The morantel sustained release bolus was administered at turnout to first-season grazing calves in order to assess its efficacy in the seasonal control of infection by nematode parasites in Ireland. The pastures grazed by control calves showed a marked increase in gastrointestinal trichostrongylid infective larvae by September, while numbers of infective larvae on pasture grazed by bolus-treated calves remained at a low level throughout the grazing season. In consequence, the controls showed significantly higher worm egg counts in late season and significantly higher worm burdens (mainly Ostertagia spp) at necropsy carried out in November on representative number of principal animals selected from each group. These reduced worm burdens were attributed to the suppression of egg output during the early part of the season as a result of treatment with the morantel sustained release bolus at turnout in the spring. Pasture contamination with Dictyocaulus viviparus larvae was present on all treatment pastures. The bolus-treated calves however were subjected to an increase in D. viviparus infection which occurred on their pasture in late season after the active life of the bolus had expired. It was concluded that bolus treatment delayed (rather than prevented) the buildup of D. viviparus infection on the pasture by 60-90 days.  相似文献

The persistent anthelmintic effect of ivermectin as a topical treatment at 500 microg/kg was evaluated against induced infections of Ostertagia ostertagi, Trichostrongylus axei and Dictyocaulus vivparus in calves. The results showed a highly significant (P<0.001) anthelmintic activity for at least 14 days against O. ostertagi and T.axei (>99 per cent efficacies) and for at least 28 days (98 per cent efficacy) against D. viviparus.  相似文献

The efficacy of the oxfendazole pulse release bolus system for the control of parasitic gastroenteritis and parasitic bronchitis in first-season grazing calves was evaluated in Belgium. Twenty-two calves were allocated to two groups. The calves in one group received a bolus at the time of turn out, while the other group remained untreated. The efficacy of the bolus was assessed by comparison of faecal worm egg counts, plasma pepsinogen concentrations, the antibody response to Ostertagia, Cooperia and Dictyocaulus species total plasma protein and albumin concentrations, and weight gains throughout the grazing season and the housing period. The oxfendazole pulse release bolus provided good control of parasitic gastroenteritis dominated by ostertagia. The effects of parasitic gastritis were greatly reduced as shown by the significantly lower values of serum pepsinogen and ostertagia antibody titres. The use of the bolus further reduced the adverse effects of parasitism as indicated by better liveweight gains and normal total plasma protein and albumin concentrations whereas in the untreated control group hypoproteinaemia and hypoalbuminaemia were observed. Most animals exhibited clinical signs of parasitic bronchitis at the end of the grazing season, and the bolus may not adequately control parasitic bronchitis in all cases at all times.  相似文献

Two trials involving a total of 36 Dorset horn lambs were conducted to assess the anthelmintic efficacy of ivermectin against experimental infections of benzimidazole-resistant strains of Haemonchus contortus and Ostertagia circumcincta. Two resistant strains of each of the two species were used and in each trial the lambs were allocated to three groups. One group was given 200 micrograms ivermectin/kg bodyweight orally, the second group was given 5 mg oxfendazole/kg bodyweight orally and the third group remained untreated as controls. Fourteen days after treatment the lambs were necropsied. Ivermectin was found to be more than 99 per cent to 100 per cent effective against all four benzimidazole-resistant strains, whereas oxfendazole was 78.6 per cent and 83.8 per cent effective against the H contortus strains, and 25.6 per cent and 39.8 per cent effective against the O circumcincta strains.  相似文献

Two studies are described which demonstrate the persistent activity of ivermectin injected subcutaneously into cattle at 200 micrograms/kg in preventing the establishment of induced infections with the gastrointestinal parasites Ostertagia ostertagi and Cooperia oncophora and the lungworm Dictyocaulus viviparus. These results indicated a reduction in mean worm count compared with the control group for O ostertagi of more than 99, 45 and 94 per cent with a seven, 14 or 21 day interval between treatment with ivermectin and the administration of infective larvae, respectively, in trial 1 and more than 99, more than 99 and 99 per cent at seven, 10 or 14 days, respectively, in trial 2. Corresponding values against C oncophora were 99, 0 and 45 per cent at seven, 14 and 21 days in trial 1 and more than 99, 84 and 31 per cent at seven, 10 and 14 days in trial 2. Against D viviparus, reduction in counts were more than 99, 98 and more than 99 per cent at seven, 14 and 21 days, respectively, in trial 1 and 100, 100 and 100 per cent at seven, 10 and 14 days, respectively, in trial 2. The relevance of these results to the build-up of infective larvae on pasture and infection in cattle is discussed.  相似文献

Under experimental conditions, fenbendazole given at doses of 0.4 and 1.0 mg/kg body weight suppressed calves' faecal output of Ostertagia and Cooperia species eggs and Dictyocaulus viviparus larvae. Both dose levels were given in the form of small daily drenches and the higher level showed greater efficacy. In a grazing experiment, medication with fenbendazole at 1.0 mg/kg/day administered intermittently to calves using an automatic dose dispenser almost completely suppressed the output of trichostrongylid eggs. As a result, infection on the pasture and in the calves remained at a low level throughout the grazing season. By contrast, control pasture and control calves showed rather heavy infection from mid-August onwards with significantly lower weight gains and widespread signs of parasitic gastroenteritis. At post mortem examination of representative calves from each group in November, the medicated animals had 99 per cent less Ostertagia species, whether adults or larvae arrested at the early fourth stage, and 95 per cent less Cooperia species compared with controls. Medication in the drinking water suppressed the faecal output of D viviparus larvae for most of the grazing season by comparison with the controls but the medicated calves became infected with this parasite towards the end of the season. Until this problem is overcome, precautions against parasitic bronchitis are advised when this system of medication is adopted.  相似文献

The residual effect of treatment with ivermectin after experimental reinfection in calves was tested. Twenty-four calves were divided into 6 groups of 4 calves each. All calves received a primary infection of 50,000 larvae of both Ostertagia ostertagi and Cooperia oncophora and 1000 Dictyocaulus viviparus larvae. Calves of group 1 remained untreated, and all other calves were treated 21 days after primary infection (0.2 mg/kg injected subcutaneously). Calves of groups 1 and 2 were slaughtered 7 days later. Calves of groups 3-6 were reinfected with the same number of larvae 3 days, 1, 3 and 6 weeks after treatment respectively. Slaughter was 21 days after reinfection. Based on post-mortem worm counts the efficacy of ivermectin after primary infection was 99.7% for O. ostertagi, 95.1% for C. oncophora and 100% for D. viviparus. A residual effect was present for at least one week, but could not be observed 3 weeks after treatment.  相似文献

A trial was conducted to determine the efficacy and pharmacokinetics of fehantel and ivermectin in six month-old red deer calves (C. eluphus). Five calves received febantel by mouth at 7.5 mg/kg, five received a subcutaneous injection of ivermectin at 200 microg/kg and five were controls. All calves were killed seven days later and total lung and gastrointestinal worm counts carried out. Febantel was 85 and 99.8% efficient in removing immature and mature Dictyocaulus viviparus, respectively, and ivermectin was 100% efficient in both cases. There was no gastro-intestinal nematodes in any of the treated calves, compared to an average of 619 in the control calves. The metabolism of febantel resulted in plasma levels of fenbendazole, oxfendazole and sulphone for which the common curves fitted by compartmental model peaked at values (standard errors)-of 0.46 (0.03), 0.41 (0.02) and 1.73 (0.07) mg/l after approximately five, nine, and thirteen hours and were undetectable at 30,72 and 120 hours respectively. There was considerable variation among animals in response to ivermectin. The fitted common curve had a peak plasma level of 15.8 (0.08) microg/l at 20 hours after injection, which had dropped to 7.9 (1.1) microg/l seven days after injection. It was estimated that after 15 days plasma levels of ivermectin would not be detectable. It is concluded that the injectable form of ivermectin tested is a highly efficient anthelmintic in deer, and that plasma levels persist for over a week after subcutaneous injection. Fehantel is very efficient against mature D. viviparus in deer, but its reduced efficiency against immature D. viviparus may relate to the deer;s ability to metabolise and excrete benzimidazoles more quickly than sheep and cattle.  相似文献

Three groups of calves were put out to graze on separate paddocks within a field known to be infected with Dictyocaulus viviparus and were also given a small initial trickle infection of the parasite. The first group were untreated controls, the second were immunised with live irradiated lungworm vaccine and the third were injected three times with ivermectin; the injections taking place after they had grazed for three, eight and 13 weeks. The subsequent infections of D viviparus were estimated by grazing a series of parasite-free tracer calves in the paddocks used by each group. The first group of such calves grazed from July 17 until August 7, the second from August 22 to September 29. During the first half of the grazing season all the untreated and three of the six immunised calves were observed to excrete D viviparus larvae, in contrast to none of the ivermectin-treated group. As a result all the tracer calves on the areas occupied by the untreated and immunised calves became infected with the parasite whereas only one worm was found in one of the 10 tracer calves grazing the area occupied by the ivermectin-treated calves.  相似文献

Twenty-eight Holstein-Friesian heifers, born the previous year and weighing between 130 and 310 kg, were allocated to one of two treatment groups by restricted randomisation, based on their initial weight. The heifers in group 1 were each treated with ivermectin in a sustained-release bolus formulation at turnout in April, and those in group 2 were each given an ivermectin bolus on July 10, 84 days after turnout. On that day the mean geometric worm egg counts of groups 1 and 2 were 0.4/g and 38.8/g, respectively, and they both had a mean plasma pepsinogen concentration of 0.59 iu/litre; in group 1, two of 14 faecal samples were positive for Dictyocaulus viviparus larvae, and in group 2 all 13 samples were positive; in group 1 eight calves were positive and three inconclusive for the presence of antibodies to D viviparus, and in group 2 the corresponding figures were 10 positive and two inconclusive; the mean liveweights of groups 1 and 2 were 274.4 kg and 262.8 kg, respectively. By December 4,231 days after turnout, the corresponding results were: mean geometric worm egg counts of 2.2/g and 0.5/g; one of 13 and none of 14 faecal samples positive for D viviparus larvae; 12 positive and two inconclusive and none positive and 10 inconclusive for the presence of antibodies to D viviparus; 214 days after turnout their mean liveweights were 361.1 kg and 358.3 kg. Although the patterns of parasitic nematode infection were different in the two groups during the grazing season, by the time they were housed both groups had achieved similar liveweights and showed evidence of an immune response to both D viviparus and gastrointestinal nematodes.  相似文献

Forty yearling calves were assigned to four equal groups; three of the groups were treated with oxfendazole at dose rates of 6.75 mg/kg, 4.50 mg/kg, or 2.25 mg/kg bodyweight while the fourth group served as an untreated control. The calves were native to north-east Mississippi, USA, and harboured natural infections of gastrointestinal nematodes. The study was conducted during July when inhibited early fourth-stage larvae may be found in large numbers after their acquisition in the spring. The calves were maintained in separate groups on concrete-floored pens for 17 days before the intraruminal administration of oxfendazole. Seven days after treatment, the calves were slaughtered and the gastrointestinal parasites counted. At all the dose rates examined oxfendazole exhibited an efficacy of at least 99.4 per cent against adults of Haemonchus placei, Trichostrongylus axei, Bunostomum phlebotomum, Cooperia species, T colubriformis, Oesophagostomum radiatum, and Trichuris ovis. The efficacy against adult Ostertagia ostertagi was at least 99.4 per cent at dose rates of 6.75 and 4.50 mg/kg bodyweight, but decreased to 93.7 per cent at 2.25 mg/kg. The efficacy of oxfendazole against inhibited larvae of O ostertagi decreased with dose rate from 78.8 per cent at 6.75 mg/kg, to 58.9 per cent at 4.50 mg/kg and 20.3 per cent at 2.25 mg/kg bodyweight.  相似文献

Four groups of six parasite-naive calves were infected at seven day intervals with three doses of infective larvae of Dictyocaulus viviparus. Twenty-one days after the first dose three of the groups were treated either with an injectable formulation of ivermectin at a dose rate of 200 micrograms/kg bodyweight, or with pour-on preparations of levamisole at 10 mg/kg or ivermectin at 500 micrograms/kg. On day 28 two calves from each group were slaughtered and their burdens of lungworms counted. On day 35 the remaining calves were reinfected with D viviparus infective larvae at a rate of 80 L3/kg. The levamisole preparation was 94.6 per cent effective and both ivermectin preparations were 100 per cent effective against the initial infections. The ivermectin-treated calves were protected from the reinfection which subsequently became patent in the levamisole-treated and control calves.  相似文献

A comparison was made of the parasitological and production responses to the prophylactic (summer) and combined prophylactic and therapeutic (summer/autumn) administrations of the morantel intraruminal, sustained-release bolus (MSRB) with responses to conventional drenching in grazing dairy calves over an 8-month period. The MSRB was designed to release morantel tartrate at 150 mg/day for 90 days. In two concurrent trials, groups of calves grazed separate replicated pastures. In Trial 1, the combined summer (December) and autumn (March) administration of the MSRB was compared with a programme of six drenches (oxfendazole at 4.53 mg/kg) at 28-day intervals from December to May. In Trial 2, the summer administration of the MSRB was compared with a programme of three autumn drenches (oxfendazole) at 28-day intervals from March to May. There were no significant between-treatment differences in mean liveweight gain in either trial. In both trials, the low numbers of eggs passed by the MSRB-treated animals during summer (maximum mean 62 eggs per gram of faeces) were sufficiently high to produce a modest autumn larval peak (maximum 1990 larvae/kg herbage). In Trial 2, the summer use of the MSRB resulted in an 83% reduction in peak larval availability compared with that on pastures grazed by untreated calves during summer. Only the 6-drench regimen (Trial 1) prevented the accumulation of appreciable worm burdens of early 4th stage Ostertagia spp.  相似文献

Three groups of ten 4-month-old red deer (Cervus elaphus) calves naturally infected with lungworm (Dictyocaulus viviparus) were treated with either oral ivermectin (200 microg/kg), topical (pour-on) ivermectin (500 microg/kg) or oral oxfendazole (5 mg/kg). Faecal larval counts for lungworm were undetectable or very low for 14 days after treatment with oxfendazole, 28 days after treatment with oral ivermectin and for 49 days after treatment with topical ivermectin. This pilot study suggests that the topical formulation of ivermectin was very effective against lungworm and had a more persistent action than the oral ivermectin formulation in young red deer.  相似文献

Two strains of Ostertagia circumcincta were isolated from sheep in Great Britain; one (CVL strain) from a breeding flock maintained at the Central Veterinary Laboratory, the other (H2 strain) from a commercial flock in southern England. Their resistance to benzimidazole anthelmintics was assessed by means of in vitro egg hatch assays and slaughter trials. In vitro egg hatch assays gave calculated ED50 estimates of 0.799 micrograms thiabendazole/ml for the CVL strain and 0.794 micrograms thiabendazole/ml for the H2 strain, compared with ED50 estimates of 0.038 micrograms thiabendazole/m and 0.036 micrograms thiabendazole/ml for two known susceptible strains of O circumcincta. There was a 40.7, 28.4 and 66.9 per cent reduction in the group mean worm burdens of lambs infected with the CVL strain following treatment with thiabendazole, fenbendazole and oxfendazole, respectively, and 23.8, 0.0, 79.6, 52.7, 99.9 and 100 per cent reduction in the group mean worm burdens of lambs infected with the H2 strain following treatment with thiabendazole, fenbendazole, oxfendazole, albendazole, levamisole and ivermectin, respectively. The anthelmintic activity of oxfendazole (Syntex) was tested in calves at dosages of 2-5 and 5-0 mg per kg. At both dose levels, oxfendazole showed 100 per cent efficacy against adult Ostertagia ostertagi, O lyrata, O cremensis, fifth stage Ostertagia spp and adult Haemonchus spp. Against adult Cooperia oncophora, efficacy was 99.8 per cent and 100 per cent at doses of 2.5 mg per kg and 5.0 mg per kg respectively while at both dose levels 100 per cent activity was recorded against C surnabada and fourth and fifth stage Cooperia spp. One hundred per cent efficacy was obtained with both dose levels against adult and fifth stage Dictyodaulus viviaprus; against Trichuris spp, percentage efficacy was 92 and 100 per cent at doses 2.5 and 5.0 mg per kg respectively. Oxfendazole showed higher efficacy than levamisole against Ostertagia spp but against the other species encountered, both anthelmintics possessed similar efficacy. Both anthelmintics significantly increased the calves' weight gains.  相似文献

Seeder calves infected with Dictyocaulus viviparus were used to contaminate a field divided into three similar paddocks. Twenty-four autumn-born calves were allocated to three matched groups; one group was given topical ivermectin treatments (0.5 mg/kg) at 3, 8 and 13 weeks after turnout (Day 0); each member of a second group was given an oxfendazole pulse-release intraruminal device (OPRB) at turnout; while a third group was kept as untreated controls to monitor the natural epidemiological pattern of events. Severe pasteurella pneumonia exacerbated by lungworm infection occurred in the controls after Day 24. Two died and repeated doses of antibiotic and anthelmintic therapy were necessary to save the remainder. Clinical signs were much milder in the ivermectin and OPRB groups and resolved with only a single dose of antibiotic. The OPRB group excreted some lungworm larvae at this time, but none was detected in the faeces of the ivermectin group during the grazing season. At housing, five calves from each group and four lungworm-naive calves were challenged with D. viviparus larvae. The infection became patent in all challenge-control calves, but no larvae were passed by any of the trial animals. Post-mortem worm-counts revealed percentage takes for the challenge controls, trial controls, ivermectin and OPRB groups of 16.7, 0.01, 0.9 and 0.2, respectively. All trial groups had therefore developed a substantial immunity.  相似文献

A field study of calves in their first grazing season tested the efficacy of four long-acting devices--a morantel sustained-release bolus, a levamisole sustained-release bolus, an oxfendazole interval bolus, and an albendazole interval bolus--against Dictyocaulus viviparus. The pasture had been previously contaminated by four calves orally inoculated with infective lungworm larvae. The calves were grazed together with four bolus-treated groups, each comprising four calves. Lungworm infection became patent in the experimentally inoculated calves between 22 and 26 days. Infection in the bolus-treated groups became patent after 54 days. The morantel bolus group excreted the most larvae, followed by the albendazole bolus group, and the levamisole bolus group. The oxfendazole bolus group excreted by far the least larvae. Eosinophil curves and ELISA titres showed that treated groups had essentially the same course of infection. The heavy infection to which the treated calves were exposed produced complete immunity in all groups. Challenge infection of 10,000 larvae at housing did not change any of the test parameters. Post-mortem examination showed only one positive calf with few worms. We concluded that when pastures are heavily infested with lungworm larvae, all boluses prevent severe clinical signs and allow build up of solid immunity, although none completely prevent excretion of larvae.  相似文献

Twelve Holstein calves were used to determine the prophylactic efficacy of ivermectin against challenge exposure with gastrointestinal and pulmonary nematodes. Two groups of 6 calves (mean body weight, 205 kg) each were formed by restricted randomization according to body weight. Group-1 calves served as nonmedicated controls. Each calf of group 2 was orally given one prototype sustained-release bolus designed to deliver ivermectin at a continuous daily dose of 8 mg. Third-stage nematode infective larvae were given to the calves on posttreatment days 28 and 42. The calves were euthanatized 77 or 78 days after treatment. Ivermectin was 100% effective (P less than 0.05) in preventing the establishment of infection by Haemonchus placei, Ostertagia ostertagi, Cooperia spp (C punctata, C oncophora, C surnabada), Nematodirus helvetianus, Oesophagostomum radiatum, and Dictyocaulus viviparus and was greater than 99% effective against Trichostrongylus axei. Sub-clinical parasitism in spring-born single suckled beef calves was investigated from the middle of their first grazing season until weaning or housing later the same year. The study was conducted on four beef suckler herds in southern England over a 3-year period and involved a total of 334 spring-born beef suckler calves and their dams. The animals were grazed extensively on pastures naturally infected with nematode larvae. At the start of each period of observation, faecal samples were taken from calves and cows and subjected to routine worm egg counts; calves were re-sampled at the end of the grazing season.In July in each year and at each location the calves were ranked by initial weight within sex, paired according to rank and randomly allocated to either an untreated control group or a group in which the calves were each treated with an ivermectin sustained-release (SR) bolus. The calves in both trial groups, and their dams, were grazed together until weaning or housing. The calves were weighed at the initial allocation and at the end of the study. The adult cows were not treated with any anthelmintic during the study.The faecal nematode egg counts (FECs) conducted in July showed that the suckler cows were excreting worm eggs at low concentrations: range 0-100 eggs per gram (epg), with one individual count of 500epg, 88% of the cows sampled had counts of <50epg. Similarly, the counts from the calf samples were fairly low in July: range 0-250epg, 73% of the calves sampled had counts of <50epg. By the end of the grazing season, the faecal samples from the untreated control calves showed higher values: range 0-650epg, with only 58% having an epg of <50.The average rate of daily liveweight gain in the untreated heifer calves was 0.79kg per day, the corresponding figure for the heifer calves treated with the ivermectin SR bolus in mid-summer was 0.88kg per day; the difference of 90g per day was significantly different (P=0.0118). The average rate of daily liveweight gain in the untreated bull calves was 0.91kg per day, the corresponding figure for the bull calves treated with the ivermectin SR bolus in mid-summer was 1.01kg per day; the difference was significantly different (P=0.0169).  相似文献
