Reason for performing study: The recovery quality scoring systems (RQSSs) in current use have not been critically reviewed for reliability. Objective: To examine reliability (reproducibility) of 4 RQSSs when applied to a ranked series. Methods: A DVD incorporating the recordings of 9 horses recovering from general anaesthesia was evaluated by final year students over 5 days. On Day 1, each evaluator ranked recoveries from 1–9 (1 = best). Over the following 4 days, each evaluator scored the same recoveries using 4 different RQSSs (3 of them in common usage and previously published) applied in random order. The scores from each RQSS were ranked and plotted against the Day 1 ranking of each evaluator to establish the extent of agreement using generalisability theory. The same 9 recoveries were also ranked by 12 experienced equine anaesthetists and the Spearman Rank Correlation coefficient calculated to determine the agreement between experienced and inexperienced evaluators. Results: The recoveries were evaluated by 117 students. All 4 RQSSs were equally reliable with low (<4%) interobserver variability. The main (80%) source of total variation arose from differences between horses. The overall ranking within each RQSS was strongly correlated with Day 1 ranking. There was strong correlation (r = 0.983) between the students' ranking and that established by experienced anaesthetists. Interobserver reliability was similar with all 4 RQSSs. Conclusion: All 4 RQSSs studied were similarly reliable. Potential relevance: The selection of a universally acceptable RQSS from amongst the 4 examined can be based on criteria other than reliability, e.g. ease of use. This will facilitate wider scale multi‐centre studies in recovery quality after anaesthesia in horses.  相似文献

Ultrasonography allows early and reliable pregnancy detection in several domestic animals. Transrectal sonography can be recommended in horses and cattle, transrectal or transcutaneous procedures in sheep, goats and pigs while transcutaneous ultrasound scanning is appropriate in dogs and cats. Three periods of time can be distinguished in the diagnosis of early pregnancy by ultrasound: the earliest phase, where signs of pregnancy can be found in some cases, but accuracy of diagnosis is very low; the succeeding period, where reliable diagnosis is possible, but results are often difficult to achieve and examination can be time consuming; the third phase in which ultrasound diagnosis is very accurate even under practice conditions and can be efficiently carried out on a large scale basis in breeding management. To achieve high accuracy in determining pregnancies a suitable ultrasound scanner with a good image quality and considerable expertise are required. Under field conditions ultrasonic pregnancy diagnosis should be possible: from Day 14 in the horse, from Day 25-28 in cattle, from Day 25 (transrectal), resp. 35 (transcutaneous) in sheep, goats and swine and from Day 24-25 (transcutaneous) in the dog and cat. Before this period of time reliable diagnosis is sometimes possible: between Day 11 and 13 in the horse, between Day 20 and 25 in cattle, between Day 20 and 25 (transrectal), resp. between Day 30 and 35 (transcutaneous) in sheep and goats, between Day 20 and 25 (transrectal) and Day 22 and 35 (transcutaneous) in swine and between Day 19-20 and 24-25 in the dog and cat. Earlier sonographic indications of pregnancy are not sufficiently reliable for large scale accurate pregnancy diagnosis.  相似文献

The SIV infection of rhesus macaques (Macaca mulatta) is the most appropriate animal model in HIV research. The permanent human T-cell line C8166 is used for in vitro SIV propagation. This paper describes ultrastructural features of the cells after infection with SIVmac. The C8166 cells are ultrastructurally characterized by a heterogenous morphology which is independent of the infection. SIV induced cell syncytia are observed 18 hours after infection. Viral particles and budding occur 48 hours p.i with a peak at the day 8. Viral particles present the typical lentiviral morphology. Using the monoclonal antibody anti SIVp28 and ultra small (0.8 nm) immunogold-silver enhancement technique, we are able to demonstrate SIV antigen immunoelectron microscopically. Therefore, this ultrastructural method is suitable to detect SIV antigen in in vivo experiments with C8166 cells from day 8 p.i. serving as positive control.  相似文献

In order to study the aerosol transmission of arthropathic and amyloidogenic Enterococcus faecalis strains, preliminary aerosol experiments were performed. The experiments were carried out in empty isolators to assess the yield and viability of E. faecalis and Newcastle disease vaccine virus (NDVV) aerosol particles with time. NDVV was aerosolized because this virus would be used in combination with E. faecalis in a subsequent study. Concentrations of about 10(5) colony-forming units (CFU) of E. faecalis/ml of air were still found 30 min after the aerosol application. At 45 min, however, E. faecalis concentrations dropped below the detection level. The average E. faecalis concentration during the aerosol experiment was estimated at 10(5) CFU/liter. The NDVV aerosol generated an average of 10(4)-10(5) 50% embryo infective dose per liter of air. In these experiments, E. faecalis and NDVV aerosols were successfully generated despite considerable initial particle loss. The bacteria and virus uptakes per chick are discussed in case day-old chicks would be exposed to these aerosols.  相似文献

为了筛选出防治葡萄茶黄蓟马效果较好的药剂,减少农药的使用量,本研究采用药膜浸叶法测定了17种药剂对茶黄蓟马成虫的室内毒力。结果表明氟虫腈、虫螨腈、高氯氰菊酯、吡虫啉、功夫等药剂对茶黄蓟马表现出较高的毒力,建议在田间防治中优先考虑使用这些药剂;毒死蜱、丁醚脲、阿维菌素、丁硫克百威和丙溴磷的毒力效果相对较低,建议在生产中尽量少用。本研究对茶黄蓟马的室内毒力测定结果可为今后化学防治该害虫提供一定的科学依据。  相似文献

Toxicity assessment of nanoparticles, now widespread in our environment, is an important issue. We have focused attention on the carcinogenic potential of copper oxide (CuO) and titanium dioxide (TiO(2)). In experiment 1, a sequential pilot study, the effectiveness of a carcinogenic bioassay featuring intraperitoneal injection (i.p.) of 20 mg 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) or 0.1% N-bis(2-hydroxypropyl)nitrosamine (DHPN) in drinking water for 2 weeks was examined. Based on the results, DHPN, as the lung carcinogen, and evaluation at week 30 were selected as the most appropriate for our purposes in Experiment 1. In experiment 2, the carcinogenic bioassay was used to assess the carcinogenic potentials of instilled nanoparticles of CuO and TiO(2). There were no significant intergroup differences in the lung neoplastic lesions induced by DHPN, although the neoplastic lesions induced by the nanoparticles in the CuO or TiO(2) intratracheal instillation (i.t.) groups, demonstrated a tendency to increase compared with the microparticles administration. At the very least, the carcinogenic bioassay with DHPN proved useful for assessment of the modifying effects of instilled particles, and further assessment of the carcinogenic potential of nanoparticles appears warranted.  相似文献

The effects of pre-ovulatory and post ovulatory insemination on pregnancy rate and embryonic-loss rate were studied in 268 mares in two experiments. Within each experiment mares were randomised within replicates as follows: to be inseminated on the day the pre-ovulatory follicle reached 35 mm (pre-ovulatory group), to be inseminated on the day of ovulation (Day 0 group), and to be inseminated on the day after ovulation (Day 1 group). Ultrasonic pregnancy diagnoses were performed on Days 11, 12, 13 and 14 (Experiment 1) and Days 11, 12, 13, 14, 15, 20 and 40 (Experiment 2). Combined for the two experiments, pregnancy rates were different (P less than 0.01) among the three groups. For Experiment 2, pregnancy rate within the pre-ovulatory group was lower (P less than 0.05) for insemination 4 days or more before ovulation than for up to 3 days before ovulation. Pregnancy rate was lower (P less than 0.05) for the Day 0 group than for the pre-ovulatory group inseminated up to 3 days before ovulation. In Experiment 2, ovulation was detected by examinations every 6 h; pregnancy rate was greater (P less than 0.05) for mares inseminated 0 to 6 h after ovulation than for mares inseminated at 18 to 24 h. No pregnancies occurred when mares were inseminated 30 h or more after ovulation. The mean day of first detection of the embryonic vesicle was different (P less than 0.0001) among the three groups. 为了获得高质量的金标孕酮单抗,通过采用柠檬酸三钠还原法,制备出符合要求的30 nm胶体金溶液,在此基础上摸索胶体金标记孕酮单克隆抗体的最佳条件,并对其进行质量鉴定。结果发现,用柠檬酸三钠还原法制备的30 nm胶体金溶液呈深红色,透射电镜观察,胶体金颗粒大小基本一致;胶体金标记孕酮单克隆抗体的最低稳定浓度为16 μg/mL,最适稳定浓度为19.2 μg/ mL,最适标记pH为8.5。制备的金标抗体复合物经电镜观察,周围有一层明显的低电子密度晕圈,且其D520 nm值在0.2~0.4之间。  相似文献

The beta-agonist clenbuterol in mane and tail hair of horses   总被引:2,自引:0,他引:2  
REASONS FOR PERFORMING STUDY: The beta2-agonist clenbuterol is commonly administered for therapeutic purposes in the horse, but its use an an anabolic agent is illegal. Clenbuterol can be detected in blood and urine for a relatively short period after administration and detection in hair could enhance the analytical range and be used to determine the history of clenbuterol application. HYPOTHESIS: That detection in mane or tail hair is possible over an extended period. METHODS: Four horses received 0.8 microg clenbuterol hydrochloride/kg bwt b.i.d. for 10 days. Four other horses were used as untreated controls. Blood, urine, mane and tail hair samples were taken on Day 0 (before) and 5, 10, 30, 35, 40, 60, 90, 120, 150 and 360 days after start of treatment. Gas chromotography/high resolution mass spectrometry (GC/HRMS) was developed for clenbuterol analysis: limit of detection was 0.2 pg/mg; intra-assay repeatability limit r = 0.06 (confidence level 95%); interassay repeatability limit r = 0.03 (confidence level 95%). Prior to treatment, clenbuterol was absent from all samples analysed. RESULTS: Clenbuterol was detectable as early as Day 5 in tail and mane hair of Segment 1 (0-20 mm from the roots) and was maximal on Day 90. However, as time progressed, shift into lower 20 mm segments was observed. On Day 360, the maximum concentration (up to 21 pg/mg) was located in Segment 13, i.e. 26-28 cm from roots of hair. Clenbuterol was not detectable in blood or urine after Day 30. Mane and tail hair results were very similar. CONCLUSIONS: The study showed that the beta-agonist clenbuterol can be found in mane and tail hair of horses after extended periods. POTENTIAL RELEVANCE: It will be possible to detect clenbuterol in breeding and show horses where anabolic drugs have been used illegally to improve conformation. This method may also be helpful to monitor therapeutic clenbuterol treatment.  相似文献

High levels of anthelmintic resistance in gastrointestinal nematodes (GIN) of small ruminants have created the need for alternative approaches to parasite control. Copper oxide wire particles (COWP; 2g) have proven effective in decreasing GIN infection in lambs. However, the risk of copper toxicity has limited the usefulness of this approach. Recently, smaller doses (0.5 and 1g) have proven effective in GIN control, reducing the risk of toxicity. The objective of this study was to examine the effectiveness and risk of toxicity using multiple small doses of COWP for GIN control in lambs between weaning and market weight. Dorper crossbred ram lambs were orally administered levamisole (Levasol, 8.0mg/kg; n=8), 0.5g (n=9), or 1g COWP (n=9) at weaning (Day 0; 118+/-2 days of age; late May 2005) and again at 6-week intervals for a total of four treatments. A pooled fecal culture determined that Haemonchus contortus was the predominant gastrointestinal parasite at weaning. Lambs grazed bermudagrass pastures and were supplemented with up to 500g corn/soybean meal and free choice trace mineralized salt. Fecal egg counts (FEC), packed cell volume (PCV), and plasma aspartate aminotransferase (AST) activity were determined every 14 days and lambs weighed every 28 days. GIN infection reached a peak at Day 42 (high FEC, low PCV). COWP effectively reduced FEC on Days 0 and 42 compared with the previous week, but did not reduce FEC on Days 84 and 126 (treatment by time interaction, P<0.005). Plasma AST activity and weight gains were similar among treatment groups throughout the study period. Concentrations of copper in the liver on Day 155 were greater in COWP-treated lambs (P<0.001), but all concentrations were normal. Multiple doses of COWP were as effective as levamisole for control of H. contortus without risk of copper toxicity.  相似文献

Fecal samples were examined immediately before and 24 to 48 h after cestocide treatment for a comparative detection of tapeworm-positive horses. In early winter, 17 weanlings, 20 yearlings, 15 2-year-old horses, 24 breeding mares, and 2 stallions were treated with praziquantel in combination with a macrocyclic lactone. The horses were presumed to be naturally infected with tapeworms after pasture grazing. Fecal samples were collected before treatment (Day 0), at 24 or 48 h after treatment (Day 1-2), and 16 to 21 d after treatment (Day 16-21). A Wisconsin test was done on all fecal samples. Odds of detection of infection for all age groups increased by a factor of 2.04 [95% confidence interval (CI): 1.30 to 3.20] from Day 0 to Day 1-2 (P = 0.002).  相似文献

Gastric ulcers are common in horses. The purpose of this study was to test the effect of porcine hydrolysed collagen (PHC) on gastric ulcer scores and gastric juice pH in horses. We hypothesise that PHC-administration will result in improved gastric lesion scores and act synergistically with omeprazole to improve treatment efficacy. Thoroughbred horses (n = 10) were studied in a 2-period, 2-treatment crossover design, where the PHC (45 g) was administered twice daily. Horses were treated for 56 days. Gastroscopy was performed and gastric juice pH measured on Days 0, 14, 28, 42, 49 and 56. Nonglandular gastric ulcer number (NGN) and severity (NGS) and glandular ulcer number (GN) and glandular severity (GS) scores were assigned by an investigator masked to treatment and serum gastrin and amino acid concentrations. By Day 42, 2 weeks after discontinuing omeprazole treatment, NGN and NGS scores returned to pretreatment values and serum gastrin was higher when compared to values measured on Day 28. By Day 49, after the feed-deprivation period, NGN and NGS were similar to pretreatment values. By Day 56, mean NGN score was significantly lower in PHC-treated horses, compared to controls. Mean gastric juice pH significantly increased in both groups on Day 28 and the pH was significantly (P = 0.0127) higher in the PHC-treated horses. Serum amino acid concentrations were not significantly different 2 h after feeding PHC and hydroxyproline was not detected. Serum gastrin concentration did not increase 2 h after feeding in the PHC-fed horses. The PHC fed to horses enhanced the effects of omeprazole on increased gastric juice pH, inhibited gastrin secretion after feeding and resulted in fewer nonglandular ulcers after long-term feeding (56 days) in stall-confined horses undergoing intermittent feeding.  相似文献

AIM: To evaluate the efficacy and tolerance of an extract of green-lipped mussel (GLME) in the management of mild-to-moderate degenerative joint disease (DJD) in dogs. METHODS: Eighty-one dogs presumptively diagnosed with DJD were treated orally daily with either GLME or a placebo for 56 days, in a double-blind, placebo-controlled study. In an uncontrolled open-label extension to the study, all dogs were treated with GLME for an additional 56 days (from Days 57-112). Clinical signs were subjectively scored by the owners, and findings of detailed musculoskeletal examinations were scored by one veterinarian. Efficacy was assessed from a qualitative comparison of the proportion of dogs with improved clinical signs, and a quantitative comparison of the scores of the musculoskeletal examinations, between groups. Haematological and biochemical analyses and reports by owners of possible adverse drug reactions were used to screen for evidence of toxicity. RESULTS: There was close agreement between assessments by the veterinarian and owners. The clinical signs of DJD in both GLME-treated and placebo groups improved significantly over baseline by Day 28; this improvement continued over the entire course of the study. There were no significant differences between groups on Day 28. On Day 56, a higher proportion of dogs in the GLME-treated group had improved clinical signs (p=0.018), and GLME-treated dogs had marginally better (p=0.053) musculoskeletal scores than dogs in the placebo group. The differences between the groups were no longer apparent by Day 112, by which time the former placebo group had been receiving GLME for 56 days in the open-label phase of the study. The proportion of dogs in the former placebo group that had improved by Day 112 (29/32; 91%) was significantly greater (p=0.012) than the proportion improved at Day 56 (15/37; 41%). No signs of toxicity were apparent. CONCLUSIONS AND CLINICAL RELEVANCE: GLME had a beneficial effect on the clinical signs of dogs presumptively diagnosed with mild-to-moderate DJD. Long-term therapy may be required before improvement is apparent.  相似文献

There is a discrepancy in the literature on potential digesta separation mechanisms in horses, with both a selective retention of fine and of large particles postulated in different publications. To assess the net effect of such mechanisms, we fed ponies on a hay‐only diet a pulse dose of whole (unchopped) marked hay together with a solute marker, collected faeces on a regular basis, measured marker concentrations in whole faeces and in their large (2.0–16 mm), medium (0.5–1.0 mm) and small (0.063–0.25 mm) particle fraction, and calculated the corresponding mean retention times (MRTs). For comparison, the same experiment was performed in goats. In goats, as expected, MRTsolute (35 hr) was significantly shorter than MRTparticle (51 hr); only a very small fraction of particle marker was excreted as large particles (2%); and the MRT of these large particles was significantly shorter than that of small particles (with a relevant difference of 8.6 hr), indicating that those few large particles that escape the rumen do so mostly soon after ingestion. In ponies, MRTsolute (24 hr) did not differ from MRTparticle (24 hr); a higher fraction of particle marker was excreted as large particles (5%); and the MRT of these large particles was longer than that of small particles (but with a non‐relevant difference of less than 1 hr). These results indicate that no relevant net separation of digesta phases occurs in horses and that selective particle retention mechanisms in the large intestine are unlikely to represent important characteristics of the horse's digestive physiology.  相似文献

The prophylactic efficacy of a single oral dose of toltrazuril against coccidiosis (mixed Eimeria infections) in naturally infected lambs was evaluated in two experiments. Toltrazuril at 20 mg kg-1, given on Day 7 or Day 10 after turnout on pasture, proved to be highly efficacious in preventing clinical coccidiosis under Norwegian conditions. Toltrazuril reduced the oocyst output to very low levels, prevented the development of diarrhoea and improved weight gain during the first 4-5 weeks after treatment. Treatment on Day 7 was superior to treatment on Day 10 with respect to improving weight gain and preventing the development of soft faeces. Lambs treated with toltrazuril on Day 7 seemed to be as immune as untreated lambs to natural reinfections with coccidia later in the grazing season. In one of the experiments, natural infections with the nematode Nematodirus battus produced a coccidiosis-like disease in some lambs simultaneously with the outbreak of coccidiosis.  相似文献

This study used a cross-over design to compare the efficacy of 3 corticosteroids for the relief of airway obstruction and inflammation in 9 heaves-affected horses. The severity of airway obstruction and inflammation was quantified by measurement of lung function and by bronchoalveolar lavage fluid (BALF) cytology, respectively. Airway obstruction was induced by stabling the horses and they remained stabled during the 10 day treatment period. Lung function was measured before treatment (baseline), at Days 3, 7, and 10 of treatment, and after 30 days at pasture. BALF cytology was investigated at baseline, Day 10, and at pasture. All 9 horses received the following 4 treatments in random order: no treatment, daily oral prednisone tablets (1 mg/kg), daily i.v. dexamethasone solution (0.1 mg/kg), and i.m. dexamethasone-21-isonicotinate (0.04 mg/kg) every 3 days. When horses received no treatment, lung function did not change significantly during stabling but improved at pasture. In all horses, daily i.v. administration of dexamethasone solution improved lung function within 3 days to levels as good as or better than those measured at pasture. Dexamethasone-21-isonicotinate was rapidly effective in 8 of 9 horses. The other horse did not respond to this drug. Prednisone tablets were without effect on Days 3 and 7 of treatment, but by Day 10, 5 of 9 horses showed some improvement in lung function. Dexamethasone i.v. solution decreased the percent neutrophils in BALF at Day 10. Other treatments had no effect on BALF cytology. These results demonstrate that dexamethasone rapidly relieved airway obstruction in heaves-affected horses. Oral prednisone had inconsistent effects but may be beneficial in some horses after more than a week of treatment.  相似文献

OBJECTIVE: To determine by pulmonary perfusion scans and ultrasonography if embolemia occurs during total hip replacement (THR) surgery in dogs. STUDY DESIGN: Prospective clinical study. ANIMALS: Forty client-owned dogs that had THR surgery. METHODS: Thoracic radiographs were taken immediately after THR and immediately after completion of (99m)Tc-MAA lung scans. Scintigraphy was performed in 28 dogs, 48 hours after THR. Intraoperative ultrasonography (intercostal or transesophageal) was performed in another 12 dogs that had THR. The right atrium and ventricle and pulmonary outflow tract were observed during and for 5 to 8 minutes after femoral component insertion into medullary canals prepared by reaming, and lavage and aspiration of debris before filling with polymethylmethacrylate in dough stage. A modified Prospective Investigation of Pulmonary Embolism Diagnosis (PIOPED) classification system was used to evaluate lung scans. RESULTS: No pulmonary radiographic abnormalities were identified. Segmental and subsegmental perfusion defects occurred in 23 (82%) dogs and were classified as severe in 9 (32%) dogs, moderate in 11, and mild in 3. There was no particular lobe predilection. Patchy mulberry-appearing defects, indicative of fat embolism, were most common. Embolemia was observed by ultrasound in 10 dogs. Variable-sized particles occurred in 8 dogs, particles and bubbles in 2 dogs, and no emboli were observed in 2 dogs. Embolemia was observed within 10 seconds after femoral stem insertion and lasted < 1 minute. Pneumoemboli remained in the right atrium for > 8 minutes before dislodgement. CONCLUSIONS: Embolemia of either air, particles, or both occurs in most dogs during THR surgery. CLINICAL RELEVANCE: Most dogs seemingly spontaneously recover from pulmonary embolism that occurs during THR. The risk of clinical complications from this pulmonary embolism should be taken seriously, even though the exact morbidity and mortality rates are unknown.  相似文献

The objective of this study was to evaluate the effect of forage concentration on sorting, nutrient intake and feeding behaviour of growing lambs. Twelve weaned lambs were exposed, in a crossover design with 7‐day periods, to each of two treatment diets: (i) lower‐concentrate diet (LC; 40.0% concentrate) and (ii) a higher‐concentrate diet (HC; 60.0% concentrate). Alfalfa hay was used as forage source. Sorting was determined by subjecting fresh feed and ort samples to particle separation and expressing the actual intake of each particle fraction as a percentage of the predicted intake of that fraction. Lambs sorted against long particles (>19 mm) on both treatments. On the LC diet, lambs sorted for medium particles, whereas animals fed the HC diet did not sort for or against medium particles. Lambs sorted for fine particles (<1.18 mm) on both treatments (p < .05). The extent of sorting for the fine particles was greater on the LC diet compared with the HC diet (p < .05). Dry matter intake was increased by increasing the concentrate content of the diet. Intake per visit and eating rate increased, and total chewing time was decreased in lambs fed HC diet. In conclusion, lambs sorted most against the longest particles and for the fine particles. Furthermore, feed sorting behaviour is affected by the forage level, and lambs sorted more for the fine particles in the LC diet.  相似文献

Radiolabeled gold colloid (198Au), which has been used to assess particle clearance in mammalian species, was used to assess blood-borne particle clearance in turkeys. When turkeys 16 weeks of age were injected intravenously with these particles, there was greater than a 98% decrease in blood gamma emission from 1 minute to 6 minutes postinjection. Uptake of particles was predominantly hepatic with minor uptake by the spleen and bone marrow. Negligible uptake was observed in lung, kidney, and skeletal muscle. Autoradiography demonstrated particles within Kupffer cells of the liver, periarteriolar macrophages of the spleen, and bone-marrow macrophages. Particles could not be demonstrated within the lung or kidney. The mononuclear phagocyte system responsible for blood particle clearance in turkeys is therefore located predominantly within the liver, spleen, and bone marrow and is similar to that of rats, mice, rabbits, and dogs. Pulmonary intravascular macrophages, which have recently been described in ruminants and pigs, are not apparent in this species.  相似文献
