From June 1987 to June 1988, a total of 180 N'Dama cattle were necropsied to identify and enumerate parasites present, including 39 sick calves and 25 dead calves from local village herds, and 116 adult cattle from a local abattoir. Of 175 animals infected with one or more helminths (97%), Haemonchus contortus was found in 67% of all necropsied animals, Cooperia punctata in 75%, Cooperia pectinata in 55%, Oesophagostomum radiatum in 71% and Bunostomum phlebotomum in 21%. There was a wide range of worm burdens (0-22 925) and a pronounced seasonal pattern occurred. Eighty-two percent of the total adult Haemonchus burden occurred during the rainy season (June-October). By the end of the rainy season, almost 100% of the Haemonchus burden was present as inhibited larvae. By contrast, a different survival strategy appeared to occur in Cooperia spp.; Oesophagostomum spp. and Bunostomum spp., which were found as adults throughout the year. Results indicated that, apart from malnutrition during the dry season, gastrointestinal nematode infections, especially haemonchosis, represent a major constraint on the health and productivity of N'Dama cattle under West African savanna conditions.  相似文献

The abomasa of sheep grazing on natural pastures in the highveld of Zimbabwe were examined for Haemonchus contortus. Of 304 abomasa, 213 (70%) harboured H. contortus. The worm burden increased during the rains to reach the peak in February-March. This was followed by a decline with low worm loads throughout the dry season. The fourth stage larvae (L4) accounted for 1-7% of the total H. contortus population, except during July and August when they comprised 22 and 54% of the worm burden, respectively. It appears that the inhibition of H. contortus is not common in the commercial farming sector where sheep are treated very frequently.  相似文献

A survey was conducted in the Nigerian derived savanna (from August 1987 to July 1988) on the seasonal fluctuations in the composition of Haemonchus contortus burden of naturally infected West African Dwarf sheep and goats. During this period 117 abomasa collected from locally slaughtered animals were processed. Male, female as well as immature worms were enumerated. The incidence of Haemonchus infection was high (77.8-100%) with no definite seasonal distinction. There was however, a clear seasonal trend in the worm burden of animals; higher burdens were evident during the rainy rather than the dry season. There was no significant hypobiosis; Haemonchus contortus survived in the host during the unfavourable dry season (November to March) as adults.  相似文献

Arrested development of Haemonchus, Cooperia and Trichostrongylus spp. was studied in (1) 14 naturally infected and eight experimentally infected West African Dwarf (WAD) goats reared in the derived savanna zone of eastern Nigeria and (2) 55 naturally infected slaughter goats obtained from the savanna and sahel regions of northern Nigeria. Six of the WAD goats carried natural infections of H. contortus and T. colubriformis and eight other (tracer) goats acquired their infections from a grass paddock artificially contaminated with H. placei, C. pectinata and C. punctata, during May to October. Another three WAD goats were artificially infected with mixed cultures of L3 of the latter three nematodes, while five goats were inoculated with 1500-2000 L3 of H. contortus harvested from cultures incubated at 25-30 degrees C for 8 days either in the dark or under normal laboratory conditions. Approximately 41% (9/22) of the infected WAD goats harboured arrested larvae of Haemonchus and/or Cooperia. No arrested larvae of Trichostrongylus were found in the six animals that were infected with this nematode. The level of inhibition varied from 0.4 to 20% and only three animals showed greater than 10% inhibition. This very low level of inhibition occurred in naturally and experimentally acquired infections, irrespective of the time of year. In the case of Haemonchus, the species and strain of the parasite and infection with L3 cultured in the dark also appeared not to influence the level of inhibition. By contrast, 65.5% (36/55) and 5.5% (3/55) of the northern savanna and sahel goats harboured arrested larvae of H. contortus and T. colubriformis, respectively. The mean percentage inhibition of the former was low (2-25%) during most of the rainy season (June-August) and high (75-90%) during the late rains and the dry season (October-April). The lowest and highest mean percentage inhibitions occurred during July and November, respectively.  相似文献

Development and survival of Haemonchus contortus larvae were studied from December 1987 to November 1988 during three different periods (dry season, first and second rainy seasons) on an experimentally infected pasture at Bunia (Ituri, Za?re). Whatever the season, eggs developed into infective larvae within six days and the largest number of larvae on the herbage occurred between the 12th and the 18th day post deposition. However, the two rainy seasons were the most favourable for transmission because of the high number of larvae on the pasture and the increased survival of these larvae after 4 weeks.  相似文献

The effect of time of day, season and stratum of herbage and soil on the availability of Haemonchus contortus and Haemonchus placei third-stage larvae (L3) on pasture was assessed. Feces from infected calves and lambs were placed on pasture plots and samples of upper herbage, lower herbage, mat and soil were collected at five intervals per day throughout the daylight hours on 18 sample days over 12 months. Using recovery rate factors derived from a preliminary investigation on the efficacy of larval recovery from each stratum, the data on larval recoveries were analyzed for the effect of season, time and stratum, and their interactions. Significant (P less than 0.05) differences were found for season, stratum and the season-with-stratum interaction for both parasites. No significant differences were detected for larval counts at different times of the day. Larval recoveries of H. contortus were larger throughout the study than those of H. placei. Most H. contortus L3 were recovered in the summer and autumn, and H. placei in the spring and summer. For both parasites, the recoveries of larvae from the upper and lower herbage were larger than those from the mat and soil. The implications of these findings are discussed in terms of control strategies.  相似文献

From autumn (April) to spring (November), groups of pregnant and dry Merino ewes grazed pasture contaminated with infective larvae of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus colubriformis. A periparturient rise in faecal egg counts occurred in the pregnant ewes, following the maturation in early spring of arrested fourth-stage larvae of H. contortus and failure of pregnant ewes to expel the resulting adult worms. Peak egg counts were seen in September, just before lambing. In dry ewes, egg counts were slightly elevated at the time of the rise in the pregnant ewes, but fell to very low levels as adult worms developing from previously arrested larvae were expelled. Lactating ewes acquired greater burdens of O. circumcincta and T. colubriformis than did dry ewes, but were equally refractory to new infections with H. contortus. It is therefore suggested that the impairment of immunity to helminth infection seen in reproductive ewes may be more specific than was previously envisaged.  相似文献

Studies on the epidemiology of gastrointestinal nematodes of sheep, conducted at two different farms in the Baghdad area, in 1979, revealed two peaks in worm egg counts in ewes, one in spring and the other in autumn; the latter coincided with the lambing season. Similarly in lambs, two peaks of infection were apparent, but the spring peak was not so well defined. During the hot and dry summer months of Iraq, helminthic infection was at a low level. Faecal culture for larvae and their identification indicated the predominance of Haemonchus contortus infection in the ewes and that of Ostertagia and Trichostrongylus spp. in the lambs examined, Trichostrongylus infection predominating during the hot, dry period of the year. The self-cure phenomenon as well as preparturient rise in egg counts, as reported in other countries, was also observed during this study.  相似文献

Efficacy of levamisole was evaluated in a suspected levamisole-resistant population of Haemonchus contortus in the Louisiana State University sheep flock. The efficacy of netobimin also was evaluated against this population of Haemonchus. In trial 1, 5 lambs naturally infected with H contortus were given 8 mg of levamisole/kg of body weight as a drench, and 5 lambs were not treated (controls). Nematode recovery after slaughter indicated 0% efficacy against H contortus. In trial 2, 30 nematode-free lambs were each given 8,300 F1 generation infective larvae of H contortus, which were derived from parent H contortus that survived 2 levamisole treatments in lambs being maintained in a nematode-free environment. Ten lambs were treated with 8 mg of levamisole/kg as a drench, 10 were treated with 20 mg of netobimin/kg as a drench, and 10 were not treated (control). Nematode recovery after slaughter revealed 62.3% and 99.8% efficacy for levamisole and netobimin, respectively, against H contortus.  相似文献

Worm counts from Yankassa ewes, lambs and tracer lambs grazing unimproved pasture at Zaria were obtained each month during the rainy season of 1976. In June and July up to 14 per cent of the Haemonchus contortus populations consisted of early fourth stage larvae (L4). In August this increased to more than 55 per cent and in October to more than 95 per cent. From June to August most of the abundant late L4 contained rodlike crystals. More than 95 per cent of the Haemonchus contortus populations of two lambs killed two weeks after infection with infective larvae, cultured for seven days at 25 degrees to 30 degrees C in the dark, were early L4. Inhibited 3rd stage Trichostrongylus spp did not exceed 26 per cent of the worm populations.  相似文献

Helminth-free lambs (approx 4 months old) were inoculated SC with a purified metabolite of exsheathed third-stage Haemonchus contortus larvae. The metabolite was obtained from in vitro cultivation and was identified as XL3FA1. After 3 periodic XL3FA1 inoculations, lambs were challenge exposed with H contortus. Multiple vaccinations with XL3FA1 did not inhibit the development of worm populations in the lambs; however, worm egg production seemed to be inhibited.  相似文献

Fifty-thousand infective larvae of Trichostrongylus axei dosed to 7-9-month-old Merinos protected them against a subsequent challenge with 50 000 infective larvae of Haemonchus contortus. Protection varied with the period between an initial dosage of T. axei and challenge with H. contortus as follows: After 90 days it was more than 80% effective in more than 80% of sheep; at 119 and 180 days there was still a significant reduction in the numbers of H. contortus recovered (P < 0,05 and P < 0,001, respectively). Prior infestation with either Ostertagia circumcincta or with Haemonchus placei was ineffective against challenge with H. contortus.  相似文献

Twenty helminth-free lambs were fed diets containing either 169 g crude protein (CP) kg-1 dry matter (DM) or 88 g CP kg-1 DM from the age of seven months. One month later five lambs from each dietary group were vaccinated against Haemonchus contortus by the oral administration of 10,000 irradiated larvae on two occasions, four weeks apart. Four weeks following the administration of the second dose of irradiated larvae both the vaccinated and unvaccinated lambs were exposed to an experimental infection of 10,000 non-irradiated H contortus larvae Faecal egg output and haematological changes were monitored throughout the study. The lambs were slaughtered 28 days after challenge when worm burdens were assessed. Vaccination was equally successful in inducing a strong resistance to the challenge infection regardless of dietary status. It was concluded that dietary protein does not influence the response to vaccination with irradiated H contortus larvae of lambs more than seven months old.  相似文献

The potential of the chitin synthesis inhibitor diflubenzuron (DFB) to alter the development of the parasitic nematodes (Ascaris suum and Haemonchus contortus was investigated. DFB given orally (10 mg kg-1 per day for 30 days) to sheep inoculated with H. contortus infective larvae did not prevent the establishment of adults or affect fecal egg output. However, there was a significant (greater than 90%) decrease in the number of infective larvae recovered from fecal cultures derived from lambs harboring H. contortus adults that were treated with DFB. DFB did not affect egg hatching. Oral administration (10 mg kg-1 per day for 20 days) of DFB to swine harboring adult A. suum adults had no effect on the adult worm burden or on egg morphology, but eggs removed from worms obtained from DFB-treated swine contained less chitin than eggs removed from untreated control swine. DFB also inhibited chitin synthesis in vitro in the isolated reproductive tract of A. suum adults. These results indicate that DFB at high doses can inhibit the subsequent development of H. contortus larvae in the feces. Since H. contortus larvae lack chitin, DFB may act on these larvae by a mechanism independent of a direct effect on chitin synthesis.  相似文献

An examination of the occurrence of crystalline inclusions in the intestinal cells of fourth and fifth stage Haemonchus contortus revealed that a high percentage of inhibited early fourth stage larvae (L4) and developing L4 and a lower percentage of early fifth stage larvae (L5) harboured these crystals. Before the fourth moult crystals were falling into pieces, indicating that worms harbouring crystals can develop further. 以培养获得的捻转血矛线虫滞育期虫体及同期正常发育的虫体为研究材料,采用设计的锚定引物和随机引物,通过mRNA差异显示PCR技术对滞育期幼虫的差异表达基因进行了筛选。结果获得了74个滞育期差异表达的基因EST。生物信息学分析表明:29个差异序列与已知的捻转血矛线虫基因组序列具有同源性;14个差异序列与已知线虫的EST具有同源性。同源基因中的rps-30、T24F1.2等已被证明参与了秀丽隐杆线虫的滞育形成。差异序列的克隆为捻转血矛线虫滞育相关基因及滞育形成机制的研究奠定了基础。  相似文献

Three paddocks were contaminated with Haemonchus contortus eggs from early spring to mid summer by yearling sheep. In Paddock 1, all contamination was derived from worms that had been previously inhibited in their development. The contamination in Paddock 2 was produced only by infections recently acquired from pasture, and in Paddock 3 contamination was attributable to both sources. When the yearling sheep were replaced in mid-summer by newly weaned lambs, severe outbreaks of haemonchosis occurred on all paddocks. It was concluded that inhibited H. contortus larvae are capable of producing sufficient eggs, when they resume development, to initiate an outbreak of haemonchosis in susceptible sheep.  相似文献

The growth, using freshly cultured Escherichia coli with ampicillin or heat-treated lyophilised E coli as a food source, of the larvae of the mouse nematode Nematospiroides dubius and the infectivity of resulting third stage larvae were determined. Concentrations of E coli between 0.5 and 1 mg dry weight ml-1 permitted optimal larval development for both N dubius and Trichostrongylus colubriformis. Development of larvae of susceptible and cambendazole-resistant strains of Haemonchus contortus in thiabendazole solutions showed clear differences between the strains and the larval development test was more sensitive than the egg hatch test. The test also detected a levamisole resistant strain of H contortus, although the degree of resistance could not be adequately measured. It is concluded that the test can be run with any anthelmintic to which resistance is suspected.  相似文献

The effects of both natural and artificial conditioning of pre-infective and infective stages of Haemonchus contortus were examined in two studies with 108 crossbred lambs, using an Ohio isolate of H. contortus known to exhibit up to 100% winter hypobiosis. The intensity of hypobiosis varied from 0 to 36% in 17 lamb groups given larvae subjected to various temperature and photoperiod conditions. The results in lambs given larvae conditioned for 4-8 weeks were not significantly different from those in lambs given freshly cultured larvae. It is suggested that winter hypobiosis of H. contortus in the northern U.S.A. is an obligatory survival mechanism that occurs without the need for external stimuli to trigger the onset of hypobiosis.  相似文献

Florida Native lambs, less than 6 months of age, were successfully vaccinated against Haemonchus contortus with a high mol. wt fraction (greater than 30,000 daltons) derived from a somatic extract of H. contortus larvae (SEL) and excretions and secretions (ES) of larvae isolated during in vitro development from the infective 3rd to 4th stage. A 59% reduction in adult worm numbers was obtained in vaccinates compared to naive lambs following challenge. The protection in vaccinated lambs was similar to that seen in lambs exposed to a primary infection of H. contortus larvae which had been cleared with anthelmintic prior to the challenge infection. The unfractionated SEL/ES preparation and a low mol. wt fraction gave no significant protection against challenge infection.  相似文献
