanese quail–Peking duck chimeras by transferring quail blastoderm cells into the subgerminal cavity of the duck. This technique will provide a basis for the investigation of fertilization barriers in interspecies germline chimeras and will aid conservation of endangered wild birds.  相似文献

简单分割7~8天奶牛胚胎59枚(117个半胚)。裸半胚成对移给59头奶牛或黄牛,80~90天有27头(45.8%)妊娠。最后24头受体产犊34头,半胚产犊率29.1%(34/117)。有10对双胎,双胎率41.7%(10/24)。比较在不同情况下—7天或8天;晚桑椹或囊胚期;透明带软化处理或不软化处理—分割的半胚,成对移植后受体妊娠率分别是40.0%和57.9%;35.7%和54.8%;48.6%和41.7%。半胚产犊率分别是26.6%和34.2%;23.6%和33.9%;30.0%和27.7%。均无显著差异(P>0.05)。分割优质胚胎得到最好的(35.7%)半胚产犊率。半胚在体外5小时内移植有较高(30.5%)的产犊率。试验探索了奶牛半胚移给远处分散的农户黄牛的可能性。11对奶牛半胚移给11头黄牛有6头(54.5%)妊娠。最后5头受体产下7头奶牛犊,两对同卵双胎。  相似文献
