The effects of various types of anticoagulants on plasma biochemistry have been studied in humans and various animals; however, limited information exists for effects on horse plasma biochemistry. Ten clinically healthy Thoroughbred horses were blood sampled in tubes containing different anticoagulants as well as no anticoagulant. The concentrations of glucose, cholesterol, triglyceride, total bilirubin, blood urea nitrogen (BUN), creatinine, total protein, albumin, calcium, inorganic phosphorus, magnesium and iron, and the activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), creatine kinase (CK), and gamma glutamyl transferase (GGT) were measured. Except for the amounts of inorganic phosphorus, magnesium, and GGT, other measured parameters were significantly lower in citrated plasma than in serum. When corrected for dilution, significant differences were seen for the amounts of glucose, cholesterol, triglyceride, bilirubin, calcium, iron, and the activity of ALT, ALP, and CK in citrated plasma. Most parameters did not show any difference; however, significant decreases in BUN, total bilirubin, ALT, and CK activity were seen when heparin was used as an anticoagulant. When compared with serum, using ethylenediaminetetra-acetic acid (EDTA) as an anticoagulant produced a significant difference in the amounts of measured plasma parameters with the exception of GGT, albumin, creatinine, inorganic phosphorus, and triglyceride.  相似文献

To assess changes in 24 blood constituents in frozen serum and heparinized plasma, blood samples were drawn from 10 clinically normal German Shepherd army dogs. The storage characteristics of nine enzymes (ALP, ALT, amylase, AST, CK, GGT, GLDH, LDH, lipase), and 15 metabolites and minerals (albumin, bile acids, bilirubin, calcium, cholesterol, creatinine, fructosamine, glucose, magnesium, phosphate, potassium, protein, sodium, triglycerides, urea) were studied. Parallel samples of serum and heparinized plasma were stored for 90 and 240 days at two different storage temperatures, -200 degrees C and -700 degrees C. Sixteen of the 24 analytes (ALP, ALT, amylase, AST, CK, GGT, GLDH, LDH, bile acids, calcium, cholesterol, creatinine, fructosamine, magnesium, phosphate, urea) showed statistically significant (p < 0.05) changes during the storage period related to storage time, storage temperature, and sample type. Seven of the analytes (amylase, GGT, GLDH, LDH, bile acids, fructosamine, magnesium) showed changes of possible clinical importance with mean differences from baseline larger than 20% for the enzymes and 10% for the metabolites and minerals during the storage periods.  相似文献

The objective of the study was to determine monthly variations in serum glucose, cholesterol, total protein (TP), urea, albumin, globulin, albumin/globulin ratio, creatinine, aspartate aminotransferase (AST), creatinine kinase (CK), alkaline phosphatase (ALP), calcium, phosphorus and magnesium in Nguni, Bonsmara and Angus beef steers raised on sweetveld. Twenty-five Nguni, 15 Aberdeen Angus and 15 Bonsmara 8-month old steers were studied from June 2006 until March 2007. Across the 9 months, Nguni had higher concentrations of glucose (P =0.019) and cholesterol (P =0.001) than the other two breeds. The overall glucose and cholesterol concentrations in the Nguni were 4 and 2.86mmol/L, respectively. There was a breedxmonth interaction on glucose, cholesterol, creatinine, calcium, albumin and phosphorus concentrations. Breed had no effect on TP, urea, globulin and AST concentrations. Breed and month differences obtained could be attributed to changes in environment temperature and nutrient content of the forage.  相似文献

The stability and storage characteristics of 24 blood constituents from dogs including nine enzymes (ALP, ALT, amylase, AST, CK, GGT, GLDH, LDH, lipase), 15 metabolites and minerals (albumin, bile acids, bilirubin, calcium, cholesterol, creatinine, fructosamine, glucose, magnesium, phosphate, potassium, protein, sodium, triglycerides, urea) were studied. Conditions studied included storing of nonanticoagulated and heparinized whole blood for 3 days (Part A), and storing of serum and heparinized plasma for 3 days (Part B). The storage temperature for both studies was +4 degrees C from day 0 to day 1, and +20 degrees C, from day 1 to day 2 and 3. Eight of 24 analytes showed no significant differences (p > 0.05) for three days in whole blood. However, the stability of all 24 analytes greatly improved by storing serum or heparinized plasma compared to nonanticoagulated or heparinized whole blood. In stored serum or heparinized plasma, 20 of 24 analytes showed no significant differences (p < 0.05) for 3 days. Nine of 24 analytes showed significant differences (p < 0.05) between serum and heparinized plasma, where CK, LDH, GGT, and potassium showed differences of possible clinical importance. This study strongly supports the practice of separating serum/plasma from clot/cells as promptly as possible to achieve improved stability for most analytes under test.  相似文献

Tick-borne fever (TBF) is a rickettsial disease of domestic and wild ruminants in temperate climates where the hard tick Ixodes ricinus is present. The disease is characterized by a high temperature and severe leukopenia. In the present study, the effects of TBF on the activity of serum alkaline phosphatase (ALP), and on the concentrations of plasma zinc, iron, total bilirubin, urea, creatinine and albumin were investigated by inoculating one group of eight sheep and one group of eight goats with the Old Sourhope (OS) strain of Ehrlichia (Cytoecetes) phagocytophila. All goats and sheep experimentally infected with E. phagocytophila reacted with fever, rickettsiaemia and leukopenia. The leukopenia was due to an acute lymphocytopenia and prolonged neutropenia. In both groups of animals. TBF was characterized by significant reductions in the activities of serum ALP and concentrations of plasma zinc, iron and albumin. However, there were significant increases in the concentrations of plasma total bilirubin, urea and creatinine in both species of animals. The reductions in ALP and iron were significantly more pronounced in sheep than in goats.  相似文献

选取9月龄欧拉型藏系绵羊羯羊(30.21 kg±1.42 kg)40只,随机分为4组,A组冷季放牧,B、C和D组冷季牧归每天每只分别补饲0.10、0.20和0.30 kg混合精料,试验期210 d,研究冷季牧归补饲精料对其血清生化指标的影响。结果表明,补饲精料提高了欧拉型藏系绵羊血清总蛋白(TP)、白蛋白(ALB)、球蛋白(GLB)、钙(Ca)的含量,钙/无机磷(Ca/IP)比值及肌酸激酶(CK)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)、谷丙转氨酶(ALT)和γ-谷氨酰胺基转移酶(GGT)活性;随着精料补饲量增大,绵羊血清葡萄糖(GLU)、胆固醇(CHO)和甘油三酯(TG)含量,超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性,生长激素(GH)、胰岛素样生长因子-1(IGF-1)、缩胆囊素(CCK)、胃泌素及免疫球蛋白A(IgA)、免疫球蛋白G(IgG)及免疫球蛋白M(IgM)的浓度增大,但补饲0.30 kg组试验羊IgG浓度较C组略有下降(P>0.05);与A组相比,补饲精料显著提高了试验羊碱性磷酸酶(AMP)活性、尿素氮(BUN)浓度和BUN/CRE的值,显著降低肌酐(CRE)和尿酸(UA)浓度(P<0.05)。综合上述结果,冷季归牧后补饲适量精料可改善欧拉型藏系绵羊能量和脂肪的代谢能力,增强肝脏蛋白合成,提高机体免疫力和抗氧化力,调节钙磷代谢。  相似文献

To study the effects of concentrate supplementation on serum biochemical indexes,40 nine-month-old Oura-type Tibetan sheep (30.21 kg±1.42 kg) were randomly divided into 4 groups.Group A grazed naturally,group B,C and D supplemented with 0.10,0.20 and 0.30 kg mixed concentrate per sheep every day after grazing,respectively.The trial lasted 210 days.The results showed that the content of total serum protein (TP),albumin (ALB),globulin (GLB),calcium (Ca),Ca/IP(inorganic phosphorus) value,creatine kinase (CK),lactate dehydrogenase (LDH),aspartate transaminase (AST),alaine transaminase (ALT) and γ-glutamine transferase (GGT) of the sheep increased with the concentrate supplemented.The content of serum glucose (GLU),cholesterol (CHO) and triglyceride (TG),the activities of superoxide dismutase (SOD) and catalase (CAT),the concentrations of growth hormone (GH),insulin-like growth facior-1 (IGF-1),cholecystokinin (CCK),IgA,IgG and IgM increased with the increasing of the amount of concentrate supplementation.But the IgG concentration of group D decreased a little compared with group C (P>0.05).Supplementary feed significantly increased the activity of alkaline phosphatase (AMP),the concentration of urea nitrogen (BUN) and the value of BUN/CRE (P<0.05),and significantly decreased the concentrations of creatinine (CRE) and uric acid (UA) (P<0.05).The results showed that supplemented with proper amount of concentrate after returning to grazing in cold season could improve the energy and fat metabolic capacity,enhance the synthesis of liver protein,improve the immunity and antioxidant capacity of the body,and regulate the metabolism of calcium and phosphorus of Oura-type Tibetan sheep.  相似文献

We studied the effects of storage time and temperature of clotted whole blood on the amounts of 17 analytes in bovine blood serum. Serum separated after blood was allowed to stand for 2, 4, 6, 12 and 24h at room temperature or on ice. Results obtained for phosphorous, magnesium, urea, cholesterol, beta-hydroxybutyrate (BHBA), triglyceride, albumin, total protein and gamma-glutamyletransferase (GGT) were not influenced by storage at room temperature or on ice for as long as 24h. Duration of the clotted whole blood storage had a significant effect on calcium, glucose concentrations, creatine kinase (CK) and aspartate aminotransferase (AST) activities and temperature had a significant effect on glucose, non-esterified fatty acids, CK and bilirubin concentrations.  相似文献

SUMMARY Effects of storage at room temperature (23–25°C) and refrigeration (4–5°C) on various biochemical constituents of camel serum were investigated. Albumin, globulin, calcium, phosphorus, cholesterol, aspartate aminotransferase (AST), alkaline phosphatase (AP) and gamma glutamyltransferase (GGT) did not change over 9 days when stored at 4–5°C. At 4–5°C, creatinine, iron and glucose in camel sera remained stable for 6 days; total protein for 7 days; and blood urea nitrogen for 8 days. Decreased activities in creatine kinase (CK), lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) were apparent after 1, 6 and 7 days, respectively. At room temperature, total protein, albumin, globulin, calcium and phosphorus were stable throughout the 9 days. Changes in glucose and iron occurred after 3 days. Stability at room temperature for LDH was 1 day; AST, 3 days; GGT and ALT, 6 days; and AP, 8 days. CK activity had already declined by 4 hours and by 9 days, only 34% activity remained.  相似文献

Since hemolysis has been a common problem in submitted ratite serum samples, a study was performed to determine interference by hemolysates. Nine chemistry analytes including glucose, total protein, albumin, creatine kinase (CK), gamma-glutamyl transferase (GGT), aspartate aminotransferase (AS7), calcium (Ca), phosphorus, and uric acid were evaluated on a wet reagent analyzer (Ciba Corning Express 550) and on a dry slide reagent analyzer (VetTest 8008). In emus, increasing hemoglobin concentrations increased total protein, albumin, and CK for both analyzers. With increasing hemoglobin concentrations, the Ciba Corning 550 analyte values were increased for AST, Ca, and uric acid and decreased for glucose and phosphorus, the opposite effect was seen in values from the VetTest 8008. GGT levels were variable or sometimes undetectable. Changes in ostrich analytes with hemolysis were similar to emus using the same analyzer. The effects of serum hemolysis often differed in magnitude and direction between the two chemistry analyzers. Interferographs were constructed to aid in rapid assessment of the effects of hemolysis on submitted serum samples.  相似文献

OBJECTIVE: To identify surrogate markers of passive transfer of immunity in kittens. DESIGN: Prospective clinical trial. ANIMALS: 55 kittens from 12 specific-pathogen-free queens. PROCEDURE: Kittens were allocated at birth into colostrum-fed (n = 27) and colostrum-deprived (28) groups. Blood was collected at birth and on days 1, 2, 4, 7, 14, 28, and 56. Serum samples were analyzed for activities of alkaline phosphatase (ALP), alanine aminotransferase, aspartate aminotransferase, creatine kinase, lactate dehydrogenase, gamma-glutamyltransferase, amylase, and lipase and for concentrations of albumin, total protein, bilirubin, urea nitrogen, creatinine, cholesterol, glucose, calcium, phosphorus, and triglycerides by use of an automated analyzer. Total serum solids concentrations were estimated by use of refractometry. Serum IgG concentrations were quantified by use of radial immunodiffusion. RESULTS: All kittens were agammaglobulinemic at birth. Colostrum-fed kittens had significantly higher IgG concentrations than did colostrum-deprived kittens from 1 though 28 days of age. Transient significant differences in serum biochemical variables between the colostrum-deprived and colostrum-fed groups were substantially resolved by day 4. Passive transfer of immunity could be reliably determined at 1 day of age and to a lesser extent at 2 days of age only by measurement of serum activity of ALP. CONCLUSIONS AND CLINICAL RELEVANCE: Adequacy of passive transfer in kittens initially correlated with serum activity of ALP, but quantification of serum IgG concentration was necessary after 2 days of age.  相似文献

Specific reference intervals are needed for each animal species for the appropriate interpretation of the results obtained from serum biochemical tests. The aim of this study was to investigate the blood composition of pure-bred Arabian mares to evaluate the need for defining reference values for different age groups. In all, 90 clinically healthy mares from five farms were used in the trial. For the determination of the effect of age, 90 mares were assigned to six groups. Groups A, B, C, D, E, and F comprised 15 foals aged ≤5 months (mean ± SD: 2.45 ± 1.88), 15 foals aged 6 to 18 months (mean ± SD: 9.33 ± 2.93), 15 mares aged 2 to 5 years (mean ± SD: 3.91 ± 0.7), 15 mares aged 6 to 12 years (mean ± SD: 8.66 ± 2.3), 15 mares aged 14 to 20 years (mean ± SD: 17.81 ± 2.13), and 15 mares aged >20 years (mean ± SD: 22.8 ± 1.93), respectively. Glucose, blood urea nitrogen (BUN), albumin, creatinine, total bilirubin, direct bilirubin, total protein, alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), calcium, iron, magnesium, and phosphorus concentrations were measured in the blood serum samples. There were significant age-related alterations for most of the biochemical parameters, except for ALT, LDH activities, and glucose, albumin, magnesium, and iron levels. The results of the present study showed that direct bilirubin (P < .01), ALP, and phosphorus decreased significantly (P < .001), whereas BUN, calcium (P < .01), creatinine, total protein, and total bilirubin increased (P < .001) with age. In foals, ALP activity, direct bilirubin, and phosphorus levels were higher, whereas BUN, creatinine, total protein, total bilirubin, and calcium levels were lower as compared with older animals. Biochemical values determined in the present study serve as reference ranges for pure-bred Arabian mares and can be used for health control and diagnosis of diseases.  相似文献

The effects of dexamethasone at different dose regimens on some haematological parameters and serum biochemistry were studied in the camel. Dexamethasone increased the serum concentration of glucose as well as neutrophils and total leukocyte count in the blood. Conversely, the serum concentrations of potassium and phosphorus and lymphocyte count in blood were decreased by dexamethasone treatment. Dexamethasone, however, had no effect on the activities of CPK and gamma-GT, on the serum concentrations of creatinine, BUN, total bilirubin, cholesterol and Na and on PCV, Hb, ESR, MCV and MCHC values.  相似文献

Abstract

Blood chemistry analytes are determined for fish from either serum or plasma samples. Serum and plasma are similar in that they both represent the fluid component of blood; however, plasma contains clotting factors that are not present in serum. This study was conducted to determine whether the type of sample—plasma or serum—had an effect on measured blood analytes in rainbow trout Oncorhynchus mykiss, channel catfish Ictalurus punctatus, hybrid tilapia Oreochromis spp., and hybrid striped bass (striped bass Morone saxatilis × white bass M. chrysops). Paired plasma and serum samples were analyzed for the following standard biochemical analytes: total protein, albumin, globulin, creatinine, total bilirubin, alkaline phosphatase, aspartate aminotransferase, sodium, potassium, chloride, calcium magnesium, phosphorus, glucose, and cholesterol. For all four taxa, values for potassium were lower in the serum and magnesium and phosphorus values were higher in the serum. Glucose values were lower in the serum from rainbow trout, hybrid striped bass, and channel catfish; whereas cholesterol values were higher in the serum of rainbow trout, channel catfish, and hybrid tilapias. The differences observed between serum and plasma were distinct from changes occurring with hemolysis and, therefore, do not represent release of erythrocyte constituents. The differences most likely represent metabolic utilization of blood constituents while the blood was clotting. This work indicates that plasma should be used preferentially to serum for biochemical analysis because analyte levels determined from serum may not accurately reflect those found in circulating blood.  相似文献

Haematological metabolic profiles in heifers could contribute to the development of proxies for oestrous detection and provide clues to further characterize biological changes during oestrus. One hundred and seven beef heifers were observed for oestrous behaviour twice daily for 124 days. Feed intake and productive performance (body weight and composition) traits were measured, and feed efficiency was determined using residual feed intake (kg DM/day). Blood plasma samples were collected when signs of oestrus were observed and every 30 ± 2 days. Heifers were considered in oestrus (n = 71) when plasma progesterone concentrations were <0.6 ng/ml. Least square means of blood metabolic parameters were compared between oestrous and non‐oestrous states and within oestrous groups according to performance traits and age. Heifers in oestrus exhibited higher concentrations of alkaline phosphatase, aspartate aminotransferase (AST), beta‐hydroxybutyric acid, creatine kinase (CK) and triiodothyronine (T3) than heifers in non‐oestrus. Heifers in oestrus revealed lower osmolality and concentrations of calcium, sodium and total protein than during non‐oestrus. Younger (and smaller) heifers had greater concentrations of CK, gamma‐glutamyl transferase (GGT), glucose and sodium than the older heifers. Heifers with lower fatness had increased osmolality and concentrations of cholesterol, CK, phosphorus, sodium and reduced T3 levels. Feed efficient heifers had greater levels of AST, cholesterol and GGT than inefficient heifers. Blood plasma parameters may be complementary to oestrous detection upon further validation; effects of age, feed efficiency, body size and body composition should be considered to optimize this haematological assessment.  相似文献

The aim of this study was to investigate the possible effects of the reproductive status and seasonal variations on the serum chemistry and vitamin status and their relationships with the thyroid hormones in Sakiz-Awassi crossbreed sheep. The sheep (n = 34) were divided into two groups. The first group (n = 22) was mated; the second group (n = 12) was not mated. Their serum samples were collected four times a year at the each season and under reproductive status. The periods are 1, early pregnancy (October); 2, late pregnancy (January); 3, lactation (April); and 4, dry season (July). The results of this study indicated that (1) total protein (TP), globulin, cholesterol, creatinine, uric acid and T4/T3 vary with reproductive status but not seasonal variations; (2) alanine aminotransferase, T4, fT4 concentrations in serum vary only with reproductive status; (3) the urea, creatine kinase, lactate dehydrogenase, alkaline phosphatase, aspartate aminotransferase, amylase, albumin, triglyceride, VLDL, Vit A-E, T3 and fT3 concentration could vary with both reproductive status and seasonal variations; (4) the glucose, gamma-glutamyl transpeptidase, folate concentrations were altered neither season of the year nor the reproductive status; (5) a single reference interval for folate, gamma-glutamyl transpeptidase (GGT), glucose, TP, globulin, cholesterol, creatinine, uric acid and T4/T3 can be used for both mated and non-mated sheep because of no differences were found due to the reproductive status. Taking the results together suggests that reproductive status and seasonal variations have to be taken into consideration for a correct interpretation of the serum chemistry values of sheep. Nutritional supplements are required for sheep during certain periods to avoid a decline of their performance, which would then represent consequent economic, loses.  相似文献

Aspartate aminotransferase, bilirubin, urea nitrogen, and creatinine were higher than in adults at birth but were similar to or lower than adult reference intervals by 8 weeks. Compared with adult reference intervals, reference intervals for calcium and phosphorus concentrations were higher and for albumin and total protein concentrations were lower throughout the study period. CONCLUSIONS AND CLINICAL RELEVANCE: Important differences exist between reference intervals for serum biochemical values of neonatal and adult cats. Age-appropriate reference intervals should be used for accurate assessment of serum biochemical test results in cats.  相似文献
