With 2 figures and 2 tables SUMMARY: Previous studies of motor unit contractile properties in the rat medial gastrocnemius revealed that these units generate higher forces in males than in females. Therefore, in the present study the number and morphometric parameters of muscle fibres and the innervation ratio of motor units in the medial gastrocnemius muscle were studied in male and female Wistar rats. The study additionally aimed at determining reasons of gender differences in motor unit force parameters, i.e. the number and diameter of muscle fibres, and mean values of the motor unit innervation ratios. Following staining of reticuline fibres by silver impregnation, the number, diameter and cross-section area of muscle fibres were determined on microscopic images of transverse muscle sections. In males, the muscles were approximately 1.5 times larger by mass and contained about 11 800 muscle fibres, whereas in females the muscles contained around 8000 fibres. In addition, the mean diameter and mean cross-section area of muscle fibres were 14 and 29% larger in males, respectively. Based on previously determined numbers of motoneurons innervating the medial gastrocnemius muscle in male and female rats, the mean innervation ratio, i.e. the number of muscle fibres innervated by one motoneuron, was estimated. This ratio was approximately 26% greater in males compared to females, with values of 207 and 153 fibres per motoneuron, respectively. Therefore, the differences in muscle fibre morphometric parameters and in the innervation ratio are responsible for higher forces of motor units in male muscles.  相似文献

The role of nerve impulses and neurogenic factors in the decrease in muscle mass following tenotomy was investigated in 30 adult (15 males, 15 females) guinea pigs. Unilateral tenotomy, neurectomy and simultaneous tenotomy and neurectomy of slow and fast hindlimb muscles were performed in 3 groups respectively. Each group comprised 10 guinea pigs. The unoperated hindlimb of each animal in each group served as control. The weight loss occurred in both slow and fast muscles but was greater in the slow soleus muscle as compared to the fast gastrocnemius muscle. In each muscle the weight loss was least in the tenotomized group. This was followed by the weight loss in the neurectomized group. The greatest weight loss was observed in the simultaneously tenotomized and neurectomized group. These observations agree with the suggestion of other workers that a possible neurogenic factor flowing from the nerves to the muscles was very significant in maintenance of muscle fibre stability. It indicated also that simultaneous neurectomy and tenotomy did not prevent degenerative changes in slow and fast muscles and that it was not nerve impulses reaching the tenotomized muscles that were responsible for degenerative changes observed but that other factors were responsible.  相似文献

This study was conducted to assess the in vitro effects of leucine and 3-hydroxy-3-methylbutyrate (HMB) on protein synthesis and degradation in isolated rat and chick muscles. Isolated muscles — extensor digitorum longus (EDL) and soleus (S) in rats and EDL and tibialis anterior (TA) in chicks — were taken out intact and incubated in Krebs—Henseleit buffer with [14C]phenylalanine. Protein synthesis was then determined by measuring [14C]phenylalanine incorporation into protein and protein breakdown was estimated by net release of tyrosine from muscle proteins. In all muscles investigated leucine stimulated protein synthesis (average 20%; p < 0.05 versus control) but only slightly reduced proteolysis (average 8%, NS). In contrast, HMB had no significant effect on protein synthesis but consistently decreased protein degradation in the range from −29% (p < 0.01 versus control) in rat extensor digitorus longus to −5% (NS) in rat soleus muscles. HMB appeared to suppress protein degradation more in white muscle fibres than in red muscle fibres (at least in the rat). These data support observations in humans indicating that HMB decreases muscle proteolysis. It is suggested that metabolism of leucine to HMB could be responsible for the reported suppressive effects of leucine on protein breakdown.  相似文献

A staining procedure used for simultaneously determining three different fibre types in single sections bovine, porcine or ovine skeletal muscle was modified for use with ostrich skeletal muscle. The muscle fibres of gastrocnemius pars externa, tibialis cranialis caput tibiale, tibialis cranialis caput femorale and fibularis longus tendo caudalis were studied. The histochemical results revealed the presence of three types of fibre only in the gastrocnemius pars externa muscle: fast-twitch glycolytic fibres (FG), fast-twitch oxidative glycolytic fibres (FOG) and slow-twitch oxidative fibres (SO), while in the other muscles the FG fibres were absent. The percentage distribution of fibres types showed a higher incidence of SO fibres compared to FOG fibres in tibialis cranialis caput femorale and tibialis cranialis caput tibiale muscles, while it was opposite in the case of the fibularis longus tendo caudalis muscle. In the gastrocnemius pars externa muscle the FG fibres outnumber the other fibres, followed by the SO and FOG fibres. The results of the analysis of variance show significant interaction between muscle x fibre type for every morphometric parameter evaluated. Differences about value of fibres area exists between tibialis cranialis caput femorale and fibularis longus tendo caudalis muscles. Both fibre types in tibialis cranialis caput tibiale muscle have mean values of transversal section area smaller than tibialis cranialis caput femorale. The other morphometric parameters show a similar trend. The gastrocnemius pars externa muscle presents similar dimensions of muscle fibres for the FG and FOG types, and significantly smaller for the SO type.  相似文献

The purpose of this study was to determine whether there were differences in skeletal muscle properties in the hindlimb muscles of different types of dogs. Muscle samples were obtained from the gracilis, sartorius cranial head, sartorius caudal head and tibialis anterior muscles of mixed-breed and hound-type dogs and Beagles. Fiber type, fiber size and capillary morphometry determinations of each muscle from each dog were made from sections stained for myofibrillar ATPase activity. Individual animals were bilaterally symmetric for all measured variables. Fiber type, fiber size and capillary geometry varied between dogs of a given type and muscles within a given dog. There were no differences between dog types for fiber type or fiber size; significant variation in log(muscle)/log(body) mass ratios between dog types was observed for all muscles. The results indicate that for a given muscle, significant variation can occur in skeletal muscle characteristics between different types of dogs and that these differences can be independent of differences in exercise history.  相似文献

The mechanism by which oestrogen inhibits development of muscle mass was investigated in the soleus muscle of 20 sexually immature female mice. Half of the population were injected weekly (from one to nine weeks old) with a physiological dose of 0.01 mg stilboestrol dipropionate (synthetic oestrogen). After each mouse was killed at 10 weeks old, the weight and muscle index was determined for each muscle. The number of muscle fibres was counted in each soleus muscle. The muscle fibre size for each muscle was determined by measuring the greatest diameter of at least 100 fibres. The number of muscle fibres was similar in both normal and oestrogen treated animals but the individual muscle fibre sizes were significantly smaller in oestrogen treated animals. These observations indicated, therefore, that oestrogen inhibits development of muscle mass in females by limiting the size of individual muscle fibres and not by inhibiting the multiplication of muscle fibres.  相似文献

The purpose of the present investigation was to determine the effects of severe dietary protein restriction on soleus (S) and tibialis anterior (TA) fiber number and S muscle fiber area, composition and length. The S and TA muscles were removed from one leg of 12 male Sprague-Dawley rats at 21 d of age. Six of these animals were placed on a 1% protein diet until 42 d of age, while six served as age-matched controls. Muscle fiber number was determined by the nitric acid digestion method for S muscles and the mean fiber dry-weight estimation method for the TA muscles. Mean fiber numbers for the S muscles were 2,655 +/- 42 and 2,669 +/- 71 for the treatment group at 21 and 42 d of age, respectively, and 15,989 +/- 899 and 16,067 +/- 695 at 21 and 42 d of age, respectively, for the TA muscle. For the age-matched control group, fiber numbers for the S muscle were 2,928 +/- 78 and 2,949 +/- 76 at 21 and 42 d, respectively, and 17,964 +/- 281 and 18,445 +/- 296 at 21 and 42 d, respectively, for the TA muscle. The S muscle fiber area, composition and length were studied using 12 male Sprague-Dawley rats. Six animals were placed on the 1% protein diet from 21 to 42 d of age, while six animals served as age-matched controls. The S muscle fiber area was 33.1 and 51.5% smaller for type I and type II fibers, respectively, for animals fed the 1% protein diet. The extensor digitorum longus (EDL) and soleus (SOL) muscle fibres from albino rats submitted to experimental chronic alcoholism were evaluated in accordance with their metabolic and morphometric profiles. Twenty-seven male animals aged 4 months and weighing approximately 400 g were used. The animals were divided into three groups: control, isocaloric and alcoholic and sacrifices were carried out after 5, 10 and 15 months. The muscles were dissected, removed, cross-sectioned in a cryostat and submitted to the NADH (nicotinamide adenine dinucleotide) reaction. The SO (slow-twitch-oxidative), FG (fast-twitch-glycolytic) and FOG (fast-twitch-oxidative-glycolytic) muscle fibre types exhibited a polygonal, triangular or rounded shape and did not present noteworthy modifications in either muscles during the study. The cross-sectional areas of the fibres from the studied muscles did not present significant differences during the observations. Fibre area behaved similarly in the alcoholic animals up to the 10th month, i.e. it was decreased, as also observed in the other groups. At 15 months, however, all fibres were increased, with a predominance of FG fibres in the SOL muscle. Changes in fibre population were observed mainly in the SOL muscle of alcoholic animals: SO fibres were initially increased in number but decreased after the 10th month, and the opposite was observed for the population of FG fibres. FOG fibres increased linearly in number throughout the experiment. The statistical analysis showed nevertheless that the fibre population and cross-sectional area changes were not significant. In the alcoholic animals quantitative variations of muscle fibres were more evident in the SOL muscle, suggesting that the SOL muscle is more sensitive to the toxic action of ethanol. The results concerning the increased fibre diameter in alcoholic animals would be associated with muscle oedema induced directly or indirectly by the ethanol.  相似文献

Computed tomographic images of the thoracic aorta of 14 German shepherd dogs were examined in order to determine the morphometry of the thoracic aorta. Examinations were carried out in the transverse plane at both intervertebral and mid-vertebral levels of each thoracic vertebra between T(5) and T(13). The dorsoventral and transversal diameters as well as cross-section area of the thoracic aorta were measured. The widest transversal diameter was observed at T(4-5), whereas the largest dorsoventral diameter was detected at T(5). The maximum cross-section area was detected at T(4-5). When dorsoventral and transversal diameters were compared between males and females, the aortic diameter was found to be smaller in males than in females. Although the shape of the thoracic aorta was transversal oval in the majority of the examined females, the shape of the thoracic aorta was dorsoventral oval in the majority of the males. There were significant differences between all levels measured for transversal (P < 0.001), dorsoventral (P < 0.001) diameters and cross-section area (P < 0.001) of the thoracic aorta. And there was a significant correlation between the three parameters examined. However, the correlation coefficient was highest in females.  相似文献

The sex differences in the number and morphometric parameters of motoneurons in motor nuclei are poorly known. The aim of this study was to determine the differences in the number and size of alpha and gamma motoneurons of the medial gastrocnemius (MG) muscle in male and female Wistar rats. Retrogradely labelled cell bodies of motoneurons of 6 months old animals were studied following a bath of the proximal stump of the transected MG nerve in a horseradish peroxidase solution. The number and soma diameters of male and female MG motoneurons were determined from serial microscopic images of sections. The weight of the brain and spinal cord was on average 17% higher in males than in females. The mean number of motoneurons was 13% higher in males than in females and amounted to 94 and 83 motoneurons, respectively. In each case, the average soma diameters and cross‐section areas of motoneurons in motor nucleus were distributed bimodally: motoneurons smaller than 27.5 μm in diameter were recognized as gamma and greater ones as alpha motoneurons. In males, the motor nucleus contained on the average 66 alpha motoneurons, whereas in females, 56 alpha motoneurons, that is the mean number of alpha motoneurons was 17% higher in males. Moreover, the soma diameters of gamma and alpha motoneurons were significantly bigger in males and the difference amounted 9 and 6%, respectively. It is concluded that the number as well as size of alpha and size of gamma motoneurons in the MG motor nucleus are greater in males.  相似文献

This study characterised muscle fibres in trunk, forelimb and hindlimb muscles of three bat species: little Japanese horseshoe (Rhinolophus cornutus), greater horseshoe (Rhinolophus ferrumequinum) and Egyptian fruit (Rousettus aegyptiacus). Twenty-seven muscles from trunk, forelimb and hindlimb were dissected, weighed and analysed by immunohistochemistry and sodium didecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and determined their cross-sectional areas (CSA). Results showed that Type IIa and Type IIa/x made the highest proportion of total muscle mass. Moderate proportion was formed by Type IIb. Type I and IIx appeared at very low levels in all bats. Type IIb was the only fibre type detected in patagial muscles in wing membrane of greater horseshoe while other fibre types were not observed. Type I muscle fibres were very few and appeared infrequently in fifteen muscles of Egyptian fruit and in only one muscle in each, greater horseshoe and little Japanese horseshoe. Type IIx was also detected in three muscles in greater horseshoe and only one muscle in Egyptian fruit but none in little Japanese horseshoe. The highest average CSA μm2 was detected in Type IIb and values were 734.2μm2 for LHB; 1537.9μm2 for GHB and 1,720.9μm2 for EFB. Lowest and undetermined values were observed for Type I and IIx. These data demonstrate that Type IIa, IIa/x and IIb form significant proportion of adult bat muscle mass and Type IIb is the largest fibre type. The distribution pattern is suggestive of specialised functions of the fibres in relation to orientation and speed of bats during flight.  相似文献

In this study, we compare the effects of pre‐ and post‐natal food deprivation on the relative proportion of fibre types and contractile responses in the extensor digitorum longus (EDL) muscle of female and male rats at different post‐natal ages. EDL muscles from undernourished male (UM) rats showed a higher proportion of Type IIB than IIA fibres and larger normalized twitch responses (with respect to muscle weight) than those of controls (CM). In contrast, EDL muscles from control (CF) and undernourished female rats (UF) showed no significant differences in their fibre type composition and normalized twitch forces at most of the ages analysed. Our data are indicative that the EDL muscles from undernourished males are more susceptible to the effects exerted by low food income than the EDL muscles from female rats. It is proposed that changes in the reactive oxygen species (ROS) concentration and hormonal factors, due to undernutrition, are involved in the alterations observed in the fibre type composition and force production of EDL muscles in undernourished male rats and that estrogens may have an antioxidant protective role on the undernourished EDL muscles in female rats.  相似文献

The greater rhea (Rhea americana) is the largest South American bird. It is a cursorial, flightless species with long powerful legs and reduced forelimbs. The goal of this study was to explore how hindlimb muscles scale with body mass during postnatal growth and to analyze whether the specialized locomotion of this species affects the growth of muscle masses. The mass of 19 muscles and body mass were weighed in 21 specimens ranging from 1‐month‐old individuals to adults. Seventeen muscles scaled with positive allometry with respect to body mass, whereas two muscles scaled isometrically . The predominance of positive allometric growth in hindlimb muscles results in a limb with massive and powerful muscles specialized to support a large body mass and to attain relatively high running speeds. Analysis of muscle mass scaling is a simple and useful way to compare possible differences between locomotor styles, and it is valuable in studies that reconstruct the paleobiology of extinct taxa.  相似文献

The possible relationship between myofiber type composition and adipose tissue development in skeletal muscle in vivo has been suggested. Recent evidence indicated that satellite cells are multipotent cells that can undergo not only myogenic, but also adipogenic differentiation. In the present study, rat satellite cells were isolated from soleus, back, extensor digitorum longus, tibialis anterior and quadriceps muscles, and their adipogenic potentials were compared by culturing them under adipogenic conditions in vitro. Cells from soleus muscle exhibited the highest adipogenic potential as judged from Oil Red-staining and immunocytochemical C/EBPalpha-staining. The adipogenic potential of satellite cells was positively correlated with type I myofiber distribution in the corresponding muscle of origin. These results demonstrated that the adipogenic potential of satellite cells differs according to the muscle of origin and suggested that its possible correlation to type I myofiber distribution may account for preferential adipose tissue development in slow oxidative muscles.  相似文献

Materials suitable for anatomical research of raptorial birds are rare. Bird‐eating raptors show distinct inter‐sexual differences in body size and parental roles. The large females catch larger prey and prepare small morsels to feed their young using their hooked beaks. Here, we investigated the architectural properties of different jaw muscles of the Japanese sparrowhawk (Accipiter gularis) and examined whether there is sexual dimorphism in their architectural design. The results showed that musculus depressor mandibulae, the opener of the lower jaw, was characterized by relatively long fascicle length, whereas musculus pterygoideus was characterized by its larger mass and physiological cross‐sectional area (PCSA) in both sexes. Females have the potential capacity to produce rapid and strong bites by their significantly longer fascicle length of M. depressor mandibulae and larger mass and PCSA of M. pterygoideus. For body size‐matched gender, jaw muscles of males had fibres of relatively longer length than females, enabling greater velocity and excursion. Architectural characteristics of jaw muscles, together with the absolute dimorphism (the fascicle length of M. depressor mandibulae, the muscle mass and PCSA of M. pterygoideus) and relative dimorphism in the muscle mass of M. pterygoideus, reflect dietary difference and asymmetric parental roles between the sexes.  相似文献

No studies have investigated the mitochondrial function in permeabilized muscle fiber from cats. The aim of this study was to investigate tissue-specific and substrate-specific characteristics of mitochondrial oxidative phosphorylation (OXPHOS) capacity in feline permeabilized oxidative muscle fibers. Biopsies of left ventricular cardiac muscle and soleus muscle, a type I-rich oxidative skeletal muscle, were obtained from 15 healthy domestic cats. Enzymatic activity of citrate synthase (CS), a biomarker of mitochondrial content, was measured. Mitochondrial OXPHOS capacity with various kinds of non-fatty-acid substrates and fatty-acid substrate in permeabilized muscle fiber was measured by using high-resolution respirometry. CS activity in the heart was 3 times higher than in the soleus muscle. Mitochondrial state 3 respiration, ADP-stimulated respiration, with complex I-linked and complex I+II-linked substrates, respectively, was significantly higher in the heart than in the soleus muscle when normalized for muscle mass, but not for CS activity, indicating that greater capacity for mitochondrial OXPHOS with these non-fatty-acid substrates in the heart may depend on higher mitochondrial content. In contrast, the soleus muscle had higher mitochondrial state 3 respiration with fatty acids than the heart when normalized for CS activity, indicating greater capacity for fatty-acid oxidation per mitochondrion in the soleus. Our findings suggest that there are tissue- specific and substrate-specific quantitative and qualitative differences in mitochondrial OXPHOS capacity between the different types of oxidative muscles from cats.  相似文献

This study investigated effects of birth weight and postnatal nutrition on growth and development of skeletal muscles in neonatal lambs. Low (L; mean +/- SD 2.289 +/- .341 kg, n = 28) and high (H; 4.840 +/- .446 kg, n = 20) birth weight male Suffolk x (Finnsheep x Dorset) lambs were individually reared on a liquid diet to grow rapidly (ad libitum fed, ADG 337 g, n = 20) or slowly (ADG 150 g, n = 20) from birth to live weights (LW) up to approximately 20 kg. At birth, weight of semitendinosus (ST) muscle in L lambs was 43% that in H lambs; aggregate weights of ST and seven other dissected muscles were similarly reduced. In ST muscle of L lambs, mass of DNA, RNA, and protein were also significantly reduced to levels 67, 60, and 34%, respectively, of those in H lambs. However, myofiber numbers of ST, tibialis caudalis, or soleus muscles did not differ between the L and H birth weight lambs and did not change during postnatal growth. During postnatal rearing, daily accretion rate of dissected muscle was lower in L than in H lambs. Accretion of muscle per kilogram of gain in empty body weight (EBW) was reduced in the slowly grown L lambs compared with their H counterparts, although the difference was less pronounced between the rapidly grown L and H lambs. Throughout the postnatal growth period, ST muscle of L lambs contained less DNA with a higher protein:DNA ratio at any given muscle weight than that of H lambs. Slowly grown lambs had heavier muscles at any given EBW than rapidly grown lambs. Content of DNA and protein:DNA ratio in ST muscle were unaffected by postnatal nutrition, but RNA content and RNA:DNA were greater and protein:RNA was lower at any given muscle weight in rapidly grown lambs. Results suggest that myofiber number in fetal sheep muscles is established before the presumed, negative effects of inadequate fetal nutrient supply on skeletal muscle growth and development become apparent. However, proliferation of myonuclei may be influenced by fetal nutrition in late pregnancy. Reduced myonuclei number in severely growth-retarded newborn lambs may limit the capacity for postnatal growth of skeletal muscles.  相似文献

An anabolic steroid, nandrolone decanoate, was shown to produce increases in the weight, cross-sectional area, tension-generating capacity and mean fibre diameter of a limb muscle, tibialis anterior (TA), in female rabbits. Despite suggestions in the literature of an exerciserelated effect of such drugs, hypertrophy was not observed in TA muscles subjected to chronic electrical stimulation, but was seen in the contralateral control muscles and in the muscles of both hind limbs in unoperated sedentary animals. The effect was confirmed in the TA of female guinea pigs, rats and mice, and could not be explained in terms of an increase in body weight. Compared to TA some other muscles, like EDL, plantaris and soleus in the rabbit, were less sensitive to treatment; others, like pubocaudalis in the rat and mouse, and temporalis in the guinea pig, were more responsive. The myotrophic effects were undiminished in adrenalectomised female rats, and could not therefore have been mediated by interference with the catabolic actions of circulating glucocorticoids, as had been proposed in an earlier hypothesis.  相似文献

This study was designed to determine the histochemical properties, size and composition of fibres in the diaphragm, intercostal and abdominal muscles of goats to clarify whether reported similarities in respiratory muscle physiology between goats and humans have a structural basis. Serial sections (10 µm) of muscular tissue from adult female goats were stained for myosin adenosine triphosphatase and reduced nicotinamide adenine dinucleotide dehydrogenase-tetrazolium reductase activities; the fibres were classified into types I, IIA and IIB; and their mean diameter and composition were determined. Abdominal and intercostal muscles contained types I, IIA and IIB fibres in the ratio 1:1:1, and the mean diameter of the fibres ranged from 49.2 to 62.2 µm. In contrast, the diaphragm contained 58.9% type I and 41.1% type II fibres, and the latter could not be differentiated into types IIA and IIB. Diaphragmatic fibres were also smaller (36.9 – 40.9 µm). These findings contrast with those in humans, where the diaphragm, intercostal and abdominal muscles contain >50% type I fibres and have fibres of identical diameter. The differences in fibre characteristics between the diaphragm, intercostal and abdominal muscles of goats and the differences between goats and humans need to be taken into consideration in interpreting the results from studies in respiratory muscle physiology.Abbreviations mATPase myosin adenosine triphosphatase - NADH-TR nicotinamide adenine dinucleotide dehydrogenase-tetrazolium reductase  相似文献
