In mammals, the canonical nuclear factor kappaB (NF-kappaB) signaling pathway activated in response to infections is based on degradation of IkappaB inhibitors. This pathway depends on the IkappaB kinase (IKK), which contains two catalytic subunits, IKKalpha and IKKbeta. IKKbeta is essential for inducible IkappaB phosphorylation and degradation, whereas IKKalpha is not. Here we show that IKKalpha is required for B cell maturation, formation of secondary lymphoid organs, increased expression of certain NF-kappaB target genes, and processing of the NF-kappaB2 (p100) precursor. IKKalpha preferentially phosphorylates NF-kappaB2, and this activity requires its phosphorylation by upstream kinases, one of which may be NF-kappaB-inducing kinase (NIK). IKKalpha is therefore a pivotal component of a second NF-kappaB activation pathway based on regulated NF-kappaB2 processing rather than IkappaB degradation.  相似文献

【目的】细胞自噬和凋亡存在着相互制约,p38MAPK信号通路作为细胞凋亡的主要调控通路之一,也对细胞自噬存在促进和抑制的双重作用。已有研究表明,促性腺激素抑制激素(gonadotropin-inhibitory hormone , GnIH)对细胞自噬与凋亡均有影响,但作用机制尚不明确。故探究GnIH通过p38MAPK信号通路对猪卵巢颗粒细胞(pGCs)自噬与凋亡的影响及其机理,为解决母猪的产子率以及同期发情等问题提供参考。【方法】min、10 min、30 min、60 min、90 min)分组,用Western blot检测猪卵巢颗粒细胞p38与p-p38的蛋白表达量变化;2、验证GnIH对p38MAPK信号通路的影响:按(空白对照、GnIH、p38激活剂(U-46619)、U-46619+GnIH)分组,用Western blot检测p38与p-p38的蛋白表达量变化;3、探究不同浓度GnIH对自噬和凋亡的影响:按(空白对照、10 -6mol·L -1 GnIH、10 -8mol·L -1 GnIH、10 -10mol·L -1 GnIH、10 -12mol·L -1 GnIH)分组,用Western blot检测自噬与凋亡标志性蛋白的表达量变化;4、验证不同浓度GnIH通过p38信号通路对自噬和凋亡的影响:将细胞分成6组(空白对照、U-46619、U-46619+10 -6 mol·L -1 GnIH、U-46619+10 -8mol·L -1 GnIH、U-46619+10 -10mol·L -1 GnIH、U-46619+10 -12mol·L -1 GnIH),用Western blot检测自噬与凋亡标志性蛋白的表达量变化。【结果】1. GnIH孵育10 min后,显著降低p38与p-p38的蛋白表达量(P<0.05),提示,GnIH对p38MAPK信号通路的最佳作用时间为10 min;2. U-46619显著促进pGCs的p38磷酸化水平(P<0.05),GnIH显著抑制pGCs的p38磷酸化水平(P<0.05),提示,U-46619使p38MAPK信号通路活化,GnIH对p38MAPK信号通路的活化有抑制作用;3. 当 GnIH的浓度为10 -6 mol·L -1时,pGCs的自噬和凋亡水平显著升高(P<0.05),随着GnIH浓度的降低,pGCs的自噬水平逐渐升高(P<0.05),pGCs的凋亡水平逐渐降低(P<0.05),提示,高浓度GnIH可以促进自噬和凋亡,随着GnIH浓度的降低,自噬水平逐渐升高,而凋亡水平逐渐下降;4. 加入U-46619后,GnIH使pGCs的自噬显著上调(P<0.05),并且使pGCs的凋亡显著下调(P<0.05),提示,不同浓度GnIH通过p38MAPK信号通路影响pGCs的自噬和凋亡。【结论】GnIH可能通过抑制p38MAPK信号通路的活化,上调pGCs的自噬,减少pGCs的凋亡。  相似文献

Cytokine signaling is thought to require assembly of multicomponent signaling complexes at cytoplasmic segments of membrane-embedded receptors, in which receptor-proximal protein kinases are activated. Indeed, CD40, a tumor necrosis factor receptor (TNFR) family member, forms a complex containing adaptor molecules TRAF2 and TRAF3, ubiquitin-conjugating enzyme Ubc13, cellular inhibitor of apoptosis proteins 1 and 2 (c-IAP1/2), IkappaB kinase regulatory subunit IKKgamma (also called NEMO), and mitogen-activated protein kinase (MAPK) kinase kinase MEKK1 upon ligation. TRAF2, Ubc13, and IKKgamma were required for complex assembly and activation of MEKK1 and MAPK cascades. However, these kinases were not activated unless the multicomponent signaling complex translocated from CD40 to the cytosol upon c-IAP1/2-induced degradation of TRAF3. This two-stage signaling mechanism may apply to other innate immune receptors, accounting for spatial and temporal separation of MAPK and IKK signaling.  相似文献

The 66-kilodalton isoform of the growth factor adapter Shc (p66Shc) translates oxidative damage into cell death by acting as reactive oxygen species (ROS) producer within mitochondria. However, the signaling link between cellular stress and mitochondrial proapoptotic activity of p66Shc was not known. We demonstrate that protein kinase C beta, activated by oxidative conditions in the cell, induces phosphorylation of p66Shc and triggers mitochondrial accumulation of the protein after it is recognized by the prolyl isomerase Pin1. Once imported, p66Shc causes alterations of mitochondrial Ca2+ responses and three-dimensional structure, thus inducing apoptosis. These data identify a signaling route that activates an apoptotic inducer shortening the life span and could be a potential target of pharmacological approaches to inhibit aging.  相似文献

[目的]克隆青蛤MAPK/p38基因,并分析其在Poly:Ic胁迫下的表达情况。[方法]利用转录组测序获得青蛤MAPK/p38基因的序列信息,采用生物信息学软件分析该基因在近缘生物间的进化关系;利用巢式及荧光定量PCR技术克隆青蛤MAPK/p38基因的结构域序列,并在Poly:Ic胁迫后立即分析该基因在青蛤各组织中的表达情况以及血淋巴中的时序性表达情况。[结果]克隆得到青蛤MAPK/p38基因的结构域序列,分析发现该基因在物种进化中很保守,其在青蛤的血淋巴、肝脏、外套膜、闭壳肌和鳃中均有表达,其中在血淋巴中表达量最高,腮中表达量最低。Poly:Ic胁迫3 h后该基因的表达量达到最大值,与对照组差异极显著(P0.01)。[结论]MAPK/p38基因所表达的产物是青蛤重要的免疫信号通路蛋白。  相似文献

目的 观察加味丹参饮含药血清对缺氧/复氧(hypoxia/reoxygenation,H/R)乳鼠心肌细胞p38 MAPK蛋白表达的影响,以探讨其保护心肌的作用机制。方法 将20只SD乳鼠的心肌细胞进行原代培养,建立H/R模型并随机分为H/R组、SB203580(p38MAPK阻断剂组)、加味丹参饮含药血清组,正常心肌细胞组作为对照组。采用T淋巴细胞化学染色法鉴定心肌细胞,罗丹明B(Rhodamine B)染色法观察细胞形态,MTT比色法检测含药血清对乳鼠心肌细胞的毒性,Western-blot法检测MKK3(促分裂原活化蛋白激酶-激酶3)、MKK6(促分裂原活化蛋白激酶-激酶6)、p38MAPK、phospho-p38MAPK蛋白表达。结果 与正常血清对照组比较,H/R组MKK3、MKK6的蛋白表达增多,p38MAPK通路阻断剂SB203580和加味丹参饮均不能减少其表达;p38MAPK、phospho-p38MAPK在H/R时表达均增加(P<0.01),而SB203580和加味丹参饮含药血清均能减少其表达(P<0.01)。结论 加味丹参饮含药血清预处理可通过抑制缺氧,复氧心肌细胞p38MAPK信号通路发挥保护作用,抑制p38MAPK表达及其磷酸化,减轻心肌细胞损伤,起到保护心肌细胞的作用。  相似文献

Homologs of the Yersinia virulence effector YopJ are found in both plant and animal bacterial pathogens, as well as plant symbionts. These YopJ family members were shown to act as cysteine proteases. The catalytic triad of the protease was required for inhibition of the mitogen-activated protein kinase (MAPK) and nuclear factor kappaB (NF-kappaB) signaling in animal cells and for induction of localized cell death in plants. The substrates for YopJ were shown to be highly conserved ubiquitin-like molecules, which are covalently added to numerous regulatory proteins. YopJ family members exert their pathogenic effect on cells by disrupting this posttranslational modification.  相似文献

Before fertilization, vertebrate eggs are arrested in metaphase of meiosis II by cytostatic factor (CSF), an activity that requires activation of the mitogen-activated protein kinase (MAPK) pathway. To investigate whether CSF arrest is mediated by the protein kinase p90Rsk, which is phosphorylated and activated by MAPK, a constitutively activated (CA) form of Rsk was expressed in Xenopus embryos. Expression of CA Rsk resulted in cleavage arrest, and cytological analysis showed that arrested blastomeres were in M phase with prominent spindles characteristic of meiotic metaphase. Thus, Rsk appears to be the mediator of MAPK-dependent CSF arrest in vertebrate unfertilized eggs.  相似文献

目的 探讨基于p38 MAPK信号通路研究养阴润目丸治疗干眼兔的作用机制。方法 采用氢溴酸东莨菪碱皮下注射途径制备干眼兔模型,按随机数字表法分成模型组、p38抑制剂组、阳性药物(杞菊地黄丸)组和低、中、高剂量养阴润目丸组,同步设不造模的正常组,6只/组。各组按相应处理方式连续干预2周后,检测各组兔泪液分泌量、泪腺病理形态、泪腺组织凋亡细胞的表达及泪腺组织中p38 MAPK、Bax、Bcl-2蛋白和mRNA表达情况。结果 养阴润目丸组和阳性药物组能增加干眼兔模型泪液分泌量,抑制泪腺细胞的凋亡,下调p38 MAPK、Bax的蛋白和mRNA表达,上调Bcl-2的蛋白和mRNA表达水平,与模型组比较,差异均有统计学意义(P<0.05)。结论 养阴润目丸能够通过纠正Bax/Bcl-2失衡,改善干眼泪腺细胞凋亡,进而减轻泪腺细胞损伤,恢复泪腺细胞的代谢功能,与调控p38 MAPK的关系极为密切,这可能是其治疗干眼的重要机制之一。  相似文献

Glycogen synthase kinase 3beta (GSK3beta) is involved in metabolism, neurodegeneration, and cancer. Inhibition of GSK3beta activity is the primary mechanism that regulates this widely expressed active kinase. Although the protein kinase Akt inhibits GSK3beta by phosphorylation at the N terminus, preventing Akt-mediated phosphorylation does not affect the cell-survival pathway activated through the GSK3beta substrate beta-catenin. Here, we show that p38 mitogen-activated protein kinase (MAPK) also inactivates GSK3beta by direct phosphorylation at its C terminus, and this inactivation can lead to an accumulation of beta-catenin. p38 MAPK-mediated phosphorylation of GSK3beta occurs primarily in the brain and thymocytes. Activation of beta-catenin-mediated signaling through GSK3beta inhibition provides a potential mechanism for p38 MAPK-mediated survival in specific tissues.  相似文献

The c-Jun NH2-terminal kinase (JNK) is activated when cells are exposed to ultraviolet (UV) radiation. However, the functional consequence of JNK activation in UV-irradiated cells has not been established. It is shown here that JNK is required for UV-induced apoptosis in primary murine embryonic fibroblasts. Fibroblasts with simultaneous targeted disruptions of all the functional Jnk genes were protected against UV-stimulated apoptosis. The absence of JNK caused a defect in the mitochondrial death signaling pathway, including the failure to release cytochrome c. These data indicate that mitochondria are influenced by proapoptotic signal transduction through the JNK pathway.  相似文献
