A serological survey of selected pathogens in wild boar in Slovenia   总被引:6,自引:0,他引:6  
Serum samples collected from 178 shot wild boars (Sus scrofa) were tested for the presence of antibodies against classical swine fever virus, Aujeszky's disease virus (ADV), porcine reproductive and respiratory syndrome virus, porcine respiratory coronavirus (PRCV), transmissible gastroenteritis virus, swine influenza virus, porcine parvovirus (PPV), swine vesicular disease virus, Actinobacillus pleuropneumoniae (APP), Mycoplasma hyopneumoniae, Salmonella spp., Brucella spp. and Haemophilus parasuis (HPS) throughout Slovenia during the hunting season 2003/2004. The number of samples corresponds to 3% of the total hunting bag. By enzyme-linked immunosorbent assay (ELISA) antibodies against ADV were detected in 55 sera (31%), against PRCV in five sera (3%), PPV in 87 sera (49%), APP in 93 sera (52%), M. hyopneumoniae in 38 sera (21%), Salmonella spp. in 85 sera (47%) and HPS in 33 sera (18%).  相似文献

The indirect fluorescent antibody test was modified to provide a rapid technique for the detection, screening and titration of antibodies to transmissible gastroenteritis of pigs. Large numbers of slides containing transmissible gastroenteritis antigen were prepared by planting mixtures of infected and uninfected swine testicular cells onto multiwelled teflon-coated slides. After overnight incubation, about one-half of the cells in each well were infected which provided contrast to aid in detecting specific fluorescence in the presence of varying degrees of background staining. Following fixation, antigen slides were stored at -20 degrees C until used. The indirect fluorescent antibody test was compared to the virus neutralization test in both the screening and titration of swine sera containing transmissible gastroenteritis antibodies. The test was found to be sensitive and reliable and to offer certain advantages over the virus neutralization test.  相似文献

Serum was obtained at weekly intervals after vaccination of 6 healthy calves with either of 2 commercially available monovalent modified-live bovine viral diarrhea (BVD) virus vaccines. Detectable neutralizing antibodies to each of 10 cytopathic and 10 noncytopathic isolates of BVD virus were produced by 1 or more of the calves by 14 days after vaccination, but no calf produced detectable neutralizing antibodies to all 20 BVD viruses. At that time, precipitating antibodies against viral-induced polypeptides of approximately 115,000; 80,000; 56,000; 48,000; 39,000; and 25,000 daltons were detected in sera from some calves. Also at that time, specificity of the antibodies for polypeptides of certain viruses was detected. At 21 days after vaccination, each calf produced neutralizing antibodies to all 20 BVD viruses. At that time, precipitating antibodies to each of the aforementioned viral induced polypeptides were detected in serum from each calf. Precipitating antibodies to viral induced polypeptides of 61,000 and 37,000 daltons were detected in samples of sera obtained from some calves at 42 days after vaccination.  相似文献

Blood samples were taken from 121 sows and gilts on 7 commercial piggeries located around Lusaka (Zambia). The samples tested negative for antibodies to Aujeszky's disease, transmissible gastroenteritis (TGE), swine influenza, hog cholera and brucellosis. Seventy-eight pigs from 5 farms had positive titres to porcine parvovirus. Eighteen sera showed positive titres to Leptospira celledoni.  相似文献

In 15 breeding and fattening pig herds, 85 mice (Mus musculus) and 40 rats (Rattus norvegicus) were captured and bacteria and viruses looked for. Bordetella bronchiseptica, Pasteurella sp., E. coli, Campylobacter jejuni and Treponema sp. were isolated from different samples. Rota-virus was also identified and neutralizing transmissible gastroenteritis antibodies were detected in the serum of one rat and mice from three different farms. Wild rats were also orally infected with Aujeszky's disease virus (ADV) and classical swine fever (CSF) virus. All the rats survived the ADV experimental infection and some of them showed ADV neutralizing antibodies in their sera. No multiplication of the SF virus was obtained.  相似文献

A serodiagnostic ELISA utilizing the recombinant nucleoprotein (rN protein) of transmissible gastroenteritis virus (TGEV) was developed, and evaluated by examining a panel of 141 virus neutralization (VN) positive and 101 negative sera. The rN protein-based ELISA (rnELISA) appeared to be highly sensitive and specific (98.6% and 98.0%, respectively) when it was compared to the VN test. The result was similar to that of an ELISA based on purified viral antigens with showing good correlation (R=0.829). No cross-reaction was detected with antisera against porcine epidemic diarrhea virus, hog cholera virus, type A rotavirus, pseudorabies virus and swine vesicular disease virus in this ELISA. The rnELISA can be an alternative for the diagnosis of TGE with a great advantage in antigen preparation.  相似文献

Samples of sera were obtained from 5,725 cows in a semiclosed herd. In each of the preceding 7 years, the herd was vaccinated against bovine viral diarrhea (BVD) with killed virus. Neutralizing antibody tests were done on all samples of sera, using cytopathic virus, BVD-TGAC virus, that was antigenically distinct from the vaccine virus. Most samples of sera had high titers of neutralizing antibodies against BVD-TGAC virus. In 48 samples of sera, neutralizing antibodies were not detected against BVD-TGAC virus, but were detected against the vaccine virus. Neutralizing antibodies against selected noncytopathic BVD viruses were not detected in several samples of serum that had neutralizing antibodies against the vaccine virus and BVD-TGAC virus. Noncytopathic BVD virus was isolated from sera obtained from 3 cows less than 4 years old. Two cows were available for further testing, and persistent infection with BVD virus was confirmed in both cows. The BVD viruses isolated from those cows were not neutralized by several samples of sera. Immunoprecipitation of polypeptides induced by the vaccine virus was done with selected samples of serum. Two patterns of immuno-precipitated viral-induced polypeptides were identified. One pattern was consistent with exposure of cows with live virus. The other pattern was consistent with exposure of cows with only the killed virus vaccine.  相似文献

The presence of serum antibodies directed against classical swine fever (CSF) virus and other pestiviruses among the wild boar (Sus scrofa) population in Croatia was investigated. During 2003, serum samples from 214 wild boars were collected in 10 hunting areas in the continental part of the country.The sera were examined by enzyme immunoassay (ELISA) and in the virus neutralization test (VNT). Out of 214 sera tested 111 (51.87 %) were positive by ELISA and regarding neutralising antibodies, against CSFV 75 (35.05 %) samples were positive. In the VNT with the C-strain (conventional live vaccine strain China) and the strain Uelzen were used. Samples were also tested for neutralizing antibodies against border disease virus (BDV) using the strain 137/4 and against bovine viral diarrhoea virus (BVDV) using the NADL strain. Neutralizing antibodies against the C-strain were detected in 36 sera (16.82 %), against strain Uelzen in 17 sera (7.94 %) and in 22 sera (10.28 %) against both strains. In five sera (2.33 %) neutralizing antibodies against BVDV and BDV were found.  相似文献

Monospecific antisera were prepared in rabbits against canine coronavirus (CCV) and transmissible gastroenteritis virus of pigs (TGEV), and in 24 pigs and 3 cats against TGEV alone. Neutralizing antibody titres were higher for the immunizing than the heterologous virus, although cross-neutralization usually was detected. This confirmed that CCV and TGEV are distinct, but antigenically related coronaviruses. In sera from 41 dogs, CCV-neutralizing titres were on average 2.7 fold higher than TGEV-neutralizing titres, suggesting that CCV was the causal agent. Sera from 29 cats in colonies with feline infectious peritonitis (FIP) and known to contain TGEV-neutralizing antibody, were found to have titres 12.3 fold higher against CCV. The FIP virus (FIPV) is probably more closely related to CCV than TGEV as judged by antigens involved in virus neutralization.Antisera to two isolates of bovine coronavirus, three isolates of haemagglutinating encephalomyelitis virus, seven strains of avian infectious bronchitis virus and the 229E strain of human coronavirus all failed to neutralize CCV and TGEV. Thus CCV, TGEV and probably FIPV fall into a group of antigenically related agents, separable from other members of the family Coronaviridae, by both virus neutralization and immunofluorescence tests.  相似文献

Staphylococcal protein-A (SpA) is known to interact with the crystallizable fragment (Fc) of IgG molecules from several species. In the present study, SpA coupled to either fluorescein isothiocyanate (FITC) or peroxidase was used in place of antisera to IgG for the fluorescent antibody (FA) techniques and the enzyme linked immunoassay (ELISA). The SpA conjugates produced low background staining when applied in these techniques, and provide a rapid, highly specific and sensitive means for the identification of viral isolates and the detection of serum antibodies. Moreover, SpA is a single reagent that replaces various preparations of anti-gamma globulin against many species. SpA-FITC conjugate was successfully applied for the identification of pseudorabies virus, hog cholera virus, swine vesicular disease virus, transmissible gastroenteritis virus, porcine parvovirus and porcine enteroviruses. Antibody titers against the mentioned viruses could be determined semi-quantitatively in the indirect FA test with SpA-FITC. In our laboratory the ELISA became a routinely practicable serological test for the detection of antibodies only after we introduced SpA-peroxidase as a marker for the IgG.  相似文献

A double-antibody ELISA for the detection of coronaviruses in intestinal contents from turkey poults with diarrhea was developed. Antibodies were raised in rabbits and guinea pigs against a Minnesota isolate of turkey enteric coronavirus (TCV) propagated in embryonating turkey eggs and were purified by density-gradient centrifugation. The specificity of antisera was confirmed by hemagglutination-inhibition and immunoelectron microscopy. Absorption of anti-TCV hyperimmune sera with egg extracts or egg ovalbumin and the use of different dilution and blocking buffers influenced the sensitivity and specificity of the ELISA. Reciprocal cross-reactivity was detected among turkey, chicken, bovine, and murine coronaviruses. Antisera to the transmissible gastroenteritis virus of swine, the rabbit enteric coronavirus, or the human coronavirus strain 299E failed to react with TCV. The TCV cross-reacted only moderately with the avian infectious bronchitis virus and the hemagglutinating encephalomyelitis virus of swine. Investigations with samples from 47 commercial turkey flocks in Quebec with episodes of transmissible enteritis revealed that the ELISA was more sensitive than was electron microscopy for detection of TCV.  相似文献

During recent years neutralizing antibodies against Border Disease Virus (BDV) were found repeatedly in German pig herds. Consequently there was a demand for a differential diagnostic system. A permanent sheep cell line and BDV reference strain Moredun were chosen and were applied in a could be used case study. A pestivirus could be isolated from piglets on a mixed farm and was characterised as 'non-Classical Swine Fever' (CSF) by using monoclonal antibodies. Due to a CSF suspicion the pig herd was destroyed immediately. Serum samples of sheep from the same farm were used for further characterisation of the new virus isolate. A neutralization test of the sheep sera was performed against different pestiviruses and the new isolate. Neutralizing antibody titres against the new virus pig isolate were significantly higher than against all other pestiviruses. BDV strain Moredun recognised the antibodies clearly, whereas CSF viral strain Alfort 187 and several isolates of bovine viral diarrhoea virus (BVDV) strains scored the lowest cross reaction.  相似文献

A long-term experiment in sheep inoculated intranasally with 2 strains of Norwegian maedi virus was carried out in 2 groups of Norwegian Dala sheep (7 sheep/group). Virus-specific cellular immune response was assayed in the lymphocyte transformation test sequentially during 3 years after sheep were inoculated in group 1 and 4 times in the 3rd year in group 2. Humoral immune response was assayed by immunodiffusion, complement-fixation, and neutralization tests on sequential serum samples collected from the 2 groups. Attempts to isolate virus were made. All group 1 sheep showed transient and irregularly recurring cellular immune responses. In group 2, 6 of the 7 sheep gave similar responses. The frequency of virus isolations was low compared with that reported by various research workers using other breeds for studying experimental maedi-visna infection. Precipitating antibodies were detected earlier, and in more animals, than were complement-fixing antibodies. Both were, however, detected later and less frequently than were reported by other research workers. There was a marked difference in the capability of the 2 maedi virus strains to induce neutralizing antibodies. The sequential sera usually showed distinct differences in neutralizing capacity of the virus strains, indicating that they are antigenically different.  相似文献

From 16 (14%) out of 112 dead or euthanized seals originating from wildlife and seal orphanages phocine morbillivirus was isolated. The majority of viral isolates in cell cultures was obtained from lung homogenates of 15 out of 71 free-ranging seals (21%). The virus was isolated by longterm cultivation in roller cultures of seal kidney cells. The phocine morbillivirus was detected by typical cytopathogenic alteration and by peroxidase-linked antibody (PLA) assay, respectively. A neutralization test based on PLA was used for antibody detection in seals using a canine distemper virus (CDV) strain and in parallel one of the phocine morbillivirus isolates. All sera tested were proven to contain neutralizing antibodies of higher titres against the latter virus than against the CDV strain. Several seals furnished morbillivirus isolates and at the same time exhibited neutralizing antibodies of low to medium titres. No viral isolates were obtained from the majority of sick animals with moderate to high neutralizing titres (greater than 1/1,000). The significance of these findings is discussed in relation to the cause of the mass mortality amongst seals observed in 1988 in the Bay of Heligoland.  相似文献

The objective of this study was to evaluate the seroprevalence and identify the strains of swine influenza virus (SwIV), as well as the seroprevalence of porcine parvovirus (PPV), transmissible gastroenteritis virus (TGEV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine respiratory coronavirus (PRCV), porcine circovirus type 2 (PCV-2), and classical swine fever virus (CSFV) in pigs in Trinidad and Tobago (T&T). Blood samples (309) were randomly collected from pigs at farms throughout T&T. Serum samples were tested for the presence of antibodies to the aforementioned viruses using commercial ELISA kits, and the circulating strains of SwIV were identified by the hemagglutination inhibition test (HIT). Antibodies against SwIV were detected in 114 out of the 309 samples (37%). Out of a total of 26 farms, 14 tested positive for SwIV antibodies. HI testing revealed high titers against the A/sw/Minnesota/593/99 H3N2 strain and the pH1N1 2009 pandemic strain. Antibodies against PPV were detected in 87 out of the 309 samples (28%), with 11 out of 26 farms testing positive for PPV antibodies. Antibodies against PCV-2 were detected in 205 out of the 309 samples tested (66%), with 25 out of the 26 farms testing positive for PCV-2 antibodies. No antibodies were detected in any of the tested pigs to PRRSV, TGEV, PRCV, or CSFV.  相似文献

Cross-protection studies between the feline infectious peritonitis (FIP) and the porcine transmissible gastroenteritis (TGE) viruses were conducted in cats, pigs and pregnant gilts. Cats vaccinated with TGE virus developed neutralizing antibodies against TGE virus and low titer antibody against FIP virus detected by an indirect fluorescent antibody technique but were not protected against a virulent FIP virus challenge. Baby pigs and pregnant gilts vaccinated with FIP virus did not develop detectable antibodies to TGE virus. Nevertheless, it appeared that vaccination of swine with FIP virus conferred some immunity against TGE virus infection. Seventeen-day-old pigs vaccinated with two doses of FIP virus had a 67% survival rate following a virulent TGE virus challenge, and 75% of the 3-day-old pigs suckling either FIP or TGE-virus-vaccinated gilts survived virulent TGE virus infection in contrast to 0% survival of baby pigs suckling unvaccinated gilts.  相似文献

Swine exposed to attenuated transmissible gastroenteritis virus had higher virus-neutralizing antibody titers than did swine exposed to virulent virus. The cellular response, measured by the direct leukocyte migration-inhibition (LMI) procedure, was greater in swine exposed to virulent virus than in swine exposed to the attenuated virus. Leukocytes from exposed swine were inhibited more in the LMI procedure in the presence of the homologeous sensitizing antigen than in the presence of the heterologous viral antigen. The humoral response measured by virus neutralizing reached a peak 21 days after exposure, and the cellular response measured by LMI reached a peak 28 days after exposure.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed for detection and quantification of serum antibodies to transmissible gastroenteritis virus (TGEV) in swine. Sera from pigs inoculated with cell culture-origin TGEV or gut-origin TGEV were tested for anti-TGEV antibody by ELISA and by serum virus-neutralization test (NT). The ELISA detected antibody 3 days (av) sooner than did the NT when sera from pigs inoculated with cell culture-origin TGEV were tested and 1 day sooner than did the NT when sera from pigs inoculated with gut-origin TGEV were tested. The ELISA appeared to be more sensitive than the NT, since ELISA was more responsive to low-level antibody and ELISA titers exceeded NT titers.  相似文献

An in vitro leukocyte-aggregation assay was developed to detect the exposure of swine to transmissible gastroenteritis virus. Leukocytes in heparinized blood samples aggregated when mixed with test antigen prepared from transmissible gastroenteritis-infected swine testicle cell cultures. Twenty-two of 23 swine exposed 3 days or more were positive or suspects in the assay; 6 nonexposed swine were negative. Aggregation was shown as early as 3 days postexposure in 1 sow and persisted for as long as 14 months in another. Persistence of the assay was proved by repeated evaluations on 2 experimentally exposed swine.  相似文献

Studies were performed to determine if passive immunization with hyperimmune sera generated to specific Newcastle disease virus (NDV) proteins conferred protection against virus challenge. Six groups of 3-wk-old chickens were passively immunized with antiserum against either hemagglutinin-neuraminidase/fusion, (HN/F) protein, nucleoprotein/phosphoprotein (NP/P), Matrix (M) protein, a mixture of all NDV proteins (ALL), intact ultraviolet-inactivated NDV (UVNDV), or negative sera. Blood samples were collected 2 days postimmunization, and the birds were challenged with Texas GB strain of NDV. Antibody titers were detected from those recipient birds that had received the antisera against the HN/F, ALL, or UVNDV by a hemagglutination inhibition test, an enzyme-linked immunosorbent assay (ELISA), and a virus neutralization test. Antibodies were detected only by the ELISA from the birds that had received antisera against NP/P and M protein. Antibody titers in the recipient birds dropped by two dilutions (log2) after 2 days postinjection. Birds passively immunized with antisera against HN/F, ALL, and UVNDV were protected from challenge, whereas chickens passively immunized with antisera against NP/P and M protein and specific-pathogen-free sera developed clinical signs of Newcastle disease. The challenge virus was recovered from the tracheas of all passively immunized groups. The presence of neutralizing antibodies to NDV provided protection from clinical disease but was unable to prevent virus shedding from the trachea.  相似文献
