The development of the parasite and lesions was studied in 32 sheep killed 10 days to 47 months after inoculation with Sarcocystis gigantea sporocysts from cats. At 21-42 days post-inoculation (d.p.i.), there was a mild encephalitis, but organisms were not seen in the brain. Immature sarcocysts were detected from 40-84 d.p.i. The cyst wall was not measurable by light microscopy at 40 d.p.i., but was 1.5-2 microns thick at 84 d.p.i. At 119 d.p.i. both immature cysts containing only metrocytes, and mature cysts containing both metrocytes and merozoites, were present. These mature cysts did not have a secondary cyst wall. A mature cyst, 350 microns in length, was found in a sheep killed at 8 1/2 months p.i. At 10 m.p.i. cysts were up to 0.5 mm long and a secondary cyst wall was present. At 47 m.p.i. cysts were 2-5 X 4.5-7.5 mm, and were found only in the muscles of tongue, oesophagus, pharynx and flank.  相似文献

Fresh preparations of micro-isolated sarcocysts from skeletal and cardiac muscle of 12 reindeer were examined by light microscopy. On the basis of cyst structure and cyst wall structure 4 Sarcocystis spp. could be differentiated. New names have been proposed for 2 previously unnamed Sarcocystis spp. of reindeer, and S. grueneri has been redefined.S. rangiferi n. sp. had macroscopic cysts in skeletal muscle measuring 2106×403 µm. The cyst wall protrusions were finger-like and measured 13.2×6.7 µm. The cysts were surrounded by a layer of fibrillar material.S. tarandi n. sp. had micro- to macroscopic cysts primarily in skeletal muscle, but a few cysts were found in the heart of one animal. In skeletal muscle the cysts measured 999×75µm; in the heart the cysts were shorter and wider. The cyst wall protrusions were fingerlike and measured 9.2×2.2 µm.S. grueneri had micro- to macroscopic cysts in cardiac muscle measuring 581×137 µm. The cyst wall was thin and relatively smooth with no visible protrusions.Sarcocystis sp. had micro- to macroscopic, slender cysts in skeletal muscle measuring 916×64 µm. The cyst wall had tightly packed, short, knob-like protrusions. The cysts of this species were previously classified as cysts of S. grueneri.  相似文献

Six 34- to 42-day-old lambs raised in coccidia-free conditions were inoculated with 70,000 sporocysts derived from sheep heart with microscopic sarcocysts. Fever and mild anorexia occurred between 25 and 33 days after inoculation. A transient anaemia was most marked 32 days after inoculation. Lambs were killed and examined 14, 25, 33, 42, 60 and 81 days after inoculation. Gross lesions were absent. First and second generation meronts were present in endothelial cells at 25 and 33 days after inoculation. Meronts were most numerous in kidney glomeruli. Developing sarcocysts were rare at 42 days after inoculation. Sarcocysts with a primary cyst wall 2 to 3 micron thick composed of palisade projections were common at 60 and 81 days after inoculation in striated muscle and brain. Mild to severe striated muscle myositis and non-suppurative encephalitis or encephalomyelitis with glial nodules were observed 25 to 81 days after inoculation. Sarcocyst frequency varied considerably; it was highest in myocardium, M vastus intermedius, M vastus medialis, M extensor carpi radialis and tongue muscle and was lowest in M masseter.  相似文献

Tissue samples comprising the oesophagus and diaphragm were collected from 400 sheep and 400 goats slaughtered at the abattoirs in the study area. Out of this number, 36 were positive for Sarcocystis cysts (sarcocysts) in sheep and 56 in goats. The sarcocysts in sheep measured 35.7 to 500 microns lengthwise and the cyst-wall 2.4 microns. They were identified to be Sarcocystis tenella. The cysts in goats measured 98 to 700 microns and the cyst-wall 2.7 microns. They were identified to be Sarcocystis capracanis. In both animals species, the sarcocysts were more frequent in the oesophagus than in the diaphragm. All sarcocysts seen were microscopic.  相似文献

Fresh preparations of microisolated sarcocysts from striated muscle of several domestic reindeer from northern Norway were examined by light microscopy. In cardiac muscle, cysts of S. grueneri were found. In skeletal muscle, cysts of S. rangiferi, S. tarandi and S. tarandivulpes were found in all samples examined. In the abdominal muscles of some reindeer, one or two other types of cysts were found.Cysts of one type were macroscopic in size, and ovoid to cylindrical in shape. The cysts were surrounded by a 8–12 µm thick layer of fibrous material, and measured 1682×910 µm. The cysts had relatively few and irregularly distributed, 20–35 µm long, and 3–5 µm wide, linguiform cyst wall protrusions, which could only be seen after removal of the fibrous layer. These cysts were classified as cysts of S. hardangeri, a species previously described from wild reindeer in southern Norway.Cysts of the other type were long and slender, measuring 5460–12700 (8994 ± 2575) × 95–280 (180 ± 50) µm. The cysts had numerous very fine, flexible, hair-like cyst wall protrusions, which were 8–10 [xm long and less than 0.5 µm thick. These cysts are considered to belong to a new Sarcocystis species of reindeer, for which the name Sarcocystis rangi n, sp. is proposed. The reindeer is recorded as the intermediate host for 6 different species of Sarcocystis.  相似文献

The susceptibility of dogs to experimental inoculation with trophozoites and cysts of human isolates of Giardia duodenalis and the clinical and laboratory profiles of infection of these animals were studied. Two groups (A and B), each comprising three dogs, were inoculated with G. duodenalis trophozoites and cysts, respectively. A third group of two dogs was not inoculated and remained as control. After inoculation feces were collected daily to determine the pre-patent period, by flotation in 33% zinc sulfate solution. Blood samples (5mL) were collected from animals at 15-day intervals during the 165 days of the experimental period and were used to carry out the hemogram and biochemical evaluation of the levels of total protein, albumin, alanine aminotransferase, gamma glutamyltransferase, urea and creatinine. A prepatent period was observed at 5-6 days post-inoculation (p.i.) in the inoculated dogs, with cysts eliminated for approximately 3 months. No alterations were seen in the clinical parameters evaluated. Anemia was observed at 15 p.i. in the inoculated dogs. The mean eosinophil count of inoculated groups was higher than that of the control (p< or =0.05) but none of the biochemical parameters analyzed presented significant differences. The results of this study show that G. duodenalis from human isolates is able to infect dogs with minimal systemic manifestations without producing clinical signs of giardiasis.  相似文献

For this investigation 50 Brown Swiss cows from 21 different farms were used. Twenty-five peripartal overconditioned cows (back fat thickness > 38 mm) were compared with 25 peripartal not overconditioned animals (back fat thickness < 38 mm). On days 20, 30 and 40 post partum the ovaries were examined sonographically and 10, 15, 20, 30 and 40 days after calving plasma concentrations of progesterone and 17-beta estradiol were determined. In peripartal overconditioned animals 12 ovarian cysts were found while only one cyst was present in not overconditioned cows (P < 0.05). At first examination all ovarian cysts were classified by ultrasound as follicle theca cysts (progesterone < 0.5 ng/ml plasma). Follow examinations resulted in 3 cysts which persisted as theca cysts while 8 cysts became luteinized and 2 cysts completely regressed. There was no indication of increased plasma progesterone and/or estradiol concentrations in overconditioned cows with higher fat deposit before of ovarian cysts had occurred.  相似文献

Fifty, 2–3 month old dairy goats were each vaccinated orally with 10000 sporocysts of Sarcocystis capracanis and 25 age-matched goats served as uninoculated controls. Groups of vaccinated and control goats were challenged with lethal doses of S. capracanis at 95, 113, 205, and 274 days post-vaccination. Vaccinated goats developed subclinical sarcocystosis. Twenty-three vaccinated goats and 1 control goat died of intestinal coccidiosis and bacterial pneumonia, 15–118 days after vaccination. Myositis and degenerating sarcocysts were seen in muscles of goats necropsied at 90–186 days postvaccination. Very few, or no sarcocysts were seen in goats necropsied at 272 and 332 days post-vaccination. Vaccinated goats survived a lethal challenge inoculation indicating persistent protective immunity.  相似文献

Sporocysts of Sarcocystis falcatula obtained from experimentally infected Virginia opossums (Didelphis virginiana) were inoculated orally to 60 wild-caught Brown-headed Cowbirds (Molothrus ater). Another 30 Brown-headed Cowbirds were not challenged and served as uninfected controls. Two inoculated and one control cowbird were necropsied every 2 weeks and the pectoral and thigh muscles were examined grossly for cyst development. Stained histologic sections of pectoral muscle, thigh muscle, and lung were examined by light microscopy and presence, density, and size of sarcocysts were determined. Sarcocysts were present by 6 weeks post-inoculation (PI) and were still growing at 40 weeks PI. The sarcocysts from birds 40 weeks post-infection were infective to an opossum. The morphology of the sarcocyst wall by transmission electron microscopy substantiated the identification as S. falcatula. Lung sections were examined for the presence of schizonts, but were seen only at 2 weeks PI. This evaluation was complicated by the presence of unidentified microfilariae. These birds are migratory and the continued growth and development of muscle cysts would allow them to be a source of infection at both extremes of their geographic range, regardless of which end of the migration at which they were infected.  相似文献

The aims of this study were to assess the accuracy of different common methods of differentiating between follicular and luteal ovarian cysts, and to monitor the response of the cysts to 12 days treatment with a progesterone-releasing intravaginal device (PRID). On the basis of agreement between the different methods, 25 of the 46 cases examined were diagnosed as follicular and 14 as luteal cysts; for the other seven cases the methods disagreed. The use of ultrasound was more accurate in diagnosing follicular cysts than luteal cysts, and combined with plasma progesterone concentrations gave the most accurate assessment of cyst type (92 per cent for follicular cysts and 82 per cent for luteal cysts). The mean (se) plasma progesterone concentration was lower in the cows with follicular cysts than in those with luteal cysts (0.29 [0.05] v 3.90 [0.63] ng/ml; P<0.05). Luteal cysts had thicker walls (5.3 [0.04] v 2.5 [0.2] mm; P<0.0001), and the wall thickness of all the cysts was positively correlated with plasma progesterone concentration (r=0.52, P<0.0004). Cows with luteal cysts had more additional follicles greater than 5 mm in diameter (P<0.01). In cows with follicular cysts and other follicles greater than 5 mm in diameter, the mean oestradiol concentration was 7.9 (1.8) pg/ml compared with 24.2 (3.1) pg/ml (P=0.002) in cows without other follicles greater than 5 mm in diameter on either ovary. At the time of PRID removal, plasma progesterone concentration had increased in the cows with follicular cysts to 1.59 (0.06) ng/ml (P<0.05) and decreased in the cows with luteal cysts to 0.87 (0.01) ng/ml (P<0.05), although there was no change in original cyst structure in 45 per cent of the cases. However, new ovarian structures were frequently observed during the treatment. The overall pregnancy rate for cows with both types of cyst after treatment was 50 per cent after three inseminations, but the first service pregnancy rate was only 18 per cent for cows with follicular cysts and 28 per cent for cows with luteal cysts. After treatment, the fertility of cows with follicular cysts was similar to that of paired herdmates, whereas cows with luteal cysts took 40 days longer to calve again than healthy herdmates. However, the culling rate was higher for cows with follicular cysts (41 v 11 per cent).  相似文献

All stages of degeneration and regeneration in chicken tracheal epithelium were studied morphologically following an intratracheal inoculation of infectious bronchitis virus (IBV). Viral antigen was detected in the cytoplasm of tracheal epithelium from 1 to 7 days post-inoculation (d.p.i.) with a peak on 3 d.p.i. At 1 d.p.i., almost all epithelial cells were involved in the degeneration. At this time, labelling index of bromodeoxyuridine (BrdU) in the basal cells showed significantly high value compared with control. At 2 and 3 d.p.i., a great number of basal cells were recognized, but the BrdU labelling index tended to decrease. At 4 and 5 d.p.i., the BrdU labelling index of basal cells significantly decreased than 1 d.p.i., and a few number of regenerated immature ciliated epithelia appeared. At 6 to 11 d.p.i., the ciliated columnar epithelia increased rapidly in number, and returned to the normal appearance except for non-ciliated patch by 13 d.p.i. These results suggested that the tracheal epithelial cells infected with IBV degenerated within 24 hours and proliferating activity of basal cells functioned immediately, and 3 to 4 days later, these basal cells were differentiated to the ciliated epithelia.  相似文献

Skeletal muscle of 5 wild reindeer was examined for sarcocysts and used for experimental infection of 6 foxes. Skeletal and cardiac muscle of another reindeer were only examined for sarcocysts. The skeletal muscle of all animals was infected with Sarcocystis sp.. In 2 of the animals cysts of S. hardangeri were also present. The single heart examined contained only cysts of S. grueneri.Four foxes given skeletal muscle containing apparently only cysts of Sarcocystis sp., started shedding Sarcocystis sporocysts, measuring on average 13.6×9.8 µm, after a prepatent period of 10–12 days. Two foxes given skeletal muscle containing cysts of both Sarcocystis sp. and S. hardangeri shed similar sporocysts, measuring on average 13.5×9.7 µm, after a prepatent period of 10–12 days.Based on the results from the present and previous investigations, Sarcocystis sp. is considered to have foxes (Vulpes vulpes and Alopex lagopus) and dogs (Ganis familiaris) as definitive hosts, becoming the second species of Sarcocystis with a known reindeer/Canidae life cycle. The name Sarcocystis tarandivulpes n. sp. is proposed as a replacement name for Sarcocystis sp. Gjerde, 1984 from skeletal muscle of reindeer.  相似文献

Sarcocystis neurona is the parasite most commonly associated with equine protozoal myeloencephalitis (EPM). Recently, cats (Felis domesticus) have been demonstrated to be an experimental intermediate host in the life cycle of S. neurona. This study was performed to determine if cats experimentally inoculated with culture-derived S. neurona merozoites develop tissue sarcocysts infectious to opossums (Didelphis virginiana), the definitive host of S. neurona. Four cats were inoculated with S. neurona or S. neurona-like merozoites and all developed antibodies reacting to S. neurona merozoite antigens, but tissue sarcocysts were detected in only two cats. Muscle tissues from the experimentally inoculated cats with and without detectable sarcocysts were fed to laboratory-reared opossums. Sporocysts were detected in gastrointestinal (GI) scrapings of one opossum fed experimentally infected feline tissues. The study results suggest that cats can develop tissue cysts following inoculation with culture-derived Sarcocystis sp. merozoites in which the particular isolate was originally derived from a naturally infected cat with tissue sarcocysts. This is in contrast to cats which did not develop tissue cysts when inoculated with S. neurona merozoites originally derived from a horse with EPM. These results indicate present biological differences between the culture-derived merozoites of two Sarcocystis isolates, Sn-UCD 1 and Sn-Mucat 2.  相似文献

Macroscopically visible Sarcocystis spp. cysts isolated from the skeletal muscle of slaughtered cattle were examined by light- and electronmicroscopy. Transmission experiments involving cats, dogs and a human volunteer were also carried out. The cysts could only be transmitted to cats which establishes them with a high degree of certainty as Sarcocystis hirsuta. The cyst wall (including protrusions) ranged from 3.3 to 7.0 micron in thickness and the individual cyst wall protrusions from 1.2 to 2.6 micron in width. Transmission and scanning electronmicroscopy revealed previously undescribed features of the cyst wall. It appears that, with increasing age, the cyst wall protrusions become larger and develop a highly irregular surface. Their attachments to the cyst wall are slender and widely spaced indicating that growth of the cyst continues without the formation of new protrusions. Within the protrusions the fibrils become disorganised and numerous osmiophilic granules appear. It is evident that major changes in the structure of sarcocysts can occur with age.  相似文献

The confirmation of a cat-sheep transmission for visible sarcocysts was achieved in specific pathogen free (SPF) animals by completing sheep-cat-sheep-cat passages during 1977-79. The study used 22 laboratory cats and 15 specially bred sheep. Experimental sheep were dosed with sporocysts recovered from cats fed visible sarcocysts. "Giant" or "fat" form of macroscopic sarcocysts commonly found on the oesophagus were first demonstrated in the SPF sheep at 17 months after dosing. The sarcocysts from these sheep were infective to cats. Thus, the cycle of transmission for cat-borne ovine sacosporidiosis was completed. Grazing sheep in some situations develop visible cysts earlier, around one year of age, hence it is considered that the infections of experimental sheep in SPF conditions may not reflect all the circumstances leading to natural infection.  相似文献

Bovine sarcocystosis is caused by Sarcocystis cruzi and is known to cause considerable morbidity and mortality in cattle. This species is distributed worldwide in cattle and is the most prevalent of the Sarcocystis species infecting cattle. There is high infection rate of sarcocyst in cattle in Iran, but to our knowledge, there is no study about identification of Sarcocystis species. This work aimed to survey prevalence of S. cruzi cyst in slaughtered cattle of Isfahan, Iran. In this study, esophageal and diaphragmatic muscles of 100 cattle were collected from Fesaran abattoir of Isfahan and examined for the presence of Sarcocystis spp. cysts macroscopically and microscopically. No macroscopic sarcocysts were found in any of the samples. In light microscopy, 89 out of 100 cattle (89%) had thin-walled cysts of S. cruzi, while 21 out of them (21%) had thick-walled sarcocysts. In addition to light microscopy, ultrastructural features of the thin-walled cyst confirmed the presence of S. cruzi.  相似文献
