The various ducts of the epididymides of four gallinaceous birds, the turkey (Meleagris gallopavo), domestic fowl (Gallus gallus), guinea-fowl (Numida meleagris) and Japanese quail (Coturnix coturnix japonica) were studied at the scanning and transmission electron microscopy levels. The tissues were fixed either by immersion or vascular perfusion, for comparative purposes. Each duct system, save for a few details, presented similar morphological features in all species. The epithelial surface of the rete testis was regular and each cell bore a single cilium, as well as numerous, or in some parts, very few, short, regular microvilli. Each of the Types I and II non-ciliated cells of the proximal and efferent ducts displayed abundant, moderately long and regular microvilli, and a solitary cilium. The ciliated cells exhibited tufts of cilia. The Type III non-ciliated cell of the connecting and epididymal ducts exhibited a solitary cilium, and numerous microvilli which were intermediate in length between those of the rete testis and those of the efferent ducts. Vascular perfusion of the avian epididymal tissue was the superior method of fixation because it minimised the developments of fixation artefacts. Apocrine secretion did not appear to occur in the epididymis of these birds as the apical blebs of Types I, II and III cells, which have previously been reported, only manifest in this study in inadequately fixed tissues, and were therefore viewed as being artefacts. The present findings suggest that the current terminology, as applied to the avian epididymis, be retained.  相似文献

Surface ultrastructural observation of the thyroid gland in donkey revealed the coexistence of certain unusual follicles or cysts, the ultimobranchial (UB) follicles, which exhibited large diameters and less regular outlines. In addition, several small follicles, cell nests or solid cellular masses were also demonstrated in association with these structures. Surface of most UB follicular cells was almost hexagonal in shape, studded with sparse or dense pleomorphic microvilli, which were much denser at the cellular borders. Each follicular cell bore single cilium that projecting over the cell surface. Interestingly, bleb-like apocrine protrusion of the apical cytoplasm of some follicular cells was frequently demonstrated. These apocrine blebs (or aposomes) were varied in shape, size and showed smooth or irregular surface. In addition to such aposomes, deep to shallow circumscribed or eroded areas of the apical cell surface was frequently demonstrated. These findings indicate that the UB follicular cells in donkeys, similar to those of camels, exhibit an apocrine activity alongside with the usual merocrine mode of secretion.  相似文献

The volumetric proportion of the various ducts of the epididymis of the emu and ostrich and the immunohistochemistry of actin microfilaments, as well as cytokeratin, desmin and vimentin intermediate filaments, were studied in the various ducts of the epididymis of the emu and ostrich. The volumetric proportions of various ducts, which are remarkably different from those of members of the Galloanserae monophyly, are as follows: the rete testis, 5.2 ± 1.4% for the emu and 2.4 ± 1.8% for the ostrich; efferent ducts, 14.2 ± 2.3% (emu) and 11.8 ± 1.8% (ostrich); epididymal duct unit, 25.8 ± 5.8% (emu) and 26.1 ± 4.1% (ostrich) and connective tissue and its content, 54.7 ± 5.8% (emu) and 60.0 ± 4.9% (ostrich). Unlike in mammals and members of the Galloanserae monophyly, only vimentin was immunohistochemically demonstrated in the rete testis epithelium of the emu, and none of the cytoskeletal protein elements in the ostrich rete testis. The epithelium of the efferent ducts of the emu co-expressed actin, cytokeratin and desmin in the non-ciliated type I cells, and vimentin in the ciliated cell component. The ostrich demonstrated only cytokeratin in this epithelium. The ratite epididymal duct unit is different from that of mammals in lacking actin (only weaky expression in the ostrich), desmin and cytokeratin, and a moderate/strong immunoexpression of vimentin in the basal cells and basal parts of the NC type III cell in the epididymal duct unit. Immunoexpression of the microfilaments and intermediate filaments varied between the two ratite birds, as has been demonstrated previously in birds of the Galloanserae monophyly, and in mammals.  相似文献

Aquaporins (AQPs) are essential membrane protein channels for the transport of water across membranes. Fluid movement in the epididymis is important for modulation of the luminal environment, in which sperm mature and reside. This study was designed to understand the morphology and localization of AQPs in ram efferent ducts (ED) and epididymis. For this purpose, the epididymis of seven animals were removed for histologic and immunohistochemical analyses. AQP1 immunoreactivity was observed in the apex of the ED, and AQP9 was found adjacent to the nuclei of the epithelial cells of the ED. The epithelial lining of ram epididymis is pseudostratified columnar and presents principal, basal, apical and narrow cells. In the initial segment (IS), a moderate reaction for AQP1 was observed in the apical cytoplasm of epithelial cells. An intense reactivity for AQP1 was noted over the microvilli of principal cells and in spermatozoa in the caput. In the corpus and cauda, AQP1 was noted only over the endothelial cells of vascular channels located in intertubular spaces. A weak‐to‐moderate reaction for AQP9 was observed in the nuclei of epithelial cells in the IS, caput and corpus of the epididymis. In the cauda, an intense reaction to AQP9 was observed in the epithelial border. In the IS, caput and corpus, the reactivity for AQP9 differed from those observed in domestic animals. The cauda showed a pattern similar to that previously described. These results indicate that AQPs 1 and 9 have reversed locations and roles in rams, suggesting activity variations related with fluid and solute absorption throughout the epididymis.  相似文献

A cell type with properties similar to those of the brush cell was recognised in the bronchial epithelium of calves. It occurred in small numbers and was not found in airways distal to the small bronchi. The ultrastructural features are described, emphasising the two constant characteristics (i) a dense population of thick apical microvilli and (ii) cytoplasmic filament bundles which extent into the microvilli.  相似文献

This remnant appeared in the form of irregular and elongated follicles of various size and shape with folded lumen. These structures occupied a peripheral location beneath the capsule, as well as being embedded mostly in the vascular connective tissue of the thyroid. Calcitonin cells were predominantly localized in the UB follicles. Scanning electron microscopy revealed that surface of most UB follicular cells was almost flat and polyhedral or hexagonal in shape. Other follicles possessed cells with dome shaped apical surface demarcated by a shallow intercellular depression. The luminal surface of the follicular cell lining was studded with microvilli that appeared with various density and length and were numerous at the cellular borders. Interestingly, most of UB follicular cells were provided with single cilium, projecting over the cell surface. Notably some UB follicles presented various stages of apocrine activities.  相似文献

Triturus torosus has a resting fundic stomach with gastric pits and glands and cell types similar to previously described amphibian species. Lining the surface and the gastric pits are columnar mucus-secreting cells. They differ in morphology and staining reactions from the mucous-neck cells, the second type of mucus-secreting cells. The latter are confined to a limited region at the necks of gastric glands where they empty into the pits and appear to “isolate” the main oxyntic, acid-secreting cells from the surface mucous cells. The gastric glands are lined by a single cell type, the oxyntic cell, which shows features of both the mammalian chief (zymogenic) and parietal (oxyntic) cells and which presumably secretes both enzymes and acid. This cell has a prominent granular endoplasmic reticulum with prezymogen and zymogen granules, numerous mitochondria, and in the apical cytoplasm an extensive system of tubules and tubulovesicles. The membrane bounding these tubules appears identical to the surface plasmalemma, and occasional continuity between the two can be demonstrated. The oxyntic cells have also a few apical microvilli and intercellular but not intracellular canaliculi. Occasional argentaffin or enterochromaffin cells with few granules are scattered among the oxyntic cells. Like other amphibia, the western newt feeds intermittently and thus is a good experimental animal for the study of gastric secretion.  相似文献

The macroscopic and microscopic anatomy of the epididymis of the sexually mature Peromyscus winkelmanni (carleton) was examined using light and scanning electron microscopy. The epididymis was divided into three regions: caput, corpus and cauda. The epididymal duct was lined with columnar and cubic epithelium with stereocilia and covered by a muscular connective tissue sheath. Capillaries appear to penetrate directly into the epithelium from the underlying connective tissue in the initial segment. The epididymal epithelium presents four cell types: principal, basal, apical and clear cells. Based on morphological differences (height of epithelial cells, length of the stereocilia, luminal area, larger diameter and spermatic index), the epididymis of P. winkelmanni, presents seven zones. The stereocilia of the epididymal ducts of zones I, II, IV and V are thick and tall, while in zone III they are thin and short. The stereocilia in zone VI are thin, while in zone VII they are short but thick. The secretory products observed in the lumen of the epididymal ducts have vesicular, granular and fibrous form in the seven zones. This study contributes to an understanding of the morphofunctional features of the epididymis in sperm maturation in a species that shows seasonal reproductive activity.  相似文献

Summary Observations were made, using scanning electron microscopy, of the surface features of the magnum in the immature ostrich during periods of ovarian inactivity, activity and regression. In birds with inactive ovaries the luminal surface of the magnum was lined with non-ciliated cells, which were densely covered by microvilli. In contrast, the magnum in birds with active ovaries was composed of ciliated and non-ciliated cells. The distribution of ciliated cells was not uniform, with clumps of cilia occurring next to non-ciliated areas. Samples collected from birds with regressing ovaries, during periods of decreasing daylength, revealed that the magnum was undergoing involution. The deciliation of ciliated cells and the presence of short microvilli on non-ciliated cells characterized magnal regression. These results suggest that ovarian activity and changes in daylength have a profound effect on the surface features of the magnum in the immature ostrich.  相似文献

A histological and immunohistochemical study of the epididymis of 21 cats which ranged from 7 to 8 months to 2 years of age was made. Five different cell types were observed in the feline epididymis: principal and basal cells, which were the most numerous, apical and narrow, PAS positive cells, which were scarce, and migratory cells, consisting of lymphocytes and macrophages. Four morphologically distinct areas of the cat epididymis were identified. Region I displayed a stellate lumen and the principal cells were tall and presented long and irregular stereocilia. In region II, the lumen was oval and the principal cells were shorter than in the inicial area. Region III displayed characteristics similar to those of region I; the principal difference was the presence of short and regular stereocilia on the surface of the principal cells. In region IV the lumen was very spacious and the epithelium shorter than in the other regions. In regions I, II, and III intraepithelial cavities were observed. With regard to the immunohistochemical results, the basal cells displayed medium immunoreactivity with vimentin. Due to its anti-desmin reactivity, the muscle wall which surrounds the epididymis was seen to become progresively thicker as it nears the tail. Broad-spectrum anti-cytokeratin serum produced intense immunostaining in the basal, apical and narrow, PAS positive cells; the principal cells of region IV also displayed a strong immunoreactivity.  相似文献

1. The structure of the chicken caecal epithelium has been investigated at macroscopic and microscopic levels (scanning and transmission electron microscopy). 2. Morphological data distinguish three regions in the caecum: proximal, with well developed villi, but smaller and less numerous than those present at mid-jejunum; medial, with longitudinal folds and small villi, and distal, with longitudinal and transverse folds and small villi similar to those of the medial region. 3. Microvilli of enterocytes from the tip of the villus of the proximal caecum are shorter and thinner than jejunal microvilli; microvilli of medial and distal regions are poorly developed. 4. There is a decrease in microvillus surface area along the villus axis as a result of the progressive decrease of microvillus length from the tip of the villus to the crypt, both in the proximal caecum and jejunum. In tip of the villus, the amplification of apical surface due to microvilli is 24, 12.7 and 14 times in proximal, medial and distal caecum, respectively, while in the jejunum the amplification factor is 33.1. 5. The higher development of proximal caecal apical surface supports the view that this region is better adapted for the absorption of nutrients that the other caecal regions.  相似文献

The use of the scanning electron microscope gave a three dimensional representation of the epithelial surface. Additionally, light microscopy revealed the representative structure of the epithelium. The rete testis showed a single layer of cubic epithelial cells. Short and dense microvilli were found on the surface. Sporadically a single, cilia-like structure was recognized. An extratesticular rete testis was identified. The flowing transition of the epithelium between the rete testis and the efferent ductuli occurred at different levels, so that both kinds of epithelial structures were recognized in the same area. The efferent ductuli were composed of a single columnar epithelium consisting of two cell types, principal cells and ciliated cells. The ciliated cells were recognized by their cilia protruding into the lumen. The principal cells showed microvilli on their surface and bleblike apical protrusions which erupt into the lumen.  相似文献

An ultrastructural study on the lymphocytes from peripheral blood samples from 20 healthy adult Arabian oryx (Oryx leucocoryx) was undertaken. Small lymphocytes ranged in size from 2–3.5 µm and exhibited the typical repertoire of organelles found in small lymphocytes of other animals but showed no evidence of azurophilic granules. Medium-sized lymphocytes were 5–6.5 µm in diameter and occasionally demonstrated azurophilic granules. Microvilli were a common finding of lymphocytes. Of particular interest was the presence of multivesicular bodies, which have previously only been described in human lymphocytes. Both small- and medium-sized lymphocytes of the Arabian oryx were smaller than those reported for other animals. Small lymphocytes exhibited short, thick microvilli, whereas medium-sized lymphocytes had long thin microvilli, a single nucleolus and occasionally azurophilic granules and multivesicular bodies.  相似文献

Among the multiple functions, which have been identified for the neonatal Fc receptor (FcRn), we study its role in the IgG transport in the mammary gland during the colostrum formation. For this reason, we have obtained several mammary gland biopsies from a pregnant sheep around parturition. The presence of the FcRn heavy chain mRNA was detected exclusively in the acinar and ductal epithelial cell by in situ hybridization (ISH). We detected strong signal in samples harvested 24 and 10 days prepartum; however, in samples we collected postpartum was barely detectable. Immunohistochemistry confirmed our ISH data. The cytoplasm of the epithelial cells of the acini and ducts in the mammary gland biopsies stained homogeneously before parturition, although a remarkable difference was observed in the pattern after lambing. The signal indicated uneven distribution of the FcRn alpha chain in the epithelial cells 1 and 5 days postpartum, since the apical sides of the epithelial cells were highlighted. The presence of the FcRn in the acinar and ductal epithelial cells and the obvious change of its distribution before and after parturition suggest that FcRn plays an important role in the IgG transport during colostrum formation. FcRn expression was also found in the lamb duodenal crypt epithelial cells, which have been previously demonstrated to secrete IgG1 in newborn ruminants, suggesting secretory role of the FcRn in ruminant epithelial cells.  相似文献

During the sexual cycle the structural changes in the cells of the glandular epithelium in the endometrium of a cow are more pronounced than those in the luminal epithelium. Microvilli appear on cell surface during pro-oestrus; in later stages the microvilli disappear and the apical parts penetrate into the lumen. The relative volume of mitochondria increases from pro-oestrus to culminate in oestrus and metoestrus and to drop in dioestrus. The values of the volume and surface area of granular endoplasmic reticulum are also at their highest level in oestrus and metoestrus. The lowest volume and surface area of smooth membranes are recorded in dioestrus, then it markedly increases to culminate in metoestrus. The incidence of lysosomes is variable and their volume culminates in metoestrus. The proliferation of the glandular epithelium has the highest intensity in oestrus and reaches its peak values in metoestrus when the apical parts of cytoplasm contain the largest amounts of smooth-walled vesicles and vacuoles, partly also secretory granules, and when the signs of secretion are observed.  相似文献

为了探明雄性雏鸵鸟生殖器官的形态学特点,采用石蜡切片和HE染色技术,对6羽45日龄雄性非洲雏鸵鸟的生殖器官进行了研究,并与家禽的相关结构进行了比较。在光镜下观察了雄雏鸟生殖器官的形态结构,结果表明:(1)鸵鸟睾丸没有睾丸纵隔和睾丸小叶,曲精小管中精原细胞排列紧密且整齐,睾丸间质较少;(2)附睾由睾丸网、输出小管和附睾管组成;(3)输精管管壁由黏膜、肌层和外膜3层组成;(4)阴茎由2个圆形的纤维性淋巴体、淋巴间隙和微血管共同构成。  相似文献

The mite Psoroptes ovis causes sheep scab disease in flocks throughout much of the world. A serious impediment to the development of novel control measures for this mite is our inability to produce in vitro colonies of the mite. Here, we describe the alimentary canal of the mite with the particular aim of determining what it feeds on, as part of a longer term goal, to develop in vitro culture techniques for P. ovis. The alimentary canal of P. ovis consists of a cuticle-lined foregut and hindgut separated by a microvilli-lined midgut. The foregut is divided into a pre-oral cavity and a muscular pharynx and oesophagus. The midgut consists of three ultrastructurally discrete areas: a stomach with bi-lobed ventriculi, a colon and a post-colon. The stomach is composed of two cell types. The most common cells (Type 1) are either cuboidal or squamous depending on the degree of gut distension and possess short microvilli, a single basally located nucleus, extensive rough endoplasmic reticulum and other components suggesting active secretion. The less common cells (Type 2), possess an extensive apical network of tubules and basally food vacuoles suggesting intracellular digestion. These cells extend into the gut lumen, become free-floating and degenerate. The colon and post-colon are composed of cells similar to Type 1 stomach cells but the post-colon epithelium possesses significantly longer microvilli. Cells from these areas have not been observed to leave the surrounding epithelium and enter the gut lumen, but it is suggested that significant absorption occurs in these two areas. Faecal pellets, often containing a significant number of bacteria, leave the digestive system through the cuticle-lined anal atrium.  相似文献

An enzymatic method of isolating primary follicles in the turkey has been described previously, but no similar work has been done in the hen. In this study, primary follicles from domestic hens (Gallus domesticus) were isolated using an enzymatic method, and the isolated follicular quantity, quality, and development in vitro were assessed. About 400 primary follicles ranging from 60 to 1125 microm in diameter were recovered with trypsin and collagenase from hen ovaries (per ovary). Of these, 76.5% were intact follicles with a complete single layer of granulose cells surrounded by the basement membrane, and their ultrastructures appeared this way in situ. Follicles (351 to 500 microm in diameter) were cultured in vitro, and 46.67% of them survived after 5 days. Ultrastructural examination showed that elongated mitochondria forming a ring were distributed to the periphery of the oocyte, the Golgi was oriented with the maturing face toward the granulosa cell layer, and the oocyte plasma membrane presented a few short microvilli lying on the oocyte surface, which confirmed that the surviving follicles were developmental. These results suggest that a simple, rapid, effective enzymatic method can be used to isolate a great number of intact primary follicles from the hen ovary.  相似文献

In this study the endometrial morphology of four dogs in oestrus and 10 dogs in late metoestrus was investigated with scanning electron microscopy. In early oestrus the cells were slightly convex with fairly long microvilli and cell boundaries that were apparent but situated on a deeper level than the cell surface. In late oestrus the cells were clearly convex with long and numerous microvilli masking the cell boundaries. At the beginning of late metoestrus the cells became less convex with many but short microvilli and prominent but lower situated cell boundaries. At the end of late metoestrus the cells were flat with very short microvilli and protruding cell borders. These morphological findings were correlated with the cyclic variations of the steroid hormone levels.  相似文献
