Immune competence, resistance to Escherichia coli and growth were measured in female chicks of broiler male parent lines from four different commercial sources. These chicks were fed with three levels of dietary crude protein (CP) from day-old. The protein contents in the diets were 18%, 20.5% and 23%; these diets are referred to as the low-, medium- and high-protein diets, respectively. There was a significant genotype by dietary protein interaction for body weight at 35 days of age but not at 14 or 28 days of age. At 14 days of age, the chicks fed on the high-protein diet weighed significantly more than those fed on the low-protein diet, but there were no differences between the chicks fed on the medium- and low-protein diets. The influence of CP content on body weight had disappeared by 28 days of age. There were significant differences between the genotypes in antibody production in response to sheep red blood cell (SRBC) inoculation, but no such differences were observed between the chicks fed the different levels of dietary protein. Chicks fed on the high-protein diet had lower lesion scores following E. coli inoculation than those fed on the low-protein diet. There were also significant differences in lesion scores among the genotypes. Genotypes with heavier body weights had significantly higher lesion scores and lower antibody titres than those with less body weight. Also, genotypes of lower body weight had a greater cutaneous basophilic hypersensitivity response to phytohaemaglutinin-P inoculation, and a better humoral response against SRBC and a lower heterophil to lymphocyte ratio.  相似文献

1. Hens from White Leghorn lines selected for high (HA) or low (LA) antibody response to sheep red blood cells (SRBC) were inoculated with 0.1 ml of either 0.25% or 2.50% SRBC suspension. Eggs laid over the next 15 d were grouped into 5, 3‐d collection periods and incubated. Maternal antibody titres were determined in chicks at hatch and 7 d after hatch.

2. In a subsequent experiment, hens of the 2 lines were inoculated with 0.1 ml of 2.50% suspension of SRBC, and eggs laid on days 10 to 13 after inoculation were incubated. Maternal antibody titres were determined in 15 and 18‐d embryos as well as in chicks at 0, 5, 10, 15, 20, and 25 d after hatch.

3. Dosage of SRBC had no effect on the antibody titres in line HA; however, the higher dosage elicited greater antibody titres than the lower dosage in line LA.

4. Maternal antibodies were detected earlier in chicks of line HA (eggs laid on days 7 to 9) than those of line LA (eggs laid on days 10 to 12) regardless of dosage administered to the hens.

5. In both lines, antibodies specific to SRBC were observed on day 15 of incubation, with the frequency of responders greatest at hatch. The high frequency of HA responders was maintained for 15 d after hatch, whereas there was an immediate decline with age in LA responders.

6. It was concluded that lines HA and LA have diverged in the pattern of maternal antibody levels as a result of the divergent selection for antibody response to SRBC.  相似文献

选用360只60~70周龄海兰褐蛋鸡,研究不同种类的多不饱和脂肪酸(PUFA)在不同添加水平下对产蛋鸡免疫功能的影响。试验共设10个处理,其中n-3PUFA的来源分别为亚麻籽油和鱼油,在日粮中的添加水平分别为20、40、60和10、30、50g/kg。n-6PUFA的来源为玉米油,在日粮中的添加量为20、40和60g/kg,另设一处理为不添加脂肪酸的对照组。在试验开始后的第5周和第7周分别给每个处理组中的6只鸡注射牛血清白蛋白抗原(BSA)和绵羊红细胞(SRBC),并分别于注射后第6,10,14天和第5,9,13天翅静脉采血,测定BSA抗体效价和SRBC抗体滴度。在试验开始后的第5周和第10周,采血测定外周全血淋巴细胞增殖速度,同时每个处理杀3只鸡,取脾脏制备单核细胞,测定其增殖速度。试验结果表明,日粮中添加PUFA具有提高产蛋鸡一免及二免BSA抗体和SRBC抗体产量的趋势,并呈现剂量依赖性。其中高鱼油添加组在一免10天时能显著(P<0.05)提高SRBC抗体滴度,二免后高鱼油添加组和高亚麻籽油添加组都能够显著(P<0.10)提高SRBC抗体滴度;对于BSA抗体效价,高n-3PUFA添加组都能够显著(P<0.10)提高。产蛋鸡外周全血淋巴细胞和脾脏单核细胞增殖速度在饲喂添加高n-3PUFA日粮后受到明显抑制。鱼油来源的n-3PUFA对产蛋鸡免疫功能影响程度比亚麻籽油的大。  相似文献

The immune responsiveness to infectious bursal disease virus (IBDV) in four native and crossbred chicken lines was compared. ELISA IBDV antibody titers in hen serum samples, yolk from matched eggs and sera from matched 1-day-old chicks from each chicken line with an identical vaccination program were measured, and plotted. There was considerable variation between lines in the measured IBDV specific antibodies, in vaccinated parent hens and in the amounts of inherited maternally derived antibodies in both yolk and progeny chicks. Differences in ratios of the inherited antibody level from hen to 1-day-old chicks were also found among different chicken lines. Breed differences in regressions of IBDV antibody levels in yolk to that of hen or progeny chicks' sera were also found, so prediction of serum titer of hen and/or progeny chicks from yolk are varied among chicken lines.  相似文献

SUMMARY: Two lines of White Leghorn chickens selected divergently for high (HH) or low (LL) antibody response 5 days after an injection with 0.1 ml of 0.25% suspension of sheep red blood cell (SRBC) antigen were used to produce parental, reciprocal F(1) , F(2) and backcross progeny. At 36 days of age males and females of the various progeny types were injected with SRBC suspension and antibody titres measured at 5 and 12 days later. Progeny of the high antibody line had higher titres at both 5 and 12 days after inoculation with SRBC than those of the low line. Reciprocal effects for SRBC titres were important only for female progeny suggesting sex-linked effects of the Z chromosome. Titres for F(1) progeny were intermediate and different from the parental lines at both 5 and 12 days after inoculation. Antibody titres 5 days after inoculation exhibited heterosis which emanated from the homogametic sex. Although maternal effects generally had no influence on antibody titres, maternal heterosis in the selected trait was due to sex-linkage. Recombination effects were negligible for both traits. In Alter von 36 Tagen wurde den Tieren eine SRBC Suspension injiziert und Antik?rpertiter 5 und 12 Tage sp?ter bestimmt. Nachkommen der HH Linie hatten an beiden Tagen h?here Titer als LL Nachkommen. Reziproke Effekte für SRBC Titer waren nur bei weiblichen Nachkommen wichtig, bedingt wohl durch geschlechtsgekoppelte Z-Chromosom Einflüsse. Die Titer der F(1) Nachkommen waren zwischen denen der Elternlinien. Antik?rpertiter 5 Tage nach Injektion zeigten Heterosis, die im homogametischen Geschlecht auftrat. Obwohl im allgemeinen maternale Wirkungen keinen Einflu? zeigten, war maternale Heterosis beim selektierten Merkmal auf Geschlechtskopplung zurückzuführen. Our group has established two lines of meat-type chickens divergently selected for early (HC line) and late (LC line) antibody responsiveness at 10 days of age to immunization with inactivated pathogenic Escherichia coli bacteria. The question addressed in the study presented here is whether this selection has changed other immunological responses, increasing the overall 'early' immunocompetence. Broilers of the third and fourth generations (S3 and S4) of the selected lines (HC and LC) and a control, unselected line (CT) were vaccinated at 10 days of age with E. coli vaccine, Newcastle virus vaccine (NDV), sheep erythrocytes (SRBC) or bovine serum albumin (BSA). Line-HC chicks exhibited higher antibody titers to E. coli, NDV and SRBC than CT or LC chicks. At 20 days of age HC chicks demonstrated a higher total protein and a higher beta- and gamma-globulin levels in their serum. At 21 days of age, HC chicks cleared carbon particles faster than LC chicks. Peripheral blood lymphocytes (PBL) from HC chicks vaccinated with E. coli vaccine, proliferated in vitro more actively in the presence of the stimulating antigen than the PBL of LC chicks. Peripheral blood lymphocytes (PBL) obtained from HC-line chicks exhibited a higher proliferative response to concanavalin A (Con A)-, phytohemagglutinin (PHA)- or pokeweed mitogen (PWM)-stimulation than LC PBL. These results demonstrate that the selection for high or low antibody response to E. coli at a young age resulted also in a significant change in the response of other parameters of the immune system. The high response to E. coli was found to be associated with a high antibody response to other antigens (NDV and SRBC), increased phagocytic activity and increased proliferative response to antigen or mitogens. Two experiments were conducted in which the effect of carrageenan (CGN) on humoral immune response of chicks selected on either high (H) or low (L) antibody production to sheep red blood cells (SRBC) was determined. H and L line chicks were injected i.p. with different doses of CGN prior to immunization with SRBC or Brucella abortus (BA). Four weeks later chicks were reimmunized with the same antigens. In general, control H and L chicks had significantly higher total anti-SRBC titers than CGN-treated chicks in primary response. Also, total anti-BA titers were significantly higher in control chicks than CGN-treated chicks on days 3 and 5 following primary immunization and on days 0 and 7 of the secondary response. Overall, the 2-mercaptoethanol (2ME)-resistant anti-SRBC titers did not differ significantly among the CGN-treated groups. However, control chicks tended to have higher anti-BA 2ME-resistant titers from day 14 p.i. of primary response on. Regardless of antigen or CGN treatment, the H line chicks had significantly higher titers than L line chicks. However, the CGN treatments did not affect the antibody response to BA nor to SRBC differently between L and H line chicks. It would appear that since CGN is cytotoxic for macrophages, selection for antibody production did not result in different abilities of the macrophages of these chicken lines to respond to an antigenic challenge.  相似文献

Twelve large white turkey hens were immunized with a commercially available Bordetella avium bacterin. Hens and eggs were tested using an enzyme-linked immunosorbent assay (ELISA) to determine the response to the bacterin. Three hundred poults were then obtained from two commercial flocks, the hens of one flock having been immunized with the same bacterin used on the group of 12 turkeys. Titers of the poults were monitored for 7 weeks, and poults were challenged by exposure to infected poults at 1, 7, 14, and 21 days post-hatch. Hens produced an antibody response following immunization, with a parallel antibody response being detected in eggs. Maternal antibodies were present in poults from immunized hens. Poult titers declined to near the level of poults from unimmunized hens by 14 days of age. Poults from immunized hens challenged at 1 and 7 days were resistant to development of clinical disease and gross lesions, whereas all poults from unimmunized hens exhibited clinical signs and gross lesions. After 14 days, the resistance of both groups to development of clinical disease, became near equal, neither group being affected as severely as the unimmunized hens challenged at days 1 and 7. Six commercial turkey breeding flocks and their progeny that had not been vaccinated for B. avium and had no history of B. avium infection were evaluated with the B. avium ELISA. There were variations between the flocks, with poult titers reflecting those found in the hens.  相似文献

To study the relation between serum complement levels and the chicken MHC (B) complex, complement haemolytic activity was measured in sera from hens from seven pure-bred B-typed White and one Brown Leghorn lines, and three ISA-Warren lines that had been divergently selected for antibody responses to sheep red blood cells (SRBC). Significant differences occurred in the serum haemolytic complement activities, both belonging to the classic (CPW) and the alternative (APW) pathways, among the 11 different haplotyped chicken lines. Hens with high CPW and high APW titres predominantly displayed the B2 or B21 haplotypes. Chickens with low CPW and APW were found in B14 and B15 haplotypes. Haplotype B14 appears to be different in complement levels when present into the pure-bred lines or into the ISA-Warren line selected for low antibody responses to SRBC. Otherwise, the presence of B21 in ISA-Warren line selected for high antibody responses to SRBC does not differ with the B21 in the inbred lines (except in the NL-line for CPW values). In general the haplotypes B2 and B21 are found in chicken lines with enhanced disease resistance, and the B15 haplotype has been connected with enhanced disease susceptibility. Our results suggest that levels of haemolytic complement activity, either from the classical or from the alternative pathways, may underlie part of the immunocompetence ascribed to the MHC (B) complex in chickens.  相似文献

1. Growth, antibody response to sheep red blood cells (SRBC) and resistance to Escherichia coli were measured in broiler female chicks received from 4 (n = 100 in each) commercial genotypes (A, B, C and D) and fed with maize-soybean-deoiled rice bran based diets containing 4 concentrations of methionine (3.91, 4.46, 5.00 and 5.54 g/kg). The diets were fed ad libitum from 1 to 49 d of age. 2. Body weight gain and weight gain/food intake at 2 week intervals, response of broilers to inoculation of 0.5 ml of SRBC (0.5 or 2.5%), 0.1 ml of E. coli (10(-4) dilutions) culture, and 100 microg phytohaemogglutinin-P (PHA-P) at 43 d of age were measured. The responses to SRBC and E. coli inoculation were recorded at 5 d post inoculation (PI), while the responses to PHA-P were recorded at 12 and 24 h PI. 3. Genotype by methionine interaction was not significant for body weight gain, but significant differences in weight gain were observed among different genotypes. Variation in methionine concentration did not influence body weight gain or weight gain/food intake at 1 to 14, or 42 d of age. At 28 d of age, chicks fed on the 3.91 g methionine/kg diet weighted significantly less than those on the other methionine concentrations. Genotype by methionine interaction was observed for food efficiency at 0 to 28 d of age but not at other ages. 4. Antibody titres against SRBC and heart and air sac lesion score to E. coli challenge were not influenced by genotype-methionine interaction. Chicks given higher concentrations of methionine had higher antibody titres and greater cutaneous basophilic hypersensitivity (CBH) response than those given low levels of methionine. Also, variation was observed in expression of CBH response to PHA-P among different genotypes. 5. It may be concluded that, although the commercial broiler chicks do not require more than 3.91 g methionine/kg for optimum growth and food efficiency, the immunity in terms of CBH response and antibody production to SRBC increased with the concentration of methionine in the diet in the majority of genotypes, indicating a higher methionine requirement for immunity than for weight gain.  相似文献

Two experiments were conducted to study the difference in humoral immune responses between lines of chickens selected for high (H) or low (L) antibody production to sheep red blood cells (SRBC). Prior to i.v. immunization with SRBC or Brucella abortus (BA), chicks of both lines were injected with either 2, 3 or 4 ml carbon suspension (59 mg carbon/ml) per kg body weight; controls were not injected. In both the H and L line, a higher dose of carbon showed a more progressive depression of the total antibody titer to SRBC during the initial stage of the primary response. The 2ME-resistant antibody titers to SRBC showed the same tendency during the latter phase of the primary response. However, chicks treated with 3 ml carbon had lower 2ME-resistant antibody titers than any other group. Following i.m. reimmunization with SRBC, the previous treatment with carbon doses enhanced total antibody titers throughout the secondary response, when compared to the controls. The 3 ml carbon-treated chicks had the highest total anti-SRBC titers in the secondary response. The secondary 2ME-resistant anti-SRBC titers were not affected by the carbon doses. Carbon treatment did not affect the antibody titers to BA. No differences between the H and L line were found in the effects of carbon on the humoral immune responses to SRBC or BA.  相似文献

The antibody response of laying hens to experimental Salmonella enteritidis infection was evaluated in microagglutination, tube agglutination, and rapid whole-blood plate agglutination assays. Hens of three different ages were infected by either oral inoculation or horizontal contact transmission. Blood was collected at weekly intervals, and the presence of specific antibodies was assessed by reaction with antigens prepared from strains of S. enteritidis and S. pullorum. The sensitivity of detection of infected hens did not vary significantly between the assays, as all three tests effectively identified most exposed hens as seropositive. Within each test, however, variation was observed in the detection sensitivity when different antigens were used. The microagglutination titers of serum samples were determined by serial dilution. Antibody titers peaked at 1 to 2 weeks postinoculation and declined steadily, although most birds were still identified as seropositive at 10 weeks postinoculation. The mean microtest titers obtained with S. enteritidis antigens were higher than with an S. pullorum antigen, indicating greater test sensitivity. However, use of the S. pullorum antigen resulted in fewer false positives when sera from uninfected control hens were tested. The titers of contact-exposed hens peaked later and at lower values than did those of inoculated hens, but these two groups of hens had similar antibody titers after the third week postinoculation.  相似文献

Phosphorus supplementation from poor quality feed-grade sources may introduce excessive levels of vanadium (V) into the diets of laying hens. Vanadium in the diet of chickens has been shown to be detrimental to egg production, albumen height, BW, and feed consumption. This study showed that dietary V also has a negative effect on the color of brown eggshells from commercial-type layers. Commercial-type brown egg layers were fed a cornsoybean meal basal diet supplemented with 0, 50, or 100 ppm of V as NH4 VO3 to determine the effect on shell pigmentation. Hens fed V at both dietary concentrations laid lighter colored eggs (less redness) after only 2 d. Another experimental group of the same type of hens that were fed lower V concentrations (0, 15, or 30 ppm) also had less shell pigmentation. After the detrimental “bleaching” effect on shell color was observed, these diets were supplemented with 1 of the following: no supplement, 100 ppm of vitamin C, 100 IU of vitamin E, or 100 ppm of β-carotene. Only vitamin C restored the eggshell color of eggs from hens fed both levels of V. When these same antioxidants were added as preventive agents to diets supplemented with 15 ppm of V before the effects of V were established, brown layers benefited again from 100 ppm of vitamin C but not from vitamin E or β-carotene.  相似文献

肉鸡日粮中分别添加7%的猪油、玉米油、亚麻子油和鱼油,检测血清抗SRBC抗体滴度和伴刀豆球蛋白A(conA)、商陆分裂素(PWM)对脾脏淋巴细胞的增殖反应。结果表明,四种脂肪对抗体生成有明显促进作用,其中以鱼油作用更显著;四种脂肪亦可明显促进脾脏淋巴细胞增殖,但脂肪种类对促进淋巴细胞增殖作用差异不大;鱼油可促进肉鸡生长,其余三种脂肪对肉鸡体重影响不大。  相似文献

A study was carried out to investigate the replacement effect of vitamin E with grape polyphenols (GPP) on antioxidant status, immune response, and organs histopathology in broilers from 1- to 35-d age. One-day-old Hubbard broiler chicks (n = 280) were randomly divided into 28 study units of 10 chicks each. A starter control diet was formulated having 22.5% CP, 3,200 kcal/kg ME, and 100 ppm vitamin E50 [50 IU DL-α-tocopheryl acetate (CON)]. Other dietary treatments were obtained by replacing vitamin E on wt:wt ratio, with 25 [low GPP (LGPP)], 50 [medium GPP (MGPP)], and 75 ppm [high GPP (HGPP)] GPP. Likewise, a control finisher diet (20% CP and 3,200 kcal/kg ME) and LGPP, MGPP, and HGPP diets were formulated having the same proportions of GPP and vitamin E as in the respective starter diets. Each dietary treatment was randomly assigned to 7 replicate pens. As a result of this study, by increasing GPP level in place of vitamin E, total phenolic contents in breast and leg muscles increased linearly (P< 0.05) whereas thiobarbituric acid reactive substances decreased quadratically with MGPP showing the lowest value (P< 0.05). Antibody titers against Newcastle disease virus were higher (P< 0.05) in broilers fed the LGPP and HGPP diets than in those fed the control diet. Antibody titers against infectious bursal disease virus were higher (P< 0.05) in broilers fed the HGPP diet than in all of the remaining dietary treatment groups. Histopathological examination of kidney and liver tissues showed no changes in birds fed LGPP and MGPP diets; however, mild effects were observed in some birds fed HGPP diet. Replacement of vitamin E with GPP resulted in lower cost per kg broiler feed. In conclusion, replacement of 75 ppm vitamin E with GPP improved antioxidant status and immune response of broilers at lower feed cost without any significant effect on histopathology of organs.  相似文献

试验用ArborAcre肉仔鸡672只,随机分成7个处理,每处理96只,雌雄各半。2×3因子设计,其中维生素E2个水平分别为20、80mg/kg;抗坏血酸3个水平依次为200、400和800mg/kg。另外设一个不添加维生素E和抗坏血酸的对照组。试验期分为0~2周龄和3~4周龄两个阶段。试验结果表明,提高日粮维生素E水平(80mg/kg)对肉仔鸡体内抗坏血酸的合成有促进作用,抗坏血酸对维生素E的作用不明显。随着日龄的增加,肉仔鸡血清维生素E的含量呈下降趋势,日粮尽早添加维生素E可以减缓血清维生素E的下降;肉仔鸡体内可以合成一定量的抗坏血酸,合成能力随日龄的增加而增强。日粮高水平的维生素E(80mg/kg)可提高28日龄肉仔鸡血液淋巴细胞转化率和血清新城疫抗体滴度,抗坏血酸对肉仔鸡细胞及体液免疫功能影响不显著。  相似文献

Broiler chickens infected at 3 weeks of age with infectious bursal disease virus (IBDV) were given Brucella abortus (BA) or sheep red blood cell (SRBC) antigens before, during, and after the acute phase of the infection. Gland of Harder (GH) extracts and serum samples were used to assay local and systemic antibody titer to each antigen 7 days after antigen was administered. Antibody titers to both BA and SRBC antigens were lower (P less than 0.05) in GH extracts and serum of IBDV-infected broilers than uninfected controls. The responses to BA, a thymus-independent antigen, took longer to become depressed than the responses to SRBC, a thymus-dependent antigen. The depression of antibody titers following IBDV inoculation suggests compromise of both local and systemic immune function, a finding of importance to the broiler industry.  相似文献

The objective of this study was to evaluate the effects of different molting methods on postmolt production, plasma corticosterone levels, and antibody production to SRBC for the welfare of laying hens. This experiment was conducted with 120 IGH-type Brown laying hens (70 wk of age), randomly divided into 3 experimental groups. The hens in one group were fed a whole-grain barley diet during the first 10 d (WB diet). On d 11, hens consumed 100 g of layer diet/d until d 28. In the second group, hens were fed a Zn diet containing 10,000 mg/kg of Zn as ZnO for 10 d (Zn diet). Hens were then provided 100 g of a layer diet from d 11 to 28. In the third group, feed was withdrawn for 10 d, and on d 11 hens were fed a cracked corn diet ad libitum until d 28 (California method; CAL diet). Hens in all groups were returned to the layer diet ad libitum on d 29. Egg weight was lower in the Zn treatment than in the other treatments. Feed intake and plasma corticosterone levels were higher and antibody production was lower in the CAL treatment than in the WB and Zn groups. The FCR was better in the WB than in the Zn group. Mortality, egg production, and egg quality were not significantly different among the molting methods. As a result, the WB molting program was the best method for postmolt production among the programs examined.  相似文献

Antibody response was rapid and high in broiler breeder chickens receiving 1 or 2 vaccinations with oil-emulsion vaccine against Newcastle disease at 23 or at 23 and 26 weeks old. The antibody titers remained high during the 41-week experimental period. At 64 weeks old, about 41 weeks after vaccination, the geometric mean hemagglutination-inhibition antibody titer was 67 from the single vaccination, and 103 from the double vaccination. The immune response to live-virus vaccine given at 2, 9, 20, 30, 42, or 54 weeks of age via the drinking water was high, but uniformity was lacking in the antibody response in the breeders and maternal antibody response in the progeny. Maternal antibody levels in one-day-old chicks were related to the titers of antibody in the dams.
