Torque teno sus virus (TTSuV) is emergent in swine herds. Recent studies have shown an increased frequency of TTSuV2 in Porcine circovirus type 2 (PCV2)-associated diseases (PCVAD), which are endemic in many swine-producing countries, including Brazil. Coinfection with several other viral and bacterial agents results in an increased incidence of more severe PCVAD. Given the limited information on TTSuV and PCV2 coinfection, especially in Brazilian swine herds, this study made a preliminary estimation of the occurrence of coinfection in swine herds by testing samples from different categories. Between 2008 and 2009, 111 samples of feces and 23 serum samples from 5 swine herds were tested for PCV2 and TTSuVs and the results analyzed for associations between these agents. No significant differences in coinfection frequency were observed for PCV2 + TTSuV1 or for PCV2 + TTSuV2 between nursery piglets (P = 0.730), growing pigs (P = 0.331), or sows (P = 0.472). However, a significant difference was observed for PCV2 + TTSuV1 + TTSuV2 between nursery piglets and growing pigs (P = 0.004; Fisher’s exact test). Phylogenetic studies agreed with the grouping of TTSuV1 and TTSuV2 into 2 different clades, with no distinct pattern of clustering of these isolates with the animal categories.  相似文献

Porcine teschovirus (PTV), Porcine sapelovirus (PSV) and Enterovirus G (EV-G) have been associated with enteric, respiratory, reproductive and neurological disorders. Although Brazil is the world’s fourth largest producer and exporter of pork, no information on the occurrence of PTV, PSV and EV-G infections is available for Brazilian pig herds. This study aimed to investigate the occurrence of Porcine enteric picornavirus infections in pig farms located in three distinct geographical regions of Brazil. Forty randomly selected diarrhoeic and normal consistency faeces of suckling (n?=?22) and nursery (n?=?18) pigs from farms located in 21 distinct cities of the Southern, Southeast, and Midwest regions of Brazil were evaluated by nested-RT-PCR assays. Suckling piglets presented the expected amplicon size for PTV (158 bp) and EV-G (313 bp) in single and mixed infections in 40.9 % (9/22) of the faecal samples. PSV amplicon (212 bp) was not detected in this age group. For nursery pigs, Porcine enteric picornaviruses amplicons were present in 77.8 % (14/18) of the faecal samples. PTV and EV-G were detected in single and mixed infections, while PSV was detected only in two samples in co-infection with PTV and EV-G in this age group. The Brazilian regions evaluated presented at least two of the tested viruses. Sequencing analysis revealed high similarities to the related viruses (95.3 to 99.2 % for PTV, 94.2 to 98.5 % for PSV and 86 to 100 % for EV-G). For the first time PTV, PSV and EV-G have been molecularly detected and characterised in pig faecal samples in Brazil.  相似文献

Seneca Valley virus (SVV) is the etiological agent of vesicular disease in pigs, clinically indistinguishable of classical viral vesicular infections, including foot-and-mouth disease. The first outbreaks of SVV infection in Brazil were reported in 2014. However, it was not known whether the virus was circulating in Brazilian pig herds before this year. This study is a retrospective serological investigation of porcine health status to SVV in Brazil. Serum samples (n = 594) were grouped in before (2007–2013, n = 347) and after (2014–2016, n = 247) SVV outbreaks in Brazil. Twenty-three pig herds were analyzed, of which 19 and 4 were sampled before and after the beginning of SVV outbreaks, respectively. Two herds sampled after 2014 presented animals with SVV-associated clinical manifestations, while the other two housed asymptomatic pigs. Anti-SVV antibodies were evaluated by virus neutralization test. The results demonstrated that pig herds of different Brazilian geographical regions and distinct pig categories were negative to anti-SVV antibodies in sera obtained before 2014. Antibodies to SVV were detected only in serum samples obtained after 2014, particularly in herds with the presence of pigs with SVV-clinical signs. These results present robust serological evidence that the SVV was not present in the major Brazilian pig producing regions prior to 2014.  相似文献

为分析我国猪细环病毒(TTSuV)和猪圆环病毒2型(PCV2)的共感染情况,利用所设计的TTSuV(TTSuV1、TTSuV2)和PCV2特异性引物对我国29个省市采集的猪群血清样品同时进行PCV2和TTSuV(TTSuV1、TTSuV2) PCR检测,分析混合感染情况.结果所检测的1898份样品中,TTSuV阳性为1103份(58%),PCV2阳性为435份(23%).阳性样品中呈混合感染的有275份(14%),其中TTSuV1和PCV2为249份(13%),TTSuV2和PCV2为200份(10%),均为阳性的有174份(9%).调查结果显示,我国猪群中TTSuV和PCV2混合感染现象较为普遍,对地区性分布特征和饲养模式等影响因素的分析表明TTSuV和PCV2混合感染情况存在地区性差异(P<0.01),但饲养模式并不是共感染的关键因素.  相似文献

Senecavirus A (SVA) has been associated with vesicular disease in weaned and adult pigs and with high mortality of newborn piglets. This study aimed to establish a nested-PCR assay for the routine diagnosis of SVA infection. Tissue samples (n = 177) were collected from 37 piglets of 18 pig farms located in four different Brazilian states. For the nested-PCR, a primer set was defined to amplify an internal VP1 fragment of 316 bp of SVA genome. Of the 37 piglets, 15 (40.5%) and 23 (62.2%) were positive for the SVA in the RT-PCR and nested-PCR assays, respectively. The SVA RNA was detected in 61/177 (34.5%) samples with the RT-PCR, while the nested-PCR assay showed 84/177 (47.5%) samples with the virus (p < 0.05). According to the herds, 11 (61.1%) and 16 (88.9%) of the 18 pig herds were positive for the SVA in the RT-PCR and nested-PCR assays, respectively. Nucleotide sequencing analysis revealed similarities of 98.7–100% among SVA Brazilian strains and of 86.6–98% with SVA strains from other countries. The nested-PCR assay in this study was suitable to recover the SVA RNA in biological specimens, piglets, and/or herds that were considered as negative in the RT-PCR assay, and is proposed for the routine investigation of the SVA infection in piglets, especially when other techniques are not available or when a great number of samples has to be examined.  相似文献

This study was performed to clarify the sow-to-fetus transmission pathway of Torque teno sus virus (TTSuV) types 1 (TTSuV1) and 2 (TTSuV2). For this purpose, detection of TTSuV1 and TTSuV2 (TTSuVs) in sera of 6 sows (Sows 1-6) at parturition and in sera of their newborn piglets immediately after birth without suckling colostrum was performed by nested polymerase chain reaction (nPCR). These sows were bred using semen that had tested negative for TTSuVs. In a TTSuV1- and TTSuV2-positive sow (Sow 1), TTSuV1 and TTSuV2 were detected in 4 and 5 of 12 newborn littermates, respectively. In a TTSuV1-positive sow (Sow 2), TTSuV1 was detected in 1 of 8 newborn littermates. In 4 TTSuV1- and TTSuV2-negative sows (Sows 3-6), TTSuV1 was detected in 6 out of the 25 newborn piglets of 3 sows (Sows 3-5), while TTSuVs were not detected in all 13 piglets of 1 sow (Sow 6). In addition, to investigate the possibility of a sow-to-piglet transmission pathway of TTSuV via colostrum, TTSuV1 and TTSuV2 in sera of 12 newborn piglets from Sows 1-3 were examined by nPCR. Immediately after birth without suckling colostrum, TTSuV1 and TTSuV2 were not detected in 10 and 8 of 12 newborn piglets, respectively; however, at 24 hr after suckling colostrum, TTSuV1 was detected in 6 piglets, while TTSuV2 was not detected in any piglets. These results confirmed the existence of a sow-to-fetus transmission pathway of TTSuV during normal pregnancy and suggested a possibility of sow-to-piglet transmission of TTSuV via colostrum.  相似文献

Bovine coronavirus (BCoV) is one of the main causes of neonatal calf diarrhoea. Several diagnostic assays have been employed to detect the presence of the virus in stool samples from calves. Despite this, the frequency of BCoV infection among Brazilian and even South American cattle herds has yet to be well characterised. This study describes the occurrence of BCoV infection among calves from dairy and beef herds in four Brazilian states. A total of 282 stool samples from 1 to 60-day-old calves were evaluated for the presence of BCoV by a semi-nested (SN) PCR assay. The animals were from herds (n = 23) located in three geographical regions in Brazil (south, southeast, and center-west). The specific BCoV amplicon was detected in 15.6% (44/282) of the faecal specimens examined, of which 95.4% (42/44) were from diarrhoeic and 4.6% (2/44) from asymptomatic calves. The specificity of the SN-PCR amplicons was evaluated by restriction fragment length polymorphism (RFLP) analysis. The results show that the BCoV is widespread, mainly among calves from 16 to 30-days-old (p = 0.0023), and verify the association between BCoV infection and clinical signs of diarrhoea (p = 0.005). These findings emphasise the importance of this virus in enteric infections of Brazilian cattle herds.  相似文献

Torque Teno Sus Virus (TTSuV) viral load in accordance with the Porcine Circovirus Type 2 (PCV2) infection and age of two pig herds was determined. A total of 100 and 155 piglets from two farms were divided into two groups. Half of the piglets from each farm were given a commercial vaccine against PCV2 (vaccinated group), and the remaining piglets were used as a control group (non-vaccinated group). An early onset and a chronic viremic state of TTSuV genogroups were present regardless of the PCV2 infection, and there were no significant differences in the dynamics of TTSuV genogroups.  相似文献

猪细环病毒1型是近年来发现的又一种环状DNA病毒,可能存在潜在致病作用。为了初步了解其流行情况,用PCR技术和ELISA技术对来自京津冀、华东与华南地区578头保育猪血清样品进行了检测。结果显示,猪细环病毒1型的核酸与抗体的猪场阳性率均为100%;核酸阳性率为73.88%,抗体阳性率为78.72%,二者之间无显著差异(P>0.05)。进一步分析发现,猪细环病毒1型的核酸阳性率和抗体阳性率都超过50%的猪场达到86.67%,都超过80%的占53.33%。调查结果表明,猪细环病毒1型感染在我国部分地区普遍流行,其在多数猪场的感染率很高。  相似文献

Background

In recent years, the occurrence and the relevance of Mycoplasma hyopneumoniae infections in suckling pigs has been examined in several studies. Whereas most of these studies were focused on sole prevalence estimation within different age groups, follow-up of infected piglets or assessment of pathological findings, none of the studies included a detailed analysis of individual and environmental risk factors. Therefore, the aim of the present study was to investigate the frequency of M. hyopneumoniae infections in suckling pigs of endemically infected herds and to identify individual risk factors potentially influencing the infection status of suckling pigs at the age of weaning.

Results

The animal level prevalence of M. hyopneumoniae infections in suckling pigs examined in three conventional pig breeding herds was 3.6% (41/1127) at the time of weaning. A prevalence of 1.2% was found in the same pigs at the end of their nursery period. In a multivariable Poisson regression model it was found that incidence rate ratios (IRR) for suckling pigs are significantly lower than 1 when teeth grinding was conducted (IRR: 0.10). Moreover, high temperatures in the piglet nest during the first two weeks of life (occasionally >40°C) were associated with a decrease of the probability of an infection (IRR: 0.23-0.40). Contrary, the application of PCV2 vaccines to piglets was associated with an increased infection risk (IRR: 9.72).

Conclusions

Since single infected piglets are supposed to act as initiators for the transmission of this pathogen in nursery and fattening pigs, the elimination of the risk factors described in this study should help to reduce the incidence rate of M. hyopneumoniae infections and thereby might contribute to a reduced probability of high prevalences in older pigs.  相似文献

Porcine circovirus type 2 (PCV2) can be vertically transmitted resulting in fetal infection with or without clinical signs and lesions. The primary objective of this study was to assess the prevalence of intrauterine PCV2 infection in clinically normal newborn piglets in conventional pork production facilities. Five commercial breeding herds located in the U.S. and Mexico were included in the study. A total of 125 sows and 3-5 neonatal piglets per sow were arbitrarily selected. Blood and colostrum samples were collected from sows. Blood was collected from piglets prior to suckling. All samples were analyzed for the presence of anti-PCV2 IgG antibodies and presence and amount of PCV2 DNA. In addition, PCV2 DNA positive samples were further subtyped into PCV2a and PCV2b. All (125/125) sow colostrum samples and 96.8% (121/125) of the sow serum samples and 21.4% (107/499) of the piglet pre-suckle serum samples were positive for anti-PCV2 IgG antibody. The overall PCV2 DNA prevalence was 47.2% (59/125) in sow serum, 40.8% (51/125) in sow colostrum, and 39.9% (199/499) in pre-suckle piglet serum. In the PCV2 DNA positive samples, PCV2b was detected at a higher frequency (69.5% for sow serum, 84.3% for sow colostrum, and 74.4% for piglet serum) compared to PCV2a (18.6% for sow serum, 9.8% for sow colostrum, and 15.6% for piglet serum). Concurrent PCV2a and PCV2b infection was detected in 11.9% of the sow serum, in 5.9% of the colostrum samples, and in 10.0% of the piglet serum samples. In conclusion, an unexpectedly high prevalence of PCV2 viremia was detected in healthy sows (serum and colostrum) and their pre-suckle piglets in the five breeding herds investigated and PCV2b was more prevalent than PCV2a. This information adds to the knowledge of PCV2 infection in breeding herds.  相似文献

Active hepatitis E virus (HEV) infections in two Brazilian swine herds were investigated. In study 1, 26 piglets born to five anti-HEV positive sows were monitored from birth to post-partum week 22. Serum samples were screened for the detection of anti-HEV antibodies and a nested RT-PCR used to examine the HEV genome. Passive transfer of immunity was confirmed. At week 22, 23/26 (88.4%) of the piglets had seroconverted. Genome amplification was achieved in a feces pool from one holding pen and in one serum sample, both from 13-week-old animals. Histology was suggestive of a potential HEV infection. In the second study, 47 piglets born to six anti-HEV-positive sows were monitored after weaning. Seroconversion was determined in eight animals at 6-8 weeks of age. HEV RNA was detected in two pools from a holding pen for 12-16-week-old animals. Brazilian isolates were classified as genotype 3. This is the first molecular evidence of HEV infection in Brazilian pig herds.  相似文献

Faecal samples from 684 litters of suckling piglets from 100 indoor swine herds from all regions of Norway were examined for the presence of Cryptosporidium and Giardia, using sucrose gradient flotation concentration and immunofluorescent staining. Thirty-one (31%) herds and 57 (8.3%) litters tested positive for Cryptosporidium, while 10 (1.5%) litters in 10 different herds (10%) tested positive for Giardia. Molecular characterisation of nine Cryptosporidium isolates demonstrated both C. suis and Cryptosporidium sp. pig genotype II. There was significantly more diarrhoea among the Cryptosporidium positive piglets than among the Cryptosporidium negative piglets. Diarrhoea was not observed amongst litters in which Cryptosporidium sp. pig genotype II was found. There was a significant difference in the Cryptosporidium prevalence between litters from different geographical areas of Norway. This study demonstrates that both Cryptosporidium (C. suis and Cryptosporidium sp. pig genotype II) and Giardia infections are prevalent among suckling piglets in Norway.  相似文献

We detected Torque teno sus virus 1 and 2 (TTSuV1 and TTSuV2) in tissue samples from 18 stillborn piglets using nested polymerase chain reaction. The detection rates of TTSuV1 and TTSuV2 were 78% and 50%, respectively, with 83% of the stillborn piglets positive for TTSuV1 or TTSuV2. TTSuV1 was detected highest in the liver (72%) followed by heart (56%), spleen (38%) and tonsils (38%) while TTSuV2 was detected highest in the tonsils (38%) followed by liver (33%), spleen (25%), and heart (17%). These results indicate that TTSuVs are commonly present but not equally distributed among the tissues of stillborn piglets.  相似文献

Bovine viral diarrhea virus (BVDV) infects ruminants as primary hosts. However, other animals like pigs are susceptible. This study was conducted to investigate seroprevalence and risk factors associated with the detection of BVDV antibodies in pig herds. A total of 1.705 serum samples of 33 finisher herds, from seven Brazilian states, were collected in slaughterhouses. The samples were tested by virus neutralization (VN) test. In total, 5.35% (91/1.705) were positive and 64% (21/33) of the herds had positive animals. A significant association with “trucks are not cleaned and disinfected” and “visitors do not respect 72-h interval between visits to farms” (P?<?0.05) was found in association with detection of BVDV-2 antibodies. This study suggests that important biosecurity gaps are present in Brazilian pig farms, as the presence of BVDV antibodies in pigs suggests (direct or indirect) contact with population(s) of ruminant species. Closing biosecurity gaps prevents spread of BVDV and other pathogens such as foot-and-mouth disease virus (FMDV) between pig and ruminant farms. This data should be taken in account by CSF surveillance programs, once cross-reaction in serologic tests between classical swine fever virus (CSFV) and BVDV antibodies has been shown to occur.  相似文献

为了解猪细环病毒(Torque teno sus virus, TTSuVs)在吉林部分规模化猪场的流行及其遗传变异情况。本研究对2017-2018年间,来自吉林地区10家规模化猪场的574份血清样本进行TTSuVs流行情况调查。并根据收地点,对20株TTSuVs进行ORF1全基因序列扩增及遗传变异性分析。结果显示:TTSuV1感染率为35.0%;TTSuV2感染率为63.2%;TTSuV1与TTSuV2混合感染率为30.3%;10株TTSuV1测序株之间的核苷酸同源性为75.7%-94.7%,氨基酸同源性为48.1%-88.2%;10株TTSuV2测序株之间的核苷酸同源性为77.5%-93.7%,氨基酸同源性为70.6%-87.4%;且吉林地区流行的TTSuV1均属于TTSuV1-lb型,而TTSuV2流行情况较为复杂,各种亚型均有涉及。  相似文献

The frequency of group A bovine rotavirus (gpA BRV) in calves from 1998 to 2002 was determined by the polyacrylamide gel electrophoresis technique in 2177 faecal samples, of which 1898 samples were diarrhoeic and 279 were of normal consistency (control group) that were collected from asymptomatic calves for comparative purposes. The animals were from beef and dairy cattle herds (n = 321) from 158 counties in seven States (Paraná, São Paulo, Minas Gerais, Mato Grosso do Sul, Mato Grosso, Goiás and Rondônia) and four Brazilian geographical regions (south, south-east, centre-west, and north). GpA BRV was detected in 19.4% (369/1898; p = 0.0001) of the samples collected in calves with diarrhoea and in only 2.2% (6/279; p = 0.0001) of the faeces with normal consistency. The proportion of positive samples collected from beef and dairy cattle herds was 22.8% (205/899; p = 0.0001) and 16.4% (169/999; p = 0.0005), respectively. In relation to age, a higher prevalence of infections was found in calves up to 30 days old, where 33.0% (189/573; p = 0.0001) and 20.2% (138/683; p = 0.0001) of the diarrhoeic faecal samples from beef and dairy cattle herds, respectively, were positive for gpA BRV. These results show the possible importance of inclusion of gpA BRV in the management of neonatal calf diarrhoea in Brazilian cattle herds.  相似文献

Sapovirus is one genus within Caliciviridae family that causes diarrhea in humans and animals. Sapovirus (SaV) has been classified into seven genogroups (GI to GVII). The GIII, GVI, and GVII, which prototype is Cowden, JJ681, and K7/JP strains, respectively, infect pigs. The objective of this study was to characterize wild-type Brazilian SaV strains from piglet stool samples and determine SaV infection frequency, age distribution and association with diarrheic disease. Stool samples from 113 piglets up to 28-days-old were collected from 34 pig farms located in the States of Minas Gerais (MG), Mato Grosso do Sul (MS), Paraná (PR), Santa Catarina (SC), and Rio Grande do Sul (RS), during 2004 and 2005. The specimens were evaluated for enteric calicivirus by RT-PCR assay with primers p289/290, designed to detect the polymerase gene of SaV and norovirus. Thirty four (30.1%) samples were positive for SaV and five amplicons were sequenced. Phylogenetic analyses placed BRA29-MS/04 and BRA52-PR/05 sequences into the GIII of SaV genus. BRA04-SC/04, BRA21-RS/04, and BRA37-MG/05 demonstrated low identity with the Cowden strain but were closely related (up to 86.3%) to the Japanese and Dutch SaV strains, grouping together in a new cluster (GVIII?) in the phylogenetic tree. SaV infection was detected more frequently (p=0.0001) in animals between 22 and 28 days of age, in equal frequencies in piglets with and without diarrhea (p=0.59), and in the five Brazilian States. In this study, such as other unclassified worldwide SaVs, the Brazilian strains showed high genetic variability. Furthermore, the distribution and frequency of SaV infection provides evidence that the virus is circulating in Brazilian pig herds.  相似文献
