Intestinal establishment and reproduction of adult Trichinella spiralis, Trichinella nativa, Trichinella britovi and Trichinella pseudospiralis were examined as single species or mixed species infections in foxes. This is the first study of intestinal dynamics of Trichinella spp. in a carnivore model and the results suggest that the intestinal phase is relatively short as only very few worms were recovered 10 days post-inoculation (dpi). In mixed species infection with equal doses of T. nativa and T. spiralis, molecular typing demonstrated that 64% of the intestinal worms and 78% of the muscle larvae were T. nativa. Conversely, T. spiralis dominated in the mixed species infections with T. pseudospiralis, constituting 66% of the intestinal worms and 94% of the muscle larvae. Although, the individual recoveries of intestinal worms were only up to 5.6% on day 1, and up to 1.5% on day 4 post-infection, the muscle larvae establishment was comparable to other fox studies. Infectivity, measured as muscle larvae burden did not differ among the four species of Trichinella, which is in contrast to other models with mice, rats, pigs or herbivores. Although statistically significant differences in intestinal worm burdens were found for some days, no distinct species were recovered in consistently higher numbers than the others.  相似文献

Over a period of one year, from March 1984 to April 1985, the abomasa of 157 dairy cows in East Flanders (Belgium) were collected and examined for the presence of nematodes. Faeces and blood were also collected from the animals. No worms were recovered from 18 cows (11.5%), 118 cows (75%) had a low to moderate worm burden (10-10,000) and 21 cows (13.5%) a high worm burden (greater than 10,000). The geometric mean total number was 2171. Ostertagia ostertagi and Trichostrongylus axei were the main species involved, with the former accounting for 90% of all trichostrongyles recovered. For both worms a seasonal pattern was evident, with the highest worm counts in December-January. Between December and February greater than 97% of the Ostertagia spp. worm burden were EL4. For all animals the average pepsinogen level was 1391 +/- 494 mU tyrosine with no statistically significant relationship between pepsinogen levels and the total number of gastric nematodes. Only 45% of the faeces samples were positive for strongylid eggs with a mean of 43 eggs per gram. The percentage generic composition of L3 larvae collected from faeces was 62% Ostertagia, 18% Trichostrongylus, 6% Cooperia, 4% Oesophagostomum and 0.2% Bunostomum.  相似文献

This survey was conducted to determine the chronobiology and seroprevalence of nasal bot infestation (Oestrus ovis) in Spain and to identify the risk factors associated with this disease in caprine herds.

A total of 1590 sera from adult goats were collected at random on 175 farms in southwestern Spain. Sera were tested by ELISA, using crude protein from second stage larvae as antigen. The mean seroprevalence was 46.04% and mean percentage of optical densities was 41.83. These data indicate a high prevalence of this parasite in the investigated areas. The serological survey revealed that goats managed at higher altitudes, at meridians latitudes and on farms with small herds had a smaller probability of infestation. Eighty goat heads, obtained from abattoirs in the central region of Spain, were collected and examined for nasal botflies from February to October 2002. O. ovis larval stages were recovered from the nasal-sinus cavities of 23 goats, reaching a prevalence of 34.94%. The mean larval burden was 3.9 larvae per infested head. No first instars were found during February and March, when the second instar reached its larger count. The third instar was observed in very small number during the whole period of study, with one peak occurring in July–August. These data show the existence of a favourable period for the development of larval instars of O. ovis in goats that starts in February and finishes in September.  相似文献

In autumn 2000, a study was carried out on 25 dairy farms in the vicinity of Utrecht with the aim to estimate infectivity levels for nematode parasites in cows. On each farm, faecal samples were collected from 15 cows, blood samples from 5 of these and herbage samples from 2 cow pastures. Faecal examination demonstrated a variation between farms and within farms in faecal egg output with a mean number of 4 eggs/g faeces (EPG) and Ostertagia spp. and Cooperia oncophora being the dominant species. In 6 out of 21 farms examined, lungworm larvae were detected in at least 1 cow. Serum pepsinogen values and serology using ELISA's with crude adult Ostertagia, crude adult C. oncophora and a specific recombinant C. oncophora protein as antigens indicated low to moderate infection levels. Pasture infectivity levels varied between farms with again Ostertagia spp. and C. oncophora as the dominant larval types and correlated with the crude worm Ostertagia ELISA, the crude worm Cooperia ELISA and the pepsinogen values. Exposure levels were high enough to enable the possible occurrence of production losses on the majority of farms.  相似文献

The chemoprophylactic efficacy of a single dose of the 10% long acting (LA) injectable formulation of moxidectin on nematode infections in calves, was evaluated. Two similar groups of 11 female, first grazing season Holstein calves were turned out in early May on separate plots of a single, naturally infected pasture. Until 56 days post-treatment (pt), the percentage reduction in faecal egg output was 100%, remaining above 90% during the entire trial, except for day 126 pt. More than 90% of the larvae in the treated group were identified as Cooperia until 140 days after treatment and more than 70% during the rest of the trial, whereas in the control group Cooperia was the most abundant species until day 84 pt and Ostertagia from 126 days pt onwards. The reduction in faecal egg output in the treated group was reflected in the mean pepsinogen levels being below the pathogenic threshold at the end of the grazing season (1.8 units of tysrosine (U tyr)) and the absence of diarrhoea during the second half of the grazing season. In the control group pepsinogen levels remained high (mean: 5.5 U tyr) and prolonged diarrhoea occurred in the second half of the grazing season. Furthermore, the weight gain for the treated group at the end of the grazing season was 41.9 kg higher than for the control group. At necropsy, the reduction in O. ostertagi worm burden in the treated group was 97.5% compared to the control group, while the reduction in C. oncophora worm burden was 57%. An additional benefit of the long acting parasitological control, was reduced pasture contamination.  相似文献

The efficacy of the morantel sustained release bolus (MSRB) in controlling gastrointestinal parasitism in beef cattle was assessed during the 1982 spring-autumn grazing season. Forty-eight cows and their calves were allotted to three equal groups. One group (T-1) served as a nonmedicated control group. One MSRB was administered to each calf of the T-2 group, and to each cow and calf of the T-3 group at the beginning of the study. The efficacy of the bolus was assessed by comparison of weight gain performance and parasitological data (fecal worm egg counts, herbage larval counts, worm counts from tracer and principal trial calves, and plasma pepsinogen level determinations). Though not statistically significant, treated calves from Group T-2 had a numerical mean weight gain advantage of 2.6 kg, and those from Group T-3 of 4.7 kg, over control calves. Average daily gains (ADG) for the three groups of calves were 0.69, 0.72, and 0.73 kg, respectively. Untreated cows from Group T-2 and treated cows from Group T-3 outperformed the control cows by 12.3 and 7.5 kg, respectively. Fecal worm egg counts from both groups of treated calves were significantly (P less than 0.01) lower than counts from control calves during the entire 169-day trial; notably, egg counts were reduced by 99% 28 days after MSRB administration to both groups of calves. There were no significant differences in the number of eggs counted from the three groups of cows, probably because of the very low numbers of eggs encountered. Mean total worm burdens of principal calves (six per group) necropsied at trial termination indicated a 91% (P less than 0.01) reduction in Group T-2 and an 87% reduction (P less than 0.01) in Group T-3. Worm-free tracer calves were introduced onto pastures every 28 days to monitor availability of infective larvae. The mean number of worms recovered at necropsy from tracer calves that grazed with control cattle increased as the season progressed. However, the numbers of parasites recovered each month from mid-August through mid-October from tracers that grazed pastures with treated cattle were lower (P less than 0.05) than those levels displayed at trial initiation. The relation between the faecal egg count and the worm burden was investigated in 104 steers of three breeds, 9–24 months of age, which were subjected to natural infection with nematode larvae from pasture at The National Cattle Breeding Station near Rockhampton in central Queensland. The worm species found were Cooperia spp., Haemonchus placei, Trichostrongylus axei, Oesophagostomum radiatum and small numbers of Bunostumum phlebotumum.

The faecal egg count was found to be a reliable measure of the size of the worm burden. Steers slaughtered at 24 months of age had 90% fewer worms and 96% fewer eggs g−1 faeces (P < 0.001) than those slaughtered at 9 months of age. Adjusted for age, the estimated faecal egg count increased 14 times (95% confidence limits 7.2, 35) for each 10-fold increase in estimated worm numbers.

The proportion of each species of larvae obtained from faecal culture (larval differential count) was related to the composition of the worm burden as measured by the proportion of worms in selected maturity categories and in selected length categories for each species (P < 0.001).  相似文献

Gastrointestinal nematode parasite burdens were monitored in a herd of field-grazed cattle from weaning to 2 years old to allow observations to be made on the dynamics of burdens of arrested Ostertagia spp. and Cooperia oncophora. Arrested Ostertagia spp. accumulated in the herd over their first autumn and winter to reach a peak in late winter/spring (August-October). From October until late January they declined at a rate comparable to that at which they had accumulated. Numbers continued to decline at a slower rate until the following autumn (May) when they began to increase again. Observations on the decline of worm burdens in a group of animals which had been moved from pasture to worm-free conditions in October indicated that intake of infective larvae over late spring/summer contributed little to the number of arrested Ostertagia spp. in the herd. No clinical Type II ostertagiosis was observed in the field-grazed animals over the time of decline of the arrested worm burden, nor was there an increase in the number of adult Ostertagia spp. present. However, two of the animals removed from pasture to worm-free conditions developed Type II ostertagiosis approximately a month after the move. Worm burdens of successive groups of "tracers" grazed with the herd every 2 months indicated that there was a small seasonal increase in the propensity of Ostertagia spp. for arrested development. However, estimated accumulation rates of arrested larvae calculated from tracer worm burdens, suggest that this seasonal increase in propensity was not adequate in itself to account for the accumulation rate observed in "resident" animals of the herd. No comparable accumulation of arrested C. oncophora took place in resident animals over the first autumn and winter of the trial despite the fact that tracer worm burdens over this period indicated a marked seasonal increase in the propensity for arrested development by this species.  相似文献

The survey was done to determine the incidence and seasonal prevalence of the gastrointestinal helminths infecting Maine dairy cattle. Approximately equal groups of calves, heifers, and cows from 13 dairy farms, managerially and geographically representative of dairying in the state, were sampled at regular 2-month intervals over the period of a year. On the basis of fecal egg counts and identification of cultured larvae, the following results were obtained: Of the 94 adult cows, 78 heifers, and 91 calves sampled continuously in the course of the survey, 95.7%, 98.7%, and 96.7%, respectively, were strongylorid positive (infected with Ostertagia, Cooperia, Trichostrongylus, Haemonchus, and Oesophagostomun), with an overall incidence of 97%. Strongyloides papillosus eggs were in 64.6% of the cattle, Bunostomum in 40.3%, Nematodirus in 27.8%, Trichuris in 27.0%, Capillaria in 9.5%, Moniezia in 25.1%, and Dictyocaulus viviparus larvae in 2.7%. Worm burdens, as reflected by mean nematode egg production per gram of feces, varied. The greatest worm burdens were in the calves, followed by the heifers and the cows. A marked variation in egg production related to season was observed, particularly with the strongylorid worms. Peak strongylorid egg production was observed in the May-June sampling period. After the peak period, worm burdens decreased in all groups of cattle to their lowest point during the winter period of January-February. This pattern of egg production was similar to that reported by workers in other parts of the world. Farm management practices were shown to be related to the degree of parasitism existing in these herds. Those herds classed as poorly managed had significantly higher levels of parasitism in young animals than those classed as having fair or good management.  相似文献

The safety of heartworm preventives in heartworm-positive cats has traditionally been evaluated using adult Dirofilaria immitis removed from infected dogs and surgically implanted into the cats. An alternate study model uses infective larvae to establish adult infections in cats. Unfortunately, the number of adult worms resulting from the latter method varies widely from none to more than 30, both unacceptable for studies of natural heartworm infection and for studies evaluating product safety in heartworm-infected cats. We sought to determine infection severity in experimental infections via echocardiography to reduce the chances of enrolling uninfected and heavily infected cats into a study. Eighty adult cats were each inoculated with 60 infective D. immitis larvae and maintained for 8 months to allow for the development of adult worms. Antigen and antibody testing, as well as echocardiographic imaging, were performed to confirm and estimate adult worm burdens. Approximately 8 and 12 months post-infection, echocardiographic examination was performed to confirm and enumerate adult D. immitis populations in the cardiovascular system. Worm burdens were stratified as 0, 1-3, 4-11, and > 11 adults, with 0 being considered uninfected and more than 11 considered too heavily infected to be relevant for anthelmintic studies. Cats with clinically relevant infections (1-10 adults) subsequently received multiple treatments with the investigational drug, and worm burdens were confirmed by necropsy 30 days following the final treatment. Worm burden estimated with echocardiography correlated well, but not precisely, with post-mortem counts (p < 0.001, r2 = 0.67). Echocardiography under-, over-, and exactly estimated heartworm burden 53%, 27%, and 22% of the time, respectively. Although the correct category (0-4) was determined by echocardiography in only 54% of cats, positive cats were distinguished from negative cats 88% of the time and the heaviest infections (> 11) were correctly categorized 95% of the time. Both false negative and false positive results were observed. We conclude that echocardiography is useful for detecting mature experimental heartworm infections, identifying cats that have rejected mature infection, and detecting very heavy heartworm burdens, but it is only moderately accurate in classifying lesser burdens. While echocardiography cannot be relied upon to consistently determine the exact heartworm burden in experimentally infected cats, it is useful in stratifying worm burdens for anthelmintic safety studies.  相似文献

The small strongylid infections of two groups of three yearling female Shetland ponies and one yearling Shetland tracer pony were studied. One group was set stocked from April to November and was treated monthly with 5 mg kg-1 albendazole from two days before turnout until July. The other group grazed similar pasture until July, was treated with 5 mg kg-1 albendazole and subsequently removed to pasture grazed by sheep from April to July. The tracer ponies were added to both groups in September. The efficacy of both methods was not completely satisfactory probably because of low efficacy of anthelmintic treatment. There were no significant differences between the cyathostome burdens of the two groups. A high proportion of the cyathostome populations of all ponies consisted of inhibited early third stage larvae (L3). The finding of low numbers of immature fifth-stage worms in the tracer ponies indicated that the considerable adult burdens in the permanent ponies originated from infection picked up before the tracer ponies were added. In the group which was removed to sheep pasture after treatment in July it was likely that the majority of the adult worm burden had been ingested as infective larvae before treatment.  相似文献

Four controlled trials were conducted to evaluate the therapeutic and persistent efficacy of a new moxidectin formulation (moxidectin 1% nonaqueous injectable) against nematode parasites in cattle. This injectable moxidectin formulation, given as a single subcutaneous injection at a dose rate of 0.02 ml/kg BW to provide 0.2 mg moxidectin/kg BW, was highly efficacious (>90–100%) against larval and/or adult stages of many species of nematodes in cattle including, Dictyocaulus viviparus, Ostertagia spp., Trichostrongylus axei, Haemonchus placei, Trichostrongylus colubriformis, Cooperia spp., Nematodirus helvetianus, Strongyloides papillosus, Oesophagostomum radiatum and Trichuris spp. This formulation had persistent efficacy of >90% against D. viviparus for at least 6 weeks post-treatment, H. placei and Oe. radiatum for 5 weeks post-treatment, and Ostertagia spp. and T. axei for 2 weeks post-treatment.  相似文献

From autumn (April) to spring (November), groups of pregnant and dry Merino ewes grazed pasture contaminated with infective larvae of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus colubriformis. A periparturient rise in faecal egg counts occurred in the pregnant ewes, following the maturation in early spring of arrested fourth-stage larvae of H. contortus and failure of pregnant ewes to expel the resulting adult worms. Peak egg counts were seen in September, just before lambing. In dry ewes, egg counts were slightly elevated at the time of the rise in the pregnant ewes, but fell to very low levels as adult worms developing from previously arrested larvae were expelled. Lactating ewes acquired greater burdens of O. circumcincta and T. colubriformis than did dry ewes, but were equally refractory to new infections with H. contortus. It is therefore suggested that the impairment of immunity to helminth infection seen in reproductive ewes may be more specific than was previously envisaged.  相似文献

Abomasa, blood samples and faecal samples for examination of nematode infections were collected from 125 dairy cows during the period November 1997-October 1998. Of these, 12 had no grazing history and were, therefore, excluded from this study. From the remaining 113, 88.5% had nematode eggs in the faeces. Larval identification of the positive cultures showed that Ostertagia spp. larvae were most frequent (97%), followed by Trichostrongylus spp. (29%), Oesophagostomum spp. (23%), Cooperia punctata (20%), Cooperia oncophora (4%), Haemonchus contortus (2%) and Bunostomum phlebotomum (1%). The geometric mean EPG was 2.4. Two cows excreted larvae of Dictyocaulus viviparus (0.1 and 0.6 LPG resp.). Worms were found in the abomasa of 108 cows (96%). In all these abomasa Ostertagia spp. was present (100%). Trichostrongylus axei was found in 47 abomasa (43.5%) and two cows (2%) were infected with Capillaria bovis. The geometric mean of the total abomasal worm counts was 1743 and of Ostertagia spp. alone 1615. Almost all male worms were Ostertagia ostertagi, only occasionally Skrjabinagia lyrata10,000) total worm burden.Ostertagia specific antibodies were highest in late summer and autumn and lowest in spring and early summer. The same pattern, although not so pronounced, was observed for the serum pepsinogen values. No clear seasonal pattern was found for the Cooperia specific antibodies. Antibodies against D. viviparus were detected in seven cows (6%).  相似文献

Over a period of 1 year, from November 1997 to October 1998, the abomasa, blood and faecal samples of 121 dairy cows in Belgium were collected and examined for nematode infections. Nematodes were present in the abomasa of 110 animals. Ostertagia was found in all 110, Trichostrongylus was seen in 65 and Haemonchus in 14 abomasa. Overall, 91% of all trichostrongyles recovered were Ostertagia. The geometric mean total number of Ostertagia was 2750, with an average of 74% inhibited early fourth stage larvae (EL4). Between November and February >90% of the Ostertagia worm burden were EL4 stages. The majority of the animals (56%) harboured a low Ostertagia burden (100-5000) and 15% had a high burden (>10,000). Sixty-four percent of the coprocultures were positive and the genera recovered were Ostertagia sp. (100%), Trichostrongylus sp. (42%), Oesophagostomum (32%), Haemonchus sp. (29%) and Cooperia sp. (16%). A seasonal pattern was evident for serum Ostertagia-specific antibodies and for serum pepsinogen concentration, with the highest levels during the summer, and low values during the winter. Dictyocaulus viviparus specific antibodies were detected in the serum of eight (7%) animals.  相似文献

From October 1980 until September 1981, 191 abomasa of naturally infected cattle were obtained from the slaughter-houses of Mürzhofen and Graz in the province of Styria, and of Wiener Neustadt in the province of Lower Austria and examined to study the monthly and seasonal occurrence of hypobiosis in Ostertagia ostertagi and Trichostrongylus axei. Infections in pastured animals (cows, steers and heifers) were compared with those in stabled animals (bulls fed on green forage and silage). Except for 10 bulls fed on silage all animals were parasitized. The results indicate that in Austria inhibited early fourth stage larvae of Ostertagia accumulate during the winter. Pastured animals showed a higher intensity of infection than stabled animals. Cows less than 3 and greater than 5 years of age had higher worm counts than those between these ages. There was a simultaneous accumulation of third stage larvae and adult worms of Trichostrongylus axei in autumn in steers and heifers. In bulls fed on green forage a higher adult worm burden was seen in winter. No significant correlation existed between the number of worm nodules in the abomasal mucosa and the various developmental stages of the worms. The count of in-utero immature eggs was higher in stabled than in pastured animals. No significant correlation between the number of in-utero eggs and that of female worms was noticeable. Numbers of mature Ostertagia eggs rose in spring, while in fall and winter the immature eggs predominated. An increase in the uterine egg count was observed in Trichostrongylus in autumn. Statistical analyses of our data show that the most decisive factor influencing the numbers of inhibited and adult Ostertagia as well as in-utero eggs is the system of animal management.  相似文献

CASE DESCRIPTION: The owner of a herd of 74 Holstein-Friesian cattle reported decreased milk production, weight loss, and coughing among lactating cows. Owner-initiated antimicrobial treatment was unsuccessful; 1 lactating cow died, and 50% of the lactating cows had clinical signs of respiratory distress, such as tachypnea and coughing. CLINICAL FINDINGS: On the basis of history, physical examination findings, and fecal examination results, affected animals were determined to have Dictyocaulus viviparus (lungworm) infestation. The disease history suggested that the herd contained cows with subclinical patent lungworm infestations; after introduction of susceptible heifers, the pastures had become heavily infested with D viviparus and clinical problems subsequently developed in both newly introduced and resident cows. TREATMENT AND OUTCOME: Affected and unaffected heifers and adult cows were treated with a pour-on formulation of eprinomectin (0.5 mg/kg [0.23 mg/lb]). One animal died, but 2 weeks after treatment, clinical signs among affected cattle were markedly improved. Ten weeks after treatment, milk production improved from 23 kg/cow/d (51 lb/cow/d) to 28 kg/cow/d (62 lb/cow/d). CLINICAL RELEVANCE: The outbreak provides additional evidence that dictyocaulosis is becoming more common among adult dairy cattle, rather than almost exclusively affecting young stock. This may be attributable to anthelmintic use and management practices on dairy farms. Combined with anecdotal reports of an increase in the incidence of dictyocaulosis among adult cattle in North America, D viviparus infestation should be included as a differential diagnosis for decreased milk production, weight loss, and coughing among adult dairy cattle.  相似文献

Tritrichomonas foetus is an extracellular parasite of the reproductive tract in cattle. The mechanism by which T. foetus causes abortion in cattle is largely unknown. There are no studies of infection in the cow oviducts, almost all published papers are related to vagina infection and few articles focusing on the uterus. The aim of the present study was to establish a working model of bovine oviduct epithelial cells and submit these cells to Tritrichomonas foetus interaction. Twenty bovine oviducts were obtained from cows at a commercial abattoir and T. foetus was injected through the isthmus into the oviduct lumen. The whole oviduct was analyzed by scanning and transmission electron microscopy. The results reported here demonstrate that: (1) fresh whole oviducts can be used as a good model to study parasite-host cell interaction; (2) cow oviduct epithelium has been shown to consist of two cell types: ciliated and nonciliated secretory cells, and T. foetus displayed great specificity for the nonciliated cells localized in the deeper oviduct folds; (3) T. foetus adheres as single separate cells, and maintains the flagella externalized; (4) differently from T. vaginalis, T. foetus does not change its shape during the adhesion process; and (5) oviduct cells exhibited morphological characteristics of apoptosis after trichomonadal interaction.  相似文献

An experimental infection of freshly cultured Haemonchus contortus larvae was given to housed, non-pregnant sheep in October. Phenothiazine was administered to all animals shortly after patency, but the egg output remained negative until mid-April indicating that removal of adult worms from the abomasum did not stimulate the resumption of development of arrested larvae. Worm burdens at post mortem suggest that adults were not being expelled to any great degree during the winter from the host, but this information is clouded by the difference in numbers of both adult worms and arrested larvae in sheep of different haemoglobin type. The results suggest that haemoglobin type may also be a factor in arrest of larvae as it is in the resistance of sheep to adult H. contortus.  相似文献
