Abstract. Juvenile Atlantic salmon from a number of freshwater hatcheries in Scotland were found to be infected by a myxosporean parasite of the genus Sphaerospora. Fish first became infected in June by extrasporogonic stages which could be found in the blood and kidney interstitium. These consisted of a primary cell containing one to over one hundred secondary cells. Some secondary cells contained one or two tertiary cells. Sporogony was disporous and occurred later in the kidney tubules. The development of both extrasporogonic and sporogonic stages was studied by light and transmission electron microscopy.  相似文献

Sphaerospora truttae is an important pathogen of Atlantic salmon parr in Scottish aquaculture. To trace the early development of S. truttae and to overcome the common problem of detecting low numbers of cryptic, early myxosporean stages, a DNA-based approach was applied in this study. Specific primers were designed for S. truttae from Atlantic salmon, based on 18S rDNA sequences, obtained from isolated myxosporean spores. These were 5' biotin-labelled and used in an optimized and rapid in situ hybridization (ISH) protocol, which provided a strong and specific signal of the parasite within host tissue sections and, at the same time, minimized structural damage to tissues due to processing. This methodology provided a reliable tool enabling the detection of S. truttae stages down to single cell level. Using ISH the epithelium of the gills was identified as the predominant entry locus of the parasite. By 3 days after infection S. truttae had penetrated the vascular epithelia and thereafter proliferated in the blood for at least 10 days before exiting the vascular system through capillary walls. From day 12 post-infection onwards, the kidney, as well as the spleen and the liver, were invaded. Numbers of S. truttae invading the kidney (37.3%) differed little from numbers found in the spleen (35.3%) and the liver (27.4%). The latter organs represented a dead end in the development of S. truttae as all stages in these organs degenerated and sporogony was found to take place exclusively inside the renal tubules. Early sporogonic stages were found from day 25 post-infection but mature spores only developed after at least 15 days of proliferation within the tubules.  相似文献

ABSTRACT:   A new myxosporean parasite was found in the body cavity and caudal peduncle of the freshwater goby Rhinogobius sp. Orange type (OR) collected from the Nagara River, Gifu Prefecture, Japan. Infected fish exhibited substantial swelling of the abdomen caused by large parasitic cysts approximately 10 mm in size, formed in the visceral cavity. The cyst was a compacted aggregate of several smaller cysts, similar to a bunch of grapes in appearance. Histological examination showed that plasmodia developed within the renal capsule, and finally occupied the visceral cavity. Spores were ovoid with an attenuated anterior end. Sutural ridges were conspicuous with several folds on the edge. Average spore size was 11.9 (10.5–13.5) μm long, 9.0 (8.0–10.0) μm wide, and 6.5 (6.0–7.0) μm thick. Two equal polar capsules were 5.5 (4.5–6 0) μm long and 3.0 (2.5–4.0) μm wide. Partial small subunit ribosomal DNA sequences of the myxosporean were distinct from those of other myxozoan species in GENBANK. A new species name, Myxobolus nagaraensis , is proposed for this parasite.  相似文献

Abstract Freshwater fish in Czechoslovakia were examined for species of the genus Sphaerospora Thélohan, 1892 and for myxosporean life cycle stages in the blood. In addition to perch infected with S. pectinacea Bocharova & Donets, 1974, renal tubules of seven host species harboured thus far unidentified Sphaerospora species. A new species, S. gobionis sp.nov. is described from renal tubules of Gobio gobio. In populations of Gobio gobio, Tinea tinea and Rutilus rutilus harbouring infections with different Sphaerospora species, organisms identified as mobile myxosporean life cycle stages were detected in the blood, where they undergo a proliferative cycle. These organisms were reminiscent of stages in the blood of common carp fingerlings, supposedly identical with Sphaerospora renicola Dyková & Lorn. It is possible that the blood stages found in the three cyprinid hosts represent stages of the life cycle of their respective Sphaerospora species. If this is correct, further studies may show if the presence of proliferative stages in the bloodstream is a character distinctive of the genus Sphaerospora.  相似文献

A Sphaerospora sp. (Myxosporea) infection (presumably S. truttae ) was identified on a trout farm in northeastern Italy. Parasites were detected in kidneys from infected brown trout, Salmo trutta L., over a 2-year period. Extrasporogonic, sporogonic stages and mature spores were simultaneously detected in the same fish. Traditional diagnostic methods for Sphaerospora spp. rely on the detection of the myxosporean developmental stages in Giemsa-stained kidney smears or haematoxylin-eosin stained tissue sections. A histochemical method was employed where 10 biotinylated lectins (Con-A, DBA, SBA, GS-I, PHA-P, LEA, PWM, RCA1, WGA and UEA-I) and the avidin-biotin-peroxidase complex (ABC) were used on Sphaerospora -infected brown trout renal tissues and kidney imprints. Five monoclonal antibodies against PKX (Mab12, MabA3, MabC5, MabD4 and MabB4) were also tested. A lectin glycoconjugate binding pattern for Sphaerospora spp. is presented. This staining method shows that SBA lectin ( Glycine max agglutinin) is a useful tool for the detection of the Sphaerospora spp. Only MabB4 bound some of the most mature sporogonic stages. In contrast Mabs12, A3, C5 and D4, and GS-I lectin ( Griffonia simplicifolia agglutinin) did not stain any of the Sphaerospora spp. stages, but did bind very specifically to the sporogonic and extrasporogonic stages of PKX, the causative agent of proliferative kidney disease (PKD).  相似文献

Cobia culture has been rapidly gaining in popularity since the early 1990s; however, the relative success of modified commercial diets in aquaculture has delayed the need for specific research into the nutritional requirements of cobia. Recent work has determined optimum dietary protein and lipid levels in juvenile cobia at 45 and 5–15% dry weight respectively. Maximum growth and feed conversion ratios have been recorded at 27–29 °C in juvenile cobia with an optimum ration level determined at 9% initial body weight per day. There is limited information on amino acid and essential fatty acids (EFA) requirements in cobia. Several studies have explored alternate protein sources in juvenile cobia with relative success observed with meat meal, yeast-based protein and various plant based sources including soybean meal. There is no literature on the vitamin or mineral requirements of cobia or the nutritional requirements of larger fish. Therefore future research should focus on the amino acid, EFA, vitamin and mineral requirements of cobia while the protein, lipid and energy requirements of larger cobia should be addressed. Additional work on feed ingredients, choice and palatability would also aid in maximizing culture production while minimizing costs thereby producing a more sustainable product.  相似文献

The development of Myxobolus bramae Reuss 1906, a myxosporean parasite of the gills of common bream Abramis brama L., was studied in experimentally infected oligochaetes. In five experiments, uninfected Tubifex tubifex (Müller) and Limnodrilus hoffmeisteri Claparéde were exposed to mature myxospores of M. bramae . In four experiments triactinomyxon type actinospores developed in Tubifex specimens but no infection was found in Limnodrilus . Actinospores were released from oligochaetes 70–81 days after initial exposure. At that time pansporocysts containing eight actinospores were located in the gut epithelium of experimental oligochaetes, but free actinosporean stages were also found in their gut lumen. Each actinospore had three pyriform polar capsules and a barrel-shaped sporoplasm with 32 secondary cells. The spore body joined the three caudal projections with a stout style. The total length of the actinospore was 139 μm on the average.  相似文献

Abstract. Laboratory examinations of pond-reared common carp, Cyprinus carpio L., revealed a close correlation between the prevalence of swimbladder inflammation (SBI), renal sphaerosporosis and infection by C-blood-protozoan among the carp fry. In impression smears as well as light and electron microscopic preparations we detected developmental stages of intercellular protozoa, mainly in the loose fibrous tissue of the swimbladder. The parasites multiplied by internal budding so that 20–46 secondary cells were formed in each primary cell and two tertiary cells were formed in each secondary cell. The final stage of development was a unit consisting of a secondary cell enclosing two tertiary cells, i.e. a so-called triple formation, which bore a close resemblance to the early sporogonic stages of the renal sphaerosporan Sphaerospora angulata Fujita, 1912. Certain morphological similarities and the frequent simultaneous presence of the swimbladder protozoan, C-bloodprotozoan, and S. angulata in hosts with clinical SBI have led us to postulate that the former two parasites could represent the hitherto unknown presporogonic stages of S. angulata. In view of the pathological changes caused by the parasites in the hosts with clinical SBI, and negative bacteriological and virological findings we have postulated that the swimbladder protozoan is the primary cause of SBI in carp fry.  相似文献

The results of comparative immunohisto-chemical staining studies with two lectin probes, GS-I and SBA, and five monoclonal antibodies raised against the PKX myxosporean are presented. In order to investigate the postulated link between PKX and Sphaerospora spp., the lectins and monoclonal antibodies were tested against PKX, Sphaerospora renicola, Sphaerospora oncorhynchi, Sphaerospora elegam, Sphaerospora truttae and Sphaero-spora sp. from Atlantic salmon, Salmo salar L., and Ceratomyxa shasta. SBA appears a useful tool for tbe diagnosis of all Sphaerospora spp. infections and was also positive for C. shasta. GS-I and monoclonal antibodies (12, C5, D4 and A3) appear to be specific for both the extrasporogonic and sporogonic stages of PKX. Although MabB4 was raised to PKX, it appears to stain some of the sporogonic stages of Sphaerospora spp. but not C. shasta. The results of the lectin and monoclonal staining of PKX and Sphaerospora oncorhynchi indicated that further work was required to prove or disprove the hypothesis that PKX and S, oncorhynchi are the same organism.  相似文献

Cobia (8.4 ± 0.1 g body weight) were fed to satiation with three test diets of high plant protein‐based ingredients and different lysine to arginine ratios, and one commercial diet (currently used for cobia rearing in Vietnam as a control for growth) for 6 weeks. The test diets contained 206 g marine ingredients kg?1, including fishmeal, krill meal and fish protein concentrate (in order of high to low inclusion), while the rest of the dietary protein was a blend of soya and pea protein concentrate, wheat protein and sunflower meal. Crystalline lysine and arginine were added in the test diets to produce either a balanced lysine to arginine ratio (BL/A; 1.1) and a high or low lysine to arginine ratio (HL/A; 1.8 and LL/A; 0.8, respectively). There were no significant differences in final body weight, weight gain, feed conversion ratio or protein gain between cobia fed BL/A‐ and commercial control diet (CCT). Cobia fed BL/A diet performed better than fish fed either HL/A‐ or LL/A diet. This was partly due to a higher feed intake and protein and lipid gain in cobia fed BL/A diet as compared to HL/A‐ and LL/A diet.  相似文献

Fish are potentially submitted to water acidification when reared in recirculating aquaculture systems. This study evaluated the responses of juvenile cobia Rachycentron canadum after acute exposure to acid water. Juvenile cobia (12.6 ± 0.5 g; 14.2 ± 0.2 cm) were acutely exposed to four pH levels (7.9 (control), 6.5, 6.0, and 5.5). After 24 h of exposure to different pH values, fish were sampled for physiological and histopathological evaluation. Acid water affected physiological parameters and induced morphological histopathologies on gill and skin of juvenile cobia, and these effects were more conspicuous with decreasing pH values. Acid stress induced blood acidosis in juvenile cobia, coupled to a decrease in bicarbonate (HCO3?) and saturated O2 (sO2) in fish blood. On the other hand, haematocrit, haemoglobin and glucose concentration increased their values (< 0.01) comparing to control level. Hyperplasia with completely fusion of secondary lamella was observed in all pH treatments (6.5. 6.0 and 5.5), while telangiectasia and proliferation of chloride cells were present for fish exposed to pH 6.0 and 5.5. In skin hyperplasia and hypertrophy of mucous cells, necrosis of these cells for fish exposed to pH 6.0 and 5.5 was observed. The results of this study demonstrate that acute acid water exposition affected physiology and histopathology in juvenile cobia, especially at pH values below 6.5. Accordingly, particular attention must be given to pH during cobia reared in recirculating aquaculture.  相似文献

Members of the myxosporean genus Kudoa occur in various marine teleosts worldwide. Several species are of concern to the fishery and aquaculture industries as they may produce unsightly cysts in the fish host's musculature or are associated with postmortem myoliquefaction of the fish muscle, commonly referred to as 'soft flesh'. This study describes the occurrence and effects on a host of a Kudoa species in Atlantic mackerel, Scomber scombrus , from the northern North Sea. Generalized postmortem myoliquefaction associated with Kudoa sp. occurred in 0.8% of the examined fish, i.e. 11 of 1339 mackerel developed 'soft flesh'. There was a significant difference in the prevalence of myoliquefaction between medium sized (400–600 g) and large mackerel (>600 g). The prevalence reached 8.9% in the latter host size group. No subclinical infections of Kudoa sp. were detected when examining fresh muscle ( n  = 103) and blood ( n  = 165) samples for spores using light microscopy. Affected mackerel developed generalized myoliquefaction after 38–56 h post-catch. No inflammatory host response was associated with the presence of plasmodia within single body muscle fibres of 'soft flesh' affected fish. Based on comparison of myxospore dimensions and analysis of the nuclear small subunit (SSU) ribosomal DNA, the present Kudoa species is assigned to Kudoa thyrsites . However, due to the species' apparently very wide geographical distribution and host range, its varying effect on different fish host species, together with the still unknown life cycle of Kudoa spp., the taxonomic status of K. thyrsites appears not to be fully resolved.  相似文献

Cobia Rachycentron canadum is a fast-growing, pelagic marine species that has recently attracted aquaculturists in both the research and commercial sectors. The typical method of grow-out for this species is in outdoor systems where production is limited to locations and seasons conducive for adequate growth and survival. Expanding the culture of cobia to indoor recirculating aquaculture systems (RAS) would allow for the production of fingerlings throughout the year and extend production to cooler regions. Two rearing trials were conducted to examine the growth and survival of cobia from hatching through 4 (trial 1, T1) or 35 (trial 2, T2) g in RAS. Cobia larvae were reared in circular tanks placed in a raceway to control water temperature and quality. During early juvenile grow-out, fish were transferred without grading to a second raceway on 29 dph (T1) or over a period of grading from 29–43 dph (T2). Larval growth (1–22 dph) measured as standard length was similar for both trials ranging from  3.9 to 14.7 mm. However, larval growth measured as wet weight (0.033 g, T1; 0.026 g, T2) or dry weight (5.7 mg, T1; 3.9 mg, T2) was significantly greater on 22 dph during T1 as was the ratio between myotome height and standard length. These differences may have resulted from an increase in initial densities from 8.7 larvae l− 1 (T1) to 14.7 larvae l− 1 (T2) which apparently caused an increase in food competition and overall aggression. During juvenile grow-out, cobia reached 4.0 g on 43 dph in T1 and 35.4 g on 71 dph in T2 matching weights achieved during grow-out in outdoor ponds. Over the course of both trials, survival was similar to that reported in outdoor ponds. Mean survival (± S.D.) during the early rearing phase (hatching through 29 or 43 dph) averaged 13.2 ± 3.2 % and 10.4 ± 3.2 % corresponding to final densities of 0.9 ± 0.2 and 1.2 ± 0.4 fish/l for T1 and T2, respectively. During the first grow-out phase (29–43 dph), survival of fish moved into the open raceway was 64.5% in T1 and 88.7 % in T2. Survival of cobia during the second grow-out phase (43–71 dph) for T2 was 92.5%. The results of this study indicate that cobia can be successfully cultured in indoor systems from hatching through at least 35 g without negatively affecting growth or survival.  相似文献

Lipid Nutrition and Feeding of Cobia Rachycentron canadum Larvae   总被引:3,自引:0,他引:3  
This study examined the fatty acid composition of cobia Rachycentron canadum eggs and yolksac larvae, as well as the ovaries of wild caught females as an initial guide to lipid nutritional requirements. A 2-wk feeding study also was conducted to investigate the effectiveness of four dietary treatments on the growth and survival of cobia larvae. Cobia eggs in the tailbud stage contained 31.4 ± 1.3 μg lipid/egg. After hatching, the amount of lipid decreased significantly (P < 0.05) from 28.3 ± 0.3 to 23.2 ± 0.1 μg lipid/larvae during the yolksac larval stage (days 1 to 3 after hatching). Ovaries from wild caught adults and captive spawned eggs and yolksac larvae contained high levels of PUFAs with docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA), and arachidonic acid (ARA) accounting for approximately 80% of the total suggesting that cobia larvae may have a high dietary requirement for these fatty acids. For the feeding study, larvae were fed: 1) Artemia only; 2) enriched rotifers for 1 d only + microparticulate diet (day 313); 3) enriched rotifers for 3 d (day 3–5) + Artemia (day 3–13); and 4) enriched rotifers for 6 d (day 3–8) + Artemia (day 3–13). Cobia larvae began feeding on rotifers 3 d after hatching and on newly hatched Artemia nauplii by the fifth day following the onset of exogenous feeding (day 7). On day 7, no differences in larval growth were found among larvae fed rotifers for 3 versus 6 d, whereas larvae fed only Artemia or rotifers for I d followed by microparticulate diet were significantly smaller (P < 0.05) and did not survive beyond day 9 and 13, respectively. The results of the feeding study indicate that cobia larvae require rotifers for a minimum of 4 d following the onset of exogenous feeding.  相似文献

Abstract. Serologic relatedness of the two life stages of the salmonid whirling disease parasite Myxosoma cerebralis Hofer, 1903 — myxosporean spores from fish cartilage and actinosporean triactinomyxon spores from aquatic tubificids — were investigated. When the direct fluorescent antibody technique was used, anti-triactinomyxon and anti- M. cerebralis rabbit sera conjugated with fluorescein isothiocyanate cross-reacted with the respective heterologous life stage. Both stages showed similar locations of specific fluorescence with conjugates of either homologous or heterologous serum. Thus, serology supports the relatedness of the myxosporean M. cerebralis and the actinosporean triactinomyxon stages.  相似文献

Abstract  This study examined stocking with three early life stages of brown trout, Salmo trutta L., in the context of rehabilitating a native lineage. Experimental sections in five streams were stocked successively with three stages (I: unfed fry at the end of the reabsorption phase, II: fed fry measuring 2–3 cm and III: fed fry measuring 4–5 cm) derived from a hatchery stock bred from wild spawners. The three stages were distinguished by single or multiple fluoromarking of the otoliths with alizarin redS. The index of relative stocking efficiency was greater for stage II than for stage I in all sections and equivalent or greater than that for stage III. Stage II achieved significantly larger mean length and weight in autumn than stage III stockings.  相似文献

Myxobolus wulii (= Myxosoma magna ) was first described from the gills of goldfish, Carassius auratus auratus, in China. Subsequently, a myxosporean infecting the hepatopancreas of allogynogenetic gibel carp, C. auratus gibelio , was designated as a different species, Myxobolus guanqiaoensis , although the morphological features were almost identical to those of M. wulii . In Japan, an unidentified Myxobolus sp. was found in the gills and hepatopancreas of goldfish. Morphological and molecular analyses in the present study identified these myxosporeans as M. wulii , which was thus shown to use different habitats in the host fish. Phylogenetic analyses of small subunit ribosomal RNA gene sequences showed that M. wulii is closely related to two gill-infecting Myxobolus species, M. ampullicapsulatus and M. longisporus . Fish infected with M. wulii in the hepatopancreas exhibit swollen abdomens and chronic mortality. Hepatopancreas tissues are virtually destroyed and replaced with plasmodia of M. wulii . A remarkable difference in susceptibility to M. wulii between two clones of allogynogenetic gibel carp was observed, suggesting that resistance to the myxosporean infection was established in a clone of fish bred by allogynogenesis.  相似文献

ABSTRACT:   The present study aimed to investigate functional changes of the kidney in mature masu salmon. The urine flow rate (UFR), kidney-somatic index (KSI), and epithelial cell height of second proximal convoluted segment of renal tubules were examined in mature and immature masu salmon Oncorhynchus masou . Two methods of calculating UFR were made as either the units per body weight, or body weight without gonads. The former calculation using body weight may be used for comparison between data obtained from immature fish, and the latter method was suitable when comparing mature sexes and assessing the transition from immature to finally mature stages. In both calculations of UFR, KSI, and epithelial cell height of the second proximal segment, the values for mature fish were significantly higher than for immature fish. These results first suggest that functional transition of the kidney occurs in maturing fish.  相似文献

ABSTRACT:   The development of embryos, larvae and juveniles of the Ryukyu-ayu, Plecoglossus altivelisryukyuensis are described based on laboratory-reared specimens.The eggs were spherical, 0.98–1.18 mm (mean 1.06 mm)in diameter with an adherent membrane. The incubation period after fertilizationwas approximately 155 h at a water temperature of 19.7–22.0°C(mean 20.7°C). Newly hatched larvae were 5.0–5.9 mm(mean 5.5 mm) in body length (BL) with 59–62 myotomes.Within 5 days after hatching, the larvae had attained 6.4–8.2 mm(mean 7.8 mm) BL and had completely consumed their yolk.Notochord flexion began at 13.7 mm BL and was completedby 16.3 mm BL. The rudimental dorsal, anal, pelvic andadipose fins appeared at 14.7, 16.3, 21.8 and 21.8 mm BL, respectively.All fin rays reached the same fixed number of adult fish at about28 mm BL. The comb-like teeth began to form at approximately30 mm BL and were fully developed at about 40–50 mm BL.The proportions of P. a. ryukyuensis specimens,which were consistent with adult fish, occurred at approximately40 mm BL.  相似文献

Abstract –  Information on larval and juvenile habitat use is virtually absent for fishes. Our objective was to document habitat use of larval and juvenile Cape Fear shiners Notropis mekistocholas , an endangered species, in both natural and lab settings so that a better understanding of habitat requirements in all life stages can be achieved. We measured habitat parameters of areas used by all life stages of N. mekistocholas in the summers of 2007–2008 in the Rocky River, North Carolina. Field data suggests larvae use more shallow depths (mean = 31.6 cm) and reduced water velocities (mean = 0.02 m·s−1) than adults (mean = 56.3 cm, 0.10 m·s−1), and juveniles 15–25 mm often school with adults. Results of lab studies support field observations. In the mesolarval and metalarval stages N. mekistocholas were selective in their use of water velocity ( P  = 0.013) and depth ( P  = 0.001). In multiple juvenile stages, N. mekistocholas were selective for water velocity ( P  = 0.015), and depth ( P  < 0.001), choosing deeper depths and higher water velocities than larvae. These results demonstrate ontogenetic shifts in habitat use of N. mekistocholas and suggest that water velocity and depth are critical to successful recruitment for this species.  相似文献
