Analytical procedures for the determination of pharmaceuticals from different therapeutic groups were proposed. These groups included the corticosteroids prednisolone and dexamethasone; the β-blockers sotalol, metoprolol, propranolol, and carvedilol; and the analgesic nonsteroidal anti-inflammatory drugs paracetamol, aspirin, metamizole, and ketoprofen. Reversed-phase ultrahigh performance liquid chromatography with an ultraviolet detector, different columns, different mobile phases, and gradient elution programmes were used to obtain the best separations within the shortest possible time. Solid-phase extraction was examined as a preconcentration step. The Oasis HLB column, with the highest recoveries (over 90% for most of the drugs), was chosen for the analysis of surface waters. Limits of detection ranged from 0.06 to 0.39 μg L?1 for all drugs after optimisation of all analytical steps.  相似文献

在不同发育阶段,睾丸合成睾酮的能力差异很大,为了阐明引起类固醇合成差异的分子机制,本实验以不同年龄的SPF昆白小鼠为研究对象,采用RT-PCR,Western Blot分析了不同年龄阶段类固醇合成酶的变化。结果表明:(1)在不同年龄阶段,P450SCCmRNA、3β-HSDmRNA、StAR mRNA和蛋白的水平无明显变化;(2)P450c17mRNA的表达水平在30d、60d、120d均处于较高的水平,但是在270d则处于较低的水平;(3)从30d到270d,ERK活性无明显变化。这表明, ERK活性可能与衰老引起睾酮合成能力下降无关。  相似文献

Measurement of shikimic acid accumulation in response to glyphosate inhibition of 5-enolpyruvylshikimate-3-phosphate synthase is a rapid and accurate assay to quantify glyphosate-induced damage in sensitive plants. Two methods of assaying shikimic acid, a spectrophotometric and a high-performance liquid chromatography (HPLC) method, were compared for their accuracy of recovering known amounts of shikimic acid spiked into plant samples. The HPLC method recovered essentially 100% of shikimic acid as compared with only 73% using the spectrophotometric method. Relative sensitivity to glyphosate was measured in glyphosate-resistant (GR) and non-GR cotton leaves, fruiting branches, and squares (floral buds) by assaying shikimic acid. Accumulation of shikimic acid was not observed in any tissue, either GR or non-GR, at rates of 5 mM glyphosate or less applied to leaves. All tissues of non-GR plants accumulated shikimic acid in response to glyphosate treatment; however, only fruiting branches and squares of GR plants accumulated a slight amount of shikimic acid. In non-GR cotton, fruiting branches and squares accumulated 18 and 11 times, respectively, more shikimic acid per micromolar of translocated glyphosate than leaf tissue, suggesting increased sensitivity to glyphosate of reproductive tissue over vegetative tissue. GR cotton leaves treated with 80 mM of glyphosate accumulated 57 times less shikimic acid per micromolar of translocated glyphosate than non-GR cotton but only 12.4- and 4-fold less in fruiting branches and squares, respectively. The increased sensitivity of reproductive structures to glyphosate inhibition may be due to a higher demand for shikimate pathway products and may provide an explanation for reports of fruit abortion from glyphosate-treated GR cotton.  相似文献

以水稻南光为材料,研究了不同铵硝摩尔比例处理时,水稻NH4^+吸收代谢基因的表达情况.档结果表明:(1)应用荧光定量PCR方法,可以精确检测水稻氮素吸收代谢基因在不同铵硝处理间的表达量变化;(2)从各基因的表达量上来看,吸收基因中以OsAMT1.1表达量最高,编码GS的基因中以OsGln1.1表达量最高,编码GOGAT的基因中,OsGlu表达量最高;(3)总体来说,不同铵硝处理对NH4^+吸收代谢基因的表达有显著影响,吸收基因对不同铵硝比例的反应要比代谢基因更敏感;(4)氮吸收基因中OsAMT4.1显著受到NO3的抑制,NH4^+的诱导.而OsAMT1.1,OsAMT1.2,OsAMT1.3和OsNRT2在铵硝摩尔比例由100:0变为50:50过程中,受到NO3^-的显著抑制,在铵硝摩尔比例由50:50变为0:100过程中OsAMT1.2和OsAMT1.3受到NO3^-的显著诱导,OsAMT1.1和OsNRT2变化不显著;(5)编码GS的基因OsGln1.1表达受NO3诱导,受NH4^+抑制.OsGln2在铵硝摩尔比例由100:0变为50:50过程中,受到NO3^-增加的诱导,同时,OsGln1.2受到NO3增加的显著抑制作用,铵硝达到50:50以后,NO3^-比例的增加对OsGln1.2和OsGln2的表达没有显著影响;(6)编码GOGAT的基因OsGlt1和OsGlu在不同铵硝摩尔比例中的变化趋势一致:铵硝比例由100:0变为50:50过程中没有显著变化,铵硝比例50:50基础上表达量受到NO3^-比例增加的显著抑制,而OsGlt2的表达受NO3^-的显著抑制,NH;的显著诱导.  相似文献

建立牛肌内前脂肪细胞体外培养方法,旨在更深入地研究组成肌肉内脂肪组织的肌内前脂肪细胞的增殖分化特性及其影响因素。选取来自成年育肥鲁西黄牛第6与第7肋骨间的肌肉内脂肪组织,利用Ⅰ型胶原酶消化法分离获得牛肌内前脂肪细胞。培养出的细胞成分均一,增殖旺盛,分化率高,经形态学动态变化观察、生长曲线、油红O脂肪染色提取法及对胰岛素和地塞米松反应的测定,证明是功能活跃的前脂肪细胞,并在体外重现了其增殖分化过程,同时经染色体核型分析证明体外培养的细胞倍性正常,可以用于后续研究。本试验成功分离得到了成年育肥牛肌内前脂肪细胞,这为下一步研究肌内脂肪的沉积机制,改善牛肉品质奠定了基础。  相似文献

Gingerols and their corresponding dehydration products shogaols were considered as the active principles of ginger, the rhizome of the plant Zingiber officinale, for its antioxidant, anti-inflammatory, and antitumor activities. Ginger (Z. officinale) has been cultivated for thousands of years as a spice and for medicinal purposes in China. Tongling (Anhui province, China) has traditionally been regarded as an ideal cultivation place. "Tongling White Ginger" enjoys a reputation for being one of the top gingers in China for its thin white peel, tender flesh, rich juice, and flavor. In this study, we have isolated and identified two novel gingerdione dimers, bisgingerdiones A (1) and B (2); two new gingerol derivatives, (5R)-5-acetoxy-1,7-bis(4-hydroxy-3-methoxyphenyl)heptan-3-one (3) and methyl (Z)-neral acetal-[6]-gingerdiol (4); and 38 known compounds (5-42) from rhizomes of Zingiber officinale collected from Tongling, China. Their structures were elucidated by means of spectroscopic methods. Compounds 1-4 showed weak cytotoxic and anti-HIV-1 activities. Compounds 6, 8, and 26 showed inhibitory activities against human and mouse 11β-HSD1 (11β-hydroxysteroid dehydrogenases) with IC(50) values between 1.09 and 1.30 μM.  相似文献

The presence of illicit drugs and their metabolites in surface waters has to be considered a new type of hazard, still unknown, for the aquatic ecosystem, due to the potent pharmacological activities of all the illicit drugs. Our research was therefore aimed at evaluating the impact of illicit drugs on the aquatic fauna, till now still undervalued. To this aim, we verified the ability of the European eel (Anguilla anguilla), a well-known biomonitor of environmental contamination, to bioaccumulate cocaine, one of the most abundant illicit drugs found in surface waters. Silver eels were exposed to a nominal cocaine concentration of 20?ng/l for 1?month; at the same time, control, carrier, and post-exposure recovery groups were made. Brains, gills, liver, kidney, muscle, gonads, spleen, digestive tract, and sections of dorsal skin were assayed by high-pressure liquid chromatography. Cocaine was found in the tissues of the treated eels and, at low concentrations, in almost all tissues of post-exposure recovery eels. These results indicate that cocaine is able to accumulate into the eel tissues; its presence suggests potential risks for eels since cocaine could affect their physiology and contribute to their decline, and for humans consuming contaminated fish.  相似文献

Global production of glyphosate-resistant (GR) soybean [Glycine max (L.) Merr.] continues to increase annually; however, there are no particular specific fertilizer recommendations for the transgenic varieties used in this system largely because reports of glyphosate effects on mineral nutrition of GR soybeans are lacking. Several metabolites or degradation products of glyphosate have been identified or postulated to cause undesirable effects on GR soybeans. In this work we used increasing glyphosate rates in different application on cv. ‘BRS 242 GR’ in order to evaluate photosynthetic parameters, macro- and micronutrient uptake and accumulation and shoot and root dry biomass production. Increasing glyphosate rates revealed a significant decrease in photosynthesis, macro and micronutrients accumulation in leaf tissues and also decreases in nutrient uptake. The reduced biomass in GR soybeans represents additive effects from the decreased photosynthetic parameters as well as lower availability of nutrients in tissues of the glyphosate treated plants.  相似文献

A field study was performed to assess the drug residue level in edible tissues after a therapeutic application of the synthetic glucocorticoids dexamethasone and flumethasone. Three diseased calves were injected intramuscularly with a commercial batch of dexamethasone esters and slaughtered 72 h after treatment. Another three calves were injected intramuscularly with an aqueous flumethasone preparation and slaughtered 24 h later. Residues of synthetic glucocorticoids in liver, muscle, kidney, and urine were assessed by competitive enzyme immunoassay. All dexamethasone concentrations exceeded the maximal residue level of 0.75 microg/kg in muscle and kidney and 2 microg/kg in the liver. The presence of both dexamethasone and flumethasone in the liver was confirmed by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). These results indicate that liver tissue provides a suitable matrix to monitor the presence of illegal residues of synthetic glucocorticoids in slaughtered animals.  相似文献

提取睾丸酮丛毛单胞菌(Comamonas testosteroni)的基因组DNA,利用PCR扩增activator基因,将扩增产物克隆到pKtac1(含强启动子tac)中,构建重组质粒pKtac1-act1和 pKtac1-act2。经酶切分析和测序,鉴定出正确的重组质粒pKtac1-act1和 pKtac1-act2。将重组质粒转化入大肠杆菌(Escherichia coli)HB101中,用酶联免疫分析方法测定细菌总蛋白质中的activator表达量;将pAX1(含3α-HSD/CR基因)分别和重组质粒(pKtac1-act1和pKtac1-act2)一起转化入E. coli HB101中,测定细菌总蛋白质中的3α-HSD/CR和activator的表达量。结果表明:重组质粒能明显提高activator的表达;pAX1与重组质粒共转化的蛋白质粗提物中,activator和3α-HSD/CR的表达量都显著提高。  相似文献

Effect of storage temperature (ST) (5, 15, and 25°C) for paddy on the consumer perception of cooked rice (CR) was investigated with six major rice cultivars in Korea (Ilmibyeo, Chucheongbyeo, Ilpumbyeo, Hwayeongbyeo, Nampyeongbyeo, and Odaebyeo) after 12 months of storage. Germination rate (GR) of the paddy, grading characteristics (percentages of head rice, broken kernels, damaged kernels, colored kernels and chalky kernels) of milled rice (MR) kernels, texture profile analysis (hardness, springiness, cohesiveness, adhesiveness, and chewiness) and color (L*, a*, and b*) of CR were measured. Also, consumer testing on CR was done with 108 consumers. Consumers evaluated acceptability for odor, appearance, taste, texture, and overall purchase intent, and willingness to pay (WTP) of CR. After 12 months of storage, physicochemical characteristics of MR and CR were significantly different depending on ST. The significant difference among three ST of paddy was noted for GR, color b* value, fat acidity of MR, and overall consumer acceptability of CR. The GR was constant during 12 months of storage at 5°C, whereas a significant decrease of GR was noted for all six cultivars within 12 months of storage at 25°C. The average WTP for rice stored at 5°C for 12 months was $U.S. 45.68/20 kg, whereas WTP for rice stored at 15°C and 25°C were $U.S. 44.19/20 kg and $U.S. 41.87/20 kg, respectively, implying the importance of ST on grain quality and product value by consumers. Overall consumer acceptability had high correlation coefficient with WTP (r = 0.985). Overall consumer acceptability had highest correlation with GR of paddy (r = 0.861), followed by b* value of CR (r = –0.826), fat acidity (r = –0.768), cohesiveness (r = 0.733), and hardness (r = –0.650) of CR by TA, implying GR of paddy and b* value of CR could be used as indicators for eating quality of rice.  相似文献

Pepper leaves treated with the herbicide J852 show an accumulation of phytoene and zeta-carotene, whereas treatment with norflurazon led to an accumulation of only phytoene. The effects of these herbicides were examined in vitro after the expression of carotenoid desaturases in Escherichia coli. Whereas norflurazon is a potent inhibitor of phytoene desaturase (PDS) (I(50) = 0.12 microM) but not of zeta-carotene desaturase (ZDS) (I(50) = 144 microM), J852 inhibits both PDS (I(50) = 23 microM) and ZDS (I(50) = 49 microM). The influence of PDS/ZDS inhibition on gene expression was examined by comparative RT-PCR. None of the examined genes, namely, encoding phytoene synthase, PDS, ZDS, or the terminal oxidase associated with phytoene desaturation, were induced upon herbicide treatment in pepper leaves or seedlings. This was unexpected because inhibition of carotene desaturation led to an up-regulation of the carotenoid biosynthetic capacity (higher amounts of accumulating precursors plus remaining colored carotenoids are present in treated tissues versus control).  相似文献

The connection between nutrient input and algal blooms for inland water productivity is well known but not the spatial pattern of water nutrient loading and algae concentration. Remote sensing provides an effective tool to monitor nutrient abundances via the association with algae concentration. Twenty-one field campaigns have been conducted with samples collected under a diverse range of algal bloom conditions for three central Indiana drinking water bodies, e.g., Eagle Creek Reservoir (ECR), Geist Reservoir (GR), and Morse Reservoir (MR) in 2005, 2006, and 2008, which are strongly influenced anthropogenic activities. Total phosphorus (TP) was estimated through hyperspectral remote sensing due to its close association with chlorophyll a (Chl-a), total suspended matter, Secchi disk transparency (SDT), and turbidity. Correlation analysis was performed to determine sensitive spectral variables for TP, Chl-a, and SDT. A hybrid model combining genetic algorithms and partial least square (GA-PLS) was established for remote estimation of TP, Chl-a, and SDT with selected sensitive spectral variables. The result indicates that TP has close association with diagnostic spectral variables with R 2 ranging from 0.55 to 0.72. However, GA-PLS has better performance with an average R 2 of 0.87 for aggregated dataset. GA-PLS was applied to the airborne imaging data (AISA) to map spatial distribution of TP, Chl-a, and SDT for MR and GR. The eutrophic status was evaluated with Carlson trophic state index using TP, Chl-a, and SDT maps derived from AISA images. Mapping results indicated that most MR belongs to mesotrophic (48.6%) and eutrophic (32.7%), while the situation was more severe for GR with 57.8% belongs to eutrophic class, and more than 40% to hypereutrophic class due to the high turbidity resulting from dredging practices.  相似文献

Residues of isopropyl (2E,4E)-11-methoxy-3,7,11-trimethyl-2,4-dodecadienoate (Altosid) insect growth regulator are determined in waters, soils, plants, milk, eggs, fish, shellfish, poultry and cattle tissues, blood, urine, and feces. Acetonitrile is the primary extraction solvent for all samples. Residues are extracted by high-speed blending followed by vacuum filtration. Fatty extracts are subjected to cold-temperature precipitation and filtration. Samples are cleaned up by petroleum ether partitioning and Florisil and neutral alumina chromatography. The concentrated eluants are analyzed by gas-liquid chromatography (GLC) on columns of differing polarity, using hydrogen flame ionization detectors. The identity of suspected residues is confirmed by additional GLC and by mass fragmentography. The lower limits of detection were: water samples, 0.0004-0.001 ppm; soils, blood, and urine, 0.001 ppm; forage grasses, forage legumes, and rice foliage, 0.005 ppm; and milk, eggs, fish, shellfish, poultry and cattle tissues, and feces, 0.010 ppm.  相似文献

Alzheimer's disease is neuropathologically characterized by amyloid β-protein (Aβ) deposition, resulting in neurotoxicity. Herein, we focused on the prevention of anthocyanins from amyloid-mediated neurodysfunction. The data demonstrated that combined exposure of Aβ(1-40) and Aβ(25-35) to Neuro-2A cells resulted in reactive oxygen species (ROS) production and perturbation of calcium homeostasis. The expressions of LXRα, ApoE, ABCA1, and seladin-1 genes were significantly down-regulated upon Aβ challenge. β-Secretase, the rate-limiting enzyme that catalyzes amyloid precursor protein transform to Aβ, was up-regulated by Aβ treatment. For the duration of Aβ stimulation, malvidin (Mal) or oenin (Oen; malvidin-3-O-glucoside) was added, and the protective effects were observed. Mal and Oen showed protective effects against Aβ-induced neurotoxicity through blocking ROS formation, preserving Ca(2+) homeostasis, and preventing Aβ-mediated perturbation of certain genes involved in Aβ metabolism and cellular defense. The present study implicates anthocyanin as a potential therapeutic candidate for the prevention of amyloid-mediated neurodysfunction.  相似文献

Inflammatory bowel diseases have been a risk factor of colorectal cancer (CRC). The reactive oxygen species (ROS) generated by inflammatory cells create oxidative stress and contribute to neoplastic transformation, proliferation, and even metastasis. Previously, resveratrol (RS) and pterostilbene (PS) had been reported to prevent chemical-induced colon carcinogenesis by anti-inflammatory and pro-apoptotic properties. In this study, we investigated whether RS and PS could prevent the azoxymethane (AOM)-induced colon tumorigenesis via antioxidant action and to explore possible molecular mechanisms. Male BALB/c mice were injected with AOM (5 mg/kg of body weight) with or without RS or PS, and at the end of the protocol, all of the mice were euthanized and colons were analyzed. Administrations of PS can be more effective than RS in reducing AOM-induced formation of aberrant crypt foci (ACF), lymphoid nodules (LNs), and tumors. We also find that PS is functioning more effectively than RS to reduce nuclear factor-κB (NF-κB) activation by inhibiting the phosphorylation of protein kinase C-β2 (PKC-β2) and decreasing downstream target gene expression, including inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and aldose reductase (AR) in mouse colon stimulated by AOM. Moreover, administration of RS and PS for 6 weeks significantly enhanced expression of antioxidant enzymes, such as heme oxygenase-1 (HO-1) and glutathione reductase (GR), via activation of NF-E2-related factor 2 (Nrf2) signaling. When the above findings are taken together, they suggest that both stilbenes block cellular inflammation and oxidative stress through induction of HO-1 and GR, thereby preventing AOM-induced colon carcinogenesis. In comparison, PS was a more potent chemopreventive agent than RS for the prevention of colon cancer. This is also the first study to demonstrate that PS is a Nrf2 inducer and AR inhibitor in the AOM-treated colon carcinogenesis model.  相似文献

FGF4已经被证明是癌基因,它涉及肿瘤的生长和转移,为了解FGF4的表达与肿瘤微环境的关系。我们利用FGF4抗体通过免疫组化对一名肺癌患者癌旁,癌组织,癌组织小鼠移植瘤,二次移植瘤以及原代培养的细胞爬片进行FGF4检测,探究其表达差异。通过对比癌组织与对照组(癌旁组织),FGF4在癌巢中高表达;同样,移植瘤与二次移植瘤的癌巢中与癌旁组织比较,FGF4表达相对较高;但是将癌组织进行原代培养后,免疫组化检测细胞爬片FGF4,发现仅有5%±0.21%的肿瘤细胞表达FGF4,对照蛋白Cytokine作为肿瘤标记物,则在100%的肿瘤细胞中表达。研究提示免疫组化检测到FGF4在体内和体外表达不同,提示肿瘤细胞FGF4的表达与肿瘤微环境调控密切相关,肿瘤微环境对肿瘤细胞的FGF4的调控有着重要作用。  相似文献

Naringenin, a well-known naturally occurring flavonone, demonstrates cytotoxicity in a variety of human cancer cell lines; its inhibitory effects on tumor growth have spurred interest in its therapeutic application. In this study, naringenin was derivatized to produce more effective small-molecule inhibitors of cancer cell proliferation, and the anticancer effects of its derivative, 5-hydroxy-7,4'-diacetyloxyflavanone-N-phenyl hydrazone (N101-43), in non-small-cell lung cancer (NSCLC) cell lines NCI-H460, A549, and NCI-H1299 were investigated. Naringenin itself possesses no cytotoxicity against lung cancer cells. In contrast, N101-43 inhibits proliferation of both NCI-H460 and A549 cell lines; this capacity is lost in p53-lacking NCI-H1299 cells. N101-43 induces apoptosis via sub-G1 cell-cycle arrest in NCI-H460 and via G0/G1 arrest in A549 cells. Expression of apoptosis and cell-cycle regulatory factors is altered: Cyclins A and D1 and phospho-pRb are down-regulated, but expression of CDK inhibitors such as p21, p27, and p53 is enhanced by N101-43 treatment; N101-43 also increases expression levels of the extrinsic death receptor Fas and its binding partner FasL. Furthermore, N101-43 treatment diminishes levels of cell survival factors such as PI3K and p-Akt dose-dependently, and N101-43 additionally induces cleavage of the pro-apoptotic factors caspase-3, caspase-8, and poly ADP-ribose polymerase (PARP). Cumulatively, these investigations show that the naringenin derivative N101-43 induces apoptosis via up-regulation of Fas/FasL expression, activation of caspase cascades, and inhibition of PI3K/Akt survival signaling pathways in NCI-H460 and A549 cells. In conclusion, these data indicate that N101-43 may have potential as an anticancer agent in NSCLC.  相似文献
