Two approaches were used to determine the relationship between salt tolerance during seed germination and vegetative growth in tomato. First, F4 progeny families of a cross between a breeding line, ‘UCT5’ (salt sensitive at all developmental stages), and a primitive cultivar, ‘PI 174263’ (salt tolerant during germination and vegetative growth), were evaluated in separate experiments for salt tolerance during germination and vegetative growth. There were significant differences among the F4 families in both the rate of seed germination and the plant growth (dry matter production) under salt stress. There was, however, no significant correlation between the ability of seeds to germinate rapidly and the ability of plants to grow under salt stress. In the second approach, selection was made for rapid germination under salt stress in an F2 population of the same cross and the selected progeny was evaluated for salt tolerance during both germination and vegetative growth. The results indicated that selection for salt tolerance during germination significantly improved germination performance under salt stress; a realized heritability estimate of 0.73 was obtained. Selection for salt tolerance during germination, however, did not affect plant salt tolerance during vegetative growth; there was no significant difference between the selected and unselected progeny based on either absolute or relative growth under salt stress. Obviously, in these genetic materials, salt tolerance during germination and vegetative growth are controlled by different mechanisms. Thus, to develop tomato cultivars with improved salt tolerance, selection protocols that include all critical developmental stages would be desirable.  相似文献

Summary The fact that the F1 progeny of a pair crossing is always of the same geneticl structure and more uniform than a F2-Fn progeny, stimulated the author to apply vegetative propagation in the breeding of beet, rye and cabbage. As early as 1917, the present writer commenced to use vegetative propagation as a method of breeding cross fertilizing plants.For more than thirty years and until now, two clones of Early Pointed Cabbage have been vegetatively maintained in the Trial Garden at Sappemeer (Prov. of Groningen). The presence of gossypol and its derivatives above the WHO/FAO standards (0.02–0.04%) in cotton seed oil and meal limits its usage as food and feed. To the contrary, the presence of pigment glands filled with gossypol and its derivatives helps to protect cotton plants from phytophageous pests. Thus a desirable cultivar would have glandless seeds on a glanded plant. This paper describes results on the successful introgression of this trait from Gossypium bickii into cultivated upland cotton. Five different tri‐specific hybrids (ABH1, ABH2, ABH3, ABH4 and ABH5) were obtained by crossing the amphidiploid F1 (G. arboreum × G. bickii) with different gland genotypes of G. hirsutum as male parent. The hybrids were highly sterile, and their chromosome configuration at meiosis metaphase 1 (M1) in pollen mother cell (PMC) was 2n = 52 = 41.04 I + 4.54 II + 0.57 III + 0.04 IV. All five hybrids were similar in morphological characters, except for the gland expression and gossypol contents. The hybrid (ABH3) derived from genotype Gl2Gl2gl3gl3 of upland cotton (a single gene dominant line) had completely introgressed the target trait of G. bickii. While ABH1 and ABH2, which derived from recessive (gl2gl2gl3gl3) or dominant (GlGl) glandless upland cotton genotypes, had glandless seeds too, but the density and size of the glands on the plant were reduced significantly.  相似文献

Sixteen white clover genotypes and their half-sib progenies were grown in pure stand. Each clover progeny was also grown in a mixed stand that also included three grass varieties of different species. Dry matter (DM) yield was measured over 2 years and seed yield at the second year. Competitive ability of clover families was defined as the ratio between mixed stand and pure stand for clover DM yield. Narrow-sense heritability from parent-offspring regression of pure stand data was high to moderate (h20.50) for DM yield, seed yield and most of their components. Persistence as predicted by stolon density showed negative genetic correlations with seed yield (rg= -0.70) and DM yield (r2= -0.60), whereas seed yield and DM yield tended towards a negative correlation (r2=0.45). Sizes of different vegetative or reproductive organs were generally correlated positively. Evidence was provided that the set of parent genotypes represents well the genetic variation available within the Ladino gene pool. The results highlighted the difficulty of combining relatively short-term DM yield, persistence and seed yield into a unique plant type. Differences in competitive ability emerged only in the second year, better ability being related mainly to petiole length among the traits assessed in pure stand.  相似文献

Most of model cotton varieties used in tissue culture have glands on both the reproductive and vegetative parts of the plant.These glands contain compounds that are toxic to human and non-ruminant animals.The presence of these compounds limits their usage as food and feed.To obtain a glandless cotton variety with high-frequency somatic embryo production ability,27 glandless varieties were screened in improved MSB media containing 0.1 mg · L-1 IAA,0.1 mg · L-1 KT,and 0.1 mg · L-1 2,4-D.Hengwu 8930,whose regeneration rate was only 2.5%,and the cycle of regeneration was about 12 months had the highest frequency.  相似文献

Two-year-grafting experiments within and between Gossypium barbadense and Gossypium hirsutum accessions were conducted to study the source, transfer, and alteration of gossypol and methylation of (+)-δ-cadinene synthase (CDN) genes along with whole chromosomes of A01, A04, A11, D01, D05, and D11, harbouring the cotton CDNs, using the whole genome bisulfite sequencing (WGBS) method. The results revealed that although a glandless line used in this study had a low level of gossypol in the seeds, it did not contain any pigment glands, indicating that expression pathways of gland and gossypol were under genetic control of different loci in cotton. Grafts with glandless rootstocks produced significant amounts of total gossypol in the seeds, clearly demonstrating that the source of gossypol biosynthesis was not specific to the root. Heterografts consisting of scion GN, a glandless line, and scion TM-1 grafted on Pima 3-79 rootstocks caused increased total seed gossypol levels whereas homograft TT had decreased total seed gossypol levels. Alteration of total gossypol levels was also associated with gland density, and DNA demethylation levels of (+)-δ-cadinene synthase genes.  相似文献

[Objective]We studied the process of pigment gland formation and the dynamic change of gossypol synthesis, the dynamic relationship and molecular mechanism between the pigment glands and gossypol during the process of embryo formation and seed germination in G. bickii Prokh. [Method] We chose G. bickii as meterial and studied the tissue section observation, determination of gossypol contents and gene expression profile analysis of pigment gland formation and gossypol synthesis genes. [Result] The results showed that a large number of pigment protoglands formed on the cotyledons during embryo development process in G. bickii. The pigment protoglands formed to cavity structure at 12 hours after seed germination, and then converted into mature pigment glands. Analysis of gene expression profiles related to the pigment gland formation and gossypol synthesis during seed germination showing that GoPGF gene was highly expressed on the cotyledons of seed germination of 12 hours, which may be related to pigment protoglands forming to cavity structure of G.bickii. Farnesyl pyrophosphate synthase gene (FPS) was highly expressed on the cotyledons of seed germination of 24 hours showing it may be associated with the gossypol transported into the cotyledon pigment glands. CYP706B1 gene was highly expressed on the cotyledons of seed germination of 0 hours and HMG1 were highly expressed on the cotyledons of seed germination of 0 hours, 36 hours and 48 hours showing these genes may be associated with the formation of pigmented cavities and the storage and transport of gossypol; Cadinene synthase genes(CAD1-A) and HMG2 genes were not closely related to the pigment gland formation. [Conclusion] The 12 hours after seed germination is the key period of the morphogenesis of the pigment glands, and the morphogenesis of the pigment glands is synchrony with the time of the occurrence of gossypol in G.bickii. GoPGF, FPS, CYP706B1 and HMG1 are associated with the formation of pigmented cavities and the storage and transport of gossypol.  相似文献

【目的】研究不同基因型棉花不同器官的色素腺体与棉酚旋光体含量之间的相关性。【方法】以6个陆地棉材料中棉所12、中棉所12(无)、珂字棉312、珂字棉312(无)、IF11和冀棉14为对象,精确测定主要器官的色素腺体密度和大小,并采用柱前衍生拆旋方法进行高效液相色谱分析,测定其相应的棉酚旋光体含量,然后对色素腺体和棉酚旋光体含量进行相关性分析。【结果】低酚棉全株无色素腺体,但在各器官中仍能检测到低于0.02%(质量分数)的少量游离棉酚存在。有酚棉各器官均具有典型的色素腺体,其中棉仁的色素腺体最密,苞叶最稀;色素腺体大小(直径)以铃壳最大,花瓣和棉仁最小。不同器官中,总棉酚含量以棉仁最高,苞叶最小;棉仁和花瓣的右旋棉酚含量大于左旋棉酚,而萼片、铃壳、上部叶、中部叶和下部叶的左旋棉酚含量大于右旋棉酚。有酚棉的色素腺体密度与大小之间呈极显著负相关;而总棉酚含量与不同棉酚旋光体含量之间,以及左右旋棉酚含量之间呈极显著正相关。色素腺体密度与总棉酚含量以及不同棉酚旋光体含量之间呈极显著的正相关,而色素腺体大小与右旋棉酚含量之间呈显著的负相关。【结论】棉仁与花瓣的总棉酚含量存在高度的相关性,因此,可以通过花瓣的棉酚含量来判断棉仁的棉酚含量。  相似文献

Common bean (Phaseolus vulgaris L.), an important food crop in Europe, America, Africa and Asia, is thought to fix only small amounts of atmospheric nitrogen. It contributes significantly to the sustainability of traditional cropping systems because of the predominance of small-scale farmers who cultivate beans in those areas. The objectives of this work were to evaluate bush bean varieties under common agronomic cropping systems and to evaluate breeding lines under low N-fertility sole cropping and intercropping systems. The purpose of the study was to characterize the genotype and cropping system's variability in symbiotic and plant characters and to identify the most suitable genotypes to establish an effective symbiosis with indigenous strains of Rhizobium. No significant differences among the bush bean varieties evaluated under typical fertilization practices were observed for N2-fixation and plant traits except for seed nitrogen. Significant differences among the bean lines studied under low N-fertilization conditions were detected for plant growth,plant component and N2-fixation traits. A significant interaction of bean genotype x cropping system was found for number of nodules per plant and nodule moisture on the bush bean varieties studied, and for days to emergence, days to flowering, end of flowering, shoot length, root dry weight and shoot nitrogen on the bean lines evaluated. Nodulation parameters were correlated positively with the yield components, shoot and root parts and duration of flowering, and correlated negatively with seed crude protein, pod and seed dimensions and seed dry weight. These observations indicate that it may be possible to increase both the symbiotic N2-fixation and seed yield through plant breeding. 莲藕是一种繁殖能力很强的植物。它可以采用无性繁殖──用芽繁地可以采用有性繁殖──用莲子繁殖。根据莲的繁殖的多样性及其繁殖器官上的特殊性,可以采用多种途径来进行莲藕品种的改良工作。(1)从自然混杂群体中单株东选育种。(2)从顶芽繁殖后代中选育。(3)从自然杂交莲子实生苗后代选育。(4)人工杂交选育新品系。由于莲藕采用人工杂交后,可以用无性繁殖方式,把杂交的优势固定下来,不需要多代自交纯化或者年年进行杂交制种,所以这将是一项行之有效的育种途径。文中并介绍了杂交后代F1代的性状表现,以及所选出的6个新品种(系)。  相似文献

Leaf senescence leads to a progressive decline in the photosynthetic competence of the leaf. This paper describes some effects of source:sink imbalance on leaf protein catabolism and senescence in soybean. We manipulated pod growth by restricting 100 or 50 % (PR-100 or PR-50, respectively) of young pods at the R4 stage in plastic drinking straws. This effectively reduces final seed mass without interrupting the vascular connections of pods. Nodulating (NOD+) and non-nodulating (NOD−) isolines of the 'Clay' soybean were grown in drainage lysimeters and three pod-restriction (PR) treatments were compared. Pod restriction decreased seed biomass per plant as a result of lower individual seed mass, which was only partially balanced by the increase in seed number. The nitrogen concentration in seeds remained unchanged in NOD+ plants, while it increased with the degree of sink restriction in seeds of NOD− plants. Leaf soluble protein, CO2 exchange rate and seed nitrogen content were consistently lower in NOD− plants; the leaf protein level remained stable with time in PR-100 plants, decreased for PR-50 and dropped for controls. Endoprotease (HBase) and carboxypeptidase (CPase) activities were significantly lower in leaves from PR-100 plants, while aminopeptidase activity was enhanced, indicating a de novo synthesis of leaf protein. This is consistent with the reported accumulation of vegetative storage proteins (VSPs) in soybean and other legumes after moderate or severe sink reduction. Thus, small modifications of the source:sink ratio such as those obtained by the non-destructive PR technique have an impact on leaf protein catabolism. Nodulating and non-nodulating soybean isolines showed similar responses to PR in terms of leaf senescence initiation and progression, but the rate of the processes appear to be largely influenced by plant N status.  相似文献

Soybean seed averages about 40% protein and is a dominant source of protein in animals and human foods. Increasing protein in soybeans is a goal in soybean breeding programs. Initial selection for protein among progeny from breeding populations is often based on non-destructive Near Infrared Reflectance (NIR) spectroscopy analysis of a 5 g or more seed sample in the F3 or later generations. NIR spectroscopy analysis for protein on single F2 seed would allow selection at the earliest possible time and improve selection efficiency for protein content; however this practice has not been evaluated. The objective of this study was to test effectiveness of NIR spectroscopy analysis of single F2 seeds to select for increased protein content from two populations developed from high and average protein content parents. F2 seeds from a single F1 plant of each population showed a normal distribution with transgressive segregation for protein concentration. In both populations, F3 seeds produced from plants from single F2 seeds with either low or high protein content were analyzed for protein by NIR spectroscopy. Protein means and ranges of F3 seed selected from high protein F2 seeds were higher in protein than F3 seed from low protein F2 seeds which produced low means and ranges in protein content. This shows that analysis of single F2 seed for protein content using NIR spectroscopy was effective in selecting for increased protein in the F3 generation. Analysis of single F2 seeds from breeding populations will improve breeding efficiency for protein in soybean breeding programs.  相似文献

Genetic variation, heritability and progeny testing in meadow bromegrass   总被引:2,自引:0,他引:2  
Meadow bromegrass is a recently introduced pasture grass in western Canada. Its leafy production and rapid regrowth have made it a major grass species for pasturing beef animals in this region. Because relatively little breeding work has been done on this species, there is little information on its breeding behaviour. The main objective of this study was to estimate genetic parameters and assess breeding methodologies for meadow bromegrass. Forty half‐sib (polycross and open‐pollinated) and full‐sib (selfed) progenies were evaluated for dry matter and seed yield, fertility index, harvest index, plant height, plant spread, crude protein, neutral detergent fibre and acid detergent fibre. Genetic variation for seed yield, harvest index, and plant height was significant in the open‐pollinated and selfed tests. Genetic variation estimates for dry matter yield were negative (polycross), not significant (open‐pollinated), or significant (selfed). Estimates of genetic variation for quality traits were not significant, except for acid detergent fibre (selfed). Correlations among characters indicated that it is possible to simultaneously improve seed and forage yield. Rankings of progenies by the half‐sib tests for forage and seed yield were not altered by the use of the synthetic parental value (SVi), which includes information from selfed progeny. The polycross progeny test did not discriminate parents as well as the open‐pollinated and selfed tests; this may have been a result of non‐random pollination in the polycross. Correlation among the progeny tests, showed that open‐pollinated and selfed progeny tests agreed for all characters, except for harvest index, while correlation between polycross and selfed tests were significant for six characters but not dry matter yield, and acid and neutral detergent fibres. The most dissimilar tests were polycross and open‐pollinated, with significant correlations found only for fertility index, height, spread and neutral detergent fibre. It was concluded that there was significant variation and moderate heritability for most traits in the meadow bromegrass populations evaluated, and that the open‐pollinated progeny test is the method of choice for selecting parents for synthetics of this species.  相似文献

Summary Autotetraploid (2n=4x=28) pearl millet inbred lines, Tift 23BE and Tift 239DB, have been developed for use in crosses with other polyploid Pennisetum species. Each line set less than 1% and 3% selfed and open-pollinated seed, respectively. Seed germination was usually less than 17%. First generation (F1) hybrids between the two inbreds set up to 61% seed while up to 40% of the seed from hybrids germinated.Seed weight per inflorescence for two planting dates averaged 0.32 g and 3.06 g for the two inbreds and the second generation F2 progeny, respectively. One hundred seed weight was also significantly higher (0.78 g vs 0.48 g) in the F2 progeny, probably due better endosperm development. Chromosome behavior and pollen stainability were similar in the inbred parents and hybrids. However, the hybrids shed more pollen than the inbred parents.Heterosis was evident in the F1 hybrids and F2 progeny which showed significant increases in plant height, leaf length, leaf width, and inflorescence length in addition to fertility improvement.  相似文献

Clearfield rice technology is a popular method for controlling noxious red rice weeds (Oryza sativa L.) in commercial rice fields in the southern U.S. Previous research has detected red rice-Clearfield variety F1 hybrids at low rates in Louisiana and Arkansas. The first research objective was to determine genetic control of imazethapyr resistance in F1 hybrids and F2 populations derived from natural and controlled hybridizations of red rice and Clearfield rice. The second objective was to characterize and compare agronomic performance of the hybrids and their progeny with Clearfield varieties. Genetic analysis showed that imazethapyr resistance was dominant in all tested F1 hybrids with single and two-gene inheritance observed across composite F2 populations. F1 hybrids exhibited high levels of variation for plant height, heading date, and seven reproductive traits. Heterosis was observed in the hybrids versus Clearfield varieties for plant height, heading date, seed-bearing tillers, panicle length, and spikelets/panicle. While seed production of the F1 hybrids was generally inferior to that of the commercial varieties, one red rice/CL121 hybrid produced greater seeds/panicle than the CL121 commercial parent. Extensive variation was observed for all measured traits in the F2 populations derived from either natural or controlled crosses. Results from this study indicate that red rice/Clearfield F1 hybrids normally exhibit low reproductive seed capacity, but a small proportion of the subsequent F2 progeny can produce high fecundity levels. Effective stewardship practices are therefore warranted to reduce the occurrence of such hybrids and their offspring to ensure continued success of the Clearfield technology. Red clover (Trifolium pratense L.) is an important perennial forage crop in Europe. Unfortunately, seed production of modern cultivars is often unsatisfactory and breeding progress for higher seed yield is slow. We evaluated whether marker assisted parentage analysis could identify progeny plants with two high seed-yielding parents in a diploid selection trial. Furthermore, we studied if breeding for high seed yield could be speeded up by selecting progeny plants with two high seed-yielding parents, compared to traditional selection on the mother plant only. In this study, 111 genotypes with excellent vegetative traits from a first cycle selection trial were allowed to pollinate each other in isolation. After seed harvest, ten high seed-yielding plants were identified and their seeds were used to grow ten half-sib progeny populations, with 1,121 plants in total. Two multiplex primer sets targeting 18 SSR loci were designed and used to fingerprint parental and progeny plants. Parentage was reliably determined for 1,083 progeny plants: 135 plants were the result of a cross between two high seed-yielding parents. In a 2-year seed yield trial, the seed yield of these 135 progeny plants was compared to that of a control group of 216 progeny plants from the same ten high seed-yielding mother plants but with no selection on the male contribution, as is current practice. Compared to the control group, progeny plants with two high seed-yielding parents had significantly higher seed yields, 23.0 and 75.9 % higher in 2012 and 2013 respectively, and a significantly higher thousand seed weight, 6.6 and 5.7 % higher in 2012 and 2013 respectively. Seed yield was strongly correlated with flower abundance (r = 0.643) and plant volume (r = 0.593 in 2012 and r = 0.509 in 2013) and negatively correlated with susceptibility to mildew (r = ?0.145). We conclude that breeding programs aiming at increasing seed yield in diploid red clover cultivars would strongly benefit from the integration of marker assisted parentage analysis to identify those progeny plants that are the result of a cross between two high seed-yielding plants. This goal cannot be achieved with traditional family selection where only the seed yield of the mother plant is known.  相似文献

 采用二倍体种子数量性状遗传模型和统计分析方法,分析了陆地棉9个亲本和30个杂交组合的子指、种仁率、仁壳比、蛋白质含量、油分含量和棉酚含量等种子物理性状和营养品质性状的遗传效应、遗传率和遗传相关性。结果表明,所有棉子品质性状均受种子直接遗传效应和母体遗传效应的共同控制,种仁率还受到细胞质效应的影响。子指和仁壳比以母体显性效应为主,蛋白质含量以显性效应为主,油分含量和棉酚含量以母体加性效应为主。遗传相关性分析表明,棉子物理性状和营养品质性状间的显性遗传相关均达到极显著水平,其中子指与蛋白质含量呈极显著显性正相关,子指与油分含量和棉酚含量呈显性负相关。种仁率和仁壳比与3个棉子营养品质性状间的母体遗传相关均达到了极显著水平。  相似文献
