In melon (Cucumis melo L.), the amount of fertile pollen decreases at low temperature during winter and early spring and thereby restricts fertilization. A promising way to overcome this problem is to develop parthenocarpic cultivars. Parthenocarpic cucumber and squash, but not melon cultivars are available. In this study, we explored 172 accessions from an East Asian melon collection and identified accessions that exhibited strong parthenocarpic ability throughout the year. Crosses between parthenocarpic accessions and a non-parthenocarpic cultivar, and among parthenocarpic accessions indicated that parthenocarpy may be inherited in a recessive manner and is likely controlled by the same gene or genes in these accessions. The parthenocarpic indices, such as the size and number of parthenocarpic fruits, differed among the cultivation periods, indicating the importance of environmental factors for parthenocarpic fruit development. We conclude that it is possible to breed new cultivars with stable parthenocarpic ability throughout the year by using the identified accessions.  相似文献

Among 371 oat accessions from the world collection of the Vavilov Institute of Plant Genetic Resources (VIR) mostly represented by the landraces from Asia (Maritime Territory of Russian Federation, Mongolia, China, Japan, North Korea, India, Iran, Afghanistan, and Bhutan) ninety five were heterogeneous for resistance to the North Caucasus populations of the greenbug (Schizaphis graminum Rondani). Damage ratings of resistance in 47 accessions were high, low level of resistance was identified in 48 genotypes. Higher frequency of greenbug resistant accessions (60.5%) was found among landraces from Mongolia. Lines with high expression of the resistance were selected from seven landraces with wide range of damage ratings. North Caucasus populations of S. graminum from Krasnodar area and Dagestan significantly differ by the frequencies of virulence to host plant. Seven virulence phenotypes differentially interacting with oat genotypes were found in the Krasnodar population and 10 phenotypes were revealed in the Dagestan population. Results of the experiments with the greenbug test clones revealed that all the lines selected from the accessions VIR-2490, VIR-2539, VIR-4074, VIR-12213, VIR-12214 (Mongolia), VIR-6688 (India) and VIR-13624 (North Korea) are protected by diverse alleles of resistance genes which differ from the earlier identified gene Grb3. These lines are supposed to have aphid resistance genes nonidentical to the Grb1 and Grb2. The accessions VIR-6688, VIR-4074 and VIR-12214 possess high resistance to Krasnodar and Dagestan greenbug populations and the accession VIR-13624 is protected by the most effective gene (genes) of resistance to the both populations from North Caucasus.  相似文献

Forty-four Asparagus officinalis cultivars, gene bank accessions and breeding lines as well as thirty-four accessions of wild relatives of Asparagus were evaluated for resistance to Asparagus virus 1. Three different test strategies were developed for the assessment of individual plants: (1) natural infection under field conditions, or two vector-mediated infection assays using the green peach aphid Myzus persicae (2) in an insect-proof gauze cage or (3) in a climate chamber. The AV-1 infections were verified by DAS-ELISA and RT-PCR approaches. All tested 660 individual plants of A. officinalis germplasm were susceptible to AV-1 infection. In contrast, in 276 plants of 29 Asparagus wild accessions no virus infection could be detected. These resistant accessions comprised of nineteen diploid, tetraploid and hexaploid species of both the Eurasian clade and the African clade of the asparagus germplasm. Data of the AV-1 resistance evaluation are discussed in relation to the genetic distance of the resistance carrier and potential application in breeding.  相似文献

The morphological diversity and relationship among 138 pear genotypes, including 11 commercial cultivars and 127 wild pear accessions from central Zagros mountains located in the west of Iran, based on 64 morphological characters were studied. The higher coefficient of variation indicated that high level of variation was belonged to the petiole stipule, fruit the relative area of the skin color, fruit relative area periphery rust dents end, fruit relative blight on an area of skin, fruit curved tail, fruit dents end, fruit depth dents end, fruit relative area periphery patina tail connection characters. Pyrus communis L. and Pyrus glabra Boiss. species were the highest and the shortest tree, respectively. According to the cluster analysis, 138 genotypes were clustered into nine main groups. Principal components analysis revealed that the first three components accounted for 68.77 % of the total variation in data. Moreover, 19 factors justified 72.57 % of the total variation based on factor analysis. The research results showed that the Zagros mountains are the centers of diversity of pear. Therefore, it is necessary to use the Iranian genotypes as the genetic resources for improving the morphological characters of pear.  相似文献

Tibet is one of the main distribution centers of wild Fragaria species in China. There are abundant, precious and rare wild strawberry resources. This paper focused on the classification, distribution and evaluation of the 65 wild strawberry accessions collected from 17 counties in Tibet. The main taxonomical characters, such as stolon branching, leaflet type, flower sex, fruit shape, status of achene and sepal, and chromosome number were observed for each accession. All accessions were identified and classified into seven Fragaria species and two forms, including F. daltoniana J. Gay (2x), F. nilgerrensis Schlecht. (2x), F. nubicola Lindl. (2x), F. pentaphylla Losinsk. (2x), F. gracilis A. Los. (4x), F. moupinensis (French.) Card. (4x) and F. tibetica Staudt et Dickoré. (4x). The two white-fruited forms were F. nubicola f. alba (2x) and F. moupinensis f. alba (4x). The analytic hierarchy process (AHP) method was used to perform a comprehensive assessment on the value of seven Fragaria species and two forms. The results showed that F. nilgerrensis had better fruit quality, disease resistance and waterlogging tolerance than any other species in Tibet. The collection and identification of wild strawberry resources in Tibet would be helpful to improve the fruit quality and stress resistance of modern strawberry cultivars.  相似文献

Genetic diversity of 139 accessions of diploid Triticum species including Triticum urartu, Triticum boeoticum and Triticum monococcum was studied using 11 SSR (simple sequence repeats) markers. A total of 111 alleles with an average of 10 alleles per locus were detected. The polymorphism information content (PIC) of each SSR marker ranged from 0.30 to 0.90 with an average value of 0.62. Among the three Triticum species T. urartu had the highest number of total alleles (Na?=?81), private alleles (Npa?=?15) and showed higher genetic diversity (Hex?=?0.58; PIC?=?0.54). The genotypes from Turkey exhibited the highest genetic diversity (PIC?=?0.6), while the least diversity was observed among 4 Georgian accessions (PIC?=?0.11). Cluster analysis was able to distinguish 139 wheat accessions at the species level. The highest genetic similarity (GS) was noted between T. boeticum and T. monococcum (GS?=?0.84), and the lowest between T. urartu and T. monococcum (GS?=?0.46). The grouping pattern of the PCoA analysis corresponded with cluster analysis. No significant differences were found in clustering of T. urartu and T. monococcum accessions with respect to their geographic regions, while within T. boeoticum species, accessions from Iran were somewhat associated with their geographical origin and clustered as a close and separate group. The results from our study demonstrated that SSR markers were good enough for further genetic diversity analysis in einkorn wheat species.  相似文献

Developing a molecular tool kit for hybrid breeding of Osmanthus species and related genera is an important step in creating a systematic breeding program for this species. To date, molecular resources have been aimed solely at Osmanthus fragrans with little work to develop markers for other species and cultivars. The objectives of this study were to (1) determine cross-transferability of O. fragrans and Chionanthus retusus derived SSRs in diverse Osmanthus taxa, (2) quantify the influence of locus-specific factors on cross-transferability, and (3) determine the genetic relationships between accessions. We tested 70 SSR markers derived from O. fragrans and C. retusus in 24 accessions of Osmanthus. Sixty-seven markers showed transfer to at least one other Osmanthus species with an overall transfer rate of 84% of loci across taxa. Genotyping with 42 microsatellite markers yielded a total of 367 loci. Number of alleles per locus ranged from 2 to 17 with a mean of 8.7 ± 4.8. Mean observed and expected heterozygosities were 0.560 ± 0.225 and 0.688 ± 0.230, respectively. Percent of polymorphic loci ranged from 40% in Osmanthus delavayi to 100% in O. fragrans. Osmanthus fragrans had the highest mean number of alleles per locus (4.2) while O. delavayi had the lowest (1.1). A reduced suite of eight-markers can distinguish between accessions with non-exclusion probabilities of identity from 3.91E?04 to 2.90E?07. The SSR markers described herein will be immediately useful to characterize germplasm, identify hybrids, and aid in understanding the level of genetic diversity and relationships within the cultivated germplasm.  相似文献

Here, two Punica species, viz., P. protopunica Balf. fil., reported as native to Socotra, and P. granatum L., were compared for the first time. Analysis of one P. protopunica and eleven P. granatum accessions was performed using three molecular markers, i.e., sequence related amplified polymorphism (SRAP), target region amplification polymorphism (TRAP), and intron targeted amplified polymorphism (ITAP), along with analysis of pgWD40 sequences, a gene involved in anthocyanin biosynthesis. All markers revealed the relationship between the two species and placed them at 33% similarity. SRAP, TRAP, and ITAP generated a total of 299, 260, and 160 bands, respectively. Of these, 78, 74, and 41 bands were specific for P. protopunica, and 92, 85, and 57 bands, respectively, were shared between both species. Sequence analysis of pgWD40~870 bp amplicons exhibited 100% identity among P. granatum accessions and 98% identity to that of P. protopunica. Phylogenetic analysis of WD40 sequences from monocot and dicot species, including both Punica species confirmed the relation between P. protopunica and P. granatum, supporting earlier reports that P. protopunica could be an ancestral species of P. granatum. Furthermore, the genetic diversity among and within P. granatum accessions from Egypt (3), Mexico (5), and Yemen (3) was assessed. Molecular marker-based relationships among region-bulked accessions was approximately the same (~90% similarity), whereas the degree of genetic variation was altered within each region. Specific bands (alleles) for accessions of each region along with those shared among them were identified. Thus, these bands could be used for pomegranate genotyping and breeding programs.  相似文献

DNA sequences of nuclear gene Got2 was studied in 60 accessions of Aegilops tauschii, 29 of subsp. tauschii and 31 of subsp. strangulata. It was found that Got2 allozyme polymorphism in Ae. tauschii is due to a single, unique, mutation which led to replacement of glutamic acid by isoleucine in residue 256 of the enzyme molecule, encoded by Got2. As revealed by Got2 DNA sequences variation, initially in its history Ae. tauschii was presented by subsp. strangulata, and among phylogenetic lineages of subsp. strangulata, the lineage “t-91s” (TauL3) is the most ancient, a relict one. Subspecies tauschii is relatively “young”. Initially it was presented by the lineage marked by combination of allozyme alleles Got2 105 and Acph1 100. In the past it inhabited the Continental area from Caucasia to Pakistan, but later on it was forced out by newly originated, now—a major lineage of subsp. tauschii, marked by Got2 100. This lineage extended the Continental area of the species up to Kirgizstan, but actually failed to penetrate into pre-Caspian area, occupied by subsp. strangulata. These results essentially differ from those obtained previously, using chloroplast DNA (cpDNA) sequences polymorphism. As revealed by cpDNA, the major, “usual”, subsp. strangulata (TauL2) is “younger” than subsp. tauschii, which resided on phylogenetic tree between relict lineage “t-91s”of subsp. strangulata—and major subsp. strangulata. But both cpDNA and Got2 DNA sequences indicate that the level of genetic variation in subsp. tauschii is much lower than in subsp. strangulata. According to Got2 DNA sequences variation, it was Ae. tauschii subsp. strangulata lineage “k-109″ which donated genome D to Triticum aestivum L. This lineage includes accessions: k-109 from South-Eastern Precaspian Azerbaijan; KU-2105, KU-2159 from Western Precaspian Iran; KU-2080 from Eastern Precaspian Iran.  相似文献

Germplasm collections of C. reticulatum and C. echinospermum are limited, while both species face threats from over-grazing and habitat change in their natural environments. Recently many new accessions of C. reticulatum and C. echinospermum were collected in east and south-east Anatolia (Turkey) but they have not yet been evaluated for agro-morphological traits. Therefore, the current study investigated agro-morphological traits of new germplasm sources of C. reticulatum and C. echinospermum and evaluated resistance to biotic and abiotic stresses for chickpea improvement. The most attractive agro-morphological traits were canopy width, number of stems and pods per plant and biological yield. The most productive accessions of C. reticulatum and C. echinospermum had 712 and 625 pods per plant, respectively. Two distinct seed, flower and leaf shapes were found in accessions of C. echinospermum. Path analyses indicated that biological yield and harvest index had the most direct influence on seed yield in both species. Factor analyses showed that high seed yield in C. reticulatum depended on high biological yield and number of pods per plant, whereas high seed yield in C. echinospermum depended on harvest index. It was concluded that most accessions of C. reticulatum and C. echinospermum were not only resistant to some biotic and abiotic stresses but also had hidden alleles that could produce transgressive segregation in crosses to cultivated material.  相似文献

Citrus depressa Hayata is an indigenous mandarin species on the Ryukyu Islands located in the subtropical region of Japan. We deduced its phylogenetic relationships by evaluating accessions grown on various Ryukyu Islands via cleaved amplified polymorphic sequence analysis of cpDNA and sequence-related amplified polymorphism (SRAP). The cpDNA results indicated that C. depressa could be classified into two types. SRAP revealed patterns of diversity within C. depressa consistent with our cpDNA results. These results indicate that maternal origin may influence or is correlated with the constitution of the nuclear genome of C. depressa. Another Japanese mandarin species, Citrus tachibana (Makino) Tanaka was distinguished from C. depressa by SRAP markers. Moreover, both C. depressa and C. tachibana could be distinguished from other Citrus species. Our results suggest that Japanese mandarin possesses a characteristic genome with the genus Citrus.  相似文献

The wild species in general is considered to be the reservoir of genes especially for biotic and abiotic stresses. In okra, the predominant biotic stresses are yellow vein mosaic disease (YVMD), shoot and fruit borer and leaf hopper. Sixty eight (68) accessions belonging to four wild Abelmoschus species [Abelmoschus caillei (A. Chev.) Stevels, Abelmoschus manihot (L.) Medik., Abelmoschus moschatus (L.) Medik. and Abelmoschus tuberculatus Pal et Singh] and eight okra varieties were characterized and evaluated for phenological characters including biotic stresses under natural epiphytotic condition. The wild species examined consisted of 18 accessions (16 exotic and 2 indigenous) of A. caillei, 29 accessions of A. manihot, 16 accessions of A. moschatus and 5 accessions of A. tuberculatus. All the wild Abelmoschus species exhibited high diversity (as measured by Shannon Diversity Index) for 3 qualitative characters viz. intensity of stem colour, leaf shape, epicalyx shape, 13 quantitative characters and 3 biotic stress parameters. Among the wild species, A. caillei and A. tuberculatus showed maximum and minimum diversity for qualitative characters, respectively. There was significant variation for 19 out of 24 quantitative characters studied. Inter-species diversity pattern as estimated through Ward’s Minimum Variance Dendrogram and Principal Component Analysis revealed clear differentiation among the species with minimum overlapping indicating close association between geographical origins and clustering pattern. Intra-species diversity indicated role of specific adaptation in sub-clustering. Resistance to YVMD was found in accessions belonging to three wild species viz. A. caillei, A. manihot and A. moschatus while resistance to shoot and fruit borer and leaf hopper was found in accessions of all the four wild species. The resistant accessions can further be used for introgressing biotic stress resistance through pre-breeding into cultivated okra species.  相似文献

Molecular characterization of germplasm is important for sustainable exploitation of crops. DNA diversity was measured using inter-retrotransposon-amplified-polymorphism and inter-simple-sequence-repeat markers in 203 Ethiopian landraces and reference varieties of linseed (flax, Linum usitatissimum) and wild Linum species. Molecular diversity was high (PIC, 0.16; GD, 0.19) compared to other reports from the species. Genotyping separated reference from landrace accessions, and clustered landrace accessions from different altitudes and geographical regions. Collections showed evidence for recent introduction of varieties in some regions. The phylogeny supported L. bienne Mill. as the progenitor of domesticated L. usitatissimum. Markers developed here will be useful for genetic mapping and selection of breeding lines. The results show the range of characters that can be exploited in breeding lines appropriate for smallholder and commercial farmers in Ethiopia, producing a sustainable, secure, high-value crop meeting agricultural, economic and cultural needs.  相似文献

The parasitic weed, Striga gesnerioides, is a major threat to cowpea productivity throughout the savannas of West and Central Africa. The identification of sources of S. gesnerioides resistance and their incorporation into breeding programs would be a beneficial strategy to combat the devastation caused by the parasite in cowpea fields. In this study we examined one hundred and ninety-four (194) accessions, four commercial varieties and two controls collected from a mini core collection of cowpea held at the International Institute of Tropical Agriculture genebank for resistance to S. gesnerioides race 3 (SG3), the most prevalent race in Nigeria, using phenotypic screening and molecular marker analysis. Our studies identified two cowpea accessions, Tvu-1272 and Tvu-16514, that are resistant to S. gesnerioides SG3. Resistance in these lines is associated with the molecular marker SSR1, known to segregate with the gene conferring resistance to SG3 in the cultivar B301. Phenotypically, resistance in Tvu-1272 and Tvu-16514 is expressed as a hypersensitive response at the site of infection on the roots. Allelism tests indicated that the gene that conferring SG3 resistance in Tvu-1272 is independent of that conferring resistance in B301. Tvu-1272 and Tvu-16514 will provide additional new sources of resistance to Striga and races prevalent in Nigeria.  相似文献

This study aims to explore the potential of Dioscorea nummularia Lam. and to gather information as a prelude to its improvement by plant breeding. After a taxonomic clarification and a review of the botanical data, its geographical areas of origin and distribution are determined. A germplasm survey conducted on nine islands of Vanuatu enabled the collection of 110 accessions corresponding to 84 cultivars. These were grouped into: (1) wild forms under domestication, (2) annual types, (3) perennial-type cultivars of D. nummularia and (4) natural interspecific hybrids with D. alata L. Within each group, remarkable morphological variation exists for stem spininess, leaf shape, emerging leaf colour, tuber shape and tuber flesh colour. Selected accessions examined for ploidy had chromosome numbers ranging from 2n = 3x = 60 to 2n = 6x = 120. Chloroplast counts conducted on stomatal guard cells confirmed the variation in ploidy levels. Eight different male cultivars were studied for pollen fertility, but <5% of the pollen grains appeared to be fertile when stained with acetocarmine. However, profuse flowering and synchrony favour pollination and result in fruit set. All accessions are resistant to anthracnose. Viruses were detected but are morphologically not discernible, with no incidence on yield. Physicochemical analyses of tubers from 16 accessions revealed that D. nummularia and interspecific hybrids have high percentages of dry matter (mean of 33.11%) and starch (82.81%). The Pacific yam could be improved through conventional hybridisation or used for interspecific crosses with related economically important species. Recommendations are made for further research.  相似文献

Asian rice (Oryza sativa L.) is widely cultivated in Asia, where it has been classified into Indica and Japonica Group, the latter is further classified into Tropical and Temperate Japonica Subgroup. O. rufipogon is believed to be the closest ancestor to O. sativa, but it remains unclear whether the two groups arose from a single ancestor or different ancestors. Therefore, here, we investigated the matrilineal ancestors of O. sativa using markers for organelle (chloroplast and mitochondrial) genomes, and 119 O. sativa landraces, 10 O. glaberrima Steud., 115 O. rufipogon Griff. from Asia, and 39 accessions from other wild rice species with AA genomes. We screened 18 organelle markers developed based on polymorphic loci in the organelle genomes. In addition, we used the open reading frame 100 of a chloroplast marker. The results indicated that O. rufipogon first differentiated into two lineages and then further differentiated into Indica and Japonica Group, respectively. Accessions of O. rufipogon (R-1f and R-2d types) from Myanmar appear to be the closest ancestors of Tropical Japonica Subgroup and Indica Group, respectively. Therefore, these wild strains may have made a strong contribution to the domestication of rice landraces in Myanmar.  相似文献

We present here the first comprehensive genetic characterization of melon landraces from the humid tropics of southern India. The genetic diversity among 50 melon landraces collected from 3 agro-ecological regions of southern India (6 agro-ecological sub-regions) was assessed by variation at 17 SSR loci, morphological traits of plant habit and fruit, 2 yield-associated traits, pest and disease resistance, biochemical composition (ascorbic acid, carotenoids, titrable acidity) and mineral content (P, K, Fe, Zn). Differences among accessions were observed in plant and fruit traits. Melon germplasm with high titrable acidity, higher than average amounts of mineral content and resistance to Cucumber mosaic virus, Zucchini yellow mosaic virus, powdery mildew (races 1, 2, 3, 5), Fusarium wilt (races 1, 2), Aphis gossypii and leafminer was recorded in the collection. A high level of genetic variability in melon germplasm was suggested by the SSR analysis. Comparative analysis using SSRs of the genetic variability between Indian melons from north, south, and east regions and reference accessions of melon from Spain, France, Japan, Korea, Iraq, Zambia showed regional differentiation between Indian melon accessions and that Indian germplasm was weakly related to the melon accessions from other parts of the world, suggesting that an important portion of the genetic variability found within this melon collection has not been used yet for the development of new cultivars. Additional collections of acidulus melon germplasm should be made in southern India and adequate management of this important genetic resource is clearly a necessity.  相似文献
