奶牛性控冻精人工授精影响因素研究   总被引:2,自引:0,他引:2  
用分离X和Y精子的性控精液进行人工授精是控制家畜性别之最简单可行的方法.然而,低密度性控精液输精效果还不如常规人工授精,许多技术环节都有待改进.以常规冻精和稀释常规冻精为对照,研究解冻方法、输精时间和部位、不同精液来源和输精员以及育成和经产牛等因素对性控冻精人工授精妊娠率的影响.结果显示,精液解冻水浴温度和持续时间对人工授精效果有显著影响,性控精液对解冻水浴温度更敏感;性控冻精和稀释常规冻精比常规冻精对输精时间要求更严格;3种精液输精到排卵卵泡同侧子宫角基部受胎率都显著高于输精于子宫体和同侧子宫角前端;3种精液育成牛受胎率(80%)都显著高于经产牛(50%);于输精同时注射促排卵素3号明显提高性控冻精受胎率;经严格挑选、能够从事胚胎移植操作的技术熟练输精员之间性控冻精受胎率差异不显著;在所设计的不同条件下,性控冻精与稀释同样倍数的常规冻精行为相似,说明精子分离过程没有对精子造成特殊损伤.研究结果说明,精确控制人工授精各个技术环节可以实现消除性控与非性控、低密度与高密度精演之间的差别,获得高妊娠率.  相似文献

Recommendations for oestrus synchronisation of dairy heifers using progesterone-containing intravaginal devices suggest re-insertion of used devices 16 days after first insemination for a period of 5 days to allow a second opportunity for artificial insemination. Controlled studies on the effectiveness of re-using intravaginal devices to synchronise returns to oestrus in non-pregnant dairy heifers are lacking. A clinical trial was conducted involving 750 Friesian heifers in 13 herds. After an initial synchronisation programme, the used intravaginal devices were re-inserted 14 or 16 days after first insemination into half of the heifers in each herd for a period of 5 days. After the first synchronisation programme, 47.5% of heifers remained non-pregnant. Re-insertion of used intravaginal devices for 5 days significantly increased the number of non-pregnant heifers detected in oestrus and inseminated by 48 hours after device removal compared to heifers in which devices were not re-inserted (45.2% v. 27.3%, p < 0.05, in herds where intravaginal devices were re-inserted on day 14; 48.8% v. 13.6%, p < 0.05, in herds where intravaginal devices were re-inserted on day 16). Re-insertion at 14 or 16 days after first insemination was equally effective in increasing visible returns to service. However, the number of non-pregnant heifers synchronised for a second round of artificial insemination was less than expected. Conception rate to the re-synchronised oestrus was unaffected by the treatment. It is concluded that the additional procedures of CIDR re-insertion, removal, tailpainting and insemination involved in there-synchrony programme, and the relatively low in-calf rate to the re-synchronised round of insemination, reduced the potential benefits of re-synchronisation.  相似文献

The study evaluated the reproductive performance of primiparous sows submitted to post‐cervical insemination (PCAI) compared with cervical artificial insemination (CAI). Difficulty with catheter introduction, the occurrence of bleeding or semen backflow during insemination, and volume and sperm cell backflow up to 60 min after insemination were also evaluated. Sows were homogenously distributed, according to body weight loss in lactation, lactation length, weaned piglets, weaning‐to‐oestrus interval and total born in previous farrowing, in two treatments: PCAI (n = 165) with 1.5 × 109 sperm cells in 45 ml (2.4 ± 0.04 doses per sow) and CAI (n = 165) with 3 × 109 sperm cells in 90 ml (2.5 ± 0.04 doses per sow). During PCAI, sows were inseminated in the absence of boars. Transabdominal real‐time ultrasonography was performed at oestrus onset, immediately before the first insemination and at 24 h after last insemination. There was no difference (P > 0.05) between treatments in farrowing rate (91.5% × 89.1%) and litter size (12.5 × 11.9 piglets born, respectively for PCAI and CAI sows). Successful passage of the intrauterine catheter in all the inseminations was possible in 86.8% (165/190) of sows initially allocated to PCAI treatment. Difficulty of introducing the catheter in at least one insemination did not affect the reproductive performance of PCAI sows (P > 0.05). Bleeding during insemination did not affect (P > 0.05) the farrowing rate in both treatments, but litter size was reduced in CAI and PCAI sows (P ≤ 0.06). Percentage of spermatozoa present in backflow within 1 h after insemination was greater in CAI than PCAI sows (P < 0.01). More than 85% of primiparous sows can be successfully post‐cervical inseminated with doses containing 1.5 × 109 sperm cells in the absence of the boar during insemination without impairing the reproductive performance.  相似文献

The objectives of this study were 1) to compare pregnancy rates resulting from 2 methods of insemination using low sperm numbers and 2) to compare pregnancy rates resulting from hysteroscopic insemination of 5 x 106 nonsorted and 5 x 106 spermatozoa sorted for X- and Y-chromosome-bearing populations (flow sorted). Semen was collected with an artificial vagina from 2 stallions of known acceptable fertility. Oestrus was synchronised (June to July) in 40 mares, age 3-10 years, by administering 10 ml altrenogest orally for 10 consecutive days, followed by 250 microg cloprostenol i.m. on Day 11. All mares were given 3000 iu hCG i.v. at the time of insemination to induce ovulation. Mares were assigned randomly to 1 of 3 treatment groups: mares in Treatment 1 (n = 10) were inseminated with 5 x 10(6) spermatozoa deposited deep into the uterine horn with the aid of ultrasonography. Mares in Treatment 2 (n = 10) were inseminated with 5 x 10(6) spermatozoa deposited onto the uterotubal junction papilla via hysteroscopic insemination. Mares in Treatment 3 (n = 20) were inseminated using the hysteroscopic technique with 5 x 10(6) flow sorted spermatozoa. Spermatozoa were stained with Hoechst 33342 and sorted into X- and Y-chromosome-bearing populations based on DNA content using an SX MoFlo sperm sorter. Pregnancy was determined ultrasonographically at 16 days postovulation. Hysteroscopic insemination resulted in more pregnancies (5/10 = 50%) than did the ultrasound-guided technique (0/10 = 0%; P<0.05) when nonsorted sperm were inseminated. Pregnancy rates were not significantly lower (P>0.05) when hysteroscopic insemination was used for sorted (5/20 = 25%) and nonsorted spermatozoa (5/10 = 50%). Therefore, hysteroscopic insemination of low numbers of flow sorted stallion spermatozoa resulted in reasonable pregnancy rates.  相似文献

The effects of artificial insemination (AI) using sexed sperm on pregnancy rates have seldom been studied in lactating dairy cows on commercial dairy farms. We evaluated pregnancy results after AI of 306 lactating dairy cows, of which 157 were inseminated with 2x10(6) frozen/thawed sexed sperm and 149 with 15x10(6) frozen/thawed unsexed sperm. The average pregnancy and calving rates were 21.0% and 20% for the sexed-sperm AIs and 46% and 45% for the unseparated control-sperm AIs respectively (p<0.001). The proportion of female calves derived from sexed-sperm AI was 82% compared with 49% for control AI (p<0.01). The proportion of live and healthy calves in single births was 100% for sexed-sperm AI and 97% for control AI (p>0.05). Our results indicate that AI with low-dose sexed sperm under field conditions in commercial dairy herds without oestrus synchronization results in significantly reduced pregnancy rates compared with normal-dose AI. Improved insemination strategies combined with increased sperm doses are needed before the use of sexed sperm can be of any significant benefit for the dairy and beef industry.  相似文献

Little information is available on the quality of stallion spermatozoa after sex sorting. The objectives of the present study were to assess the quality of sex‐sorted stallion spermatozoa and determine its fertilizing ability after hysteroscopic low dose insemination. Ejaculates from four stallions were collected and sorted by a MoFlo SX® flow cytometer/sperm sorter. Before and after sorting, spermatozoa were evaluated for motility by Computer Assisted Sperm Analysis, viability (SYBR 14‐propidium iodide), mitochondrial function (JC‐1) and acrosomal status (fluorescein isothiocyanate Pisum sativum agglutinin conjugated). A fertility trial was carried out on four mares (seven oestrous cycles) by hysteroscopic insemination, depositing 5 × 106 X‐bearing spermatozoa. Sex sorting resulted in a significant decrease (p < 0.001) in all motility characteristics. Sperm viability and percentage of spermatozoa with functional mitochondria were not affected by the sorting process, while the percentage of reacted spermatozoa was higher (p < 0.01) for non‐sorted than sorted spermatozoa. Pregnancy rate was 28.6% (2/7) after low dose hysteroscopic insemination. Only one pregnancy was carried to term with the birth of a healthy filly. In conclusion, despite the reduction in sperm motility, sex sorting did not impair stallion sperm viability and mitochondrial activity immediately post‐thaw; moreover, the sexed spermatozoa retained the ability to fertilize in vivo.  相似文献

A pilot experiment with heifer heat synchronization with chlorsuperlutin-soked intravaginal tampons was conducted with 254 heifers in six herds in the North Moravian region. The rubber-foam tampons, cylindrical in shape (diameter 70 mm, height 70-90 mm), were introduced into the cranial part of the vagina, using an applicator and a vaginal speculum. Sixteen days after introduction, the tampons were removed by pulling the silon thread with which the tampons were cross-stitched. The heifers were inseminated on the second to fifth day after the removal of the tampons. The average performance in the application of the tampons was 38.5 tampons per hour, and in their removal 20.7 tampons per hour. The retention rate of the tampons was 97.6%; out of this, 79.8% of the tampons were pulled out by the silon thread. The signs of heat were good, 87.9% of the heifers could be inseminated intrauterinally. All the heifers with retained tampons were inseminated. The conception rate after the first insemination was 32.7%, after the second insemination 56.4%, and after two inseminations 68.1%. Out of the synchronized heifers, 8.1% were culled for slaughter. The drawbacks of this treatment include the low conception rate of the synchronized heifers, a high requirement for repeated insemination in the synchronized oestrus, and, consequently, a high sperm consumption.  相似文献

SUMMARY Effects of body weight, condition score, ovarian cyclic status and insemination regimen on pregnancy rates were investigated in 164 Bos indicus heifers synchronised with norgestomet-oestradiol and pregnant mare serum gonadotrophin (PMSG). Oestrus detection techniques were also compared. Heifers were inseminated at either a fixed time (group 1, n = 83) of 48.0 ± 0.2 h (mean ± SEM) after implant removal or at 8.9 ± 0.5 h after oestrus was detected (group 2, n = 81). Group 2 heifers that were not detected in oestrus by 72 h after implant removal were inseminated at that time. Oestrus was detected for the purpose of insemination using heatmount detectors. Tail-paint and oestrogen treated, chin-ball harnessed steers were used to compare the efficiency of oestrus detection. The probability of ovarian cyclicity increased with increasing body weight and condition score (P < 0.001). A higher proportion of heifers that were acyclic at the commencement of treatment, compared with cyclic heifers, were detected in oestrus at the time of insemination in the fixed-time inseminated group (P <0.01). Analysis of covariance revealed that intervals from implant removal to oestrus were influenced by ovarian cyclic status (P < 0.01) and insemination group (P < 0.05). A higher pregnancy rate (%± SEM) was obtained in acyclic compared with cyclic heifers in the group 1 heifers (50.0 ± 10 vs 28.1 ± 6; P = 0.055) but not among the group 2 heifers (45.8 ± 10 vs 49.1 ± 7; P = 0.787). The probability of pregnancy was found to be associated negatively with body weight (P = 0.01) while a higher pregnancy rate was obtained in the group 2 compared with group 1 heifers (48.2%vs 34.9%; P = 0.093). The efficiency of oestrus detection was highest using heatmount detectors compared with tail-paint and chin-ball harnessed steers (90.7%vs 37.0% and 23.5%, respectively; P < 0.0001). We conclude that pregnancy rates can be increased in extensive environments when insemination follows oestrus detection using heatmount detectors compared with a fixed-time insemination. The fertility of heifers inseminated at a fixed time is influenced by ovarian cyclic status due to its influence on oestrus-to-insemination intervals.  相似文献

The primary objective of this study was to determine whether a single measurement of intravaginal electrical resistance (VER), using the commercially available Ovatec® probe, can discriminate between dioestrus and oestrus in Bos indicus females, which had been treated to synchronize oestrus. Santa Gertrudis heifers (n = 226) received one of three oestrous synchronization treatments: double PGF 10 days apart, 8‐day controlled internal drug release (CIDR) treatment or CIDR pre‐synchronization + PGF 10 days after CIDR removal. The heifers were inseminated within 12 h following observed oestrus, or, if not observed, at a fixed time approximately 80 h, following the last synchronization treatment. They were palpated per rectum for signs of pregnancy 9 weeks after artificial insemination (AI). Vaginal electrical resistance measurements were taken at the completion of synchronization treatments (presumed dioestrus), immediately prior to AI (oestrus), and then at 3 and 9 weeks post‐AI. Mean VER differed between presumed dioestrus and oestrus (113.7 vs 87.4, p < 0.001). The area under the receiver operating characteristics (ROC) curve was 0.925, indicating that VER was highly discriminatory between dioestrus and oestrus. Vaginal electrical resistance at time of AI was negatively associated with odds of conception when all inseminations were included in the analyses [odds ratio (OR) = 0.97; 95% CI 0.95–1.00; p = 0.018], but not when fixed time AIs were excluded (OR = 1.00; 95% CI 0.97–1.03; p = 0.982). Mean VER readings differed between pregnant and non‐pregnant animals at both 3 weeks (120.5 vs 96.7, p < 0.001) and 9 weeks (124.0 vs 100.3, p < 0.001) post‐AI. However, 3‐ and 9‐week VER measurements were not highly discriminatory between pregnancy and non‐pregnancy (area under ROC curve = 0.791 and 0.736, respectively). Mean VER at time of AI for animals diagnosed in oestrus differed between each of the oestrous synchronization treatments (84.7, 73.6 and 78.9, groups 1–3 respectively, p < 0.001). These findings suggest that measurement of VER may improve accuracy of oestrus diagnoses when selecting cattle for AI following oestrous synchronization programmes involving tropically adapted cattle.  相似文献

The objective was to compare embryo yield and quality in lactating dairy cows superovulated (SO) with varying amounts of gonadotropins and FSH:LH ratios and inseminated with SexedULTRA? sex‐sorted semen. The SO treatments (n = 77) involved 3 protocols: groups F700 and F1000 were given total doses of 700 and 1,000 IU of Folltropin (FSH:LH ratio 49:1), respectively, whereas group F700P300 was given 700 IU of Folltropin + 300 IU of Pluset (FSH:LH ratio 1:1). Cows were artificially inseminated 3 times over a 10‐hr interval with frozen‐thawed SexedULTRA? sex‐sorted semen (total of 10 × 106 sex‐sorted sperm), starting 18 hr after onset of oestrus, with embryos/ova recovered 7 d after oestrus. Total number of recovered structures and transferable embryos were lower (p < 0.05) in F700 (4.7 ± 3.0 and 1.9 ± 1.7, respectively; mean ± SD) compared to F1000 (8.1 ± 3.8 and 4.4 ± 2.6) and F700P300 (8.5 ± 6.4 and 4.5 ± 3.3). Percentage of cows ovulating >50% of follicles ≥0.8 cm in diameter was lower (p < 0.05) in F700 (35.5%) than in F1000 (82.4%) and F700P300 (73.1%). Percentage of unfertilized oocytes was higher (p < 0.05) in F700 (45.0% vs. 27.7% for F1000 and 29.0% for F700P300) whereas percentage of morulae was higher (p < 0.05) in F1000 (19.3% vs. 8.7% for F700 and 12.2% for F700P300). Embryo quality was similar among groups (p > 0.05). In conclusion, embryo production in lactating dairy cows was improved by increasing total dose of gonadotropins from 700 to 1,000 IU, with SexedULTRA? sex‐sorted semen yielding satisfactory fertilization rates and embryo quality.  相似文献

SUMMARY Oestrus was synchronised in 57 Bos indicus heifers using norgestometoestradiol and pregnant mare serum gonadotrophin. Oestrus was detected by observations made at six-hourly intervals, using oestrogen-treated and chin-ball harnessed steers, heatmount detectors, tail-paint and visual observation. Heifers were inseminated once at either a fixed time of 49.2 ± 0.4 h (mean ± SE; n = 29) after implant removal or 12.6 ± 1.5 h (n = 28) after oestrus was detected. The mean (± SE) time to the onset of oestrus was 47.1 ± 1.9 h, while 90% of heifers recorded in oestrus were detected within 66 h of implant removal. Heatmount detectors were significantly more efficient at detecting oestrus than chin-ball harnessed steers, tail paint or visual observation (P < 0.001). A higher pregnancy rate was obtained in heifers inseminated after oestrus detection compared with heifers inseminated at a fixed-time (57.1 vs 34.5%; P = 0.043) and a higher pregnancy rate was obtained in heifers classified as easy to inseminate compared with heifers classified as difficult to inseminate (57.8 vs 0%, P < 0.001). We conclude that heatmount detectors are an efficient means of detecting oestrus in synchronised B indicus heifers and that pregnancy rates can be increased when insemination follows oestrus detection compared with a fixed-time insemination regimen.  相似文献

A re-evaluation of results from ten trials conducted in commercial dairy herds between 1974 and 1981 in which lactating dairy cows were injected once or twice with either of two forms of prostaglandin F2alpha (PGF) showed that: the variation in the post-injection interval to oestrus was influenced by the stage of the oestrous cycle at which treatment was administered; this variation was sufficient to reduce pregnancy rates to set-time inseminations; an accurate aid for oestrus detection, such as tail painting, should be used routinely with PGF; if PGF-treated cows were detected in oestrus before being inseminated, the pregnancy rates to first insemination were usually 10% higher than in untreated herd mates. The results show that PGF systems in dairy herds should not be used as au alternative to accurate oestrus detection. These systems can be used to condense the breeding programme and increase pregnancy rates to first insemination. The exploitation of this fertility effect justifies the reassessment of PGF usage in New Zealand dairy herds.  相似文献

Flow cytometrically sex‐sorted sperm have been widely used for improving reproductive management in the dairy industry. However, the industrial application of this technology in other domestic species is largely limited by the lower fertility after insemination. The aim of this study was to investigate effects of antioxidant supplementation during the sex‐sorting and freezing process on the quality and functions of sorted sperm from Liaoning Cashmere goats. We tested the effects of antioxidant supplementation during sex‐sorting and freezing process, including ascorbic acid‐2‐glucoside AA‐2G, glutathione, melatonin and vitamin C (VC), on the quality and functions of sex‐sorted fresh and frozen‐thawed sperm. Based on these experiments, we performed deep insemination with sex‐sorted sperm using our improved strategy, in comparison to unsorted sperm. In Experiment 1, compared with control group and other antioxidants, AA‐2G supplementation significantly alleviated the degradation of motility and viability of fresh sperm after sorting and showed the highest percentage of sperm with normal morphology. In addition, AA‐2G supplementation showed an evident protection against the sorting process‐induced membrane and acrosome damage. In Experiment 2, AA‐2G supplementation was most effective in protecting motility, while melatonin supplementation appears to facilitate the degradation of quality of frozen‐thawed sex‐sorted sperm. In Experiment 3, we performed deep insemination with sperm that were sorted and frozen in the presence of AA‐2G and obtained a satisfying pregnancy rate comparable to that from unsorted sperm. The results showed that AA‐2G supplementation efficiently protects quality and function of both fresh and frozen‐thawed sex‐sorted sperm of Cashmere goats, thus obtaining a satisfying pregnancy outcome.  相似文献

Successful sex‐sorting of goat spermatozoa and subsequent birth of pre‐sexed kids have yet to be reported. As such, a series of experiments were conducted to develop protocols for sperm‐sorting (using a modified flow cytometer, MoFlo SX®) and cryopreservation of goat spermatozoa. Saanen goat spermatozoa (n = 2 males) were (i) collected into Salamon's or Tris catch media post‐sorting and (ii) frozen in Tris–citrate–glucose media supplemented with 5, 10 or 20% egg yolk in (iii) 0.25 ml pellets on dry ice or 0.25 ml straws in a controlled‐rate freezer. Post‐sort and post‐thaw sperm quality were assessed by motility (CASA), viability and acrosome integrity (PI/FITC‐PNA). Sex‐sorted goat spermatozoa frozen in pellets displayed significantly higher post‐thaw motility and viability than spermatozoa frozen in straws. Catch media and differing egg yolk concentration had no effect on the sperm parameters tested. The in vitro and in vivo fertility of sex‐sorted goat spermatozoa produced with this optimum protocol were then tested by means of a heterologous ova binding assay and intrauterine artificial insemination of Saanen goat does, respectively. Sex‐sorted goat spermatozoa bound to sheep ova zona pellucidae in similar numbers (p > 0.05) to non‐sorted goat spermatozoa, non‐sorted ram spermatozoa and sex‐sorted ram spermatozoa. Following intrauterine artificial insemination with sex‐sorted spermatozoa, 38% (5/13) of does kidded with 83% (3/5) of kids being of the expected sex. Does inseminated with non‐sorted spermatozoa achieved a 50% (3/6) kidding rate and a sex ratio of 3 : 1 (F : M). This study demonstrates for the first time that goat spermatozoa can be sex‐sorted by flow cytometry, successfully frozen and used to produce pre‐sexed kids.  相似文献

The aim of this study was to investigate the expression of progesterone receptor (PR) in the utero‐tubal junction (UTJ) of sows at 24 h after intra‐uterine insemination (IUI) and deep intra‐uterine insemination (DIUI) compared with conventional artificial insemination (AI) in pigs. Fifteen multiparous sows were used: AI (n = 5), IUI (n = 5) and DIUI (n = 5). The sows were inseminated with a single dose of diluted semen during the second oestrus after weaning at 6–8 h prior to ovulation (AI: 3000 × 106 spermatozoa, IUI: 1000 × 106 spermatozoa and DIUI: 150 × 106 spermatozoa). The UTJ was collected and subject to immunohistochemical staining using avidin‐biotin immunoperoxidase technique with mouse monoclonal antibody to PR. In the oviductal part of the UTJ, the intensity of PR in the tunica muscularis and the proportion of PR‐positive cells in the surface epithelium after DIUI were lower than AI (p < 0.05). The intensity and the proportion of PR‐positive cells between AI and IUI in all compartments of the UTJ did not differ significantly (p > 0.05). When comparing between tissue compartments, prominent staining was observed in the muscular layer of the UTJ. It could be concluded that the expression of PR in the UTJ prior to fertilization after DIUI with a reduced number of spermatozoa was lower than that after AI. This might influence sperm transportation and the fertilization process.  相似文献

PGF2α either as a subcutaneous injection (8 or 10 mg) or intrauterine infusion (dose 5 mg) successfully synchronised oestrus in beef cattle AI programs. The beef cattle programs were managed by 1 of the following 2 systems.
  • 1 Two 10-day insemination periods, 1 for the first and the second for the repeat inseminations. Oestrus was detected and oestrus cows inseminated for the first 4 days of the first period. On the fifth day all cows not inseminated were treated with PGF2α. The treated cattle came into oestrus and were inseminated over the following 2 to 5 days.
  • 2 Two PGF2α treatments 10 days apart followed by two 4-day insemination periods. During the first 4-day period, cows exhibiting synchronised oestrus were inseminated. Cows returning to oestrus were inseminated during the second 4-day period, about 18 days after the first period.
Synchronisation of oestrus was not sharp, oestrus being distributed over 7 days, with the majority of cows being in oestrus on the third (42.3%) and fourth (22.4%) days. The route of administration of PGF2α (injection or infusion) did not effect oestrus synchronisation. PGF2α by either route of administration did not effect the fertility of cows inseminated at either the synchronised or subsequent oestrus. Pregnancy rates at the synchronised oestrus were higher when cows were inseminated after detection of oestrus than when they were inseminated without reference to oestrus at a fixed time 3 days after PGF2α treatment (mass injection). Oestrus synchronisation greatly reduced the input of time, labour and feed required during the total program. However, oestrus synchronisation did not alter the overall efficiency of artificial breeding programs measured in terms of the proportion of cattle entering a program that were inseminated and became pregnant. The main problem encountered was drug wastage through unknowingly treating cows and heifers that were not cycling.  相似文献

The aim of this study was to compare pregnancy per artificial insemination (P/AI) after timed AI with sex‐sorted sperm (SS) or conventional semen (CS) in lactating dairy cows. Cyclic cows (n = 302) were synchronized by Ovsynch and randomly assigned into two groups at the time of AI. Cows with a follicle size between 12 and 18 mm and clear vaginal discharge at the time of AI were inseminated with either frozen‐thawed SS (n = 148) or CS (n = 154) of the same bull. A shallow uterine insemination was performed into the uterine horn ipsilateral to the side of probable impending ovulation. Pregnancy per AI on Day 31 tended (p = 0.09) to be less for SS (31.8%) than CS (40.9%). Similarly, P/AI on Day 62 was less (p = 0.01) for cows inseminated with SS (25.7%) compared with CS (39.0%). The increased difference in fertility between treatments from Days 31 to 62 was caused by the greater (p = 0.02) pregnancy loss for cows receiving SS (19.2%) than CS (4.8%). Cow parity (p = 0.02) and season (p < 0.01) when AI was performed were additional factors affecting fertility. Primiparous cows had greater P/AI than multiparous cows both on Day 31 (41.7% vs 25.0% in SS and 53.0% vs 31.8% in CS groups) and on Day 62 (33.3% vs 20.5% in SS and 48.5% vs 31.8% in CS groups). During the hot season of the year, P/AI on Day 31 was reduced (p = 0.01) in the SS group (19.6%) when compared with the rates during the cool season (38.1%). In conclusion, sex‐sorted sperm produced lower fertility results compared to conventional semen even after using some selection criteria to select most fertile cows.  相似文献

Artificial insemination (AI) in pigs has been established for about four decades but ejaculates are still used insufficiently. Higher demand of semen for AI and new techniques that involve low sperm concentration require the optimization of insemination protocols. Based on the knowledge of the physiology of sperm transportation and events in the female genital tract prior to fertilization, new strategies are under development to minimize sperm losses. One goal is to deposit the semen into the uterine horn rather than into the proximal cervix. It was shown that the minimal number of spermatozoa necessary for surgical AI at the utero‐tubal junction (UTJ) were at least 1 × 106 diluted in 0.5 ml of a special extender. Artificial insemination into the distal part of the uterine horn required about 1 × 107 million sperm in 20 ml of extender. Meanwhile, first insemination devices for non‐surgical intra‐uterine AI are commercially available. Using similar sperm concentrations as for surgical AI, non‐surgical uterine insemination did not differ significantly from control inseminations in terms of pregnancy rate and litter size. With respect to the fertilizing capacities of their ejaculates, boars have to be selected more strictly for sperm quality parameters as most of the compensatory effects of sperm cells disappear in maximally extended semen samples.  相似文献

Deep intra‐uterine insemination is commonly accepted as a routine procedure for artificial insemination in horses. The motives and principles of deep insemination are well described, but the equipment used may differ. In this trial, the efficiency of two different insemination pipettes for deep intra‐uterine insemination in the mare was compared with insemination into the uterine body using commercially available frozen–thawed semen of two stallions of proven fertility. These inseminations were performed using two different doses. The semi‐flexible Minitube pipette was compared with a newly designed insemination device with a more flexible telescopic insemination catheter (Ghent device). The semi‐flexible Minitube pipette performed better than the newly designed insemination device with respect to pregnancy outcome (p = 0.008). The superiority of deep horn insemination over uterine body insemination was reflected by the better pregnancy rates obtained after deep insemination using the same low doses (30.6% better pregnancy rates) (p = 0.0123).  相似文献

Flow cytometrically sex sorted spermatozoa are reduced in their fertilizing capacity, particularly when stored either in cooling extender or after freezing in liquid nitrogen. So far, preservation methods for sorted spermatozoa have differed only marginally from procedures used for unsorted semen. In the present study, a TRIS extender was modified to balance major cell damage caused by the sorting process and by liquid storage of the sorted spermatozoa. The new extender, containing a combination of antioxidants (AO) and bovine serum albumin (BSA), significantly increased the lifespan and fertilizing capacity of sex sorted spermatozoa. No significant differences were observed between unsorted controls and sorted samples for motility and status of sperm membranes as tested by fluorescein-isothiocyanat-peanut agglutinin/propidium iodide (FITC-PNA/PI). Acrosome integrity of spermatozoa was significantly better when semen was stored at 15 degrees C for 24 and 48 h in an extender containing AO with or without BSA as compared with controls (p < 0.05). There were no significant differences, in pregnancy rates of heifers inseminated at a natural oestrus, between unsorted controls (16/24, 66.7%) and both sorted groups (AO + BSA: 18/31, 58.1% and AO-BSA: 12/22, 54.5%). Additionally, it was shown for the first time that artificial insemination (AI) with liquid sexed bull spermatozoa stored for 72 h after sorting can result in pregnancy rates similar to AI with non-sorted semen.  相似文献
