The level ofFusarium dry-rot resistance found in important potato varieties has been improved with agronomically acceptableSolanum tuberosum genotypes. Three breeding selections, A74114-4, A76260-16, and BR6316-7 were highly resistant to the two commonFusarium dry-rot species (F. roseum Lk ex Fr. f. sp.sambucinum [Fs], andF. solani var.coeruleum (Sacc), Booth [Fc]). Resistance was determined separately for each of these species. Resistance to Fs was genetically distinct from resistance to Fc since there was no correlation between Fs and Fc reaction for 180 progeny clones from six crosses representing all combinations of resistance. Resistance to each of theseFusarium species was readily transmitted to progeny. Seventy-five percent of the progeny from resistant X resistant parents were also resistant to Fs, while 95% of the progeny from susceptible X susceptible parents were susceptible. Mixed infections withErwinia atroseptica and eitherFusarium species resulted in a qualitative change from a resistant reaction to a susceptible reaction. Bacterial interaction withFusarium spp. is, therefore, an important consideration in determining storage-rot resistance.  相似文献

Dry rot is a serious potato disease and causes significant losses in China. Research efforts on potato dry rot have been very limited as well as the attempts to characterize the pathogen in the major potato production regions of China. A total of 260 Fusarium isolates were identified in 698 potato tubers collected in six important potato production regions of northern China, out of which five different Fusarium species, Fusarium sambucinum, F. avenaceum, F. oxysporum, F. equiseti, and F. acuminatum, were isolated. The identification of each species was confirmed by sequencing analysis of ∼700 bp DNA fragment derived from the translation elongation factor-1 alpha gene. F. sambucinum was found to be the predominant species accounting for 56% of the isolates. Different pathogenicity was found to be associated with five most common Fusarium species. Sixty-seven clones used in China were identified as susceptible to F. sambucinum, indicating little scope for developing resistant cultivars using the currently available potato germplasm.  相似文献

Fusarium sambucinum is one of the most important causal agents that not only cause the dry rot disease of potato tubers in fields and stores worldwide but also capable of producing secondary metabolites toxic for people and animals. Here we present the first draft genome sequence of the strain (F-4) estimated to be around appx. 42.0 Mb. The genome has 12,845 protein coding genes with more than 35,900 exons and gene density of 3.13 per 10Kb. F. sambucinum is evolutionary more close to the F. graminearum among the Fusarium species complex. The genome sequence represents a valuable resource for understanding the pathogenecity and virulence factors, and their evolution within the complex and highly plastic genus Fusarium.  相似文献

Progeny tubers from seed potatoes originating from either a traditional clonal selection method or the more modernin vitro tissue culture method of propagation were tested for storage rot response following inoculation of three tuber sites with twoFusarium species. Significant differences were found among disease responses for the twoFusarium species and for the four cultivars tested. Disease symptoms were less severe forF. solani var.coeruleum than forF. sambucinum. Fusarium sambucinum caused less disease in Superior than Kennebec, Russet Burbank and Sebago. Seed propagation method did not significantly affect disease response except for one case;in vitro culture of Kennebec had less disease due toF. sambucinum at the tuber side inoculation site than clonal selection. Kennebec and Superior inoculatedF. sambucinum had significant differences with respect to the number of years of field propagation for the tuber side inoculation site and for the tuber mean rot index. Significant differences in number of years of field propagation were also found for eye-ends of Superior tubers and stem ends of Russet Burbank tubers inoculated withF. solani var.coeruleum.  相似文献

This paper describes a new source of host-plant resistance to the Colorado Potato Beetle (CPB;Leptinotarsa decemlineata (Say)) in a tetraploid potato (Solanum tuberosum L.) selection, ND2858-1. This hybrid was recorded as a cross betweenS. fendleri ssp.fendleri (A. Gray) and NDSU selection ND1215-1. Field defoliation of this and progeny clones from a subsequent cross to an cultivar was reduced by 50% to 90% in open-choice field screening plots. Progeny of ND2858-1 are vigorous, with high yield potential, and some clones have tuber glycoalkaloid content equivalent to commercial cultivars. These clones are male and female fertile and may be useful to other potato breeding programs. Further characterization of this resistance source is in progress.  相似文献

Wild potato species contain many traits of economic importance. Late blight (LB) resistance and cold chipping are traits desired in potato cultivars. These traits could be co-currently introgressed if they occurred together in wild potato species. Our research objectives were (1) to determine if variation for cold chipping exists between potato species, accessions within species, and plants within accessions all having foliar LB resistance, and (2) to identify wild potato genotypes combining LB resistance and cold chipping. Materials include 665 genotypes from 43 LB-resistant accessions of 12 potato species having Endosperm Balance Numbers (EBN) of 1, 2, and 4, and 59 LB-resistant genotypes retained from these accessions for breeding. Potato chips were made from greenhouse-grown tubers following storage at 4 C for 6 months. Chip color was scored 1–10, ≤ 4 is acceptable by industry standards. Most of the variation for chip color was due to differences between species. Species ranged in the percentage of acceptably chipping genotypes (0% – 67%) with nine of 12 species having cold-chipping genotypes. Appreciable variation was present within accessions as well. The best chipping accessions wereS. verrucosum plant introduction (PI) 161173 – 4.33 / 0.67 (mean / proportion acceptable genotypes),S. stoloniferum PI 250510 -4.36 / 0.64,S. pinnatisectum PI 347766 -4.65 / 0.35 and 275233 -4.73 / 0.44, andS. megistacrolobum PI 195210 -5.14 / 0.29. Eleven 1EBN genotypes fromS. pinnatisectum andS. trifidum and five 2EBN genotypes fromS. verrucosum, S. fendleri,S. stoloniferum, andS. microdontum were identified that combined LB resistance and cold chipping. Co-current introgression would require fewer breeding cycles than other breeding methods to identify hybrid genotypes possessing both traits.  相似文献

Thiabendazole-resistant strainsof Fusarium sambucinum andF. solani var.coeruleum threaten to negate chemical control options for post harvest treatment of Fusarium dry rot. Biological control of dry rot of storage tubers is feasible using bacterial antagonists (25). The impact of yeasts on dry rot has not been investigated. Initial biological control tests employed strains of twenty species of yeasts from the ARS Culture Collection (NRRL). Strain selection was based on strain isolation from plant matter or from environments that would indicate a high likelihood of strain survival in soil. The control potential of these and 29 additional strains isolated from soil adhering to recently harvested tubers was evaluated using a whole Russet Burbank tuber bioassay. At 2xl06 cells/ml, only two unidentified strains andCryptococcus laurentii strain NRRL Y-2536 reduced disease (P=0.05, P=0.10, respectively) while bacterial strainPseudomonas fluorescens 2-79 (NRRL B-15132) was more effective (P=0.01). Conversely,Debaryomyces robertsiae increased disease (P=0.05). No yeast strains significantly controlled disease in a subsequent trial. One of six additional strains ofC. laurentii (P=0.10), none of five strains ofPichia farinosa and neither unidentified strain controlled disease at 5 x 107 cells/ml whereasP. fluorescens again reduced disease (P=0.01). After 6 h, four yeast strains decreased and three increased conidial germination ofF. sambucinum R-6380 though there were no differences after 18 h. Five yeast strains, including two strains ofC. laurentii (NRRL Y-2536, NRRL Y-7139) were marginally effective in controlling disease incited byF. solani var.coeruleum S-1257. Though additional testing may identify yeast strains with considerable promise as biological control agents active against Fusarium dry rot, evidence to date indicates bacterial agents have a greater potential for commercial development.  相似文献

Eleven accessions belonging to six species ofSolanum (S. bulbocastanum,S. chacoense, S. gourlayi, S. microdontum, S. papita, and S.sparsipilum) were tested for resistance to salt. Resistance was measured by survival and growth (dry weight of tops) of plants treated with solutions of NaCl and Na2SO4. The six species could be grouped into four classes based on good to poor resistance to salt solutions (1.S. chacoense, 2.S. gourlayi, 3.S. microdontum, S. sparsipilum, S. bulbocastanum and 4.S. papita). There was a close relationship (r=0.94**) between the survival of plants and growth of surviving plants treated with salt solutions. There was no relationship between known heat or drought resistance of various accessions and salt resistance. Results indicate there is a large genetic base available to improve the salt resistance of the cultivated potato.  相似文献

Wild potato (Solanum sect.Petota) germplasm has been collected in Mexico on nine major expeditions, as determined by 20 collections or more from each expedition currently at the United States potato genebank, the National Research Support Program-6 (NRSP-6). These have resulted in 609 accessions with good collection data. In addition, NRSP-6 has germplasm of approximately 90 other Mexican collections that are unspecific regarding date or place of collection. This expedition was funded to collect those remaining collections with no or little germplasm:Solanum clarum,S. xedinense, S. hintonii, S. hjertingii var.physaloides, S. leptosepalum, S. lesteri, S. macropilosum, S. xmichoacanum, S. xsambucinum, andS. stenophyllidium. In addition, some species and species groups (species groups indicated in parentheses) have unresolved taxonomic problems that needed clarification by additional field collections. These are (S. agrimonifolium and S.oxycarpum), (S. brachycarpum, S. guerreroense, S. hougasii, andS. iopetalum - theS. brachycarpum complex), (S. fendleri, S. papita, S. stoloniferum - theS. stoloniferum complex),S. leptosepalum, andS. macropilosum. We conducted a wild potato germplasm collecting expedition in Mexico from August 22 to October 31, 1997. Our 103 collections, 71 as germplasm collections, provide the first germplasm samples forS. hjertingii var.physaloides,S. leptosepalum, andS. macropilosum. They provide additional germplasm of the rare speciesS. clarum, S. xedinense, S. lesteri, S. xmichoacanum, S. xsambucinum, andS. stenophyllidium. We additionally gathered germplasm and field data to help resolve taxonomic difficulties inS. agrimonifolium andS. oxycarpum, theS. brachycarpum complex, and theS. stoloniferum complex.  相似文献

The recent US-8 clonal lineage ofPhytophthora infestans, the pathogen causing late blight (LB) disease, is one of the most widespread and difficult to control. Sources of foliar resistance to US-8 were evaluated in 1927 seedling genotypes by sampling 49 plant introductions (PIs) representing 13 Mexican and South AmericanSolanum species having an Endosperm Balance Number (EBN) of 1, 2, or 4, and one species with an unknown EBN. Species evaluated were 2x(1EBN)S. bulbocastanum, S. cardiophyllum, S. commersonii, S. pinnatisectum, andS. trifidum; 2x(2EBN)S. berthaultii, S. megistacrolobum, S. microdontum, andS. verrucosum; 4x(2EBN)S. fendleri andS. stoloniferum; 6x(4EBN)S. guerreroense;2x unknown EBNS. polyadenium; as well as4x S. tuberosum families as controls. Foliar resistance in the field was monitored in 1998 and 1999. Resistance differed relative to species geographic origin, EBN, species themselves, PIs within a species, and genotypes within a PI. In 1998 Mexican germplasm had a lower area under the disease progress curve (AUDPC, 1140) and greater resistance than South American germplasm (1601), while the 1EBN (1119) and unknown EBN (1075) species had greater resistance than 2EBN (1470) and 4EBN (1888) species. AUDPC ranged among species from 555 (S. cardiophyllum) to 1932 (S. megistacrolobum) and among PIs within a species most widely forS. bulbocastanum (267–1007) and least widely forS.polyadenium (1019–1179). In 1999 LB symptoms were not observed on the species before an early freeze; however, all 1999S. tuberosum families showed LB symptoms and segregated for resistance, pointing to greater resistance in the species than in cultivated germplasm. Resistance found in diploid 2EBN germplasm is more accessible due to greater crossability with cultivated germplasm; however, though more difficult to introgress, resistance found in some of the more reproductively isolated 1EBN germplasm is worth pursuing.  相似文献

Most wild potato germplasm in genebanks is collected, preserved, and evaluated as botanical seed populations that may be highly heterozygous and heterogeneous. However, some species are selfers so potentially very homozygous, perhaps also homogeneous. Intrapopulation heterogeneity increases sampling error that can undermine consistency in seed regeneration in the genebank, screening results, germplasm collecting, and estimates of taxonomic relationships. Thus, knowledge of genetic heterogeneity (GH) can predict the need to commit more resources for larger sample sizes or replication when populations of a given species are being regenerated, evaluated, collected, and classified. This study investigated withinpopulation GH in 32 potato populations comprising four different breeding systems observed inSolatium species:S. fendleri (2n=4x=48, disomic selfer),S. jamesii (2n=2x=24, outcrosser),S. sucrense (2n=4x=48, tetrasomic outcrosser), andS. verrucosum (2n=2x=24, selfer). RAPD markers were used to estimate heterogeneity among 24 individuals per population. Populations ofS. verrucosum were quite homogeneous with an average GH of 6.0%. Similarly low heterogeneity was detected among the eight populations ofS. fendleri (average GH=7.1%). In contrast,S. jamesii andS. sucrense had a much higher GH of 29.4% and 44.1%, respectively. These results demonstrate and quantify the great difference in intrapopulation heterogeneity among wild potato species. Calculations based on intrapopulation heterogeneity indicate that samples should be composed of 25 to 30 random plants for low sample variation that is uniform for all species.  相似文献

Resistance to Columbia Root-knot nematode was discovered in three wild species in a polyploidy series, Solanum bulbocastanum (2n?=?2x), .S. fendleri (2n?=?4x), and S. hougasii, 2n?=?6x, respectively. After several generations of introgression into the cultivated gene pool, a single dominant monogene conditioning resistance to root propagation of race 1 of CRKN was theorized to be acting in breeding lines derived from each species. Coincidently, sequence tagged sites (STS) and cleaved amplified polymorphic site (CAPS) markers that associated with resistance with S. bulbocastanum were found to be informative in other segregating breeding populations regardless of the source of the gene. The utility of the markers across ploidies suggests that the original gene and genome of S. bulbocastanum or a modern derivative, is present in S. fendleri and S. hougasii. In this polyploidy series, knowledge that the S. bulbocastanum genome is the original source of useful genes guides future germplasm surveyors to search first and most intensively in the primitive Mexican diploid species that may have served as the contributor of a genome to descendants harboring this genome at higher ploidies.  相似文献

A representative sample of the US potato germplasm collection was screened in the greenhouse for ability to accumulate tuber calcium. Germplasm with very high and very low calcium accumulation capacity has been identified by progressively screening for extremes among species, among populations (accessions), and finally among genotypes (i.e., fine screening). Among accessions, the bestS. gourlayi andmicrodontum populations averaged more than six-fold greater tuber calcium than the poorestS. kurtzianum population. When screening was performed at the genotype level, the best individuals ofgourlayi andmicrodontum had over eight-fold greater calcium than the poorestkurtzianum individual. When grown in the field, calcium levels ofS. microdontum selections were confirmed to be significantly higher thantuberosum cultivars. When selected genotypes were intercrossed and their progeny were compared to their original populations, mean tuber calcium was significantly changed in the intended direction of selection forS. kurtzianum (made lower) andS. microdontum (made higher). These stocks are expected to be useful tools for study of the genetic and physiological bases of tuber calcium accumulation, refining screening methods, and breeding improved cultivars.  相似文献

The rate of selfing in the wild, tuber-bearing speciesS. gourlayi Hawkes (2n = 4x = 48) andS. oplocense Hawkes (2n = 6x = 72) was estimated from progeny of plants nulliplex for the “parallel spindles (ps) allele. The estimated values for plants grown in experimental plots were 22% forS. gourlayi and 1% forS. oplocense. The quantitative method used appears to be a simple and efficient way to calculate selfing rates in polyploids. Knowledge of selfing rates is useful for basic population genetic studies and for plant breeding and seed production of economic crops. There is a need for genetic markers inSolanum species if selfing rates are to be taken into consideration for the selection of an appropriate breeding method for the production of true potato seed.  相似文献

Blue light fluorescence microscopy was found to be a quick and easy procedure for assessing pollen tube development in interspecific pollinations ofSolanum. Based on penetration of pollen tubes to the base of the style, several combinations of green-peach-aphid-resistant species and potential bridging species to S.tuberosum ssp.tuberosum should be considered for embryo culture. Sterile or highly infertile hybrids were obtained between green-peach-aphid-resistantS. tuquerrense andS. tuberosum ssp.tuberosum and are described. Slightly fertile hybrids were obtained betweenS. tuquerrense andS. polytrichon, a species compatible withS. tuberosum ssp.tuberosum.  相似文献

Resistance to both thiabendazole (TBZ) and thiophanate-methyl (TPM) was observed in Canadian isolates ofFusarium sambucinum andHelminthosporium solani recovered from diseased potato tubers. Resistance was not found in isolates ofFusarium avenaceum, Fusarium culmorum, Fusarium equiseti andFusarium solani. A high level of resistance to both TBZ (EC50 180 to ?500 mg/l) and TPM (EC50 ?500 mg/l) was observed forH. solani isolates, whereasF. sambucinum exhibited a relatively low level of resistance to TBZ (EC50 34 to 71 mg/l) and a high level of resistance to TPM (growth uninhibited by 2,500 mg/l). A joint Danish, Mexican, United States wild potato (Solanum sect.Petota) germplasm collecting expedition was conducted in Mexico between Aug. 21-Oct. 20, 1988. The trip resulted in 93 seed and 25 tuber collections of 18 species and one putative natural hybrid,S. xmichoacanum. Rare species collected include:Solanum darum,S. hintonii,S. lesteri, andS. xmichoacanum. First germplasm collections were made of the disjunct populations ofS. fendleri from Baja California. Greenhouse assays were carried out to characterizeSolanum accessions previously reported to be late blight resistant and to identify individuals within the accessions with high levels of resistance to late blight. Foliage of wildSolanum species accessions or hybrid cultivated potato x wild species (diploid or tetraploid) from Mexico, Russia, or South America was inoculated withPhytophthora infestans (US-8) and was scored for severity of foliar symptoms at 7 and 14 days after inoculation (DAI). Mexican accessions (S. brachycarpum,S. pinnatisectum,S. guerreroense, andS.fendleri) were significantly more resistant (1.1% infection at 14 DAI) than either Russian (59.1% infection at 14 DAI) or South American (53.4% infection at 14 DAI) accessions. Moreover, the genotypes within the Mexican accessions tended to be more uniformly resistant whereas genotypes within the Russian and South American accessions tended to segregate for resistance. The more resistant genotypes of Russian and South American accessions (176/546) were retested, and 56 genotypes were identified as having potential for use in potato breeding programs for resistance to late blight. Fifty percent of the selected genotypes were fromS. microdontum accessions PI498124, PI595510, and PI595511. The potential of these species for incorporation into breeding programs for late blight resistance is discussed.  相似文献

Volatile metabolites from ‘Russet Burbank’ potatoes inoculated withErwinia carotovora ssp.carotovora (ECC),Erwinia carotovora ssp.atroseptica (ECA), andFusarium sambucinum (FSA) were analyzed by sampling the headspace at 3 and 6 days after inoculation and then using a gas chromatograph/mass spectrometer (GC/MS) to identify the compounds. Non-wounded noninoculated and wounded non-inoculated tubers served as checks. Compounds with an abundance of ≥105 and with frequency of ≥3 out of 20 replicates (10 replicates × 2 incubation times) were subjected to further analysis. A total of 81 volatile metabolites were detected, of which 58 were specific to one or common to a few, but not to all inoculations/diseases. Acetic acid ethenyl ester was unique to ECA, while cyclohexene, diazene, and methoxy-(1,1-dimethyl-2-dihydroxy-ethyl)-amine were unique to ECC, and 2,5-norbornadiene and styrene were unique to FSA. Several metabolites were common only to tubers inoculated with ECC and ECA and were not detected in fungus-inoculated or in control tubers. High abundances of acetone and butane were detected in ECC- and ECA-inoculated tubers, respectively. The possible use of differences in volatile metabolic profiles to discriminate diseases of potato tubers in storage is discussed.  相似文献

AC Belmont is adapted as a second early cultivar in Eastern Canada. It has a short dormancy. The white-skinned round oval tubers size and bulk early, but more similar to Superior than to Jemseg. The specific gravity of AC Belmont is lower than Superior. When boiled, the tubers of AC Belmont have a good, somewhat moist texture; when baked the texture of AC Belmont is relatively dry. AC Belmont is highly resistant toVerticillium albo-atrum, moderately resistant to PVY,Fusarium coeruleum, F. sambucinum f.6. andRhizoctonia solani.  相似文献
