肉鸡热休克蛋白70 mRNA荧光定量PCR方法的建立和优化   总被引:1,自引:0,他引:1  
对适应性饲养到30日龄时的60只肉鸡进行热应激处理,通过热休克蛋白70(heatshockprotein70,HSP70)mRNA荧光定量PCR(fluorescencequantitativePCR,FQ-PCR)方法检测热应激处理肉鸡组织中HSP70mRNA含量。虽然受试鸡肝脏和心脏的HSP70mRNA水平在热应激6h时略低于对照组(P>0.05),但随持续性高温应激时间的延长,HSP70mRNA水平逐渐升高,热应激18h时应激肉鸡肝脏和心脏HSP70mRNA水平达最高水平,明显高于对照组(P<0.01)。实验选用3-磷酸甘油醛脱氢酶基因(GAPDH)作为内参照,体外转录的RNA和阳性质粒作为两种标准品。结果显示,实验所建立和优化的FQ-PCR反应体系是理想的。  相似文献

体外培养的奶牛,绵羊和小鼠的附植前胚胎在热应激条件下能够合成热休克蛋白70以增强胚胎的耐热性保护其不受热损伤.而钙调蛋白(CaM)是细胞内最重要的Ca2+受体蛋白并参与细胞活动中重要基因的转录活动.本实验旨在研究CaM参与小鼠(Mus musculus)胚胎热应激诱导型热休克蛋白70(HSP70)的表达,且明确CaM参与HSP70表达的具体途径.将发育至早期囊胚的胚胎分成空白对照(37℃)组、W7处理非热应激(37℃+W7)组、热应激(39℃)组和W7处理热应激(39℃+W7)组.提取总RNA并分别检测HSP70、CaM和热休克转录因子1(HSFl) mRNA表达;提取胞浆蛋白用Western blot技术检测HSP70、CaM和HSF1表达;用免疫共沉淀法检测HSP70-CaM和HSP70-HSF1复合物.结果发现,W7能显著抑制39℃热应激1h小鼠胚胎HSP70 mRNA和蛋白的表达(P<0.05); W7对其他各组胚胎HSF1mRNA表达的影响不显著(P>0.05),但能显著降低39℃热应激1h小鼠胚胎HSF1蛋白表达(P<0.05); 39℃热应激时,胚胎HSP70-CaM复合物增多,HSP70-HSF1复合物减少.本研究表明,小鼠胚胎受热应激时,CaM通过与HSP70竞争性结合,解离出HSF1,从而使HSP70大量表达.本研究揭示了CaM参与小鼠胚胎热激反应中HSP70表达的一种途径.可为研究胚胎耐热性以及热激信号转导提供基础资料.  相似文献

热休克蛋白60(HSP60)不仅在细胞保护方面发挥作用,也参与雄性动物生殖生理的调控。为研究HSP60基因序列特性及其在天祝白牦牛下丘脑-垂体-睾丸轴(HPT-axis)上的表达情况,探寻其在白牦牛睾丸发育、精子发生过程中的作用机制,本研究采用RT-PCR和RACE技术获得白牦牛HSP60基因全长cDNA序列,采用生物信息学方法分析HSP60蛋白的理化性质、结构及不同物种之间的同源性等,并利用qPCR、Western blotting、免疫组化法分析HSP60在白牦牛下丘脑-垂体-睾丸轴中的表达及定位。结果表明,白牦牛HSP60基因cDNA全长为2 300 bp,开放阅读框为1 722 bp,编码572个氨基酸;其理论分子量为60.977k Da、等电点为5.69,编码的蛋白为非跨膜可溶性蛋白。氨基酸序列比对结果显示,白牦牛HSP60氨基酸序列与牛、瘤牛、绵羊、藏羚羊、骆驼、白犀牛、兔和黑猩猩的氨基酸序列同源性高、进化水平相近。HSP60基因及蛋白在白牦牛的下丘脑、垂体及睾丸组织中均有表达,其中下丘脑及垂体组织表达量显著高于睾丸,下丘脑和垂体之间差异不显著。免疫组化结果显示,HSP60蛋白定位表达于白牦牛下丘脑组织的室旁核大细胞、室旁核小细胞和神经角演网、垂体组织的腺细胞、睾丸组织的精原细胞、精母细胞、支持细胞和间质细胞,而精子细胞中表达较弱。通过对健康成年白牦牛HSP60在下丘脑-垂体-睾丸轴的表达与定位检测,推断HSP60参与雄性白牦牛生殖轴调控,并参与睾丸发育与精子发生。本研究结果为进一步研究HSP60对雄性生殖调控奠定了基础。  相似文献

灯盏花乙素(scutellarin,Scu)具有清热解毒、扩张心脑血管、改善微循环等作用。通过观察不同浓度Scu对猪肾小管上皮细胞(pig kidney proximal tubular,LLC-PK1)热休克蛋白72(heat shock protein72,HSP72)、B细胞淋巴瘤/白血病-2(B-cellymphoma/leukemia-2,Bcl-2)及Bcl-2相关X蛋白(Bcl-2associated X protein,Bax)表达的影响,探讨灯盏花乙素提高细胞耐热性的可能机制。将培养的LLC-PK1细胞随机分为37℃空白对照(Ⅰ组),42℃单纯热应激1 h(Ⅱ组),以及分别用不同浓度(1×10-5、1×10-6和1×10-7mol/L)Scu处理并42℃热应激1 h组(分别为Ⅲ、Ⅳ和Ⅴ组),提取各组细胞总RNA和总蛋白,用实时荧光定量PCR和Western blot分别检测HSP72、Bcl-2和Bax基因及蛋白的表达量。结果显示,与Ⅰ组相比,Ⅱ组LLC-PK1细胞HSP72和Bax的mRNA及蛋白表达量显著升高(P0.05),Bcl-2/Bax mRNA和蛋白比值显著降低(P0.05),Bcl-2 mRNA和蛋白表达量并无显著差异(P0.05)。与Ⅱ组相比,Ⅳ和Ⅴ组LLCPK1细胞HSP72 mRNA和蛋白的表达量均显著升高(P0.05),Ⅲ组无显著差异(P0.05);Ⅳ组细胞Bcl-2 mRNA和蛋白表达量均显著升高(P0.05),Ⅲ和Ⅳ组并无显著差异(P0.05);Ⅲ、Ⅳ和Ⅴ组Bax mRNA和蛋白表达量均显著降低(P0.05);Ⅳ和Ⅴ组Bcl-2/Bax蛋白比值显著升高(P0.05)。研究结果表明,热应激条件下Scu可诱导LLC-PK1细胞HSP72表达,提高Bcl-2/Bax比值。结果提示,Scu可增强细胞的抗热休克作用。本研究丰富了体外培养细胞热应激反应的理论,为在实践中增加细胞抗热休克能力提供了理论依据。  相似文献

高温诱导体外培养奶牛乳腺上皮细胞的应激响应   总被引:1,自引:0,他引:1  
实验以体外培养的奶牛乳腺上皮细胞为模型,研究不同时间高温(42℃)热处理对细胞热休克蛋白HSP、细胞凋亡、乳脂肪和乳蛋白合成相关基因mRNA表达丰度的影响.结果发现,热休克转录子hsf-1和热休克蛋白基因hsp27、hsp70和hsp90在高温处理0.5 h后mRNA表达上调,其中hsp70转录水平有急剧上调过程,且随热处理时间延长均表现出先上升后下降的趋势;标志细胞凋亡的Bax基因从处理的0.5 h开始mRNA表达下调,随后升高;但细胞凋亡Bcl-2 mRNA表达上调;在检测的热处理过程中,αS1-酪蛋白(CSN1S1)基因转录先上调后下降,而β-酪蛋(CSN2)和嗜乳脂蛋白(BTN)基因转录均下调;与脂肪酸合成和调控相关的基因乙酰辅酶A羧化酶(ACACA)、过氧化物酶激活受体.-γ(PPARG)和固醇调节元件转录因子-1(SREBF-1)基因表达显著上调,而过氧化物酶激活受体-α(PPARA)基因mRNA转录没有明显变化.本实验结果证明热应激诱导了奶牛乳腺上皮细胞的应激反应,对细胞的乳蛋白和乳脂肪合成功能产生了显著影响,细胞产生热耐受.  相似文献

利用间接法免疫组织化学技术和RNA探针原位杂交技术,研究了HSP70 及HSP70 mRNA在热应激6 h处理肉鸡心脏、肝脏、肺脏、肾脏、脾脏、胸腺和法氏囊等组织中的分布。结果显示,热应激6 h时HSP70 mRNA主要分布在肉鸡肝脏和肺脏中的血管壁及其周围,心肌纤维也有少量阳性信号,而脾脏、法氏囊和胸腺等组织未检出现阳性信号;热应激6 h时HSP70主要分布在所检肉鸡各种组织的血管壁。提示HSP70与热应激肉鸡心血管系统具有相关性,可能与心血管系统的功能和损伤有关。  相似文献

热激蛋白90家族(heat shock protein 90(HSP)family)是一种进化保守的蛋白质,在分子伴侣功能、细胞循环调控、抗原传递方面发挥着重要作用。本研究用RT-PCR和RACE技术,克隆得到了松材线虫(Bursaphelenchus xylophilus)、拟松材线虫(Bursaphelenchus mucronatus)和豆伞滑刃线虫(Bursaphelenchus doui)热激蛋白90基因的cDNA全长序列,分别命名为Bx-hsp90、Bm-hsp90和Bd-hsp90,(GenBank登录号分别为:GU013561、HMO34943和HM347331),其cDNA全长分别为2255、2193和2232bp,开放阅读框均为2127bp,编码708个氨基酸,5'端非编码区的长度分别为33、13和13bp,3'端非编码区的长度分别为95、53和92bp。其DNA全长分别为2440、2388和2407bp,包含3个内含子。三者氨基酸序列的一致性为98.78%。进化分析表明,松材线虫、拟松材线虫、豆伞滑刃线虫与秀丽小杆线虫(Caenorhabditis elegans)的亲缘关系较近,推测三...  相似文献

根据GenBank发表的鸡和鸭等物种的脂联素基因序列的同源保守区域,设计了3对特异性引物,以鹅(Anser domeatica)血液基因组DNA为模板,经扩增、测序及拼接得到了部分DNA序列,全长1062bp,包含2个外显子和1个内含子(GenBank Accession No.EU370686)。该基因编码区长738bp,编码245个氨基酸,编码区核苷酸序列及所编码氨基酸序列与鸭的同源性最高,分别为94.85%和95.51%,与鸡的同源性次之,而与哺乳类的同源性较低。蛋白预测的分子量和等电点分别为26603.3D和5.19,与鸡、鸭、人及小鼠的脂联素蛋白相似。半定量PCR结果显示鹅脂联素基因在骨骼肌、脂肪、心脏和肌胃中高度表达,在小肠、腺胃、肾和肺中中度表达,而在肝脏、脾、卵巢和间脑中低度表达。  相似文献

用RT-PCR方法,从黄颡鱼(Pelteobagrus fulvidraco)肝脏中克隆获得了hepcidin cDNA序列,并对其进行序列测定,在此基础上分析了hepcidin基因在不同组织中的表达差异和不同细菌感染后对其表达的影响。结果表明,黄颡鱼hepcidin的cDNA片段(GenBank登陆号EU257703)长为282bp,编码93个氨基酸,由信号肽(23个氨基酸)、前肽(45个氨基酸)和成熟肽(25个氨基酸)组成,成熟肽含有8个保守的半胱氨酸(cys)残基,可形成4个链内二硫键。与已报道的其他鲶形目鱼类hepcidin核苷酸序列的同源性为80-82%,推导的氨基酸序列的同源性为69-71%。RT-PCR半定量分析显示黄颡鱼hepcidin基因在不同组织中普遍表达,其中肝脏表达水平最高,头肾、肠道、肌肉、脑、胃、鳃、心脏、卵巢表达次之,皮肤、脾脏、肾脏表达较低。感染嗜水单胞菌和腊样芽孢杆菌的黄颡鱼,肝脏hepcidin mRNA水平显著升高。  相似文献

热激蛋白90(heat shock protein 90,Hsp90)调控生物体的细胞代谢、生长发育和逆境适应等一系列生物过程.本研究通过cDNA末端快速扩增技术(rapid amplification of cDNA ends,RACE)分离获得了一个水稻干尖线虫(Aphelenchoides besseyi)Hsp90基因,命名为Ab-hsp90.Ab-hsp90全长cDNA序列含有69bp的5’非翻译区、编码712个氨基酸2 139 bp的开放阅读框以及137 bp的3'非翻译区UTR.Ab-hsp90DNA序列包含9个外显子和8个内含子.Ab-hsp90蛋白序列与其他植物寄生线虫Hsp90s高度相似,最大似然树显示与植物寄生线虫位于同一进化分支;结构上具有5个保守的Hps90蛋白家族序列标签和特异MEEVD基系,表明Ab-hsp90为胞质型Hsp90.qRT-PCR显示,Ab-hsp90在水稻干尖线虫各个龄期均表达,在卵和成虫表达水平最高.当线虫暴露于干燥、低渗透胁迫以及短时间阿维菌素溶液中时Ab-hsp90均上调表达;Ab-hsp90还受到短时低温(4℃)诱导,在高温(40℃)环境持续高水平表达.在侵染抗感不同的水稻(Oryza sativa)植株时,Ab-hsp90的表达水平具有差异性:在线虫接种抗性水稻72 h内,Ab-hsp90一直高度表达,而在接种感病水稻上Ab-hsp90表达水平显著升高随后下降.取食灰葡萄孢(Botrytis cinerea)的线虫Ab-hsp90在9d内始终高度表达,而胡萝卜(Daucus carota)愈伤组织培养的线虫6d内Ab-hsp90表达上调但此后逐渐下调.这些数据表明,Ab-hsp90可能参与水稻干尖线虫的抗逆适应、早期侵染以及取食行为.本研究有助于拓展植物寄生线虫Hsp90蛋白的功能,为进一步研究线虫与植物互作机制提供了有益借鉴.  相似文献

Climate change and rising carbon dioxide (CO2) levels are expected to reduce the mineral nutrient content of soybean seeds. The main objective of this study was to survey diverse soybean germplasm for variation in seed elemental concentrations and their relationships between elements, protein content, and individual seed weight. Seeds from 90 soybean genotypes were weighed and subjected to inductively coupled plasma-mass spectrometry (ICP-MS) ionomics analysis and Carbon/Nitrogen (C/N) analysis to determine protein. The results demonstrated substantial variation with the possibility of significantly improving most mineral nutrients, especially selenium (Se), copper (Cu), iron (Fe), and manganese (Mn). This diverse survey identifies genotypes that can complement existing soybean breeding programs for improving seed nutritional quality. Correlation analysis identified two clusters of co-variant elements: zinc (Zn), phosphorus (P), and sulfur (S) as well as Zn, Cu, Se, and rubidium (Rb) were positively correlated with each other. Tolerable upper limits of Rb intake are not defined for humans illustrating the need to monitor trace elements along with desirable nutrients.  相似文献

90-day oral toxicity study of a grape seed extract (IH636) in rats   总被引:1,自引:0,他引:1  
To assess the safety of grape seed extract with less than 5.5% catechin monomers (IH636), 4 groups of male and female Sprague-Dawley rats were provided grape seed extract in the diet at levels of 0 (control), 0.5, 1.0, or 2.0% for a period of 90 days. All animals survived the duration of the study, and no significant changes in clinical signs, hematological parameters, organ weights, ophthalmology evaluations, or histopathological findings were observed. A significant increase in food consumption was observed in male and female rats provided the grape seed extract diets compared to that of the control rats, especially in male rats consuming 2.0% grape seed extract. This effect was not accompanied by increases in body weight gains. Grape seed extract appeared to increase the insoluble fraction of the diet. Male rats in the high-dose group exhibited decreased serum iron levels and decreased serum iron/total iron binding capacity ratio compared to those of the controls, although all values were within historical ranges for Sprague-Dawley rats. In conclusion, administration of the grape seed extract IH636 to male and female Sprague-Dawley rats in the feed at levels of 0.5, 1.0, or 2.0% for 90 days did not induce any significant toxicological effects.  相似文献

Wheat flour and water mixtures at 90% absorption (dry flour basis) prepared at various mixing times were examined using Fourier transform infrared (FT‐IR) reflectance spectroscopy. Spectra were obtained using a horizontal attenuated total reflection (ATR) trough plate. The apparent amount of protein and starch on the surface of the dough varied with mixing time but this was likely due to the polyphasic nature of the substrate and the changing particle distributions as the batter matrix was developed. Deconvolution of the Amide I band revealed contributions from alpha helical, β‐turn, β‐strand, β‐sheet, and random conformations. The ratio of β‐sheet to nonsheet conformations reached its greatest value about the same time that the mixture was most effectively separated by a laboratory‐scale, cold‐ethanol‐based method but before the peak consistency measured by a microfarinograph.  相似文献

本文报道了两种淡水腹足类动物对90 Sr的浓集与分布的过程 ,大瓶螺 (Am pullariagigasSpix) 4h开始便迅速浓集90 Sr并转至螺体各个部位 ,小个体各器官组织对90 Sr的浓集系数大于大个体 ,刚孵化的幼螺在 1 5min内便对90 Sr有浓集作用 ,并随时间的延长而增加。石螺 (Bellarnyapurificata)对90 Sr于 4h开始有吸收浓集作用 ,但其浓集系数低于大瓶螺 ,在各自积累高峰期大瓶螺的贝壳对90 Sr的浓集系数为石螺的 1 4倍。两种母螺的卵和子螺的90 Sr比活度均高于同时间两种螺贝壳的比活度 ,说明90 Sr通过母体迅速转移至子体  相似文献

为获得可以区分自然感染和疫苗免疫且毒力较弱的布鲁氏菌候选疫苗株,本实验研究了磷酸葡萄糖变位酶基因(pgm)对布鲁氏菌(Brucella melitensis)M5-90疫苗株毒力的影响,并对其进行了免疫评价。本实验构建了重组质粒pGEM-7zf-Δpgm,电转化布鲁氏菌M5-90感受态细胞,筛选获得布鲁氏菌疫苗株M5-90的pgm基因缺失株(Δpgm),并对获得的M5-90 Δpgm株进行遗传稳定性、免疫原性检测。结果显示,Δpgm能稳定传15代;Δpgm组的抗体含量较亲本株M5-90组差异不显著;Δpgm在诱导机体产生IL-2的能力要强于M5-90,产生INF-γ的能力低于M5-90;该基因缺失株采用虎红平板凝集试验和试管凝集试验不发生凝集反应;Western blot证实,Δpgm不能诱导小鼠产生抗pgm蛋白的抗体。本研究构建的Δpgm具有很好的遗传稳定性和免疫原性,为研制布鲁氏菌基因缺失疫苗提供了实验依据。  相似文献

The monoculture cropping system causes significant changes within the soil ecosystem, which constitutes a habitat for soil-dwelling springtails. Focusing on the response of soil fauna to 90 years of potato cultivation in monoculture the study investigates the abundance and diversity of soil-dwelling springtails, considering changes in the soil environment in relation to five-crop rotation. Another point was the soil quality evaluation using Collembola as bioindicators (QBS-c index). A long-term monoculture experiment was established in Poland in 1923 and has continued uninterruptedly to the present time. Soil samples were taken over a period of three years (2011–2013) to determine collembolan abundance and composition, as well as physical and chemical soil properties.

The study demonstrated that there were greater numbers of Collembola in the long-term monoculture of potatoes, especially before planting time, compared to numbers in a five-field crop rotation. At the same time apparently greater species diversity was found in potato culture within crop rotation. The biological indicator of soil quality based on the occurrence of springtails (QBS-c) has proved useful in assessing changes in soil caused by agrotechnical activities. This index indicated better biological soil quality in the five-field rotation system compared to monoculture.  相似文献
