The presence of lignin within biomass impedes the production of liquid fuels. Plants with altered lignin content and composition are more amenable to lignocellulosic conversion to ethanol and other biofuels but may be more susceptible to insect damage where lignin is an important resistance factor. However, reduced lignin lines of switchgrasses still retained insect resistance in prior studies. Therefore, we hypothesized that sorghum lines with lowered lignin content will also retain insect resistance. Sorghum excised leaves and stalk pith Sorghum bicolor (L.) Moench (Poales: Poaceae) from near isogenic brown midrib (bmr) 6 and 12 mutants lines, which have lowered lignin content and increased lignocellulosic ethanol conversion efficiency, were examined for insect resistance relative to wild-type (normal BTx623). Greenhouse and growth chamber grown plant tissues were fed to first-instar larvae of corn earworms, Helicoverpa zea (Boddie) and fall armyworms Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), two sorghum major pests. Younger bmr leaves had significantly greater feeding damage in some assays than wild-type leaves, but older bmr6 leaves generally had significantly less damage than wild-type leaves. Caterpillars feeding on the bmr6 leaves often weighed significantly less than those feeding on wild-type leaves, especially in the S. frugiperda assays. Larvae fed the pith from bmr stalks had significantly higher mortality compared with those larvae fed on wild-type pith, which suggested that bmr pith was more toxic. Thus, reducing lignin content or changing subunit composition of bioenergy grasses does not necessarily increase their susceptibility to insects and may result in increased resistance, which would contribute to sustainable production.

Toxoneuron nigriceps Viereck (Hymenoptera: Braconidae), a koinobiont endoparasitoid of the tobacco budworm, Heliothis virescens F. (Lepidoptera: Noctuidae), derives nutrition from the host hemolymph during the internal portion of its larval development but feeds destructively on host tissues externally after egression. To investigate the importance of this tissue-feeding phase, and to evaluate the behaviors associated with postegression feeding, T. nigriceps larvae were subjected to one of four treatments: 1) allowed to carry out normal tissue feeding, 2) deprived of tissue feeding, 3) presented with tissues scraped away from the host remains, and 4) fed tissues scraped from an unparasitized H. virescens larva. Additionally, total carbohydrates, lipids, and proteins were quantified from pre and posttissue feeding T. nigriceps larvae to examine the effect of postegression feeding on parasitoid nutritional physiology. Parasitoids that received no tissues after egression, or that received tissue from an unparasitized H. virescens larva, had significantly smaller body masses at all stages than those allowed to feed naturally or fed tissues scraped from a parasitized host. Parasitoids that underwent normal host feeding after egression also reached larger masses then those fed scraped host tissue. Parasitoids that received no tissue after egression survived to adulthood significantly less often than those that were presented with any H. virescens tissue. This suggests that postegression tissue feeding is a vital developmental step for T. nigriceps, and that T. nigriceps will not only feed when normal postegression behavior is disrupted, but will also feed on unparasitized tissue. The quantification of macronutrients in the tissues of pre and posttissue feeding T. nigriceps larvae showed significantly elevated proportions of proteins, lipids, and carbohydrates in the tissues of larvae that had completed feeding, with the greatest difference being in total lipids.

This study aimed to evaluate the feeding responses and digestive proteolytic and amylolytic activity of Helicoverpa armigera (Hübner) on 11 corn (Zea mays L.) hybrids at 25 ± 1°C, 65 ± 5% relative humidity (RH), and a photoperiod of 16:8 (L:D) h. The fourth- and fifth-instar larvae fed on hybrid K47*K19 had the highest weight of food consumption and those reared on hybrid KSC705 had the lowest value of food consumption. The highest weight gain of the larvae was observed when H. armigera were fed hybrid KLM78*MO17 and lowest when they were fed hybrids K36 * MO17, KSC705, and K35 * K36. Pupal weight of H. armigera was heaviest when larvae were fed hybrid K47*K19 and lightest when they were fed hybrid KSC705. The highest proteolytic activity of the fourth-instar larvae was observed when they were fed hybrid KSC705, and the lowest activity was observed when they were fed hybrid K47*A67. Fifth-instar larvae that fed on hybrid K47*K19 showed the highest proteolytic activity. Fourth-instar larvae that fed on hybrid K36*MO17 showed the highest amylase activity. The fifth-instar larvae fed on hybrid K47*A67 showed the maximum amylase activity and those reared on the K48*K18 showed the minimum activity. Our results indicated that K36 * MO17, KSC705, and K48 * K18 were the most unsuitable hybrids for feeding H. armigera.

Newly hatchedLeptinotarsa decemlineata larvae were less likely to initiate feeding onSolanum berthaultii than onS. tuberosum. Among larvae that fed there was no difference in the time to initiation of feeding; however, the duration of the initial feeding bout was less onS. berthaultii than onS. tuberosum. Larvae spent less time feeding and more time walking onS. berthaultii than onS. tuberosum. All larvae caged onS. tuberosum fed within 24 hours, but larvae were less likely to feed on a F6 S. tuberosum XS. berthaultii hybrid and least likely to feed onS. berthaultii. Lack of feeding during the first 24 h was highly correlated with mortality at 72 h. Whether or not larvae feed within the first five minutes of being placed on a leaflet is a reliable predictor of larval mortality at 72 h and is a useful test for determining the susceptibility of plants to feeding byL. decemlineata. The amount of trichome exudate accumulated on the tarsi over a 72 h period is correlated with resistance. Resistance ofS. berthaultii to neonateL. decemlineata is characterized by a refusal to initiate feeding and a reduced feeding rate by larvae that do feed.

Viable biocontrol agents for mosquito control are quite rare, therefore improving the efficacy of existing biological agents is an important study. We need to have a better understanding of the predation-risk behavioral responses toward prey. This research examined prey choices by Toxorhynchites splendens by monitoring the behavioral responses of Aedes aegypti, Aedes albopictus, and Anopheles sinensis larvae when exposed to the predator. The results show that Tx. splendens prefers to consume Ae. aegypti larvae. The larvae exhibited different behavioral responses when Tx. splendens was present which suggest vulnerability in the presence of predators. “Thrashing” and “browsing” activities were greater in Ae. aegypti larvae. Such active and risky movements could cause vulnerability for the Ae. aegypti larvae due to increasing of water disturbance. In contrast, Ae. albopictus and An. sinensis larvae exhibited passive, low-risk behaviors, spending most of the time on the “wall” position near the edges of the container. We postulated that Ae. aegypti has less ability to perceive cues from predation and could not successfully alter its behavior to reduce risk of predation risk compared with Ae. albopictus and An. sinensis. Our results suggest that Tx. splendens is a suitable biocontrol agent in controlling dengue hemorrhagic vector, Ae. aegypti.

The aim of this study was to evaluate the effectiveness of the entomopathogenic fungi (EPF), Beauveria bassiana (Bals.) Vuill. (Deuteromycotina: Hyphomycetes) strain PPRI 5339 [BroadBand, an emulsifiable spore concentrate (EC) formulation] and Metarhizium anisopliae (Metsch.) Sorokin (Hypocreales: Clavicipitaceae) strain F52 [Met52, both EC and granular (GR) formulations] against the larvae of Polyphylla fullo (L.) (Coleoptera: Scarabaeidae). Larvicidal bioassays were performed in foam boxes (100 by 75 by 50 cm; length by width by height), containing moist soil medium with some humus and potato tubers as food. Although the B. bassiana product (min. 4 × 109 conidia/ml) was applied at 100, 150, and 200 ml/100 l water; M. anisopliae strain F52 was applied at 500, 1,000, and 1,500 g/m3 of moist soil medium for GR (9 × 108 cfu/g) and 75, 100, and 125 ml/100 l water for EC (5.5 × 109 conidia/ml) formulation. Both fungi were pathogenic to larvae of the pest; however, young larvae (1st and 2nd instars) were more susceptible to infection than older ones (3rd instar). Mortality rates of young and older larvae varied with conidial concentration of both fungi and elapsed time after application. The B. bassiana product was more effective than both of the formulations of the M. anisopliae product, causing mortalities up to 79.8 and 71.6% in young and older larvae, respectively. The highest mortality rates of young and older larvae caused by the M. anisopliae product were 74.1 and 67.6% for the GR formulation, 70.2 and 61.8% for the EC formulation, respectively. These results may suggest that both fungi have potential to be used for management of P. fullo.

Cotton bollworm (Heliothis armigera) is a major pest of cotton and other crops. It is important to understand the mechanisms of insecticide tolerance of cotton bollworm on cotton cultivars with host plant resistance to this insect pest. The objectives of this study were to investigate the effects of cotton pigment glands and their gossypol on the growth, development and insecticide tolerance of cotton bollworm. Three pairs of cotton isogenic lines with glanded versus glandless leaves, as well as artificial diets with 5 levels of gossypol, were used to raise cotton bollworm larvae for five generations. The growth, development and insecticide tolerance of larvae were studied. The results indicated that the cotton pigment glands and higher levels of gossypol resulted in a significant decrease in larval weights and moth eclosion rates and delayed the development of larvae and pupae. Larvae that fed on glanded cotton leaves were significantly more tolerant to two insecticides, cyhalothin and monocrotophos, than those fed on glandless cotton leaves. LD50 values were only increased where they were in amount per unit body weight, and not where were calculated in amount per individual. Also the insecticide tolerance of cotton bollworm larvae increased as the gossypol content was raised from 0 to 0.225% in artificial diets. Meanwhile, the activities of two detoxifying enzymes, carboxylesterase and glutathione s-transferase, were significantly higher in the larvae fed on glanded cotton leaves than those fed on glandless cotton leaves. The activities of two enzymes also increased greatly with the increase of gossypol content from 0 to 0.225% in artificial diets. Across 5 generations of feeding and investigation, the significant inhibition effect on larval growth and larval tolerance to two pesticides were found to be only associated with the feeding by current generation, but were not related to previous diets. The activities of two detoxifying enzymes in larvae were also not enhanced significantly when they were fed continuously on glanded cotton leaves or artificial diet with high gossypol. These results indicated that pigment glands and higher levels of gossypol not only inhibited the growth of cotton bollworm larvae but also enhanced their insecticide tolerance. However, the inhibition effect and enhanced insecticide tolerance were inducible but could not be accumulated or inherited. These results will help us better understand the interaction and co-evolution of insecticide tolerance in larvae of cotton bollworm and host chemical components, and also has provided useful information on cotton bollworm management in cotton production, especially in glandless cotton.

The minute pirate bug Orius insidiosus (Say) is an important predator in mango agroecosystems. It attacks several species of thrips, particularly Frankliniella invasor Sakimura, which is considered a species of economic importance in mango. We investigated the effect of six diets on the development, survival, and reproduction of O. insidiosus: 1) first instars of F. invasor, 2) second instars, 3) adults, 4) pollen, 5) pollen plus thrips larvae, and 6) water. Individuals fed on thrips larvae, with or without pollen, completed their immature development significantly faster. Nymphs of O. insidiosus were able to complete their development feeding on pollen only, while individuals that received water as a diet were unable to reach the adult stage. The highest intrinsic growth rate was obtained when O. insidiosus were fed on pollen plus thrips larvae, and the lowest when individuals were fed on thrips adults. Our studies show that a diet of pollen plus F. invasor larvae is optimal for O. insidiosus development and population growth.

The effect of the whole methanol extracts of five Chrysanthemum species on feeding and performance of Spodoptera littoralis (Boisduval) larvae has been investigated in vitro. The extracts exhibited an anti-feeding and phagostimulating activities against cotton leafworm larvae when applied either on leaf discs or incorporated into an artificial diet. Under chosen conditions, the antifeedant index calculated over 24 h for sixth instar larvae significantly varied from 78.55 for Chrysanthemum segetum L. to −44.18 for Chrysanthemum fuscatum Desf. at the dose of 1000 ppm. Toxicity of the extracts was manifested by a high mortality, reduced growth rates, and low weight gain by larvae fed on diets containing 1000–10,000 ppm of the extract. Anyone of the larvae treated with Chrysanthemum macrotum (D.R.) Ball. leaves crude extract survived to pupation at the two higher concentrations. The time to pupation increased for Chrysanthemum grandiflorum flowers crude extract from 11.40 ± 0.93 to 28.93 ± 10.92 days as the extract concentration in the diet increased from 0 to 10,000 ppm. The ingestion of crude extract by the third instar larvae reduced significantly the consumption, growth and utilisation of the ingested and digested food, and reduced digestibility.

The cotton bollworm Helicoverpa armigera (Hubner) is one of the most destructive pest insects in Iran and many other countries. In this study, lethal and sublethal effects of methoxyfenozide, and thiodicarb were evaluated against H. armigera larvae that fed on insecticide-treated artificial diet. The effects of methoxyfenozide and thiodicarb were assessed in 3rd instars. Methoxyfenozide and thiodicarb showed LC50 values of 4 and 639 mg a.i./ml, respectively, in H. armigera larvae. Sublethal effects on development, adult longevity, and reproduction were observed in H. armigera larvae that survived exposure to an LC30 of the insecticides. Larvae that were exposed to an LC30 concentration of methoxyfenozide exhibited lower pupal weight and increased larval and pupal developmental times compared with thiodicarb treated larvae or control larvae. Adults that were exposed as larvae to an LC30 concentration of methoxyfenozide or thiodicarb showed reduced fecundity (35% and 30%, respectively) compared to control adults. The tested insecticides significantly reduced adult longevity. The longevity of adult females that as larvae were treated with an LC30 concentration of methoxyfenozide or thiodicarb was reduced by 28% and 23%, respectively, in comparison to control females. We predict that the combination of lethal and sublethal effects of the insecticides, especially methoxyfenozide, will induce significant effects on field population dynamics of H. armigera.

Four neem-based insecticides, Neemix® (0.25% EC @ 20 mg azadirachtin/liter), Ecozin® (3% EC @ 20 mg azadirachtin/liter), Agroneem® (0.15% EC @ 4.8 mg azadirachtin/liter) and Neem oil (0.25% EC azadirachtin @ 20 mg azadirachtin/liter) and a non-commercial neem leaf powder, were evaluated for oviposition deterrence, antifeedant effect to larvae and toxicity to eggs and larvae of Pieris brassicae (Linn.) on cabbage leaves in the laboratory. The concentrations tested were with in the ranges of recommended field rates. Oviposition deterrence in no-choice, two-choice and six-choice assays, was observed for all the treatments. They exhibited significant (P < 0.01) oviposition deterrence on P. brassicae when compared with a non-treated control. Cabbage leaves treated with the neem-based insecticides were used as an egg-laying substrate. Numbers of eggs oviposited by P. brassicae adults on treated cabbage leaves were significantly lower then those treated with water, but no significant differences were detected among the neem insecticides. They also deterred feeding by Pieris larvae and exhibited significant antifeedant effects. Larvae of P. brassicae on treated leaves stopped feeding and dropped from the leaf, resulting in no or minimal damage. Direct contact with neem-based insecticides decreased the survival of eggs. Survival of larvae fed for 9 days on leaves treated with neem-based insecticides was reduced to 51%, 49%, 48%, 24% and 18% in the Neem oil, Neemix, Agroneem, Ecozin and neem leaf powder treatments, respectively. Therefore, it can be concluded from the present investigations that neem-based insecticides had oviposition deterrence, antifeedant and toxic effect to P. brassicae.

The cotton bollworm Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) is a widespread pest of many cultivated and wild plants in Europe, Africa, Asia, and Australia. In 2013, this species was reported in Brazil, attacking various host crops in the midwestern and northeastern regions of the country and is now found countrywide. Aiming to understand the effects of different host plants on the life cycle of H. armigera, we selected seven species of host plants that mature in different seasons and are commonly grown in these regions: cotton (Gossypium hirsutum, “FM993”), corn (Zea mays, “2B587”), soybean (Glycine max, “99R01”), rattlepods (Crotalaria spectabilis), millet (Pennisetum glaucum, “ADR300”), sorghum (Sorghum bicolor, “AGROMEN70G35”), and cowpea (Vigna unguiculata, “SEMPRE VERDE”). The development time of immatures, body weight, survivorship, and fecundity of H. armigera were evaluated on each host plant under laboratory conditions. The bollworms did not survive on corn, millet, or sorghum and showed very low survival rates on rattlepods. Survival rates were highest on soybean, followed by cotton and cowpea. The values for relative fitness found on soybean, cotton, cowpea, and rattlepods were 1, 0.5, 0.43, and 0.03, respectively. Survivorship, faster development time, and fecundity on soybean, cotton, and cowpea were positively correlated. Larger pupae and greater fecundity were found on soybean and cotton. The results indicated that soybean, cotton, and cowpea are the most suitable plants to support the reproduction of H. armigera in the field.

Enset root mealybug, Cataenococcus ensete Williams and Matile-Ferrero, (Homoptera: Pseudococcidae) is an economically important pest on enset. Adults and nymphs of overlapping generations feed on the corms and roots of enset and are difficult to control once established. Isolates of Beauveria bassiana (Balsamo) Vuillemin and Metarhizium anisopliae (Metschnickoff) Sorokin that originated from Ethiopia were evaluated for their efficacies against enset root mealybugs under laboratory, pots and field conditions. Of the tested isolates, two strains (FF and PPRC-56) of B. bassiana were found to be highly pathogenic to adults, causing 97 and 100% mortality, respectively 20 days after inoculation under laboratory conditions. On the other hand, the isolates PPRC-56, FF, PPRC-6 and Mm induced mean mortalities of 97, 95, 96 and 83%, respectively, to the adult enset root mealybugs on the pot experiment, and 51.33, 38.67, 29.33 and 19.33% mortality at the first site and 54.00, 42.67, 32.00 and 25.33% at the second site, respectively, in field experiments. The study showed B. bassiana and M. anisopliae can be used in the integrated management of enset root mealybug.

A field survey was conducted in Ataulfo mango (Mangifera indica L.) orchards in Chiapas, Mexico, with the objective of determining the natural enemies of the Frankliniella complex species (Thysanoptera: Thripidae). Seven species of this genus feed and reproduce in large numbers during the mango flowering. Two representative orchards were selected: the orchard “Tres A” characterized by an intensive use of agrochemicals directed against thrips, and the orchard “La Escondida” that did not spray insecticides. During mango flowering, five inflorescences were randomly collected every 5 d in both orchards, for a total of 18 sampling dates. Results revealed the presence of 18 species of arthropods that were found predating on Frankliniella. There were 11 species in the families Aeolothripidae, Phlaeothripidae, Formicidae, Anthocoridae and Chrysopidae; and seven species of spiders in the families Araneidae, Tetragnathidae, and Uloboridae. Over 88% of predators were anthocorids, including, Paratriphleps sp. (Champion), Orius insidiosus (Say), Orius tristicolor (White), and O. perpunctatus (Reuter). The orchard that did not spray insecticides had a significantly higher number of predators suggesting a negative effect of the insecticides on the abundance of these organisms.

Bradysia odoriphaga Yang and Zhang (Diptera: Sciaridae), the Chinese chive root maggot, is a destructive pest of Allium vegetables and flowers that causes severe losses in northern China. Novel biological control technologies are needed for controlling this pest. We identified a new entomopathogenic fungus isolated from infected B. odoriphaga larvae and evaluated the susceptibility of the biological stages of B. odoriphaga and the effects of temperature on fungus growth and pathogenicity. Based on morphological characteristics and molecular phylogeny, the fungus was identified as Mucor hiemalis BO-1 (Mucorales: Mucorales). This fungus had the strongest virulence to B. odoriphaga larvae followed by eggs and pupae, while B. odoriphaga adults were not susceptible. A temperature range of 18–28°C was optimum for the growth and sporulation of M. hiemalis BO-1 and virulence to B. odoriphaga larvae. At 3 and 5 d after inoculation with 105 spores/ml at 23°C, the survival rates were 24.8% and 4.8% (2nd instar larvae), respectively, and 49.6% and 12.8% (4th instar larvae), respectively. The potted plant trials confirmed that M. hiemalis BO-1 exerted excellent control efficiency against B. odoriphaga larvae, and the control exceeded 80% within 5 d when the spore concentration applied exceeded 107 spores/ml. In conclusion, these findings supported the hypotheses that this fungus could serve as an effective control agent against B. odoriphaga larvae and is worth being further tested to determine its full potential as a biocontrol agent.

Cardiovascular diseases, especially atherosclerosis, have found to be the dreadful diseases worldwide and therapeutic interventions using plant sources have wide therapeutic value. Vigna unguiculata (VU) leaves have been used as food and therapeutics. Hence, our study was designed to evaluate the hypolipidemic as well as anti-atherogenic potential of VU leaves in normalizing atherogenic gene expression, cholesterol profile, generation of reactive oxygen species (ROS) and antioxidant enzyme system on cholesterol fed rabbit model. For the study New Zealand white rabbits were randomly divided into four groups of six animals each and experimental period was three months; group -i - ND [normal diet (40 g feed)], group-ii- ND (normal diet) +EAVU [ethyl acetate fraction of Vigna unguiculata (150 mg/kg body weight)], group -iii- ND [normal diet ]+ CFD [cholesterol fed diet (cholesterol 1 % of 40 g feed and cholic acid 0.5 % of 40 g feed)] and group-iv - ND [normal diet] +CFD [cholesterol fed diet ]+EAVU [ethyl acetate fraction of Vigna unguiculata (150 mg/kg body weight)]. Atherosclerosis was induced by feeding the rabbit with cholesterol (1 % of 40 g feed) and cholic acid (0.5 % of 40 g feed). Supplementation of EAVU normalized cholesterol profile, generation of reactive oxygen species (ROS), lipid peroxidation products like thiobarbituric acid reactive substance (TBARS), antioxidant system and important genes of cardiovascular diseases like interleukin-10 (IL 10), paraoxanase-1 (PON I), interleukin-6 (IL 6), and cyclooxygenase-2 (Cox 2) to near normal level as compared with normal diet. The result obtained showed the antioxidant as well as anti-atherogenic potential of Vigna unguiculata leaves in ameliorating cholesterol induced atherosclerosis, and thus it is good task to include VU leaves in daily diet for the prevention of cardiovascular diseases especially atherosclerosis.

The internal colonization of Brassica napus plants by Metarhizium anisopliae was demonstrated and the endophytic and direct effects of the fungus against Plutella xylostella larvae were assessed. Internal colonization of the plant by the fungus was shown by re-isolating the fungus from leaves, petioles and stems of the plant that were not the initial site of inoculation. Results indicated that significant differences (at P = 0.05) were obtained in mean % successful re-isolation of the fungus from leaves, petioles and stems at 2, 3 and 4 weeks after inoculation. To assess the endophytic effect of the fungus, larvae of P. xylostella were introduced onto plant leaves distant from the initial site of inoculation with the fungus (one leaf per plant). Results indicated that significant differences (at P = 0.05) were obtained in mean % mortality of larvae that were introduced at 2, 3 and 4 weeks after inoculation compared to the non-inoculated control. Similar significant results were obtained when the direct effect of the fungus against the larvae was assessed. In conclusion, this research provides evidence for the first time of the endophytic action of M. anisopliae against P. xylostella larvae and the ability of the fungus to colonize the internal tissues of B. napus plants.
