The effects of both temperature and humidity on the survival of the mites Psoroptes ovis and Psoroptes cuniculi were considered in laboratory assays. When P. ovis and P. cuniculi were maintained at 95% humidity, maximum survival decreased linearly with increasing temperature, from approximately 15 days at 9 degrees C to 5 days at 30 degrees C. There was no significant difference between P. ovis and P. cuniculi in the effects of temperature on maximum survival. Adult male P. ovis and P. cuniculi had lower mean maximum survival than any of the other life cycle stages. There was a small but significant effect of humidity on survival for P. cuniculi; LT50 values were greater at 75-85% r.h. than at 55-65% r.h. The influence of off-host survival and the infestation of naive sheep from mites in the environment on the epidemiology of sheep scab are discussed.  相似文献

Rabbits infested with Psoroptes ovis (Hering) or with P cuniculi (Delafond) were injected IM or subcutaneously with ivermectin or with vehicle. A single injection of ivermectin (200 micrograms/kg) either IM or subcutaneously was inadequate to eliminate all mites in the ears of the rabbits. However, a single injection of ivermectin at 400 micrograms/kg, regardless of the route of injection, eliminated all P cuniculi mites, but eliminated P ovis mites in only 50% of the rabbits. Ivermectin was more effective against P cuniculi than against P ovis at both doses tested, regardless of the route of injection.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed for the detection of specific antibodies against crude Psoroptes antigen. The diagnostic sensitivity was 93.7% in 191 sheep with clinical signs associated with mange. These animals originated from 29 flocks in which psoroptic mites were detected. All of 59 sheep infested with Psoroptes ovis were seropositive. Additionally, in 49% of 70 clinically unaffected sheep originating from P. ovis-infested flocks, specific antibodies could be detected, suggesting that asymptomatic infestations can be diagnosed by serology. The specificity of the ELISA was 96.5% as determined with 254 sheep originating from 44 flocks without clinical mange. Cross-reactivity in a low range was detected with selected sera of sheep with clinical chorioptic or forage mite infestations. Four sheep seroconverted 2 weeks after experimental P. ovis infestation, i.e. 2 weeks before clinical signs became obvious. After successful doramectin treatment of 14 sheep with naturally acquired P. ovis infestation, the ELISA values declined slowly but remained positive in seven cases beyond 17 weeks.  相似文献

Goats and sheep were pastured together and held in close contact in yards twice a month for 21 months. The goats had a high prevalence of both the rabbit ear canker mite, Psoroptes cuniculi and the chorioptic mange mite, Chorioptes bovis. The sheep were not infested with P. cuniculi at the beginning of the study and viable C. bovis could not be detected. At no time during the study did P. cuniculi establish in the ears of sheep, despite a maximum prevalence of >95% P. cuniculi in goats. This may have been related to the viscous nature of sheep cerumen. Levels of cerumen in goats' ears increased with increasing prevalence of the mite in the flock. The prevalence of C. bovis in the sheep flock did not exceed 10%, despite a maximum prevalence of >90% infested goats. The difficulty of detecting light infestations of mites means that it was not clear whether the C. bovis infestation of sheep was derived from goats or was present when the study began. Whatever the source, close contact with goats with an active and extensive C. bovis infestation seems unlikely to seriously influence populations of the ectoparasite in sheep. Psoroptes cuniculi was found to he established in goats as young as five days and post-mortem examination of goats' ears was more successful at revealing P. cuniculi than was use of an otoscope during life.  相似文献

Sheep scab mites, Psoroptes ovis, collected from a Merino donor sheep, were used to infest Merino and Dorper sheep, and Angora and Boer goats. Mites were placed on the sheep on 1 or 2 occasions and on 5 occasions on the goats. All the animals were examined at regular intervals for the presence of scab lesions and living mites. Both sheep breeds developed lesions, but those on the Merino sheep were always larger than those on the Dorper sheep at the same intervals after infestation. None of the goats developed lesions or showed signs of irritation, or harboured any mites.  相似文献

In sheep, lesions caused by Psoroptes ovis, the sheep scab mite, may become colonized by Staphylococcus aureus. The present study compares clinical signs, lesional area and the immune response to P. ovis and S. aureus in P. ovis-infested sheep with and without secondary S. aureus infection. No differences were detected in the clinical signs or lesional areas in the S. aureus-positive and -negative sheep. However, 6 weeks after infestation an IgG but not IgE isotype antibody response to S. aureus was detected in the S. aureus-positive but not the S. aureus-negative group of sheep. This response targeted S. aureus antigens with molecular weights of approximately 36, 38, 50 and 65 kDa. In addition, 6 weeks after infestation an IgE response to P. ovis was detected in the S. aureus-positive but not the S. aureus-negative group of sheep.  相似文献

The field efficacy of pour-on formulation of flumethrin 1 per cent was assessed in cattle naturally infested with Psoroptes ovis. In a finishing unit, 18 infested animals were selected, left untreated for one month and examined twice to monitor the progress of the infestation. They were treated twice 10 days apart with a pour-on formulation of flumethrin 1 per cent (2 mg/kg bodyweight). Skin samples were collected from the animals on days 0, 7, 14, 28 and 42 and examined for the presence of live mites. The clinical condition of the cattle was evaluated at the beginning and at the end of the trial using a standardised chart. Bodyweights were recorded on days -32, 0, 28 and 50. In addition the 65 cows in a calving unit were given the same treatment regimen and its efficacy was evaluated on days 0, 7, 14 and 28 in 11 P ovis infested animals. On the day of treatment most of the infested animals had a severe pyodermatitis. From day 7 onwards no live mites were found in the treated animals and there was a marked clinical improvement. The percentage efficacy on day 28 was 100 per cent in both units. In the finishing animals treatment was associated with a marked improvement in daily weight gains between day 0 and day 28.  相似文献

Bovine scabies in a clinical problem during the cool autumn and cold winter months. In south Texas it is necessary to stanchion animals in order to elicit patent infestation during summer months. Environmental factors and grooming have been purported to account for summer resistance to infestation. However, stanchioned animals exhibit varied levels of susceptibility to infestation, suggesting that the noted variability may be influenced by the host immune response. In this study, animals were infested with Psoroptes ovis while in stanchions. The development of anti-P. ovis antibody activity was measured by the enzyme-linked immunosorbent assay and cellular immune function was monitored with mitogens and P. ovis antigens. A correlation was noted between the development of anti-P. ovis antibody activity and the increase in mite numbers and dermatitis. Coincident with the developing infestation was a suppression of T-cell function that appeared to be stress-related as a result of stanchioning. A working hypothesis is presented that attempts to correlate the developing humoral response and the depressed T-cell response with both an increasing dermatitis and mite population.  相似文献

Lipid ingestion from sheep epidermis by Psoroptes ovis (Acari: Psoroptidae)   总被引:1,自引:0,他引:1  
Skin biopsies from three groups of sheep infested with Psoroptes ovis were cryofixed in liquid nitrogen to preserve outer epidermis with its lipid. From one group of five sheep (Group A), biopsies were taken from relatively healthy skin near the edge of scab lesions where mites had congregated. Frozen vertical sections from these biopsies were stained with Oil Red O and haematoxylin before mounting. Red lipid globules were plentiful within the body cavity of sectioned mites, but ingested lipid could not be distinguished from endogenous mite body stores by this technique. In a second group of five sheep (Group B), enclosed lumbar skin areas were coloured red with the lipophilic stains Oil Red O or Sudan IV. These enclosed coloured skin areas were inoculated with P. ovis and sampled by skin biopsy 1 or 4 days later. Cryofixed biopsies were cut into vertical frozen sections and mounted without staining for examination. Red-coloured lipid within mites, matching red-coloured lipid on outer epidermis, was evidence for the epidermal origin of P. ovis ingesta in the early stages of an infestation. From two other sheep (Group C), cryofixed biopsies were examined by scanning electron microscope and mites were seen with mouthparts embedded in abraded outer epidermis, but the precise depth of epidermal penetration was not determined. A light microscope survey of 3198 frozen vertical skin sections from the first 10 sheep (Groups A and B) showed that inner stratum corneum of the epidermis was the deepest penetration recorded for gnathosomes of P. ovis cryofixed in situ by liquid nitrogen. No structure of P. ovis was identified in dermal tissues.  相似文献

Calves infested with Psoroptes ovis (Hering, 1838) for the first time (naive) or previously infested calves were examined for serum antibody activity to P. ovis (obtained from rabbits) antigens by enzyme-linked immunosorbent assay (ELISA) to determine the temporal appearance of specific serum antibody activity. The development of the serum antibody activity to P. ovis antigens was then correlated with the development of lesions (% scab) and with changes in the number of P. ovis. The serum antibody activity [ELISA OD414 value (EODV) greater than 0.290] to P. ovis antigens in naive calves, in most cases, is first detected at the same time or slightly after the detection of mites and mite-caused lesions; and the development of specific serum antibody activity paralleled the increase in the P. ovis population and the percentage scab until 7 weeks post-infestation (PI). In previously infested calves, if serum antibody activity to P. ovis antigens was not already present from the previous infestation, specific serum antibody activity was detected at the same time and developed in a similar manner as in the naive calves. The serum antibody activity to P. ovis antigens could be detected after calves were relieved of their P. ovis burdens by pesticide treatment or after the P. ovis population began to decline when the calves were allowed to groom themselves. Serum antibody activity to P. ovis antigens was not detected in any of the control calves during the test period.  相似文献

Four trials including 11,266 sheep were conducted in South Africa to evaluate the efficacy of the systemic parasiticide ivermectin against field outbreaks of sheep scab (Psoroptes ovis) when two doses of approximately 200 micrograms/kg were administered subcutaneously seven days apart (days 0 and 7). As sheep scab is a notifiable disease in South Africa, it was not possible to include an untreated control group. The prevalence of clinically affected animals in the four treated flocks varied from 0.4 per cent to 99 per cent before the two treatments. After the treatments, there were no signs of active clinical infection in any of the sheep between days 28 and 30, or at subsequent examinations. P ovis mites were recovered from scrapings from 114 of 127 indicator sheep before the treatment but no mites were recovered from them between days 28 and 30 or 42 and 58 after the treatments.  相似文献

In the present study, the efficiency of the administration of doramectin (DOR) and DOR+vitamin AD3E (VIT), and the influence of these agents on oxidative stress parameters in rabbits infested by Psoroptes cuniculi (P. cuniculi) were investigated. DOR (200 microg/kg) and DOR (200 microg/kg)+VIT AD3E were administered to infested rabbits intramuscularly (IM). The administration of DOR and DOR+VIT improved the healing of ear lesions on day seven. Increase in the plasma malondialdehyde (MDA) level and erythrocyte catalase (CAT) activity, and decrease in glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities were determined in infested rabbits. In the rabbits which were administered DOR+VIT, plasma MDA levels decreased, and erythrocyte GSH-Px and SOD activities increased on day seven. In conclusion, DOR and DOR+VIT combination were effective against P. cunuculi infestation. Infestation stimulated oxidative stress. VIT treatment resulted in antioxidant activity against oxidative stress induced by P. cuniculi infestation.  相似文献

The pattern of population growth of the ectoparasitic mite, Psoroptes ovis (Acari: Psoroptidae), on its ovine host is considered through the development of a Leslie matrix-based, simulation model. The model is parameterised using experimental data in conjunction with reanalysis of published data. The model shows that on sheep P. ovis populations grow at a rate of approximately 11% per day and the population doubles every 6.3 days. Additional rates of adult mortality, in excess of 50% per day, need to be imposed to prevent population growth. The predictions of the model are tested by comparison of the expected numbers of mites with the numbers recorded in lesions either on naturally infested sheep where the date of infestation can be estimated or on one artificially infested animal, where the initial number of mites and date of infestation are known precisely. In both cases the observed number of mites in lesions relate closely to the numbers expected from the simulations. The model simulations do not support the concept of a 'lag' phase as distinct from the 'growth' phase in the changing pattern of mite abundance on an infested sheep and suggests that the observed pattern of growth is a natural function of an exponential increase in numbers. The development of such models and their use in explaining the demographic processes which drive mite population dynamics are discussed.  相似文献

In this paper the taxonomy and relative importance of various species within the genus Psoroptes are reviewed with respect to inter- and intra-specific variations in cross infestivity and host specificity, site of infestation and morphological characters. Intra-specific variation within populations of the sheep scab mite, Psoroptes ovis will then be discussed critically with respect to virulence (the relative growth of the scab lesion and mite burdens), epidemiology, susceptibility to the systemic acaricide, ivermectin, and resistance to synthetic pyrethroid and organophosphate acaricides. Finally, it is suggested that the ear mite, P. cuniculi, and the sheep scab mite, P. ovis are variants of the same species. Information was collected from published literature and from the results of unpublished studies carried out at the VLA, Weybridge over the last 10 years.  相似文献

OBJECTIVE: To evaluate the efficacy and safety of topical administration of selamectin in rabbits naturally infested with Psoroptes cuniculi. DESIGN: Randomized controlled trial. ANIMALS: 48 mixed-breed domestic rabbits with active P. cuniculi mite populations and clinical ear lesions. PROCEDURES: Rabbits were randomly allocated to 1 of 6 treatment groups. On day 0, rabbits in groups 1 and 2 were given vehicle, rabbits in groups 3 and 4 were given selamectin at a dose of 6 mg/kg (2.7 mg/lb), and rabbits in groups 5 and 6 were given selamectin at a dose of 18 mg/kg (8.2 mg/lb). On day 28, rabbits in groups 2, 4, and 6 were given a second dose of vehicle or selamectin. Otoscopic examinations were performed and ear lesion size was measured weekly for 8 weeks. Quantitative viable mite counts were performed on day 56. RESULTS: On days 7 through 56, lesion sizes for all selamectin-treated groups were significantly lower than sizes for control groups; there were no significant differences in lesion sizes among selamectin-treated groups. All rabbits in the 2 control groups had viable adult P. cuniculi mites for the duration of the study, as determined by otoscopic examination, whereas all rabbits in the 4 selamectin-treated groups were free from P. cuniculi mites on days 7 through 56. No adverse reactions associated with selamectin treatment were observed. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that topical application of selamectin at a dose of 6 or 18 mg/kg can completely eliminate mites from rabbits naturally infested with P. cuniculi.  相似文献

Influence of Psoroptes ovis on the energy metabolism of heifer calves   总被引:2,自引:0,他引:2  
To determine the effect of Psoroptes ovis on the energy metabolism of heifers, 32 calves were randomly assigned to four treatments in a 2 X 2 factorial arrangement. Calves were fed the same diet at two intake levels, high or low, and were either infested or were not infested with P. ovis mites. Calves were housed in antigrooming stanchions. Body composition was determined by urea dilution on Days 0 and 63. Ration digestibility was determined on fecal grab samples using acid-insoluble ash as a marker. Infested calves had developed a severe P. ovis infestation 7 weeks following exposure and had significantly lower daily gain, gain:feed, and energy retention and higher serum glutamic-oxaloacetic transaminase than control calves. P. ovis infestation increased the maintenance energy requirement of calves by greater than 50% (79 vs. 123 kcal of net energy kg-1 body wt.75). For each 10% increase in the body surface affected by P. ovis, maintenance energy requirement increased 0.5 mcal day-1.  相似文献

The influence of subcutaneously administered ivermectin on the specific immune response was studied in rabbits infested with mites (Psoroptes cuniculi) and in rats infested with lice (Polyplax spinulosa). A pronounced specific antibody activity and a change in immunoblotting pattern was observed in rabbits after the ivermectin treatment. However, in rats the antibody activity decreased and the profile of specific antibodies, tested by immunoblotting, remained the same as before the treatment. The specific immune response in rabbits artificially immunized with whole-body Psoroptes cuniculi extract was not affected by ivermectin. It was concluded that ivermectin has no direct effect on the immune response of rabbits and rats and that the enhanced immune response in the mite-infested rabbits was caused by the massive release of antigens associated with the synchronous death of the mites.  相似文献

We have previously shown that infestation with Psoroptes ovis induces an IgE response and intense tissue eosinophilia, typical of a Type I hypersensitivity response [Parasite Immunol. 22 (2000) 407]. Intradermal tests (IDSTs) suggest that there are also delayed and Arthus-type responses to this parasite. In order to study the nature of ovine cutaneous reactions to P. ovis, na?ve controls and experimentally infested sheep (n = 5) were challenged intradermally with mite antigen. Challenge elicited immediate (P < 0.001) and delayed (P < 0.005) wheal reactions in sensitised sheep. At 6 (P < 0.02) and 30 h (P < 0.001) the predominant infiltrating cells were eosinophils. To explore the role of circulating antibodies, na?ve sheep (n = 5) were subjected to Prausnitz-Kustner (PK) tests. These elicited immediate (P < 0.02) but not delayed wheal reactions. At 6 h eosinophils (P < 0.001) dominated the infiltrate. These results suggest that P. ovis allergens provoke an IgE-dependent immediate and late phase response and a cell-mediated eosinophil-rich delayed-type hypersensitivity response (ER-DTH).  相似文献

Ivermectin at 200 micrograms/kg bodyweight given either as a single subcutaneous injection or as an oral drench failed to eradicate Psoroptes ovis from artificially infested sheep. The oral drench reduced the mite populations by 43 per cent within 24 hours but no further significant decline was recorded over 38 days. The subcutaneous injection reduced the mite populations by 90 per cent after 10 days but live P ovis were present on all the treated animals 84 days after treatment. The efficacy of treatment was less the higher the initial mite burden. The injection had no effect on clinical sheep scab, and the disease continued to progress despite the mite mortality.  相似文献

The response of sheep scab mites to pyrethroid insecticides and organophosphate compounds was studied in vitro with the objective of finding a simple test for detecting insecticide resistance in scab mites. Psoroptes cuniculi from rabbits or P. ovis from sheep were enclosed in small 'tea bags' made from heat sealable paper prior to dipping in insecticide. Mites failed to die 24 h after a 1 min dip in working concentrations of insecticidal sheep dips. With flumethrin a variety of different conditions were tested but most failed to improve the efficacy of flumethrin. It is suggested with freshly collected mites pyrethroids may be taken up by ingestion.  相似文献
