试验研究了乙酰半胱氨酸(NAC)对脂多糖(LPS)多次刺激造成的免疫应激仔猪炎性介质的影响,旨在探讨NAC是否有缓解仔猪免疫应激的作用。选取来源一致、体重相近的18头健康仔猪[杜洛克×长白×大白,体重(11.58±0.26)kg],随机分成3个处理(对照组、LPS组和NAC组),每个处理6个重复,每个重复1头猪。对照组和LPS组饲喂基础日粮,试验组(NAC组)为基础日粮+500 mg/kg NAC,LPS组和NAC组仔猪分别于试验的第101、3、20 d腹膜注射100μg/kg BW的LPS,对照组注射等量的灭菌生理盐水。试验第10、20 d在注射LPS 3 h后前腔静脉采血,测定血浆TNF-α、CORI、L-6和PGE2含量。试验结果显示:日粮中添加NAC显著缓解了LPS刺激导致的血浆(第10 d)和血浆(第20 d)中TNF-α、CORI、L-6和PGE2含量的升高(P<0.05)。由此可见,在本试验条件下,LPS刺激引起仔猪血浆中炎性介质升高,日粮中添加500 mg/kg NAC缓解LPS导致的血浆中炎性介质的升高,缓解免疫应激,最终可能缓解LPS刺激引起的生长抑制。  相似文献

试验选用 72头 (2 8± 2 )d断奶的仔猪 ,采用 2× 2因子试验设计 ,研究共轭亚油酸 (CLA)是否有缓解仔猪免疫应激的作用。结果显示 ,添加CLA缓解了因注射脂多糖 (LPS)引起的日增重降低 (P <0 .0 5 ) ,并改善了试验全期的饲料转化效率 (P <0 .0 5 )。两次LPS刺激后 ,CLA抑制 (P <0 .0 5 )了由LPS诱导的血浆白细胞介素 6 (IL 6 )、肿瘤坏死因子 α(TNF α)和α 乙酰糖蛋白 (AGP)浓度的上升。在第 14d和 2 1d ,LPS刺激提高 (P <0 .0 5 )了血浆IL 1β和皮质醇含量 ,而CLA则降低了IL 1β和皮质醇含量。本试验证明 ,CLA能缓解免疫应激引起的仔猪生长抑制 ,其防止免疫应激诱导的生长抑制作用可能与CLA抑制炎性细胞因子的分泌有关  相似文献

本试验旨在探讨饲粮中添加刺五加多糖(Acanthopanax senticosus polysaccharide,ASPS)对脂多糖(LPS)免疫应激断奶仔猪生长性能和血液生理生化指标的影响。试验采用2×2两因素设计,即饲粮处理(添加0或800 mg/kg ASPS)和免疫应激处理(注射LPS或生理盐水)。将64头(28±3)日龄、平均体重为(7.22±0.46)kg的"杜×长×大"三元杂交断奶仔猪随机分为4个处理,其中处理1和2饲喂基础饲粮(添加0 mg/kg ASPS),处理3和4饲喂试验饲粮(添加800 mg/kg ASPS),试验第14和21天,处理2和4仔猪腹腔注射100μg/kg BWLPS,处理1和3仔猪腹腔注射等量生理盐水。注射后3 h采血,测定血液生理生化指标。试验期21 d。结果表明:试验1~14 d,饲喂ASPS对未注射LPS的仔猪生长性能无显著影响(P>0.05);试验15~21 d,饲喂ASPS显著增加仔猪的平均日增重(ADG)和平均日采食量(ADFI)(P<0.05),且饲喂ASPS使注射LPS的仔猪ADG和ADFI显著增加(P<0.05),但对注射生理盐水的仔猪无显著影响(P>0.05)。饲喂ASPS对仔猪ADG的影响与LPS刺激存在显著互作关系(P<0.05)。试验第14天,饲喂ASPS显著提高了仔猪外周血淋巴细胞数量(P<0.05),对外周血淋巴细胞数量的影响与LPS刺激存在显著的互作关系(P<0.05),且饲喂ASPS能显著增加注射LPS的仔猪外周血淋巴细胞数量(P<0.05),但对注射生理盐水的仔猪无显著影响(P>0.05)。试验第14和21天,饲喂ASPS显著降低了仔猪血浆α-酸性糖蛋白(α-AGP)、葡萄糖、前列腺素E2(PGE2)含量(P<0.05),显著提高了血浆白细胞介素-2(IL-2)含量(P<0.05),且其对α-AGP、IL-2、PGE2含量的影响与LPS刺激存在显著的互作关系(P<0.05),饲喂ASPS能显著降低注射LPS的仔猪血浆α-AGP、PGE2含量(P<0.05),但对注射生理盐水的仔猪无显著影响(P>0.05)。由此可见,饲喂ASPS对非免疫应激断奶仔猪的生长性能无影响,但可以缓解免疫应激断奶仔猪的生长抑制,ASPS缓解免疫应激断奶仔猪生长抑制与降低其血浆α-AGP和PGE2含量,提高外周血淋巴细胞数量和血浆IL-2含量有关。  相似文献

试验研究了免疫应激对断奶仔猪免疫和神经内分泌激素的影响。选用 6头体重为 (7.6± 0 .3)kg的(2 8± 3)d的达兰断奶仔猪 ,绝食 12h后 ,随机选取 4头猪从腹膜注射 2 0 0 μg/kg(BW)的脂多糖 (LPS) ,另 2头注射等量生理盐水作对照。于注射后 0、1、2和 3h ,分别采血测定血浆白细胞介素 1β(IL 1β)、前列腺素E2 (PGE2 )、皮质醇、生长激素 (GH)和类胰岛素生长因子 I(IGF I)的含量。结果表明 :(1)注射LPS激活了应激轴 :与对照组相比 ,LPS提高了注射后 1h(P <0 .10 )、2h(P <0 .0 1)和 3h(P <0 .0 1)的IL 1β水平 ,提高了注射后 1h(P <0 .0 5 )和 3h(P <0 .0 1)的皮质醇和PGE2 水平 ;(2 )注射LPS抑制了生长轴 :LPS降低了注射后 3h的GH水平 (P <0 .10 ) ,降低了注射后 2h (P <0 .0 5 )和 3h(P <0 .0 1)的IGF I水平。结果显示 ,免疫应激激活了仔猪应激轴 ,而抑制了生长轴 ,在一定程度上揭示了免疫应激抑制生长的机制  相似文献

研究脂多糖(Lipopolysaccharide, LPS)对断奶仔猪外周血白细胞、胸腺、脾脏和肠道淋巴结过氧化物酶体增殖物活化受体(PPARγ) mRNA表达水平的影响,探讨免疫应激是否影响免疫细胞和免疫器官PPARγ mRNA的表达.选取12头健康的(21±1)d断奶仔猪,分成2个处理,每个处理6个重复.试验组注射100μg/kg BW的LPS,对照组注射等量的生理盐水.于注射LPS后1.5和3h,采血分离血浆、白细胞;注射LPS后3h屠宰仔猪,取胸腺、脾脏和肠道淋巴结进行相关检测.结果表明:(1)LPS刺激后1.5和3h,肿瘤坏死因子TNF-α和皮质醇含量均急剧上升(P<0.001);LPS刺激后3h,胰岛素含量显著下降(P<0.001),胰高血糖素含量显著上升(P<0.001).此外,LPS刺激也导致血液生化指标发生了显著变化.这表明LPS刺激导致机体处于急性免疫应激状态.(2)LPS刺激导致外周血白细胞(P<0.001)、胸腺(P<0.05)和肠道淋巴结(P<0.05)的PPARγ mRNA表达量显著升高,表明免疫应激诱导了免疫细胞和免疫器官中PPARγ mRNA的表达.提示PPARγ可能参与仔猪免疫应激的调控,可能成为缓解免疫应激的一个新靶点.  相似文献

本试验旨在研究N-乙酰半胱氨酸(NAC)对脂多糖(LPS)单次刺激仔猪肠黏膜免疫应激的影响。选取24头健康仔猪,随机分成4组,每组6个重复,每个重复1头猪。对照组和LPS组饲喂基础饲粮,250和500 mg/kg NAC组在基础饲粮中分别添加250和500 mg/kg NAC。试验第17天,LPS组、250和500 mg/kg NAC组仔猪腹膜分别注射100μg/kg BW的LPS,对照组注射相应剂量的灭菌生理盐水。注射后6 h屠宰,取肠黏膜。结果表明:与对照组相比,饲粮中添加250或500 mg/kg NAC缓解了LPS刺激导致的肠黏膜中白细胞介素-2、白细胞介素-6和前列腺素E2水平的升高及热应激蛋白70(HSP70)表达量的升高(P<0.05)。由此可见,饲粮中添加250和500 mg/kg NAC可有效抑制LPS刺激导致的肠黏膜中炎性因子的升高,缓解急性免疫应激。  相似文献

试验旨在探讨牛膝多糖(ABPS)对脂多糖(LPS)刺激的断奶仔猪生长性能、炎性介质和内分泌激素的影响。选用48头(28±3)d(8.45±0.14)kg的杜洛克×长白×大白断奶仔猪,采用2×2因子设计,包括日粮处理(0或500 mg/kg ABPS)和免疫应激(注射LPS或生理盐水)。试验期28 d。在第14天和第21天,每日粮组一半的猪注射100μg/kg BW的LPS,另一半注射生理盐水作对照。注射后3 h,采血测定血浆肿瘤坏死因子-α(TNF-α)、前列腺素E2(PGE2)、皮质醇、生长激素(GH)和类胰岛素生长因子-I(IGF-I)含量。结果表明:LPS刺激显著降低了第14~21、22~28天的日增重(ADG)和日采食量(ADFI)(P<0.05或P=0.05);ABPS显著提高了22~28 d的ADG(P<0.05)。LPS刺激显著提高了14 d和21 d的TNF-α、PGE2和皮质醇的含量,降低了IGF-I的含量(P<0.01);ABPS显著缓解了LPS刺激导致的TNF-α(14 d和21 d)和皮质醇(14 d)含量的上升以及IGF-I(14 d和21 d)含量的下降(P<0.05)。说明ABPS缓解了免疫应激仔猪生长的抑制,其机制可能与其抑制了炎性介质的分泌有关。  相似文献

为研究猪生长激素释放因子(pGRF)基因质粒对由脂多糖或三联苗诱导的免疫应激仔猪生长性能和免疫指标的影响,选用36头(28±2)d断奶的杜长大(Duroc×Landrace×Yorkshire,DLY)三元杂交仔猪,按体重相近、阎公猪和母猪各1/2的原则,分为对照组(不注射pGRF基因质粒、大肠杆菌LPS或三联苗)、注射pGRF组、注射脂多糖(LPS)组、注射pGRF+LPS组、注射三联疫苗组(猪瘟兔化弱毒,猪丹毒杆菌G4T10弱毒,猪源多杀性巴氏杆菌E0630弱毒)和注射pGRF+三联疫苗6个处理,每个处理6个重复,每个重复1头猪.试验开始时,pGRF组、pGRF+LPS组、pGRF+三联疫苗组的仔猪大腿肌肉分别注射1.0 mg pGRF基因质粒;试验的第11天给LPS组、pGRF+LPS组仔猪注射100 ug/kg LPS生理盐水溶液,三联疫苗组以及pGRF+三联疫苗组注射2头份三联苗生理盐水溶液,对照组和pGRF组注射生理盐水;第18天时按照第11天的操作重复进行.结果表明,在2次注射LPS或三联苗后,即10~17 d和17~24 d阶段,pGRF+LPS组仔猪的ADG显著(10~17 d,P<0.05)或极显著(17~24 d,P<0.01)高于LPS组,F/G均显著低于LPS组(P<0.05);pGRF+疫苗组仔猪ADG均显著高于疫苗组(P<0.05),F/G显著低于疫苗组(P<0.05).在第11天和第18天,pGRF+LPS组仔猪的血清细胞因子(IL-1、IL-6)水平均极显著低于LPS组(P<0.01),类胰岛素生长因子(IGF-I)和IgG浓度均极显著高于LPS组(P<0.01);pGRF+疫苗组仔猪的血清IL-1和IL-6浓度显著低于疫苗组(P<0.05),lgG浓度极显著(11 d,P<0.01)或显著(18 d,P<0.05)高于疫苗组,IGF-I浓度显著(11 d,P<0.05)或极显著(18 d,P<0.01)高于疫苗组.证明pGRF基因质粒能缓解由LPS或三联疫苗免疫应激引起的仔猪生长抑制;能够抑制由脂多糖或三联苗诱导的血清IL-1和IL-6浓度的上升和血清IGF-I浓度的降低,同时能提高免疫应激仔猪的血清IgG浓度.  相似文献

In late December 1983, 2 800 veterinarians across Canada were sent a questionnaire in order to assess the status of heartworm disease in Canada in 1983 and 26% of them responded. Veterinarians reported that 59 504 dogs were blood-tested to check for microfilariae and 771 dogs (1.30% of those tested) were found with Dirofilaria immitis. Heartworm disease was diagnosed in all provinces except New Brunswick and Newfoundland but most (733) of the cases were in Ontario.

Heartworm disease was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 31% of the cases were observed with clinical signs of heartworm disease and 64% had a history of not having left Canada. Southwestern Ontario continues to be the focus of the infection and most of the dogs there had not left the province previously.

In late December 1985, 1485 institutional veterinarians and small and mixed animal clinics across Canada were sent a questionnaire in order to assess the status of Dirofilaria immitis in Canada in 1985 and 44% of them responded. Veterinarians reported that 137,300 dogs were blood-tested to check for microfilariae and 1210 dogs were found with heartworm. Another 36 dogs were amicrofilaremic but diagnosed with heartworm disease to give the total number diagnosed in 1985 as 1247 (0.91%).

Heartworm was reported from all provinces except Prince Edward Island, Newfoundland and Saskatchewan but most (1126) of the cases were in Ontario. Southwestern Ontario continued to be the primary focus of the infection in Canada. From Quebec, 91 cases were reported mostly from and around Montreal. From Manitoba, 19 cases were reported from Winnipeg and surrounding areas. Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 28% of the cases were observed with clinical signs of heartworm disease and 78% had a history of not having left Canada.

Heartworm was reported from all provinces except Prince Edward Island and Newfoundland but most (1310) of the cases were in Ontario. In Quebec, 126 cases were reported mostly from west of Montreal.

Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 27% of the cases were observed with clinical signs of heartworm disease and 72% had a history of not having left Canada. In late November 1991, 1883 clinics in Canada were sent a questionnaire to assess the status of Dirofilaria immitis in dogs in 1991 and there was a 60.0% response. There were 344,031 dogs tested for heart-worm (HW), 627 were found infected and the prevalence of HW infection was 0.18%. There were 417 dogs with HW in Ontario, 116 in Manitoba, 38 in Quebec, 53 in British Columbia, three in Alberta, and one in Nova Scotia. In British Columbia, all of the infected dogs but one were from the Okanagan valley which, as from 1991, is a new focus of infection in Canada. Most dogs with HW had not been on preventive medication in 1990, and the prevalence among dogs tested and unprotected was 0.59%. That prevalence was considerably higher in endemic areas. Companion dogs, over three years of age and maintained primarily outdoors in rural areas, were most frequently infected. One cat was diagnosed with D. immitis and 33 dogs had Dipetalonema reconditium.  相似文献

In late November 1989, 1732 clinics and institutional veterinarians were sent a questionnaire to assess the status of Dirofilaria immitis, and 51.7% responded. Of 247,716 dogs tested, 394 had D. immitis microfilariae and 51 were amicrofilaremic for a total of 445 cases and heartworm prevalence of 0.17%. Most (408) of these dogs had no preventive medication and the prevalence among dogs tested and unprotected was 1.01%. That prevalence was considerably higher in endemic areas. Thirty-seven dogs with heartworm had preventive medication. Heartworm was most frequent in companion dogs over three years of age maintained outdoors in rural areas. About 75% of the cases had never left Canada, 26% had clinical signs and 125 were not treated.

Heartworm was reported from British Columbia, Manitoba, Ontario, Quebec, Nova Scotia and Newfoundland, but 383 cases were in Ontario. South-western Ontario was the primary focus of infection. There were 33 cases in Quebec and 24 in Manitoba, mainly found in and around Metropolitan Montreal and Winnipeg respectively.

Another 65 dogs were amicrofilaremic but diagnosed with heartworm disease and one was found with heartworm at necropsy to give the total number diagnosed in 1986 as 935 (0.62%).

Heartworm was reported from Manitoba, New Brunswick, Ontario and Quebec, but most (810) of the cases were from Ontario. South-western Ontario continued to be the primary focus of the infection in Canada. There were 103 cases reported from Quebec, mostly from and around Montreal, and 21 cases from Manitoba, from Winnipeg and surrounding areas. Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 33% of the cases were observed with clinical signs of heartworm disease and 81% had a history of not having left Canada.
