Parameters derived from the non-invasive and simple monofrequency forced oscillation technique were compared with classical parameters of ventilatory mechanics in order to assess its usefulness for the investigation of pulmonary function in calves. To facilitate this comparison, theoretical derivations were coupled with in vitro measurements, using an artificial lung model, and with in vivo studies. These studies compared the oscillatory resistance parameters (Ros and Re) and the respiratory system compliance (Crs) against the classical pulmonary resistance (RL) and the dynamic compliance (Cdyn), respectively. Ros and Re were highly correlated (r0·87) with RL and the comparison between Crs and Cdyn gave a similarly high correlation (r0·88). Given its simplicity, its correspondence with classical parameters and its rapidity and reproducibility, monofrequency forced oscillation technique seems well suited for the investigation of pulmonary function under field conditions.  相似文献

Multifrequency impulse oscillometry ( ) was compared with a monofrequency forced oscillation technique ( ) in calves undergoing experimentally induced bronchoconstriction and subsequent bronchodilatation. The dynamic lung compliance (Cdyn) was also measured by conventional methods. For each test, the baseline mean and the responses to saline, a bronchoconstrictive agent (carbachol) and a bronchodilator (fenoterolhydrobromide) were calculated. Using the 1os, the information was markedly frequency-dependent. The resistance (R) and the magnitude of respiratory impedance (Z) were only sensitive at 5 Hz, leading to negative frequency dependence of these parameters as an indicator of peripheral airway obstruction. A high sensitivity for reactance (X) and phase angle (0) values was observed between 5 and 20 Hz. For MFO (10 Hz), the parameters Ros (which includes resistive and capacitive components of the respiratory system), phase shift (psi), and the oscillatory derived compliance of the respiratory system (Crs) were of the greatest clinical potential. Crs showed a significant coefficient of linear correlation (r=0.88, P<0·001) with Cdyn. At the 10 Hz test frequency similar results were observed with MFO and 105, suggesting that for healthy calves the measurement effect of an impulse is not significant. With respect to peripheral airway calibre, a test frequency less than 10 Hz appeared to be most sensitive and least variable.  相似文献

The effect of clenbuterol hydrochloride on the course of disease in calves with a natural bovine respiratory syncytial virus infection was examined. Six calves (three to nine months of age) originating from four herds with respiratory tract disease and serological evidence of a bovine respiratory syncytial virus infection were used in this study. The calves were injected intravenously with clenbuterol hydrochloride. The effect of clenbuterol on the course of disease was measured using the PO2 in blood taken from an indwelling canula inserted in the caudal auricular artery and by clinical signs. Clenbuterol did not improve clinical signs. After clenbuterol administration arterial PO2 values decreased significantly in five out of six patients. Six to eight hours after medication the mean arterial PO2 values were higher than initial values. The moderate positive effect of clenbuterol after six to eight hours may be caused by enhancing ciliary activity and by the secretolytic activity of clenbuterol.  相似文献

Calves perorally administered the beta-adrenergic agonist (beta-A) clenbuterol for 28 d were studied before, during, and after a 12-min treadmill exercise. During exercise on d 1 of clenbuterol administration, respiratory rate, respiratory minute volume, and heart rate and blood glucose, lactate, and insulin concentrations increased more in beta-A-treated calves than in controls. Oxygen extraction rate and growth hormone concentrations were lower in clenbuterol-treated calves, whereas oxygen consumption, carbon dioxide production, and blood cortisol concentration increased similarly in the absence and presence of the beta-A. After 2 wk of daily clenbuterol administration, respiratory rate and respiratory minute volume during exercise were still higher and oxygen extraction was still lower, whereas all other measures were similar to those in controls. The increased heart rate in response to isoproterenol after 3 wk of clenbuterol administration was reduced markedly in resting but only slightly in exercising animals, whereas heart rate reduction by propranolol during exercise was similar to that in controls. Seven days after withdrawal of clenbuterol, newly administered clenbuterol evoked the same effects as on d 1. In conclusion, there were marked reactions to the first clenbuterol treatment that were in part enhanced during treadmill exercise. After 2 wk of beta-A administration, animals responded much less to the beta-A and changes were not different from those in controls. Resensitization to the beta-A was observed 7 d after its withdrawal.  相似文献

The resistance (R rs) and reactance (X rs) of the total respiratory system were determined at various frequencies in 14 healthy conscious beagle dogs. A pseudorandom noise pressure wave was produced at the nostrils of the animals by means of a loudspeaker adapted to the nose by a tightly fitting mask. A Fourier analysis of the pressure and flow signals yielded meanR rs andX rs, over 16 s, at frequencies from 2 to 26 Hz. The influence of the posture of the dog, the position of its head, the linearity of the respiratory system, the reproducibility of the method and the effects of upper and lower airway obstructions were studied. In sitting and standing healthy dogs with the head in the extended position,R rs values increased progressively with frequency from 5.4±0.4 (SEM) cmH2O L–1 s at 6 Hz up to 8.8±0.7 cmH2O L–1 s at 26 Hz, the mean resonant frequency being 6.1±0.5 Hz. No significant differences were observed between measurements performed with the head in the normal or the extended position. In a recumbent posture, allR rs values were increased butR rs was still dependent on the frequency in the same way (7.1±0.7 cmH2O L–1 s at 6Hz up to 10.0±0.5 cmH2O L–1 s at 26 Hz). Tracheal compression also induced higherR rs values without changes in the frequency dependence or in the resonant frequency.In anaesthetized dogs, airway obstruction was induced by inhalation of histamine (4 mg/ml for 5 min; theR rs values tended to decrease with increasing frequency, and the resonant frequency was markedly increased.Abbreviations FOT forced oscillation technique - IV intravenous(ly)  相似文献

This study was to investigate whether the monofrequency forced oscillation (MFO) technique could be accurately used for pulmonary function testing in calves. The airflow resistance measured by this technique was compared to the resistance measured by the classical reference method by an in vitro study, using an artificial lung model, and by an in vivo study, using 11 healthy calves. The effect of the reference impedance of this oscillation equipment was also investigated. The results show linear relationships (r2 > 0.924, P < 0.001) in the resistance ranges 0 to 0.7 kPa litre-1 s and 0 to 1.0 kPa litre-1 s with a reference impedance of 1.0 kPa litre-1 s and 2.0 kPa litre-1 s, respectively. The MFO resistance values lower and higher than 0.4 kPa litre-1 s are slightly overestimated and underestimated, respectively. It was shown that, if some technical requirements are met, the MFO technique is a simple, reproducible, fast and accurate method which allows measurement of airflow resistance even in field conditions.  相似文献

The aim of the present study was to carry out molecular epidemiological investigation on enterotoxigenic Escherichia coli (ETEC) K99 and Salmonella spp. in diarrheic neonatal calves. Fecal samples were obtained from 220 diarrheic calves at 9 farms related to four governorates in central and northern Egypt. E. coli and Salmonella spp. isolates were examined for E. coli K99 and Salmonella spp. using PCR. ETEC K99 was recovered from 20 (10.36 %) out of 193 isolates, whereas Salmonella spp. was recovered from nine calves (4.09%).Multivariable logistic regression was used to evaluate the risk factors associated with both infections. ETEC K99 was significantly affected by age (P < 0.01; OR: 1.812; CI 95%: 0.566–1.769), colostrum feeding practice (P < 0.01; OR: 5.525; CI 95%: 2.025–15.076), rotavirus infection (P < 0.001; OR: 2.220; CI 95%: 0.273–1.251), vaccination of pregnant dams with combined vaccine against rotavirus, coronavirus and E. coli (K99) (P < 0.001; OR: 4.753; CI 95%: 2.124–10.641), and vitamin E and selenium administration to the pregnant dam (P < 0.01; OR: 3.933; CI 95%: 0.703–1.248).Infection with Salmonella spp. was found to be significantly affected by the animal age (P < 0.05; OR: 0.376; CI 95%: 0.511–1.369), Hygiene (P < 0.05; OR: 0.628; CI 95%: 1.729–5.612), and region (P < 0. 01; OR: 0.970; CI 95%: 0.841–1.624).The results of the present study indicate the importance of PCR as rapid, effective and reliable tool for screening of ETEC and Salmonella spp. when confronted with cases of undifferentiated calf diarrhea. Moreover, identification of the risk factors associated with the spreading of bacteria causing diarrhea may be helpful for construction of suitable methods for prevention and control.  相似文献

In an open, controlled, multi-centre clinical field trial, seven ‘naturally occurring’ outbreaks of acutefebrile (rectal temperature ≥ 39·5°C) respiratory disease in housed calves were treated with a single antimicrobial agent, and either the non-steroidal anti-inflammatory drug (NSAID) carprofen (n=95) or flunixin meghunine (n=92) on an alternate basis. Carprofen was administered as a single subcutaneous injection at a mean dosage of 1·4 mg kg−1 (range 1·2 to 1·9 mg kg−1) body weight on the first day and flunixin meglumine by intravenous injection at a mean dosage of 2·0 mg kg−1 (range 1·2 to 2·6 mg kg−1) body weight on the first 3 consecutive days. All calves were examined clinically immediately prior to initial treatment and on three occasions up to 1 week after the end of treatment. There were no statistically significant differences between NSAID groups in reduction of clinical parameters between examinations, or in overall efficacy. This trial demonstrated that a single dose of carprofen was equally effective as three daily closes of flunixin meglumine as adjunctive therapy to antimicrobial treatment in acute respiratory disease in calves.  相似文献

The objective of this study was to investigate the pharmacokinetics and effect of nalbuphine administered intravenously to calves immediately prior to surgical castration. Ten healthy calves were randomly assigned to two treatments (n = 5): (i) 0.9% sodium chloride (CONT) placebo, (ii) nalbuphine hydrochloride (NAL) (0.4 mg/kg). Blood samples collected over 10 h postcastration were analyzed for nalbuphine and cortisol concentrations. Additionally, heart rate, respiratory rate, rectal temperature, and step count was compared between groups using a random‐effects mixed model. Changes in behavior and attitude were assessed using a six‐point ordinal scoring system and compared using chi‐square analysis. Plasma NAL concentrations were only detectable for 3 h postadministration (T½ = 0.68 h; Range: 0.53–0.79 h). There was no effect of NAL treatment prior to castration on cortisol concentrations (P = 0.99), heart rate (P = 0.73), respiratory rate (P = 0.59), rectal temperature (P = 0.22), and step count (P = 0.08) but fewer calves showed signs of head shaking, kicking, and tail flicking in the NAL group compared with the CONT group (P = 0.036). Therefore, we conclude that a single intravenous injection of nalbuphine at 0.4 mg/kg reduced some pain‐related behaviors but did not significantly eliminate the physiological signs of distress in calves after surgical castration.  相似文献

The aim of the study was to assess the relationship between acute and subacute metabolic and endocrine effects after intravenous administration of the 2-adrenergic agonist clenbuterol in a growth-promoting dose to female pigs. Acute metabolic and endocrine effects were assessed by measuring the blood glucose, serum insulin and nonesterified fatty acid (NEFA) concentrations during 300 min after a single administration of clenbuterol. Significantly higher serum insulin and NEFA concentrations (19.90±2.50 U/ml, p<0.01, and 0.69±0.04 mmol/L, p<0.001, respectively) were measured 30 min after the preprandial administration of clenbuterol in female pigs. Over the same period, the levels of blood glucose (4.42±0.30 mmol/L) showed no difference from those of control pigs. The postprandial serum NEFA concentration decreased moderately during 210 min after feeding. Postprandial blood glucose and insulin concentrations increased and reached maximal levels 120 min after clenbuterol administration (10.91±0.60 mmol/L and 85.22±7.24 U/ml, respectively), and returned to basal levels at 300 min (4.20±0.21 mmol/L and 7.75±1.60 U/ml, respectively) after the administration of clenbuterol. Subacute metabolic and endocrine effects were assessed by measuring the blood glucose, serum insulin and NEFA concentrations for 21 days after the repeated doses of clenbuterol. In addition, the influence of clenbuterol administration on the endocrine regulation of the onset of the next expected oestrus in female pigs was assessed by measuring their serum 17-oestradiol and progesterone concentrations. Blood glucose, serum insulin and NEFA concentrations after the last administration of clenbuterol did not differ significantly from those in control animals. The onset of the next expected oestrus occurred regularly without any significant difference in serum 17-oestradiol or progesterone concentrations between the treated (9.83±2.60 pg/ml and 0.15±0.03 ng/ml) and control pigs (8.52±2.70 pg/ml and 0.25±0.06 ng/ml). The study results suggest the duration of intravenous administration of clenbuterol in a growth-promoting dose necessary to influence the metabolic and endocrine activities in female pigs.  相似文献

The chiral inversion of ketoprofen has been previously demonstrated in cattle, but no studies have been performed on different ages and metabolic situations in the animals. The aim of this work was to study any modifications of the stereoconversion of ketoprofen that occur by reason of age, lactation or gestation in dairy cows. Holando Argentino cattle were divided into three groups: 8 cows in early lactation, 8 pregnant cows and 8 newborn calves. Four animals from each group received the enantiomer R-(–)-ketoprofen by intravenous administration; the other four animals received the S-(+) enantiomer, all at doses of 0.5 mg/kg. Blood samples were collected at standardized times after dosing and assayed for ketoprofen by high-performance reversed-phase liquid chromatography (HPLC). The percentage inversion of R-(–)-ketoprofen to S-(+)-ketoprofen was 50.5% (SD ± 2.4) in the preruminants, 33.3% (SD ± 1.7) in cows in early lactation and 26.0% (SD ± 5.1) in cows in gestation. These results indicate a differing enantioselective metabolic behaviour for one compound in one species under different physiological situations.  相似文献

The effect of Brucella on the generation of microbicidal reactive oxygen and nitrogen metabolites by bovine peripheral polymorphonuclear cells (PMNs) was investigated. The PMNs were recovered from the peripheral blood of control calves and experimental calves previously vaccinated against brucellosis. Significantly larger quantities of NO and H2O2 were generated by PMNs from control and experimental calves following activation by heat-killed whole cells or outer membrane protein of Brucella abortus than by non-activated cells (p<0.05–0.01). In contrast, generation of H2O2 and NO decreased when PMNs were exposed to the lipopolysaccharide of Brucella. However, the generation of H2O2 and NO by activated PMNs from the control and experimental calves did not differ significantly.  相似文献

Five French Alpine goats received 2 mg kg–1 of dichlorvos (DDVP) by intravenous injection and 0.15 mg kg–1 of atropine sulphate 5–10 min later. Ventilatory mechanics, gas exchanges, pulmonary haemodynamics and pulmonary vascular resistance (PVR) were measured before treatment, 5 min after DDVP injection and 5 min after atropine injection.Within 2 min of DDVP administration, all the goats showed acute respiratory distress, excitation and slight muscle fasciculations. A post-inspiratory pause was recorded in 3 goats. Hypersecretion of saliva or nasal discharge was not observed. Dynamic compliance and heart rate decreased significantly and total pulmonary resistance, pulmonary artery and wedge pressures increased significantly. On the other hand, minute ventilation, arterial oxygen and carbon dioxide tensions were not significantly altered by DDVP.Atropine treatment reversed all the clinical and functional parameters, with the exception of the central nervous and muscular signs, which disappeared within 12 h.It was concluded that experimental DDVP toxicosis induced changes in the mechanics of breathing and pulmonary haemodynamics associated with diffuse bronchoconstriction and cardiac insufficiency respectively.  相似文献

The disposition kinetics and urinary excretion of pefloxacin after a single intravenous administration of 5 mg/kg were investigated in crossbred calves and an appropriate dosage regimen was calculated. At 1 min after injection, the concentration of pefloxacin in the plasma was 18.95±0.892 g/ml, which declined to 0.13±0.02 g/ml at 10 h. The pefloxacin was rapidly distributed from the blood to the tissue compartment as shown by the high values for the initial distribution coefficient, (12.1±1.21 h–1) and the constant for the rate of transfer of drug from the central to the peripheral compartment, K 12 (8.49±0.99 h–1). The elimination half-life and volume of distribution were 2.21±0.111 h and 1.44±0.084 L/kg, respectively. The total body clearance (ClB) and the ratio of the drug present in the peripheral to that in the central compartment (P/C ratio) were 0.454±0.026 L/kg h) and 5.52±0.519, respectively. On the basis of the pharmacokinetic parameters obtained in the present study, an appropriate intravenous dosage regimen for pefloxacin in cattle for most of the bacteria sensitive to it would be 6.4 mg/kg repeated at 12 h intervals.  相似文献

The purpose of the study was to correlate electroretinogram (ERG) parameters with increasing levels of plasma, erythrocyte and ocular tissue cholinesterase inhibition using the beagle dog as a model for human neurovisual toxicity. The anticholinesterase compound physostigmine was administered at various steady-state intravenous infusion rates based on pharmacokinetic estimates of plasma and red blood cell cholinesterase inhibition. The most sensitive parameter was the b-wave amplitude of the rod response, which was significantly depressed compared to pretreatment at all levels of acute cholinesterase depression. The overall maximal ERG response demonstrated a trend of declining a-and b-wave amplitudes, which corresponded with the increased levels of cholinesterase depression, but these differences were not significant. The depression of the electroretinogram rod and cone amplitudes appeared to parallel plasma cholinesterase inhibition more closely than erythrocyte cholinesterase activity. Ocular tissue cholinesterase activity was significantly depressed in the retina (70%), cornea (60%) and dorsal rectus extraocular muscle (46%). Five field trials evaluated whether immunization of beef cattle prior to weaning, at weaning, or immediately upon arrival at the feedlot with a commercial bovine respiratory syncytial virus (BRSV) vaccine would reduce subsequent treatment for respiratory disease.

Bovine respiratory syncytial virus vaccination was associated with a significant (p<0.05) reduction in treatment rate in one of three groups of calves immunized prior to weaning (−12%) and in calves immunized upon arrival at the feedlot (−4%).

There was no significant (p>0.05) effect of the BRSV vaccine on treatment rate in calves immunized at weaning, in calves immunized upon arrival at the Saskatoon bull test station, or in yearlings immunized upon arrival at the feedlot.

Although the trend in these field trials was to a sparing effect of the BRSV vaccine, the small reduction in treatment rate may not justify the cost of the vaccination program.

The influence of fasting on the bioavailability and disposition kinetics of albendazole (ABZ) and its metabolites in cattle was investigated. ABZ (10 mg/kg) was given by intraruminal (i.r.) (Experiment 1) and intravenous (i.v.) (Experiment 2) administration to Holstein calves either fed ad libitum (control) or subjected to a 48 h fasting period (fasted group) prior to treatment. The rate of passage of digesta through the gastrointestinal (GI) tract was evaluated by measurement of cobalt faecal excretion following the oral administration of the sodium-cobalt-ethylendiamine-tetracetic acid complex to calves subjected to the feeding conditions above described. Jugular blood and abomasal fluid (via cannula) samples were collected over 120 h post-treatment; samples were analysed by high performance liquid chromatography (HPLC) for ABZ, ABZ sulphoxide (ABZSO) and ABZ sulphone (ABZSO2). Fasting the animals prior to the i.r. treatment resulted in pronounced modifications to the plasma and abomasal fluid disposition kinetics of ABZ and its metabolites. A greater extent of GI absorption with significantly higher Cmax (150%) and AUC (310%) values for ABZSO in plasma, was observed in fasted compared to fed animals following the i.r. administration of ABZ. Extended detection of ABZ metabolites resulting in significantly longer plasma t½el and MRT was also obtained in fasted compared to fed calves. These results correlated with the substantially enhanced availability of ABZ and its metabolites (AUCs over 200% greater) in the abomasal fluid of the fasted animals. Fasting did not induce changes to the plasma disposition of either ABZ or its metabolites after the i.v. treatment. The digesta passage rate, measured by the amount of cobalt excreted in faeces, was significantly lower in fasted compared to animals fed ad libitum. A delayed GI transit time that decreases the rate of passage of the drug down the digestive tract, may have accounted for enhanced ABZ dissolution and absorption in fasted compared to fed calves. The findings reported in this article show that fasting prior to treatment notably affects the bioavailability and disposition kinetics of ABZ and its metabolites in cattle.  相似文献

The aim of the study was to evaluate the pharmacokinetics of flurbiprofen (FBP) in different age groups and physiological status groups in dairy cattle. Ten Argentine Holstein bovines were divided into three different groups: 3 cows in early lactation, 3 cows in gestation and 4 newborn calves. Based on previous experience, all the animals received racemic FBP (50:50) at a dose of 0.5 mg/kg by intravenous administration. Blood samples were taken at predetermined times after administration of flurbiprofen. Plasma enantiomer concentrations were measured by HPLC. Total body clearance (ClB) of (S)-(+)-FBP was higher in calves than in cows (114.5, 136.4, 121.4, 128.9 μg/ml vs 22.0, 24.2, 46.5 μg/ml and 27.6, 25.3, 34.6 μg/ml). In calves the disposition kinetics showed stereoselective behaviour. Area under the concentration–time curve (AUC) was higher and ClB and steady-state volume of distribution (Vss) were lower for (R)-(−)-FBP than for (S)-(+)-FBP. In cows, stereoselectivity was observed in ClB and elimination half-life (fract12)(frac{t_1}{2}) only in the early lactation group. In this study, enantioselective metabolic behaviour of FBP under the physiological situations studied was found. Hence, it is possible that both enantiomers of flurbiprofen may contribute to the drug's therapeutic effects, but further studies with the administration of separate enantiomers will be required to elucidate their metabolism.  相似文献

The purpose of this study was to investigate the effects of some a and β sympathomimetic and sympatholytic drugs on respiratory impedance in healthy conscious calves. Ten Friesian calves were investigated in this study. The forced oscillation technique was used to measure the resistance ( R rs) and the reactance ( X rs) of the respiratory system at frequencies ranging from 4 to 26 Hz. Isoprenaline (1 μg/kg i.v.), propranolol (3 μg/kg i.v.), noradrenaline (2 μg/kg i.v.), xylazine (20 μg/kg i.v.) and yohimbine (0.25 mg/kg i.v.) were administered. Isoprenaline induced a significant decrease of R rs. An increase of R rs after administration of propranolol was observed but without any change of the frequency dependence of R rs. A small increase in the resonant frequency was also recorded. A decrease of R rs was recorded after yohimbine injection. Noradrenaline and xylazine administration increased the resistances and the resonant frequency and induced a negative frequency dependence of R rs. These results suggest that (1) the major effects of β adrenergic drugs are on the central airways, (2) the a adrenergic system may play a role on the regulation of bronchomotor tone in calves, (3) the effects of a adrenergic drugs are on both central and peripheral airways and (4) the forced oscillation technique allows the differentiation of calibre changes occurring in small and large airways.  相似文献

Effects of histamine inhalation were investigated with two different techniques in nine conscious, healthy calves. The oesophageal balloon technique was used to measure the dynamic respiratory compliance (Cdyn) and the pulmonary resistance (RL). The reactance (Xrs) and the resistance (Rrs) of the respiratory system were measured at high frequencies by the forced oscillation technique. These parameters were recorded before and after histamine inhalation. Three histamine dihydrochloride solution concentrations were used (16 mg/ml; 32 mg/ml; 64 mg/ml). Histamine inhalation induced a decrease in Cdyn and Xrs an increase in Rrs, RL and of the resonant frequency and a negative frequency dependence of Rrs. Some of these changes were satisfactorily correlated with the histamine solution concentrations. The resonant frequency was well correlated with Cdvn. It was concluded that histamine inhalation induces a decrease of the calibre of small and large airways and a non-homogeneous behaviour of the pulmonary ventilation in awake calves.
