Inter-sectional hybrids were successfully obtained by the reciprocal crosses between 11 cultivars (including 6 diploids and 5 tetraploids) of Begonia semperflorens (SS & SSSS genomes) and B. ‘Orange Rubra’ (RR genome) with the aid of in vitro culture of mature or immature seeds on MS medium containing 0.1 mg l−1 α-naphthylacetic acid, 0.1 mg l−1 6-benzyladenine, 10 mg l−1 gibberellic acid, 30 g l−1 sucrose and 2.5 g l−1 gellan gum. Embryo rescue as ovary culture with immature seeds 12th–16th day after pollination (DAP) generally gave higher efficiency of plantlet formation, but in some cross combinations, culture of mature seeds (30 DAP) resulted in higher yield of plantlets. Flow cytometric analysis revealed that they were consisted of the plants with various genomic combinations (RS, RR, RSS, RRS, RRSS and RRRRSS) as estimated by the DNA contents of both parents. Hybridity of these plants with various genomic combinations including RR was confirmed by random amplified polymorphic DNA analysis. These results suggested that unreduced gamete formation and spontaneous chromosome doubling were involved in the hybrid formation of various ploidy levels and genomic combinations. These hybrids showed various levels of intermediate traits between both parents according to the genomic compositions, and some of them had desirable characters of both parents.  相似文献

Starch properties are major determinants of grain quality and food characteristics in rice (Oryza sativa L.). Control of starch properties will lead to the development of rice cultivars with desirable characteristics. We performed quantitative trait locus analysis and detected a putative region on chromosome 2 associated with phenotypic variation of starch properties in two glutinous rice varieties developed in the Hokkaido region of Japan: ‘Kitayukimochi’, which has a low pasting temperature and creates soft rice cakes, and ‘Shirokumamochi’, which has a high pasting temperature and creates hard rice cakes. Starch branching enzyme IIb (SbeIIb) was identified as a candidate gene within the region. Sequence analysis of SbeIIb in parental lines identified two single-nucleotide polymorphisms (SNPs) with non-synonymous mutations in the coding region of the ‘Shirokumamochi’ genotype (SbeIIbsr). We genotyped over 100 rice cultivars, including 28 rice varieties in the Honshu region of Japan, using the CAPS marker, which was designed using one of the SNPs. However, SbeIIbsr was not found in rice cultivars in Honshu. Distribution analysis indicated that SbeIIbsr was introduced to the rice breeding population in Hokkaido from the American variety ‘Cody’ via the Hokkaido cultivar ‘Kitaake’. As a result, SbeIIbsr was distributed only in progenies of ‘Kitaake’.  相似文献

Suitability of wheat (Triticum aestivum L.) for many food products depends on its unique protein. Elevated grain protein content (GPC) and its quality influences the bread making properties of wheat. The objective of this study was to examine the association of elevated GPC with agronomic and end-use quality in two hard red spring wheat recombinant inbred (RI) populations derived from wild emmer (Triticum turgidum L. var. dicoccoides). The two hard red spring populations (ND683/Bergen and Glupro/Bergen) were developed using a single-seed-descent method. ND683 and ‘Glupro’ are high in GPC (180 g kg-1), presumably due to the introgression of gene(s) from Triticum turgidum L. var. dicoccoides and ‘Bergen’ is low in GPC (145 g kg-1). From each of the two populations 12 high- and 12 low-GPC RI lines (F5:7) were selected for replicated testing at two North Dakota (ND) locations in 1995. In both populations, the high-GPC lines had significantly (p < 0.05) higher values compared to the low-GPC lines for mean GPC and water absorption. Mean grain yield of the high-GPC lines was not significantly different from the low-GPC lines in both populations. In the ND683/Bergen population, the high-GPC lines had significantly (p < 0.05) higher values than the low-GPC lines for mean plant height, days to heading, and flour extraction. GPC was significantly (p < 0.05)and negatively associated with test weight and also significantly (p < 0.01) and positively associated with water absorption in the Glupro/Bergen population. In these populations, results suggested that it may be possible to select lines that combine higher GPC and acceptable yield level, but later in maturity and taller in plant height. Resistance to soybean mosaic virus (SMV) is imperative for soybean (Glycine max (L.) Merr.) production in the Tohoku region. Molecular markers for SMV resistance were previously reported for U.S. SMV strains, but they cannot be applied because of the differences in strain classification between Japan and the U.S. A U.S. variety ‘Harosoy’ has been used mainly as a donor of resistance to SMV strains C and D in a Japanese breeding program, resulting in resistant varieties such as ‘Fukuibuki.’ Because ‘Harosoy’ harbors the Rsv3 gene conferring resistance to the virulent SMV strain groups, G5 through G7, it appears that the Rsv3 gene confers resistance to strains C and D. In this study, we introduced resistance to the two strains from ‘Fukuibuki’ into a leading variety ‘Ohsuzu’ by recurrent backcrossing with marker-assisted selection. All lines selected with markers near Rsv3 showed resistance to the strains, suggesting that the Rsv3 locus is responsible for the resistance. Three years of trials showed that one of the breeding lines, ‘Tohoku 169,’ was equivalent to ‘Ohsuzu’ with respect to agricultural characteristics such as seed size, maturity date, and seed yield, except for the SMV resistance.  相似文献

Green stem disorder (GSD) is one of the most serious syndromes affecting soybean (Glycine max) cultivation in Japan. In GSD, stems remain green even when pods mature. When soybean plants develop GSD, seed surfaces are soiled by tissue fluid and seed quality is deteriorated during machine harvesting. We performed quantitative trait locus (QTL) analyses for GSD insensitivity using recombinant inbred lines (RILs; n = 154) derived from a cross between an insensitive line (‘Touhoku 129’) and a sensitive leading cultivar (‘Tachinagaha’) during a 6-year evaluation. Three effective QTLs were detected. The influences of these QTLs were in the following order: qGSD1 (LG_H) > qGSD2 (LG_F) > qGSD3 (LG_L). At these three QTLs, ‘Touhoku 129’ genotypes exhibited more GSD insensitivity than ‘Tachinagaha’ genotypes. The lower incidence of GSD for ‘Touhoku129’ was attributable primarily to these three QTLs because RILs harboring a ‘Touhoku 129’ genotype at the three QTLs exhibited a GSD incidence similar to that of ‘Touhoku 129.’ Although a limitation of this study is that only one mapping population was evaluated, this QTL information and the flanking markers of these QTLs would be effective tools for resolving GSD in soybean breeding programs.  相似文献

Summary Genetic relationships of condensed tannins (CT) with other forage quality parameters have not been adequately studied in birdsfoot trefoil (Lotus corniculatus L.). The objectives were to bidirectionally select for CT concentration in birdsfoot trefoil to determine selection response and to create populations for examination of genetic relationships of CT with forage quality parameters, principally lignin. High-and low-tannin parental clones were selected from NC-83 birdsfoot trefoil germplasm and intercrossed to produce Syn1 populations. Herbage samples, harvested for two years at two locations, were analyzed from high-tannin, low-tannin, and parental populations for CT concentration, herbage yield, and forage quality parameters. The mean condensed tannin concentrations in the high-and low-tannin populations were 69.3±0.8 and 21.1±0.8 g catechin equivalent (CE) kg-1 dry matter (DM) compared with 36.4±0.8 g CE kg-1 DM in the parental population. The selection response exhibited a quadratic relationship, with selection for increased CT concentration more effective than for reduced CT concentration. Compared with the parental population, acid detergent lignin (ADL) was higher, and crude protein (CP) and in vitro digestible dry matter (IVDDM) concentrations were lower for the high-tannin population and the converse was true for the low-tannin population. Environmental sustainability concerns make improving yield under lower N input a desirable breeding goal. To evaluate genetic variation and heterosis for low-N tolerance breeding, 28 F1 hybrids from a diallel scheme, along with their eight parental lines, were tested for agronomic traits including kernel number per ear (KNE) and grain yield per plant (GY), in replicated plots over two years under low-nitrogen (LN, without nitrogen application) and normal-nitrogen (NN, 220 kg N ha−1) conditions. Taken together the heritability in this and our previous studies, the correlation with grain yield, and the sensitivity to the stress for target trait selection, KNE was a good secondary target trait for LN selection in maize breeding. KNE also showed much higher mid-parent heterosis than hundred-kernel weight under both nitrogen levels, particularly under LN, indicating that KNE contributed the majority of GY heterosis, particularly under LN. Therefore, KNE can be used as a positive target trait for hybrid performance prediction in LN tolerance breeding. Our results also suggest that breeding hybrids for LN tolerance largely relies on phenotypic evaluation of hybrids under LN condition and yield under LN might be improved more by selection for KNE than by direct selection for GY per se.  相似文献

The effect of drought on barley leaf epicuticular wax load (EWL), residual transpiration rates (RTR) and grain yield was examined by subjecting 12 barley genotypes to controlled irrigation and terminal water stress conditions. The mean leaf epicuticular wax load was found to be 9% greater in the plants subjected to terminal water stress than in those provided irrigation, while the mean residual transpiration rate of the irrigated plants was 20% higher than in those subjected to water stress. Under these stress conditions, the correlation between grain yield and the epicuticular wax load was positive (P < 0.01), while that between the grain yield and the residual transpiration rate was negative (P < 0.05). Under the water stress conditions, the breeding lines studied showed a greater mean epicuticular wax load than the commercial varieties, while the residual transpiration rate was greater in these varieties than in the breeding lines. The greater epicuticular wax load of the breeding lines favoured their tolerance of drought, improving their yields over those of the commercial varieties.  相似文献

Kenaf (Hibiscus cannabinus L.; Family: Malvaceae), is multipurpose crop, one of the potential alternatives of natural fiber for biocomposite materials. Longer fiber and higher cellulose contents are required for good quality biocomposite materials. However, average length of kenaf fiber (2.6 mm in bast and 1.28 mm in whole plant) is below the critical length (4 mm) for biocomposite production. Present study describes whether fiber length and cellulose content of kenaf plants could be enhanced by increasing GA biosynthesis in plants by overexpressing Arabidopsis thaliana Gibberellic Acid 20 oxidase (AtGA20ox) gene. AtGA20ox gene with intron was overexpressed in kenaf plants under the control of double CaMV 35S promoter, followed by in planta transformation into V36 and G4 varieties of kenaf. The lines with higher levels of bioactive GA (0.3–1.52 ng g−1 fresh weight) were further characterized for their morphological and biochemical traits including vegetative and reproductive growth, fiber dimension and chemical composition. Positive impact of increased gibberellins on biochemical composition, fiber dimension and their derivative values were demonstrated in some lines of transgenic kenaf including increased cellulose content (91%), fiber length and quality but it still requires further study to confirm the critical level of this particular bioactive GA in transgenic plants.  相似文献

Summary Resistance to fusarium wilt, incited by Fusarium oxysporum (Schlecht.) f. sp. lycopersici (Sacc.) Snyder & Hansen race 3 in tomato (Lycopersicon esculentum Mill.) was discovered in LA 716, a L. pennellii accession. A resistant BC1F3 breeding line, E427, was developed from LA 716. E427 was crossed with the susceptible cv. Suncoast and F1, BCP1, BCP2 (to Fla 7155, a susceptible parent) F2, F3, and BCP2S1 seeds were obtained. Segregation for resistance following root dip inoculation over three experiments indicated a single dominant gene controlled resistance. Five of the 12 BCP1S1's segregated more susceptible plants, whereas one of the 12 segregated more resistant plants than expected (P<0.05). Three of 23 F3 lines segregated more susceptible plants than expected while 1 of the 23 had more resistant plants than expected (P<0.05). Segregation in all other lines fit expected ratios. Five of the 23 F3's were homozygous resistant which was an acceptable fit to expectations (P=0.1–0.5). The gene symbol I 3 is proposed for resistance to race 3 of the wilt pathogen. Deviations from expected ratios in data reported here and for other breeding lines indicate an effect of modifier genes and/or incomplete penetrance. The yellowing strain of Soybean dwarf virus (SbDV-YS) causes yellowing and yield loss in common bean (Phaseolus vulgaris). The most effective control is achieved through breeding for resistance. An indeterminate climbing cultivar with a white seed coat, ‘Oofuku’, is resistant to SbDV-YS in inoculation tests. We crossed ‘Oofuku’ with an elite cultivar, ‘Taisho-Kintoki’, which is SbDV-YS-susceptible, determinate dwarf with a red-purple seed coat, and performed amplified-fragment-length polymorphism analysis of F3 lines. From nucleotide sequences of the resistant-specific fragments and their flanking regions, we developed five DNA markers, of which DV86, DV386, and DV398 were closely linked to Sdvy-1, a resistance gene. Using the markers, we developed ‘Toiku-B79’ and ‘Toiku-B80’, the near-isogenic lines (NILs) incorporating Sdvy-1 in the background of ‘Taisho-Kintoki’. The NILs had similar growth habit, maturity date and seed shape to those of ‘Taisho-Kintoki’. The quality of boiled beans was also similar, except that the NILs had more seed coat cracking than ‘Taisho-Kintoki’. The NILs showed no SbDV-YS infection in inoculation tests. We suggest that Sdvy-1 is a useful source of SbDV-YS resistance in common bean.  相似文献

Imisun and CLPlus are two imidazolinone tolerance traits in sunflower (Helianthus annuus L.) determined by the expression of two alleles at the locus Ahasl1. Both traits differed in their tolerance level to imazapyr —a type of imidazolinone herbicide— when aboveground biomass is considered, but the concomitant herbicide effect over the root system has not been reported. The objective of this work was to quantify the root biomass response to increased doses of imazapyr in susceptible (ahasl1/ahasl1), Imisun (Ahasl1-1/Ahasl1-1) and CLPlus (Ahasl1-3/Ahasl1-3) homozygous sunflower genotypes. These materials were sprayed at the V2–V4 stage with increased doses of imazapyr (from 0 to 480 g active ingredient ha−1) and 14 days after treatment root biomass of each plant was assessed. Genotype at the Ahasl1 locus, dose of imazapyr and their interaction significantly contributed (P < 0.001) to explain the reduction in root biomass accumulation after herbicide application. Estimated dose of imazapyr required to reduce root biomass accumulation by fifty percent (GR50) differed statistically for the three genotypes under study (P < 0.001). CLPlus genotypes showed the highest values of GR50, 300 times higher on average than the susceptible genotypes, and almost 8 times higher than Imisun materials, demonstrating that both alleles differ in their root biomass response to foliar application of increased doses of imazapyr.  相似文献

Summary The fatty acid composition of seed-oil of breeding lines and F1 hybrids of Matthiola incana was analyzed, using direct esterification and gas chromatography. The breeding lines tested differed significantly with respect to the levels of palmitic, oleic, linoleic and linolenic acids. Embryonic-stage heterosis in linolenic acid concentration was demonstrated by F1 hybrid seeds, derived from mating horticulturally different lines of M. incana. Linolenic acid content was negatively correlated with both oleic acid content (r=–0.85) and linoleic acid content (r=–0.66). None of the breeding lines or the F1 hybrids significantly passed the limit of 67% linolenic acid. Possible genetic and biochemical explanations for the above phenotypic data are discussed.  相似文献

It has long been known that a bacterial leaf blight-resistant line in rice obtained from a crossing using ‘Asominori’ as a resistant parent also has resistance to blast, but a blast resistance gene in ‘Asominori’ has not been investigated in detail. In the present study, a blast resistance gene in ‘Asominori’, tentatively named Pias(t), was revealed to be located within 162-kb region between DNA markers YX4-3 and NX4-1 on chromosome 4 and to be linked with an ‘Asominori’ allele of the bacterial leaf blight resistance gene Xa1, tentatively named Xa1-as(t). An ‘Asominori’ allele of Pias(t) was found to be dominant and difference of disease severity between lines having the ‘Asominori’ allele of Pias(t) and those without it was 1.2 in disease index from 0 to 10. Pias(t) was also closely linked with the Ph gene controlling phenol reaction, suggesting the possibility of successful selection of blast resistance using the phenol reaction. Since blast-resistant commercial cultivars have been developed using ‘Asominori’ as a parent, Pias(t) is considered to be a useful gene in rice breeding for blast resistance.  相似文献

The appearance of brown rice grown under high temperature conditions is an important characteristic for improvement in Japanese rice breeding programs. We performed a QTL analysis of the appearance quality of brown rice using chromosome segment substitution lines of the indica cultivar ‘Habataki’ in the ‘Koshihikari’ genetic background. A line carrying a ‘Habataki’ segment on chromosome 7 showed a high percentage of perfect grains produced under high temperature conditions during the ripening period. To verify the role of this segment, and to narrow down the region containing the useful allele, substitution mapping was performed using multiple paired lines. As a result, the chromosomal location of a gene that we named Appearance quality of brown rice 1 (Apq1) was delimited to a 48-kb region. In addition, we developed an Apq1-near isogenic line (NIL) to evaluate the effect of Apq1 on various agronomic traits. Under high temperature conditions during the ripening period, the Apq1-NIL produced significantly higher percentages of perfect grains than ‘Koshihikari’. Other agronomic traits, including yield and palatability, were similar between the Apq1-NIL and ‘Koshihikari’. Therefore, the ‘Habataki’ allele of Apq1 will be useful in breeding programs aimed at improving the quality of grains ripened under high temperature conditions.  相似文献

The pea crop (Pisum sativum L.) is a convenient source of plant protein for animal feeding. The objective of this research was to evaluate field pea breeding lines for agronomic performance and seed quality focussed to their use in a sustainable production. Thirty-five field pea breeding lines and six elite cultivars were evaluated for their agronomic value in field in Spain upon 20 traits related to flower, cycle, plant architecture, productivity and seed quality. The lines showed significant differences in all the quantitative traits evaluated and three of them, namely MB-0307, MB-0308, and MB-0319 were chosen to be evaluated, together with the advanced cultivar ZP-1233, in field and in growing chamber for agronomic performance, seed quality and ability for sustainable production. The four accessions displayed high seed protein content that had significant effect of cropping density with averages of 253.6 g kg−1 under low cropping density of 60 plants m−2 and 259.1 g kg−1 under high cropping density of 90 plants m−2, therefore, the low cropping density should be regarded as the most convenient. Average yield of the lines MB-0307, MB-0308 and MB-0319 and the cultivar ZP-1233 was fair (197.5 g m−2), probably due to the absence of fertilizers and irrigation, aiming for the sustainability of the crop. Intercropping with rye and herbicide application resulted in no differences on the seed yield; therefore, the ability of the breeding lines to grow without herbicide seems to be demonstrated, while the germination of the seeds at low temperature was very good. These results indicate that field pea could be a new protein crop in the North of Spain to satisfy the demand of plant protein for animal feed, based upon adapted breeding lines that combine the ability for growing under sustainable conditions with other desirable agronomic traits maintaining an adequate productivity.  相似文献

Four breviaristatum (short awned and semi-dwarf) barley mutants; ari-e.1, ari-e.119, ari-e.156 and ari-e.228 were compared with other semi-dwarf mutants; Golden Promise, Alf, Pallas and Diamant along with their non-mutant parents; Bonus, Foma, Maythorpe, Bomi and Valticky, for response to salt stress. Plants were exposed to hydroponic salt treatments (NaCl at 25 and 175 mol m-3) for 4 weeks, after which response was measured in terms of shoot dry weight, sodium content and δ13C. In general ari-e mutants and Golden Promise had significantly lower Na+ contents than the other mutants. They also had significantly more negative δ13C values than the other lines in stressed (175 mol m-3 NaCl) conditions. There was a positive correlation (r = 0.71, p < 0.01) between shoot Na+ and δ13C values so that δ13C became less negative with increasing Na+ content. Shoot dry weights were compared to shoot Na+ and δ13C values. The ari-e and Golden Promise mutants showed less reduction in dry matter production in salt stress relative to the control treatment than all the other lines. The data suggest that ari-e mutants and Golden Promise are better adapted to salt stressed environments than the other lines examined. Tests for gibberellic acid sensitivity revealed that ari-e mutants and Golden Promise responded weakly to GA3, while other dwarf mutants Pallas, Diamant and Alf along with their parents Bonus, Foma, Maythorpe, Valticky and Bomi were highly sensitive. The level of N fertilization and the content of leaf N in Cynodon dactylon × C. transvaalensis Burtt Davy cv. ‘Tifway 419’ bermudagrass were evaluated non-destructively with a fluorescence-based method. It was applied directly into the field by using the Multiplex portable fluorimeter during two consecutive seasons (2010 and 2011). In the 2010 experiment, the nitrogen balance index (NBI1) provided by the sensor was able to discriminate (at P < 0.05) six different N levels applied, up to 250 kg ha−1, with a precision (root mean square error, RMSE) in the rate estimate of 3.29 kg ha−1. In 2011, the index was insensitive to the N treatment between 150 kg ha−1 and 250 kg ha−1 N rates, and its precision was 39.98 kg ha−1. Calibration of the sensor by using the destructive analysis of turf samplings showed a good linear regression between NBI1 and the leaf N content for both 2010 (R2 = 0.81) and 2011 (R2 = 0.93) experiments. This allowed mapping of the leaf N spatial distribution acquired by the sensor in the field with a prediction error of 0.21%. Averaging the overall estimates of leaf N content per N treatment provided an upper limit of 200 kg ha−1 for the required fertilization, corresponding to a critical level of leaf N of about 2.3%. Our results confirm the usefulness of the new fluorescence-based method and sensor for a precise management of fertilization in turfgrass.  相似文献

利用已开发的面粉色泽相关性状基因的分子标记,检测了182份新疆小麦品种(系)的Psy-A1、Ppo-A1、Ppo-D1、Ta Lox-B1等位变异,并结合表型鉴定结果,分析不同等位变异对面粉色泽的影响。结果表明,单基因等位变异对面粉色泽的影响不同,其中4个基因等位变异L*值的差异均不显著;对a*值的效应,Psy-A1a高于Psy-A1b(P0.01),Ppo-A1b高于Ppo-A1a(P0.05),Ta Lox-B1a高于Ta Lox-B1b(P0.05);Psy-A1a基因型的b*值高于Psy-A1b基因型(P0.01);Psy-A1a基因型的Wht值低于Psy-A1b基因型(P0.01)。说明Psy-A1是影响面粉色泽(红度、黄度和白度)的重要基因,而Ppo-A1和Ta Lox-B1基因的等位变异只对面粉红度有显著影响。4个检测基因共有12种等位变异组合,对a*、b*和Wht值有显著效应(P0.01),但对L*值影响不显著(P0.05);Psy-A1b/Ppo-A1a/Ppo-D1b/Ta Lox-B1b组合具有最高的Wht值和最低的a*、b*值,Psy-A1a/Ppo-A1a/Ppo-D1b/Ta Lox-B1a组合具有最低的Wht值和最高的a*、b*值。182份品种的Wht值平均为86.86,其中审定品种平均为86.87,冬小麦品种为86.42,春小麦品种为87.32。青春5号具有优良的基因型和较高的面粉白度,应加强利用。总体来看,改良新疆小麦面粉的白度,应重点加强对Psy-A1基因的选择,提高Psy-A1b比例。  相似文献
