A single intraperitoneal injection of a sterile extract of sonically disrupted group A streptococcal cells induced an inflammatory process in the hearts of mice. 将松材线虫携带的致病细菌无细胞滤液、无菌松材线虫和致病细菌混合物、致病细菌鞭毛蛋白无细胞滤液、无菌松材线虫、无菌松材线虫和非致病细菌混合物接种黑松导致的松树萎蔫的病状与野生松材线虫导致松树萎蔫的病状进行观察和对比,结果为:(1)经培养的致病荧光假单孢菌GcM5-1A菌株的无细胞滤液、无菌松材线虫和致病细菌GcM5-1A混合物接种导致的黑松的病状变化趋势与松材线虫致死的黑松病状是一致的;无菌松材线虫、无菌松材线虫和非致病细菌AM2C混合物不能使黑松发病;(2)致病细菌GcM5-1A无细胞滤液和野生松材线虫导致的黑松病状和病程天数无显著差异;(3)接种无菌松材线虫和致病细菌GcM5-1A混合物的黑松病程显著长于接种野生松材线虫的黑松.结果可以用松萎蔫病是松材线虫和其携带的细菌的复合侵染病害的假说和松材线虫与其携带的细菌之间的互惠共生关系的论点来解释,说明松材线虫携带的致病细菌无细胞滤液是松萎蔫病致病的真正原因.  相似文献

【目的】目前使用的基因Ⅰ型猪瘟兔化弱毒C株疫苗是公认的安全、有效疫苗,但近年来基因Ⅱ群的猪瘟流行毒株比例增高,因此对猪瘟(CSF)的有效防控提出了更高要求。本研究旨在通过反向遗传学技术构建适应CSFV基因Ⅱ型的新型C株疫苗,为标记疫苗的开发奠定基础。【方法】在构建的C株感染性克隆基础上,拯救嵌合CSFV基因Ⅱ型HZ1-08毒株E2抗原区的重组C株病毒RecCHZE2,并分析其致病性和免疫原性。【结果】重组嵌合病毒RecCHZE2接种家兔后出现典型的定型热,兔脾脏肿大;重组嵌合病毒RecCHZE2接种猪只后没有发热和明显的白细胞减少,在接种后第12天检测到一过性的病毒血症;对接种猪血清检测结果表明,针对2型毒株的相应抗体在接种后的20 d达到高峰;接种猪血清针对C株和HZ1-08毒株的中和抗体效价相当,但C株免疫猪血清对流行毒株的中和抗体水平降低。【结论】重组嵌合病毒RecCHZE2不仅保留C株病毒弱毒的特性,且可提高针对Ⅱ型流行毒株的抗体水平。  相似文献

采用离体培养法探讨了人参根系分泌物中有机酸类物质(苯甲酸、邻苯二甲酸、己二酸、丁二酸)及皂苷类物质(人参皂苷Rb2、Rd、Rc、Rg1、Re)对人参主要病原菌及其生防菌的化感作用差异。结果显示,有机酸及皂苷对微生物的化感作用表现出受体多、阈值低(1 μg·L-1)、作用质量浓度范围大(1 μg·L-1~1 mg·L-1),但无明显浓度效应的特点;有机酸及皂苷的化感作用强度介于-132.94%~31.33%,对生防菌的促进作用大于病原菌,对部分病原菌生长具有直接的抑制活性,二者对病原菌与生防菌表现出一定的偏害/偏利作用;人参皂苷Rd、Re具有抑制病原菌、促进生防菌的作用,是人参根系分泌物中重要的萜类防御物质。  相似文献

 报道三七根腐病复合侵染中病原细菌的研究结果。病菌从根部侵入,引起根腐,叶片褪绿变黄或青枯,病菌随病残体、土壤和种苗传播。病菌在金氏B培养基上菌落圆形,灰白色,略凸起,边缘整齐,半透明,湿润光滑,不产生荧光色素,菌体杆状,直或稍弯,革兰氏染色阴性,具1~6根极生鞭毛。积聚β羟基丁酸盐颗粒,氧化酶、精氨酸双水解酶呈阳性,烟草过敏反应,马铃薯软腐阴性,人工接种发病症状与田间症状相似,发病率达90%以上。经鉴定,该菌为假单孢杆菌(Pseudomonas sp.)。  相似文献

【目的】从表达M2e-EYFP融合蛋白转基因柔嫩艾美耳球虫的稳定性,及其在环境中的传播能力、竞争能力和存活能力方面,分析转基因球虫对生态环境的潜在风险。【方法】稳定性试验包括:转基因球虫感染非靶动物,观察临床症状和卵囊产出情况;转基因球虫继代繁殖,检测荧光率;转基因球虫与毒害艾美耳球虫混合感染,PCR检测卵囊中的外源基因。传播能力试验主要是分析感染鸡和不感染同居鸡的卵囊产量和荧光率。竞争性试验则是用不同混合比例的2种球虫感染鸡,检测卵囊产量和荧光率;存活能力试验是用不同处理方式的卵囊感染鸡,检测卵囊是否产出以分析卵囊存活能力。【结果】转基因球虫不感染小鼠和家鸽,与野生型柔嫩艾美耳球虫宿主特异性相同;连续繁殖传17代后外源基因稳定表达。与毒害艾美耳球虫混合寄生时在异种球虫体内未检测出外源基因。传播能力试验中,46d攻毒时转基因球虫感染与不感染同居组的卵囊产量差异不显著,但与不感染对照组相比均显著减少,且1∶1感染和不感染同居组卵囊荧光率检测结果维持在51.6%67.9%。竟争性试验中,攻毒时各比例感染组卵囊产量均极显著少于不感染攻毒对照组,荧光率检测结果各比例组变化幅动不大,且1∶1组哨兵鸡卵囊荧光率检测结果为41.2%61%。存活能力试验,转基因球虫与野生型球虫在夏季室温条件下均不能存活90d,且与其他不同方式处理的卵囊外界存活时间接近。【结论】说明转基因球虫具有良好的稳定性,在环境中的传播能力、竞争能力和存活能力未呈现显著优于野生球虫的趋势。  相似文献

Newly formed small lymphocytes with a short life-span in the blood are the only cells from thoracic duct lymph which accumulate in acutely inflamed tissue. This conclusion is drawn from studies in which rats with induced peritonieal exudates were injected intravenously with radioactively labeled thoracic duct cells. Radioactivity, originally vested in newly formed donor small lymphocytes, was found later in a small number of similar exudate cells. Small lymphocytes generated 10 days or more before the thoracic ducts were cannulated failed to localize in peritoneal exudates, although the cells moved in large numbers from the blood in to lymph nodes.  相似文献

【目的】从患真菌性皮肤病肉牛的毛发、皮屑、痂皮等病料样本中分离皮肤病原真菌并对其进行种属分类鉴定,为牛真菌性皮肤病的诊断及防治提供参考依据。【方法】无菌采集宁夏地区患皮肤病肉牛的毛发、皮屑、痂皮,将病料样本置于载玻片上,滴加10% KOH溶液1滴,静置5 min后进行镜检观察;将病料样本接种于沙氏葡萄糖琼脂培养基(SDA)中进行真菌的分离、纯化培养,对分离的真菌用乳酸酚棉兰染液进行染色观察,根据真菌形态学特征对其进行初步分类;用分离的真菌制备浓度为1×108 cfu·mL-1的孢子悬液涂抹健康ICR小鼠皮肤,根据其致病性筛选病原真菌;提取病原真菌DNA,对其内部转录间隔区(ITS)进行PCR扩增并测序,登录NCBI GenBank数据库,对病原真菌ITS序列进行同源性比较并构建系统发育树,根据其同源性比较及系统发育分析结果,结合真菌形态学特征对病原真菌作出种属分类鉴定。【结果】病料中含真菌菌丝及链状厚垣孢子,毛发中存在大量小分生孢子;从病料样本中共分离、纯化到23株真菌,分属于毛癣菌属(Trichophyton)、曲霉属(Aspergillus)、横梗霉属(Lichtheimia)、链格孢属(Alternaria)、镰刀菌属(Fusarium)、附球菌属(Epicoccum)、毛霉菌属(Mucor);小鼠皮肤感染试验表明,分离的毛癣菌属真菌NXGY1、NXGY2具有较强的致病性,试验组小鼠均出现瘙痒、皮屑增多、形成结痂、脱毛等临床症状;毛癣菌属NXGY1、NXGY2 的ITS序列长度大小分别为660、662 bp,其同源性比较及系统发育分析结果表明,NXGY1、NXGY2与疣状毛癣菌(Trichophyton verrucosum)同源性高达99% ,在系统发育树上属于同一分支,遗传距离最近。【结论】根据真菌形态特征及系统发育分析结果,鉴定分离的肉牛皮肤病原真菌NXGY1、NXGY2为疣状毛癣菌(T. verrucosum)。  相似文献

以植物内生真菌球毛壳菌ND35菌株和病原真菌立枯丝核菌Rs-1菌株及毛白杨树组培苗为试材,分析检测了接种球毛壳菌后立枯丝核菌侵染杨树组培苗的发病情况和4种不同处理的植物产生过氧化物酶(POD)、多酚氧化酶(PPO)和苯丙氨酸解氨酶(PAL)的活性变化。结果表明:与对照相比接种球毛壳菌后的植物在一定程度上能够阻止立枯丝核菌的扩展,发病率和病情指数均有所降低,明显提高植物POD、PPO和PAL的酶活性。说明接种球毛壳菌后诱导植物产生的3种防御酶对提高植物抗病性有一定的影响。  相似文献

Friable callus was induced when immature seeds of floribunda rose were inoculated on MS medium supplemented with 2,4-D 3.0 mg.L^-1. When transfered onto subculture media, friable callus developed into embryogenic callus, which was used to establish cell suspension lines. Cell suspensions had to be subcultured at a interval of 4-5 days at the first several culture cycles. The best subculturing cycle for the stable cell suspensions was 8-10 days. The best inoculum quantity was 1 mL PCV(Packed Cell Volume) per 40 mL culture fluid.  相似文献

Sarcocysts morphologically similar to Sarcocystis muris were observed in mice after inoculation with Toxoplasma-like oocysts found in feces of a stray cat. Cats that were fed mice infected with the oocysts shed similar oocysts in their feces. Sarcocysts were found histologically in about 50 percent of mice inoculated with 40,000 or more oocysts and examined 42 days or longer after exposure. Most inoculated mice developed low Toxoplasma dye-test antibody titers 3 to 4 weeks after exposure, but Toxoplasma antibody was usually not detectable in infected cats.  相似文献

Cells derived from lung biopsies and pleural effusions from AIDS patients with Kaposi's sarcoma (KS) of the lungs were established in long-term culture with the aid of conditioned medium from HTLV-II-transformed T cells (HTLV-II CM). These AIDS-KS cells were similar to the so-called spindle cells in KS lesions and had some of their features. They produced factors that supported their own growth (autocrine) and the growth of other cells (paracrine), including umbilical vein endothelium and fibroblasts. That the AIDS-KS cells also expressed potent angiogenic activity was demonstrated by the chorioallantoic membrane assay and by subcutaneous inoculation of AIDS-KS cells into nude mice, which resulted in the development of angiogenic lesions composed of mouse cells and showing histological features similar to those of human KS lesions. These data suggest that AIDS-associated KS and possibly other types of KS may be initiated by signals that induce the growth of particular cells (spindle cells of lymphatic or vascular origin) and the expression of autocrine and paracrine activities.  相似文献

在无鉴别寄主体系的现状下,为研究不同地区的玉米南方锈菌Puccinia polysora Underw. 的相对寄生适合度(relative parasitic fitness)和致病性,以及不同玉米品种对其抗性水平,本试验以采自海南三亚、广东河源、广西河池、广西桂林、湖南邵东、云南玉溪6 个地区的可能存在致病性分化的同种玉米南方锈菌为研究对象,将其分别接种到28 个玉米品种上,分析其病情指数和相对寄生适合度。结果表明:抗病性最高的玉米品种为登海605,致病性最强的菌株来源于广西河池;不同来源的玉米南方锈菌在同一玉米品种上的相对寄生适合度存在较大差异;相同来源的菌株在不同玉米品种间相对寄生适合度之间也存在显著差异;玉米南方锈菌存在致病性分化。本研究可为抗玉米南方锈病品种选育及玉米南方锈菌致病类型与生理小种鉴定提供科学依据。  相似文献

【目的】分离鹅源坦布苏病毒,并研究其对雏鹅的致病性。【方法】用RT-PCR方法对江苏省3个鹅场送检的产蛋期病鹅样品进行检测;对坦布苏病毒核酸阳性的样品进行病毒分离,测定分离病毒的生物学特性和E蛋白基因序列。【结果】这些样品中坦布苏病毒呈阳性;从病料中分离鉴定一株病毒,命名为SHYG。E蛋白基因序列同源性比对显示其与中国鸭源坦布苏病毒同源性达到99.3%;生物学特性研究表明该病毒可致Vero细胞病变,对小鼠无致病性,接种2周龄雏鹅出现精神沉郁、腹泻、神经症状甚至死亡;组织学变化可见肝脏淤血、胆小管扩张、脂肪变性;肺淤血、炎性渗出及含铁血黄素沉着,脾脏网状内皮细胞空泡化,肾出血,脑充血、胶质细胞增生。【结论】坦布苏病毒SHYG株对雏鹅有较强的致病性。  相似文献

探讨不同免疫程序对MDV(Marek's DiseaseVirus,MDV)抗独特型抗体苗抗感染保护的影响.将MDV抗独特型抗体苗用胚内.皮下与单纯皮下相结合的方法免疫SPF鸡,以第二次免疫接种5 d攻毒后, 进行抗感染保护测定,比较各实验组淋巴细胞增生病变,病毒分离及大肠杆菌感染指标等的差异,确定MDV抗独特型抗体苗的免疫程序.结果表明,二种免疫效果相同,抗MD感染保护力达90%.  相似文献

通过小白鼠醋酸扭体法、家兔蛋白胨致热法对氟尼辛葡甲铵(FM)解热、镇痛作用进行了药效学研究.结果表明:氟尼辛葡甲铵具有明显的解热、镇痛作用.与对照组相比,氟尼辛葡甲铵4个剂量组(1.25、2.5、5、10 mg/kg)对醋酸所致的小鼠扭体反应均有极强的抑制作用,抑制率最高达100%.2.5 mg/kg的FM镇痛率即达82.7%,明显强于双氯芬酸钠(65.4%)和安乃近(58.7%).对蛋白胨引起的家兔发热的解热效果,氟尼辛葡甲铵高剂量组(4 mg/kg)优于安乃近组(0.2 g/kg)(P<0.05)和氨基比林组(0.2 g/kg)(P<0.01).中剂量氟尼辛葡甲铵组(2 mg/kg)解热作用稍逊于安乃近组,但差异不显著.低剂量氟尼辛葡甲铵组(1 mg/kg)解热作用与氨基比林组相当.  相似文献

Goat antibodies to idiotypes (anti-idiotypic antibodies; Ab2) that recognize an idiotype associated with the combining site of a BALB/c mouse IgG2a monoclonal antibody (Ab1) to human gastric carcinoma were used to immunize BALB/c mice and rabbits. A monoclonal anti-anti-idiotypic antibody (Ab3) of IgG1 isotype was obtained after immunization of mice. The Ab3 and the Ab1 showed identical binding specificities and bound with similar avidities to the same tumor antigen. The induction of Ab1-like Ab3 by Ab2 was not restricted to mice, since Ab3 could also be induced in rabbits. Both the mouse- and the rabbit-derived Ab3 bound the same gastrointestinal cancer-associated antigen as Ab1. These findings indicate that Ab2 induced the formation of antigen-specific Ab3, probably because it bears the internal image of the tumor-associated antigen. This Ab2 may therefore have potential for modulating the immune response of cancer patients to their tumors.  相似文献

Since May 2006,a highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV) variant characterized by 30 amino acids deletion within its NSP2-coding region emerged and caused extensive economic losses to China's pig industry.To investigate the in vivo pathogenicity and immune responses of the newly emerging PRRSV,3 groups of 60-d-old conventional piglets were inoculated intranasally with a representative strain of the HP-PRRSV variant HuN4 with 3 different infection doses (3× 103-3× 105 TCID50).The results revealed that the virus variant caused severe disease in piglets and the significant clinical characteristics consisted of persistently high fever (41.0-41.9℃) and high morbidity and mortality (60-100%),the marked clinical signs of PRRS and severe histopathogenic damages in multiple organs.It induced rapid and intense humoral immune responses and seroconversion was detected in most infected pigs at 7 d post-infection (DPI).The virus vigorously replicated in vivo and the highest virus average titer was 9.7 log copies mL-1 serum at 7 DPI.Elevated levels of IFN-y and IL- 10 cytokine production in serum in this study were also observed.Taken together,our results demonstrated that the HP-PRRSV variant HuN4 strain is highly pathogenic for piglets and suitable to be a reference strain of highly virulent PRRSV for evaluating the efficacy of the new vaccines.  相似文献
