Several conclusions of importance to studies on the epidemiology of heartwater were drawn from an investigation in which the numbers of adult Amblyomma hebraeum ticks, to which a closed herd of Hereford cattle were exposed over a period of 6 1/2 years, were manipulated. With a tick Cowdria ruminantium infection rate of 3-5%, an endemically stable situation was created by dipping the herd only when an average of 10 adult male and female A. hebraeum ticks were counted on 10 animals. When the average was increased to 15 during the calving period, 97% of calves acquired a tick-mediated immunity at the age of 6 months. Because only adult ticks confined to the hindquarters are counted, this procedure is recommended as a feasible and practical guideline to stock owners wishing to determine a dipping programme that would ensure endemic stability. The indirect fluorescent antibody test gave a true reflection of the infection rate through ticks in calves 3-6 months old, but not in older animals that had been re-infected more than once. This is because on one hand antibody may persist for 2 years after withdrawal from tick exposure and on the other the artificial re-infection of cattle with a tick acquired immunity is not always followed by a rise in antibody titres and may even result in seronegativity. Four cows infected and re-infected through ticks, remained immune to challenge for 2 years after withdrawal from tick exposure. Within the confines of one farm 3 isolates of C. ruminantium that differed in pathogenicity and immunogenicity were recovered from ticks. Serum samples collected in a cross-sectional survey of grazing cattle on Manyara Ranch, Monduli district, Tanzania, were tested by indirect major antigenic protein 1 fragment B (MAP 1-B) ELISA to determine the seroprevalence of Ehrlichia ruminantium and to assess ranch-level risk factors for heartwater. Heartwater-exposed cattle were widespread on the ranch and overall seroprevalence was 50.3% (95% CI, 44.9-55.6), enough to indicate an endemically unstable situation. Multivariate logistic regression modelling was used to identify risk factors associated with seropositivity. Two factors appeared to increase the herd's risk for contracting heartwater. Seroprevalence increased significantly with age (beta = 0.19 per year of age, P < 0.001) and animals carrying ticks of any species were associated with an increased risk of infection with E. ruminantium (Odds ratio, OR = 3.3, P < 0.001). The force of infection based on the age seroprevalence profile was estimated at 18 per 100 cattle year-risk. The current tick control measures on the ranch were associated with a decreased risk of infection with E. ruminantium (OR = 0.25 for no dipping and OR = 0.31 for low dipping, P < 0.001). Six tick species were identified; in order of frequency these were: Ambylomma variegatum 59.9%, Rhipicephalus evertsi evertsi 13.9%, Rhipicephalus pulchellus 12.5%, Hyalomma truncatum 7.03% and Rhipicephalus appendiculatus 6.07%. The least encountered tick was Rhipicephalus simus, which accounted for 0.38%. The cattle seemed well adapted to their environment and capable of resisting the tick burden under this extensive wildlife/livestock grazing and interaction system.  相似文献

We report Marie Galante as one of the Caribbean islands most heavily infested by the tropical bont tick (TBT) Amblyomma variegatum which is associated with two major diseases of ruminants: heartwater and dermatophilosis. In 2005, a survey was undertaken to assess the prevalence of TBT infestation in cattle, the prevalence of Ehrlichia ruminantium infection in TBTs, and the tick control measures implemented by livestock owners. A random sample of 195 cattle herds out of 1885 recorded on the island was investigated by thoroughly counting adult ticks on each animal and filling a questionnaire. A randomly collected sample of 136 TBTs was tested for infection by E. ruminantium by pCS20 nested PCR. Cattle herd prevalence (hp) was 73.8% for infestation by at least one TBT, 17.9% for infestation by at least one engorged female TBT, and 8.2% for clinical dermatophilosis. Cattle individual prevalence was 42.3% for infestation by at least one TBT, 6.6% for infestation by at least one engorged female TBT, and 2.2% for clinical dermatophilosis. The minimum, maximum and average numbers of TBTs per infested animal were, respectively 1, 108 and 11.5. Prevalence of TBT infection by E. ruminantium was 19.1%. No significant difference in herd prevalence was found among parishes or among ecological zones. For cattle owners treating against ticks (97.9% of all owners), all used aspersion of amitraz and herd prevalence was significantly different among those treating every 1-2-week (hp=69.6%, n=148), and less often than every 2-week (hp=88.6%, n=35) (P=0.031). Of the 42 herd subunits treated less than 4 days before the survey, 27 (64%) were infested with at least one TBT, and 6 (14%) with at least one engorged female TBT. These results indicate a high level of TBT infestation in Marie Galante, the inefficacy of tick treatments currently performed, and the need for an improved tick control strategy. Persisting high levels of infestation in Marie Galante threaten the success of on-going TBT eradication programs in the Caribbean because TBT can spread through migrating birds and trade of animals or of animal hides to other islands and potentially the American continent.  相似文献

Blood samples and ticks were collected from 48 cattle and 74 horses from seven sites in the Peten region of Guatemala. Data on body condition, mucous membrane capillary refill time and tick infestation levels were recorded for each animal in the study. Horses had significantly higher levels of tick infestation than cattle, as well as poorer body condition scores. Seroprevalence of Babesia spp. was 95.8% for B. bovis in cattle, 89.6% for B. bigemina in cattle, and 92.7% for B. equi in horses. Seroprevalence of Anaplasma marginale in cattle was 87.5%, similar to reports in animals from other regions of Central America. This is the first time that A. phagocytophilum has been reported in animals from this region, with overall PCR-prevalence of 27.6% in cattle and horses, and seroprevalence of 28.4% (52% in cattle and 13% in horses). An agent was identified with serological cross-reactivity and close genetic relatedness to Ehrlichia ruminantium, but further testing confirmed that the agent in Guatemalan cows was not the agent of heartwater. Ticks were identified to species with the predominant species identified on cattle as Boophilus microplus and Amblyomma cajennense, while Anocentor nitens and A. cajennense were most commonly found on horses. Prevalence of infection, tick infestation levels, host factors and environmental data were analyzed for association; A. nitens was significantly associated with A. phagocytophilum prevalence by village.  相似文献

In 1994 a batch of apparently healthy goats was selected for intended export to the USA from a heartwater-free and vector tick-free region of South Africa. The animals were tested serologically for heartwater, using either or both an IFA and an ELISA test, and 52% were found to be serologically positive. A PCR assay based on Ehrlichia ruminantium 16S gene sequences gave positive results for 54% of the animals, suggesting that apparently non-pathogenic E. ruminantium variants existed in this heartwater-free area. To identify and characterise the agents responsible for the positive serological and PCR results, ticks and animal blood samples were collected from two of the three farms involved in the original survey during two successive seasons of expected peak tick activity. Ticks were kept alive for a minimum of 3 weeks to allow digestion of any blood meal before being processed. Over the two seasons, 28% of the livestock and 15% of the ticks sampled were found to be carrying E. ruminantium. E. ruminantium 16S and pCS20 sequences were detected in all of the four tick species collected from the livestock (Rhipicephalus evertsi evertsi, Rhipicephalus evertsi mimeticus, Hyalomma truncatum, Hyalomma marginatum rufipes), suggesting that some of the species may act as vectors. Animals generally carried multiple E. ruminantium 16S genotypes, whereas ticks rarely carried more than one. Infection levels in both animals and ticks were too low to generate a marked response when a blood stabilate was sub-passaged in a clean sheep, preventing the subsequent establishment of any of the organisms in culture.  相似文献

At present, heartwater in the Caribbean is known with certainty only on Guadeloupe, Marie Galante and Antigua; the first 2 islands are widely infected. The most important factors responsible for particular aspects of heartwater in Guadeloupe are: Cowdria ruminantium of high virulence. A very resistant cattle population (Creole), not normally clinically affected. A fairly susceptible goat population (Creole) (22% goats born in endemic areas die after experimental inoculation) which, fortunately, includes breeding lines with inherited resistance characteristics. Amblyomma variegatum which is present all over the island and all through the year, but with a low infection rate (1-2% of adult ticks are infected) because of the short period of rickettsemia in infected animals. The low rate of tick infection results in a low endemicity of the disease. For goats, the epidemiologic situation can be regarded as unstable because the low rate of infection in ticks does not allow a natural immunization of the majority of young kids when they still have a non-specific resistance. Experimental infection trials were conducted to investigate susceptibility of leopard tortoises (Geochelone pardalis) and helmeted guineafowl (Numida meleagris) to infection with Cowdria ruminantium, the causative agent of heartwater, a tickborne disease of domestic and wild ruminants. Ten guineafowl were inoculated intravenously with a virulent dose of C. ruminantium derived from bovine endothelial cell cultures, and four leopard tortoises were exposed to C. ruminantium infection by the feeding of infected Amblyomma hebraeum ticks. Uninfected A. hebraeum ticks (on both tortoises and guineafowl) and Amblyomma marmoreum ticks (on tortoises only) were fed on the animals during weeks 2 and 3 post-exposure in an attempt to detect infection. These ticks were analyzed for C. ruminantium infection by xenodiagnosis and with the C. ruminantium-specific pCS20 polymerase chain reaction (PCR) assay. Attempts to detect infection in ticks fed on either species were negative by both tests. These results suggest that leopard tortoises and helmeted guineafowl are refractory to C. ruminantium infection and, therefore, are unlikely to be capable of introducing heartwater directly into new areas. However, leopard tortoises are efficient hosts of A. marmoreum and A. hebraeum and are likely to be important epidemiologically in the transport and maintenance of these tick vector species.  相似文献

In order to detect the prevalence of Cowdria ruminantium in the vector tick, Amblyomma hebraeum, free-living, unfed adult ticks were collected with the aid of pheromone/CO2 traps. Ticks were collected at the Rietgat communal grazing area, as well as in the southwestern Kruger National Park and in the Songimvelo Game Reserve, all located in heartwater-endemic areas of South Africa. The presence of C. ruminantium in these ticks was determined by polymerase chain reaction (PCR) analysis. Ticks from the Rietgat communal grazing area were assayed in 2 batches and 4.7% of the one and 11.3% of the other were positive for infection, while 5.7% of the ticks collected in the Kruger National Park and 25% in the Songimvelo Game Reserve were positive. These results support the contention that a vector-wildlife cycle of transmission of C. ruminantium, the cause of heartwater in domestic ruminants, can be maintained in the absence of the latter animals.  相似文献

Creole goats and cattle in Guadeloupe can be carriers of cowdriosis (heartwater: Cowdria ruminantium) after recovery for a period as long as 11 months in goats and 2 months in cattle. The carrier status was demonstrated by feeding Amblyomma variegatum nymphs on recovered animals and the resulting adult ticks on susceptible goats. Cowdria ruminantium was not detected permanently during the carrier status.  相似文献

Two main methods are currently being used by farmers in the heartwater regions to control this disease in cattle and other stock, namely strategic control of ticks and total control of ticks. Some farmers make successful use of the latter method, particularly in areas which are marginal for the disease. Total tick control requires top managerial skills, which includes continual expert supervision, a relatively rare asset. The majority of farmers, often unintentionally, apply strategic control of ticks to control heartwater, either without or with vaccination against the disease. In this case the objective of tick control is to minimize "tick worry" without interfering too drastically with natural transmission of the disease. This method would make provision for natural immunization of young animals (a more uncertain method than vaccination) and the maintenance of immunity. In practice these methods appear to diminish deaths from heartwater appreciably, but do not protect all animals from the disease. Furthermore, they appear to be more effective in cattle than other stock. Although strategic control has an evolutionary background and considerable merit under African conditions, it is being applied in an empirical way because of paucity of information on the epidemiology of heartwater. More information on the infection rate in and infectivity of ticks (percentage of infected ticks) offer challenging research possibilities.  相似文献

In the Central Guinea savannah of C?te d'Ivoire, cattle breeding started only approximately 30 years ago. The impact of parasitism on the overall health status and productivity of the trypanotolerant N'Dama cattle in this area is unknown. In close collaboration with national veterinary institutions and local farmers, we studied spectrum, burden and seasonal dynamics of ticks (including aspects of cowdriosis) on N'Dama village cattle. In a longitudinal study, three randomly selected cattle herds (traditional farming type) of one village were examined repeatedly for ticks. Spectrum, burden, seasonal epidemiology of ticks were assessed. In these traditional herds (which lack (ecto)parasite management), all animals were infested by ticks at monthly counts. Five different tick species were identified; the four genera in order of frequency were: Amblyomma (overall prevalence 96%), Boophilus (47%), Hyalomma (<1%) and Rhipicephalus (<1%). Amblyomma variegatum was the most-abundant tick on cattle in all seasons. Seroprevalence of Cowdria ruminantium was 31% (95% CI: 26, 36%). Most of the animals typically carried low tick burdens. N'Dama cattle seem well adapted to their environment and can resist the tick burdens under this traditional farming system.  相似文献

Detection of heartwater is not always easy especially because all the serological assays so far available either have poor sensitivity or specificity. The indirect MAP-1B ELISA has been reported to be the most specific test for heartwater, although it does also detect antibodies to some closely related ehrlichial agents. This study was undertaken to compare two methods for the detection of heartwater infection caused by the ehrlichial agent Ehrlichia (Cowdria) ruminantium. Fifteen cattle on a heartwater-endemic farm infested with high numbers of Amblyomma hebraeum ticks, and hence exposure to E. ruminantium infection were monitored over an 8-week period by pCS20 PCR and an indirect MAP-1B ELISA. Infection was detected by pCS20 PCR in most animals with the highest number of positives (60%) in week 6 of the study. Similarly, exposure to E. ruminantium was detected by indirect MAP-1B ELISA in some animals, with the highest number of seropositives (27%) at weeks 2-6 of the study. The data demonstrated a fluctuating rickettsaemia in cattle in a heartwater-endemic area. Comparison of the two tests indicated that the pCS20 PCR assay was more reliable because it detected more infections than the indirect MAP-1B ELISA and would therefore be the method of choice for detection of E. ruminantium infection.  相似文献

The Theileria parva carrier-state in cattle on commercial farms on Zimbabwe was investigated using parasitological and serological methods. The proportion of cattle showing Theileria piroplasms on two farms, which had recent histories of disease outbreaks, were 64% (n = 106, total of heifers and weaned calves examined) and 71.5% (n = 60) while the proportion of T. parva antibodies for the same animals were 59% and 98.5%, respectively. On four farms where no cases of the disease occurred for over 10 years, the average proportion of animals showing piroplasms and antibodies were 55.4% (range 32-82, n = 223) and 73% (range 47-91, n = 223), respectively. However, on another three farms which had no history of theileriosis outbreaks these proportions were very low, being 11.4% (0-24, n = 157) for piroplasms and 12.2% (5-23, n = 157) for antibodies. The mean infection rate in unfed Rhipicephalus appendiculatus adults collected from farms with a high prevalence of cattle which were carriers of Theileria piroplasms during the tick activity season was 29% (range 12-60%) with 9.3 (range 2-18.7) mean infected acini per infected tick. The infectivity of different tick batches to susceptible cattle produced a wide spectrum of theileriosis reactions. Laboratory controlled experiments were carried out to study the persistence of T. parva (Boleni) piroplasms in cattle immunized with this strain as well as its infectivity for ticks and its subsequent transmissibility to cattle. Examination of the salivary glands of 15 batches of ticks collected from six immunized cattle on three different occasions over 18 months showed that none were infected with Theileria parasites. However, the infectivity of other ticks in the same batches to susceptible animals was demonstrated 6, 10 and 18 months after cattle had been immunized with Boleni stabilate.  相似文献

Ixodid ticks were collected from domestic animals, mainly cattle, in 11 administrative zones covering 84 districts in central Ethiopia over a period of 2 years (July 1996 to June 1998). Nineteen tick species were identified. Four of these belonged to the genus Amblyomma, one to Boophilus, two to Haemaphysalis, three to Hyalomma and nine to Rhipicephalus. Amblyomma variegatum and Rhipicephalus evertsi evertsi were present in all 11 administrative zones and, with the exception of Afar, Boophilus decoloratus was present in nearly every district in which collections were made. These three species constituted more than 50% of all ticks collected. Amblyomma cohaerens and Rhipicephalus bergeoni were common in the west of the survey region and Rhipicephalus pulchellus in the east. Except for B. decoloratus, of which more females than males were collected, the numbers of male ticks recovered were equal to or exceeded those of females. Mortality in crossbred dairy cattle caused by heartwater (Cowdria ruminantium infection) was reported during the survey period. An integrated approach to tick control is suggested.  相似文献

A survey was carried out to define the distribution of heartwater in goats that originated from six districts in communal grazing semi-arid areas of Zambia. A total of 181 samples (40.1%) out of 451 serum samples from adult goats were positive for Ehrlichia ruminantium antibodies after screening using indirect MAP-1B antigen ELISA technique with statistically significant differences (P < 0.01) between the six districts. Out of 1 036 adult goats examined for tick infestation, 105 were infested by ticks, with Amblyomma species being the most dominant tick encountered. Amblyomma variegatum, which is the vector for heartwater transmission in Zambia constituted 42.4% of the tick species, identified. The overall tick infestation rate was 10% while the tick:goat ratio was 2.1:1. Amblyomma variegatum appears to be widespread throughout the study area, as are antibodies to E. ruminantium.  相似文献

The importance of equine piroplasmosis control in endemic countries has increased in recent years and plays an important role to maintain the international market open to the horse industry. The purpose of this study was to demonstrate the occurrence of equine piroplasmosis (Babesia equi or Theileria equi and Babesia caballi) in Brazil--a country where the disease occurs endemically--in different climatic conditions, and to evaluate the results of a strategy for tick control in order to decrease infection rates. Blood samples were taken from 720 horses on 28 farms from different regions and subjected for complement fixation testing. The strategy was based on the control of the tick population by spraying the horses with acaricides, treating positive horses and preventing cattle and horses from grazing together. A significant association was found in the prevalence of antibody titres in tropical and subtropical areas. A significantly lower prevalence rate occurred on those farms where measures to control tick population were established. Farms in endemic countries may significantly reduce the prevalence of equine piroplasmosis by establishing measures to control the tick population and treating chronic carrier horses. Additional measures for controlling ticks in tropical areas are also discussed.  相似文献

Adult Amblyomma hebraeum ticks, the principle vector of heartwater (cowdriosis) of domestic ruminants in southern Africa, were collected in pheromone traps placed in Kruger National Park, an exclusively wildlife sanctuary in South Africa. These ticks transmitted Cowdria ruminantium, the rickettsial agent causing heartwater, to a susceptible goat, resulting in acute, fatal disease. C ruminantium was isolated in bovine endothelial cell culture from the plasma of this animal during the febrile stage of the disease and transmitted to susceptible goats, causing fatal heartwater. The prevalence of C ruminantium infection in 292 ticks was determined by polymerase chain reaction (PCR) analysis to be 1.7 per cent (95 per cent confidence interval 0.71 to 4.0 per cent). A DNA probe analysis, which is less sensitive than PCR, detected infection in three of the five PCR-positive ticks. The remaining infections were below the detection limit of the DNA probe, which is approximately 70,000 organisms. This is the first evidence that a vector-wildlife cycle of transmission of C ruminantium can be maintained independently of domestic ruminants.  相似文献

Five groups of Tswana-cross castrated male cattle between 20 and 30 months of age (a total of 158 animals) were transported from a ranch in a heartwater-free area of south Botswana to a feedlot near Gaborone in the east of Botswana where heartwater is endemic. On arrival, one group was vaccinated intravenously with the Onderstepoort sheep blood heartwater vaccine, one group was vaccinated intravenously with the new Onderstepoort tick-derived heartwater vaccine and a third group was vaccinated subcutaneously with this tick-derived vaccine. Vaccine reactions were blocked with long acting oxytetracycline on the first day of fever. A fourth group had a series of injections of long acting oxytetracycline on days 0, 7, 14 and 21 after arrival, and a fifth served as untreated controls. The animals remained at the feedlot for 65 days during which time they faced a low level of challenge by Amblyomma hebraeum ticks. None contracted heartwater and so they were then challenged, together with a further group of control cattle, with a dose of the sheep blood vaccine. Some animals in all groups had severe heartwater reactions and died despite therapy, but 76.7 per cent, 64.5 per cent and 74.3 per cent of the cattle in the blood vaccine, intravenous tick vaccine and long acting oxytetracycline groups respectively were resistant to challenge, compared with 48.3 per cent of the subcutaneous tick vaccine group and 36.4 per cent of the controls. It was concluded that intravenous vaccination of susceptible adult cattle with either the blood or the tick-derived vaccine needs careful monitoring in the month after vaccination and does not necessarily result in immune animals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Cattle, vaccinated as calves with Cowdria ruminantium-infected tick stabilate, were challenged 6, 12 and 24 months later. In the absence of tick challenge, vaccination of calves induced a partial immunity against subsequent challenge at 12 and 24 months. In animals exposed to ticks, the resistance was no better than that of control, unvaccinated cattle. When they were challenged at 6 months of age there was no difference between vaccinated and unvaccinated calves, either in the absence or presence of tick challenge, and all the animals manifested a high degree of natural resistance. This study therefore suggests that the value of vaccinating Afrikander-cross calves in heartwater endemic areas should be further investigated. The indirect fluorescent antibody (IFA) test proved to be a valuable means of monitoring the serological response of vaccinated animals and detecting the sero-conversion of animals exposed to tick infection. On one hand, there was good correlation between the febrile reaction and the results of the IFA test on the sera of vaccinated and control cattle challenged with the heartwater agent, in that all sero-positive animals were resistant to challenge. On the other hand, though, a considerable percentage of the animals that were serologically negative were also resistant to challenge.  相似文献

We present a simple model of the dynamics of heartwater that we use to explore and better understand various aspects of this disease. We adapted the Ross-Macdonald model for malaria epidemiology so that we could consider both host and vector populations, and evaluate the interactions between the two. We then use two more biologically detailed models to examine heartwater epidemiology. The first includes a carrier state and host mortality, and the second includes density dependence. The results from all three models indicate that a stable equilibrium with high disease levels is probably the standard situation for heartwater (R0 between 5.7 and 22.4). More than 80% of cattle become infected with heartwater if only 12% of infected tick bites produce an infection in cattle, if tick burdens are as low as only five ticks per host per day, or if tick lifespans are as short as 7 d. A host recovery rate of 30 d results in over 50% of the cattle becoming infected with heartwater. Our analyses indicate that it is quite difficult to prevent the establishment and maintenance of high levels of heartwater in a herd, thereby supporting previous suggestions that any attempts at controlling this disease through stringent tick control regimens are not warranted.  相似文献
