The effects of heavy metals (Hg2+, Cd2+, Co2+, Cr3+, Pb2+, Cu2+, Fe2+. Ni2+, Zn2+, Mn7+, Mn4+ and Mn2+) on the relative hatching percentage of brine shrimp, Artemia salina were assessed. Artemia salina cysts (San Francisco Bay Brand) hatched in seawater containing various heavy metal concentrations. After 48 hours, the number of hatched nauplii were counted. A negative linear relationship between hatching rate of Ariemia salina cysts and heavy metal concentration occurred, except copper which conformed to a cubic model. The relative toxicities of heavy metals on Artemia salina cysts were as follows (Cu2+, most toxic; Mn2+, least toxic): Cu2+ > Zn2+ > Ni2+ > Co2+ > Mn7+ > Cr3+ > Cd2+ > Fe2+ > Hg2+ > Mn4+ > Pb2+ > Mn2+.  相似文献

The composition of sediment pore water was determined for ponds constructed on clayey Ultisols at Auburn, Alabama. Pore water was anaerobic and contained much higher concentrations of ferrous iron (Fe2+), soluble reactive phosphorus (SRP), total phosphorus (TP), total ammonia-nitrogen (TAN), and sulfide (S2−) than surface or bottom waters. Concentrations of SRP and TP in pore water were higher in ponds with high soil phosphorus concentrations than in a new pond with less soil phosphorus. Increased concentrations of organic matter in soil or larger inputs of feed to ponds favored greater microbial activity in soils and higher concentrations of TAN in pore water. The pH of pore water was 6.5–7.0, and pH was apparently controlled by the equilibrium:

Movement of Fe2+, SRP, and S2− from pore water into pond water apparently was prevented by the oxidized layer of soil just below the soil-water interface. Pond managers should concentrate on maintaining this oxidized layer to reduce the tendency for toxic substances to diffuse into the pond water.  相似文献

The aim of this study was to determine whether magnesium (Mg2+) in seawater is required for egg activation of the black tiger shrimp Penaeus monodon and whether manipulation of Mg2+ levels can be used to delay the process and thereby synchronize egg activation. Female P. monodon broodstock were allowed to spawn in artificial seawater containing Mg2+ at varying levels with respect to the normal (100%) level: 100%, 50%, 20% and 0%. Egg activation occurred normally at 100% Mg2+, incompletely at 50% and 20% Mg2+ levels and did not occur at all with 0% Mg2+. The fertilization rate with 100% Mg2+ was observed to be 83%, but fertilization failed to take place in all the other groups. The fertilization rate was restored from 0% to 76% following the 20% Mg2+ level treatment when Mg2+ levels returned to normal (100%) as soon as spawning was completed. This study suggests that the level of Mg2+ in seawater plays a vital role in P. monodon egg activation, and that commencement of this process could be delayed by manipulation of the Mg2+ level during and immediately after spawning.  相似文献

ABSTRACT:    This study incorporated the 43 kDa Zn-binding membrane protein isolated from common carp into liposome. The specificity and strength of the binding of 65Zn to the 43 kDa protein-liposomes, and the binding of the 65Zn-labeled 43 kDa protein-liposomes to laminin were studied. It was found that 65Zn was bound to the external side of the 43 kDa protein-liposomes. Specific binding of 65Zn to the protein-liposomes was detected. The binding parameter of Zn to the protein was found to be: maximum binding site (Nmax), 76.7 pmole/µg protein (approx. 3 mole of Zn2+/mole); and equilibrium dissociation constant (Kd), 0.19 µM. Of the cations introduced (Ca2+, Cd2+,Co2+, Cr2+, Cu2+, Fe2+, Hg2+, Mg2+, Mn2+, Ni2+, Pb2+), only Co2+ competed significantly with Zn. The protein-liposomes were also found to bind specifically to laminin with a Nmax of 1.1 pmole/µg laminin, and Kd of 4.79 µM. No significant protein-liposome binding occurred to other extracellular matrix proteins (fibronectin, fibrinogen or vitronectin). Furthermore, the binding was specifically inhibited by the Arg-Gly-Asp (RGD) peptide or GRGDSPG, while two other analogs (GRGESPG and GRADSPG) were without effect.  相似文献

This present study was designed to investigate the effects of stocking density and water exchange on the growth rate, survival and performance index of L. setiferus postlarvae under controlled laboratory conditions. The experiment was done with postlarvae (PL10 to PL40) at densities of 50, 150, 250 and 350 shrimp/m2 and various different water exchanges rate per day (0, 6, 12 and 18%). The maximum growth rate was obtained for shrimp with 12% water exchange per day at all densities. A reduction of the maximum growth rate was observed in relation to density with the highest values in shrimp stocked in a density of 50 and 150 shrimp/m2 (mean value of 0.53 mg/d) and the lowest in shrimp stocked in a density of 350 shrimp/m2 (0.24 mg/d). The multiple regression equation obtained to relate performance index (growth rate* survival : PI), shrimp density (X1) and water exchange (X2) was: PI = 0.31 + (0.001) X1+ 0.039 X2+ 2.28 × 10−6 X12+ (−0.0017) X22+ (0.000026)X1X2, R 2= 0.78; P > 0.03. According to this equation the optimum shrimp density-water exchange comhination was between 5 to 12% of water exchange at stocking density of between 50 and 150 shrimp/m2. Salinity, ammonia-N and nitrite-N increased according to the time spent in tanks without water exchange. With no (0%) water exchange, water quality parameters measured were outside the optimum for L. setiferus postlarvae. The use of optimum density and water exchange in a nursery system for L. setiferus with optimum variables established is proposed.  相似文献

This study investigated infection dynamics of infectious salmon anaemia virus (ISAV) by conducting two experiments to examine minimum infective dose and viral shedding of ISAV. In terms of minimum infective dose, the high variability between replicate tanks and the relatively slow spread of infection through the population at 1 × 101 TCID50 mL−1 indicated this dose is approaching the minimum infective dose for ISAV in seawater salmon populations. A novel qPCR assay incorporating an influenza virus control standard with each seawater sample was developed that enabled the quantity of ISAV shed from infected populations to be estimated in values equivalent to viral titres. Viral shedding was first detected at 7 days post-challenge (5.8 × 10−2 TCID50 mL−1 kg−1) and rose to levels above the minimum infective dose (4.2 × 101 TCID50 mL−1 kg−1) on day 11 post-challenge, 2 days before mortalities in ISAV inoculated fish started. These results clearly demonstrate that a large viral shedding event occurs before death. Viral titres peaked at 7.0 × 101 TCID50 mL−1 kg−1 15 days post-infection. These data provide important information relevant to the management of ISA.  相似文献

To determine the changes in blood chemistry associated with sequential transfer of summer flounder Paralichthys dentatus (320–480 g), 300 hatchery-reared fish were moved to three different environmental conditions during a 20-d period. Fish were transferred in progression from a recirculating seawater system (22 ppt, 22.5 C) to a flow-thru seawater system (31 ppt, 20.0 C), to three small coastal net pens (33 ppt, 15.5 C), and finally to a large open ocean net pen (33 ppt, 16.0 C). For this study, eight random fish were captured at each progressive step (environmental condition), anesthetized (MS222), and bled from the caudal vein (2 mL). Transferred flounder were bled every 12 h for 48 h to collect plasma cortisol and glucose samples. Fish were bled 24 h after transport and every 3 d thereafter for osmolarity, hematocrit, hemoglobin concentration, mean corpuscular hemoglobin content, glucose, cortisol, and the electrolytes Cl- Na+, K+ and Ca+. The most significant perturbations to blood chemistry (P < 0.05) occurred within 24 h of initial transfer from the recirculating to flow-thru seawater systems, suggesting an osmoregulatory rather than handling or transfer related stress. Osmolarity, electrolyte, and hematological parameters fluctuated and then recovered to stable levels by day 8 in the flow-thru seawater system. However, unlike the initial transfer, successive movement to the coastal and then the open ocean net pens produced transient increases in both plasma cortisol and glucose levels, suggesting a high level of stress associated with extended flounder handling and transfer.  相似文献

Abstract. A preliminary study was conducted to assess the performance of the tilapia, Oreochromis spilurus (Günther), cultured in seawater cages at different stocking densities during the nursing and rearing phases. The stocking densities tested were 200, 400 and 600 fish/m3 during the nursing phase and 100, 200, and 300 fish/m3 during the rearing phase.
In both growth phases, no significant differences (P>0.05) were observed among the different stocking densities on the mean individual final weight, daily growth rate, feed conversion ratio and survival rate. Yields in cages stocked with 400 and 600 fish/m3 during the nursing phase, however, were significantly (P<0.05) higher compared with cages stocked with 200 fish/m3. No significant differences (P>0.05) were observed between 400 and 600 fish/m3. A density of 600/m3 is therefore considered to be optimum for the nursing phase.
Yields of cages during the rearing phase increased significantly (P<0.01) with the increase in stocking density. After grading the fish, however, no significant differences (P>0.05) were observed when only fish bigger than 150g were considered. The occurrence of exophthalmia (cataract) was observed in two of the four replicates at the highest stocking density (300 fish/m3) during the rearing stage. Therefore, a stocking density of 200 fish/m3 is considered optimum for the rearing phase.  相似文献

ABSTRACT: The present study used squid gill as a source of transglutaminase (TGase) because it has extremely high TGase activity compared with other tissues. The enzyme was purified using successive chromatographies of Sephacryl S-300 and hydroxyapatite columns. The yield and purification-fold of the enzymatic activity was 12.6% and 14.1-fold, respectively. The molecular mass of the purified enzyme was estimated to be 94 kDa by using sodium dodecylsulfate–polyacrylamide gel electrophoresis analysis. Enzyme activity was enhanced 15-fold with an increase in NaCl concentration. Although the activity was dependent on Ca2+ concentration, it was not sufficiently activated even by 50 mM CaCl2 in the absence of NaCl, but could be fully activated with 10 mM CaCl2 in 0.7 M NaCl. However, in the absence of substrates, the enzyme was rapidly inactivated. The pH and temperature optima of the enzyme were approximately pH 8.0 and 20°C, respectively. It was stable in the absence of Ca2+ at pH 7.5–9.0 and had a rate constant (K D ) of 1.6 × 10–5 s–1 for thermal inactivation at 50°C. These results in which squid gill TGase could be activated at higher concentrations of Ca2+ and NaCl than at a physiological concentration, suggest that contact with seawater or body fluid seems to activate the enzyme if the tissue is disrupted.  相似文献

ABSTRACT:   Amylase, with MW of 59 kDa, was purified from small abalone Haliotis sieboldii by ammonium sulfate fractionation, CM Sepharose Fast Flow and Sephacryl S-100 HR chromatographies. The optimal pH and temperature of purified amylase were 6.0 and 37°C, respectively. The purified enzyme was stable at pH 6.0–8.0 and low temperatures. It was activated by Ba2+, Mg2+, Ca2+, Co2+, Ni2+, Mn2+, K+, Ag+, Na+ and Li+, but completely or partially inhibited by Al3+, Cu2+, Cd2+, Hg2+ and Zn2+. EDTA could completely inhibit, while iodoacetamide, N-ethylmaleimide and urea partially inhibit the purified amylase. According to the digestion mode of various polysaccharides, the purified enzyme was considered to be an α-amylase.  相似文献

ABSTRACT:   The effects caused by suspended particles in seawater and sediment cover of the substrate on the attachment to the substrate of zoospores, and on the subsequent growth and survival of gametophytes of the brown alga Eisenia bicyclis were examined in the laboratory. The attachment rate to the substrate of the zoospores was remarkably reduced by the sediment cover on the substrate, and it decreased to 3.8% of the no-sediment control by a slight sediment cover of 3.0 mg/cm2 (0.048 mm in thickness). The growth of gametophytes was not inhibited at 10 mg/cm2 or less of sediment cover, though it was stopped at 30 mg/cm2. The survival rate of gametophytes became 39.4% at 5 mg/cm2 on the substrate, and gametophytes were all blighted at 30 mg/cm2. The ratio of male to female of the surviving gametophytes became 81.5:18.5, when the sediment cover was 10 mg/cm2. The rate of total loss ( TL [%]) of zoospores and gametophytes of Eisenia bicyclis was obtained from the following equation: TL  = 100 (1 − exp[−0.0339  C ] exp[−1.24  Q ]), where, C (mg/L) and Q (mg/cm2) indicate the concentration of suspended particles in seawater and the sediment on the substrate, respectively.  相似文献

The present study was carried out to investigate the influence of water temperature on the growth performance and digestive enzyme (pepsin, trypsin and lipase) activities of Chinese longsnout catfish. Triplicate groups of Chinese longsnout catfish (35.6±0.48 g, mean±SE) were reared at different water temperatures (20, 24, 28 and 32 °C). The feeding rate (FR), specific growth rate (SGR) and feed efficiency ratio (FER) were significantly affected by water temperatures and regression relationships between water temperature and FI, SGR as well as FER were expressed as FR=−0.016 T 2+0.91 T −10.88 ( n =12, R 2=0.8752), SGR=−0.026 T 2+1.39 T −17.29 ( n =12, R 2=0.7599) and FER=−0.013 T 2+0.70 T −8.43 ( n =12, R 2=0.7272). Based on these, the optimum temperatures for FR, SGR and FER were 27.66, 26.69 and 26.44 °C respectively. The specific activities of digestive enzymes at 24 or 28 °C were significantly higher than that at 20 or 32 °C. In addition, there was a significant linear regression between FR or SGR and specific activities of pepsin and lipase, which indicated that pepsin and lipase played important roles in regulating growth through nutrient digestion in Chinese longsnout catfish.  相似文献

Artemia franciscana were hatched and tested for tolerance to ammonia at pH 6.5 and 8.5 in artificial seawater with a salinity of 17 ‰. Nauplii seemed to be less tolerant of ammonia at pH 6.5 despite the fact that more of the ammonia is in the ionized form (NH4+). It is suggested that this greater sensitivity is a result of the NH4+ competing with the Na+ during gut transfer. Nauplii hatched from decapulated cysts were more tolerant of ammonia than those hatched from whole cysts, probably because of a greater energy reserve.  相似文献

A generalized nitrogen budget was constructed to evaluate the potential role of mangrove sediments as a sink for dissolved inorganic nitrogen (DIN) in shrimp pond effluents. DIN concentrations were measured in pond effluents from three semi-intensive shrimp ponds along the Caribbean coast of Colombia between 1994–1995. Mean NH4+ concentrations in the discharge water for all farms were significantly higher (67 × 12 μg/L) than in the adjacent estuaries (33 × 8 μg/L). Average NH4+ concentrations in the pond discharge over all growout cycles were similar, representing an approximate doubling in relation to estuarine water concentrations. In contrast, NO2-+ NO3- concentrations were similar in both pond effluent and estuarine waters. Dissolved inorganic nitrogen loading of the ponds was similar. The estimated reduction of DIN in pond effluents by preliminary diversion of outflow to mangrove wetlands rather than directly to estuarine waters would be × 190 mg N/m2 per d. Based on this nitrogen loss and depending upon the enrichment rate, between 0.04 to 0.12 ha of mangrove forest is required to completely remove the DIN load from effluents produced by a 1-ha pond.  相似文献

Abstract— Feed intake, specific growth rate and changes in body composition were studied in age 1+ (140-190 g) brown trout ( Salmo trutta L.) reared at three temperatures (2, 4 and 6°C) under continuous light conditions. Feed in take increased from 35.7 kJ-kg−1.day−1 at 2°C to 95.7 kJ-kg−1-day−1 at 6°C, and the growth rate increased from 0.19%-day−1 to 0.42%-day−1 over the same temperature range. The estimated lower temperature limits for feeding and growth were slightly above 0°C. For all groups of fish, the majority (about 75%) of the weight gain comprised water and protein, but lipid deposition tended to increase with increasing temperature. The deposition of lipid accounted for about 50% of the body energy gain at all temperatures.  相似文献

The spawning ground of the Japanese sardine, Sardinops melanostictus (Schlegel), was distributed over the oceanic waters as well as the coastal waters along the Pacific coasts of western and eastern Japan during 1978–1992. The area of the spawning ground in the coastal waters on the continental shelf has ranged from 95 000 km2 in 1992 to 143 000 km2 in 1988, constituting 44–77% of the total area of the spawning ground. The area of the coastal spawning ground was relatively constant in spite of the large fluctuations in egg abundance, i.e. size of the spawning population, from 88 trillion (1987) to 668 trillion (1989) in the waters. Spawning adults seemed to extend over the coastal waters irrespective of the size of the spawning population. In contrast to the coastal waters, the spawning area in the oceanic waters offshore of the continental shelf increased from 31 000 km2 in 1978 to 183 000 km2 in 1988 and then shrank to 83 000 km2 in 1992, as a function of the spawning population size. The egg distribution density in the coastal waters stayed less than 6000 m−2 mo−1, but it reached as high as 27 400 m−2 mo−1 in the expanded spawning ground in the oceanic waters. The oceanic waters seemed to function as a reserve spawning ground for the sardine in years of extremely high spawning population.  相似文献

This study was conducted to evaluate three canola protein products as partial replacements of steam-dried whole herring meal (HM) in a practical diet for juvenile chinook salmon in seawater. Groups of 30–40 g chinook salmon held in 9.5–12°C seawater on a natural photoperiod were fed one of seven equivalent protein (390 g kg−1) and isoenergetic (20 MJ gross energy kg−1) diets to satiation. Each of three test protein sources, namely, commercial canola meal (CM), low-temperature extruded CM (90°C; LT), and high-temperature extruded CM (150°C; HT) comprised about 120 and 240 g kg−1 of dietary protein by replacement of 15 and 30%, respectively, of the HM protein in the control diet. Chinook salmon growth and feed intake were markedly depressed by replacing HM protein with a high level of CM, but these responses were not lowered by replacement with LT and HT. The phytic acid content in commercial canola meal was reduced by about 10 and 30% from the original level by extrusion cooking at low (90°C) and high (150°C) temperature, respectively. Therefore, the diets containing HT still had significant amounts of phytic acid. Terminal whole-body zinc contents were inversely related to the dietary levels of the different canola protein products. Thyroid function was altered by CM but this effect was offset by heat treatment. The results suggest that extrusion cooking of CM improves its nutritive value for chinook salmon in seawater. Indeed, it was found that HT could comprise 240 g kg−1 of the dietary protein without adversely affecting performance.  相似文献

ABSTRACT: To examine the involvement of chloride cells in the uptake of Ca2+ in freshwater (FW) killifish, chloride cell morphology was compared in fish acclimated to different defined FW environments with Ca2+ concentrations of either 0.1 mM, 0.5 mM, or 2.5 mM. Numerous chloride cells were detected in whole-mount preparations of the gill filaments, which were stained with an antiserum specific for Na+, K+-ATPase. Chloride cells, located mostly on the afferent–vascular edge of the filaments, were larger at lower Ca2+ concentrations. Electron microscopic observations showed that in the 0.1 mM and 0.5 mM Ca2+ experimental groups, the apical membrane of chloride cells were flat or slightly projecting and equipped with numerous microvilli. In the 2.5 mM Ca2+ group, some chloride cells formed an apical pit, whereas other cells were similar to those observed in the 0.1 mM and 0.5 mM Ca2+ groups. Plasma osmolality decreased with decreasing ambient Ca2+ concentration, suggesting that environmental Ca2+ affects the permeability of the body surfaces. Gill Na+, K+-ATPase activity in the 0.1 mM and 0.5 mM Ca2+ groups were significantly higher than that in the 2.5 mM Ca2+ group, implying the involvement of the Na+–Ca2+ exchanger in Ca2+ uptake in the gills. These findings suggest that chloride cells function as the site for Ca2+ uptake in killifish acclimated to low Ca2+ environments.  相似文献

The present study evaluates if abdominal dis-tension caused by a water-filled stomach in seawater farmed rainbow trout, Oncorhynchus mykiss (Walbaum), and Atlantic salmon, Salmo salar L., could be provoked experimentally by live chilling in sea water (0.5 °C). Fifty rainbow trout and 50 salmon were visually classified, either as normal or suffering from the condition. Prior to chilling, no rainbow trout or salmon suffered from a water-filled stomach. During chilling, 25% of the rainbow trout and 2.5% of the salmon developed water-filled stomach. Affected rainbow trout had significantly higher blood plasma osmolality and significantly lower body weight than normal trout. The frequency of trout with a water-filled stomach increased significantly with increasing chilling time and increasing plasma osmolality. The regression coefficients revealed that the proportion of affected rainbow trout in-creased by 0.82% h−1 of chilling and by 0.46% per unit increase in mosmol L−1. The present study revealed that abdominal distension in seawater farmed rainbow trout was far more severe than in Atlantic salmon and could be provoked experimentally by osmoregulatory stress. This is consistent with the observation that water-filled stomach appear only occasionally in seawater farmed Atlantic salmon.  相似文献

Abstract. The aim of this research was lo evaluate the effect of density and feeding level (initially 8 or 16kg/m3 and 1·3% or 2% of present biomass respectively) on productive traits, meat composition and water quality output in farmed rainbow trout, Oncorhynchus mykiss (Walbaum). Individual fish growth shows significant differences in relation to density and feeding level; feed conversion rate is positively Influenced by the lower feeding level and low rearing density. At low density, specific growth rate, protein efficiency ratio and coefficient of variation of individual final weights are more favourable than at high density. Dry matter and lipid composition of meat are positively affected by feeding level; whereas no differences are recorded on the amino acid and fatty acid profiles. Water quality shows differences due to density (NO3 and NH4+) and feeding level (NO2, NH4+ and P).  相似文献
