Two types of triploid hard clams Mercenaria mercenaria were produced by inhibiting polar body I (PB1) or polar body II (PB2) with cytochalasin B. Treatments were applied at 22–23°C, with PB1 inhibition starting at 4–7 min postfertilization and ending when PB2 was first observed in control groups, and with PB2 inhibition starting at 17–23 min postfertilization and ending when 80% of control eggs released PB2. Triploid induction success was evaluated by chromosome counting in 2–4 cell embryos and by flow cytometry at larval and juvenile stages. PB2 inhibition produced more triploids (82%–100%) than PB1 inhibition (71%–83%), although the difference was not significant ( .088). Triploid percentages in PB1‐ or PB2‐inhibited groups showed a small but insignificant decline during the first 6 months. At month 3, PB1 and PB2 triploids were not different from their within‐group diploids, but significantly larger than control diploids; PB1 triploids were significantly larger than PB2 triploids ( .003). At month 6, PB1 triploids were not different from either within‐group or control‐group diploids, while PB2 triploids were significant larger than both within‐group and control diploid; PB1 triploids were smaller than PB2 triploids. At month 16, PB1 and PB2 triploids in one remaining replicate were not different from their within‐group diploids. Overall, this study shows that triploids can be efficiently produced by PB1 or PB2 inhibition, and their growth performance relative to diploids is variable depending on age and replicates or parental genotype.  相似文献

Live hatchery feeds were assayed for fatty acids (FA), amino acids (AA), and their ability to support growth and survival of larval and postlarval mahimahi Coryphaena hippurus at two different hatchery stages. Euterpina acuritrons copepods (C), mahimahi yolk-sac larvae (YSL), Artemia parthenogenica brine shrimp nauplii (BSn), A. parthenogenica juveniles (BSj), and Brachionus plicatilis rotifers (R) were assayed, using several enrichment media. There was little difference in AAs among feeds.
Levels of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were about 10 times higher in YSL than in their feeds. This explains previous findings where first stage larval survival (0–9 days) was not affected by feed HUFA levels. Second stage survival was significantly higher when larvae were fed copepods. Enrichment with 100 ppm SuperSelco greatly improved the survival of larvae that were fed brine shrimp. The even higher omega-3 fatty acids found in copepods appear to be important for survival of larvae under more stressful conditions.
Brine shrimp juveniles enriched with SuperSelco are a good food for postlarval mahimahi. Yolksac larvae of mahimahi are an even better food, promoting faster growth at less cost, for large scale mahimahi aquaculturists. Different batches of yolksac mahimahi larvae varied by a factor of 10 in their concentration of DHA, but always had the highest level of DHA as much as 40% of total fatty acids (FAs). These "high HUFA" batches of YSL also had the highest levels of EPA and total fatty acids. The data suggest that climate and broodstock age may have considerable influence on larval nutrition.  相似文献

Larval stages of the Pacific white shrimp, Litopenaeus vannamei (Boone) were fed standard live diets of mixed microalgae from the first to the third protozoea (PZ1 to PZ3), followed by Artemia nauplii until post‐larvae 1 (PL1). Trypsin enzyme activity for each larval stage was determined using N‐α‐p‐toluenesulphonyl‐l ‐arginine methyl ester (TAME) as a substrate. Results were expressed as enzyme content to assess ontogenetic changes during larval development. Tissue trypsin content (IU µg?1 DW for each larval stage) was significantly highest at the PZ1 stage and declined through subsequent stages to PL1. This contrasts with previously observed patterns of trypsin development in Litopenaeus setiferus (Linnaeus) and other penaeid genera, which exhibit a peak in trypsin activity at the third protozoea/first mysis (PZ3/M1) larval stage. Litopenaeus vannamei larvae transferred to a diet of Artemia at the beginning of the second protozoea (PZ2) stage were significantly heavier on reaching the first mysis stage (M1) than those fed algae, while survival was not significantly different between treatments. At both PZ2 and PZ3 stages, trypsin content in larvae feeding on Artemia was significantly lower than in those feeding on algae. The rapid decline in trypsin content from PZ1 and the flexible enzyme response from PZ2 suggest that L. vannamei is physiologically adapted to transfer to a more carnivorous diet during the mid‐protozoeal stages.  相似文献

Wild caught Asian catfish were spawned manually following HCG injection, and a portion of the eggs were subjected to cold-shock at 4 C for 15 min within 2-min post-fertilization. Nuclear diameter measurements of cold-shocked fish revealed that 96% were triploids (3N), while non-shocked fish were all diploids (2N). During larval and fry culture (first 26 d), triploid fish mortality was =50%, while diploid mortality was =25%. Following 8-mo culture in tanks at three stocking densities, triploid fish survival was significantly greater ( P < 0.05), than diploids, with 84.0% and 57.3%, respectively. Triploid live weight was also significantly greater than diploids, with 69.2 and 45.9 g averages, respectively. Ninety-two percent of diploids had welldeveloped gonads after 8 mo; whereas none of the triploids had mature gonads. Gonads were undifferentiated with 31% of the triploids. These sexually undifferentiated fish had greater growth rates than male or female triploids, and greater growth than all diploids. Carcass weight (gutted) of triploids was 95.8% of live weight, compared with 92.5% for diploids. Lastly, triploids had very few deformities compared with diploids, with 1.3% and 17.6%, respectively. Deformities included curved spines, and humped backs just posterior of the head.  相似文献

为了评估熊本牡蛎(Crassostreasikamea)中国群体与美国群体的杂交效应与杂交三倍体优势,构建了杂交群体和自交群体,并使用细胞松弛素B诱导了杂交三倍体,比较分析了幼虫、稚贝与成贝的生长与存活优势。自交群体和杂交群体的幼虫、稚贝与成贝的培养条件均相同,室内培育幼虫,培育密度为4~5个/mL,自然海区养殖稚贝与成贝,养殖密度为40~45个/吊。结果表明,与自交组相比,杂交二倍体具有较高的卵裂率(13.61%)与D幼率(5.67%)(P0.05),但杂交二倍体幼虫的壳高生长表现杂种劣势(–0.43%),而稚贝、成贝的壳长与壳高的生长表现杂种优势,平均优势率分别为3.96%与6.65%。杂交三倍体诱导组幼虫的壳高生长优势率为3.69%,稚贝及成贝的壳长与壳高平均生长优势率分别为12.69%与13.64%,并且日龄越大生长优势越显著。总体上,杂交三倍体诱导组在3~180日龄具有存活劣势,并且15日龄存活劣势率最大(–48.72%),而在360日龄存活优势率为6.70%。杂交二倍体幼虫与成贝均具有存活优势,平均优势率分别为10.44%与4.59%。本研究表明熊本牡蛎中美地理群体杂交二倍体的生长和存活优于自交二倍体,而杂交三倍体诱导组的生长优势较显著,并且在成贝期具有显著的成活率优势,表明杂交三倍体诱导组的优势来源于三倍体优势和部分杂种优势。  相似文献

Triploidy may provide a means to neuter the Pacific oyster, C. gigas, genetically thereby increasing survival and marketability during periods of reproduction. Pressure treatments administered 10 min after fertilization for 10 min duration at 6000–8000 psi consistently produced triploids. The highest proportion obtained was 57%. Triploidy was assayed in all experiments at the larval stage and again as spat (metamorphosed larvae) using flow cytometry. A technique to assay larvae as small as 250 μm is described. Results obtained by flow cytometry were verified by chromosome counts. Analysis of triploidy at the larval stage provides reliable estimates of the proportion of triploids, eliminating several weeks of culture time.  相似文献

The effect of ploidy on the mortality of Crassostrea gigas spat caused by the ostreid herpesvirus (OsHV‐1) genotype μVar was investigated at five sites along the Atlantic coast in France in 2011. Sibling diploids and triploids were produced using either unselected or selected OsHV‐1‐resistant oysters. No significant interactions were found between the factors of environment, genotype and ploidy at the endpoint dates. The mean mortality rates at the sites were 62% and 59% for diploids and triploids, respectively, and the two rates were not significantly different. The mean mortality rates were 33% and 32% for sibling diploids and triploids, respectively, when OsHV‐1‐resistant parents were used and 91% and 85%, respectively, when unselected parents were used. The results were confirmed through other broodstocks tested in 2013. Our study is the first to clearly show that mortality related to OsHV‐1 is similar between diploids and triploids in C. gigas when the same germplasm is used for both ploidy. Furthermore, OsHV‐1 resistance was not substantially altered by triploidization, indicating that the achieved selective breeding of diploid oysters for OsHV‐1 resistance can be translated into improved survival in triploids.  相似文献

This investigation examined the effects of temperature, density and early weaning on the survival and growth of Palaemonetes varians larvae. Survival of larvae raised at 17.5 °C was not significantly different (average + standard deviation) (94 ± 5%) from the survival of those raised at 19.5 °C (95 ± 5%) and at 21.5 °C (94 ± 4%). However, the duration of the larval stage was significantly longer for shrimp reared at 17.5 °C (17.3 ± 0.8 days) compared with shrimp reared at 19.5 °C (14.3 ± 0.7 days) and at 21.5 °C (11.3 ± 0.6 days). No significant differences ( P >0.05) were found in the survival rate, final weight and length of larvae reared at the densities of 5, 10, 20 and 50 larvae L−1. The survival of P. varians larvae fed solely on Artemia was significantly higher ( P <0.05) than larvae weaned with an artificial practical diet from Zoea II stage (94 ± 4% and 82 ± 1%, respectively, for Artemia and artificial diet-fed larvae), but no significant differences ( P >0.05) were observed in the final larval weight or length between these two treatments. The survival and growth of the larvae fed with the practical diet tested is a promising step ahead in the development of the culture of this species as it eliminates both the need for Artemia throughout all larval stages, and the need for more expensive artificial diets.  相似文献

Three‐summers‐old all‐female triploid and diploid rainbow trout were compared after one on‐growing season in sea net cages. Slaughter traits of round weight, gutted weight, fillet weight, carcass% and fillet% were measured at three times in November 2017, January and April 2018. The triploid group had lower daily growth coefficient mean (4.25) and higher feed conversion ratio (1.18) than diploids (4.48 and 1.05, respectively) during on‐growing (June–November). In November, no difference of means was found between mature or immature diploids and triploids for any of the weight traits when the effect of vertebrae defects was statistically removed. However, the triploids had attained higher means than mature or immature diploids in gutted and fillet weight by January, suggesting that the loss of muscle mass during early winter was lower in triploids. Sexually maturing diploids (46%) had lower slaughter yield means compared to triploids or immature diploids at each measurement time, and these differences also increased during overwintering. Instead, the means of yield traits remained similar between the triploid and immature diploid groups through the winter. Likewise, fillet redness remained at equally high level in triploids and immature diploids, whereas in maturing diploids this attribute decreased substantially during overwintering. The triploid group had a higher incidence of vertebral defects (12.0%) than diploids (5.3%). The present results demonstrate the potential of triploid trout in producing large‐sized (>2 kg) fillet fish until spring markets. However, more detailed investigations are needed, particularly regarding the animal health and growth efficiency in triploids, relative to their diploid counterparts.  相似文献

Abstract. The present study attempts to investigate the effect of Penaeus monodon -type baculovirus (MBV) infection on growth and survival of the larval stages of Penaeus monodon Fabricius. The results showed that MBV may significantly retard larval growth and cause mortalities during the larval stage. The MBV-infected larvae of P. monodon varied more in body size than uninfected control shrimp, and were discoloured. To detect the presence of MBV in larval prawn, histopathological techniques and an enzyme-linked immunosorbent assay ( elisa ) were used. Positive diagnoses were routinely detected as early as the zoea 1 stage when ELISA was used.  相似文献

Triploid female fish show impaired gametogenesis and are unable to produce viable offspring. The reproductive physiology of artificially-induced triploid female salmonids has been well described up until the time of first sexual maturation in diploids, but few reports exist for older triploids. This study reports the influence of triploidy on growth, ovarian development and reproductive endocrinology among three age classes of female brook trout (Salvelinus fontinalis) in comparison to sibling diploids. Triploids were larger than diploids for most of the study period, but the difference was statistically significant only during maturation and spawning of 2+ diploids. Plasma estradiol-17 (E2), testosterone (T) and vitellogenin (VTG) levels in triploids were generally lower than in diploids, and VTG was the only parameter to show seasonal fluctuations resembling those of diploids. Triploids showed significantly lower GSI and total oocyte number than diploids of similar age, and only half of all triploids sacrificed during the study (n=56) had developing oocytes in their ovaries. At age 3+, 13 of 19 triploid females had oocytes at various stages of development, including perinucleolar, yolk vesicle and yolk globule stages. In addition, three of these fish had collectively produced 72 mature stage oocytes. Thus, whereas diploid brook trout can produce mature oocytes as two-year-olds, triploids cannot do so until four years of age, with the number of mature oocytes being greatly reduced.  相似文献

An alternative larval shrimp feeding regime, in which umbrella‐stage Artemia were constituting the first zooplankton source was evaluated in the culture of Litopenaeus vannamei. In a preliminary experiment, umbrella‐stage Artemia were fed to larvae from zoea 2 (Z2) to mysis 2 (M2) stages to identify the larval stage at which raptorial feeding starts and to determine daily feeding rates. The following experiment evaluated the performance of two feeding regimen that differed during the late zoea/early mysis stages: a control treatment with frozen Artemia nauplii (FAN), and a treatment with frozen umbrella‐stage Artemia (FUA). The ingestion rate of umbrella‐stage Artemia increased from nine umbrella per larvae day?1 at Z2 stage to 21 umbrella per larvae day?1 at M2. A steep increase in ingestion and dry weight from Z3 to M2 suggests a shift to a raptorial feeding mode at the M1 stage. Treatment FUA exhibited a significantly higher larval stage index (P < 0.05) during the period that zoea larvae metamorphosed to the mysis stage, and a higher final biomass, compared with treatment FAN. Based on these results and on practical considerations, a feeding regime starting with umbrella‐stage Artemia from Z2 sub‐stage can be recommended for L. vannamei larvae rearing.  相似文献

One approach to ongrow juvenile European lobster, Homarus gammarus, is to utilize land based rearing systems, incorporating automated feeding, individual culture and provision of stable pelleted feeds, preferably using sustainable ingredients. We initiated three feeding experiments to investigate the general suitability of ingredients produced from seafood by‐products as novel feeds for H. gammarus, in terms of promoting survival, development and growth of post‐larval lobsters from post‐larvae (PL) stage IV to the first juvenile stage (stage V). The first experiment was designed to screen an array of candidate, locally produced, novel protein sources on growth performance parameters. This initial experiment revealed that PL reared on a raw (i.e. wet, unprocessed shrimp) feed used as a reference showed superior performance to those reared on experimental feeds containing fishmeal, herring protein isolate or mussel meal; however, a novel type of shrimp meal, produced by flocculation from waste water, promoted the best PL performance of any experimental feed. A second experiment was designed to test the effect of drying method and to optimize the form of a wet shrimp reference feed used by lobster hatcheries. This showed that the performance of PL reared on experimental freeze‐dried shrimp feed was not significantly different to those reared on the wet, unprocessed shrimp used as a reference feed. However, lobsters offered experimental oven‐dried shrimp feed (with or without an immune supplement) resulted in significantly lower survival or growth performance. A third and final experiment was designed in an attempt to improve a candidate herring‐based protein source, by supplementing with nutrients found in shrimp. However, the results showed that PL reared on the wet reference shrimp feed still showed superior growth and survival than those reared on a herring feed alone, or supplemented with additives found in shrimp meal (either glucosamine, astaxanthin or both supplements combined). The high survival and growth, low incidence of moulting problems and high availability of waste shrimp material, suggest that non‐heat‐treated shrimp products are a promising feed ingredient for post‐larval European lobsters.  相似文献

Morphological development and allometric growth patterns of Seriola lalandi larvae were assessed to characterize normal growth patterns under culture conditions. Early ontogenetic stages of yellowtail kingfish exhibited an exponential growth in terms of standard length as a function of age. Five development stages were characterized from hatching to the juvenile stage: larval stage I (0–2 days post hatch, dph) with endogenous feeding, characterized by a small yolk sac, unpigmented eyes, primordial finfold surrounding the body and a closed mouth; larval stage II (2–15 dph) characterized by mouth opening, complete pigmentation of eyes and the beginning of the exogenous feeding; subsequently, in the larval stage III (15–25 dph) the posterior tip of notochord of the larvae bended upward and the first rays appeared in fins, concomitant with a change in swimming behaviour; thereafter, larval stage IV (post‐flexion stage; 25–30 dph) began when larvae resembled in morphology to a juvenile organism; however, caudal and dorsal fins were not completely development. Lastly, the juvenile stage was reached 30 dph characterized by a morphology and fin structures similar to those of the adults. Growth and development of structures and organs associated with vital functions such as feeding, sensorial and breathing systems seemed to be more critical previous to 23 dph, which was reflected with a positive allometric growth of head and eyes during this period. The results from this study can be used as a tool‐guide to assess normal development in larval research with S. lalandi to improve existing rearing protocols in hatchery production.  相似文献

Because of the filter‐feeding behavior of shrimp larvae, it is important to define precisely the size of the particle ingested in the different stages until postlarval stage where raptorial habits are more evident than the filter‐feeding lifestyle. Selectivity assays were conducted by using Polystyrene DVB particles with diameter between 1 and 50 μm as food. A group of organisms from each stage were put into the particle suspension for 15 min to let the polystyrene particles be ingested. The particle distribution in the media and the content of the gut of the larvae were characterized with digital image processing analysis. The results were compared using Ivlev selectivity formula, which compares the frequency distribution of each size of the particle in the media and in the gut of larvae. The results of selectivity were adjusted with a third‐order polynomial regression to determine the optimum and preferred size of the food particles for each larval stage between Zoea I and Postlarva I. It is concluded that the different larval stages of Litopenaeus vannamei may be considered as a single group of larvae who ingest foods with size between 5.71 and 20.33 μm. The optimal size of the food ingested was 14.42 μm wide.  相似文献

Two trials were conducted to evaluate the effects of chemically synthesized astaxanthin (Ax) on growth performance, survival, and stress resistance in larval and post‐larval kuruma shrimp (Marsupenaeus japonicus). Six dietary levels of Ax (0, 50, 100, 200, 400 and 800 mg/kg diet) were added to a baseline diet. As a first study, an 8‐day feeding trial was conducted on larval kuruma shrimp, with results showing that larvae‐fed diets supplemented with different Ax levels exhibited better performance during developmental and metamorphosis to postlarvae. Broken‐line regression analysis indicated that the optimal levels for growth and stress resistance of larvae were 168.9 mg/kg and 82.1 mg/kg diet, respectively. Second, a 30‐day feeding trial was conducted on post‐larval shrimp. Supplementation with 100 and 200 mg/kg Ax yielded significantly higher final body weight, body weight gain and specific growth rate than that in a control group. A cumulative mortality index for osmotic stress revealed significantly better performance in a group fed 200 mg/kg supplementation than in controls. Broken‐line regression analysis indicated that the optimal levels for growth and stress resistance of postlarvae were 108.7 mg/kg and 178.1 mg/kg diet, respectively. Based on these results, we suggest that carefully dosed Ax supplementation is a beneficial nutritional strategy for the early developmental stages of kuruma shrimp.  相似文献

This study determined the effect of triploidy on the survival, growth and gonadal development of turbot from 6 to 48 months of age. From 6 to 24 months of age (first sexual maturity), survival was similar in both ploidies (P > 0.05). From 24 to 48 months of age, after the first sexual maturity, survival was 91.9% in diploids and 100% in triploids, which did not exhibit the post-spawning-associated mortality. Growth was similar for both ploidies during the first year of life. After that, triploids grew significantly (P < 0.05) more that diploids, with more marked differences after each spawning season. From 24 to 48 months, the average weight difference between both ploidies was 11.4 ± 1.9%, ranging from 4.3 to 23.0%. At 47 months of age, the biomass of triploids was 10.3% greater in total weight and 14.3% greater in eviscerated weight. Gonads of triploid males were similar to that of diploids, whereas in triploid females, they were significantly smaller and rudimentary. A histological analysis carried out at 47 months of age showed complete sterility of triploids in both sexes. Sex ratio was 1 male (M):0.6 female (F), for diploids, significantly (P < 0.05) different from 1:1, and 1 M:3.3 F for triploids, significantly (P < 0.05) different from 1:1 and from the diploids. Since females grow more than males, culture of triploids benefited from the high female ratio, which helped to reduce size dispersion. In addition, their sterility allowed better performance by avoiding the reduction in growth that takes place during the spawning periods. Together, these observations indicate that triploidy induction can be an interesting option for turbot aquaculture, especially for the production of large-size fish of more than 2 years of age.  相似文献

Triploid Pacific oysters Crassostrea gigas farmed in Port Stephens, NSW had an exceptionally fast growth rate and reached a whole weight of 55 g in 13 months versus 20 months for diploids. Mortality of the triploids (24.5±2.94%) was significantly lower (P<0.05) than that of the diploids (40.0±2.26%) over the duration of the experiment (July 2002–February 2004). Unfortunately, this advantage was offset by discoloration of the meats of the triploids when they were in better condition than the diploids over summer (October 2003–March 2004). However, discoloration of meat of triploids had cleared up by April 2004 and neither did they suffer this problem from April–September 2003. The triploids also had a lower peak condition than the diploids. Oysters in peak meat condition, i.e. spawning condition, are preferred for the half shell trade in Australia and in this study, there was at least a six‐month period prior to discoloration, when the triploids were large enough and had sufficient meat condition for marketing on the half shell.  相似文献

The effects of thermal treatments on induction of triploidy in Atlantic cod have been investigated. Cold shock [−1.7±0.1°C at 20 min post fertilization (PF) for 2 h] was based on a previously developed protocol, and heat shocks, below the lethal threshold of 24°C, were at 16, 18 or 20°C applied 20, 30 or 40 min PF for 20 min. Cold shock did not affect larval survival and was ineffective for producing triploids (range 0–4%). A heat shock of 20°C at 20 min PF generated the highest percentages (range 66–100%) of triploid larvae at hatching, with survival ranging from 10% to 20% relative to the controls. Lower heat shock temperatures or delayed shocks increased survival but decreased the number of triploids, providing no net gain in triploid yield (range 1–9%). Heat shocks applied later than 20 min PF produced 2–4% tetraploid larvae at hatching. A thermal shock of 20°C initiated at 20 min PF and lasting 20 min proved to be the most generally efficient treatment for induction of triploidy in Atlantic cod.  相似文献
