用有机溶剂萃取法从三种木莲叶片中分离获得的提取物,经GC/MS进行成分分析和定量测定.再进行抗氧自由基活性测定.从红花木莲(Manglietia insignis (Wall) Blume)、桂南木莲(Manglietia chingii Dandy)和乳源木莲(Manglietia yuyuanensis Law)中分别鉴定出21、36和20种化合物,其中11种成分为三种木莲所共有,12种成分为两种木莲所共有.各成分的相对含量为红花木莲:萜烯类化物38.93%,烷烃类28.18%,含氮化合物15.73%,芳香族化合物7.23%.桂南木莲:萜烯类30.22%,酸类14.17%,烷烃类13.87%,芳香族化合物13.29%;乳源木莲:醇类28.00%,萜烯类25.38%,芳香族化合物18.00%. 研究结果还表明,三种木莲的提取物均有较强的抑制氧自由基的功能.在稀释100倍时,抑制超氧阴离子活力最强,稀释20倍时,抑制羟自由基活力达最大.本研究结果为进一步探索三种木莲的保健功能提供科学依据.图1表5参15.  相似文献

Four new compounds, maninsigins A–D (14), including two new neolignans (12) and two new sesquiterpenes (34), as well as ten known compounds (514), were isolated from the leaves and stems of Manglietia insignis. Their structures were established on the basis of extensive spectroscopic analyses. In addition, some compounds were tested for their cytotoxic and neurite outgrowth-promoting activities, as well as their antagonistic activity toward FXR ligand.  相似文献

Twenty plant materials collected from the islands of Java and Kalimantan in Indonesia were extracted with 50% aqueous ethanol (crude extract). The crude extracts were assayed for antimicrobial activities against Streptococcus sobrinus and for glucosyltransferase (GTase) inhibition. Fourteen extracts inhibited the growth of S. sobrinus by more than 50% and six extracts inhibited GTase activity by more than 50% at a concentration of 100 μg/ml. Koompassia malaccensis (kempas) extracts showed 90% depression of S. sobrinus growth and 80% inhibition of GTase activity at a concentration of 100 μg/ml. Kempas crude extracts were subjected to column chromatography using Sephadex LH-20 and then preparative high-performance liquid chromatography to isolate four compounds A, B, C, and D. These compounds were identified as taxifolin and the flavanonol rhamnoside isomers neoastilbin, astilbin, and isoastilbin, respectively, from 1H and 13C nuclear magnetic resonance (NMR) spectra and other two-dimensional NMR techniques (COSY, HMBC, and HMQC). Each compound depressed the growth of S. sobrinus over a concentration range of 9.3242.7 μg/ml and showed GTase inhibitory activity with IC50 values in the range 27.4–57.3 μg/ml. Taxifolin and flavanonol rhamnoside isomers isolated for the first time from kempas could be potent compounds for preventing dental caries. Metarhizium anisopliae sensu stricto (ss) (Metsch.) Sorok. isolate ICIPE 07 is being developed as biopesticide for the control of ticks. In addition, leaf extracts of Calpurnia aurea Benth, and the attraction aggregation and attachment pheromone (AAAP) are being used as ticks’ attractant. The three agents are being considered for use in combination in an autodissemination approach, whereby ticks that are attracted to semiochemicals are infected with the inoculum. Experiments were therefore conducted to evaluate in vitro the compatibility between C. aurea, AAAP, and the M. anisopliae on vegetative growth, conidial production, and spore viability. Calpurnia aurea leaf extract was compatible with the fungus at all the concentrations tested, whereas AAAP inhibited all the fungal growth parameters. The virulence of M. anisopliae formulated in emulsifiable extracts of C. aurea was also tested against different developmental stages of Rhipicephalus appendiculatus in laboratory bioassays. No significant differences in virulence were observed between M. anisopliae applied alone and M. anisopliae formulated in different concentrations of C. aurea leaf extracts. These results suggest that C. aurea leaf extracts is compatible with M. anisopliae and could be mixed together for “spot-spray” treatments as low-cost and environmental-friendly technology to control ticks in grazing field, while AAAP should be used separately.  相似文献

Heartwood extracts from Amazonian trees cumaru-ferro (Dipteryx odorata), jatoba (Hymenaea courbaril), and guarita (Astronium lecointei) exhibit antioxidant activities comparable with that of α-tocopherol, a well-known antioxidant. This article reports the characterization of the antioxidant compounds in the extracts of the three heartwoods. Silica gel column chromatography of the cumaru-ferro EtOAc extract yielded (−)-(3R)-7,2′,3′-trihydroxy-4′-methoxyisoflavan and (+)-(3R)-8,2′,3′-trihydroxy-7,4′-dimethoxyisoflavan. Silica gel column chromatography followed by preparative high-performance liquid chromatography of the jatoba EtOAc extract yielded (−)-fisetinidol and (+)-trans-taxifolin. Chemical structures were assigned using electron-ionization mass spectrometry, 1H and 13C nuclear magnetic resonance (NMR) spectroscopy including nuclear Overhauser effect spectroscopy (NOESY), as well as optical rotation and circular dichroism. Gas chromatography-mass spectrometry demonstrated that the isolated compounds were predominant in the EtOAc extracts. In the guarita EtOAc extract, catechin and gallic acid were identified by comparing their retention times and mass fragmentation patterns with those of authentic samples. Antioxidant activity determined by the 1,1-diphenyl-2-picrylhydrazyl assay demonstrated that all these compounds had activities comparable with that of α-tocopherol. Hot water extract (HWE) from Pinus radiata bark was fractionated into monomeric polyphenol (MPP), oligomeric proanthocyanidin (OPA), and polymeric proanthocyanidin (PPA) fractions by monitoring the UV-visible spectrum of the eluted fractions. Nine polyphenols were identified in MPP: three phenolic acids and six flavonoids. The major compounds of MPP were taxifolin (456 μg/mg), catechins (240 μg/mg), and protocatechuic acid (46.2 μg/mg). The OPA components ranged from dimers to hexamers. PPA comprised procyanidin (PC, 94%), a small amount of prodelphinidin (PD, 6%), and also the higher trans configuration, rather than the cis configuration, of C2-C3. The average molecular weight (Mw, Mn), polydispersity (Mw/Mn), and DP of PPA were 3,800 (Mw), 1,200 (Mn), 3.2 and 13, respectively. The potential antioxidant activities of HWE and the three fractions were estimated using proton- or electron-donating assays containing 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) (ABTS) radical cation, superoxide anion (O2•−), hydrogen peroxide (H2O2), reducing power and lipid peroxidation. All the bark fractions exhibited potent quenching abilities against both commercially available radicals (DPPH•, ABTS•+) and chemically induced radicals (H2O2, O2•−), as well as strong reducing power and inhibitory activity of lipid peroxidation. Particularly, OPA showed a hydrogen peroxide scavenging activity significantly higher (p < 0.05) than that of the other fractions. From 2-thiobarbituric acid reactive substances and ferric thiocyanate assays, PPA was suspected to act by essentially delaying the formation of hydroperoxide by quenching free oxygen radicals that operates in oil-in-water emulsion.  相似文献

Extractives can affect the vibrational properties tanδ (damping coefficient) and E′/ρ (specific Young’s modulus), but this is highly dependent on species, compounds, and cellular locations. This paper investigates such effects for African Padauk (Pterocarpus soyauxii Taub.), a tropical hardwood with high extractives content and a preferred material for xylophones. Five groups of 26 heartwood specimens with large, yet comparable, ranges in vibrational properties were extracted in different solvents. Changes in vibrational properties were set against yields of extracts and evaluation of their cellular location. Methanol (ME) reached most of the compounds (13%), located about half in lumen and half in cell-wall. Water solubility was extremely low. tanδ and E′/ρ were very strongly related (R 2 ≥ 0.93), but native wood had abnormally low values of tanδ, while extraction shifted this relation towards higher tanδ values. ME extracted heartwood became in agreement with the average of many species, and close to sapwood. Extractions increased tanδ as much as 60%, irrespective of minute moisture changes or initial properties. Apparent E′/ρ was barely changed (+2% to −4%) but, after correcting the mass contribution of extracts, it was in fact slightly reduced (down to −10% for high E′/ρ), and increasingly so for specimens with low initial values of E′/ρ.  相似文献

Two diarylheptanoids, 1,7-bis-(3,4-dihydroxyphenyl)-heptane-3-one-5-O-β-d-xylopyranoside (I) and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-O-β-d-xylopyranoside (II), were isolated from the bark of Alnus hirsuta. Compounds I and II exhibited strong antioxidative activity against 1,1-diphenyl-2-picrylhydrazyl radicals, with IC50 values of 8.36 ± 0.54 and 8.67 ± 1.46 μM, respectively. In addition, we demonstrated that compounds I and II inhibited the production of nitric oxide and reactive oxygen species and the expression of proinflammatory molecules such as inducible nitric oxide synthase and cyclooxygenase-2 in lipopolysaccharide-induced macrophages. According to our results, the two diarylheptanoids isolated from the bark of A. hirsuta exhibited significant antioxidative activity and anti-inflammatory effects and may be useful in the pharmaceutical industry for alleviating oxidative stress.  相似文献

The components of the essential oils from seeds of Azadirachta indica, Azadirachta siamensis, and Azadi-rachta excelsa were studied by gas chromatography and gas chromatography-mass spectrometry. The main components of A. indica oil were hexadecanoic acid (34.0%), oleic acid (15.7%), 5,6-dihydro-2,4,6-triethyl-(4H)-1,3,5-dithiazine (11.7%), methyl oleate (3.8%), and eudesm-7(11)-en-4-ol (2.7%). The major components of A. siamensis oil were hexadecanoic acid (52.2%), tricosane (10.5%), tetradecanoic acid (6.8%), oleic acid (4.9%), and pentacosane (4.9%). Azadirachta excelsa oil contained oleic acid (31.3%), hexadecanoic acid (14.2%), octadecanoic acid (13.0%), 4-octylphenol (9.7%), and O-methyloximedecanal (6.8%) as the main constituents. The essential oils from A. indica, A. siamensis, and A. excelsa were found to contain fatty acids (52.6%–72.3%) as major components. The minor components of the oils were n-alkanes, aromatics, esters, sulfur and nitrogen compounds, and terpenoids. Differences in oil composition were observed between the three species.  相似文献

[目的]研究桂西南岩溶区不同恢复模式群落的生物量及其林下植物多样性的特点,为该区域的生物多样性保护、生态功能恢复效果评价积累基础数据。[方法]以桂西南岩溶区4种不同恢复模式为研究对象,采用样方法对林下植物多样性进行研究;采用收获法研究灌木层与草本层的地上、地下生物量及凋落物层现存量;采用异速生长模型来估算乔木层的地上生物量,并参考IPCC根茎比来量化乔木的地下生物量。[结果]表明:共调查记录林下植物85种,隶属于46科,81属,其中,灌木植物25科,46属,50种;草本植物21科,35属,35种;不同恢复模式群落生物量的变化趋势为自然恢复林(166.66 t·hm-2)任豆林(48.61 t·hm-2)吊丝竹林(36.54 t·hm-2)灌草坡(0.96 t·hm-2)。[结论]不同恢复模式灌木层物种丰富度差异不显著,最高的为灌草坡(16种),其次为任豆林(15种),最低为自然恢复林(12种);草本层物种丰富度最高的为任豆林(12种),其次为灌草坡(10种),最低为自然恢复林(4种)。自然恢复林乔木不同组分生物量与任豆林、吊丝竹林之间差异显著(P0.05);灌草坡的灌木、草本地上生物量与吊丝竹林、任豆林、自然恢复林之间差异显著(P0.05)。吊丝竹林、任豆林的灌草生物量表现为草本层灌木层,而灌草坡与自然恢复林的灌草生物量则表现为灌木层草本层。  相似文献

In screening experiments for antiacne activity, methanolic and 50% ethanolic extracts of Caesalpinia sappan wood showed the most potent activity out of 28 species of plants extracts. These extracts showed inhibition of Propionibacterium acnes growth, lipase inhibitory activity, and antioxidant activity. In order to isolate the active compound from C. sappan, separation of the extract components was performed by column chromatography and preparative high-performance liquid chromatography (HPLC). Brazilin, protosappanin A, and sappanone B were isolated from methanolic extracts. Brazilin showed better antibacterial activity [minimum inhibitory concentration (MIC) = minimum bactericidal concentration (MBC) = 0.50 mg/ml] than protosappanin A (MIC = MBC = 1.00 mg/ml) and sappanone B (MIC = MBC > 2.00 mg/ml). The 50% inhibitory concentration (IC50) for lipase inhibition was lowest for brazilin (6 μM), which showed strong inhibition compared with protosappanin A (100 μM) and chloramphenicol (677 μM, positive control). The antioxidant activity of brazilin (IC50 8.8 μM) was not significantly different from protosappanin A (9.1 μM) and (+)-catechin (10.2 μM). The antioxidant activity of brazilin and protosappanin A were higher than sappanone B (IC50 14.5 μM). Brazilin is considered to have suffi ciently potent activity for use as an antiacne agent.  相似文献

Hiba (Thujopsis dolabrata) wood was treated with supercritical carbon dioxide (scCO2) at stepwise temperature increments from 50° to 400°C continuously so that extractives (dichloromethane-soluble and -insoluble phases) and solid residues were obtained. The yield of extractives from hiba wood increased with increasing extraction temperature. The volatile compounds in the dichloromethane-soluble phase from scCO2 extraction at 50°C contained only terpenoids. However, the volatile compounds in the dichloromethane-soluble phase from scCO2 extraction at 300°C not only contained terpenoids but also phenols, furans, hydrocarbons, and organic acids. The yield of β-thujaplicin, which is a useful compound in hiba wood, increased with increasing extraction temperature from 50°C to 300°C; the optimal conditions for extracting β-thujaplicin were 300°C and 19.61 MPa. Further study of degradated compounds from the cellulosic and lignic materials of hiba wood after stepwise high-temperature scCO2 treatment above 300°C may provide clues to its efficient use.  相似文献

Red flour beetle Tribolium castaneum (Herbst) and rice weevil Sitophilus oryzae (L.) are considered to be the major insect pests in storage. Essential oils from aromatic plants are recognized as proper alternatives to fumigants. Thymus persicus (Ronniger ex Rech. f.) is one of these plants that have medicinal properties and is indigenous to Iran. The essential oil was obtained from aerial parts of the plant and analyzed by GC and GC–MS. Carvacrol (44.69%) and thymol (11.05%) were the major constituents of the oil extracted. In this experiment, fumigant toxicity of the essential oil was studied against T. castaneum, S. oryzae at 27 ± 1°C and 60 ± 5% RH in dark condition. The adult insects were exposed to the concentrations of 51.9, 111.1, 207.4 and 370.4 μl/l air to estimate median lethal time (LT50) values. The fumigant toxicity was increased in response to increased essential oil concentrations. The LT50 values at the lowest and the highest concentrations tested were ranged from 28.09 to 13.47 h for T. castaneum, and 3.86 to 2.30 h for S. oryzae. It was found that S. oryzae adults were much more susceptible to the oil than T. castaneum. After 24 h of exposure, the LC50 values (95% fiducial limit) for T. castaneum and S. oryzae were estimated to be 236.9 (186.27–292.81) and 3.34 (2.62–4.28) μl/l air, respectively. These results suggest that T. persicus essential oil merits further study as potential fumigant for the management of these stored-product insects.  相似文献

A study was conducted to explore the mechanism that emissions of volatile organic compounds(VOC) from heated needles and twigs(200°C,within 15 min) of Pinus pumila affect fire behaviours using the technology of Thermal Desorption-Gas Chromatography-Mass Spectrometry(TD-GC-MS).The results indicated that the main components of VOC from heated needles and twigs are terpenoids.Most of these terpenoids are monoterpenes.Terpenoids account for 72.93% for the needles and 92.40% for the twigs of the total VOC,and their emission ratios are 61.200 μg·g-1 and 217.060 μg·g-1 respectively.Heated twigs can emit more terpenoids than heated needles because twigs had more volatile oils than needles.In actual fires,these large amounts of terpenoid emissions,especially the monoterpene emissions,have strong effects on fire behaviors that are not only in the initial stage but also in the fast propagation stage of fires.These flammable gases are capable of causing violent combustion and creating crown fires.In addition,if these gases accumulate in an uneven geographical area,there will be a possible for eruptive fires and/or fires flashover to occur.  相似文献

Sawdusts of sugi (Cryptomeria japonica) wood prepared before and after steam drying at 120°C in a kiln were extracted with n-hexane and ethyl acetate to give n-hexane extracts and ethyl acetate extracts. From gas chromatography-mass spectrometry analysis of the ethyl acetate extracts from woods before and after steam drying, the components of 4-epi-cubebol, cubebol, and 2,7(14),10-bis-abolatrien-1-ol-4-one, which existed in the raw sugi wood, were proved to disappear in the steam-dried wood. These components were also absent in the ethyl acetate extract of the steam-condensed solution of waste steam from the kiln outlet. When these three components were treated with 0.2% (v/v) acetic acid solution at 120°C, δ-cadinene was produced as a major product from both 4-epi-cubebol and cubebol by dehydration and cleavage of the cyclopropane ring, and cryptomerone from 2,7(14),10-bisabolatrien-1-ol-4-one by hydration. The chemical changes of the three components presumably occur during steam drying of the sugi wood. Superoxide dismutase‐like activity of crude Eucalyptus globulus extracts obtained from new wound wood was measured using the water‐soluble tetrazolium salt (WST)‐1 assay. This in vitro assay determined the activity of superoxide radicals scavenging in a xanthine oxidase system. The 50% inhibition (IC50) of formazan formation was estimated for fractionated samples and purified compounds isolated from the crude wound wood extract, i.e. engelitin and pedunculagin. Standards of gallic acid and pentagalloyl glucose were also measured. Fraction II as well as the compounds pentagalloyl glucose and pedunculagin showed the highest levels of antioxidant activity. Subsequent analysis of fraction II by high‐performance liquid chromatography (HPLC) coupled with negative ion electrospray mass spectrometry revealed the fraction to be dominated by hydrolysable tannins including pedunculagin, di‐, tri‐ and tetragalloylglucoses. These preliminary results provide some evidence for an antioxidant role of secondary metabolites in tree wound repair and defence.  相似文献

Calocedrus formosana Florin (Cupressaceae) is an endemic tree to Taiwan. To evaluate the antioxidant activity of the plant extracts from heartwood, bark, and leaf of C. formosana, assays for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide anion scavenging activities, as well as prevention of DNA strand cleavage were performed in this study. Similar IC50 values against the DPPH radical were found for the heartwood and bark extracts at approximately 23µg/ml. Moreover, the heartwood extract exhibited the highest inhibitory activity against superoxide radicals among the test samples; a 2.3-fold lower value of IC50 for superoxide radical inhibition was found in the heartwood extract relative to that of (+)-catechin. Much less effect on inhibition of DPPH and superoxide radicals was found from the leaf extract of C. formosana. More than 70% of superoxide radicals were inhibited in the presence of 10µg/ml heartwood extract, whereas only 15% inhibition was obtained from the leaf extract. The heartwood extract, at a dose of approximately 0.5mg/ml, apparently completely prevented the X174 supercoiled DNA cleavage induced by ultraviolet photolysis of H2O2, as judged by agarose gel electrophoresis. This report also suggests that the antioxidant activities of the plant extracts of C. formosana are in good correlation with their phenolic contents.  相似文献

The purpose of this study was to investigate the sorption of selected volatile substances from oak wood-chip samples (Quercus pyrenaica Willd. and Quercus petraea L.) subjected to different toasting levels, namely, without toasting, with medium toasting, and with strong toasting, through the use of solid-phase microextraction (SPME). The main volatile compounds identified as a function of the toasting level and botanical species were furfural, hexanal, α-pinene, d-limonene, decanal, vitispirane, ethyl hexanoate, cis-3-methyl-γ-octalactone (“oak lactone” or “whisky lactone”), α-terpineol, p-xylene, and nonanal. Considering the data obtained from the toasted woods (medium and strong intensity) in comparison with those of nontoasted woods, it can be pointed out that the average peak area and the number of compounds identified in the gas chromatogram decreased during the toasting process. In general, regarding the compounds analyzed, quantitative differences were found between the two oak wood species under study. High values of volatile compounds were found in Quercus pyrenaica oak wood chips. In addition, for the number of compounds identified in oak wood extracts and directly extracted from solid oak wood chips by SPME, it is concluded that the best extraction process for volatile compounds from oak wood is the use of oak wood-chip liquid extracts.  相似文献

 The reaction behavior and kinetics of lignin model compounds were studied in supercritical methanol with a batch-type supercritical biomass conversion system. Guaiacol, veratrole, 2,6-dimethoxyphenol, and 1,2,3-trimethoxybenzene were used as model compounds for aromatic rings in lignin. In addition, 5-5, β-1, β-O-4, and α-O-4 types of dimeric lignin model compounds were used as representatives of linkages in lignin. As a result, aromatic rings and 5-5 (biphenyl)-type structures were stable in supercritical methanol, and the β-1 linkage was not cleaved in the β-1-type structure but converted rapidly to stilbene. On the other hand, β-ether and α-ether linkages of β-O-4 and α-O-4 lignin model compounds were cleaved rapidly, and these compounds decomposed to some monomeric compounds. Phenolic compounds were found to be more reactive than nonphenolic compounds. These results indicate that cleavages of ether linkages mainly contribute to the depolymerization of lignin, whereas condensed linkages such as the 5-5 and β-1 types are not cleaved in supercritical methanol. Therefore, it is suggested that the supercritical methanol treatment effectively depolymerizes lignin into the lower-molecular-weight products as a methanol-soluble portion mainly by cleavage of the β-ether structure, which is the dominant linkage in lignin.
